Abstract

"TESTES DE TRANSFERêNCIA DE ENERGIA DE RESSONâNCIA DE FLUORESCêNCIA (FRET) à BASE DE CéLULAS PARA TOXINAS CLOSTRIDIANAS". A presente invenção refere-se a um método para determinar a atividade de toxina clostridiana por (a) contatar com uma amostra uma célula contendo um substrato de toxina clostridiana que inclui um fluoróforo doador; um aceitador tendo um espectro de absorbância soprepondo o espectro de emissão do referido fluoróforo doador; e uma seq³ência de reconhecimento de toxina clostridiana contendo um sítio de clivagem que intervém entre fluoróforo doador e um aceitador, onde a transferência de energia de ressonância é demonstrada entre fluoróforo doador e um aceitador sob as condições apropriadas; (b) excitar o fluoróforo doador; e (c) determinar a transferência de energia de ressonância da célula contatada com relação a uma célula de controle, onde a diferença na transferência de energia de ressonância da célula contatada como comparada com a célula de controle é indicativa de atividade de toxina clostridiana.
