Abstract

Description

本发明一般涉及制造和分装药物领域,更具体地说,涉及一种改进的、在一次性单个药剂的铝凸泡形包装(blister        pack)内的、制造冷冻干燥药物片剂的方法。近年来,药物制造商已转而用凸泡形包装供药物片剂成形和分装使用,这些凸泡形包装一般由凸泡形片或凸泡形膜和盖片组成。凸泡形片包含容纳单个药剂的一些凹陷。在用于制造冷冻干燥片剂的标准方法中,液状的单个药剂引入凸泡形片的每个凹陷中。与液状药剂一起,凸泡形片然后放入冷冻环境内,在此环境中,药剂遇到低温以将它们冷冻。之后,再将凸泡形片移入冷冻干燥器,在此,冰由于升华而去掉。当冷冻干燥完成时,凸泡形片从干燥室内取出并用一张粘性的盖片复盖,此盖片把固体的药剂密封在各个凹陷中。美国专利US4,305,502在此作为教导,特别是作为用于制造冷冻干燥片剂的一种已知的方法的参考文献引入。
然而,先前已经用于冷冻或冷冻干燥方法的凸泡形片有若干不足之处。首先,凸泡形片一般用聚合物质制造,它过一段时间会使潮气渗入凸泡形包装内并达到里面所储存的药剂。为解决这个问题,已经开发出一些凸泡形片,在这些凸泡形片中,把一层铝层压在两个聚合物层之间。铝层的存在防止了潮气渗入凸泡形包装,但它又带来了第二个问题。亦即,在冷冻过程中,当遇到温度变化时,传统的铝/聚 合物层压制件由于该层压制件相对层在热膨胀或收缩程度上的差异将趋于卷曲。这就使它们在冷冻过程中使用时造成困难,因为液体制品可能很容易地从所形成的凹陷中流出,或者可能在灌注和冷冻操作时不均匀地处在凹陷中。进而,凸泡形片的卷曲可能造成药剂受到冷冻或升华,因为某些凹陷不可以与冷冻器或冷冻干燥器的冷表面进行物理的接触。唯一的解决办法是在层压制件的边上使用重物,以便使它们所保持足够的平。但是,这样的办法在大批量的制造工作中是不实际的,且可能干扰冷冻过程。
因此,这就需要有一种作为在制造冷冻干燥片型中避免凸泡形片的卷曲问题的、使用高阻挡物铝层压制件的方法。
在一个基本的方面,本发明是一种改进的用于制造在铝凸泡形包装内的冷冻干燥剂型。药剂作为一种液体引入凸泡形片的凹陷中。凸泡形片包括一个位于第一和第二外层之间的密封的中间层,且每一个外层实质上具有同样整个的热膨胀系数,如在此所定义的术语那样。层压制件的外层的性质是这样的,使得当它在冷冻干燥过程中遇到温度变化时,不存在引起层压制件弯曲的层间应力。外层对这种温度变化的对称响应可以通过对于两个外层使用相同薄膜材料实现,或者通过使用不同的材料实现,所说的不同的材料借助于内在特性或者厚度,呈现类似的热膨胀或收缩程度。外层可以每一个由分离的次层组成,只要在一个外层中的各层是这样的,使得外层作为一个整体,显示与另外的外层有同样整个的膨胀或收缩度。在把药剂引入凸泡形片的凹陷内后,就使药剂被冷冻和冷冻干燥。之后,把盖片附着在凸泡形片上,以便把固体药剂密封在凸泡形片内。

如上所述,本发明是一种改进的、用于制造在铝凸泡形层压制件内冷冻干燥药剂型的方法。回到图1和图2,为了形成凸泡形包装,凹陷10是在所要求的层压制件的条带12上通过传统的冷成形方式形成。凹陷的大小和形状只是一个选择问题,这由要形成的片剂的大小和性质、以及还有本专业的普通技术人员熟知的其它考虑决定。
回到图3,层压制件的条带12包括中间层14,该层基本上是不透过潮气的。用于中间层的材料优选的是具有厚度为10至100μm的铝,优选的厚度为45μm,当然,也可以用其它合适的材料替代。中间的铝层14在第一外层16和第二外层18之间。外层可以涂复或层压在中间层上,但各层不必需粘在一起。第一和第二外层优选地由聚合物质制造,包括聚酰胺、聚氯乙烯、聚丙烯或其它这样的物质。第一和第二外层可以用相同的材料制造,或用不同的材料制造,且可以具有不同的厚度,只要它们基本上具有类似的热膨胀系数,亦即,当层压制件遇到温度变化时,特别是在冷冻过程中所遇到的温度范围内,在这个范围内温度可以低达-196℃,它们可以由这样的材料制造并具有这样的厚度,即第一和第二外层在薄膜的平面内基本上显示出相同的膨胀度或收缩度。例如,层压制件薄膜12包括位于聚丙烯的第一和第二外层16和18之间的中间的铝层14,每个 外层大约50μm厚。
回到图4,可以看出,外层之一或两个外层也可以由分离的次层组成,且每个次层可以是聚合物,也可以是非聚合物。例如,第一外层16可以由两个或多个次层组成,如聚酰胺次层20和聚氯乙烯次层22。第二外层18可以由一种同样的次层组成,或有两个或多不次层组成,如图中所示的24、26和28,它们与在第一外层中的各次层不同。可以用作次层的材料包括上面所说的聚合物,以及漆、铝或纸张。也可以包括底漆层。主要关心的仍然是第一外层16和第二外层18基本上整个地显示了在响应温度变化时,具有相同的膨胀度或收缩度,以便防止凸泡形片卷曲。
回到图1,液体状的单个药剂30以传统的方式被引入凸泡形片中的每个凹陷内。然后,凸泡形片放在冷冻装置中,例如,氮喷雾冷冻室,在该冷冻室内,凸泡形片和药剂经受到足以使药剂快速冷冻的温度,典型地低到-196℃。一旦药剂已经冷冻,凸泡形片转移到冷冻干燥室内。在冷冻干燥室内,药剂典型地受到0.1至1.0毫巴的真空,历时180至500分钟。与此同时,温度迅速地从约-30℃上升到约60℃。如在图5中所示的,一旦药剂已经冷冻干燥,粘性的盖片32复盖在凸泡形片上,使药剂密封在凸泡形片的各个凹陷内。有关将药剂引入凸泡形片、药剂物冷冻和冷冻干燥以及附着盖片的方法,这些对本专业的普通技术人员是公知的,在此不需要作更深入的讨论。
在前面已经说明了本发明的优选实施例,应该明白,本专业的普通技术人员还可以作出各种改进和变化,但这些都在权利要求书中所述的本发明的构思和范围之内。

Claims (11)

1、一种用于制造在凸泡形包装内的冷冻干燥剂型的方法,包括
a)把液体的药剂引入多层分层的凸泡形薄片的凹陷内,所述的薄片包括第一和第二外层,和一个位于第一和第二外层之间的密封的中间层,第一和第二外层实际上具有相同的热膨胀系数;
b)冷冻药剂;
c)把盖片附着在凸泡形薄片上,以便把药剂密封在凸泡形片的凹陷内。
2、如权利要求1所述的方法,其中凸泡形片的中间层是铝。
3、如权利要求1所述的方法,其中凸泡形薄片的第一和第二外层是用相同的物质制造的。
4、如权利要求1所述的方法中期凸泡形薄片的第一和第二外层至少一个是聚合物质。
5、如权利要求4所述的方法,其中聚合物质主要由包括聚乙烯、聚酰胺、聚氯乙烯和聚丙烯的组中选取的一种或多种聚合物组成。
6、如权利要求1所述的方法,其中凸泡形薄片的第一和第二外层实质上具有相同的厚度。
7、如权利要求1所述的方法,其中凸泡形薄片的第一和第二外层至少一个进一步包括多个次层。
8、如权利要求7所述的方法,其中所述那些次层之一是漆或底漆层。
9、如权利要求7所述的方法,其中所述那些次层之一是聚合物质。

11、一种用于制造在凸泡形包装的冷冻干燥剂型的方法,包括:
a)把液体的药剂引入多层分层的凸泡形薄片内的凹陷中,该薄片包括第一和第二聚合物外层,它们实际上具有类似的热膨胀系数,且该薄片包括一个位于第一和第二外层之间的、并连接到它们上面的中间层;
b)冷冻药剂;
c)冷冻干燥药剂;和
d)把盖片附着在凸泡形片上,以便把药剂密封在凸泡形片的凹陷内。
