Abstract

La presente invención se refiere a composiciones y métodos para reducir el nivel de nornicotina y N´-nitrosonornicotina (NNN) en plantas de tabaco y partes de planta de las mismas. Las composiciones comprenden polinucleótidos y polipéptidos aislados para nicotina demetilasas, CYP82E10, específicas de raíz y variantes de las mismas, que están involucradas en la conversión metabólica de nicotina a nornicotina en estas plantas. Las composiciones de la invención también incluyen planta de tabaco, o partes de planta de las mismas, que comprenden una mutación en un gen que codifica una nicotina demetilasa CYPS2E10, en donde la mutación da por resultado la expresión o función reducida de la nicotina demetilasa CYP82E10. También se proporcionan semilla de estas plantas de tabaco, o progenie de las mismas, y productos de tabaco preparados a partir de las plantas de tabaco de la invención, o a partir de partes de planta o progenie de las mismas. También se proporcionan métodos para reducir el nivel de nornicotina, o reducir la tasa de conversión de nicotina a nornicotina, en una planta de tabaco, o parte de planta de la misma. Los métodos comprenden introducir en el genoma de una planta de tabaco una mutación dentro de por lo menos un alelo de cada uno de los tres genes de nicotina demetilasa, en donde la mutación reduce la expresión del gen de nicotina demetilasa y en donde un primero de estos genes de nicotina demetilasa codificada una nicotina demetilasa específica de raíz involucrada en la conversión metabólica de nicotina a nornicotina en una planta de tabaco una parte de planta de la misma. Los métodos encuentran uso en la producción de productos de tabaco que tienen niveles reducidos de nornicotina y su metabolito carcinogénico, NNN, y de esta manera potencial carcinogénico reducido para individuos que consumen estos productos de tabaco o expuestos al humo secundario derivado de estos productos.
