在农业和林业领域,人们一直长期努力提供产量增加的植物,这特别是为了保证持续增长的世界人口的食物供应和保证再生原材料的供应。传统作法是试图通过育种获得高产植物。然而,这种方法既费时又成本高。而且,对于每一个目标植物种都要进行相应的育种计划。
在一定程度上,通过植物的遗传操作即在植物中有目的地导入和表达重组核酸分子,已获得了进展。该方法的优点是它们通常不限于一个植物种,也可转移至其它植物物种。
例如,在EP-A 0 511 979中,描述了原核天冬酰胺合成酶在植物细胞中表达导致生物量生产增加。WO 96/21737描述了例如通过表达不受调节的果糖-1,6-二磷酸酶,由于光合速率增加而增加植物产量。然而,仍然需要普遍适用的改善农业和林业上目标植物产量的方法。
而且,随着植物所含物质作为原材料的可再生来源起着越来越重要的作用,生物技术研究中的一个问题是调整所述植物原材料以满足加工业的要求。为了使再生原材料可在尽可能多的领域应用,更需要获得大范围的物质。此外,为了提高从植物中生产可再生来源的原材料的效率,必需增加所述植物内含物质的产量。
除了脂肪和蛋白质外,油和多糖是主要的再生植物原材料。除了纤维素以外,多糖的主要形式是作为高等植物最重要的贮藏物质之一的淀粉。其中,特别是马铃薯和玉米是人们感兴趣的植物,因为它们是生产淀粉的重要栽培作物。
除了在食品工业中应用之外,植物界最重要的贮藏物质之一多糖淀粉广泛用作生产工业产品的再生原材料。

对于工业加工马铃薯淀粉,干重(淀粉含量)是一个关键值,因为它决定加工的成本。干重(淀粉含量)增加意味着产量相同时马铃薯块茎的含水量降低。含水量降低导致运输费用降低,烹调时所需确切烹调时间减少。
因此,看起来期望提供淀粉含量增加的植物细胞和植物,以及生产这种植物细胞和植物的方法。而且,看起来期望提供其直链淀粉和支链淀粉含量符合加工业要求的淀粉。在本文中,直链淀粉含量增加的淀粉和直链淀粉含量降低的淀粉都是有利的,因为它们可各自特别适用于特别的用途。

此外,可使用所谓的“激活标签”方法(见例如,Walden等,植物杂志(Plant J.)(1991),281-288;Walden等,植物分子生物学(Plant Mol.Biol.)26(1994),1521-1528)。所述方法基于通过增强子元件如花椰菜花叶病毒35S RNA启动子的增强子或章鱼碱合成酶增强子激活内源启动子。
但在一个优选实施方案中,遗传修饰包括向植物细胞基因组导入编码质体ADP/ATP转运蛋白的外源核酸分子。
因此,术语“转基因”是指本发明的植物细胞包含至少一个在基因组中稳定整合的编码质体ADP/ATP转运蛋白的外源核酸分子,优选核酸分子。
优选地,术语“外源核酸分子”是指编码具有质体ADP/ATP转运蛋白生物活性的蛋白质的核酸分子,该核酸分子在相应植物细胞中不是天然存在的,或在植物细胞中不是天然以该精确的空间次序存在,或者所述核酸分子定位于植物细胞基因组中非天然存在的一个位置。优选地,外源核酸分子是包含各种元件的重组分子,而且其组合或特定空间排列在植物细胞中不是天然存在的。本发明的转基因植物细胞含有至少一个编码具有质体ADP/ATP转运蛋白生物活性的蛋白质的外源核酸分子,其中所述核酸分子优选与确保在植物细胞中转录的调节DNA元件特别是启动子相连。
原则上,外源核酸分子可以是编码表达后定位于质体内膜上的ADP/ATP转运蛋白的任何核酸分子。在本文中,质体ADP/ATP转运蛋白是催化转运ATP进入质体和ADP排出质体的蛋白质。这种核酸分子是已知的,来自例如拟南芥(Kampfenkel等,FEBS lett.374(1995),351-355;Genebank Acc.No.X94626和Acc.No.Z49227)或马铃薯(Genebank Acc.No.Y10821)。利用所述已知核酸分子本领域技术人员可根据标准方法如异源筛选从其它生物特别是植物分离相应序列。特别地,也可使用编码ADP/ATP转运蛋白并与确保在质体内膜上定位的导向序列连接的非植物核酸分子。在本文中,例如已知ADP/ATP转运蛋白来自普氏立克次氏体(Williamson等,基因(Gene)80(1989),269-278)和砂眼衣原体。
在一个优选实施方案中,外源核酸分子编码Arabidopsis thaliana的质体ADP/ATP转运蛋白,特别是Kampfenkel等(1995,loc.cit.)所述的蛋白质AATP1。
本发明的细胞可与天然存在的植物细胞区分开来,特别因为它们含有所述细胞中非天然存在的外源核酸分子,或所述分子整合在细胞基因组中非天然存在的位置,即处于另一种基因组环境中。此外,本发明所述转基因植物细胞可与天然存在的植物细胞区分开来,是因为它们含有至少一个拷贝的在其基因组中稳定整合的外源核酸分子,可选择地还有在细胞中天然存在所述分子的拷贝。如果导入细胞的核酸分子是细胞中天然存在分子的额外拷贝,则本发明的植物细胞可与天然存在的植物细胞区分开来,特别是因为所述额外拷贝位于其非天然存在的基因组位置。例如这可通过Southern印迹分析测定。
本发明植物细胞还可优选通过至少一种下列特征与天然存在的植物细胞区分开:如果核酸分子是植物细胞异源的,则转基因植物细胞显示导入核酸分子的转录本,这可用例如Northern印迹分析检测。优选地,本发明的植物细胞含有导入核酸分子编码的蛋白质。这可通过免疫方法特别是Western印迹分析来检测。
如果核酸分子是植物细胞同源的,则本发明的细胞可通过例如导入的外源核酸分子的额外表达与天然存在细胞区分开。优选地,转基因植物细胞含有外源核酸分子的更多转录本。这可通过例如Northern印迹分析检测。
术语“遗传修饰”是指通过导入外源核酸分子使植物细胞在其遗传信息上被修饰,而且外源核酸分子的存在或表达导致表型改变。在本文中,表型改变优选指细胞一或多项功能的可测量的改变。例如由于导入的外源核酸分子的存在或表达,本发明的遗传修饰植物细胞显示质体ADP/ATP转运蛋白活性增加。
在本发明中,术语“活性增加”是指细胞中质体ADP/ATP转运蛋白基因表达增加、质体ADP/ATP转运蛋白蛋白质数量增加和/或质体ADP/ATP转运蛋白活性增加。
表达增加可通过例如由Northern印迹分析测量编码ADP/ATP转运蛋白转录本的量来检测。在本文中,增加优选指与相应非遗传修饰的细胞相比转录本的量增加至少10%、优选至少20%、更优选至少50%、特别优选至少75%。ADP/ATP转运蛋白量的增加可通过例如Western印迹分析来测定。在本文中,增加优选指与相应非遗传修饰的细胞相比ADP/ATP转运蛋白的量增加至少10%、优选至少20%、更优选至少50%、特别优选至少75%。
质体ADP/ATP转运蛋白活性可通过例如从相应组织分离质体并采用硅油过滤法测定ATP转入的Vmax-值来测定。各种类型质体的纯化参见Neuhaus等(生物化学杂志(Biochem.J.)296(1993),395-401)。硅油过滤法参见Quick等(植物生理学(Plant Physiol.)109(1995),113-121)。
令人惊奇地发现,对含有质体ADP/ATP转运蛋白活性增加的植物细胞的植物,内含物质和/或生物量的产量与相应未修饰野生型植物相比增加了。例如,发现与未修饰的野生型植物相比根据本发明的植物的油含量和/或淀粉含量增加了和/或这些淀粉的直链淀粉含量也增加了。
在本发明中,术语“野生型植物”是指用作产生所述植物的起始材料的植物,即除了导入的遗传修饰外其遗传信息与本发明植物遗传信息相同的植物。
术语“产量增加”是指与未修饰的野生型植物的植物细胞相比本发明植物细胞中内含物质部分优选淀粉或油增加至少10%、优选至少20%、更优选至少30%、最优选至少40%。
术语“淀粉含量增加”是指与未修饰的野生型植物的植物细胞相比根据本发明的植物细胞中淀粉部分增加至少10%、优选至少20%、更优选至少30%、最优选至少40%。淀粉部分的测定根据附带的实施例所述方法进行。
按所附实施例描述的方法测定淀粉部分。
术语“直链淀粉含量增加”是指与未修饰的野生型植物的植物细胞相比本发明植物细胞所合成淀粉的直链淀粉含量增加至少10%、优选至少20%、更优选至少30%、最优选至少40%。直链淀粉含量通过所附实施例所述方法测定。
如上所述,质体ADP/ATP转运蛋白是定位于质体内膜上(Heldt等,FEBS Lett.5(1969),11-14;Pozueta-Romero等,美国国家科学院院刊(Proc.Nat.Acad.Sci.USA)88(1991),5769-5773;Neuhaus,植物生理学101(1993)573-578;Schünemann等,植物生理学103(1993),131-137)催化转运ATP进入质体和ADP排出质体的转运蛋白质。因此,质体ADP/ATP转运蛋白为质体基质提供胞质ATP。
Kampfenkel等(FEBS Lett.374(1995),351-355)最先从拟南芥(Neuhaus等,植物杂志11(1997),73-82)分离了编码ADP/ATP转运蛋白(AATP1)的cDNA,该cDNA显示与革兰氏阴性细菌普氏立克次氏体的ADP/ATP转运蛋白有很高的相似性(66.2%相似)。A.thaliana的AATP1-cDNA编码由589个氨基酸组成的强烈疏水蛋白质,它显示12个潜在的跨膜螺旋(Kampfenkel等FEBS Lett.374(1995),351-355)。所述cDNA可在面包酵母和大肠杆菌中功能性表达。提取该蛋白质并在蛋白脂质体中重建之后,可测到ATP转运速率的增加(Neuhaus等,植物杂志11(1997),73-82)。利用抗拟南芥AATP1肽片段的抗体,可显示ADP/ATP转运蛋白AATP1位于叶绿体内被膜中(Neuhaus等,植物杂志11(1997),73-82)。
迄今,质体ADP/ATP转运蛋白对植物代谢的功能未能彻底阐明。已考虑了各种功能,如给质体基质供应胞质ATP可能对蛋白质转入质体内、氨基酸生物合成、脂肪酸代谢或淀粉代谢有影响(Flügge和Hinz,Eur.J.Biochem.160(1986),563-570;Tetlow等,Planta 194(1994),454-460;Hill和Smith,Planta 185(1991),91-96;Kleppinger-Sparace等,植物生理学98(1992),723-727)。
然而,完全令人惊奇的是,质体ADP/ATP转运蛋白活性的增加导致在相应转基因植物中淀粉含量增加。同样令人惊奇的是,发现质体ADP/ATP转运蛋白活性增加对所产生淀粉的分子组成有影响。例如,与未修饰的马铃薯植物块茎的淀粉相比,根据本发明的马铃薯植物块茎的淀粉显示直链淀粉增加。
迄今为止,一直假定淀粉的分子特性只是由淀粉合成酶如分支酶(E.C.2.4.1.18)、淀粉合成酶(E.C.2.4.1.21)和ADP-葡萄糖焦磷酸化酶(E.C.2.7.7.27)的相互作用决定。然而,完全令人吃惊的是,质体转运蛋白的表达对淀粉的结构有影响。
本发明的植物细胞可来自任何植物物种,即既包括单子叶植物也包括双子叶植物。优选地,植物细胞来自农业上的作物,即人类为了营养或技术特别是工业目的而栽培的植物。通常优选来自合成油和/或淀粉或者贮藏油和/或淀粉的植物的植物细胞。因此,本发明优选涉及合成淀粉或贮藏淀粉的植物的植物细胞,所述植物例如谷类(黑麦、大麦、燕麦、小麦、小米、西米等)、水稻、豌豆、玉米、medullar pea、木薯、马铃薯、油菜、大豆、大麻、亚麻、向日葵或蔬菜(番茄、菊苣、黄瓜、生菜等)。优选马铃薯、向日葵、大豆、水稻的植物细胞。特别优选玉米、小麦、油菜和水稻的植物细胞。
另外,本发明主题是含有上述转基因植物细胞的转基因植物。所述植物可通过例如从本发明的植物细胞再生产生。原则上,转基因植物可以是任何植物,包括单子叶植物和双子叶植物。优选地,它们是有用的植物,即人类为了营养或技术特别是工业目的而栽培的植物。这些植物可以是合成油和/或淀粉或者贮藏油和/或淀粉的植物。本发明优选涉及诸如谷类(黑麦、大麦、燕麦、小麦、小米、西米等)、水稻、豌豆、玉米、medullar pea、木薯、马铃薯、油菜、大豆、大麻、亚麻、向日葵或蔬菜(番茄、菊苣、黄瓜、生菜等)的植物。优选马铃薯、向日葵、大豆、水稻。特别优选玉米、小麦、油菜和水稻。
如上所述,令人吃惊地发现,与野生型植物相比在含有质体ADP/ATP转运蛋白活性增加的本发明植物细胞的淀粉贮藏植物中淀粉含量增加了,和/或与相应未修饰野生型植物相比这些淀粉的直链淀粉含量也增加了。
因此,在一个优选实施方案中,本发明还涉及含有本发明植物细胞的淀粉贮藏植物,其与未修饰的野生型植物相比显示淀粉含量增加和/或与相应的未修饰野生型植物相比所述淀粉的直链淀粉含量增加。
术语“淀粉贮藏植物”包括所有具有淀粉贮藏组织的植物如玉米、小麦、水稻、马铃薯、黑麦、大麦、燕麦。优选水稻、大麦和马铃薯。特别优选玉米和小麦。

与其它合成直链淀粉含量增加的淀粉的植物如玉米的amylose-extender和dull突变体相比,根据本发明的所述植物具有如下优点:除了直链淀粉含量增加外,它们显示淀粉含量不降低甚至增加。
另外,本发明的主题是含有本发明植物细胞的油贮藏植物,其与未修饰的野生型植物细胞相比油含量增加,优选在贮藏油的组织中油含量增加。
术语“油贮藏植物”包括所有能贮藏油的植物如油菜、芸苔、大豆、向日葵、玉米、花生、小麦、棉花、油棕、橄榄树和鳄梨。优选玉米、小麦和大豆。特别优选油菜和芸苔。

为了降低本发明细胞中的ADP/ATP转运蛋白活性,优选表达反义RNA。
为了该表达,可使用含有编码ADP/ATP转运蛋白的全长序列的DNA分子,包括可能存在的侧翼序列,或使用仅含有编码序列一部分的DNA分子,其中这些部分必须足够长以在细胞中导致反义效应。通常,序列长度可达最小15bp长,优选100-500bp长,特别地,为了有效反义抑制,序列长度超过500bp。通常采用比5000bp短的DNA分子,优选小于2500bp。
也可使用与植物细胞中内源存在的编码质体ADP/ATP转运蛋白的序列高度同源的DNA序列。最小同源性应大于约65%。优选采用同源性在95%到100%之间的序列。

RNA-DNA寡核苷酸的一部分DNA成分与内源ADP/ATP转运蛋白的核酸序列同源,但与内源ADP/ATP转运蛋白核酸序列相比显示突变或含有包含于同源区中的异源区。通过RNA-DNA寡核苷酸的同源区与内源核酸分子的碱基配对,然后同源重组,RNA-DNA寡核苷酸的DNA成分中包含的突变或异源区可转入植物细胞的基因组中。这会导致质体ADP/ATP转运蛋白活性降低。

另外,本发明的主题是含有上述转基因植物细胞的转基因植物。所述植物可从本发明的植物细胞再生产生。该转基因植物原则上可是任何植物物种的植物,即包括单子叶植物和双子叶植物。优选地,它们是来自农业上的作物的植物细胞,即来自人类为了营养或技术特别是工业目的而栽培的植物。优选地,它们是合成淀粉或贮藏淀粉的植物,例如谷类(黑麦、大麦、燕麦、小麦、小米、西米等)、水稻、豌豆、玉米、medullarpea、木薯、马铃薯、油菜、大豆、大麻、亚麻、向日葵、豇豆和竹芋。特别优选马铃薯。
与野生型植物的淀粉相比,本发明所述植物合成的淀粉显示直链淀粉含量降低。术语“直链淀粉含量降低”和“野生型植物”定义见上。
而且,本发明还涉及产生转基因植物的方法,与相应野生型植物的淀粉相比所述转基因植物的淀粉显示直链淀粉含量降低,其中
(a)通过导入外源核酸分子对植物细胞进行遗传修饰,而且该遗传修饰导致植物细胞中内源存在的质体ADP/ATP转运蛋白活性降低;和
(b)从根据步骤(a)产生的细胞再生植物;和选择性地
(c)从根据(b)产生的植物产生后代植物。
对于根据步骤(a)导入植物细胞的修饰,上述关于本发明的植物细胞和植物的讨论同样应用如此。
根据步骤(b)的植物再生可根据本领域技术人员已知的方法进行。
根据本发明方法步骤(c)的后代植物的产生可通过例如营养繁殖(如通过插枝、块茎、或愈伤组织培养和全株再生)或有性繁殖来实现。优选地,有性繁殖以控制的方式进行,即选择有特定性状的植物相互杂交和繁殖。
在一个优选实施方案中,本发明的方法用于产生转基因马铃薯植物。
本发明还涉及可通过本发明的方法获得的植物。
本发明还涉及根据本发明的植物以及根据本发明方法产生的含有本发明遗传修饰细胞的转基因植物的繁殖材料。在本文中,术语繁殖材料包括那些适于通过营养或有性方式产生后代的植物的组成部分。例如,适于营养繁殖的有插枝、愈伤组织培养物、根状茎或块茎。其它繁殖材料包括例如果实、种子、幼苗、原生质体、细胞培养物等。优选地,繁殖材料是种子,特别优选块茎。
另外,本发明还涉及编码质体ADP/ATP转运蛋白的核酸分子、其互补序列或所述分子的部分在产生与野生型植物的淀粉相比合成直链淀粉含量减少的淀粉的植物中的应用。优选采用与显示ADP/ATP转运蛋白活性增加的本发明细胞相关的上述核酸分子。
已知多种技术可用于在植物宿主细胞中导入DNA。这些技术包括采用根癌农杆菌(Agrobacterium tumefaciens)或毛根农杆菌(Agrobacterium rhizogenes)作为转化剂用T-DNA转化植物细胞、原生质体融合、注射、DNA电穿孔、通过生物轰击途径导入DNA或其它可能的方法。
农杆菌属(Agrobacterium)介导的植物细胞转化的使用已有详细分析,并在下列文献中有充分的描述:EP 120516;Hoekema:The BinaryPlant Vector System Offsetdrukkerij Kanters B.V.,Alblasserdam(1985),第V章;Fraley等,Crit.Rev.Plant Sci.4,1-46和An等,EMBO J.4(1985),277-287。关于马铃薯的转化,见例如Rocha-Sosa等,EMBO J.8(1989),29-33。
采用基于农杆菌属的载体转化单子叶植物也有描述(Chan等,植物分子生物学22(1993),491-506;Hiei等,植物杂志6(1994)271-282;Deng等,中国科学(Science in China)33(1990),28-34;Wilmink等,植物细胞报道(Plant Cell Reports)11(1992),76-80;May等,Bio/Technology13(1995),486-492;Connor和Domisse,Int.J.Plant Sci.153(1992),550-555;Ritchie等,转基因研究(Transgenic Res.)2(1993),252-265)。单子叶植物转化的另一个系统是通过生物轰击途径转化(Wan和Lemaux,植物生理学104(1994),37-48;Vasil等,Bio/Technology 11(1993),1553-1558;Ritala等,植物分子生物学24(1994),317-325;Spencer等,Theor.Appl.Genet.79(1990),625-631)、原生质体转化、部分透化细胞的电穿孔、通过玻璃纤维导入DNA。特别地,玉米的转化在文献中有多次描述(见例如WO 95/06128,EP 0513849,EO 0465875,EP 292435;Fromm等,生物技术(Biotechnology)8(1990),833-844;Gordon-Kamm等,植物细胞(PlantCell)2(1990),603-618;Koziel等,生物技术11(1993),194-200;Moroc等,Theor.Appl.Genet.80(1990),721-726)。
其它谷类的成功转化也有描述,例如大麦(Wan和Lemaux,loc.cit.,Ritala等,loc.cit.,Krens等,自然(Nature)296(1982),72-74)和小麦(Nehra等,植物杂志5(1994),285-297)。
为了在植物细胞中表达编码ADP/ATP转运蛋白的有义或反义方向的核酸分子,所述核酸分子优选与确保在植物细胞中转录的调节DNA元件相连。特别地,所述元件包括启动子。通常任何在植物中有活性的启动子都是适合的。
可选择启动子以使表达组成型发生,或仅在特定组织、植物发育的特定时间点或外部因素确定的时间点发生。对于植物和核酸分子,启动子均可是同源或异源的。
适合的启动子是例如用于组成型表达的花椰菜花叶病毒35S RNA启动子和玉米泛素启动子、用于马铃薯块茎特异表达的patatin基因启动子B33(Rocha-Sosa等,EMBO J.8(1989),23-29)和保证只在光合活跃组织中表达的启动子如ST-LS1启动子(Stockhaus等,美国国家科学院院刊84(1987),7943-7947;Stockhaus等,EMBO J.8(1989),2445-2451)或用于胚乳特异表达的小麦HMG启动子、USP启动子、菜豆蛋白启动子、玉米的玉米蛋白基因启动子(Pedersen等,细胞(Cell)29(1982),1015-1026;Quatroccio等,植物分子生物学15(1990),81-93)、谷蛋白启动子(Leisy等,植物分子生物学14(1990),41-50;Zheng等,植物杂志4(1993),357-366;Yoshihara等,FEBS Lett.383(1996),213-218)或shrunken-1启动子(Werr等,EMBO J.4(1985),1373-1380)。然而,也可使用仅在外部因素确定的时间点激活的启动子(见例如WO 9307279)。在本文中,特别感兴趣的是允许简单诱导的热激蛋白启动子。而且,可采用种子特异的启动子如Vicia faba的保证在Vicia faba和其它植物种子中特异性表达的USP启动子(Fiedler等,植物分子生物学22(1993),669-679;Bumlein等,Mol.Gen.Genet.225(1991),459-467)。
上述胚乳特异启动子的实施方案特别适于在胚乳中增加淀粉含量。与此相反,感兴趣的是使用胚特异启动子增加油的含量,因为油主要贮藏在胚中。
因此,优选地,根据本发明采用保证在胚或种子中表达的启动子。在本发明一个优选实施方案中,启动子是玉米的球蛋白-1(glb1)启动子(Styer和Cantliffe,植物生理学6(1984),196-200)。在本发明另一个实施方案中,胚特异启动子来自植物,优选Cuphea lanceolata、Brassica rapa或Brassica napus。特别优选的是启动子pCIFatB3和pCIFatB4(WO95/07357)。它们分别是基因CIFatB3和CIFatB4的启动子,它们已经在转基因油菜中成功地用于中等链脂肪酸的生物合成,因此对于本发明的方案具有适合的表达窗口。
在另一个优选实施方案中,采用pCIGPDH启动子(WO 95/06733)、napin(参见例如Kridi,种子科学研究(Seed Sci.Res.)1(1991),209-219;Ellerstrom等,植物分子生物学32(1996),1019-1027;Stalberg等,Planta199(1996),515-519)或oleosin启动子(见例如Keddie,植物分子生物学(1994)327-340;Plant等,植物分子生物学25(1994),193-205)。
而且,可存在一个终止序列用作转录的正确终止和给转录本添加被认为能稳定转录本的poly-A尾。所述元件见文献(见例如Gielen等,EMBOJ.8(1989),23-29),并可按需要互换。
本发明的转基因植物细胞和植物合成的淀粉与野生型植物合成的淀粉相比在理化上特别是在直链淀粉/支链淀粉比率上改变,优选由于质体ADP/ATP转运蛋白活性增加或降低引起。特别地,与野生型淀粉相比,可针对所述淀粉胶的粘性和/或凝胶形成特性来改变所述淀粉。
因此,本发明涉及产生修饰淀粉的方法,包括从上述植物之一和/或所述植物淀粉贮藏部分提取淀粉的步骤。优选地,所述方法还包括提取淀粉之前收获栽培的植物和/或所述植物的淀粉贮藏部分,特别优选地,还包括收获前栽培本发明植物的步骤。从植物或植物贮藏淀粉的部分提取淀粉的方法是本领域技术人员已知的。而且,从各种淀粉贮藏植物提取淀粉的方法都有描述,见如“淀粉:化学和技术(Starch:Chemistry andTechnology)”(编者:Whistler,BeMiller和Paschall(1994),第2版,Academic Press Inc.London Ltd;ISBN 0-12-746270-8;见例如第XII章,第412-468页:玉米和高粱淀粉:生产(maize and sorghum starch:production);Watson著;第XIII章,第469-479页:木薯、竹芋和西米的淀粉:生产(starches from tapioca,arrowroot and sago:production);Corbishley和Miller著;第XIV章,第491-506页:小麦的淀粉:生产、修饰和用途(starch from wheat:production,modification and uses);Knight和Oson著;第XVI章,第507-528页:水稻的淀粉:生产和用途(starch from rice:production and uses);Rohmer和Klem著;玉米的淀粉(starch from maize):Eckhoff等,谷类化学(Cereal Chem.)73(1996)54-57,按工业标准从玉米提取淀粉通常通过所谓“湿磨”来实现)。通常采用的用于从植物材料提取淀粉之方法的器具是分离器、倾析器、水力旋流器、喷射干燥器和流化床干燥器。
另外,本发明的主题是可从本发明的转基因植物细胞、植物和繁殖材料获得的淀粉和可通过上述本发明方法获得的淀粉。
本发明的淀粉可根据本领域已知的方法修饰并以未修饰或修饰形式适于食品或非食品工业中的多种用途。
原则上,可能的用途可分为两个领域。一个领域包括淀粉的水解产物,主要是通过酶或化学方法获得的葡萄糖和葡聚糖结构单元。它们用作供进一步化学修饰和加工如发酵的初始材料。为了降低成本,水解方法的简便和便宜可能是重要的。目前,该方法基本是使用淀粉葡糖苷酶酶解。通过减少酶的使用将有可能节约成本。这可通过改变淀粉的结构来实现,如谷粒表面扩大、低分支程度引起的更容易消化或限制所用酶接近的空间结构。另一个领域即由于其聚合结构淀粉与所谓的天然淀粉一样使用,可再进一步划分成两个应用领域:
1.在食品中的应用
淀粉是各种食品的典型添加物,其中它基本用作水性粘合添加物和/或引起粘性或凝胶形成提高。重要特性是流动和吸附性能、溶胀和糊化温度、粘性和增稠性、淀粉的溶解度、透明度和糊剂结构、热、剪切和酸抗性、退化倾向、膜形成能力、耐冻/融性、可消化性以及与例如无机或有机离子形成复合物的能力。
2.在非食品中的应用
另一个主要应用领域是在各种生产过程中使用淀粉作为辅助剂或在技术产品中用作添加剂。使用淀粉作为辅助剂的主要应用领域首先是纸和纸板工业。在该领域淀粉主要用于滞留(滞留固体)、对填充物和细微颗粒大小分级、使物质凝固和脱水。此外,还利用淀粉在稠度、硬度、声音、手感、光泽、平滑、撕裂强度以及表面的优点。
2.1纸和纸板工业
在纸生产过程中,可区分出4个应用领域,即表面、涂层、备料和喷雾。表面处理对淀粉的要求主要是高亮度、相应粘性、高粘性稳定性、良好的膜形成以及低产尘。当用于固体内容物涂层时,相应粘性、高结合能力以及高色素亲合力有重要作用。作为备料添加剂,在纸浆中快速、均一、不损耗的分散,高机械稳定性和完全滞留是重要的。当使用淀粉喷雾时,固体的相应含量、高粘性和高结合能力也是重要的。
2.2粘合剂工业
一个主要应用领域是例如粘合剂工业,其中该应用领域再分为4个领域:用作纯淀粉胶、在以特殊化学制品制备的淀粉胶中的应用、使用淀粉作为合成树脂和聚合分散相的添加剂以及使用淀粉作为合成粘合剂的增充剂。所有基于淀粉的粘合剂的90%用于生产瓦楞纸板、各种纸袋、纸和铝的复合材料、纸盒和信封、邮票等的湿胶。
2.3纺织品和织物保养品
作为辅助剂和添加剂的另一个用途是生产纺织品和织物保养品。在纺织品工业中,可区分下列4个应用领域:将淀粉用作上浆剂,即作为平滑处理和加强去毛刺性能的辅助剂以对抗纺织中牵引的张力以及提高纺织过程中的耐磨强度,用作主要在质量降低性预处理如漂白、染色等之后纺织品的整理剂,用作生产染料糊剂的增稠剂以避免染料扩散,和用作缝纫纱线束缚剂的添加剂。
2.4建筑业
而且,淀粉可用作建筑材料的添加剂。一个例子是石膏板板材的生产,其中在稀石膏浆中混合的淀粉与水一起糊化、在石膏板表面扩散因而使纸板与该板粘合。其它应用领域是将淀粉与泥灰和矿物纤维混合。在混合好的混凝土中,淀粉可用于减缓硬化过程。
2.5土壤稳定化
另外,淀粉在生产土壤稳定剂中是有利的,可用于在人工侵拢土壤时临时性保护土壤颗粒免于失水。根据本领域现有知识,由淀粉和聚合物乳剂组成的混合产品可认为具有与现有产品相同的降低冲蚀和结壳的作用;然而,它们便宜得多。
2.6在植物保护剂和肥料中的应用
另一个应用领域是淀粉用于植物保护剂中以改变这些制品的特定性质。例如,可使用淀粉改善植物保护剂和肥料的可润湿性以便定量释放有效成分,将液体、挥发性和/或不良气味的有效成分转换成微晶、稳定、可变形的物质,混合不相容组合物和由于分解减缓而延长作用持续时间。
2.7药品、医疗和化妆品工业
淀粉也可用于药品、医疗和化妆品工业领域。在制药工业中,淀粉可用作片剂粘合剂或胶囊粘合剂的稀释。此外,淀粉适于用作片剂的崩解剂,因为吞服后它吸收液体并在短时间后溶胀以致活性成分释放。由于性质上的原因,其它应用领域是医疗润滑和疗伤扑粉。在化妆品领域,例如淀粉可用作粉末添加物如香水和水杨酸的载体。淀粉的一个相当广泛的应用领域是牙膏。
2.8淀粉作为煤和煤球的添加物
淀粉也可用作煤和煤球的添加物。通过加入淀粉,煤可定量地结块和/或制成高质量的煤球,从而防止煤球过早分解。烧烤用煤添加了4%-6%淀粉,高热煤含有约0.1%-0.5%淀粉。而且,淀粉适于作为粘结剂,因为在煤和煤球中加入淀粉可显著减低有害物质的释放。
2.9矿石和煤浆的加工
另外,淀粉可用作矿石和煤浆的加工过程中的凝聚剂。
2.10铸造材料的添加物
另一个应用领域是用作加工铸造材料的添加物。对于各种铸造工艺,需要从沙石混合粘合剂产生的核心。目前最常用的粘合剂是混合了改性淀粉、主要是溶胀淀粉的膨润土。加入淀粉的目的是增加流动阻力以及改善结合强度。而且溶胀淀粉可符合更多的生产工艺的必要条件如冷水中的可分散性、再水合能力、与沙石的良好混合性和高度水结合能力。
2.11橡胶工业
在橡胶工业中淀粉可用于改善技术和光学质量。其原因是改善表面光泽、手感和外观。为此,在冷固化之前将淀粉分散在粘性的橡胶物质的橡胶处理表面。也可用于改善橡胶的可印刷性。
2.12皮革替代品的生产
改性淀粉的另一个应用领域是生产皮革替代品。
2.13合成聚合物中的淀粉
在塑料市场存在下列应用领域:淀粉产品在工艺中的应用(淀粉只是填充剂,合成聚合物和淀粉之间没有直接键合),或淀粉产物在聚合物的生产中应用(淀粉和聚合物形成稳定的键)。
使用淀粉作为纯填充剂不能与其它物质如滑石竞争。当特定淀粉性质变得有效因而终产物的性质明显改变时这种情况就不同了。一个例子是淀粉产品在热塑性材料如聚乙烯的加工过程中的应用。由此,淀粉和合成聚合物通过共作用以1∶1的比例结合形成“母材料”,由此使用制成粒状的聚乙烯采用常规技术可生产各种产品。淀粉在聚乙烯膜中的整合可造成增加空心体中物质通透性,改善水蒸气通透性,改善抗静电性能,改善抗阻塞性能以及改善水染料的可印刷性。
另一种可能是淀粉在聚氨酯泡沫胶中的应用。由于淀粉衍生物的适应性以及加工技术的优化,有可能特定控制合成聚合物和淀粉羟基之间的反应。结果是由于淀粉的使用聚氨酯膜具有下列特性:降低热膨胀系数、减小收缩性、改善压力/张力性能、增加水蒸气通透性而不改变水的吸收、降低可燃性和破裂密度、可燃部分无液滴形成、无卤化物和减缓老化。目前仍然存在的缺点是压力和冲击强度降低。
开发膜产品不是唯一选择。也可利用含量为50%以上的淀粉生产固体塑料产品如罐、盆、碗等。而且,淀粉/聚合物混合物具有更容易生物降解的优点。
另外,由于它们极易结合水,淀粉接枝聚合物具有极大的重要性。这些产品具有淀粉骨架和根据自由基链反应机制在其上接枝的合成单体的侧链。目前已有的淀粉接枝聚合物的特征是结合和保持水的能力改善,达到高粘度时每g淀粉1000g水。这些超吸收剂主要用于卫生领域,如尿布和床单等产品,以及农业部门如种子小球。
一方面,重组DNA技术修饰的新淀粉的应用取决于结构、含水量、蛋白含量、脂含量、纤维含量、灰/磷酸含量、直链淀粉/支链淀粉比、相对摩尔质量的分布、支化度、颗粒大小和形状以及结晶性,另一方面取决于导致下列性质的特性:流动和吸附性能、糊化温度、粘性、增稠性能、溶解度、糊剂结构、透明性、热、剪切和酸的耐性、退化倾向、凝胶形成的能力、耐冻/融性、形成复合物的能力、碘的结合、膜形成、粘合强度、酶稳定性、可消化性和反应性。
通过转基因植物遗传操作产生修饰的淀粉可修饰该植物淀粉的特性,从而不再需要通过多种化学或物理方法进行进一步修饰。另一方面,用重组DNA技术修饰的淀粉可进行进一步化学修饰,这将为某些上述应用领域产生进一步的质量改善。这些化学修饰是基本上是已知的。这些修饰特别是下列方法:
-热处理
-酸处理
-氧化和
-酯化作用
这些修饰导致形成磷酸、硝酸、硫酸、黄原酸、乙酸和柠檬酸淀粉。其它有机酸也可用于酯化:
-形成淀粉醚
淀粉烷基醚、O-烯丙基醚、羟烷基醚、O-羧甲基醚、含氮淀粉醚、含磷淀粉醚和含硫淀粉醚。
-形成分支淀粉
-形成淀粉接枝聚合物

下列实施例进一步阐述本发明。
实施例1
细菌表达载体pJT118的构建和大肠杆菌转化
拟南芥的AATP2蛋白在N端融合含有10个氨基酸的“组氨酸标签”。为此,通过PCR方法分离编码拟南芥的全长AATP2蛋白的cDNA。下列寡核苷酸用作有义引物,此外它还含有一个XhoI限制性位点:cgtgagagatagagagctcgagggtctgattcaaacc(SEQ ID NO:1);包含碱基对66-102。
用另外携带一个BamHI限制性位点的寡核苷酸作为反义引物:gatacaacaggaatcctggatgaagc(SEQ ID NO:2);包含碱基对1863-1835。获得的PCR产物用琼脂糖凝胶法纯化,用限制性酶XhoI/BamHI切割,导入质粒pET16b(Novagene,Heidelberg,德国)的阅读框内。这导致在编码全长拟南芥的AATP2蛋白质的cDNA的N-端显示一个10个氨基酸的组氨酸标签(His-AATP2)。所述载体称为pJT118。PCR产物的序列通过测定两条核苷酸链的序列来确定(Eurogentec)。大肠杆菌C43的转化(Miroux和Walker,分子生物学杂志260(1996),289-298)按照标准方法进行。大肠杆菌C43允许动物(Miroux和Walker,loc.cit.)和植物(Tjaden等,J.Biol.Chem.(1998))膜蛋白的异源表达。
载体pJT118转化所述菌株后,用放射性标记的ADP和ATP进行摄取研究。可通过这些研究说明His-AATP2可在大肠杆菌C43的细胞质膜中功能性表达。这说明AATP2实际上编码ADP/ATP转运蛋白。N末端组氨酸标签的存在导致大肠杆菌中拟南芥来源的AATP2的转运活性较无N末端组氨酸标签的AATP2的转运活性高(2x-3x)。
实施例2
构建质粒pJT31并将该质粒导入马铃薯植物的基因组中
为了构建植物转化载体,将拟南芥的AATP1-cDNA(Kampfenkel等,FEBS Letters 374(1995),351-355)的长2230bp的EcoRV/BamHI片段连入用SmaI/EeoRV和BamHI切割的载体pBinAR中(Hfgen和Willmitzer,植物科学66(1990),221-230)。通过插入该cDNA片段,形成由如下片段A、B和C构成的表达盒(pJT31)(见图1):

质粒pJT31的大小约14.2kb。
按Rocha-Sosa等(EMBO J.8(1989),23-29)所述利用农杆菌将该质粒导入马铃薯植物。转化结果,转基因马铃薯显示质体ADP/ATP转运蛋白mRNA的增加。这用Northern印迹分析检测(见图6)。按标准方法从马铃薯植物的叶和块茎组织分离RNA,将50ug RNA在琼脂糖凝胶(1.5%琼脂糖,1x MEN缓冲液,16.6%甲醛)上分离。电泳后通过毛细管印迹法用20xSSC将RNA转移至尼龙膜Hybond N(Amersham,UK)上。通过UV照射将RNA固定在膜上。将膜在磷酸杂交缓冲液中预杂交2小时(Sambrook等,loc.cit.),然后加入放射性标记探针杂交10小时。
实施例3
构建质粒pJT32并将该质粒导入马铃薯植物基因组
为构建植物转化载体,将S.tuberosum AATP1-cDNA编码区(Genbank Y10821)1265bp长的BamHI/NdeI片段连入用Smal/NdeI和BamHI切割的载体pBinAR中(Hfgen和Willmitzer,植物科学66(1990),221-230)。
通过插入该cDNA片段,形成由如下片段A、B和C构成的表达盒(见图2):
片段A(540bp)含有花椰菜花叶病毒的35S启动子。
片段B含有S.tuberosum ADP/ATP转运蛋白(AATPI S.t.)的一个长1265bp的区域。该区域以反义方向与pBinAR的35S启动子融合。
片段C(215bp)含有根癌农杆菌章鱼碱合成酶基因的聚腺苷酸化信号。
质粒pJT32的大小约13.3kb。

分析转基因马铃薯植物的淀粉、直链淀粉和糖含量
可溶性糖含量的测量按Lowry和Passonneau在“灵活的酶法分析系统(A Flexible System of Enzymatic Analysis)”,Academic Press,NewYork,USA(1972)中所述进行。淀粉含量的测定按Batz等(植物生理学100(1992),184-190)所述进行。

实施例5
产生表达盒并转化油菜植物
pBluescript衍生质粒的表达盒pTE200(Short等,核酸研究(Nucl.Acid Res.)16,(1988),7583-7600)携带Cuphea lanceolata的硫酯酶基因CIFatB4(GenBank号:AJ 131741)的启动子和终止子序列和用于插入各种有用基因的适当多接头序列。在可变识别区具有不相容核苷酸的外围SfiI识别位点允许将包括有用基因的整个表达盒定向转移至二元质粒载体pMH000-0(pLH9000的进一步发展)(Hausmann和Tpfer,(1999):第9章:″Entwicklung von Plasmid-Vektoren″in Bioengineering fürRapssorten naeh Maβ,D.Brauer,G.Rbbelen和R.Tpfer(编),Vortrge für Planzenzüchtung,第45卷,155-172)的相应限制位点中,并防止受体载体中DNA的重新环化。
为了产生表达盒pTE200,首先从含有C.lanceolata完整CIFatB4基因的基因组克隆CITEg16(W095/07357)分离含有启动子的长约3.3kb的SalI-BbvI片段。为此,切割并修饰启动子3’端的BbvI限制性位点,以使该片段能被SalI和SmaI切割的pBluescript(stratagene)接受。通过切割、用T4聚合酶修饰然后再连接缺失该片段5’端第1211位核苷酸处的内部EcoR限制性位点。
利用聚合酶链式反应和特异寡核苷酸引物从CITEg16模板(WO95/07357)中扩增终止子序列,并通过引物提供多接头限制性位点(MCS)。引物的序列是:5′GAATTCCTGCAGCCCGGGGGATCCACTAGTCTCGAGAAGTGGCTGGGGGCCTTTCC3′(SEQ ID NO:3)=5′-引物:(MCS:EcoRI,PstI,SmaI,BamHI,SpeIl XhoI;CIFatB4终止子:位置35-56)和5′TCTAGAGGCCAAGGCGGCCGCTTCAACGGACTGCAGTGC3′(SEQ ID NO:4)=3′-引物:CIFatB4终止子:位置22-39,MCS:NotI,StyI,SfiI,XbaI。用EcoRI和NotI切割扩增产物并插入pBlueSfi BA(Hausmann和Tpfer,见上)的相应限制性位点。携带启动子的片段用BamHI打开、修饰、然后用SalI切割以通过终止子前面的SalI和HindIII限制性位点将其置于pBlueSfi BA载体中。结果获得表达盒pTE200(见图7)。
为了构建植物转化载体,将Solanum tuberosum的AATP1cDNA(pTMI,Tjaden等,The Plant Journal 16(1998)531-540)长2270bp的EcoRI片段连入用EcoRI打开的载体pTE200中。方向通过限制性消化控制。结果是质粒pTE208(图8)。在后面的步骤中,pTE208的SfiI片段以定向方式插入二元载体pMH000-0的多接头限制性位点中(图9)。结果获得载体pMH 0208。
二元质粒载体pMH000-0从pLH9000(Hausmann和Tpfer,见上)进一步发展而来,具有供植物转化的替代筛选标记。在Asp718修饰至XhoI限制性位点后,将磺胺类药物基因(sul)与二磷酸核酮糖羧化酶小亚基转入质体的信号肽(tp)一起从前体质粒pS001(Reiss等,美国国家科学院院刊93,(1996),3094-3098)分离出来。在质粒经SalI和BamHI修饰后,将XhoI-SalI片段插入pBluescript衍生质粒的胭脂碱合成酶基因(pAnos)终止子前面的XhoI和BamHI限制性位点。在随后的三片段连接中,将获得的tpsul-pAnos片段(XhoI-XbaI)和pRT103pat(Tpfer等,酶学方法(Methods in Enzymol.)217,(1993),66-78)的XhoI-HindIII片段和用XbaI和HindIII打开的质粒pK18(Pridmore,基因56,(1987),309-312)连接。结果,含有来自pRT103pat的CaMV35S rna基因终止子的膦丝菌素乙酰转移酶基因以与tpsul-pAnos单位相反方向放置。将CaMV35S基因双启动子作为pROA93子代(Ott et al.,Mol.Gen.Genet.221,(1990),121-124)的XhoI片段插入抗性介导基因序列之间的XhoI位点以完成所述双选择单位(抗除草剂Basta和磺胺类药物磺胺嘧啶的抗性)。在邻近多接头中进行相应修饰后,获得的双选择盒通过XbaI和HindIII与pLH9000前体质粒(Hausmann和Tpfer,见上)中的卡那霉素盒交换。结果获得二元质粒载体pMH000-0。
油菜品种Drakkar胚轴外植体的转化按照De Block(Plant Physiol.91(1989),694-701)的程序利用携带二元载体pMH0208(ATP/ADP转运蛋白,有义)的农杆菌(株系GV 3101 C58C1 Rifr)进行。在选择营养培养基(磺胺类药物)上再生芽,并种植在温室中直到种子成熟。采用PCR和叶片检测(对glufosinatammonium(Basta)的耐受性),测定哪些植物含有转基因。从所述植物收获各种发育阶段的成熟胚并保存在液氮中。
为了测定油含量,用非侵入近红外光谱法分析转基因油菜系和对照系的成熟种子(见例如Schulz等,J.Near Infrared Spectrosc.6,(1998),A125-A130;Starr等,J.Agric.Sci.104(2),(1985),317-323)。

Claims (10)
1.遗传修饰的转基因植物细胞,其中该遗传修饰是导入外源核酸分子,与来自野生型植物的相应未遗传修饰植物细胞相比所述核酸分子的存在或表达导致质体ADP/ATP转运蛋白活性增加,其中与相应未遗传修饰的植物细胞相比,所述转基因植物细胞显示淀粉含量增加和/或其淀粉中直链淀粉含量增加。
2.根据权利要求1的转基因植物细胞,其中外源核酸分子编码质体ADP/ATP转运蛋白。
3.根据权利要求2的转基因植物细胞,其中核酸分子编码拟南芥的质体ADP/ATP转运蛋白。
4.根据权利要求1-3任一项的转基因植物细胞,其是淀粉贮藏植物的细胞。
5.根据权利要求4的转基因植物细胞,其是玉米、小麦或马铃薯植物的细胞。
6.产生与野生型植物相比显示淀粉含量增加和/或其淀粉中直链淀粉含量增加的转基因植物的方法,其中
(a)通过导入外源核酸分子对植物细胞进行遗传修饰,所述外源核酸分子的存在或表达可导致细胞中质体ADP/ATP转运蛋白活性增加;
(b)从根据步骤(a)产生的细胞再生植物;和
(c)可选择地从根据步骤(b)产生的植物产生后代植物。
7.由权利要求6的方法可以获得的转基因植物的繁殖材料在生成进一步的植物中的应用。
8.编码质体ADP/ATP转运蛋白的核酸分子在产生与野生型植物相比显示淀粉含量增加和/或其淀粉中直链淀粉含量增加的转基因植物中的应用。
9.产生修饰淀粉的方法,包括从根据权利要求1-5之任一项的植物细胞提取淀粉。
10.产生修饰淀粉的方法,包括从由权利要求6的方法可获得的植物提取淀粉。
