Description
KAISERLICHES
PATENTAMT.
Die den Gegenstand des Patents 108223 bildenden Alkalimetallverbindungen der y-Oxypiperidine
sind aufserordentlich reactionsfähige Körper. Durch Einwirkung von Halogenalkylen
oder Sä'urechloriden, Halogenfettsäure-
CH-ONa
estern, Harnstoffchloriden etc. erhält man die entsprechenden Hydroxylwasserstoffsubstitutionsproducte
der Alkamine bezw. Alkamincarbonsäureester gemäfs folgender Gleichung:

A. Einführung von Alkylen.
ι. Darstellung des Methyläthers des n-Methyltriacetonalkamins.
Man versetzt die ätherische Lösung des Natriumalkaminats aus 171g n-Methyltriacetonalkamin
mit 142 g Jodmethyl und kocht längere Zeit am Rückflufskühler. " Die ätherische Lösung
wird sodann mit verdünnter Sodalösung geschüttelt. Nach dem Abdunsten des Aethers

hinterbleibt der Methyläther des η - Methyltriacetonalkamins als dünnflüssiges, piperidinähnlich
riechendes OeI vom Siedepunkt 217
bis 22i°. Der Methyläther ist schwer löslich in Wasser und wird durch concentrirte Natronlauge
oder Pottasche aus der wässerigen Lösung abgeschieden; er bildet leicht lösliche, zum
Theil gut krystallisirende Salze.
Die vorstehende Reaction wird durch folgende Gleichung veranschaulicht:

In derselben Weise kann man die Aether des n-Methylvinyldiacetonalkamins und
seines geometrischen Isomeren darstellen. Der Methyläther des labilen n-Methylvinyldiacetonalkamins
ist ein gelbes, unangenehm riechendes OeI, das leicht in Alkohol und Aether,
nicht aber in Wasser löslich ist und bei gewöhnlichem Luftdruck unter Zersetzung gegen
2oo° siedet.
2. Darstellung des Methyläthers des n-Methyltetram ethyl -γ- oxypiperidincarbonsä'uremethylesters.
Man erhitzt die Lösung des im Patent 108223 erwähnten Natriumalkaminats aus 229 g n-
NaO-C-COOCHo

Methyltetramethyl -γ- oxypiperidincarbonsäuremethylesters
mit 142 g Jodmethyl längere Zeit am Rückflufskühler. Nach dem Abdestilliren
der vorerst mit Sodalösung gewaschenen ätherischen Lösung hinterbleibt der Methyläther des
ii-Methyltetramethyl-y-oxypiperidincarbonsäuremethylesters
in öliger Form. Er enthält noch kleine Mengen unveränderten η - Methyltetramethyl
-γ- oxypiperidincarbonsäuremethylesters, welche zweckmä'fsig, dadurch beseitigt werden,
dafs man das OeI kurze Zeit am Rückflufskühler mit Wasser kocht.
Die Reaction verläuft nach folgender Gleichung:

Den Methyläther des Tropins erhält man genau in der vorbeschriebenen Weise.
Er stellt ein weifses, krystallinisches Pulver dar, das über 3000 schmilzt, in Wasser leicht löslich
ist, ebenso in heifsem Alkohol, schwerer hingegen in Aether.
B. Einführung von Säureradikalen.
Darstellung von Benzoyl - η - methyltriacetonalkamin.
In die ätherische Lösung des aus 171 g n-Methyltriacetonalkamin
hergestellten Natriumalkaminats läfst man eine mit Aether verdünnte
Lösung von 140,5 g Benzoylchlorid einlaufen. Die Reaction geht momentan vor sich. Nach
dem Abdestilliren der mit verdünnter Sodalösung gewaschenen ätherischen Lösung hinterbleibt
das Benzoyl - η - methyltriacetonalkamin als dickflüssiges OeI, das durch
Waschen mit Wasser von unverändert gebliebenem η - Methyltriacetonalkamin getrennt
wird.
In derselben Weise erhält man z. B. das Cinnamyl - η -methyltriacetonalkamin,
indem man Zimmtsäurechlorid auf das entsprechende Alkaminat einwirken läfst. Es bildet
ein farbloses, dickes OeI, das gegen 2600 siedet.
Durch Einwirkung von Chlorkohlensäureäther erhält man das Aethylcarbonat des
n-Methyltriacetonalkarßins. Es bildet
weifse Krystalle vom Schmelzpunkt 2630, die sich leicht in Alkohol lösen, woraus sie durch
Aether wieder abgeschieden werden.
Zu den analogen Derivaten des n-Methylvinyldiacetonalkamins, sowie des Tropins kann
man in der gleichen Weise gelangen.
Die Benzoyl-n-methyl-Verbindung des
bei i6i° schmelzenden Vinyldiacetonalkamins
(vergl. auch Patent 95620) ist ein roth-CH3
CH,
C/CH,
C\CH
NaJ.
N-CH
braunes, dickes OeI von nicht unangenehmem Geruch, das in Wasser zu Boden sinkt, in
Alkohol und in Aether sehr leicht löslich ist. Der Destillation unterworfen, geht es unter
Zersetzung oberhalb 2500 über.
Das Benzoyltropin stellt ein gelbes, klares
OeI von stechendem Geruch dar; es besitzt sonst dieselben Eigenschaften wie das vorbev
schriebene Benzoyl - η - methylvinyldiacetonalkamin. Bei 175 bis i8o° destillirt, bildet es
ein farbloses, dünnflüssiges OeI, das beim Kochen mit Wasser Zersetzung erleidet.
Auch die Urethane lassen sich sehr bequem in der Weise erhalten, dafs man Harnstoffchloride
auf die Alkaminate einwirken läfst.
Beispiel: Man versetzt die ätherische Lösung des aus 171 g η - Methyltriacetonalkamin
hergestellten Natriumalkaminats mit 155,5 g
Phenylharnstoffchlorid und erhitzt längere Zeit am Rückflufskühler. Nach dem Abdestilliren
der mit Soda gewaschenen ätherischen Lösung hinterbleibt das Pheny Iu r ethan des n-Methyltriacetonalk
amins als ein dickflüssiges OeI. Durch Krystallisation aus Benzol-Ligro'in
erhält man das Phenylurethan in nadeiförmigen Krystallen, die bei 111 bis 1130
schmelzen. Die Verbindung entspricht der Formel:

In derselben Weise erhält man aus dem Alkaminat des η - Methyltetramethyl - γ - oxypiperidincarbonsäuremethylesters
und Phenyl-
harnstoffchlorid das entsprechende Urethan
von der Formel:

CH.
Es schmilzt bei 132 bis 1330. Der Körper ist
unter die in dem Patent 92589 beschriebenen Verbindungen einzureihen.
Die Hydroxylwasserstoffsubstitutionsproducte
der Alkamine sollen medicinische Verwendung finden.
Claims (1)
- Patent-Anspruch:Verfahren zur Darstellung von in der Hydroxylgruppe substituirten Alkaminen und AIkamincarbonsäureestern, darin bestehend, dafs man in den den Gegenstand des Patents 108223 bildenden Alkalimetallverbindungen von y-Oxypiperidinen für das Alkalimetallatom ein geeignetes Radikal einführt.
