Abstract

Reivindicación 1: Un método para obtener ediciones genéticas in vitro en una célula o en un embrión de vertebrados en una pluralidad de sitios de ADN cromosómico de interés que comprende introducir en una célula o en un embrión de vertebrados: una primera endonucleasa direccionada dirigida a un primer sitio de ADN cromosómico de interés y un primer templado para una reparación dirigida por homología (HDR) que es homólogo de la primera secuencia del sitio de interés; y una segunda endonucleasa direccionada dirigido a un segundo sitio de ADN cromosómico de interés y un segundo templado de HDR homóloga de la segunda secuencia del sitio de interés, donde la primera secuencia del templado de HDR reemplaza a la secuencia de ADN cromosómico nativa en el primer sitio de interés y la segunda secuencia del templado de HDR reemplaza a la secuencia de ADN cromosómico nativa en la segunda secuencia del sitio de interés. Reivindicación 16: Un método para obtener noqueos de genes multiplexados en una célula o embrión primario de vertebrado que comprende introducir en la célula o el embrión una pluralidad de TALEN dirigidos a diferentes genes blanco en la presencia de templados de HDR que presentan homología con dichos genes blanco diferentes. Reivindicación 17: Un embrión quimérico de vertebrado para obtener células huésped y células donantes que comprende un embrión huésped con una pluralidad de ediciones genéticas de la célula huésped en diferentes sitios de ADN cromosómico, y una célula donante integrada con las células huésped para formar el embrión quimérico. Reivindicación 27: Un animal quimérico de vertebrado para obtener células huésped y células donantes que comprende una pluralidad de ediciones genéticas de la célula huésped en diferentes sitios de ADN cromosómico, y una célula donante integrada con las células huésped para formar el animal quimérico. Reivindicación 31: Un animal vertebrado grande quimérico que comprende células huésped y células donantes, donde las células huésped comprenden por lo menos una edición genética no meiótica de un gen gametogénico o espermatogénico que es complementado por un gen de las células donantes, donde el animal comprende gametas con el genotipo de las células donantes.
