Abstract
Métodos y composiciones para la integracion dirigida de una secuencia exogena en un sitio blanco predeterminado del genoma de plantas. Reivindicacion 1: Un vector donante que comprende una primera y segunda secuencia de ADN; en donde la primera secuencia es homologa de una tercera secuencia y la segunda secuencia es homologa de una cuarta secuencia; en donde la tercera y cuarta secuencias son secuencias de ADN cromosomico; y en donde los bordes cercanos de tercera y cuarta secuencias están separados por al menos 1 par de nucleotidos. Reivindicacion 2: El vector de la reivindicacion 1, en donde la tercera y cuarta secuencias son secuencias endogenas. Reivindicacion 3: El vector de la reivindicacion 1 o reivindicacion 2, en donde al menos una entre la primera o segunda secuencias tiene una longitud de 100 nucleotidos. Reivindicacion19: Un método para expresar una proteína en una célula vegetal, donde dicho método comprende los pasos de (a) poner la célula en contacto con un vector direccionador de acuerdo con la reivindicacion 8; y (b) expresar una o mas nucleasas en la célula, en donde dichas una o más nucleasas clivan el ADN cromosomico dentro de los 3 pares de kilobases de ya sea la tercera o cuarta secuencia; de modo tal que el clivaje de ADN cromosomico en el paso (b) estimula la incorporacion del vector direccionador en el genoma por recombinacion de homologos.
