Abstract
Η παρούσα εφεύρεση σχετίζεται με μια μέθοδο για την παραγωγή μιας τοξίνης αλλαντίασης και περισσότερο συγκεκριμένα με μια μέθοδο για την παρασκευή τοξίνης αλλαντίασης, όπου η μέθοδος περιλαμβάνει τα στάδια: (α) της επεξεργασίας μιας καλλιέργειας ενός στελέχους παραγωγής τοξίνης αλλαντίασης με οξύ για την κατακρήμνιση μιας τοξίνης αλλαντίασης· (β) της προσθήκης ρυθμιστικού διαλύματος στην κατακρημνισμένη τοξίνη αλλαντίασης και της ακόλουθης επεξεργασίας με έναν αναστολέα πρωτεάσης και νουκλεάση, εκχυλίζοντας έτσι την τοξίνη αλλαντίασης· (γ) της επεξεργασίας της εκχυλισμένης τοξίνης αλλαντίασης με οξύ για την κατακρήμνιση της τοξίνης αλλαντίασης και της διάλυσης του κατακρημνίσματος σε ρυθμιστικό διάλυμα· και (δ) του καθαρισμού της τοξίνης αλλαντίασης με ανιονανταλλακτική χρωματογραφία. Η χρήση της μεθόδου της τοξίνης της εφεύρεσης καθιστά δυνατή την παραγωγή μιας υψηλής καθαρότητας τοξίνης αλλαντίασης με μια απλή διαδικασία, πράγμα το οποίο υποδηλώνει ότι η μέθοδος είναι ιδιαιτέρως οικονομική και αποτελεσματική. Η τοξίνη αλλαντίασης που παράγεται με τη μέθοδο της εφεύρεσης έχει υψηλή καθαρότητα σε σύγκριση με τις τοξίνες αλλαντίασης που παράγονται με συμβατικές μεθόδους, και έτσι έχει αυξημένη δυνατότητα να δρα σε μια τοπική περιοχή. Έτσι, η κυκλοφορία στο σύστημα της τοξίνης αλλαντίασης, που μπορεί να έχει ως αποτέλεσμα παρενέργειες, είναι μειωμένη για να αυξηθεί η ασφάλεια. Αντίστοιχα, η τοξίνη αλλαντίασης της εφεύρεσης μπορεί να χρησιμοποιηθεί για διάφορους σκοπούς, συμπεριλαμβανομένων της αγωγής νευρομυϊκών διαταραχών, αφαίρεσης των ρυτίδων και της αγωγής της σπαστικής ημιπληγίας και της εγκεφαλικής παράλυσης.
