Abstract
Ein b-Lactam der Formel I: wobei R1 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist; R2 Wasserstoff, Alkyl, Acyl, Acetal, Ethoxyethyl oder eine andere Hydroxyl-Schutzgruppe ist; und R3 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist; und ein Verfahren zur Herstellung von Taxol, bei dem besagtes b-Lactam und ein Alkohol in Gegenwart eines Aktivierungsmittels miteinander in Kontakt gebracht werden zur Herstellung einer Taxol-Zwischenverbindung und Umwandlung der Zwischenverbindung in Taxol. wobei besagtes A, B, D, E, F, G, I, J, K, L, M, P und Q wie oben definiert sind, der Kontakt zwischen besagtem ß-Lactam und besagtem Alkohol in Gegenwart einer ausreichenden Menge Aktivierungsmittels unter effektiven Bedingungen erfolgt, damit das ß-Lactam mit dem Alkohol reagieren kann und ein ß-Amidoester gebildet wird, der als Zwischenverbindung bei der Taxolsynthese eingesetzt werden kann, und die besagte Zwischenverbindung in Taxol umgewandelt wird
18. Verfahren gemäß Anspruch 17, wobei der besagte Alkohol folgende Formel hat:

19. Verfahren gemäß Anspruch 18, wobei R4 aus Ethern, Estern, Carbonaten und Silylgruppen ausgewählt wird.
20. Verfahren gemäß Anspruch 18, wobei besagtes Ri Phenyl und besagtes R3 Phenyl ist.
21. Verfahren gemäß Anspruch 18, wobei R4 Ethoxyethyl, Trimethylsilyl od3rTriethylsilyl ist.
22. Verfahren gemäß Anspruch 17, wobei besagtes Aktivierungsmittel Triethylamin, Diisopropylethylamin, Pyridin, N-Methylimidazol oder4-Dimethylaminopyridin ist.
Die vorliegende Erfindung bezieht sich auf neuartige ß-Lactame, ein Verfahren zu deren Herstellung und ein Verfahren zur Herstellung von Taxol unter Verwendung solcher ß-Lactame.
Die Taxanfamilie der Terpene, zu der Taxol gehört, hat sowohl in der biologischen als auch in der chemischen Wissenschaft beträchtliche Aufmerksamkeit erregt. Ta> _>l ist ein vielversprechendes Krebschcjmotherapeutikum mit einem breiten Spektrum an leukämie- und tumorhemmenden Wirkungen und weist folgende Struktur auf:

Aufgrund dieser vielversprechenden Wirkung werden gegenwärtig in Frankreich und den USA klinische Tests mit Taxol durchgeführt.
Für diese klinischen Versuche wird das Taxol gegenwärtig aus der Borke verschiedener Eibenarten gewonnen. Taxol kommt jedoch nur in sehr kleinen Mengen in der Borke dieser langsam wachsenden, immergrünen Bäume vor, so daß ernsthafte Besorgnis besteht, daß die begrenzte Bereitstellung von Taxol nicht den Bedarf decken wird. Demzufolge bemühen jich Chemiker seit einigen Jahren ernsthaft darum, eine praktizierbare Methode zur Herstellung synthetischer Taxole zu entwickeln.
Bisher können die Ergebnisse jedoch insgesamt nichi befriedigen.
Eine vorgeschlagene Methode bezieht sich auf die Synthese des tetracyclischenTaxankerns aus Grundchemikalien. Holten et al.
berichtete in JACS 110, 6558 (1988) über die Synthese des Taxols, eines Artverwandten des Taxusin. Trotz der bei diesem Verfahren erzielten Fortschritte stellt die Vollsynthese des Taxols einen mehrstufigen, langwierigen und kostspieligen Prozeß
Ein alternatives Herangehen an die Taxolherstellung wurde von Greene et al. in JACS 110,5917 (1988) beschrieben, bei dem ein Artverwandter des Taxols, das 10-Deacetyl-Baccatin III, eingesetzt wird, das folgende Struktur aufweist:

Das lO-Deacatyl-Baccatin III ist besser verfügbar als Taxol, da es aus den Blättern des Taxus baccata gewonnen wird. Entsprechend der Methode von Greene et ai. wird das lO-Deacetyl-Baccatin III durch Anlagerung der ClO-Acetylgruppe und der C 13-Amidoester-Seitenkette durch Veresterung der C-13-Ott-Gruppe mit einer ß-Amido-Carbonsäureeinheit in Taxol umgewandelt. Obwohl dieses Herangehen relativ wenige Schritte umfaßt, ist die Synthese der ß-Amido-Carbonsäureeinheit ein mehrstufiger Prozeß mit geringer Ausbeute. Die Kupplungsreaktion ist langwierig und verläuft mit geringer Ausbeute. Diese Kupplungsreaktion stellt jedoch einen wichtigen Schritt dar, der bei jeder Synthese von Taxol oder eines biologisch aktiven Derivats des Taxol erforderlich ist, da die Anwesenheit der ß-Amidoester-Seitenkette am C13 für die Antitumorwirkung unerläßlich ist, wie von Wani et al. in JACS 93, 2325 (1971) nachgewiesen wurde.
Ein weiterhin bestehendes Hauptproblem bei der Synthese von Taxol und anderen potentiellen Antitumormitteln besteht im Fehlen einer leicht verfügbaren Einheit, die problemlos an den C 13-Sauerstoff angelagert werden könnte, um die ß-Amidoester-Seitenkette zu gewährleisten.
Die Entwicklung einer solchen Einheit und eines Verfahrens zu deren Anlagerung mit hoher Ausbeute würde die Sythese von Taxol und verwand'er Antitumormittel ermöglichen, die eine modifizierte Gruppe von Kern-Substituenten oder eine modifizierte C13-Seitenkette aufweisen. Dies wurde möolich durch die Entdeckung einer neuen, leicht verfügbaren chemischen Einheit einer Seitenkettevorstufe und eines leistungsfähigen Verfahrens zu dessen Anlagerung an den C 13-Sauerstoff.
Abriß der Erfindung
Zu den Zielen der vorliegenden Erfindung zählt demgemäß die Herstellung einer Seitenkettevorstufe für die Synthese von Taxolen und die Bereitstellung eines Verfahrens zur Anlagerung der Seitenkettevorstufe mit relativ hoher Ausbeute zur Erzeugung einer Taxol-Zwischenverbinduno.
Kurz ausgedrückt bezieht sich die vorliegende Erfindung auf eine Seitenkettevorstufe, ein ß-Lactam 1 mit folgender Formel: O
wobei Ri Aryl, substituiertem Aryl, Alkyl, Alkenyl oder Alkinyl ist; R2 Wasserstoff, Ethoxyethyl, Acetal oder eine andere Hydroxyl-Schutzgruppe ist; und R3 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist.
Die vorliegende Erfindung bezieht sich auch auf ein Verfahren zur Herstellung einer Taxol-Zwischenverbindung, bei dem ein Alkohol in Gegenwart einer ausreichenden Menge eines Aktivators unter effektiven Bedingungen mit ß-Lactam 1 in Kontakt gebracht wird, damit das ß-Lactam mit dem Alkohol reagiert und ein ß-Amidoester gebildet wird, der als Zwischenverbindung in der Taxolsynthese eingesetzt werden kann.
Die vorliegende Erfindung bezieht sich außerdem auf ein Verfahren zur Herstellung von Taxol, bei dem ein Alkohol in Gegenwart einer ausreichenden Menge eines Aktivators unter effektiven Bedingungen mit ß-Lactam 1 in Kontakt gebracht wird, damit das ß-Lactam mit dem Alkohol reagiert und die Zwischenverbindung eines ß-Amidoestertaxols gebildet wird. Die . Zwischenverbindung wird dann bei der Taxolsynthese eingesetzt.
Weitere Gegenstände und Merkmale dieser Erfindung werden zum Teil offensichtlich und zum Teil nachfolgend erläutert sein.
Detaillierte Beschreibung
Die vorliegende Erfindung bezieht sich auf ein ß-Lactam 1 und dessen Derivate, dessen Struktur nachfolgend dargestellt ist.

Wie oben dargestellt, ist R1 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl; R2 Wasserstoff, Ethoxyethyl, Acetal oder eine andere Hydroxyl-Schutzgruppe; und R3 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl. Gemäß den lUPAC-Regeln lauten die Bezeichnungen der ß-Lactame 2 und 3 1-8enzoyl-4-phenyl-3-{1-ethoxyethoxy)azetidin-2-on 2 und 1-3enzoyl-4-phenyl-3-(2,2,2-trichlorethoxymethyl)azetidin-2-on 3. Am meisten wird das ß-Lactam 2 bevorzugt. In Übereinstimmung mit der vorliegenden Erfindung wird ein Verfahren zur Herstellung von Taxol-Zwischenverbindungen, natürlichem Taxol und nicht natürlich vorkommenden Taxolen vorgestellt, die folgende Strukturformel aufweisen:

Die Alkylgruppen des Taxols sind entweder allein oder mit den verschiedenen, oben dargelegten Substituenten vorzugsweise niederes Alkyl, das ein bis sechs Kohlenstoffatome in der Hauptkette und bis zu 10 Kohlenstoffatome enthalt. Die Aryl-Anteile des Taxols enthalten allein oder mit den verschiedenen Subsxituenten 6 bis 10 Kohlenstoffatome sowie Phenyl, α-Naphthyl, ß-Naphthyl usw.
Zu den Substituenten gehören Alkanoxy, Hydroxy, Halogen, Alkyl, Aryl, Alkenyl, Acyl, Acyloxy, Nitro-, Amino-, Amidogruppen usw. Phenyl stellt das stärker bevorzugte Aryl dar.

Gemäß dem erfindungsgemäßen Verfahren werden ß-Lactsme 1 in Gegenwart eines Alkohols und eines Akiivierungsmittels, vorzugsweise eines tertiären Amins, wie z.B. Triethylamin, Diisopropylethylamin, Pyridin, N-Methylimidazol und 4-Dimethylaminopyridin (DMAP) in ß-Amidoester umgewandelt. Beispielweise reagieren ß-Lactame 1 in Gegenwart von 4-Dimethylaminopyridin (DMAP) mit Verbindungen, die einen tetracyclischen Taxankern und eine C13-Hydroxylgruppe haben, so daß Substanzen mit einer ß-Amidoestergruppe bei C13 entstehen.
Bei dem Alkohol sollte es sich vorzugsweise um 7-0-Triothylsilyl-Baccatin III handeln, dessen Herstellung von Greene et al. in JACS 110,5917 (1988) beschrieben wurde oder auf andere Weise erfolgen kann. Nach Greene et al. wird 10-Dercetyl-Baccatin III gemäß dem folgenden Reaktionsschema in 7-0-Triethylsilyl-Baccatin III umgewandelt.

Wie dargelegt wird, reagiert das 10-Deacetyl-Baccatin III unter sorgfältig zu optimierenden Bedingungen mit 20 Äquivalenten von (C2H5I3SiCI bei 23°C in Argonatmosphäre in 20 Stunden in Gegenwart von 50 ml Pyridin/mmol 10-Deacetyl-Baccatin III, ν. odurch 7-Triethylsilyl-10-deacetyl-Baccatin III (32 a) als Reaktionsprodukt bei einer Ausbeute von 84-86% nach der Reinigung entsteht. Das Reaktionsprodukt wird dann mit 5 Äquivalenten CH3COCI und 25 ml Pyridin/mmol der Verbindung 32 a bei 0°C in Argonatmosphäre 48 Stunden acetyliert unter Bildung von 7-0-Triethylsilyl-Baccatin III (32 b) mit einer Ausbeute von 86%. Greene et al. in JACS 110, 5917, unter 5918 (1988).
Wie im folgenden Reaktionsschema dargestellt, kann 7-0-Triethylsilyl-Baccatin III (32 b) bei Raumtemperatur mit einem ß-Lactam der vorliegenden Erfindung unter Bildung einer Taxolzwischenverbindung reagieren, in der die C-7- und C-2-Hydroxylgruppen durch Triethylsilyl- bzw. Ethoxyethyl-Gruppen geschützt sind. Diese Gruppen werden da/in unter milden Bedingungen hydrolysiert, um die Esterbindung oder die Taxolsubstituenten nicht zu beeinträchtigen.

Obwohl sich das vorliegende Schema auf die Synthese des natürlichen Produkts Taxol bezieht, kann es mit Modifikationen beim ß-Lactam oder tetracyclischen Alkohol natürlicher oder synthetischer Herkunft zur Herstellung anderer synthetischer Taxole im Rahmen der vorliegenden Erfindung verwendet werden.
Im anderen Falle kann ein ß-Lactam 1 in Gegenwart eines Aktivierungsmittels und eines anderen Alkohols als 7-0-Triethylsilyl-Baccatin III in einen ß-Amidoester umgewandelt werden,, um eine Taxolzwischenverbindung zu bilden. Die Taxolsynthese kann dann unter Einsatz der Taxolzwischenverbindung entsprechend einem geeigneten Reaktionsschema verlaufen. Die ß-Lactam-Alkylgruppen sind entweder allein oder mit den verschiedenen, oben dargelegten Substiuenten vorzugsweise niederes Alkyl, dasein bis sechs Kohlenstoffatome in der Hauptkette und bis zu 15 Kohlenstoffatome enthält. Die Substituenten enthalten Alkanoxy, Hydroxy, Halogen, Alkyl, Aryl, Alkenyl, Acyl, Acyloxy, Nitro, Amino und Amido usw. Phenyl stellt das stärker bevorzugte Aryl dar.
Wie oben festgestellt wurde, kann R2 des ß-Lactam 1 Alkyl, Acyl, Ethoxyethyl, 2,2,2-Trichlorethoxymethyl oder eine andere Hydroxyl-Schutzgruppe sein wie Acetale und Ether, d. h. Methoxymethyl, Benzyloxymethyl; Ester wie Acetate; Carbonate wie Methylcarbonate; o.a. Eine Vielzahl von Schutzgruppen für die Hydroxylgruppe und deren Synthese ist in „Protective Groups in Organic Synthesis" von T. W. Greene, John Wiley and Sons, 1981 aufgeführt. Die gewählte Hydroxyl-Schutzgruppe sollte leicht eliminierbar sein unter Bedingungen, die ausreichend mild sind, damit die Esterbindung oder andere Substituenten der Taxolzwischeriverbindung nicht beeinträchtigt werden. R2 ist jedoch vorzugsweise Ethoxyethyl oder 2,2,2-Trichlorethoxymethyl und am günstigsten Ethoxyethyl.

Das ß-Lactam 1 ν 'eist mehrere asymmetrische Kohlenstoffe auf, und Fachleuten ist bekannt, daß die im Rahmen der vorliegenden Erfindung behandelten Verbindungen mit asymmetrischen Kohlenstoffatomen in diastereomeren, racemischen oder optisch aktiven Formen auftreten können. Alle diese Formen gehören zum Umfang dieser Erfindung. Präziser ausgedrückt, die vorliegende Erfindung umfaßt Enantiomere, Diastereomere, racemische Gemische und andere, damit zusammenhängende Mischungen.
Die ß-Lactame 1 können aus leicht verfügbaren Materialien gewonnen werden, wie das folgende Schema für ß-Lactam 2 zeigt:

Die Ausgangsstoffe sind leicht verfügbar. a-Acyloxy-Acetylchlorid wird aus Glykolsäure gewonnen und in Gegenwart von tertiärem Amin mit Iminen, die aus Aldehyden und p-Methoxyanilin hergestellt werden, cyclokonderisiert unter Bildung von i-p-Mothoxyphenyl-S-acyloxy^-arylazetidin^-onen.
Die p-Methoxyphenylgruppe kann leicht durch Oxidation mit Ceriumammoniumnitrat eliminiert worden, und die Acyloxygruppe kann unter den bekannten Standardbedingungen hydrolisiert werden, um 3-Hydroxy-4-arylazetidin-2-one zu erhalten.
Die 3-HydroxylgrUppe kann durch eine Reihe von Standardschutzgruppen wie die 1-Ethoxyethylgruppe geschützt werden. Das racemische 3-Hydroxy-4-arylazetidin-2-on wird vorzugsweise in reine Enantiomere aufgespalten, bevor es durch Rokristallisation der entsprechenden 2-Methuxy-2-(tri(luormethyl)phenylessigester geschützt wird, wobei lediglich das rechtsdreher.de Enantiometer bei der Taxolherstellung eingesetzt wird.
Das 3-(1-Ethoxyethox/)-4-phenylazetidin-2-on kann auf jeden Fall in ß-Lactam 2 umgewandelt werden durch Behandlung mit einer Base, vorzugsweise n-Butyllithium, und einem Aroylchlorid bei -78X oder darunter.
Die folgenden Beispiele sollen die Erfindung veranschaulichen.
Herstellung von Cis-1-benzoyl-3-(1-ethoxyethoxy)-4-phenylazotidinon 2 Cis-i-p-methoxyphenyl-S-acetoxy^-phenylazetidin^-on.
Einer Lösung aus 962mg (4,56mmol) des aus Benzaldehyd und p-Me!hoxyanilin gewonnenen Imins und 0.85ml (6,07 mmol) Triethylamin in 15 ml CH2CI? mit einer Temperatur von -20°C wurde tropfenweise eine Losung aus413mg (3,04 mmol) Acetoxyacetylchlorid in 15ml CH2CI2 zugesetzt. Das Reaktionsgemisch wurde über einen Zeitraum von 18h auf 250C erwärmt. Anschließend wurde das Reaktionsgemisch mit 100ml CH2CI2 verdünnt, die Lösung wurda mit 30ml wäßriger 10%iger HCI extrahiert. Die organische Schicht wurde mit 30ml Wasser und 30 ml gesättigten wäßrigen Natriumhydrogencarbonats gewaschen, über Natriumsulfat getrocknet und zur Erzielung eines Feststoffes konzentriert. Der Feststoff wurde mit 50 ml Hexan versetzt, und das Gemisch wurde filtriert. Der verbleibende Feststoff wurde aus Ethylacetat/hexan rekristallisiert, es wurden 645mg (68%) Cis-i-p-methoxyphenyl-S-acetoxy^-phenylazetidin^-on in Form weißer Kristalle mit einem Schmelzpunkt von 163°C erhalten.
Cis-S-acetoxy^-phenylazetidin^-on. Einer Lösung aus 20,2g Cis-1-p-methoxyphenyl-3-acetoxy-4-phenylazetidin-2-on in 700ml Acetonitril mit einer Temperatur von -1O0C wurde langsam eine Lösung aus Ceriumammoniumnitrat in 450 ml Wasser über einen Zeitrcum von 1 h zugesetzt. Das Gemisch wurde bei - 10°C 30 Minuten gerührt und mit 500ml Ether verdünnt. Die wäßrige Schicht wurde zweimal mit je 100ml Ether extrahiert, und die vereinigten organischen Schichten wurden zweimal mit je 100 ml Wasser, je 100ml gesättigter, wäßriger Natriumhydrogensulfitlösung und je 100ml gesättigter, wäßriger Natriumhydrogencarbonatlösung gewaschen und nach Einengungen wurden 18,5g Feststoff erhalten. Bei der Rekristallisation des Feststoffes aus Aceton/Hexan wurden 12,3g (92%) Cis-3-acetoxy-4-phenylazetidin-2-on in Form weißer Kristalle mit einem Schmelzpunkt von 152-1540C erhalten.
Cis-S-hydroxy^-phenylazetidin^-on. Einer Mischung aus 200 ml THF und 280 ml 1 m wäßriger Kalilauge mit einer Temperatur von O0C wurde eine Lösung aus 4,59 g (22,4 mmol) Cis-S-acetoxy^-phenylazetidin^-on in 265 ml THF mit Hilfe eines Tropftrichters über einen Zeitraum von 40 Minuten zugesetzt. Die Lösung wurde bei O0C eine Stunde gerührt, dann wurden 100 ml Wasser und 100 ml gesättigten Natriumhydrogencarbonats zugegeben. Die Mischung wurde viermal mit je 200 ml Ethylacetat extrahiert, und die vereinigten organischen Schichten wurden über Natriumsulfat getrocknet und konzentriert. Es wurden 3,54 g (97 %) des racemischen Cis-3-hydroxy-4-phenylazetidin-2-on in Form weißer Kristalle mit einem Schmelzpunkt von 147-1490C erhalten. Dieses Material wurde durch Rekristallisation seines 2-Methoxy-2-(tr',', luormethyDphenylessigesters aus Hexan/Aceton und anschließende Hydrolse /x/25Hg 1770C in seine Enantiomere aufgespalten.
Cis-3-(1-ethoxyethoxy)-4-pherylazetidin-2-on. Einer Lösung aus 3,41 g (20,9mmol) Cis-3-hydroxy-4-phenylazetidin-2-on in 15ml THF mit einer Temperatur von O0C wurden 5 ml Ethylvinylether und 20 mg (0,2 mmol) Methansulfonsäure zugegeben. Die Mischung wurde bei O0C über einen Zeitraum von 20 Minuten gerührt, mit 20ml gesättigten wäßrigen Natriumhydrogencarbonats verdünnt und dreimal mit je 40ml Ethylacetat extrahiert. Die vereinigten Ethylacetatschichten wurden über Natriumsulfat getrocknet und konzentriert, die Ausbeute betrug 4,87 g (99%) Cis-3-(1-ethoxyethoxy)-4-phenylazetidin-2-on in Form eines farblosen Öls
Cis-1-benzoyl-3-(1-ethoxyethoxy)-4-phenylazetidin-2-on.
Einer Lösung aus 2,35g (lOmmol) Cis-3-(1-ethoxyethoxy)-4-phenylazetidin-2-on in 40ml THF mit einer Temperatur von -78°C wurden 6,1 ml (10,07mmol) einer 1,65m Lösung von n-Butyllithiumin Hexan zugegeben. Die Mischung wurde bei -78°C 10 Minuten gerührt, dann wurde eine Lösung von 1,42g (10,1 mmol) Benzoylchlorid in 10ml THFzugesetzt. Die Mischung wurde bei -78°C eine Stunde lang gerührt und mit 70 ml gesättigten wäßrigen Natriumhydrogencarbonats verdünnt und dreimal mit je 50 ml Ethylacetat extrahiert. Die vereinigten Ethylacetatextrakte wurden über Natriumsulfat getrocknet und konzentriert, die Ausbeute waren 3,45 g eines Öls. Bei der Chromatografie des Öls auf mit Ethylacetat/Hexan eluierten Kieselgel wurden 3,22 g (95%) Cis-1-benzoyl-3-(1-ethoxyethoxy)-4-phenylazetidin-1-on (2) in Form eines farblosen Öls erhalten.
Herstellung von ß-Amidoesten aus Cls-1-benzoyl-3-(1-ethoxyethoxy)-4-phenylazetldln-2-on 2 Benzyl-S-.benzamido-S-phenyl^-hydroxypropionat.
Einer Lösung aus 88mg (0,26mmol)Cis-1-benzoyl-3-(1-ethoxyethoxy)-4-phenylazetidin-2-on in 0,3ml THF wurden 28mg (0,26mmol) Benzylalkohol und 32g (0,26mmol) 4-Dimethylaminopyridin (DMAP) zugesetzt. Nach 5 Stunden wurde die Mischung bei 25°C mit 10 ml gesättigter wäßriger Natriumhydrogencarbonatlösung verdünnt und dreimal mit je 20ml Ethylacetat extrahiert. Die vereinigten Ethylacetatschichten wurden mit TmI 5%iger wäßriger HCI und 10ml gesättigten Natriumhydrogencarbonats extrahiert, über Natriumsulfat getrocknet und konzentriert, die Ausbeute betrug 112mg (100%)
Benzylesterin Form eines Öls, das nach c'erNMR-Analyse>97%ig rein war. Einer Lösung dieses Öls in 4ml THF wurde 1ml 10%iger wäßriger HCI-Lösung zugesetzt. Die Mischung wurde bei 250C 30 Minuten gerührt, mit 20ml gesättigter wäßriger Natriumhydrogencarbonatlösung verdünnt und viermal mit je 30ml Ethylacetat extrahiert. Die vereinigten Ethylacetatextrakte wurden über Natriumsulfat getrocknet und durch Konzentration in einen Feststoff überführt. Nach Rekristallisation des Feststoffes aus Chloroform wurden 92 mg (95%) Benzyl-3-benzamido-3-phenyl-2-hydroxypropionat in Form weißer Kristalle mit einem Schmelzpunkt von 129-131 "C erhalten.
Taxol. In ein kleines Reaktionsgefäß wurden 109mg (0,320mmol) (+)-Cis-1-benzoyl-3-(1-ethoxyethoxy)-4-phenylazetidin-2-on, 45mg (0,064mmol) 7-O-Triethylsilyl-Baccatin III, 7,8mg (0,064mmol) 4-Dimethylaminopyridin (DMAP) und 0,032ml Pyridin gegeben. Bei 25°C wurde das Gemisch 12 Stunden gerührt und mit 100ml Ethylacetat verdünnt. Die Ethylacetatlösung wurde
mit 20 ml 10%igerwäßriger Kupfersulfatlösung extrahiert, über Natriumsulfat getrocknet und konzentriert. Der Rückstand wurde durch Kieselgel filtriert, das mit Ethylacetat eluiert wurde. Nach Chromatografieren auf mit Ethylacetat/Hexan eluiertem Kieselgel und anschließender Rekristallisation aus Ethylacetat/Hexan wurden 61 mg (92%) 2'-(1-Ethoxyethoxy)-7-O-triethylsilyltaxol in Form eines Gemisches der Diastereomere im Verhältnis 2:1 erhalten.
Eine Probe von 5mg 2'-(1-Ethoxyethoxy)-7-0-triethylsilylta·- öl wurde in 2 ml Ethanol gelöst, und 0,5ml wäßriger 0,5%iger HCI-Lösung wurden zugegeben. Bei 00C wurde das Gemisch 30 Stunden gerührt und dann mit 50ml Ethylacetat verdünnt. Die Lösung wurde mit 20ml gesättigter, wäßriger Natriumhydrogencarbonatlösung extrahiert, über Natriumsulfat getrocknet und konzentriert. Der Rückstand wurde mittels Säulenchromatografie auf mit Ethylacetat/Hexan eluiertem Kieselgel gereinigt, und es wurden 4,5 mg (ca. 90%! Tpxol erhalten, das in jeder Hinsicht mit einer authentischen Probe übereinstimmte
Aus dem oben dargelegten i&t ersichtlich, daß die einzelnen Ziele der Erfindung erreicht worden sind.
Da bei den zuvor dargelegten Verbindungen und Verfahren verschiedene Veränderungen vorgenommen werden könnten, ohne den Bereich der Erfindung zu verlassen, sind sämtliche obige Beschreibungen und Darstellungen als illustrativ und nicht in einem eingeschränkten Sinne zu verstehen,
Claims (4)
1. ß-Lactam der Formel:

wobei R1 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist; R2 Wasserstoff, Ethoxyethyl, 2,2,2-Trichlorethoxymethyl oder eine andere Hydroxyl-Schutzgruppe ist; und R3 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist.
2,2,2-TrichlorethoxymethyI ist; und R3 Aryl oder substituiertes Aryl ist. 10. Verfahren gemäß Anspruch 8, wobei der Alkohol die Formel hat:
AcO
wobei R4 eine Hydroxyl-Schutzgruppe ist.
11. Verfahren gemäß Anspruch 10, wobei R4 Ethoxyethyl, Trimethylsüyl oder Triethylsilyl ist.
12. Verfahren gemäß Anspruch 10, wobei ein tertiäres Amin das Aktivierungsmittel ist.
13. Verfahren gemäß Anspruch 10, wobei das Aktivierungsmittel Triethylamin, Diisopropylethylamin, Pyridin, N-Methylimidazol oder4-Dimethylaminopyridin ist.
14. Verfahren zur Herstellung von Taxol, bei dem ein Alkohol mit einen ß-Lactam dar folgenden Formel in Kontakt gebracht wird:

R1 Aryl, substituiertes Aryl, ΑΙΙοΊ, Alkenyl oder Alkinyl ist;
R2 eine Hyroxyl-Schutzgruppe iat; und
Ra Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist;
der besagte Alkohol und das ß-tactam in Gegenwart einer ausreichenden Menge Aktivierungsmittel unter effektiven Bedingungen miteinander in Kontakt gebracht werden, das ß-Lactam mit dem Alkohol reagiert und ein ß-Amidoester ansteht, der als Zwischenverbindung bei der Taxolsynthese eingesetzt werden kann, und die besagte Zwischenverbindung in Taxol umgewandelt wird.
15. Verfahren gemäß Anspruch 14, wobei die Hydroxyl-Schutzgruppeaus Acetalen, Ethern, Estern und Carbonaten ausgewählt wird.
16. Verfahren gemäß Anspruch 14, wobei R2 Ethoxylethyl ist.
17. Verfahren zur Herstellung eines Taxols der Formel:
A und B unabhängig Wasserstoff oder niederes Alkanyloxy, Alkenyloxy, Alkinyloxy oder Aryloxy sind oder
A und B zusammen eine Oxogruppe bilden;
L und D unabhängig Wasserstoff oder Hydroxyl oder niederes Alkanyloxy, Alkenyloxy, Alkinyloxy oder Aryloxy sind;
E und F unabhängig Wasserstoff oder niederes Alkanyloxy ,Alkenyloxy, Alkinyloxy oder Aryloxy sind oder
E und F zusammen eine Oxogruppe bilden;
G Wasserstoff oder Hydroxyl oder niederes Alkanyloxy, Alkenyloxy, Alkinyloxy oder Aryloxy ist
G und M zusammen eine Oxogruppe oder Methylen darstellen oder G und M zusammen einen Oxocyclopropyl-Ring bilden oder M und F zusammen einen Oxocyclobutyl-Ring bilden;
J Wasserstoff, Hydroxyl oder niederes Alkanyloxy,
Alkenyloxy, Alkinyloxy oder Aryloxy ist oder
I Wasserstoff, Hydroxyl oder niederes Alkanyloxy, Alkenyloxy, Alkinyloxy oder Aryloxy ist; oder I und J zusammen eine Oxogruppe darstellen; und
K Wasserstoff, Hydroxyl oder niederes Alkoxy, Alkanyloxy, Alkenyloxy, Alkinyloxy oder Aryloxy ist; und
P und Q unabhängig Wasserstoff oder niederes Alkanyloxy, Alkenyloxy, Alkinyloxy oder Aryloxy sind oder
P und Q zusammen eine Oxogruppe darstellen; und
S und T unabhängig Wasserstoff oder niederes Alkanyloxy, Alkenyloxy, Alkinyloxy oder Aryloxy sind oder
S und T zusammen eine Oxogruppe darstellen; und
Uund V unabhängig Wasserstoff oder niederes Alkyl, Alkenyl, Alkinyl, Aryl oder substituiertes Aryl sind; und
W Aryl, substituiertes Aryl, niederes Alkyl, Alkenyl oder Alkinyl ist, bei dem
ein ß-Lactam folgender Formel
2. Verbindung gemäß Anspruch 1, wobei die Hydroxyl-Schutzgruppe aus der Gruppe bestehend aus Acetalen, Ethern, Estern und Carbonaten, ausgewählt wird.
3. Verbindung gemäß Anspruch 1, wobei R1 Phenyl, substituiertes Phenyl oder Aryl ist; R2 Ethoxyethyl oder 2,2,2-Trichlorethoxymethyl ist; und R3 Phenyl, substituiertes Phenyl oder Aryl ist.
4. Verbindung gemäß Anspruch 1, wobei R2 Ethoxyethyl oder 2,2,2-Trichlorethoxymethyl ist.
5. ß-Lactam c "ormel:
wobei R2 eine Hydroxyl-Schutzgruppe ist.
6. Verbindung gemäß Anspruch 5, wobei R2 Ethoxyethyl ist.
7. Verbindung gemäß Anspruch 5, wobei R2 aus einer Gruppe, bestehend aus Acetalen, Ethern, Estern und Carbonaten, ausgewählt wird.
a) Verfahren zur Herstellung von ß-Lactam der Formel
wobei
R1 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist; R2 Wasserstoff, Acetal, Ethoxyethyl, 2,2,2-Trichlorethoxymethyl oder eine andere Hydroxyl-Schutzgruppe ist; und
R3 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist;
• das Verfahren die Schritte der Reaktion eines a-Acyloxyacetylchlorids mit einem Imin in Gegenwart eines Amins zur Erzeugung einer ß-Lactam-Zwischenverbindung und der Umwandlung der Zwischenverbindung in das ß-Lactam umfaßt, b) Verfahren nach Ansprucn 7 a, wobei das a-Acyloxy-Acetylchlorid die Struktur:

und die Zwischenverbindung die Struktur:

OAC
hat.
c) Verfahren nach Anspruch 7a und 7b, wobei R, und R3 Phenyl sind.
d) Verfahren nach Anspruch 7 a, 7 b und 7 c, wobei R2 Ethoxyethyl oder 2,2,2-TrichlorethoxymethyI ist.
8. Verfahren zur Herstellung einer Taxol-Zwischenverbindung, bei dem ein Alkohol mit einem ß-Lactarn der folgenden Forme! in Kontakt gebracht wird:

wobei
R1 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist,
R2 eine Hydroxyl-Schutzgruppe ist, und
R3 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist,
der besagte Alkohol und das ß-Lactam in Gegenwart einer ausreichenden Menge Aktivierungsmittel unter effektiven Bedingungen miteinander in Kontakt gebracht werden, damit das ß-Lactam mit dem Alkohol reagiert und ein ß-Amidoester entsteht, der als Zwischenverbindung bei derTaxolsynthese eingesetzt werden kann. 9. Verfahren gemäß Anspruch 8, wobei Ri Aryl oder substituiertes Aryl ist; R2 Ethoxyethyl oder
R2 eine Hydroxyl-Schutzgruppe ist, und
R3 Aryl, substituiertes Aryl, Alkyl, Alkenyl oder Alkinyl ist,
der besagte Alkohol und das ß-Lactam in Gegenwart einer ausreichenden Menge Aktivierungsmittel unter effektiven Bedingungen miteinander in Kontakt gebracht werden, damit das ß-Lactam mit dem Alkohol reagiert und ein ß-Amidoester entsteht, der als Zwischenverbindung bei derTaxolsynthese eingesetzt werden kann. 9. Verfahren gemäß Anspruch 8, wobei Ri Aryl oder substituiertes Aryl ist; R2 Ethoxyethyl oder
