背景技术
跨膜蛋白CEACAM1[也称为胆汁糖蛋白(BGP)、CD66a和C-CAM1]是癌胚抗原家族(CEA)的成员,也属于免疫球蛋白超家族。CEACAM1与包括CD66a、CD66c和CD66e蛋白在内的其它已知CD66蛋白相互作用。它表达于从上皮细胞到造血来源的那些细胞(例如,免疫细胞)的各种各样的细胞上。
许多不同功能归因于CEACAM1蛋白。已证明,CEACAM1蛋白对结肠癌、前列腺癌以及其它类型的癌症表现出抗增殖特性。另外的数据证实CEACAM1主要参与血管生成和转移。CEACAM1也在先天性和适应性免疫应答的调节中发挥作用。例如,CEACAM1被证明是包含在人肠上皮内的活化T细胞的抑制性受体[参见,WO99/52552和Morales等人,J.Immunol.163(1999),1363-1370]。另外的报道已表明,CEACAM1通过与mAb交联的TCR或通过淋病奈瑟氏菌Opa蛋白抑制T细胞活化和增殖。
黑素瘤是色素产生细胞(黑素细胞)的恶性肿瘤,全球皮肤癌相关的发病率的75%由黑素瘤引起,其主要是由广泛转移引起。转移性黑素瘤(MM)对大多数抗癌方案的反应都很微弱,MM患者的平均总存活时间为8.5个月。正常黑素细胞很少表达CEACAM1,而在黑素瘤细胞上经常发现CEACAM1。原发性皮肤黑色素瘤病变中的CEACAM1表达强烈地预示着不良预后的转移性疾病的发展。而且观察到,来源于一些转移性黑素瘤患者的NK细胞上的CEACAM1表达相比健康供体有所增大。

本发明人经过艰苦的实验和筛选生产出对CEACAM1具有选择性的单克隆抗体。这种抗体已被证明优于其它抗CEACAM1单克隆抗体,如通过功能阻断试验所表明的。

非人(如鼠类)抗体的人源化形式(人化形式,humanized form)为免疫球蛋白、免疫球蛋白链或其片段(如Fv、Fab、Fab'、F(ab')2或其它抗体的抗原结合序列)的嵌合分子,其包含极小的源于非人免疫球蛋白的序列。人源化抗体包括人免疫球蛋白(受体抗体),其中形成受体互补决定区(CDR)的残基被来自非人物种(供体抗体)如小鼠、大鼠或家兔的CDR、具有期望特异性、亲和性和接受力(能力、容量,capacity)的残基所替代。在一些情况下,人免疫球蛋白的Fv框架残基被相应的非人残基替代。人源化抗体还可包含既不能在受体抗体中找到又不能在所输入的CDR或框架序列中找到的残基。一般而言,该人源化抗体包含基本所有的至少一个,典型地为两个可变结构域,其中对应于非人免疫球蛋白CDR区的所有或基本所有的CDR区,以及所有或基本所有的FR区是人免疫球蛋白共有序列的那些。该人源化抗体还最佳地包含免疫球蛋白恒定区(Fc),典型地为人免疫球蛋白的恒定区的至少一部分[Jones等人,Nature,321:522-525(1986);Riechmann等人,Nature,332:323-329(1988);和Presta,Curr.Op.Struct.Biol.,2:593-596(1992)]。
用于人源化非人抗体的方法是本领域熟知的。一般地,人源化抗体具有一个或多个从非人来源引入其中的氨基酸残基。这些非人氨基酸残基通常称为输入残基(import residues),其典型地取自输入可变结构域。基本可按Winter和同事[Jones等人,Nature,321:522-525(1986);Riechmann等人,Nature332:323-327(1988);Verhoeyen等人,Science,239:1534-1536(1988)]的方法,通过将啮齿动物CDR或CDR序列替换为人抗体的相应序列进行人源化。因此,此种人源化抗体为嵌合抗体(美国专利4,816,567),其中不足一个完整的人可变结构域基本上已被来自于非人物种的相应序列所替代。实际上,人源化抗体典型地为这样的人抗体,其中一些CDR残基和可能的一些FR残基被来自于啮齿动物抗体相似位点的残基所替代。

本发明抗体可以降低抑制性CEACAM1嗜同性(或同型)相互作用或异型相互作用,从而增强淋巴细胞的活性。CEACAM1嗜同性相互作用通过N-结构域发生。数种氨基酸对这种相互作用起着关键作用,包括R43、Q44、D64和R82。该相互作用导致招募SHP-1磷酸酶的胞浆(细胞质)酪氨酸残基磷酸化。这在淋巴细胞内启动了抑制级联,其靶向邻近的介体(介质,mediator),如ZAP70。

本发明的药物组合物可通过本领域熟知的工艺制造,如通过常规的混合、溶解、造粒、包糖衣、研粉、乳化、制成胶囊、包埋(entrapping)或冻干工艺制造。
因此,根据本发明使用的药物组合物可用常规方法,使用一种或多种使活性成分容易加工成可以药用的制剂的生理可接受载体(包括赋形剂和助剂)而制造。合适的剂型取决于所选的给予途径。
对于注射,该药物组合物的活性成分可调配在水溶液中,优选调配在生理上相容的缓冲液如汉克溶液(Hank’s solution)、林格溶液(Ringer’ssolution)或生理盐缓冲液中。对于经粘膜给予,可以在配方中使用适合于透过屏障的穿透剂(penetrant)。此种穿透剂在本领域一般是已知的。

对于通过鼻吸入给予,根据本发明使用的活性成分可以喷雾剂(aerosol spray)形式,从利用适合的推进剂如二氯二氟甲烷、三氯氟甲烷、二氯四氟乙烷,或二氧化碳的加压包装或喷雾器中方便地递送。在加压喷雾剂的情况下,剂量单位可通过提供阀门确定从而递送计量的量。可将用在分配器中的如明胶的胶囊和药筒,调配成包含该化合物与合适的粉末基质如乳糖或淀粉的粉末混合物。
本文描述的药物组合物可调配用于肠胃外给予,如通过推注和连续输注给予。注射用制剂可以单位剂量的形式呈现,如,在安瓿(ampoules)或多剂量容器中,在其中可选地加入防腐剂。该组合物可以是油性或水性媒剂(媒介物,vehicles)中的悬浮液、溶液,或乳液,并可含有调配剂如悬浮剂、稳定剂,和/或分散剂。

本发明的药物组合物还可利用如常规栓剂基质如可可油或其它甘油酯,调配成直肠组合物如栓剂或保留灌肠剂(retention enemas)。
适用于本发明上下文的药物组合物包括其中活性成分的含量对达到预期目的有效的组合物。更具体地,治疗有效量是指对预防、缓和或改善紊乱(如,癌症)的症状或延长受治疗受验者的存活时间有效的活性成分量。

对于用于本发明方法中的任何制剂,治疗有效量或剂量最初可根据体外和细胞培养物分析进行估计。例如,可以在动物模型中调配剂量,以获得期望的浓度或滴定度。此类信息可用来更准确地确定对于人有用的剂量。

实施例1
生成单克隆抗体
生成MRG1单克隆抗体
生成以纳摩尔浓度(nanomolar concentration)在体外有效地阻断CEACAM1嗜同性相互作用的单克隆抗体。简言之,使用5微克重组人CEACAM1(完整蛋白,购自R&D System)免疫小鼠,每隔2周免疫一次,共免疫3次。收获脾细胞,并使其与SP2/0细胞融合,从而生成杂交瘤文库。
将产生阻断CEACAM1的抗体(MRG1 mAb)的杂交瘤再克隆数次从而得到稳定的克隆。
其它单克隆抗体
Kat4c mAb和家兔多克隆抗CEACAM(抗体)购自DAKO(Glostrup,Denmark)。
实施例2
抗CEACAM1mAb的特异性
材料和实验程序
生成表达CEACAM的细胞
通过电穿孔用CEACAM1、CEACAM5、CEACAM6或CEACAM8稳定转染CEACAM-阴性721.221人细胞(亲代B细胞),并用G418进行筛选。
用包含融合到鼠类ζ链(murine zeta chain)的跨膜和胞质尾区上的人CEACAM1胞外部分的嵌合分子转染鼠类胸腺瘤BW亲代细胞(缺少TCRα和β链,但完全保留IL-2的分泌机制的细胞)。转染通过电穿孔执行,用G418筛选。
FACS法抗体筛选
通过流式细胞术如下筛选出具有CEACAM1结合活性的杂交瘤:
(a)将50,000个转染的CEACAM细胞置于96-U形孔中。
(b)用冷FACS缓冲液(PBS,BSA0.5%,叠氮化物(Azide)0.05%)洗涤该细胞。
(c)将该细胞与染色mAb(MRG1或Kat4c):0.1微克mAb/100微升一起在冰上温育30分钟。
(d)离心该细胞,除去上清液,将该细胞以1:200的稀释度重悬在100微升FITC-结合的山羊抗小鼠抗体(Jackson Immunoresearch)中。
(e)温育30分钟(在冰上于黑暗条件下温育)后,离心该细胞,洗涤并将其重悬在FACS缓冲液中。
(f)利用FACScalibur和CellQuest软件分析细胞。
结果
因为721.221亲代细胞不表达CEACAM蛋白中的任何一种,所以用CEACAM1、CEACAM5、CEACAM6或CEACAM8稳定转染这些细胞以便测试CEACAM1单克隆抗体(mAb)的特异性。然后通过流式细胞术筛选具有CEACAM1结合活性的杂交瘤。如图1A所示,根据本教导生成的MRG1mAb对人CEACAM1具有特异性。它与CEACAM5的交叉反应性不显著,而且不与CEACAM6或CEACAM8结合。图1B示出,所有转染子都表达了CEACAM分子,其中CEACAM1最低,这强调了MRG1的特异性模式。
实施例3
该mAb能够抑制CEACAM1嗜同性结合
材料和实验程序
ELISA法抗体筛选
利用BW功能系统测试CEACAM1阻断活性。该BW功能系统包括用包含融合到鼠类ζ链(BW/CEACAM1-ζ,参见上面的实施例2)上的人CEACAM1胞外结构域的嵌合分子稳定转染的小鼠细胞系(BW)。BW/CEACAM1-ζ细胞与其它CEACAM1阳性细胞的共温育导致可测量浓度的小鼠IL-2的分泌。
因而,将BW/CEACAM1-ζ(效应细胞)或221/CEACAM1(靶细胞)各自单独地与10-40ng/ml MRG1mAb预温育。在冰上温育1小时后,加入易位细胞(221/CEACAM1或BW/CEACAM1),并通过夹心ELISA(R&Dsystems)测量小鼠IL-2的分泌。
细胞毒性分析
在存在或不存在1μg/ml MRG1mAb的情况下,执行测试肿瘤浸润淋巴细胞对各种黑素瘤细胞系的杀伤的细胞毒性分析。CEACAM1High526mel、624mel和CEACAM1dim09mel黑素瘤细胞用作靶细胞。TIL014细胞用作效应细胞,E:T比例为10:1。在与MRG1mAb在冰上温育1小时后,加入易位细胞,并在37℃下共温育5小时。靶细胞用绿色荧光染料(CFSE)预先标记,并通过流式细胞术碘化丙啶(PI)共染色法测定特异性溶解。扣除自然死亡。
结果
证实了纯化的MRG1mAb抑制CEACAM1嗜同性结合的能力。如图2所示,纯化的mAb MRG1对CEACAM1嗜同性结合表现出剂量依赖性抑制。该mAb在浓度为10ng/ml时高效地降低了CEACAM1相互作用,在浓度为20ng/ml时有效地达到平稳状态(plateau)。重要地,该两个实验状况,即,将MRG1mAb加入到效应细胞BW/CEACAM1-ζ中,或者将MRG1mAb加入到靶细胞221/CEACAM1中,表现出类似的结果(有效地阻断小鼠IL-2的分泌)。
MRG1mAb的阻断作用进一步在细胞毒性分析中得到证明。如图3所示,对CEACAM1High526mel和624mel细胞的杀伤可通过将该抗体与效应细胞一起温育而增强(但不适用于靶细胞)。MRG1mAb的存在并不影响对CEACAM1dim09mel细胞的杀伤(图3)。
实施例4
抗CEACAM1mAb抑制癌症细胞迁移和增殖
材料和实验程序
侵袭分析
在侵袭分析中测试抗体的阻断作用。简言之,在存在或不存在1μg/mlMRG1mAb的情况下预温育黑素瘤细胞(08mel或09mel),然后通过基质胶侵袭试验测试。使侵袭进行24小时,利用标准化XTT量化侵袭细胞量。
净增殖分析
在第0天将CEACAM1High526mel细胞接种在48孔板(2,500个细胞/孔)中。在接种后,加入三种不同浓度(0.5、1或3μg/ml)的MRG1,或根本不加入MRG1。在接种2或5天后对总活细胞进行计数。利用标准化的XTT,通过直接细胞计数确定增殖。
结果
如图4所示,MRG1阻断了CEACAM1阳性08mel细胞的侵袭(CEACAM1表达水平为中等,即,CEACAM1表达的中位荧光强度(median fluorescence intensity)为50),而对CEACAM1dim09mel细胞的影响很小或没有影响(CEACAM1表达水平低,即,CEACAM1表达的中位荧光强度为15)。
也在净增殖分析中测试了MRG1。观察到它以剂量依赖方式抑制526mel细胞的净增殖(图5)。处理5天后,增殖降低超过60%(用3μg MRG1mAb处理)。
实施例5
MRG1抑制动物实验模型中的癌症细胞增殖
材料和实验程序
黑素瘤异种移植模型
将5x106个CEACAM1+人黑素瘤细胞皮下注射到7周龄的SCID-NOD小鼠侧腹中。肿瘤块(肿瘤组织tumor masses)在14~17天内形成在100%的小鼠中,并继续生长。用测径器非侵袭地监测肿瘤尺寸,每周监测3次,并按(d1xd2xd3/2)计算近似体积。
通过腹膜内注射用0.5ml无菌PBS稀释的0.5mg抗体,给予MRG1。注射PBS作为对照。
通过将用200μl无菌PBS稀释的20x106个细胞静脉注射到尾静脉中,给予反应性人抗黑素瘤淋巴细胞。
结果
与上面证明的阻断功能一致,给予MRG1抗体抑制了肿瘤生长。当在肿瘤细胞接种时给予该抗体(图6A,“预防方案”)或在可测量肿瘤块已经形成时给予该抗体(图6B,“治疗方案”),这种作用是明显的。在8天内注射4次并接着非侵袭性监测之后,这些作用是明显的(见图6中的箭头)。应当注意,这种作用与任何免疫调节作用无关,因为SCID-NOD小鼠是免疫缺陷的。
通过单独静脉注射反应性人抗黑素瘤淋巴细胞(其抑制肿瘤生长)对抗黑素瘤免疫应答进行刺激(图7)。通过每周腹膜内注射一次MRG1可显著增强这种作用。
实施例6
MRG1优于前面描述的抗CEACAM1抗体
材料和实验程序
ELISA法抗体筛选
利用上文实施例3详细描述的BW功能系统测试CEACAM1阻断活性。
将100,000个BW/CEACAM1-ζ细胞与15ng/ml MRG1mAb、2600ng/ml Kat4c mAb或600ng/ml家兔多克隆抗CEACAM抗体一起预温育。在冰上温育1小时后,加入50,000个721.221/CEACAM1细胞,并通过夹心ELISA(R&D Systems)测量小鼠IL-2的分泌。
结果
如上文实施例3所描述的,本发明人证明了使用15ng/ml MRG1mAb几乎可以完全阻断CEACAM1活性。相比之下,所测试的抗CEACAM1单克隆抗体Kat4c仅在浓度高200倍时才能够产生较小的阻断作用,浓度高40倍的多克隆家兔抗CEACAM抗体产生类似的抑制作用(分别为2600ng/ml和600ng/ml,图8)。

1.一种以ATCC登录号PTA-9974保藏的杂交瘤细胞。
2.一种由根据权利要求1所述的杂交瘤细胞生产的分离的抗体。
3.根据权利要求2所述的分离的抗体,其附接在细胞毒性部分上。
4.根据权利要求3所述的分离的抗体,其中,所述细胞毒性部分包括细胞毒素、趋化因子、化疗药、促凋亡剂、干扰素、放射性部分、或它们的组合。
5.根据权利要求2所述的分离的抗体,其附接在可鉴定部分上。
6.根据权利要求2所述的抗体在制备用于免疫调节表达CEACAM1的淋巴细胞的药物中的应用。
7.根据权利要求2所述的抗体在制备用于抑制表达CEACAM1的肿瘤细胞的迁移或增殖的药物中的应用。
8.根据权利要求2或5所述的抗体在制备用于诊断需要其的受验者中的癌症的药物中的应用,所述诊断包括使源自所述受验者的生物样品与根据权利要求2或5所述的抗体接触,其中,超出预定阈值的复合物形成表示所述受验者患有癌症。
9.治疗有效量的根据权利要求2所述的抗体在制备用于治疗需要其的受验者中表达CEACAM1的癌症的药物中的应用。
10.根据权利要求9所述的应用,进一步包括向所述受验者给予淋巴细胞。
11.根据权利要求10所述的应用,其中,所述淋巴细胞包括T细胞或NK细胞。
12.根据权利要求2所述的抗体在制备用于抑制CEACAM1同型或异型的蛋白质-蛋白质相互作用的药物中的应用,其中,根据权利要求2所述的抗体用于与表达CEACAM1的淋巴细胞接触。
13.根据权利要求6或12所述的应用,其中,所述表达CEACAM1的淋巴细胞是肿瘤浸润淋巴细胞或NK细胞。
14.根据权利要求6或12所述的应用,其中,所述表达CEACAM1的淋巴细胞是细胞毒性T细胞。
15.根据权利要求7所述的应用,其中,所述肿瘤细胞包括黑素瘤肿瘤细胞。
