Abstract
本发明公开了一种昆虫细胞无血清培养基及其应用,该培养基由氨基酸、维生素、无机盐、微量元素、碳水化合物和其他分子化合物组成,不仅能够满足各种亚型昆虫细胞的培养需求,还能够有效提高AAV的表达量并降低空壳率,且可以维持更长时间更高活率的细胞状态,从而能够提高生产效率;该培养基价格低廉,从而可以降低生产成本,具有广泛的应用前景和价值。

技术领域
本发明涉及细胞培养技术领域,具体为一种昆虫细胞无血清培养基及其应用。
背景技术
昆虫细胞表达系统(杆状病毒表达系统),作为真核细胞系统,具有其独特的生物学特性,它是表达大片段DNA的理想载体,其重组蛋白也具有完整的生物学功能,相对于其他表达系统(如CHO等哺乳动物细胞),表达水平更高,能同时表达多个基因,且较低的免疫原性,不会引起炎症反应,故已应用于多个领域,如基因治疗及疫苗等方面的研究。如何快速、安全、有效且低成本开发适用性预防疫苗,成为一个阶段性的挑战。昆虫细胞表达系统能够快速获得高产的表达产物、对人体安全,并且极其适用于规模化放大生产,无疑是快速、安全且低成本生产新冠预防性疫苗的有效途径之一。
腺相关病毒(adeno-associated virus,AAV)是基因治疗领域应用最为广泛的载体,也是经过FDA认证的,对人体极其安全的生物载体。目前生产AAV的主流工具细胞,包括人源的HEK293和各种亚型的昆虫细胞。以HEK293作为AAV病毒的生产宿主,很难实现低成本的规模化生产,其多质粒瞬转的生产方法,极大的限制了工业化的生产规模(一般在500L以下,多数为200L),造成目前AAV相关疗法成本极高,特别是对于全身性用药的病症或者患者,治疗费用无法承担,临床可及性极差。而以昆虫细胞作为AAV病毒的生产宿主,可以有效解决上述弊端,其工业化放大生产规模可至数千升,且工艺较HEK293稳定可控,极大的压缩了AAV病毒生产成本,扩大了AAV相关疗法或者药物的临床可及性,以造福社会和广大患者。
以昆虫细胞作为AAV生产宿主,一个高表达的细胞系,搭配合适的培养基,对提高AAV病毒产量和方便后期相关质量的鉴定至关重要。而目前培养昆虫细胞的培养基,大多会添加5%甚至更多含量的血清,使其增加了很多的风险,也有一些进口培养基(如Gibco的SF900 III)或者国产培养基,虽然不添加血清,但依然含有少量水解物成分,且价格昂贵。
因此,昆虫细胞无血清培养基及其应用,是本行业目前研究的热点和痛点。
发明内容
针对上述问题,发明人开发了一种无任何动物性来源成分的、无蛋白的、化学成分明确的昆虫细胞无血清培养基,不仅能够满足各种亚型昆虫细胞的培养需求,还能够有效提高AAV的表达量并降低空壳率,且价格低廉。
本发明的目的在于提供一种昆虫细胞无血清培养基及其应用,以解决上述背景技术中提出的问题。

一种昆虫细胞无血清培养基,由氨基酸、维生素、无机盐、微量元素、碳水化合物和其他分子化合物组成;

与现有技术相比,本发明所达到的有益效果是:
1、本发明提供的昆虫细胞无血清培养基,无任何动物性来源成分的、无蛋白成分、化学成分明确。
2、本发明提供的昆虫细胞无血清培养基,不仅能够满足多种亚型昆虫细胞高密度的悬浮培养,而且可以维持更长时间更高活率的细胞状态,从而能够提高生产效率,降低生产成本,具有广泛的应用前景和价值。
3、本发明提供的昆虫细胞无血清培养基,能够显著提高病毒产量,降低病毒空壳率,有利于降低病毒规模化生产和质量检测成本。
综上,本发明的培养基弥补了现有昆虫培养基的不足,极大程度上改善了国内昆虫细胞无血清培养基在规模化应用过程中的性能不足,能够为基因治疗,特别是新冠疫苗等商业化生产提供质量稳定的高性能昆虫细胞无血清培养基。

以下实施例中所述的昆虫细胞培养基组成(包括氨基酸、维生素、无机盐、微量元素、碳水化合物和其他分子化合物各成分)均采购自西格玛(Sigma);商业化培养基Sf-900III SFM采购自赛默飞(Gibco);细胞株Sf-9、Sf-21、High Five均购自赛默飞(Gibco);qPCR检测试剂盒(厂家:百奥莱博,货号BTN14-62400)。
一、培养基的配置:
实施例1~7的昆虫细胞无血清培养基制备方法如下:将1L用量对应的氨基酸、维生素、无机盐、微量元素、碳水化合物和其他分子化合物组分加入800mL的注射用水中,室温搅拌30分钟后,加入一定量的氢氧化钠助溶,用盐酸调节pH到6.7~7.0,定容至1L后,用0.22µm无菌膜过滤,4℃长期保存备用,即可。
实施例1:由氨基酸、维生素、无机盐、微量元素、碳水化合物和其他分子化合物组成,

二、病毒表达和检测:
步骤1、细胞培养基的制备:
实施例1~7的昆虫细胞无血清培养基作为实验组;
商品化培养基Sf-900 III SFM作为对照组。
步骤2、驯化昆虫细胞:
将昆虫细胞(Sf-9、Sf-21或者High Five细胞)以0.4×106细胞数/mL的密度接种至含20mL实验组培养基和对照组培养基的125mL三角摇瓶中,将三角摇瓶置于28℃摇床中培养,转速120rpm;当细胞密度约3×106细胞数/mL时,以0.8×106细胞数/mL的密度接种传代,直至细胞传代10次以上。
步骤3、检测病毒滴度以及空壳率:
步骤3.1、将步骤2驯化并培养好的昆虫细胞(Sf-9、Sf-21或者High Five细胞)以0.8×106细胞数/mL的密度分别接种至含30mL对照组培养基和实验组培养基的125mL三角摇瓶中,将三角摇瓶置于28℃摇床中培养,转速120rpm。
步骤3.2、将步骤3.1的细胞培养2天后,此时细胞密度约为2.5~4×106细胞数/mL。以感染复数2MOI(即病毒与细胞个数比值为2)接种重组杆状病毒reAcMNPV06(申请人自行构建保有,能够稳定表达重组AAV病毒,其中AcMNPV由华南农业大学孙京臣教授赠予)至对照组培养基和实验组培养基培养的细胞中,继续28℃摇床中培养72小时,转速120rpm。其他能够表达AAV病毒的重组杆状病毒,也可进行病毒滴度以及空壳率的验证实验,并获得类似的验证结果。
步骤3.3、收集3.2步骤中培养的细胞上清,qPCR检测试剂盒检测培养上清中的病毒滴度以及病毒空壳率。
三、结果和分析:
1、根据步骤2驯化Sf-9细胞,每隔24小时取样计数,如图1所示实验组和对照组细胞培养至第6天(D06)达到密度峰值,实验组的实施例3活细胞峰值密度为11×106细胞数/mL,如图2所示,活率大于95%,明显优于对照组;
2、根据步骤2驯化Sf-21细胞,每隔24小时取样计数,如图3所示,实验组和对照组细胞培养至第6天(D06)达到密度峰值,实验组的实施例3活细胞峰值密度为13×106细胞数/mL,如图4所示,活率大于95%,明显优于对照组;
3、根据步骤2驯化High Five细胞,每隔24小时取样计数,如图5所示,实验组和对照组细胞培养至第5天(D05)达到密度峰值,实验组的实施例3活细胞峰值密度为12×106细胞数/mL,如图6所示,活率大于95%,明显优于对照组;
4、以Sf-9细胞为例,根据步骤3检测实验组的实施例3的培养基和对照组培养基表达病毒,病毒感染后培养3天取培养上清,检测上清中病毒滴度和病毒空壳率,如图7所示,实验组的实施例3上清中病毒滴度可达19×106TU/mL,如图8所示,病毒空壳率远低于5%,整体性能优于对照培养基。
上述实验结果表明:(1)本发明公开的昆虫细胞无血清培养基,是完全化学成分确定的,不含有任何动物来源性成分的无血清配方,不仅能够满足Sf-9、Sf-21和Higfh Five等多种昆虫细胞高密度的悬浮培养,而且整体性能表现优于对照的进口培养基,可以维持更长时间更高活率的细胞状态,具有广泛的应用前景和价值;(2)本发明公开的昆虫细胞无血清培养基,能够有效促进病毒的包装,提高培养病毒包装滴度,降低空壳病毒的组装概率,降低后期的质量检测和控制风险。
本发明的培养基不含血清和蛋白质,所有组分化学成分确定,能够满足多种昆虫细胞的高密度悬浮培养,并且本发明的培养基能够支持病毒的高滴度表达,降低病毒空壳率;本发明的培养基为国内研发产品,性能优于主流进口培养基,可以从整体上降低生产成本,适用于大规模生产。本发明的培养基弥补了现有昆虫细胞培养基的不足,填补了国内化学成分确定的昆虫细胞培养基的空白。

5.根据权利要求4所述的一种昆虫细胞无血清培养基的应用,其特征在于,所述的病毒为腺相关病毒。
6.根据权利要求1~3中任一所述的一种昆虫细胞无血清培养基的应用,其特征在于,该昆虫细胞无血清培养基用于生物类药物的生产。
7.根据权利要求6所述的一种昆虫细胞无血清培养基的应用,其特征在于,所述的生物类药物为抗体、融合蛋白、重组蛋白、酶、疫苗中的任意一种。
