Abstract

Métodos, usos y animales para la introgresión de alelos entre animales, incluidos los de SNP. En una forma de realización se introduce un sistema de endocucleasa blanco y una plantilla de HDR en una célula con una falta de coincidencia en la unión de la endonucleasa direccionadora y el sitio blanco. Reivindicación 1: Un método basado en la reparación dependiente de la homología (HDR) para introducir un alelo exógeno en el ADN cromosómico de una célula, que comprende introducir un sistema de endonucleasas dirigidas y un molde para la HDR, que comprende el alelo exógeno, en la célula, donde el sistema de nucleasas dirigidas comprende un componente que puede unirse de manera específica a una secuencia endógena conocida en el ADN cromosómico, donde el sistema de nucleasas dirigidas y el molde para la HDR son útiles para conferirle al ADN cromosómico identidad con la secuencia del molde para la HDR y para introducir el alelo exógeno en lugar de un alelo endógeno en el ADN cromosómico, y donde el sistema de nucleasas dirigidas y/o el molde para la HDR presentan características que son apropiadas para disminuir la unión específica entre el sistema de nucleasas dirigidas y el ADN.
