Abstract
本发明公开了一种富硒海蓬子保健酒及其生产工艺,该发明选用海蓬子、枸杞、决明子、茯苓、绞股蓝、五味子、当归、葛根、杜仲、何首乌、红花、甘草、甜叶菊等中草药,通过富硒酿酒酵母发酵生产复合发酵型保健酒,产品酒质温和爽口、风味独特,营养价值高,基本保持海蓬子及中草药中的天然营养成分,且富含人体所需的有机硒,具有降血压、降血脂、抗疲劳、延缓衰老、提高机体免疫力、营养皮肤、润肺、健脾养胃、预防毛细血管疾病、增强心脏功能、预防老年痴呆等功效。本发明优势在于:一是通过酵母二步发酵提高了保健酒中的有机硒含量;二是科学配伍多种中草药,显著提升产品功效;三是通过酵母发酵和超声波联合作用,中草药活性成分提取率高。

技术领域
本发明涉及一种富硒海蓬子保健酒及其生产方法,属于保健食品领域。
背景技术
近年来,随着生活水平的日益提高,人们的健康意识越来越强,保健品市场的消费及购买力也不断的提升,特别是海洋保健产品因其含有极其丰富的营养成分、具有多种保健功效、无副作用等而深受消费者的青睐。发酵型保健酒不但含有植物中草药中对人体极具保健功效的活性营养成分,而且在发酵过程中产生大量的各种营养成分(如糖、蛋白质、人体必需氨基酸等),合理适度饮用能起到较好的保健作用,成为中草药保健食品领域乃至整个保健食品领域最具发展前途的产品。海蓬子又称海芦笋,是一种肉质化真盐生植物。其干燥植株含纤维素5~20%,蛋白质9.0%,富含人体必需的8种氨基酸、微量元素和维生素,其中胡萝卜素含量超过普通蔬菜的40倍,还含有天然的植物保健盐和天然的植物碱( 微角皂苷),具有降低胆固醇、降压、降血脂等功效。因此,开发海蓬子保健酒具有广阔的应用前景。
硒是维持人体正常生命活动所必需的元素之一,缺硒会导致多种疾病的发生。天然食品中硒含量普遍较低,紧靠天然食物中的硒并不能满足人的正常需要,膳食硒的低水平摄入对人体健康有潜在威胁,要保证人体健康,防治有关疾病,就应当有充分的硒摄入,通过摄取足够的硒来预防有关疾病的发生显得尤其重要,而无机硒毒性较高不能被人类所利用,通过植物和微生物将无机硒转化成有机硒是目前研究的热点,但植物体内的硒含量普遍较低,因而通过微生物转化可以为人类提供高产价廉的有机硒产品。
本发明以海蓬子为组方基础,配伍多种具有降血压、降血脂、抗氧化、抗衰老功效的中草药,利用高活性富硒酿酒酵母发酵具有降血压、降血脂、抗氧化、抗衰老、提高机体免疫力等多种保健功效的中草药,制备一种新的富硒海蓬子保健酒,既保留了海蓬子、枸杞等中草药的医学保健作用,拓展了中草药保健酒的应用途径,又提高了保健酒中的有机硒含量,是一种营养丰富、方便食用、风味独特的特色中草药保健酒,适合多种人群饮用,满足人们对健康保健食品的需求。
发明内容
本发明的目的在于提供一种富硒海蓬子保健酒及生产工艺,该发明选用海蓬子、枸杞、决明子、茯苓、绞股蓝、五味子、当归、葛根、杜仲、何首乌、红花、甘草、甜叶菊等中草药为原料通过富硒酿酒酵母发酵生产复合发酵型保健酒,该产品酒质温和爽口、风味独特,营养价值高,基本保持海蓬子及中草药中的天然营养成分,且富含人体所需的有机硒,具有降血压、降血脂、抗疲劳、延缓衰老、提高机体免疫力、营养皮肤、润肺、健脾养胃、预防毛细血管疾病、增强心脏功能、预防老年痴呆等功效。
为实现上述目的,本发明采用的技术方案是:
本发明提供一种富硒海蓬子保健酒,其特征在于:富硒海蓬子保健酒是以海蓬子、枸杞、决明子、茯苓、绞股蓝、五味子、当归、葛根、杜仲、何首乌、红花、甘草、甜叶菊等中草药为原料通过富硒酿酒酵母二步发酵工艺制得。
本发明提供一种富硒海蓬子保健酒,所述的中草药配伍比例为:海蓬子40~50份、枸杞20~30份、决明子15~20份、茯苓12~15份、绞股蓝8~10份、五味子8~10份、当归6~8份、葛根4~6份、杜仲3~5份、何首乌5~10份、红花15~20份、甘草5~10份、甜叶菊10~15份。
上述海蓬子原料为新鲜的海蓬子茎叶。
上述原料中海蓬子、枸杞预处理工艺为:称取一定量的海蓬子或枸杞,加3倍的水,放入75℃水浴中浸泡2~3h,然后打浆。
上述原料中其他原料的预处理工艺为:原料混合后粉碎过40目筛,加入5倍用Na2CO3调节pH为7.5~8.0的纯净水,然后加入中草药质量比1~2%的纤维素酶,在超声波辅助条件下浸提20~30min,浸提温度60~70℃。
上述原料预处理均不进行过滤。
本发明提供一种富硒海蓬子保健酒,其特征在于是按如下的方法制备:
(1)菌种活化:将保存的高活性富硒酿酒酵母在30~40℃温水中活化30min;
(2)发酵液配制:配制酿酒酵母一步发酵培养基,培养基葡萄糖浓度5%、酵母膏浓度1%,121℃灭菌30min;
(3)一步发酵:将活化后高活性富硒酵母接种到发酵液,接种比例2%,在28~32℃条件下培养48h,在培养12h、24h、36h时分别向培养基中添加10g/L的亚硒酸钠;
(4)过滤:发酵结束后过滤,分离酿酒酵母;
(5)中草药发酵培养基配制:将预处理后的中草药混合,添加0.15% (NH4)2SO4、0.15%K2HPO4、0.1%MgSO4;
(6)接种:将步骤(4)过滤后的酿酒酵母按10%的接种量接种中草药发酵培养基;
(7)控温发酵:在缓慢搅拌条件下控温发酵,发酵温度控制在26~28℃,发酵时间为36~48h;
(8)超声波破壁:发酵结束后采用超声波方式对酵母破壁20~30min。破壁时搅拌转速300r/min以上;
(9)过滤:发酵结束后过滤除去中草药渣和发酵菌体等固体不溶物;
(10)陈酿:为促进就提的稳定和澄清,将酒液置于0~10℃条件下陈酿3~5天;
(11)调配:调整富硒海蓬子保健酒的酒精体积浓度10~12%,加入1~1.5g/L木糖醇、3~5g/L山梨酸钾、1~2g/L柠檬酸钾、5~10g/L低聚异麦芽糖、5~10g/L蜂蜜;
(12)膜过滤:用孔径为0.15μm的微孔膜过滤海蓬子保健酒;
(13)无菌灌装:在无菌灌装生产线中灌装封口制得富硒海蓬子保健酒。
本发明提供一种富硒海蓬子保健酒及其制备工艺,通过动物试验进行富硒海蓬子保健酒的毒理学试验及安全性评价。小鼠急性经口毒性试验结果表明,本富硒海蓬子保健酒对小鼠急性经口最大耐受剂量大于高剂量组75mg/kg·bw; 遗传毒性试验小鼠骨髓细胞微核试验和小鼠精子畸形试验两项试验结果均为阴性,表明该富硒海蓬子保健酒无致突变作用;由大鼠30d喂养试验可知,大鼠体征行为正常、体态健康、血液及血液生化指标也无异常变化,因此75mg/kg·bw的剂量(人体推荐量的150倍)属于安全剂量;遗传毒性试验和大鼠30d喂养试验结果均为阴性,因此确定本富硒海蓬子保健酒属无毒类食品,人可以安全放心食用。
本发明提供一种富硒海蓬子保健酒及生产工艺,其中海蓬子保健酒的产品特征在于海蓬子保健酒的酒度为10%~12%/(v/v), 含糖量≤15g/L, 总酸( 以苹果酸计)6~10g/L, 挥发酸( 以乙酸计)< 1.2 g/L,干浸出物≥17 g/L,有机硒含量150~180μg/L;海蓬子保健酒色泽呈棕红色、晶莹透明、无悬浮物和沉淀物;入口绵软淳厚,舒适爽口,诸味协调。
本发明提供一种富硒海蓬子保健酒及生产工艺,其中富硒海蓬子保健酒的产品具有降血压、降血脂、抗疲劳、延缓衰老、提高机体免疫力、营养皮肤、润肺、健脾养胃、预防毛细血管疾病、增强心脏功能、预防老年痴呆等功效;本发明的转化应用不仅可以改变酒类消费结果,满足人民群众对绿色、安全保健食品的需求,有益于国民健康,而且可以高值化充分利用盐碱地植物资源,推动海蓬子加工业发展,有利于改善农业生态环境,推进循环经济发展。
本发明提供一种富硒海蓬子保健酒及生产工艺,其优势在于:一是采用高活性富硒酵母的二步发酵发酵工艺将无机硒转化为安全性和营养型更好的有机硒,提高了保健酒中的有机硒含量,经检测保健酒中有机硒的含量达到150~180μg/L,取得了预料不到的技术效果;二是科学配伍海蓬子和多种中草药,通过海蓬子和中草药之间的协同作用具有较好的降血压、降血脂、抗疲劳、延缓衰老、提高机体免疫力、营养皮肤、润肺、健脾养胃、预防毛细血管疾病、增强心脏功能、预防老年痴呆等功效,安全无副作用,食用方便。不同的中草药性味各异,功效存在着巨大的差异,不同的中草药组合后存在复杂的相互作用关系,且中草药之间的配比比较密切,不同配比也将使君臣佐使关系发生实质性的变化,从而使复合中草药的关系发生实质性改变。本申请通过海蓬子与枸杞、决明子、茯苓、绞股蓝、五味子、当归、葛根、杜仲、何首乌、红花、甘草、甜叶菊等中草药彼此间存在复杂的相互作用关系,合理配比,取得了预料不到的技术效果,使富硒海蓬子保健酒的关系发生实质性改变,产品较仅含有本申请中单一原料保健酒的抗氧化能力总和提高38.6%以上,具有较好的免疫调节活性;采用试剂盒法测定实验小鼠皮肤组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)及羟脯氨酸(Hyp)等指标,结果表明SOD、GSH-Px、Hyp等指标显著高于空白对照组(P<0.01),MDA显著低于空白对照组(P<0.01),说明产品具有较好的延缓衰老活性;改变原料组成或配比会降低富硒海蓬子保健酒的抗氧化能力和抗衰老能力(P<0.01),动物实验结果表明相同制备工艺条件下仅含有海蓬子原料的保健酒与不含有海蓬子原料保健酒(其余配方同实施例1)小鼠皮肤组织SOD指标分别为本申请产品的24.6%和58.9%;三是通过高活性酵母的发酵和后续超声波破壁联合作用,显著提高了中草药中皂甙、黄酮、生物碱等活性物质的提取率,检测结果表明皂甙、黄酮、生物碱提取率达到95%以上;四是采用微孔膜过滤的生产工艺,产品澄清度高,除菌、除杂效果好,灭菌效果彻底,替代了传统的巴氏灭菌法,产品中营养成分损失小。
具体实施方式
实施例1:
一种富硒海蓬子保健酒,其特征在于是按如下的方法制备:
(1)菌种活化:将保存的高活性富硒酿酒酵母在30~40℃温水中活化30min;
(2)发酵液配制:配制酿酒酵母一步发酵培养基,培养基葡萄糖浓度5%、酵母膏浓度1%,121℃灭菌30min;
(3)一步发酵:将活化后高活性富硒酵母接种到发酵液,接种比例2%,在28~32℃条件下培养48h,在培养12h、24h、36h时分别向培养基中添加10g/L的亚硒酸钠;
(4)过滤:发酵结束后过滤,分离酿酒酵母;
(5)中草药配伍:将中草药按海蓬子50份、枸杞20份、决明子20份、茯苓12份、绞股蓝10份、五味子8份、当归8份、葛根4份、杜仲5份、何首乌5份、红花20份、甘草5份、甜叶菊15份比例配伍;
(6)原料预处理:
①上述原料中海蓬子、枸杞预处理工艺为:称取一定量的海蓬子或枸杞,加3倍的水,放入75℃水浴中浸泡2~3h,然后打浆;
②上述原料中其他原料的预处理工艺为:原料混合后粉碎过40目筛,加入5倍用Na2CO3调节pH为7.5~8.0的纯净水,然后加入中草药质量比2%的纤维素酶,在超声波辅助条件下浸提20min,浸提温度60~70℃;
(7)中草药发酵培养基配制:将预处理后的中草药混合,添加0.15% (NH4)2SO4、0.15%K2HPO4、0.1%MgSO4;
(8)接种:将步骤(4)过滤后的酿酒酵母按10%的接种量接种中草药发酵培养基;
(9)控温发酵:在缓慢搅拌条件下控温发酵,发酵温度控制在26~28℃,发酵时间为36~48h;
(10)超声波破壁:发酵结束后采用超声波方式对酵母破壁20~30min。破壁时搅拌转速300r/min以上;
(11)过滤:发酵结束后过滤除去中草药渣和发酵菌体等固体不溶物;
(12)陈酿:为促进就提的稳定和澄清,将酒液置于0~10℃条件下陈酿3~5天;
(13)调配:调整富硒海蓬子保健酒的酒精体积浓度10~12%,加入1.5g/L木糖醇、3g/L山梨酸钾、2g/L柠檬酸钾、5g/L低聚异麦芽糖、10g/L蜂蜜;
(14)膜过滤:用孔径为0.15μm的微孔膜过滤海蓬子保健酒;
(15)无菌灌装:在无菌灌装生产线中灌装封口制得富硒海蓬子保健酒。
实施例2:
一种富硒海蓬子保健酒,其特征在于是按如下的方法制备:

(7)中草药发酵培养基配制:将预处理后的中草药混合,添加0.15% (NH4)2SO4、0.15%K2HPO4、0.1%MgSO4;
(8)接种:将步骤(4)过滤后的酿酒酵母按10%的接种量接种中草药发酵培养基;
(9)控温发酵:在缓慢搅拌条件下控温发酵,发酵温度控制在26~28℃,发酵时间为36~48h;
(10)超声波破壁:发酵结束后采用超声波方式对酵母破壁20~30min。破壁时搅拌转速300r/min以上;
(11)过滤:发酵结束后过滤除去中草药渣和发酵菌体等固体不溶物;
(12)陈酿:为促进就提的稳定和澄清,将酒液置于0~10℃条件下陈酿3~5天;
(13)调配:调整富硒海蓬子保健酒的酒精体积浓度10~12%,加入1g/L木糖醇、5g/L山梨酸钾、1g/L柠檬酸钾、10g/L低聚异麦芽糖、5g/L蜂蜜;
(14)膜过滤:用孔径为0.15μm的微孔膜过滤海蓬子保健酒;
(15)无菌灌装:在无菌灌装生产线中灌装封口制得富硒海蓬子保健酒。
实施例3:
一种富硒海蓬子保健酒,其特征在于是按如下的方法制备:
(1)菌种活化:将保存的高活性富硒酿酒酵母在30~40℃温水中活化30min;
(2)发酵液配制:配制酿酒酵母一步发酵培养基,培养基葡萄糖浓度5%、酵母膏浓度1%,121℃灭菌30min;
(3)一步发酵:将活化后高活性富硒酵母接种到发酵液,接种比例2%,在28~32℃条件下培养48h,在培养12h、24h、36h时分别向培养基中添加10g/L的亚硒酸钠;
(4)过滤:发酵结束后过滤,分离酿酒酵母;
(5)中草药配伍:将中草药按海蓬子45份、枸杞25份、决明子18份、茯苓14份、绞股蓝9份、五味子9份、当归7份、葛根5份、杜仲4份、何首乌8份、红花18份、甘草8份、甜叶菊12份比例配伍;
(6)原料预处理:
①上述原料中海蓬子、枸杞预处理工艺为:称取一定量的海蓬子或枸杞,加3倍的水,放入75℃水浴中浸泡2~3h,然后打浆;
②上述原料中其他原料的预处理工艺为:原料混合后粉碎过40目筛,加入5倍用Na2CO3调节pH为7.5~8.0的纯净水,然后加入中草药质量比1.5%的纤维素酶,在超声波辅助条件下浸提25min,浸提温度60~70℃;
(7)中草药发酵培养基配制:将预处理后的中草药混合,添加0.15% (NH4)2SO4、0.15%K2HPO4、0.1%MgSO4;
(8)接种:将步骤(4)过滤后的酿酒酵母按10%的接种量接种中草药发酵培养基;
(9)控温发酵:在缓慢搅拌条件下控温发酵,发酵温度控制在26~28℃,发酵时间为36~48h;
(10)超声波破壁:发酵结束后采用超声波方式对酵母破壁20~30min。破壁时搅拌转速300r/min以上;
(11)过滤:发酵结束后过滤除去中草药渣和发酵菌体等固体不溶物;

(13)调配:调整富硒海蓬子保健酒的酒精体积浓度10~12%,加入1.2g/L木糖醇、4g/L山梨酸钾、1.5g/L柠檬酸钾、7.5g/L低聚异麦芽糖、7.5g/L蜂蜜;
(14)膜过滤:用孔径为0.15μm的微孔膜过滤海蓬子保健酒;
(15)无菌灌装:在无菌灌装生产线中灌装封口制得富硒海蓬子保健酒。
以下以试验数据说明本申请取得的技术效果。
1、保健酒延缓衰老功能活性研究
动物实验方法:150只昆明种老龄小鼠随机分成5组:即对照组、实施例3试验组、对照试验1组(原料仅含海蓬子,其余同实施例3)、对照试验2组(原料仅含枸杞、决明子、茯苓、绞股蓝、五味子、当归、葛根、杜仲、何首乌、红花、甘草、甜叶菊等中草药原料,不含海蓬子,其余原料配方及制备工艺同实施例3)、对照试验3组(发酵菌种选用普通酵母菌,其余同实施例3),每组30只。保健酒样品按人体(60kg)推荐剂量为每人每天60ml ,试验时灌胃10ml/kg.d(相当于人体推荐用量)。对照组给予等量生理盐水,各组均采用灌胃法,每日2次,连续90天。实验期间各组每周重新称重1次,按体重的变化重新计算保健酒的剂量;
测试指标:采用试剂盒法测定实验小鼠皮肤组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)及羟脯氨酸(Hyp)等指标;
从上述试验结果中可以发现,本申请保健酒能够显著提升抗衰老保健功效,且改变原材料组成或发酵菌种会显著降低产品功效(P<0.01)。
2、保健酒降血脂功能活性研究
(1)试验动物
试验动物均为SD雄性大鼠,体重200±30克;
(2)模型制备
降血脂试验中采用高脂饲料喂饲制备高脂血症大鼠模型。高脂饲料配方:2%胆固醇,10%猪油,0.2%丙硫氧嘧啶,87.8%基础饲料,分别于第2、3周末大鼠禁食12 h,尾静脉取血检测血脂,较正常组血脂升高且稳定后,确认为高脂血症模型;
(3)试验分组
将试验大鼠分为五组,分别为模型对照组、实施例3试验组、对照试验1组(原料仅含海蓬子,其余同实施例3)、对照试验2组(原料仅含枸杞、决明子、茯苓、绞股蓝、五味子、当归、葛根、杜仲、何首乌、红花、甘草、甜叶菊等中草药原料,不含海蓬子,其余原料配方及制备工艺同实施例3)、对照试验3组(发酵菌种选用普通酵母菌,其余同实施例3),每组30只;对照组给予等量生理盐水,各组均采用灌胃法,每日2次,连续90天。实验期间各组每周重新称重1次,按体重的变化重新计算保健酒的剂量;
(4)试验方法
降血脂试验:各组动物分别于给药前、给药后30 d取空静脉血,肝素抗凝,立即3 500r/min离心5min提取血清,按要求保存,测定大鼠的TC、TG、HDL-C等指标;
(5)试验结果
由表2可以发现本申请对降低大鼠血脂具有较显著的功效(与模型对照组相比差异有统计学意义,P<0.01);实施例3试验组的降血脂效果显著优于对照试验组,差异有统计学意义(P<0.01),这表明中草药配方和保健酒加工工艺对保健酒的降血脂效果有显著的影响。
以上实施例仅用于说明本发明的技术方案,而非对其进行限制;尽管参照前述实施例对被发明进行了详细的说明,但对于本领域的普通技术人员来说,依然可以对前述实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换;而对这些修改或者替换,并不使相应技术方案的本质脱离本发明所要求保护的技术方案的精神和范围。
Claims (6)
1.一种富硒海蓬子保健酒,其特征在于富硒海蓬子保健酒是海蓬子及中草药原料通过富硒酿酒酵母二步发酵工艺制得;所述海蓬子及中草药原料配伍比例为:海蓬子40~50份、枸杞20~30份、决明子15~20份、茯苓12~15份、绞股蓝8~10份、五味子8~10份、当归6~8份、葛根4~6份、杜仲3~5份、何首乌5~10份、红花15~20份、甘草5~10份、甜叶菊10~15份。
2.如权利要求1所述的富硒海蓬子保健酒,其特征在于所述海蓬子原料为新鲜的海蓬子茎叶。
3.如权利要求1所述的富硒海蓬子保健酒,其特征在于所述海蓬子、枸杞的预处理工艺为:称取一定量的海蓬子或枸杞,加3倍的水,放入75℃水浴中浸泡2~3h,然后打浆。
4.如权利要求1所述的富硒海蓬子保健酒,其特征在于所述原料中除海蓬子和枸杞外的其他原料预处理工艺为:混合后粉碎过40目筛,加入5倍用Na2CO3调节pH为7.5~8.0的纯净水,然后加入中草药质量比1~2%的纤维素酶,在超声波辅助条件下浸提20~30min,浸提温度60~70℃。
5.如权利要求3-4所述的富硒海蓬子保健酒,其特征在于所述原料预处理工艺均不进行过滤。
6.如权利要求1所述的富硒海蓬子保健酒,其特征在于所述富硒酿酒酵母二步发酵工艺具体包括以下步骤:
(1)菌种活化:将保存的高活性富硒酿酒酵母在30~40℃温水中活化30min;
(2)发酵液配制:配制酿酒酵母一步发酵培养基,培养基葡萄糖浓度5%、酵母膏浓度1%,121℃灭菌30min;
(3)一步发酵:将活化后高活性富硒酵母接种到发酵液,接种比例2%,在28~32℃条件下培养48h,在培养12h、24h、36h时分别向培养基中添加10g/L的亚硒酸钠;
(4)过滤:发酵结束后过滤,分离酿酒酵母;

(10)陈酿:为促进就提的稳定和澄清,将酒液置于0~10℃条件下陈酿3~5天;
(11)调配:调整富硒海蓬子保健酒的酒精体积浓度10~12%,加入1~1.5g/L木糖醇、3~5g/L山梨酸钾、1~2g/L柠檬酸钾、5~10g/L低聚异麦芽糖、5~10g/L蜂蜜;
(12)膜过滤:用孔径为0.15μm的微孔膜过滤海蓬子保健酒;
(13)无菌灌装:在无菌灌装生产线中灌装封口制得富硒海蓬子保健酒。
