Description
Verfahren zur Herstellung von organischen Phosphorverbindungen Das Hauptpatent beschreibt organische Phosphorverbindungen der Formel EMI1.1 In dieser Formel bedeuten RI und R2 gleiche oder verschiedene Alkylgruppen von 1 bis 6 Kohlenstoff- atomen, von welchen Alkylgruppen mindestens eine durch ein Halogenatom substituiert ist ; X ein Sauerstoffoder Schwefelatom ; Yt und y2, welche gleich oder verschieden sind, je ein Wasserstoff-oder Halogenatom, eine Alkylgruppe von l bis 4 Kohlenstoffatomen oder einen Benzyl-oder Phenylrest, welch letztere im Kern mit Halogenatomen, Alkylgruppen von 1 bis 4 Kohlen- stoffatomen oder Nitrogruppen substituiert sein kömnen, oder Yi und y2 bilden zusammen eine Alkylenkette -Yl Y-von 3 bis 6 Kohlenstoffatomen. Die Verbindungen der Formel I sind wirksam bei der Bekämpfung eines Befalles durch Nematoden der Arten Aspiculuris tetraptera, Nematospiroides dubius und Syphacia obvelata im Verdauungstrakt von Mäusen, der Gattungen Coopenia, Haemonchus, Oesophagoste- mum, Strongyloides, Trichostrongylus und Nematodirus im Verdauungstrakt von Schafen und Rindern, Ancylo stoma und Toxa caris im Verdauungstrakt von Hunden und der Art Capillaria obsignata in Hühnern. Es zeigte sich, dal3 organische Phosphorverbindun- gen der Formel II, welche denjenigen der Formel I nahe verwandt sind, gegen die gleichen gastro-intestinalen Nematoden wirksam sind. Die vorliegende Erfindung betrifft nun ein Verfahren zur Herstellung solcher Verbindungen der Formel EMI1.2 worin R3 und R4 gleiche oder verschiedene, mit Halogen substituierte Alkylreste von I bis 4 Kohlenstoffatomen und Y3 und R4 gleiche oder verschiedene Alkoxycarbo nyl-1-oder Alkoxycarbonylalkylgruppen bedeuten, oder einer den Substituenten Y3 und Y4 Wasserstoff oder der Methylrest ist, wobei die Alkoxycarbonylgruppen oder die Alkoxycarbonylreste der Alkoxycarbonylalkylgruppen 2 bis 6 und die Alkylreste in den Alkoxycarbonyl- alkylgruppen 1 bis 4 Kohlenstoffatome aufweisen. Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man eine Phosphorverbindung der Formel EMI1.3 mit einem Cumarinderivat der Formel EMI1.4 umsetzt, in welchen Formeln Z ein Halogenatom und M Wasserstoff oder ein MetaHatom, oder Z und M beide Wasserstoffatome sind. Man kann z. B. eine Verbindung der Formel II herstellen, indem man eine Phosphorverbindung der Formel III, in der Z ein Chloratom ist, mit einem Cumarinderivat der Formel IV, worin M Wasserstoff ist, in Gegenwart eines Säunebindemittels, wie Alkalicarbonat, z. B. Soda, oder einer organischen Base, z. B. Tri äthylamin, und eines Katalysators, z. B. fein verteiltes Kupfer, in einem Lösungsmittel, wie Aceton, Athylmethylketon, Methylpropylketon, Toluol oder Benzol, erwärmt. Wenn z. B. M ein Alkalimetall ist, kann die Verwendung des Säurebindemittels entfallen. Man kann auch derart vorgehen, dass man eine Phosphorverbindung der Formel III, in der Z ein Wasserstoffatom ist, mit einem Cumarinderivat der For, mel IV, in welcher M ebenfalls ein Wasserstoffatom ist, in Gegenwart eines Säurebindemittels, z. B. Pottasche, oder einer organischen Base, z. B. Pyridin, bei einer zwischen 10 und 50 C liegenden Temperatur umsetzt. Bei dieser Durchführungsart arbeitet man zweckmässig in einem Lösungsmittel, welches gleichzeitig als Oxydationsmittel dient, z. B. Kohlenstofftetrachlorid, aber man kann auch ein nichtoxydierendes Lösungsmittel, z. B. Benzol, verwenden, und setzt dann ein Oxydationsmittel, z. B. Kohlenstofftetrabromid, zu. Die Verbindungen der Formel II können zusammen mit einem Trägerstoff zur Verwendung gelangen. Ein solches Präparat kann ein Verdünnungsmittel und ein Dispergiermittel enthalten. Beispiel 1 Zu einer Mischung von 11, 7 g 4-Athoxycarbonyl-7- hydroxycumarin, 14, 0 g wasserfreiem Kaliumcarbonat und 250 ml Methyläthylketon, die eine Stunde erhitzt und gerührt worden ist, gibt man 12, 0 g bis- (2-Chlor- äthyl)-phosphorchloridsalz während 1/2 Stunde zu und erhitzt die Mischung 3 Stunden lang unter Rückfluss. Nachdem sich die Suspension abgekühlt hat, wird sie filtriert und das Lösungsmittel durch Destillation ent fernt. Den Rückstand löst man in 100 ml Chloroform und wäscht die Lösung zweimal mit einer Lösung von 100 ml Natriumcarbonat, dann mit 50 ml Wasser, trocknet über wasserfreiem Natriumsulfat und dampft das Lösungsmittel ein. Das zurückbleibende gelbe 01 löst sich leicht in Athanol, und die Zugabe von Diisopropyl äther liefert Kristalle von O, O-bis (2-Chlor-äthyl)-O- (4- äthoxycarbonyl-cumarin-7-yl)-phosphat vom Schmelzpunkt 68-70 C. Gemäss Analyse enthält es 7, 44 % Phosphor (theoretische Menge 7, 06 %). Beispiel 2 Zu einer Mischung von 26, 2 g 3-Athoxycarbonyl- methyl-4-methyl-7-hydroxy-cumarin und 20, 7 g bis-(2 Chloräthyl)-phosphit in 200 ml Tetrachlorkohlenstoff gibt man langsam unter Rühren 13, 5 g Triäthylamin und hält die Temperatur der Mischung durch geeignetes Kühlen unter 35 C. Nach Beendigung der Zugabe wird die Mischung einige Stunden bei Zimmertemperatur gerührt und 100 ml Chloroform und 100 ml Wasser zugesetzt, um den ausgefallenen Feststoff zu lösen. Die untere Chloroformschicht wird nacheinander mit ver dünnter Salzsäure, Natriumcarbonatlösung und Wasser gewaschen, die gewaschene Lösung über wasserfreiem Natriumsulfat getrocknet und eingedampft. Den öligen Rückstand löst man in ¯thanol, und nach Zugabe von Diisopropyläther scheiden sich farblose Kristalle von O, O-bis-(2-Chlor-äthyl)-0-(3-äthoxycarbonylmethyl-4- methyl-cumarin-7-yl)-phosphat vom Schmelzpunkt 68 bis 69 C ab. Gemäss Analyse enthält es 6, 57 % Phosphor, 46, 37 % Kohlenstoff, 4, 65 % Wasserstoff und 14, 62% Chlor (theoretische Mengen Phosphor 6, 64 %, Kohlenstoff 46, 37 %, Wasserstoff 4, 53 %, Chlor 15, 18 %). In Beispiel 8 wird das Produkt als gelbes öl erhal- ten, dessen Brechungsindex n 1D in der folgenden Tabelle unter der Spalte unkorrigierter Schmelzpunkto ange- geben ist. Die erhaltene Mischung wird auf 45 C abgekühlt, mit 3n Schwefelsäure angesäuert und mit 1 Liter Eiswasser ver dünnt. Das Gemisch lässt man 30 Minuten stehen und giesst es dann in dünnem Strom in 1, 5 Liter gutgerührtes Eiswasser. Nach lOminütigem Rühren wird der Feststoff abfiltriert, mit Wasser gewaschen und bei ungefähr 110 C getrocknet. Das als 7-Hydroxy-4-äthoxycarbonylcumarin identifizierte Produkt hat einen Schmelzpunkt von 149-151 C. Zu einer gerührten Mischung von 135 g 7-Hydroxy 4-äthoxycarbonyl-cumarin, 120 g 0,0-bis-(2-Chlor- äthyl)-phosphit und Tetrachlorkohlenstoff setzt man 73, 5 g Triäthylamin mit solcher Geschwindigkeit zu, dass die Temperatur bei ungefähr 35 C gehalten wird. Nach beendeter Zugabe wird das Gemisch über Nacht bei Zimmertemperatur gerührt und ergibt eine feste Masse. Diese wird mit 300 ml Wasser behandelt und 10 Minuten gerührt, bevor genügend Chloroform zugegeben wird, um den ganzen Feststoff in Lösung zu bringen. Die Lösung wäscht man dann nacheinander mit 2n Salzsäure, Natriumbicarbonatlösung, Wasser und Salzlösung und trocknet sie dann über wasserfreiem Natriumsulfat. Das überschüssige Lösungsmittel wird unter ver mindertem Druck abgedampft und der ölige Rückstand in einem gleichen Volumen von I. M. S. gelöst. Zu der Ollosung setzt man Holzkohle zu, filtriert und verdünnt mit einem gleichen Volumen Diisopropyläther. Das Produkt ist ein farbloser kristalliner Feststoff vom Schmelzpunkt 70-71 C und erweist sich als 0, 0-bis-2 (Chlor äthyl-O-(44Åahoxydarbonyl-cumari'n-7-yl)-phosphat.
Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von Organophosphorverbindungen der Formel EMI3.1 worin R3 und R4 gleiche oder verschiedene, mit Halogen substituierte Alkylreste von 1 bis 4 Kohlenstoffatomen und Y3 und Y4 gleiche oder verschiedene Alkoxycarbo- nyl-oder Alkoxycarbonylalkylgruppen bedeuten, oder einer der Substituenten Y3 und Y4 Wasserstoff oder der Methylrest ist, wobei die Alkoxycarbonylgruppe 2 bis 6 und der Alkylrest in der Alkoxycarbonylalkylgmppe 1 bis 4 Kohlenstoffatome aufweist, dadurch gekennzeichnet, dass eine Phosphorverbindung der Formel EMI3.2 mit einem Cumarinderivat der Formel EMI3.3 umsetzt,in welchen Formeln Z ein Halogenatom und M Wasserstoff oder ein Metallatom ist oder sowohl Z als auch M Wasserstoffatome sind.UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass Z ein Chloratom und M Wasserstoff ist.2. Verfahren nach Unteranspruch 1, dadurch gekennzeichnet, dass man die Umsetzung in Gegenwart eines Säurebindungsmittels und eines Katalysators durchführt.3. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass Z Chlor und M Natrium ist.4. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die Umsetzung in Gegenwart eines Säurebindemittels und eines perhalogenierten Kohlenwasserstoffes durchführt.5. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die Umsetzung in Gegenwart eines die Ausgangsstoffe lösenden Mittels durchführt.6. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass sowohl R3 als auch R4 2-Chloräthylreste sind.7. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass O, O-bis- (2-Chlor-äthyl)-O- (4-äthoxycarbo- nyl-cumarin-7-yl)-phosphat hergestellt wird.
