Abstract

Se proveen composiciones y métodos para reducir el nivel de nornicotina y N’-nitrosonornicotina (NNN) en plantas de tabaco y partes de las mismas. Las composiciones comprenden polinucleótidos y polipéptidos aislados para nicotinas demetilasas específicas para la raíz., CYP82E10, y variantes de las mismas, que intervienen en la conversión metabólica de nicotina en nornicotina en estas plantas. Las composiciones de la presente además incluyen plantas de tabaco, o partes de las mismas, que comprenden una mutación en un gen que codifica una nicotina demetilasa CYP82E10, en tanto la mutación da como resultado una expresión o función reducida de la nicotina demetilasa CYP82E10. Además se proveen semillas de estas plantas de tabaco, o progenie de las mismas, y productos de tabaco preparados a partir de las plantas de tabaco de la misma, o de partes de las mismas o progenie de las mismas. Además se proveen métodos de reducción del nivel de nornicotina, o de reducción de la tasa de conversión de nicotina en nornicotina, en una planta de tabaco, o parte de la misma. Los métodos comprenden introducir en el genoma de una planta de tabaco una mutación dentro de por lo menos un alelo de cada uno de por lo menos tres genes de nicotina demetilasa, en tanto la mutación reduce la expresión del gen de nicotina demetilasa, y el primero de estos genes de nicotina demetilasa codifica una nicotina demetilasa específica de la raíz que interviene en la conversión metabólica de nicotina en nornicotina en una planta de tabaco o una parte de la misma. Los métodos son de aplicación en la producción de productos de tabaco que poseen niveles reducidos de nornicotina y su metabolito carcinogénico, NNN, y así un potencial carcinogénico reducido para aquellos individuos que consumen estos productos de tabaco o que están expuestos al humo secundario derivado de estos productos.
