Description
본 발명은 신규 β-락탐, 그 제조방법 및 이러한 β-락탐의 사용을 포함하는 탁솔 제조방법에 관한 것이다.
탁솔이 속하는 테르펜의 탁산족은 생물학 및 화학분야에 중대한 관심을 끌어왔다. 탁솔은 하기 구조를 갖는, 넓은 스펙트럼의 항백혈병 및 종양 억제 활성을 가진 유망한 암화학요법제이다.
이 유망한 활성으로 인해, 탁솔은 프랑스 공화국 및 미합중국에서 임상시험을 행하고 있다.
이들 임상시험을 위한 탁솔의 공급은 현재 여러 종류의 주목으로부터의 나무껍질에 의해 제공된다. 그러나, 탁솔은 느리게 성장하는 이들 상록수의 나무껍질에서 소량으로만 발견되므로, 제한된 탁솔공급이 수요를 만족하지 못하는 중대한 우려를 야기한다. 마침내, 최근 화학자들은 탁솔 제조를 위한 실현 가능한 합성경로를 찾는 노력에 힘을 기울여왔다. 현재까지, 결과들은 완전히 만족스럽지 않았다.
제안된 한 합성경로는 시판 화학약품으로부터 테트라시클릭 탁산핵의 제조에 관한 것이다. 탁솔 협동작용물 탁수신의 합성이 홀톤(Holton)일행에 의해 문헌[JACS 110, 6558 (1988)]에 보고되어 있다. 이 연구에서 이루어진 진보에도 불구하고, 탁솔의 최종전체 합성은 다단계의 지루하고 경비가 드는 공정이다.
탁솔제조에 대한 또 다른 연구가 그린(Greene)일행에 의해 문헌[JACS 110,5917(1988)]에 기재되어 있으며, 탁솔의 협동작용물인 하기 구조를 갖는 10-데아세틸박카틴Ⅲ의 사용이 포함되어 있다.
10-데아세틸 박카틴Ⅲ 은 탁수스 박카타(Taxus baccata)의 잎으로부터 얻을 수 있기 때문에 탁솔보다 더욱 쉽게 입수할 수 있다. 그린 일행의 방법에 따라서, 10-데아세틸박카틴Ⅲ은 C10아세틸기를 부착하고 C-13알코올을 β-아미도카르복실산 단위에 의해 에스테르화 시킴으로서 C13 β-아미도에스테르 측쇄를 부착시키는 것에 의해 탁솔로 전환된다. 이 접근방식은 비교적 적은 단계를 필요로 하나, β-아미도카르복실산 단위의 합성은 낮은 수율로 진행되는 다단게 공정이며 커플링 반응은 지루하고 역시 낮은 수율로 진행된다. 그러나, 이 커플링 반응은 탁솔 또는 탁솔의 생물학적 활성 유도체의 모든 복잡한 합성법에 필요한 핵심단계이며, 그 이유는 와니(Wani) 일행에 의해 문헌 [JACS 93,2325(1971)]에 C13에서의 β-아미도에스테르 측쇄의 존재가 항종양 활성에 요구된다고 나와 있기 때문이다.
탁솔과 기타 잠재적 항종양제의 합성에 존재하는 주요 차이점은 C13산소에 쉽게 부착되어 β-아미도 에스테르 측쇄를 제공할 수 있는 용이하게 구입가능한 단위가 없는 점이다. 이러한 단위와 그것을 고수율로 부착시키는 방법을 개발한다면 탁솔 뿐 아니라 변성된 핵치환기조(set) 또는 변성된 C13 측쇄를 갖는 관련 항종양제의 합성이 용이해질 것이다. 이러한 요구는 신규의 쉽게 구입가능한 측새 전구체 화학단위 및 C13 산소에 그 부착을 위한 효율적 방법을 발견함으로써 충족되어 왔다.
따라서, 본 발명의 목적은 탁솔 합성을 위한 측쇄 전구체의 제공 및, 탁솔 중간체를 제공하기 위하여 비교적 고수율의 측쇄 전구체를 부착시키는 방법의 제공이다.
간략하게는, 본 발명은 하기 식의 측쇄 전구체, β-락탐 (1)에 관한 것이다.
상기식에서, R₁ 아릴, 치환된 아릴, 알킬, 알케닐 또는 알키닐이며, R₂는 수소, 에톡시에틸, 아세탈 또는 기타 히드록실보호기이고, R₃는 아릴, 알킬, 알케닐 또는 알키닐이다.
본 발명은 또한 β-락탐을 알코올과 반응시키기에 효과적인 조건하에 충분한 양의 활성화제의 존재하에 알코올을 β-락탐 (1)과 접촉시켜 탁솔 합성에서 중간체로서 사용될 수 있는 β-아미도에스테르를 형성하는 것으로 이루어지는 탁솔 중간체의 제조방법에 관한 것이다.
본 발명은 또한 β-락탐을 알코올과 반응시키기에 효과적인 조건하에 충분한 양의 활성화제 존재하에 알코올을 β-락탐(1)과 접촉시켜 β-아미도에스테르 탁솔중간체를 형성하는 것으로 이루어지는 탁솔의 제조방법에 관한 것이다. 중간체는 다음에 탁솔의 합성에 사용된다.
본 발명의 목적 및 특징은 이후에서 일부는 자명해지고 일부는 지적될 것이다.
본 발명은 β-락탐(1) 및 그 유도체에 관한 것이며, 그 구조는 아래에 나타내어진다.
앞에서 지적된 바와 같이, R₁은 아릴, 치환된 아릴, 알킬, 알케닐 또는 알키닐이며, R₂는 수소, 에톡시에틸, 아세탈 또는 기타 히드록실보호기이고, R₃은 아릴, 치환된 아릴, 알킬, 알케닐 또는 알키닐이다. 바람직하게는 R₁은 페닐, 치환된 페닐 또는 아릴이며, R₂는 에톡시에틸, 2,2,2-트리클로로에톡시메틸 또는 기타 아세탈히드록실보호기이고, R₃는 페닐, 치환된 페닐 또는 아릴이다. R₁과 R₃가 페닐인 바람직한 2가지 β-락탐의 구조를 아래에 나타낸다.
IUPAC 원칙에 따라서, β-락탐(2 및3)의 명칭은 1-벤조일-4-페닐-3-(1-에톡시에톡시)아제티딘-2-은(2) 및 1-벤조일-4-페닐-3-3-(2,2,2-트리클로로에톡시메톡시)아제티딘-2-은(3)이다. 가장 바람직한 β-락탐은 β-락탐(2)이다.
본 발명에 따라서, 하기 구조식을 갖는 탁솔 중간체, 천연탁솔 및 비천연적으로 발생하는 탁솔의 제조 방법이 제공된다.

탁솔알킬기는 단독으로 또는 상기 정의된 다양한 치환기와 함께 주사슬내에 1∼6탄소원자 및 10이하 탄소원자를 함유하는 저급알킬기가 바람직하다. 이들은 직쇄 또는 분지쇄이고 메틸, 에틸, 프로필, 이소프로필, 부틸, 이소부틸, t-부틸, 아릴, 헥실 등이 포함된다.
탁솔 알케닐기는 단독으로 또는 앞에서 정의된 다양한 치환기와 함께 주사슬내에 2∼6탄소원자 및 10이하의 탄소원자를 함유하는 저급알케닐이 바람직하다. 이들은 직쇄 또는 분지쇄이고 에테닐, 프로페닐, 이소프로페닐, 부테닐, 이소부테닐, 아릴, 헥세닐등이 포함된다.
탁솔 알키닐기는 단독으로 또는 앞에서 정의된 다양한 치환기와 함께 주사슬내에 2∼6탄소원자 및 10이하의 탄소원자를 함유하는 저급알케닐이 바람직하다. 이들은 직쇄 또는 분지쇄이고, 에테닐, 프로티닐, 부티닐, 이소부티닐, 아릴, 헥시닐등이 포함된다.
알카노일옥시의 예로는 아세테이트, 프로피오네이트, 부티레이트, 발레레이트, 이소부티레이트 등이 포함된다. 더욱 바람직한 알카노일옥시는 아세테이트이다.
탁솔 아릴성분은 단독으로 또는 다양한 치환기와 함께 6~10탄소원자를 함유하며 페닐, 알파-나프틸 또는 β-나프틸 등이 포함된다. 치환기로는 알카녹시, 히드록시, 할로겐, 알킬, 아릴, 알케닐, 아실, 아실옥시, 니트로, 아미노, 아미도 등이 포함된다.
페닐이 더욱 바람직한 아릴이다.
치환기 A,B,D,L,E,F,G,M,I,J,K,P,Q,S,T,U,V 및 W의 바람직한 것들을 아래 표 1에 열거한다.

본 발명의 방법에 따라서, 알코올과 활성화제, 바람직하게는 트리에틸아민, 디이소프로필에틸아민과 같은 3차 아민, 피리딘, N-메틸이미다졸 및 4-디메틸아미노피리딘(DMAP)존재하에 β-락탐(1)을 β-아미도에스테르로 전환시킨다. 예를 들어, β-락탐(1)을 4-디메틸아미노피리딘(DMAP)돈재하에, 탁산테트라시클릭핵과 C13 히드록실기를 갖는 화합물과 반응시켜 C13에서 β-아미도에스테르기를 갖는 물질을 얻는다.
가장 바람직하게는, 알코올은 문헌 [Greene et al., JACS 110, 5917(1988)]에 기재된 대로 또는 다른 경로에 의해 수득될 수 있는 7-O-트리에틸실릴박카틴 Ⅲ이다. 그린일행의 문헌에서 보고된 바와 같이 10-데아세틸박카틴 Ⅲ은 하기 반응도식에 따라서 7-O-트리에틸실릴박카틴 Ⅲ으로 전환된다.
주의깊게 최적화한 조건하에, 50ml의 피리딘/mmol 의 10-데아세틸박카틴Ⅲ 존재하에 10-데아세틸박카틴Ⅲ 을 아르곤 분위기하에 23℃에서 20시간동안 20당량의 (C₂H5)₃SiCl과 반응시켜, 반응생성물로서 7-트리에틸실릴-10-데아세틸박카틴Ⅲ(32a) 을 정제후 84~86% 수율로서 수득한다. 다음에 반응생성물을 0℃ 아르곤 분위기하에 48시간동안 5당량의 CH3COCl과 25ml의 피리딘/mmol의 32a로서 아세틸화시켜 86 % 수율의 7--트리에틸실릴박카틴 을 수득한다. 하기 반응도식에 나타내어진 바와 같이, 7-트리에틸실릴박카틴Ⅲ(32b) 을 실온에서 본 발명의 β-락탐과 반응시켜 C-7및 C-2′히드록실기가 각각 트리에틸실릴 및 에톡시에틸보호기로써 보호된 탁솔 중간체를 제공할 수 있다. 이들 기는 다음에 에스테르결합 또는 탁솔 치환기들을 방해하지 않는 순한 조건하에 가수분해된다.
본 도식은 천연 탁솔 생성물의 합성에 관한 것이지만, 본 발명내에 포함되는 다른 합성 탁솔을 제조하기 위하여, 천연 또는 비천연 원천으로부터 유도될 수 있는 β-락탐 또는 테트라시클릭 알코올에도 변형하여 쓸 수 있다.
또 다르게는, 7-0-트리에틸실릴박카린Ⅲ외의 알코올과 활성화제의 존재하에 β-락탐(1)을 β-아미도 에스테르로 전환시켜 탁솔 중간체를 형성할 수 있다. 다음에 적절한 반응도식하에 탁솔 중간체를 사용하여 탁솔의 합성을 진행할수 있다.
β-락탐 알킬기는, 단독으로 또는 앞에서 정의된 다양한 치환기와 함께, 주사슬 내 1∼6 탄소원자 및 15 이하의 탄소원자를 함유하는 저급알킬이 바람직하다. 이들은 직쇄 또는 분지쇄일 수 있고, 메틸, 에틸,프로필, 이소프로필, 부틸, 이소부틸, t-부틸, 아릴, 헥실 등이 포함된다.
β-락탐 알케닐기는, 단독으로 또는 앞에서 정의된 다양한 치환기와 함께, 주사슬내 2~6 탄소원자 및 15 이하의 탄소원자를 함유하는 저급알케닐이 바람직하다.
이들은 직쇄 또는 분지쇄일 수 있고 에테닐, 프로페닐, 이소프로페닐, 부테닐, 이소부테닐, 아리, 헥세닐 등이 포함된다.
β-락탐 알키닐기는, 단독으로 또는 앞에서 정의된 다양한 치환기와 함께 주사슬내2~6 탄소원자 및 15 이하의 탄소원자를 함유하는 저급알키닐이 바람직하다. 이들은 직쇄 또는 분지쇄일 수 있고, 에테닐, 프로페닐, 부티닐, 이소부티닐, 아릴, 헥시닐 등이 포함된다.
β-락탐 알카노일오시의 예로는 아세테이트, 프로피오네이트, 부티레이트, 발레레이트, 이소부티레이트 등이 포함된다. 더욱 바람직한 알카노일옥시는 아세테이트이다.
기재된 β-락탐 아릴 성분은, 단독으로 또는 다양한 차환기와함께 6 -15 탄소원자를 함유하여 페닐, 나프틸, 또는 나프틸 등이 포함된다. 치환기로는 알카녹시, 히드록시, 할로겐, 알킬, 아릴,알케닐, 아실, 아실옥시, 니트로, 아미노, 아미도 등이 포함된다. 페닐이 더욱 바람직한 아릴이다.
앞에서 지적된 바와 같이 β-락탐 (1)의 R2는 알킬, 아실, 에톡시에틸, 트리틀로로에톡시메틸 또는 기타 히드록실보호기로서 아세탈 및 에테르, 즉 메톡시메틸, 벤질옥시메틸; 에스테르, 예컨대 아세테이트;카보네이트, 예컨대 메틸카보네이트등이다. 히드록실기에 대한 각종 보호기 및 그 합성은 문헌 (T.W.Greene, John Wiley and Sons, Protective Groups in Organic Synthesis, 1981)에서 찾을 수 있다.
선택된 히드록실보호기는 탁솔 중간체의 에스테르 결합 또는 기타 치환기들을 방해하지 않는 충분히 순한 조건하에 용이하게 제거되어야 한다. 그러나, R₂는 바람직하게는 에톡시에틸 또는 2,2,2-트리클로로에톡시메틸이고 가장 바람직하게는 에톡시에틸이다.

화합물의 예를 일반식으로 아래에 나타낸다.
β-락탐(1)은 여러 개의 비대칭 탄소를 갖기 때문에 비대칭 탄소원자를 갖는 본 발명의 화합물은 디아스테레오머, 라세미체 또는 광학활성 형태로 존재할수 있음이 이 분야 숙련인들에게 공지되어 있다. 이들 모든 형태는 본 발명이 영역내에 들어간다. 더욱 상세하게는, 본 발명은 에난티오머, 디아스테레오머, 라세미혼합물 및 그의 이들의 혼합물을 포함한다.
β-락탐(1)은 아래 도식에서β -락탐(2)에 대해 도시된 바와 같이 용이하게 입수 가능한 물질로부터 제조할 수 있다.
시약 (a) 트리에틸아민, CH2Cl2, 25℃, 18시간: (b) 4 당량 제2세륨 암모늄니트레이트, CH3CN, -10℃, 10분; (c) KOH, THF, H2O, 0℃, 30분; (d) 에틸비닐에테르, THF, 톨루엔 술폰산 (촉매), 0℃, 1.5시간; (e) CH₃Li에테르, 10분; 벤조일클로라이드, -78℃, 1 시간
출발물질들은 쉽게 구입할 수 있다. α-아실옥시아세틸클로라이드는 글리콜산으로부터 3차 아민 존재하에서 제조되며, 알데히드 및 p-메톡시아닐린으로부터 제조된 아민과 고리축합하여 1-p-메톡시페닐-3-아실옥시-4-아릴아제티딘-2-온을 얻는다.
p-메톡시페닐기는 제2세륨암모뉴 니트레이트에 의한 산화로써 쉽게 제거될 수 있고, 아실옥시기는 이 분야 숙련인에게 친숙한 표준 조건하에 가수분해되어 3-히드록시-4-아릴 아제티딘-2-온을 제공할 수 있다.
3-히드록실기는 1-에톡시에틸과 같은 각종 표준 보호기로써 보호될 수 있다. 바람직하게는, 라세미화 3-히드록시-4-아릴아제티딘-2-온을 순수한 에난티오머로 용해시킨 후, 상응하는 2-메톡시-2-(트리플루오로메틸)페닐아세트산 에스테르의 재결정화에 의해 보호하고 우선성(dextrorotatory) 에난티오머만을 탁솔의 제조에 사용한다. 어떤 경우에도, 3-(1-에톡시에톡시)-4-페닐아제티딘-2-온은 -78℃ 이하에서 염기, 바람직하게는 n-부틸리튬과 아로일클로라이드로 처리함으로써 β-락탐(2)로 전환될 수 있다.
하기 실시예로써 본 발명을 예시한다.
실시예 1
시스-1-벤조일-3-(1-에톡시에톡시)-4-페닐아제티디논(2)의 제조
시스-1-p-메톡시페닐-3-아세톡시-4-페닐아제티딘-2-온;
-20℃에서 CH2Cl2 15 mL에 용해시킨, 벤즈알레히드와 p-메톡시아닐린으로부터 유도된 이민 962mg (4.56mmol)과 트리에틸아민 0.85mL(6.07mmol)의 용액에, CH2Cl215mL에 알파-아세톡시아세틸클로라이드 413mg(3.04mmol)을 용해시킨 용액을 적가한다. 반응 혼합물을 18시간에 걸쳐 25℃ 까지 가온한다. 반응 혼합물을 다음에 100mL의 CH ₂Cl₂로 희석하고 용액을 30mL의 10% HCL수용액으로 추출한다. 유기층을 30㎖30mL의 포화 중탄산나트륨 수용액으로 씻고, 황산나트륨으로 건조시키고 농축하여 고형물질을 얻는다. 고체를 50mL의 헥산으로 분쇄하고 혼합물을 여과한다. 남은 고체를 에틸 아세테이트/헥산으로 재결정하여 645mg(68%)의 시스-1-p-메톡시페닐-3-아세톡시-4-페닐아제티딘-3-온을 백색결정으로서 수득한다. 융점 163℃ 시스-3-아세톡시-4-페닐아제티딘-2-온:
-10℃에서 아세토니트릴 700mL에 20.2g의 시스-1-p-메톡시페닐-3-아세톡시-4-페닐아지티딘-2-온을 용해시킨 용액에 물 450mL에 제2세륨암모늄 니트레이트를 용해시킨 용액을 1시간에 걸쳐 서서히 가한다. 혼합물을 30분간 -10℃에서 교반하고 500mL의 에테르로 희석한다. 수성층을 2 ×100mL 분량의 에테르로 추출하고, 합친 유기층을 2×100mL에 분량의 물, 2 × 100mL분량의 포화 중아황산나트륨 수용액, 2 ×100mL분량의 포화 중탄산나트륨 수용액으로 씻고 농축하여 18.5g의 고체를 수득한다. 아세톤/헥산으로 고체를 재결정하면 12.3g(92%)의 시스-3-아세톡시-4-페닐아제티딘-2-온을 백색결정으로서 수득한다. 수득한다. 융점 152∼154℃.
시스-3-아세톡시-4-페닐아제티딘-2-온:
0℃에서 THF 200mL의 1M수산화칼륨 수용액 280mL의 혼합물에 THF265mL에 4.59g(22,4mmol)의 시스-3-아세톡시-4-페닐아제티딘-2-온을 용해시킨 용액을 적가 깔때기를 통해 40분간에 걸쳐 가한다. 용액을 0℃에서 1시간동안 교반하고 100mL의 물과 100mL의 포화 중탄산나트륨을 가한다. 혼합물을 4 × 200mL분량의 에틸 아세테이트로 추출하고 합친 유기층을 황산나트륨으로 건조시키고 농축하여 3.54g(97%)의 라세미화 시스-3-히드록시-4-페닐아제티딘-2-온을 백색 결정으로서 수득한다. 융점 147∼149℃. 이 물질을 헥산/아세톤에 의한 그 2-메톡시-2-(트리플루오로메틸)페닐아세트산 에스테르의 재결정화 후 가수분해로써 그 에난티오머로 분해시킨다. [α]25Hg 177°.
시스-3-(1-에톡시에톡시)-4-페닐아제티딘-2-온:
0℃에서 THF 15mL에 시스-3-히드록시-4-페닐아제티딘-2-온 3.41g (20.9mmol)을 용해시킨 용액에 5mL의 에틸비닐 에테르와 20 mg(0.2 mmol)의 메탄술폰산을 가한다. 혼합물을 0℃에서 20분간 교반하고, 20mL의 포화 중탄산나트륨 수용액으로 희석하고, 3 × 40mL 분량의 에틸 아세테이트로 추출한다. 합친 에틸 아세테이트층을 황산나트륨으로 건조시키고 농축하여 4.87g(99%)의 시스-3-(1-에톡시에톡시)-4-페닐아제티딘-2-온을 무색 오일로서 수득한다.
시스-1-벤조일-3-(1-에톡시에톡시)-4-페닐아제티딘-2-온:
-78℃에서 THF 40mL에 시스-3-(1-에톡시에톡시)-4-체닐아제티딘-2-온 2.35g(10mmol)을 용해시킨 용액에, 6.1mL(10.07mmol)의 1.65M n-부틸리튬의 헥산 용액을 가한다. 혼합물을 -78℃에서 10분간 교반하고 THF 10mL에 벤조일클로라이드 1.42g(10.1mmol)을 용해시킨 용액을 가한다. 혼합물을 -78℃에서 1시간 동안 교반하고 포화 중탄산나트륨 수용액 70mL로써 희석하고 3 × 50mL분량의 에틸 아세테이트로 추출한다. 합친 에틸 아세테이트 추출액을 황산나트륨으로 건조시키고 농축하여 3.45g의 오일을 수득한다.. 이 오일을 에틸 아세테이트/헥산으로 용출시켜 실리카겔에 크로마토그래피하여 3.22g(95%)의 시스-1-벤조일-3-(1-에톡시에톡시)-4-페닐아제티딘-2-온(2)을 무색오일로서 수득한다.
실시예 2
시스-1-벤조일-3-(1-에톡시에톡시)-4-페닐아제티딘-2-온 (2)으로부터 β-아미도에스테르의 제조
벤질-3-벤즈아미도-3-페닐-2-히드록시프로피오네이트:
THF 0.3mL에 시스-1-벤조일-3-(1-에톡시에톡시)-4-페닐아제티딘-2-온 88mg(0.26mmol)을 용해시킨 용액에 벤질 알코올 28mg(0.26mmol)과 4-디메틸아미노피리딘(DMAP) 32 mg(0.26mmol)을 가한다. 25℃에서 5시간후, 혼합물을 10 mL의 포화 중탄산나트륨 수용액으로 희석하고 3×20 mL 분량의 에틸 아세테이트로 추출한다.
합친 에틸아세테이트층을 10 mL의 5% HCI 수용액과 10㎖의 포화 중탄산나트륨으로 추출하고, 황산나트륨으로 건조시키고 농축하여 112 mg(100%)의 벤질 에스테르를 오일로서 수득하며 이것은 NMR분석에 의하면 순도 97%이상이다.
이 오일을 THF 4 mL에 용해시킨 용액에 1 mL의 10% HCI수용액을 가한다. 혼합물을 25℃에서 30분간 교반하고 20 mL의 포화 중탄산나트륨 수용액으로 희석하고, 4×30 mL 분량의 에틸 아세테이트로 추출한다. 합친 에틸 아세테이트추출액을 황산나트륨으로 건조시키고 농축하여 고체를 얻는다. 이 고체를 클로로포름으로 재결정하여 92 mg(95%)의 벤질-3-벤즈 아미드-3-페닐-2-히드록시프로피오네이트를 백색 결정으로서 수득한다. 융점 129-131℃.
탁솔:
작은 반응 용기에 109 mg(0.320mmol)의 (+)-시스-1-벤조일-3-(1-에톡시에톡시-4-페닐-아제티딘-2-온, 45 mg(0.064mmol)의 7-0-트리에틸실릴 박카틴Ⅲ, 7.8mg(0.063mmol)의 4-디케틸아미노피리딘(DAMP) 및 0.032 mL의 피리딘을 가한다. 혼합물을 25℃에서 12시간동안 교반하고 100 mL의 에틸 아세테이트로 희석한다. 에틸 아세테이트 용액을 20 mL의 10%황산구리 수용액으로 추출하고, 황산나트륨으로 건조시키고 농축한다. 잔사를 에틸 아세테이트로 용출시켜 실리카겔플러그(plug)를 통해 여과한다. 에틸 아세테이트/헥산으로 용출시켜 실리카겔에 플래시 크로마토그래피한 후에 에틸 아세테이트/헥산으로 재결정하여 61 mg(92%)의 2'-(1-에톡시에톡시)-7-0-트리에틸실릴 탁솔을 디아스테레오머의 2:1 혼합물로서 수득한다.
2'-(1-에톡시에톡시)-7-0-트리에틸실릴 탁솔 5 mg 시료를 2 mL의 에탄올에 용해시키고 0.5 mL의 0.5 % HCI수용액을 가한다. 혼합물을 0℃에서 30시간동안 교반하고 에틸 아세테이트 50 mL로 희석한다. 용액을 20 mL의 포화 중탄산나트륨 수용액으로 추출하고, 황산나트륨으로 건조시키고 농축한다. 잔사를 에틸 아세테이트/헥산으로 용출하여 실리카겔에 컬럼 크로마토그래피하여 4.5 mg(약 90%) 의 탁솔을 얻으며, 이것은 공인샘플과 모든 면에서 동일하다.
이상에서 볼 때, 본 발명의 여러 목적들은 성취되었음을 알 수 있다.
상기 조성물 및 방법은 본 발명의 영역을 벗어나지 않고서 다양한 변화를 기할 수 있으므로, 이상의 기술 내용에 포함된 모든 것은 예시적일 뿐 제한적 의미가 아닌 것으로 이해되어야 한다.
Claims (15)
- 제1항에 있어서, R₁은 아릴 또는 치환된 아릴이며, R₂는 에톡시에틸 또는 2,2,2-트리클로로에톡시메틸리고, R₃은 아릴 또는 치환된 아릴인 방법.
- 제3항에 있어서, R₄는 에톡시에틸, 트리메틸실릴 또는 트리에틸실릴인 방법.
- 제3항에 있어서, 활성화제가 3차 아민인 방법.
- 제3항에 있어서, 활설화제가 트리에틸아민, 디이소프로필에틸아민, 피리딘, N-메틸이미다졸 또는 4-디메틸아미노피리딘인 방법.
- 제 7항에 있어서, 히드록실 보호기가 아세탈, 에테르, 에스테르 및 카보네이트 중에서 선택되는 방법.
- 제7항에 있어서, R₂가 에톡시에틸인 방법.
- 하기 식의 β-락탐을(상기 식에서, R₁은 페닐, 치환된 페닐, 아릴, 알킬, 알케닐 또는 알키닐이며, R₂는 히드록실 보호기이고, R3은 페닐, 치환된 페닐, 아릴, 알킬, 알케닐 또는 알키닐이다.)알코올과 접촉시킴에 있어서, β-락탐을 알코올과 반응시키기에 효과적인 조건하에 충분한 양의 활성화제 존재하에 상기 β-락탐과 상기 알코올의 접촉을 수행하여 탁솔 합성에 중간로서 사용하기 적합한 β- 아미도 에스테르를 형성하고, 상기 중간체를 탁솔로 전환시키는 것으로 이루어지는 하기식의 탁솔의 제조방법.(상기 식에서 A와 B는 독립적으로 수소 또는 저급알카노일옥시, 알케노일옥시, 알키노일옥시 또는 아릴로일옥시이거나 A와 B는 함께 옥소를 형성하며; L과 D는 독립적으로 수소 또는 히드록시 또는 저급 알카노일옥시, 알케노일옥시, 알키노일옷 또는 아릴로일옥시이며; E와 F는 독립적으로 수소 또는 저급알카노일옥시, 알케노일옥시, 알키노일옥시 또는 아릴로일옥시이거나 E와 F는 함께 옥소를 형성하고; G는 수소 또는 히드록시 또는 저급알카노일옥시, 알케노일옥시, 알키노일옥시 또는 아릴로일옥시이며; G와 M이 함께 옥소 또는 메틸렌을 형성하거나 G와 M이 함께 옥소리클로프로필고리를 형성하거나 M과 F가 함께 옥소시클로부틸고리를 형성하며; J는 수소 또는 히드록시 또는 저급 알카노일옥시, 알케노일옥시, 알키노일옥시 또는 아릴로일옥시이며; I는 수소 또는 히드록시 또는 저급 알카노일옥시, 알케노일옥시, 알키노일옥시 또는 아릴로일옥시이며; I와 J는 함께 옥소를 형성하고; K는 수소, 히드록시 또는 저급알콕시, 알카노일옥시, 알케노일옥시, 알키노일옥시 또는 아릴로일옥시이고; P와 Q는 독립적으로 수소 또는 저급알카노일옥시, 알케노일옥시, 알키노일옥시 또는 아릴로일옥시이거나 P와 Q는 함께 옥소를 형성하고; S와 T는 독립적으로 수소 또는 저급알카노일옥시, 알케노일옥시, 알키노일옥시 또는 아릴로일옥시이거나 S와 T가 함께 옥소를 형성하고; U와 V는 독립적으로 수소 또는 저급알킬, 알케닐, 알키닐, 아릴 또는 치환된 아릴이고; W는 아릴, 치환된 아릴, 저급알킬, 아케닐 또는 알키닐이다 )
