Abstract
本发明涉及编码植物中参与淀粉合成的酶的核酸分子。所述酶是新的淀粉合成酶同工型。本发明还公开了用于产生合成改性淀粉的转基因植物细胞和植物的载体。此外,还公开了产生所述转基因植物细胞和植物的方法,和制备改性淀粉的方法。
Description
本发明涉及植物中编码参与淀粉合成的酶的核酸分子。该酶是新的淀粉合成酶同工型。
本发明还涉及载体和用所述核酸分子或载体转化的宿主细胞,尤其是植物细胞和能够从这些细胞再生的植物。
而且,本发明描述了用于产生如下转基因植物细胞和植物的方法,该转基因植物细胞和植物由于编码淀粉合成酶的DNA分子的引入而合成性质发生改变的淀粉。本发明还涉及能够从根据本发明的这些植物细胞和植物中获得的淀粉,和产生该淀粉的方法。
由于注意到近来植物成分作为可再生原材料其重要性正日益增加,生物技术研究的任务之一是努力使这些植物原材料适合于加工工业的要求。因此,为了使可再生原材料可以应用到尽可能多的应用领域中,必须获得多种多样的材料。
油类、脂肪和蛋白质,以及多糖,构成了来自植物的重要可再生原材料。除了纤维素外,高等植物中最重要的贮藏物质之一,淀粉,起着重要的作用。在这方面,玉米是最令人感兴趣的植物之一,因为它是全球最重要的淀粉生产作物。
多糖淀粉是化学上一致的基本单元葡萄糖分子的聚合物。然而,它是由聚合程度和葡萄糖链中分支发生程度不相同的不同形式分子组成的高度复杂混合物。因此淀粉不是均一的原材料。具体地,我们区分了直链淀粉和支链淀粉,直链淀粉是由α-1,4-糖苷键连接的葡萄糖分子形成的基本上不分支的聚合物,而支链淀粉是具有不同分支的葡萄糖链的复杂混合物。这些分支是通过额外α-1,6-糖苷键的出现产生的。在用于淀粉生产的典型植物例如玉米或马铃薯中,所合成的淀粉由约20%-25%的直链淀粉和约75%-80%的支链淀粉组成。
为了使淀粉尽可能广泛地得到应用,提供能够合成尤其适合于多种目的的改性淀粉的植物显然是理想的。除了植物育种方法外,提供此类植物的一种可能方式是,通过重组方法定向遗传改变产淀粉植物的淀粉代谢。然而,这样做的前提条件是鉴定和表征参与淀粉合成和/或淀粉修饰的酶,并分离编码这些酶的相关DNA分子。
导致淀粉合成的生物化学合成途径基本上是已知的。在植物细胞中,淀粉合成发生在质体中。在光合作用活跃的组织中,它们是叶绿体,在光合作用不活跃的淀粉贮藏组织中是造粉体。
最重要的参与淀粉合成的酶是淀粉合成酶和分支酶。在淀粉合成酶中,已经有多种同工型的描述,所有这些同工型均通过将ADP-葡萄糖的葡糖残基转移至α-1,4-葡聚糖上催化聚合反应。分支酶催化在线性α-1,4-葡聚糖中引入α-1,6-分支。
可以区分出两类淀粉合成酶:颗粒结合性淀粉合成酶(GBSS)和可溶性淀粉合成酶(SS)。然而,这种区别并不是在每种情况下都是泾渭分明的,因为一些淀粉合成酶既以颗粒结合形式存在又以可溶性形式存在(Denyer等,Plant J.4(1993),191-198;Mu等,Plant J.6(1994),151-159)。
除了颗粒结合性淀粉合成酶GBSSI类型外,基于cDNA和氨基酸序列比较,至今在玉米植物中描述了属于可溶性淀粉合成酶类型的至少3种不同的同工型。
淀粉合成酶同工型I(SSI)包括在玉米中编码约76kDa蛋白质zSSI(Mu等,Plant J 6,(1994),151-159)和至今仅在单子叶植物例如水稻(Baba等,Plant Physiol.103,(1993),565-573)中描述过的基因,该同工型主要在胚乳中表达。通常,这些蛋白质可以受到柠檬酸盐的刺激,并且不依赖所谓的引发分子(primer molecules)。
相反,淀粉合成酶同工型II(=SSII)通常依赖于引发分子,并表现出与豌豆(Dry等,Plant J.2,(1992),193-202)和马铃薯(Edwards等,Plant J.8,(1995),283-294)的SSII同工型(其中一些过去被命名为GBSSII)具有最大的序列同源性。
当考虑玉米的SSII时,必须将文献中称作zSSIIa和zSSIIb的基因或cDNA(Harn等,Plant Mol.Biol.37,(1998),639-649;Imparl-Radosevich,Arch.Biochem.Biophys,362,(1999),131-138),与基于较早生化研究命名的(Boyer和Preiss,Plant Physiol.67,(1981),1141-1145;Mu等,Plant J.6,(1994),151-159)、来自玉米胚乳约180kDa的所谓SSII蛋白质(分子量是通过凝胶过滤确定的(Mu等,Plant J.6,(1994),151-159))区分开来。实际上哪个基因相应于这个180 kDa蛋白质的问题目前仍没有结论性的答案(Imparl-Radosevich,Arch.Biochem.Biophys.362,(1999),131-138)。Cao等(Plant Physiol.120,(1999),205-215)认为所谓的du1基因是相应于该180kDa蛋白质的基因。
目前已描述过的称作SSIII的第三类淀粉合成酶基因,在马铃薯中编码一个139kDa的蛋白质(Abel等,Plant J.10,(1996),981-991;Marshall等,植物细胞(Plant Cell)8,(1996),1121-1135),在马铃薯的块茎中占总淀粉合成酶活性的80%。由于与马铃薯SSIII氨基酸序列的比较中该C端某些序列区域具有高度保守性,因此原先称作du1基因的该玉米基因被提议重新命名为“zSSIII”(Cao等,Plant Physiol.120,(1999),205-215),前缀“z”代表其生物来源是玉蜀黍(Zea mays)。
至今仅确定了同工型GBSSI在淀粉合成中的详细功能。该酶活性极大地或完全地降低的植物合成不被淀粉酶消化的“蜡样”淀粉(Shure等,细胞(Cell)35(1983),225-233;Visser等,Mol.Gen.Genet.225(1991),289-296;WO9211376A1),因此该酶在直链淀粉合成中起着重要作用。在雷氏衣藻(Chlamydomonas reinhardtii)的细胞中同样观察到该现象(Delrue等,J.Bacteriol.174(1992),3612-3620)。此外在衣藻属(Chlamydomonas)中,还可以证明GBSSI不仅参与直链淀粉合成,还影响支链淀粉合成。不具有GBSSI活性的突变体缺少通常合成的含有较长葡聚糖链的支链淀粉特定部分。
可溶性淀粉合成酶同工型的功能仍不清楚,推测这些可溶性淀粉合成酶与分支酶一起参与支链淀粉的合成(见例如Ponstein等,Plant Physiol.92(1990),234-241),并且在淀粉合成速率的调节中起着重要作用。
除了玉米外,还在一系列其它植物物种中鉴定到可溶性淀粉合成酶。例如,已经从豌豆(Denyer和Smith,Planta 186(1992),609-617)和马铃薯(Edwards等,Plant J.8(1995),283-294)中分离到均质的可溶性淀粉合成酶。发现在这些情况中鉴定为SSII的该可溶性淀粉合成酶同工型与颗粒结合性淀粉合成酶GBSSII是相同的(Denyer等,Plant J.4(1993),191-198;Edwards等,Plant J.8(1995),283-294)。借助层析方法已经描述了一些其它的植物种类,例如大麦(Tyynela和Schulman,Physiologia Plantarum 89(1993),835-841;Kreis,Planta 148(1980),412-416)和小麦(Rijven,Plant Physiol.81(1986),448-453)中,存在多种SS同工型。编码这些蛋白质的DNA序列也已有描述(见例如GenBankAcc.No.U48227;Vrinten等,Mol.Gen.Genet.261(3),(1999),463-471)。
为了提供关于改变任何淀粉贮藏植物以便使它们合成改性淀粉的更多选择,必须鉴定在各种情况中编码这些淀粉合成酶的其它同工型的DNA序列。
因此,本发明的目标是提供编码参与淀粉生物合成的酶的核酸分子,借助于这些核酸分子可以制备这些酶的活性表现出增加或降低的重组植物,由此导致在这些植物中合成具有改变的化学和/或物理性质的淀粉,从而更好地适合于一般和/或特殊目的。
通过提供本专利申请中描述的实施方案可实现该目标。
因此,本发明涉及编码具有淀粉合成酶生物活性的蛋白质或该蛋白质的生物活性片段的核酸分子,这些分子优选编码具有Seq ID No.2所示氨基酸序列的蛋白质。
具体地,本发明涉及含有Seq ID No.1所示核苷酸序列或其部分的核酸分子,优选含有Seq ID No.1中所示编码区或相应的核糖核苷酸序列的分子。
本发明还涉及具有与Seq ID No.1中所示序列的全部或部分互补的序列的核酸分子。
本发明主题还有编码淀粉合成酶或其生物活性片段、并/或由于遗传密码的简并性其序列不同于上述分子的核苷酸序列的核酸分子。
本发明还涉及编码淀粉合成酶或其生物活性片段、并且与上述分子之一杂交的核酸分子。
根据本发明的核酸分子既可以是DNA又可以是RNA分子。适合的DNA分子是例如基因组或cDNA分子。RNA分子可以是例如mRNA或反义RNA分子。
因此,本发明还涉及相应于SEQ ID NO.1所示cDNA序列的基因组序列的内含子部分的核苷酸序列。适合的内含子序列可以例如采用SEQ IDNO.1所示核酸分子,通过例如筛选基因组DNA文库来分离和鉴定。
为了本发明的目的,术语“杂交”是指在常规杂交条件下,优选在例如Sambrook等(分子克隆,实验室手册(Molecular Cloning,ALaboratory Manual),第2版(1989),Cold Spring Harbor LaboratoryPress,Cold Spring Harbor,NY)所描述的严紧条件下进行的杂交。“杂交”尤其优选指在以下条件下发生的杂交:杂交缓冲液:2×SSC;10×Denhardt溶液(Ficoll 400+PEG+BSA;比例为
1∶1∶1);0.1% SDS;5mM EDTA;50mM Na2HPO4;

原则上,与根据本发明的核酸分子杂交的核酸分子可以来源于任何具有此类分子的生物(即原核生物或真核生物,尤其是细菌、真菌、藻类、植物或动物生物)。它们优选来源于单子叶或双子叶植物,尤其是有用的植物,特别优选来源于淀粉贮藏植物,尤其是玉米。
与根据本发明的分子杂交的核酸分子可以从例如各种生物的基因组或cDNA文库中分离获得。或者,它们可以通过重组方法或化学合成法来制备。
这些核酸分子可以采用根据本发明的分子或这些分子的部分或这些分子的反向互补分子,通过例如按标准方法(见例如Sambrook等,1989,分子克隆,实验室手册,第2版,Cold Spring Harbor Laboratory Press,Cold Spring Harbor,NY)进行杂交,从植物或其它生物体中获得鉴定和分离。
可以采用的杂交探针有例如完全或基本上具有SEQ ID NO.1所示核苷酸序列或该序列的部分的核酸分子。用作杂交探针的片段还可以是借助常规合成技术制备的、其序列基本上与根据本发明的核酸分子的序列一致的合成片段。一旦鉴定和分离了与根据本发明的核酸分子杂交的基因后,必须进行序列测定并分析该序列编码的蛋白质的性质。
本发明还涉及1999年8月5日保藏在德国Braunschweig的德意志微生物保藏中心、具有保藏号DSM 12970的质粒IR 65/87,并涉及包含在质粒IR 65/87插入片段中编码具有淀粉合成酶酶活性的蛋白质的核酸分子。此外,本发明还涉及包含在质粒IR 65/87插入片段中的核酸分子的片段,优选含有该编码区或其部分的片段。而且,本发明还涉及与质粒IR 65/87插入片段中包含的核酸分子杂交的核酸分子,并涉及具有与质粒IR 65/87中所包含核酸分子插入片段的全部或部分互补的序列的核酸分子。此外,本发明还涉及与质粒IR 65/87的插入核酸分子相比较,其核苷酸序列由于遗传密码的简并性而发生偏离的核酸分子。
本发明还涉及上述根据本发明的核酸分子的片段和等位变体。
在本文中,片段是根据本发明的核酸分子的部分,其编码根据本发明的蛋白质或该蛋白质的部分,而且通常是由根据本发明的核酸分子的约25-150个,优选至少150个,特别优选至少500个,尤其是至少1000个,特别优选至少3500个核苷酸组成的寡核苷酸或多核苷酸。
术语“片段”在本文中是指编码根据本发明的蛋白质的部分、并具有功能活性的本发明核酸分子的部分。而且,该片段还可以编码反义mRNA,或包含在介导共抑制效应或体内诱变效应的分子中。“具有功能活性”在本文中是指本发明核酸分子编码的蛋白质的生物活性在根据本发明的植物细胞中发生增加或降低。
所述等位变体不仅可以是天然存在的变体,还可以是合成的变体或通过重组DNA技术制备的变体。
本发明还涉及根据本发明的上述核酸分子的衍生物。术语“衍生物”在本文中是指这些分子的序列在一或多个位置上不同于上述核酸分子,但与这些序列,尤其是SEQ ID NO.1所示核苷酸序列的编码区有高度同源性。本文中同源性是指至少50%的序列一致性,尤其是至少70%、优选85%以上、特别优选95%以上的一致性。与上述核酸分子的偏离可能是由于缺失、替代、插入或重组造成的。
为了本发明的目的,“同源性”是指所讨论的核酸分子之间和它们编码的蛋白质之间存在功能和/或结构等同性。与上述分子同源并且构成这些分子的衍生物的核酸分子,通常是执行相同生物学功能、含有修饰的这些分子的变体。它们可以是天然存在的变体,例如来自其它生物体的序列,或是突变体,这些突变可以是天然存在的或可以通过定向诱变引入。
由本发明核酸分子的不同变体(片段、衍生物、等位变体)编码的蛋白质与SEQ ID NO.2中定义的氨基酸序列共同具有某些特征。这些特征可以包括例如酶活性、分子量、免疫学反应性、构象等,和物理性质例如凝胶电泳中的迁移行为、层析行为、沉淀系数、可溶性、光谱性质、稳定性、最适pH、最适温度等。
淀粉合成酶的重要特征是:1)其定位在植物细胞质体的基质中;ii)其能够采用ADP-葡萄糖作为底物合成线性α-1,4-连接的葡聚糖。该活性可以按Denyer和Smith(Planta 186(1992),609-617)所述方法测定。
根据本发明的核酸分子可以来源于表达所述基因的原核或真核生物,优选来源于植物,尤其是来源于淀粉合成或淀粉贮藏植物。这些植物可以是单子叶植物也可以是双子叶植物。在本文中特别优选例如谷类植物物种(例如大麦、黑麦、燕麦、小麦等)、玉米、稻、豌豆、木薯、马铃薯等。
根据本发明的核酸分子编码的蛋白质是至今从未进行过鉴定和性质分析的植物淀粉合成酶同工型。这些蛋白质具有淀粉合成酶的酶活性,并在SEQ ID NO.2所示氨基酸序列的第740位-第1170位氨基酸区域中表现出与马铃薯SSIII(Marshall等,Plant Cell 8,(1996),1121-1135)和称作zSSIII的玉米同工型(du1)(Cao等,PLant Physiol.120,(1999),205-215)显著同源。根据本发明的核酸分子编码的蛋白质与马铃薯SSIII和玉米zSSIII在N端显著不同。而且,SEQ ID NO.2所示蛋白质的计算等电点也与马铃薯SSIII和玉米zSSIII的计算等电点显著不同。而且,SEQ ID NO.2所示蛋白质与zSSIII(计算的分子量为约188kDa)相比具有明显低的约132kDa的计算分子量。与zSSIII不同,根据本发明的同工型的基因表达在幼叶中比在胚乳中高。
因此,在又一实施方案中,本发明涉及编码具有淀粉合成酶生物活性的蛋白质的上述根据本发明的核酸分子,这些分子优选编码在N端区域与SEQ ID NO.2所示氨基酸序列有至少50%、优选至少65%、尤其是至少80%、特别优选至少95%同源性的蛋白质。术语“N端”在本文中是指SEQ ID NO.2所示氨基酸序列的第1-150位氨基酸、优选第1-300位氨基酸、特别优选第1-480位氨基酸。
在本发明的再一实施方案中,根据本发明的核酸分子编码具有淀粉合成酶生物活性的蛋白质,这些分子编码的蛋白质具有6.95pH±1.00pH、优选6.95pH±0.75pH、特别优选6.95pH±0.50pH的计算等电点pI。

在本发明的再一优选实施方案中,根据本发明的核酸分子编码具有淀粉合成酶功能、且在Seq ID No.3-29所示一或多个基序中具有至少2个氨基酸、特别优选至少5个氨基酸、尤其是至少10个氨基酸、尤其优选至少20个氨基酸的缺失的蛋白质。
在本发明再一尤其优选的实施方案中,根据本发明的上述核酸分子编码的蛋白质的所述序列基序VII具有以下氨基酸序列:(1)SH---AMRYG-(11),该基序的第3、4、5和11位可以被任何氨基酸占据。在Seq ID No.2中,序列基序VII起始于第1067位氨基酸(=S)并终止于第1077位氨基酸(=S)。
在第3位,特别优选选自T、L、M、Q或R的氨基酸。在第4位,优选I、L或M的氨基酸。在第5位,优选选自I、Y、M、T或V的氨基酸。在第11位,优选选自S、T、C或A的氨基酸。
本发明还涉及上述根据本发明的核酸分子在筛选核酸文库,尤其是cDNA和基因组文库中的应用和/或作为杂交探针的应用。此外,本发明还涉及上述根据本发明的核酸分子在制备转基因植物细胞或转基因植物中的应用。而且,本发明还涉及含有上述根据本发明的核酸分子的载体,尤其是质粒、粘粒、病毒、噬菌体和其它通常在遗传工程中使用的载体。
在一个优选实施方案中,这些载体中包含的核酸分子与在原核或真核细胞中或在无细胞系统中确保转录和合成可翻译RNA的调节元件连接。用于在微生物(例如大肠杆菌(E.coli.)、酿酒酵母(S.cerevisiae))中表达根据本发明的核酸分子的调节元件已在文献中有大量的描述。允许下游基因获得尤其强表达的启动子有例如T7启动子(Studier等,酶学方法(Methods in Enzymology)185,(1990),60-89)、lacuv5、trp、trp-lacUV5(DeBoer等,Rodriguez和Chamberlin(编),《启动子、结构和功能》(Promoters,Structure and Function);Praeger,纽约,(1982),462-481;DeBoer等,Proc.Natl.Acad.Sci.USA(1983),21-25)、lpl、rac(Boros等,基因(Gene)42(1986),97-100)。通常,蛋白质的量从微生物生长周期的对数中期至对数末期达到最大。因此,优选采用诱导型启动子来合成蛋白质。这些启动子经常比组成型启动子导致更高的蛋白产量。由于克隆基因的持继转录和翻译,强组成型启动子的使用时常导致用于其它必需细胞功能的能量丧失,由此使细胞生长变慢(Bernard R.Glick/Jack J.Pasternak,Molekulare Biotechnologie(1995),Spektrum Akademischer Verlag GmbH,Heidelberg Berlin Oxford,第342页)。因此,为了获得最佳的蛋白量,通常采用两步骤法。首先,在最佳条件下使宿主细胞生长达到相对高的细胞密度。然后在第二步中,依据所采用的启动子类型,诱导转录。在本文中尤其适合的是乳糖或IPTG(=异丙基-β-D-硫代吡喃型半乳糖苷)诱导型tac启动子(deBoer等,Proc.Natl.Acad.Sci.USA 80(1983),21-25)。转录终止信号在文献中也已有描述。
用于在植物细胞中表达根据本发明的核酸分子的调节元件与根据本发明的植物细胞结合进行描述。
根据本发明的核酸分子在原核细胞例如大肠杆菌(Escherichia coli)中的表达是重要的,因为这使得可以对这些分子编码的酶的酶活性进行更为详细的表征。尤其是,可以在缺少植物细胞中参与淀粉合成的其它酶时对所讨论的酶合成的产物进行性质分析。这就使得可以对所讨论蛋白质在植物细胞的淀粉合成过程中具有的功能作出推断。
此外,还可以通过分子生物学常规技术(见例如Sambrook等,1989,分子克隆,实验室手册,第2版,Cold Spring Harbor Laboratory Press,Cold Spring Harbor,NY)将各种突变引入根据本发明的核酸分子中,这就导致合成生物学性质发生潜在改变的蛋白质。一方面,可以产生缺失突变体,其核酸分子通过从该编码DNA序列的5’或3’末端开始的渐进缺失来制备,这就导致合成适合的缩短蛋白质。例如,在该核苷酸序列5’末端的缺失使得可以鉴定出负责将该酶运输至质体的氨基酸序列(转运肽(transit peptides))。这就使得可以定向产生由于除去了所述序列而不再定位于质体而定位于细胞质中的酶,或定向产生由于加入其它信号序列而定位在其它区室中的酶。
还可以将原来的转运肽转换为介导在质体中定位的另一转运肽。可以采用的质体信号序列有例如菠菜铁氧还蛋白:NADP+氧化还原酶(ferrodoxin:NADP+ oxidoreductase)(FNR)。该序列含有质体蛋白质菠菜铁氧还蛋白:NADP+氧化还原酶cDNA的5’非翻译区和侧翼转运肽序列(核苷酸第-171位-第+165位;Jansen等,当代遗传学(Current Genetics)13,(1988),517-522)。
可以采用的另一质体信号序列是例如玉米蜡质蛋白(waxy protein)的转运肽加上该成熟蜡质蛋白的头34个氨基酸(Klosgen等,Mol.Gen.Genet.217,(1989),155-161)。此外,还可以采用不带该成熟蜡质蛋白头34个氨基酸的该玉米蜡质蛋白的转运肽(见上)。
另一方面,还可以在根据本发明的核酸分子的特定位置中引入点突变,以便改变的氨基酸序列对例如酶活性或酶调节产生影响。这使得可以产生具有改变的Kcat和/或Km值的突变体,或产生不再接受细胞中正常存在的通过变构调节或共价修饰进行的调节机制的突变体。
而且,还可以制备具有改变的底物或产物特异性的突变体,例如采用ADP-葡萄糖-6-磷酸而非ADP-葡萄糖作为底物的突变体。还可以制备具有改变的活性-温度曲线的突变体。
为了在原核细胞中进行重组操作,可以将根据本发明的核酸分子或这些分子的部分引入允许通过DNA序列重组发生诱变或序列修饰的质粒中。可以通过标准方法进行碱基替换,或添加天然或合成序列(参见Sambrook等,1989,分子克隆,实验室手册,第2版,Cold Spring Harbor LaboratoryPress,NY,USA)。为了将这些DNA片段彼此连接在一起,可以在这些片段上加上转接器(adapter)或接头(linker)。而且,可以进行操作以提供适合的限制性切割位点或除去多余的DNA或限制性切割位点。如果适合进行插入、缺失或替代,则可以采用体外诱变、引物修复、限制性酶切或连接。一般采用的分析方法是序列分析、限制性分析和其它生化和分子生物学方法。
而且,本发明涉及含有根据本发明的上述核酸分子的载体,该核酸分子与保证在原核或真核细胞中转录和合成可翻译RNA的调节元件按有义方向连接。术语“有义方向”的意义是本领域技术人员已知的。
在再一实施方案中,本发明涉及根据本发明的上述核酸分子或根据本发明的载体转化的宿主细胞,尤其是原核或真核细胞,以及来源于这些转化细胞并含有根据本发明的核酸分子或根据本发明的载体的细胞。这些细胞优选是细菌细胞、尤其优选是植物细胞。
本发明还涉及制备宿主细胞的方法,其中通过引入根据本发明的核酸分子和/或根据本发明的载体对宿主细胞进行遗传修饰。该宿主细胞可以是原核或真核生物来源的。该细胞优选是细菌细胞,尤其优选是植物细胞。
本发明中术语“遗传修饰”是指通过引入根据本发明的核酸分子改变宿主细胞、尤其是植物细胞的遗传信息,而且根据本发明的核酸分子的存在或表达导致表型的改变。在本文中表型改变优选是指一或多项细胞功能的可测量到的改变。例如,根据本发明的遗传修饰植物细胞表现出根据本发明的蛋白质的活性降低,或根据本发明的蛋白质的活性增加。
本发明的主题还有根据本发明的核酸分子编码的蛋白质或其生物活性片段,以及它们的制备方法,其中在允许该蛋白质合成的条件下培养根据本发明的宿主细胞,随后从该培养细胞和/或培养基中分离该蛋白质。根据本发明的蛋白质的特征性特性已结合相应的根据本发明的核酸分子在上面进行了描述。
现在通过提供根据本发明的核酸分子使得可以借助重组方法以一种前所未有的方式影响植物的淀粉代谢,和改变此代谢以致合成与野生型植物中合成的淀粉相比在例如其物理化学性质方面发生改变的改性淀粉,这些物理化学性质尤其是指直链淀粉/支链淀粉比率、分支程度、平均链长度、磷酸含量、糊化性能、和淀粉颗粒的大小和/或形状(淀粉颗粒的形态)。由于例如通过超量表达适合的核酸分子,或通过提供不再接受细胞中天然调节机制作用、和/或对于其活性而言具有不同温度依赖性的突变体,造成根据本发明的蛋白质的活性增加,由此可以增加适当遗传改造的植物中的产量。
仅是影响玉米中淀粉合成这一可能性就具有明显的经济重要性:玉米是全球最重要的淀粉植物。全球每年生产的淀粉大约80%来自玉米。
因此,为了增加所述淀粉合成酶的活性,可以在植物细胞中表达根据本发明的核酸分子。而且可以通过本领域技术人员已知的方法修饰根据本发明的核酸分子,以便获得不再接受细胞中天然存在的调节机制作用、或表现出改变的温度依赖性或底物或产物特异性的根据本发明的淀粉合成酶。
当在植物中表达根据本发明的核酸分子时,原则上可以使合成的蛋白质定位在植物细胞的任何区室中。为了实现在特定区室中的定位,必须缺失掉保证质体定位的序列,并任选地将剩余的编码区与确保在所指定的区室中定位的DNA序列连接。这些序列是已知的(见例如Braun等,EMBOJ.11(1992),3219-3227;Wolter等,Proc.Natl.Acad.Sci.USA 85(1988),846-850;Sonnewald等,Plant J.1(1991),95-106)。质体信号序列的例子已在上面不同部分中提及过了。
因此本发明还涉及根据本发明的核酸分子或根据本发明的载体转化的转基因植物细胞、和来源于这些转化细胞的转基因植物细胞。这些细胞含有根据本发明的核酸分子,该核酸分子优选与确保在植物细胞中转录的调节DNA元件,尤其是启动子连接。
有多种技术可以用于将DNA引入植物宿主细胞中。这些技术包括采用根癌农杆菌(Agrobacterium tumefaciens)或毛根农杆菌(Agrobacteriumrhizogenes)作为转化手段通过T-DNA转化植物细胞、利用聚乙二醇转化原生质体、注射、DNA的电穿孔、利用生物轰击法(biolistic approach)引入DNA、和其它可能的方法。
对农杆菌介导的植物细胞转化已经有深入的研究,并充分描述于EP-A2-0120516;Hoekema,《二元植物载体系统》(The Binary Plant VectorSystem),Offsetdrukkerij Kanters B.V.,Alblasserdam(1985),第V章;Fraley等,Crit.Rev.Plant Sci.4,1-46和An等,EMBO J.4,(1985),277-287。关于马铃薯的转化,见例如Rocha-Sosa等(EMBO J.8,(1989),29-33)。
通过基于农杆菌属(Agrobacterium)的载体进行的单子叶植物转化也已有描述(Chan等,Plant Mol.Biol.22,(1993),491-506;Hiei等,Plant J.6,(1994)271-282;Deng等,Science in China 33,(1990),28-34;Wilmink等,Plant Cell Reports 11,(1992),76-80;May等,Bio/Technology 13,(1995),486-492;Conner和Domisse,Int.J.PlantSci.153(1992),550-555;Ritchie等,Transgenic Res.2,(1993),252-265)。用于转化单子叶植物的另一系统是通过生物轰击方法转化(Wan和Lemaux,Plant Physiol. 104,(1994),37-48;Vasil等,Bio/Technology 11(1993),1553-1558;Ritala等,Plant Mol.Biol.24,(1994),317-325;Spencer等,Theor.Appl.Genet.79,(1990),625-631)、原生质体转化、部分透化细胞的电穿孔、和利用玻璃纤维引入DNA。尤其是玉米的转化已在文献中有多次描述(参照例如WO9506128A2、EP-A2-0513849、EP-A1-0465875、EP-A1-292435;Fromm等,生物技术(Biotechnology)8,(1990),833-844;Gordon-Kamm等,植物细胞(Plant Cell)2,(1990),603-618;Koziel等,生物技术11(1993),194-200;Moroc等,Theor.Appl.Genet.80,(1990),721-726)。
其它谷类物种的成功转化也已有描述,例如大麦(Wan和Lemaux,见上;Ritala等,见上;Krens等,自然(Nature)296,(1982),72-74)和小麦(Nehra等,Plant J.5,(1994),285-297)。
原则上,用于在植物细胞中表达根据本发明的核酸分子的调节元件可以是在植物细胞有活性的任何启动子、增强子、终止子等。可以选择启动子以使在根据本发明的植物中的表达组成型发生,或仅在特定组织、植物发育的特定时间点或外部因素确定的时间点发生。相对于该植物而言,该启动子可以是同源的或异源的。
适合启动子的例子有用于组成型表达的花椰菜花叶病毒35S RNA的启动子(见例如US5352605)和泛素启动子(见例如US5614399),用于在马铃薯中块茎特异性表达的patatin基因启动子B33(Rocha-Sosa等,EMBO J.8(1989),23-29),或保证仅在光合作用活跃组织中表达的启动子例如ST-LS1启动子(Stockhaus等,Proc.Natl.Acad.Sci.USA 84(1987),7943-7947;Stochhaus等,EMBO J.8(1989),2445-2451)、Ca/b启动子(见例如US 5656496;US 5639952;Bansal等,Proc.Natl.Acad.Sci.USA 89,(1992),3654-3658)和核酮糖二磷酸羧化酶-加氧酶SSU启动子(见例如US 5034322、US 4962028)。然而,也可以使用仅在外来因素确定的时间点上激活的启动子(见例如WO9307279A1)。在本文中,特别感兴趣的是允许简单诱导的热激蛋白启动子。而且,种子特异性启动子例如蚕豆(Vicia faba)USP启动子保证了在蚕豆和其它植物中的种子特异性表达(Fiedler等,Plant Mol.Biol.22,(1993),669-679;Baumlein等,Mol.Gen.Genet.225,(1991),459-467)。可以使用的其它启动子有果实特异性启动子,例如描述在WO9101373A1中的那些。
尤其优选使用的启动子是用于胚乳特异性表达的那些启动子,例如谷蛋白启动子(leisy等,Plant Mol.Biol.14,(1990),41-50;Zheng等,Plant J.4,(1993),357-366)、小麦HMG启动子、USP启动子、菜豆蛋白启动子、或玉米的玉米醇溶蛋白基因启动子(Pedersen等,细胞(Cell)29,(1982),1015-1026;Quatroccio等,Plant Mol.Biol.15(1990),81-93)。借助于胚乳特异性启动子,可以使根据本发明的核酸分子在胚乳中的转录本量与相应野生型植物的胚乳相比获得增加。
尤其优选使用玉米shrunken-1启动子(sh-1)(Werr等,EMBO J.4,(1985),1373-1380)。
还可以存在用于正确终止转录和在转录本上添加据认为具有稳定转录本功能的polyA尾的终止序列。这些元件在文献中已有描述(参见例如Gielen等,EMBO J.8(1989),23-29)并且可以随意地互换。
此外,借助根据本发明的核酸分子,还可以制备其中本发明蛋白质的活性降低的植物细胞和植物。这就导致合成与来自野生型植物细胞的淀粉相比具有改变的化学和/或物理性质的淀粉。
由此本发明的另一主题是与野生型植物的相应非遗传修饰植物细胞相比其中本发明蛋白质活性降低的转基因植物细胞。
为了本发明的目的,术语“野生型植物”/“野生型植物细胞”是指用作产生根据本发明的植物/植物细胞的起始材料的植物/植物细胞,即除了引入的遗传修饰外,其遗传信息与根据本发明的植物/植物细胞的遗传信息相符。
例如,可以通过表达相应的反义RNA、或通过表达适当构建的特异切割编码本发明蛋白质之一的转录本的核酶,成功地产生其中根据本发明的蛋白质活性降低的植物细胞。而且,还可以通过引入借助共抑制效应导致编码本发明蛋白质的内源性基因表达降低的DNA分子,实现活性的降低。而且,还可以通过体内诱变制备具有降低活性的本发明蛋白质的植物细胞。为此,可以通过同源重组在编码本发明蛋白质的内源性基因中引入突变或插入。该突变或插入导致编码本发明蛋白质的内源性基因的表达降低,或导致合成无活性的根据本发明的蛋白质。
为了降低植物细胞中根据本发明的蛋白质的活性,优选表达反义RNA。
为此,一方面,可以采用含有编码本发明蛋白质的完整序列的DNA分子,包括存在的任何侧翼序列在内,另一方面,可以采用仅含有编码序列的部分的DNA分子,但这些部分必须足够长以在细胞中引起反义效应。然而,也可以使用编码例如内含子的基因组DNA分子。一般,为了获得有效的反义抑制,可以采用最小长度到15bp的序列,优选长100-500bp的序列,尤其是长度大于500bp的序列。通常,采用短于5000bp的DNA分子,优选短于2500bp的序列。优选采用与待转化的植物物种同源的DNA分子。
还可以采用表现出与根据本发明的DNA分子的序列高度同源,但又不完全一致的DNA序列。最小同源性应大于约65%。优选采用具有95%-100%同源性的序列。
或者,还可以通过共抑制效应实现根据本发明的蛋白质在植物细胞中的活性降低。该方法是本领域技术人员已知的,描述于例如Jorgensen(Trends Biotechnol.8(1990),340-344)、Niebel等(Curr.Top.Microbiol.Immunol.197(1995),91-103)、Flavell等(Curr.Top.Microbiol.Immunol.197(1995),43-46)、Palaqui和Vaucheret(PlantMol.Biol.29(1995),149-159)、Vaucheret等(Mol.Gen.Genet.248(1995),311-317)、de Borne等(Mol.Gen.Genet.243(1994),613-621)和其它文献中。
同样,如同在上述反义技术中所述的一样,在本文中既可以采用包括根据本发明的核酸分子的完整编码区的DNA分子,也可以采用仅包括该编码序列的部分的DNA分子。还可以使用内含子。
表达核酶以降低细胞中某些酶的活性是本领域技术人员已知的,描述于例如EP-B1-0321201中。植物细胞中核酶的表达描述于例如Feyter等(Mol.Gen.Genet.250,(1996),329-338)。
而且,还可以通过“体内诱变”,即通过转化细胞在细胞中引入杂合RNA-DNA寡核苷酸(“嵌合体(chimeroplast)”),实现根据本发明的核酸分子在植物细胞中的活性降低(Kipp,P.B.等,第5届国际植物分子生物学大会的海报部分,1997年9月21-27,新加坡;R.A.Dixon和C.J.Arntzen,关于“转基因植物中代谢改造”的会议报告,KeystoneSymposia,Copper Mountain,CO,USA,TIBTECH 15,(1997),441-447;国际专利申请WO 9515972A1;Kren等,肝病学(Hepatology)25,(1997),1462-1468;Cole-Strauss等,科学(Science)273,(1996),1386-1389)。
该RNA-DNA寡核苷酸的一部分DNA成分与编码根据本发明的核酸分子的内源性基因的核酸序列同源,但与该内源性基因的核酸序列相比具有突变,或者含有同源区包围的异源区。
该RNA-DNA寡核苷酸和该内源性核酸分子在同源区域的碱基配对,以及之后的同源重组,使得存在于该RNA-DNA寡核苷酸的DNA成分中的突变或异源区域可以被转移至植物细胞的该内源性基因中。这就导致根据本发明的蛋白质活性降低。
本领域技术人员还知道,可以通过表达本发明蛋白质的无功能衍生物,尤其是反显性(transdominant)突变体,和/或表达这些蛋白质的拮抗剂/抑制剂,实现该蛋白质的活性。这些蛋白质的拮抗剂/抑制剂包括例如抗体、抗体片段或具有相似结合性质的分子。例如,细胞质scFv抗体被用于在遗传修饰的烟草植物中调节植物光敏素A蛋白质的活性(Owen,Bio/Technology 10,(1992),790-794;综述:Franken,E.,Teuschel,U.和Hain,R.,生物技术当代见解(Current Opinion in Biotechnology)8,(1997),411-416;Whitelam,Trends Plant Sci.1,(1996),268-272)。
为了本发明的目的,术语“活性降低”是指细胞中编码根据本发明的蛋白质的内源性基因的表达降低和/或根据本发明的蛋白质的量减少,尤其是指细胞中根据本发明的蛋白质的酶活性降低。
表达的降低可以通过例如借助Northern印迹分析测量编码根据本发明的蛋白质的转录本的量来确定。在此“降低”优选指与相应的非遗传修饰细胞相比转录本的量降低至少50%、优选至少70%、特别优选至少85%、极为特别地优选至少95%。
根据本发明的蛋白质的量的减少可以例如通过Western印迹分析来确定。在此“减少”优选指与相应的非遗传修饰细胞相比,根据本发明的蛋白质的量减少至少50%、优选至少70%、特别优选至少85%、极为特别地优选至少95%。
根据本发明的蛋白质的酶活性降低可以例如按Denyer和Smith(Planta 186(1992),609-617)所描述的方法进行测定。在本文中,与相应的非遗传修饰细胞相比,酶活性的降低优选指至少50%、优选至少70%、特别优选至少85%、极为特别优选至少95%的降低。
以上在不同部分提及的关于术语“遗传修饰”的说明也适用于此处。
因此,特别地,本发明的主题还有转基因植物细胞。
a) 其含有导致合成至少一种反义RNA的至少一个DNA分子,其中该反义RNA可以引起编码本发明蛋白质的内源性基因表达降低;
b) 其含有至少一个DNA分子,该DNA分子通过共抑制效应导致编码本发明蛋白质的内源性基因表达降低;

而且,借助根据本发明的核酸分子,还可以制备其中根据本发明的蛋白质活性增加的植物细胞和植物。这就导致合成与来自野生型植物细胞和野生型植物的淀粉相比较具有改变的化学和/或物理性质的淀粉。
本发明的另一主题是与野生型植物的相应非遗传修饰植物细胞相比其中根据本发明的蛋白质活性增加的转基因植物细胞。
为了制备与相应的非遗传修饰野生型植物细胞相比具有增加活性的本发明蛋白质的植物细胞,按有义方向使用含有根据本发明的淀粉合成酶编码区的本发明核酸分子。在再一实施方案中,还可以使用该编码区的部分,条件是它们编码具有催化活性的淀粉合成酶蛋白质。在一个尤其优选的实施方案中,使用Seq ID No.1所示核酸分子。
为了本发明的目的,术语“活性增加”是指细胞中编码根据本发明的蛋白质的内源性基因的表达增加,和/或根据本发明的蛋白质的量增加,尤其是指细胞中根据本发明的蛋白质的酶活性增加。
表达的增加可以通过例如借助Northern印迹分析测量编码根据本发明的蛋白质的转录本的量来确定。在此“增加”优选指与相应的非遗传修饰细胞相比较转录本的量增加至少50%、优选至少100%、尤其优选至少500%、特别地优选至少1500%。
根据本发明的蛋白质的量的增加可以例如通过Western印迹分析来确定。在此“增加”优选指与相应的非遗传修饰细胞相比,根据本发明的蛋白质的量增加至少50%、优选至少100%、尤其优选至少500%、特别地优选至少1500%。
根据本发明的蛋白质的酶活性的增加可以例如按Denyer和Smith(见上)所描述的方法进行测定。与相应的非遗传修饰细胞相比,酶活性的增加在此优选指至少50%、优选至少100%、尤其优选至少500%、特别地优选至少1500%的增加。
在本发明的一个优选实施方案中,与野生型植物的植物细胞相比具有增加活性的本发明蛋白质的本发明转基因植物细胞是来源于淀粉贮藏组织、优选块茎和胚乳、尤其是玉米植物的胚乳的植物细胞。
令人惊奇的是,已经发现,与相应的野生型植物相比其中根据本发明的蛋白质活性增加的植物细胞,尤其是胚乳的植物细胞,合成与野生型植物中,尤其是胚乳中合成的淀粉相比其物理化学性质发生改变的淀粉,以致该淀粉可以更好地适用于特定目的。
根据本发明的植物细胞可以属于任何植物物种,即单子叶植物或者双子叶植物。它们优选是来自农业上有用植物(即为了营养目的或技术目的,尤其是工业目的人为栽培的植物)的植物细胞。本发明优选涉及纤维形成植物(例如亚麻、大麻、棉)、储油植物(例如油菜、向日葵、大豆)、储糖植物(例如甜菜、甘蔗、糖粟(sugar millet)、香蕉)和储蛋白植物(例如豆科植物)。
在再一优选实施方案中,本发明涉及饲料植物(例如用作动物饲料的草、牧草、紫花苜蓿、三叶草等)、蔬菜植物(例如番茄、莴苣、苣荬菜)的植物细胞。
在一个尤其优选的实施方案中,本发明涉及淀粉贮藏植物(例如小麦、大麦、燕麦、黑麦、马铃薯、玉米、稻、豌豆、木薯、绿豆)的植物细胞,尤其优选玉米、稻、小麦和马铃薯的植物细胞。
根据本发明的植物细胞可以用于再生全株。通过根据本发明的转基因植物细胞的再生可以获得的植物也是本发明的主题。
本发明的主题还有含有根据本发明的上述植物细胞的植物。原则上,根据本发明的植物可以是任何植物物种的植物,即单子叶植物或双子叶植物。它们优选是来自农业上有用植物(即为了营养目的或技术目的,尤其是工业目的人为栽培的植物)的植物细胞。本发明优选涉及纤维形成植物(例如亚麻、大麻、棉)、储油植物(例如油菜、向日葵、大豆)、储糖植物(例如甜菜、甘蔗、糖粟、香蕉)和储蛋白植物(例如豆科植物)的植物细胞。

在其它优选实施方案中,根据本发明的植物在淀粉贮藏组织的植物细胞中,优选在块茎或胚乳的植物细胞中,尤其优选在玉米植物的胚乳植物细胞中,与来自野生型植物的相应非遗传修饰植物细胞相比,表现出增加活性的根据本发明的蛋白质。
在本发明的再一实施方案中,含有其中本发明蛋白质活性增加的本发明植物细胞的植物,与非修饰野生型植物相比,表现出更高的产量和/或更高的淀粉含量。
术语“更高的产量”在此优选指组分,尤其是淀粉,和/或生物质的产量增加,尤其是在后者按每株植物鲜重测量时如此。
术语“增加的淀粉含量”在此是指与未修饰的野生型植物的植物细胞相比,根据本发明的植物细胞中淀粉含量高出至少10%、优选至少20%、尤其是至少30%、特别优选至少40%。
测定淀粉含量的方法是本领域技术人员已知的。
在产量和/或淀粉含量上的这种增加优选涉及可收获的植物器官例如种子、果实、储藏根、块茎、根、花、芽、枝条、茎或木材。
根据本发明,当在相同条件下生长时,与相同基因型的相应非转化植物相比较,基于该生物质和/或组分的产量增加至少3%,优选与野生型植物相比增加至少5%、尤其是至少10%、特别优选至少20%或甚至40%。
本发明还涉及产生转基因植物的方法,其中
a) 通过引入根据本发明的核酸分子和/或根据本发明的载体,对植物细胞进行遗传修饰;并且
b) 从细胞再生植物;并任选地,
c) 从b)的植物产生更多的植物。
在以上不同部分已经解释过的遗传修饰的说明也适用于步骤a)中所述引入的遗传修饰。例如,遗传修饰的植物细胞表现出根据本发明的蛋白质的活性降低,或根据本发明的蛋白质的活性增加。
可以通过本领域技术人员已知的方法根据步骤b)再生出植物。
依据本发明方法的步骤c),通过例如无性繁殖(如采用插条、块茎,或通过愈伤组织培养和再生全株)或有性繁殖可以产生更多的植物。有性繁殖优选在受控条件下进行,即在具有特定性状的所选植株之间互相进行杂交并繁殖。
本发明还涉及通过根据本发明方法可以获得的植物。
本发明还涉及根据本发明的植物的繁殖材料,和依据本发明方法产生的转基因植物的繁殖材料。术语繁殖材料包括适合于通过无性或有性繁殖途径产生后代的那些植物部分。适合于无性繁殖的例子有插条、愈伤组织培养物、根茎或块茎。其它繁殖材料包括例如果实、种子、幼苗、原生质体、细胞培养物等。优选地,该繁殖材料是块茎和种子。
在再一实施方案中,本发明涉及根据本发明的植物的可收获部分,例如果实、储藏根、根、花、芽、枝条或茎,优选种子或块茎,并涉及含有这些植物可收获部分,优选种子或块茎的食物。根据本发明的植物的可收获部分,优选种子或块茎,和/或食物可以用改变的能量值,优选增加的能量值来表征。
术语“能量值”在本文中尤其是指“可消化的能量”。术语“可消化的能量”定义为:可消化的能量=供给的总能量-排泄物的热量值(Landwirtschaftliches Lehrbuch 2:Tierzucht,Ed.:D.Schmidt,第5版,1982,Eugen Ulmer GmbH & Co,第244页)。总能量是指可以测量到的以焦耳为单位的食物总热量值。确定“能量值”或“可消化的能量”的方法是本领域技术人员已知的。

由于根据本发明的核酸分子的表达或额外表达,和/或由于根据本发明的蛋白质的活性增加或降低,根据本发明的转基因植物细胞和植物合成的淀粉与野生型植物合成的淀粉相比,在例如其物理化学性质方面,尤其是直链淀粉/支链淀粉比例、分支程度、平均链长度、磷酸含量、糊化性能、淀粉颗粒大小和/或淀粉颗粒形状方面,发生改变。尤其是,与野生型淀粉相比,该淀粉可以在该淀粉胶料所具有的粘度和/或胶体形成性质方面发生改变。
本发明的主题还有可从根据本发明的转基因植物细胞和/或根据本发明的植物和/或根据本发明的繁殖材料获得的淀粉。
由于改变的理化性质,根据本发明的淀粉表现出改变的功能性质。淀粉或其水溶液的重要功能性质有退化性能、膜形成性质、胶体强度、粘度、颜色的稳定性、酶可消化性、冻融稳定性、酸稳定性、剪切稳定性、透明度、水结合能力、糊化性质、和结合与粘着性质。根据本发明的淀粉可以通过本领域技术人员已知的方法进行修饰,并以其未修饰或修饰的形式适合于食品或非食品部门中的多种应用。
原则上,可以将淀粉的可能用途分为两个重要方面。一个方面包括通过酶学或化学方法获得的淀粉水解产物,主要是葡萄糖和葡聚糖单元。它们被用作其它化学修饰和加工例如发酵的起始材料。在此为了降低费用重要的是水解方法的简单性和廉价设计。目前该方法基本上是采用淀粉葡萄糖苷酶通过酶学方法来进行的。通过采用较少的酶来节省经济开支将是合理的。这可以通过改变淀粉的结构来实现,例如通过颗粒表面面积的增加、由较低的分支程度导致的更好的可降解性、或限制所用酶接近的立体结构来实现。在另一方面,淀粉由于其聚合结构而以所谓天然淀粉的形式被应用,该方面可以进一步划分为两个应用领域:1.食品工业
淀粉是各种食品的典型添加物,其中它基本上用作水性粘合添加物或
引起粘度的增加或胶体形成的增加。重要的特性有流动和吸附性能、
溶胀和糊化温度、粘性和增稠性能、淀粉的溶解度、透明度和糊剂结
构、热、剪切和酸抗性、退化倾向、膜形成能力、冻/融抗性、可消
化性、以及与例如无机或有机离子形成复合物的能力。2.非食品工业
在这个大领域中,淀粉可以在各种生产过程中用作辅助剂或在技术产
品中用作添加剂。淀粉用作辅助剂的主要领域有纸和纸板工业。在该
领域中,淀粉主要用于滞留(滞留固体)、对填充物和细微颗粒进行
大小分级、使物质固化和脱水。此外,还利用淀粉在稠度、硬度、声
音、手感、光泽、平滑性、撕裂强度以及表面方面具有的有利性质。2.1纸和纸板工业
在纸生产过程中,可区分出4个应用领域,即表面、涂层、备料和喷
雾。表面处理对淀粉的要求主要是高亮度、相应粘性、高粘性稳定性、
良好的膜形成以及低产尘。当用于固体内容物涂层时,相应粘性、高
结合能力以及高色素亲合力有重要作用。作为备料添加剂,在纸浆中
快速、均一、不损耗的分散,高机械稳定性和完全滞留是重要的。当
使用淀粉喷雾时,固体的相应含量、高粘性和高结合能力也是重要的。2.2粘合剂工业
一个主要应用领域是例如在粘合剂工业中,其中淀粉应用于4个方
面:用作纯淀粉胶、在以特殊化学制品制备的淀粉凝胶中的应用、使
用淀粉作为合成树脂和聚合分散相的添加剂以及使用淀粉作为合成粘
合剂的增充剂。在所有基于淀粉的粘合剂中90%用于生产瓦楞纸板、
各种纸袋、纸和铝的复合材料、纸盒和信封、邮票等的湿胶。2.3纺织品和织物保养工业
淀粉作为辅助剂和添加剂的另一可能用途是生产纺织品和织物保养
品。在纺织品工业中,可区分出下列4个应用领域:将淀粉用作上浆
剂,即作为平滑处理和加强去毛刺性能的辅助剂以对抗纺织中牵引的
张力以及提高纺织过程中的耐磨强度,主要在质量降低性预处理如漂
白、染色等之后用作纺织品的整理剂,在染料糊剂生产中用作增稠剂
以避免染料扩散,和用作缝纫纱线束缚剂的添加剂。2.4建筑业
淀粉的第4个应用领域是用作建筑材料中的添加剂。一个例子是石膏
板板材的生产,其中在稀石膏浆中混合的淀粉与水一起糊化、在石膏
板表面扩散因而使纸板与该板粘合。其它应用领域有将淀粉与泥灰和
矿物纤维混合。在混合好的混凝土中,淀粉可用于减缓硬化过程。2.5土壤稳定化
而且,淀粉在生产土壤稳定剂中是有利的,可用于在人工侵拢土壤时
临时性保护土壤颗粒免于失水。根据本领域现有知识,由淀粉和聚合
物乳剂组成的混合产品可认为具有与现有产品相同的降低冲蚀和结壳
的作用;然而,它们便宜得多。2.6淀粉在植物保护剂和肥料中的应用
另一个应用领域是淀粉用于植物保护剂中以改变这些制品的特定性
质。例如,可使用淀粉改善植物保护剂和肥料的可润湿性,定量释放
有效成分,将液体、挥发性和/或不良气味的有效成分转换成微晶、
稳定、可变形的物质,混合不相容组合物和由于分解减缓而延长作用
持续时间。2.7药品、医疗和化妆品工业
淀粉也可用于药品、医疗和化妆品工业领域。在制药工业中,淀粉可
用作片剂粘合剂或用于胶囊粘合剂的稀释。此外,淀粉适于用作片剂
的崩解剂,因为吞服后它吸收液体并在短时间后溶胀以致活性成分释
放。由于性质上的原因,医用润滑粉末和疗伤用药粉也以淀粉为基础。
在化妆品领域,例如淀粉可用作粉末添加物如香水和水杨酸的载体。
淀粉的一个相当广泛的应用领域是牙膏。2.8淀粉作为煤和煤球的添加物
淀粉也可用作煤和煤球的添加物。通过加入淀粉,煤可定量地成块和
/或形成高质量的煤球,从而防止煤球过早分解。烧烤用煤添加了
4%-6%淀粉,高热煤含有约0.1%-0.5%淀粉。而且,淀粉作为结合剂
变得愈来愈重要,因为在煤和煤球中加入淀粉可显著降低有害物质的
释放。2.9矿石和煤浆的加工
而且,淀粉可用作矿石和煤浆加工过程中的凝聚剂。2.10淀粉在铸造过程中作为添加物
另一个应用领域是淀粉在铸造过程中用作加工材料的添加物。对于各
种铸造工艺,需要从沙石混合粘合剂产生的核心。目前最常用的粘合
剂是混合了改性淀粉、主要是溶胀淀粉的膨润土。加入淀粉的目的是
增加流动阻力以及改善结合强度。而且溶胀淀粉可符合该生产工艺的
其它必要条件如冷水中的可分散性、再水合能力、与沙石的良好混合
性和高度水结合能力。2.11淀粉在橡胶工业中的应用
在橡胶工业中淀粉可用于改善技术和光学质量。其原因是改善表面光
泽、手感和外观。为此,在冷固化之前将淀粉分散在粘性的橡胶物质
的橡胶处理表面上。淀粉还可用于改善橡胶的可印刷性。2.12皮革替代品的生产
改性淀粉的另一应用领域是生产皮革替代品。2.13合成聚合物中的淀粉
在塑料市场存在下列应用领域:淀粉产品在加工工艺中的应用(淀粉
只是填充剂,合成聚合物和淀粉之间没有直接键合),或淀粉产物在
聚合物生产中的应用(淀粉和聚合物形成稳定的键)。
使用淀粉作为纯填充剂是不能与其它物质如滑石竞争的。当特定淀粉性质变得有效因而终产物的性质明显改变时这种情况就不同了。一个例子是淀粉产品在热塑性材料如聚乙烯的加工过程中的应用。由此,淀粉和合成聚合物可以通过共作用以1∶1的比例结合形成“母材料”,由此使用制成粒状的聚乙烯采用常规技术可生产各种产品。淀粉在聚乙烯膜中的整合可造成空心体中物质通透性的增加,水蒸气通透性的改善,抗静电性能的改善,抗阻塞性能的改善以及水性染料可印刷性的提高。
另一种可能是淀粉在聚氨酯泡沫胶中的应用。由于淀粉衍生物的适应性以及加工技术的优化,有可能特异地控制合成聚合物和淀粉羟基之间的反应。结果是由于淀粉的使用,聚氨酯膜获得下列特性:降低的热膨胀系数、减少的收缩性能、改善的压力/张力性能、增加的水蒸气通透性而又不改变对水的吸收、降低的可燃性和破裂密度、可燃部分无液滴形成、无卤化物和老化的减缓。目前仍然存在的缺点是压力和冲击强度的降低。
膜产品的开发不再是唯一的选择。也可以生产具有50%以上淀粉含量的固体塑料产品如罐、盘和碗。而且,淀粉/聚合物混合物具有在更大程度上可被生物降解的优点。
而且,由于它们极易与水结合,淀粉接枝聚合物已经具有了极大的重要性。这些产品具有淀粉骨架和根据自由基链反应机制在其上接枝的合成单体的侧链。目前已有的淀粉接枝聚合物的特征是改善的水结合和保持能力,在高粘度时每克淀粉结合和保持高达1000g的水。在过去的几年,这些超吸收剂已获得了更为广泛的应用-主要应用于卫生领域,如尿布和床单等产品,以及农业部门如种子小球。
应用重组DNA技术修饰的新淀粉的决定性因素,一方面是结构、含水量、蛋白含量、脂含量、纤维含量、灰/磷酸含量、直链淀粉/支链淀粉比、相对摩尔质量的分布、支化度、颗粒大小和形状以及结晶性,另一方面是导致下列性质的特性:流动和吸附性能、糊化温度、粘度、增稠性能、溶解度、糊状结构和透明性、热、剪切和酸的耐受性、退化倾向、凝胶形成能力、冻/融抗性、复合物形成能力、碘结合能力、膜形成能力、粘合强度、酶稳定性、可消化性和反应性。
通过采用转基因植物进行遗传操作产生改性淀粉,可以改变从该植物获得的淀粉的性质,以致不再需要通过化学或物理方法进行进一步修饰。另一方面,可对通过重组DNA技术修饰的淀粉进行进一步化学修饰,这将为某些上述应用领域产生进一步的质量改善。原则上,这些化学修饰是本领域技术人员已知的。这些修饰尤其是指通过下列方法进行的修饰:
-热处理
-酸处理
-氧化和
-酯化作用
这些修饰导致形成磷酸、硝酸、硫酸、黄原酸、乙酸和柠檬酸淀粉。其它有机酸也可用于酯化:
-形成淀粉醚

-形成淀粉接枝聚合物。

(a)细胞系DSM 6009原生质体的制备
原生质体分离
在最后一次更换原生质体悬浮培养物中的培养基后2-4天,优选
3天,将该液体培养基吸出,用500ml原生质体洗涤溶液1洗涤留
下的细胞,并通过吸出该洗涤溶液作再一次干燥。在每个收获的2g 细胞生物质中加入10ml原生质体分离介质。将该重悬细胞和细胞团聚体在暗处于27±2℃孵育4-6小时,此过程中伴随轻柔振摇(30-40rpm)。原生质体纯化当每ml已释放了1百万个原生质体(显微镜下观察)时,将该悬浮液通过网目大小分别为200μm和45μm的不锈钢筛和尼龙筛。将一个100m和一个60μm的筛组合在一起也同样适合于除去细胞团聚体。在显微镜下评价含有原生质体的滤出液。通常,该滤出液含有98-99%的原生质体。其余是未消化的单细胞。此纯度的原生质体制备物无需额外的梯度离心即可用于转化实验。通过离心(在吊桶式转头中100rpm(100×g,3min))沉淀原生质体。丢弃上清液并将原生质体重悬在洗涤溶液1中。重复该离心步骤,然后将原生质体重悬在转化缓冲液中。(b)原生质体转化将重悬在转化缓冲液中的原生质体以10ml为一份的量分装在50ml同质异晶聚合物管中,滴度为0.5-1×106个原生质体/ml。将用于转化的DNA溶解在Tris-EDTA(TE)缓冲液中。向每ml原生质体悬浮物中加入20μg质粒DNA。所用载体为介导膦丝菌素抗性的质粒(参见例如EP-A2-0513 849)。加入DNA后,小心摇动该原生质体悬浮物,以使DNA均匀地分散在该溶液中。之后立即滴加5mlPEG溶液。小心地转动管子以使该PEG溶液均匀分布。然后,再加入5ml PEG溶液,并重复该混合步骤以获得均一化。将原生质体在该PEG溶液中于25±2℃保持20分钟。然后通过离心3分钟(100g;1000rpm)沉淀原生质体。丢弃上清液。通过在20ml W5溶液中小心振摇对原生质体进行洗涤,之后再次离心。然后将它们重悬在20ml原生质体培养基中,再次离心并再次重悬在培养基中。将滴度调节至6-8×105个原生质体/ml,并将这些原生质体以3ml为一 部分培养在培养皿(φ60mm,高15mm)中。用Parafilm密封这些培养皿,然后将其于25±2℃置于暗处。(c)原生质体培养在分离和转化后的头2-3周内,在不添加新鲜培养基的情况下培养原生质体。一旦从原生质体再生出的细胞发育成超过20-50个细胞的细胞团聚体后,添加含有蔗糖(90g/l)、渗透压相同的1ml新鲜原生质体培养基。(d)筛选转化的玉米细胞,和植株的再生加入新鲜培养基后3-10天,可以将从原生质体形成的细胞团聚体接种在补加了100mg/l L-膦丝菌素的琼脂培养基上。含有原生质体培养基中的维生素、90g/l蔗糖和1.0mg/l 2,4-D的N6培养基,与例如MS培养基(Murashige和Skoog(1962),见上)中的大量和微量元素一起,同样地适合作为类似培养基。来源于稳定转化原生质体的愈伤组织可以在该选择培养基上无阻碍地持续生长。3-5周,优选4周后,可以将该转基因愈伤组织转移至同样含有100mg/l L-膦丝菌素、但不再含有任何生长素的新鲜选择培养基中。在3-5周的期间内,50%在其基因组中整合有L-膦丝菌素乙酰转移酶基因的转基因玉米愈伤组织在含有L-膦丝菌素的该培养基上分化出第1批植物。(e)培养转基因再生植株在含有5×10-4M L-膦丝菌素的无激素N5培养基(Chu C.C.等,Sci.Sin.16(1975),659)上培养胚胎发生的转化转基因玉米组织。在该培养基上,充分表达膦丝菌素乙酰转移酶基因(PAT基因)的玉米胚发育成植株。未转化的胚、或仅具有很弱的PAT活性的胚将死亡。当体外植物的叶达到4-6cm长时,可将它们转移至土壤中。将根上的琼脂残余物洗掉后,将植株种植在壤土、沙、蛭石和标
准土壤的3∶1∶1∶1混合物中,并在移植到90-100%相对空气湿度
中后头3天中使其适应土壤培养。在具有14小时光周期、按植物
水平约25,000Lux的人工气候室中,以23±1/17±1℃的日/夜温
度,对这些植株进行培养。将这些适应植物培植在65±5%的空气
湿度中。4.DNA片段的放射性标记
利用Boehringer(德国)的DNA随机引物标记试剂盒,按照厂家说明
书对DNA片段进行放射性标记。以下实施例在于举例说明本发明,而不构成任何形式的限制:实施例1:鉴定、分离和表征编码新的玉蜀黍(Zea mays)淀粉合成酶同工
型的cDNA
为了鉴定编码新的玉米淀粉合成酶同工型的cDNA,首先按照Logemann等(Anal.Biochem.163,(1987),21-26)的方法,从玉米仁(授粉后15-20天)中分离总RNA。然后利用Oligotex-mRNA纯化试剂盒(Quiagen),按照厂家说明书,将1mg总RNA用以制备polyA+-RNA。
然后利用Stratagene的ZAP cDNA合成试剂盒从5μg polyA+-RNA构建cDNA文库。该文库含有约9×105个独立的重组噬菌体,cDNA插入片段的平均大小为约1.3kb。
然后对约4×105个噬菌体进行菌斑影印(plaque-lifting)。为此,采用Hybond N滤膜(Amersham)。在缓冲液A(5×SSC,0.5%BSA,5×Denhardt,1%SDS,40mM磷酸缓冲液,pH7.2;100mg/l鲑精DNA,25%甲酰胺)中42℃预杂交4个小时,然后采用放射性标记(随机引物DNA标记试剂盒,Boehringer Mannheim)的马铃薯(Solanum tuberosum)SSIII cDNA(GenBank登录号X 94400)的EcoRI/XhoI片段(总cDNA)对该滤膜进行杂交。12小时后,在含有3×SSC、0.5%SDS的缓冲液中55℃20分钟洗涤该滤膜3次。然后,在其上放置一张X光片约14个小时。
分离强杂交噬菌斑,并将其稀释、更新并转移至滤膜上。按上述再次进行杂交和洗涤。按照厂家说明书(Stratagene)体内切割这些分离的噬菌体后,分离各种质粒,并通过限制性分析对它们进行表征。然后确定具有最长的cDNA插入片段的质粒DNA序列。其中一个质粒命名为pZm_SS6,含有SEQ ID NO.1中所示核苷酸序列。实施例2:制备用于转化植物的载体
从含有SEQ ID NO.1所示核苷酸序列完整编码区的pZm_SS6质粒的NotI/Bsp 120I片段起始,通过标准方法构建载体IR65/87(见图1),该载体尤其适合于产生根据本发明具有增加活性(过量表达方案)或降低活性(共抑制解决方案)的淀粉合成酶的转基因玉米植物,并于1999年8月5日被保藏在德意志微生物保藏中心(Deutsche Sammlung fürMikroorganismen),保藏号为DSM 12970。
根据上述方法采用载体IR65/87转化玉米植物。
然后筛选与相应未转化玉米植物相比表现出本发明蛋白质活性增加或活性降低的转基因植物。
Claims (22)
1.编码具有淀粉合成酶生物活性的蛋白质或该蛋白质的生物活性片段的核酸分子,其选自
(a)编码具有SEQ ID NO.2所示氨基酸序列的蛋白质的核酸分子;
(b)含有SEQ ID NO.1所示核苷酸序列或其互补序列的核酸分子;
(c)含有质粒IR 65/87(保藏号DSM 12970)中所存在的cDNA核苷酸
序列的编码区、或其互补序列的核酸分子;
(d)其核苷酸序列由于遗传密码的简并性而不同于(a)、(b)或(c)所述
核酸分子的序列的核酸分子;和
(e)构成(a)、(b)、(c)或(d)所述核酸分子的片段、等位变体和/或衍
生物的核酸分子。
2.根据权利要求1的核酸分子,其是DNA分子。
3.根据权利要求1的核酸分子,其是RNA分子。
4.含有根据权利要求1-3之一项或多项的核酸分子的载体。
5.根据权利要求4的载体,其含有在原核和/或真核细胞中保证转录所述核酸分子和/或合成可翻译RNA的一或多个调节元件。
6.根据权利要求4-5之一项或多项的载体,其中所述核酸分子以有义方向和在原核和/或真核细胞中保证可翻译RNA转录和合成的调节元件相连接,或其中所述核酸分子以反义方向和在原核和/或真核细胞中保证非可翻译RNA转录和合成的调节元件相连接。
7.用根据权利要求1-3之一项或多项的核酸分子或根据权利要求4-6之一项或多项的载体转化的宿主细胞,或来源于该细胞的细胞。
8.根据权利要求7的宿主细胞,其是植物细胞。
9.根据权利要求1-3之一项或多项的核酸分子编码的蛋白质或其生物活性片段。
10.产生根据权利要求9的蛋白质或其生物活性片段的方法,其中在允许合成该蛋白质的条件下培养根据权利要求7-8之一项或多项的宿主细胞,并从该培养细胞和/或培养基中分离该蛋白质。
11.根据权利要求8的转基因植物细胞,其中编码具有淀粉合成酶生物活性的蛋白质或其生物活性片段的所述核酸分子处于允许在植物细胞中转录可翻译mRNA的调节元件的控制之下。
12.根据权利要求8的转基因植物细胞,其中根据权利要求9的蛋白质或其生物活性片段的活性在该植物细胞中与在来自野生型植物的相应非遗传修饰植物细胞中相比发生降低。
13.根据权利要求8的转基因植物细胞,其中根据权利要求9的蛋白质或其生物活性片段的活性在该植物细胞中与在来自野生型植物的相应非遗传修饰植物细胞中相比发生增加。
14.含有根据权利要求8和11-13之一项或多项的植物细胞的植物。
15.根据权利要求14的植物,其是作物植物。
16.根据权利要求14-15之一项或多项的植物,其是淀粉贮藏植物。
17.根据权利要求14-16之一项或多项的植物,其是玉米植物。
18.产生根据权利要求8的转基因植物细胞的方法,其中通过引入根据权利要求1-3之一项或多项的核酸分子和/或根据权利要求4-6之一项或多项的载体,对植物细胞进行遗传修饰。
19.产生根据权利要求14的转基因植物的方法,其中
(a)通过引入根据权利要求1-3之一项或多项的核酸分子和/或根据
权利要求4-6之一项或多项的载体,对植物细胞进行遗传修饰;
并且
(b)从该细胞再生植物;并任选地,
(c)从(b)的植物产生更多的植物。
20.根据权利要求14-17之一项或多项的植物的繁殖材料,其含有根据权利要求8和11-13之一项或多项的植物细胞。
21.从根据权利要求8和11-13之一项或多项的植物细胞、根据权利要求14-17之一项或多项的植物、或根据权利要求20的繁殖材料中获得的淀粉。
22.产生根据权利要求21的改性淀粉的方法,包括步骤:从根据权利要求8和11-14之一项或多项的植物细胞、根据权利要求14-17之一项或多项的植物和/或根据权利要求20的繁殖材料中提取淀粉。
