また、特定のペプチドを濃縮する方法としては、逆浸透膜や限外濾過膜を用いて特定のペプチドを得る方法(特許文献2、3)や、カラム樹脂に吸着させて溶出する方法(特許文献4)などが考えられるが、いずれも工業的な製造では高額な設備投資が必要となる。
このように、特殊な膜処理や分画処理なしに、安価にGTWY含量の高い素材を効果的に製造する方法は未だ知られていない。

そのうちでも、特に、本発明の製造方法は、GTWY配列を含むタンパク質素材をアスペルギルス・メレウス由来のプロテアーゼで加水分解する際の酵素反応時のpH条件を制御することでホエイタンパク質からのGTWYの生成量を高めると共に、GTWYの分解を抑制することができる方法である。したがって、本製造方法によれば風味が良好でGTWY含量の高い素材を提供することができる。以下、本発明について詳細に説明する。
本発明の原料としては、ホエイタンパク質含有素材が用いられ、このうちでもGTWY配列を含むタンパク質素材が好ましく用いられる。GTWY配列を含むタンパク質素材とは、グリシン-スレオニン-トリプトファン-チロシンからなるアミノ酸配列のペプチド(GTWY)をその構成の一部に含むタンパク質であり、典型的にはβラクトグロブリンを含有する素材が挙げられる。GTWY配列を含むタンパク質素材には、タンパク質の分解物も含まれる。βラクトグロブリンを含有する素材としては、例えば、牛乳、乳製品、ホエイ、ホエイ粉末、ホエイタンパク質濃縮物(WPC)、ホエイタンパク質分離物(WPI)などが挙げられる。本発明のタンパク質の加水分解反応を行う際の原料の濃度は特に制限するものではないが、通常の製造工程を考慮するとタンパク質量として0.01%~50%(w/w)程度が好ましく、より好ましくは0.1%~20%程度が好ましい。
本発明においてホエイタンパク質含有素材の加水分解に用いるプロテアーゼとしては、アスペルギルス・メレウス由来のプロテアーゼが好ましい。アスペルギルス・メレウス由来のプロテアーゼとしては、例えば、プロテアーゼ P「アマノ」3SD(天野エンザイム)、プロテアーゼ(Protease from Aspergillus melleus (シグマ社))、スミチームMP(新日本化学工業)などの市販の酵素が知られている。酵素の添加量は特に制限するものではないが、適当な反応時間と製造コスト等を考慮して、反応液中の酵素濃度が0.001%~10%程度が好ましく、より好ましくは0.01%~5%、更に好ましくは0.01%~2%である。
本発明者らの検討により、酵素反応中のpHが中性~アルカリ性の条件下において、GTWYペプチドなどの特定ペプチドが効率よく生成し、かつその分解が抑えられることが見出された。一般に、タンパク質をプロテアーゼにより加水分解すると、生成したペプチドの影響によりpHが急激に低下することが知られており、素材製造のために数%以上のタンパク質溶液を加水分解する場合には、通常の酵素反応試験で用いる0.01~0.2M程度の緩衝液ではpHを維持することは困難である。よって、反応中のpHが中性~アルカリ性で維持されるように工夫して酵素反応を行う必要がある。
反応開始時のpHとしては、pH7.0より大きいことが好ましく、より好ましくはpH7.5以上である。好ましい範囲としてpH7.0より大きく10.0以下であることが好ましく、より好ましくはpH7.5以上9.5以下である。
また、反応中のpHとしては、6.6以上を保持することが好ましく、6.7以上がより好ましく、7.0以上がよりいっそう好ましい。好ましい範囲としてpH6.6以上10.0以下であることが好ましく、より好ましくはpH6.7以上9.5以下であり、さらにいっそう好ましくは7.0以上9.0以下である。
反応中のpHを調整する方法としては、酵素反応開始時のpHをNaOH、KOH、(NH4)CO3などにより中性~アルカリ性に調整してから反応を開始してもよいし、反応中に間欠的にアルカリ性溶液を添加してpHをアルカリ性に戻してもよいし、中和剤としてアルカリ性溶液を用いてpHを一定に保ちながら反応を行ってもよい。
また、特定ペプチドが生成後、特定ペプチドの顕著な分解が起こらない範囲内であれば、酵素反応時間を延長したり、温度等の条件を変えて反応したり、他の酵素を添加して反応してもよい。
本発明の製造方法を用いることにより、βラクトグロブリンなどのホエイタンパク質を含む原料から特定ペプチドを効率よく回収することが可能となる。例えば特定ペプチドが、GTWYの場合、原料に含まれるGTWY配列を含むペプチドのうち、15~90%のGTWYを回収することが可能である。また、本発明の製造方法によれば、GTWYの分解が抑制されるため、その分解産物であるWYの生成が抑制され、WYに対するGTWYのモル比率(以下、単にGTWY/WYのモル比率、GTWY/WYモル比率などということがある)を高めることができる。本発明のGTWY含有組成物のGTWY/WYのモル比率は約2以上であり、好ましくは3以上であり、さらに好ましくは4以上である。GTWY/WYのモル比率が2未満である場合は、生成したGTWYの3分の1以上が分解を受けていることになり、GTWYを得る方法として効率的な方法ではない。
また、本発明のペプチド組成物の平均分子量は、500以上であることが好ましく、700以上がよりいっそう好ましい。一般的にタンパク質は、プロテアーゼにより分解を受けることで苦味や旨味などの呈味を示すペプチドやアミノ酸を生成することが知られており、分子量500未満のアミノ酸やジペプチド、トリペプチドが多いほどその呈味は強く感じられる。
本発明の製造方法を用いることで、ホエイタンパク質の過剰な分解を抑制しつつ、GTWYなどの特定のペプチドを生成することが可能となるため、平均分子量が500以上の風味の良いペプチド組成物を得ることが可能である。
ペプチド組成物がGTWY含有組成物の場合、好ましくは、GTWY/WYモル比率が2以上で、かつ、平均分子量が500以上であり、さらにいっそう好ましくは、GTWY/WYモル比率が3以上で、かつ、平均分子量が500以上であり、もっとも好ましくはGTWY/WYモル比率が4以上で、かつ、平均分子量が700以上である。
本発明のGTWY含有組成物を用いた素材、飲食品、医薬品等については、含有するGTWY量とWY量を測定し、その比率を求めることで検出が可能である。
また、本発明のペプチド組成物は、飲食品に配合して使用することができる。飲食品に配合する場合は、本発明のペプチド組成物をそのまま配合することができる。また、当該ペプチド組成物を透析膜やイオン交換樹脂等によって脱塩を行ったもの、さらに、凍結乾燥や噴霧乾燥等によって乾燥を行い粉末化したものも配合することができる。
本発明のペプチド組成物を配合した飲食品としては、清涼飲料、乳飲料、発酵乳、チーズ、バター、スキムミルク、ジュース、ゼリー、パン、アイスクリーム、麺、ソーセージ、育児用調製乳や離乳食等を挙げることができる。
また、本発明のペプチド組成物は医薬品や機能性食品として利用することもできる。この場合、本発明のペプチド組成物は、錠剤、カプセル剤、細粒剤、散剤、丸剤、トローチ、舌下剤または液剤等の経口投与用の製剤に加工して利用することができる。
1.試験方法
βラクトグロブリンを基質として、酵素反応の初期pH(反応開始時のpH)がGTWYの生成に与える影響を調べた。βラクトグロブリン(SIGMA)を8%(w/w)となるように20mMの緩衝液に溶解して、pHを6.6、6.9、7.1、7.5、7.8に調整した。50℃に加温した後、プロテアーゼ P「アマノ」3SDを0.05%(w/v)となるように添加して反応を開始し、反応30分後、1時間後にサンプルを採取して反応液のpHとGTWY含量を測定した。GTWYの測定は、阿野らの方法(非特許文献2)に従い、液体クロマトグラフ‐タンデム型質量分析装置(LC-MS/MS) にて定量した。
反応液中のGTWY量の経時変化を図1に、pHの経時変化を図2に示す。
初期pHが6.6~7.1の水準では、反応開始30分後をピークにGTWYが生成してその後減少しており、GTWYの生成と共に分解が起きていることが確認された。また、反応開始30分後にpHが急激に低下してpHが6.0~6.5程度となっていた。
一方、初期pHが7.5~7.8の水準では、反応開始30分後のGTWY量が多く、反応開始1時間後でもGTWY量が維持されていた。反応中のpHは、反応開始30分後で低下したが、pHが6.6以上を維持していた。
以上の結果から、プロテアーゼによる酵素反応液中のpHを6.6以上で保持することにより、GTWYの生成を高め、かつ、GTWYの分解を抑えられることが確認された。
1.試験方法
WPCを基質として、酵素反応の初期pHがGTWYの生成に与える影響を調べた。市販のWPCを13%(w/w)となるように50℃に加温したイオン交換水に溶解し、NaOHにてpHを6.5~9.0に調整した。プロテアーゼ P「アマノ」3SDを0.1%(w/v)となるように添加して反応を開始し、反応開始1時間後にサンプルを採取して反応液のpHとGTWY含量を測定した。
反応後のGTWY量とpHを表1に示す。初期pHが6.5、7.0の水準では、反応終了後のGTWY量が79.8、148.8μg/mlと低く、反応終了後のpHは6.26~6.53に低下していた。
一方、初期pHが7.7~9.0の水準では、GTWY量が236.8~471.7μg/mlと高く、反応後のpHも6.73~7.02とpH6.6以上を維持していた。
以上の結果から、プロテアーゼによる酵素反応開始時のpHを7.0より大きいアルカリ側にすることにより、酵素反応中のpHを高く維持することができ、GTWYの生成量を高められることが確認された。
1.試験方法
GTWYを基質として、酵素反応時のpH条件がプロテアーゼによるGTWYの分解へ与える影響を調べた。pHが6.4~8.9となるように調製した20mMリン酸バッファーに、化学合成したGTWYを0.2mg/mlとなるように添加し、プロテアーゼ P「アマノ」3SDを0.01%添加して50℃で2時間反応した。GTWYを基質とした場合は、タンパク質の分解によるpH低下は起こらず、反応前後でpHの変化は認められなかった。反応後に残存するGTWYおよびその分解産物であるWYの量を高速液体クロマトグラフ(HPLC)にて測定した。
反応開始時のGTWY量、反応後のGTWY量およびWY量の測定結果と、GTWYの分解率((反応開始時のGTWY量-反応後のGTWY量)/反応開始時のGTWY量)を表2に示す。
反応中のpHが6.4の条件では、反応2時間でGTWYの50%程度が分解され、その分解産物であるWYが81.3μM生成した。
また、反応中のpHが7.4の条件では、反応2時間でGTWYの40%程度が分解され、その分解産物であるWYが57.3μM生成した。
一方、pHが8.1~8.9の条件では、GTWYの分解が9~23%程度に抑制されており、WYの生成量も低かった(8.3~37.5μM)。
以上の結果から、プロテアーゼによる酵素反応液中のpHをアルカリ側に保持することによりGTWYの分解を抑制することができることが見出された。
1.試験方法
GTWY配列を含むタンパク質素材としてWPCを用いて、酵素反応時のpHをアルカリ側で一定に保持した条件で、GTWY含有組成物を試作した。
市販のWPCを16%(w/w)となるようにイオン交換水に溶解し、50℃に加温した後、NaOHにてpHを7.5に調整した(WPC溶液)。前記WPC溶液に、プロテアーゼ P「アマノ」3SDを0.1%(w/v)となるように添加して反応を開始した。ジャーファメンター(全容2L微生物培養システムBMJ-02NC;ABLE社)を用いて、pHが7.5に維持されるように中和剤(NaOH)を加えながら50℃で5時間反応を行った。反応開始1時間後および5時間後にサンプルを採取し、反応液中のGTWY量を測定した。
反応液中のGTWY量は、1時間後で701.6μg/ml、5時間後で851.6μg/mlとなった。これらを凍結乾燥したところ、GTWY含量はそれぞれ4.4mg/g、5.3mg/gとなった。したがって、GTWYを高含有する組成物が得られた。
1.試験方法
(1)本発明の試験方法
GTWY配列を含むタンパク質素材としてWPCを用いて、酵素反応開始時のpHをアルカリ側に高めた条件で、GTWY高含有素材を試作した。試作は2水準行った(表3)。市販のWPCを16%(w/w)となるようにイオン交換水に溶解し、50℃に加温した後、NaOHにてpHを8.5に調整した。プロテアーゼ P「アマノ」3SDを水準1は0.06%(w/v)、水準2は0.1%(w/v)となるように添加し、水準1は48℃で2時間、水準2は50℃で30分酵素反応を行った。90℃で20分間加温して酵素を失活させた後、凍結乾燥し、GTWY高含有素材1および2を得た。
比較対象として、特許文献1の方法を参考にして既存の製造方法でGTWY素材を製造した。市販のWPC素材を10%(w/w)となるようにイオン交換水に溶解し、50℃に加温した後、NaOHにてpHを7.0に調整した(WPC溶液)。前記WPC溶液にプロテアーゼ P「アマノ」3SDを0.2%(w/v)となるように添加し、50℃にて5時間酵素反応を行った。その後、反応液を90℃で20分間加温して酵素を失活させた。この反応液を凍結乾燥し、既存製法のGTWY含有素材(比較例のGTWY含有素材)を得た。
各素材中のGTWY含量、WY含量、GTWY/WYのモル比率、平均分子量を表3に示す。GTWY/WYのモル比率は、GTWYの分子量525.54、WYの分子量を367.41として算出した。
本発明のGTWY高含有素材中のGTWY含量は、水準1は5.1mg/g、水準2は3.4mg/gと非常に高く、素材中のWY含量は、水準1は0.2mg/g、水準2は0.1mg/gとどちらも低かった。そのため、GTWY/WYのモル比率は、水準1は17.0、水準2は23.5であり、どちらも高値を示した。また、GTWY高含有素材の平均分子量は、水準1が786Da、水準2が1024Daであった。
比較例のGTWY素材中のGTWY含量は0.8mg/g、WY含量は0.3mg/gであり、GTWY/WYのモル比率は1.9であった。当該素材の平均分子量は309Daであった。酵素反応開始30分後のpHが6.2、5時間後のpHが6.0であったことから、酵素反応中にGTWYの分解が起こっていると考えられた。
以上の結果から、本発明技術を用いることにより、より短い時間でGTWY含量、GTWY/WYのモル比率および平均分子量が高い素材を提供できることが示された。
1.試験方法
実施例5で調製したGTWY高含有素材2と、既存製法(比較例)で製造したGTWY素材について、苦味の比較を行った。9名のパネラーに素材粉末20mgを舌上に置いて味わってもらい、その苦味を以下の基準に示す4点法で評価した。

3点 苦味を強く感じる
2点 苦味を感じる
1点 苦味をわずかに感じる
0点 苦味を全く感じない
10名の平均点を比較した結果、比較例のGTWY素材では平均点が2.4と苦味が強かったのに対し、本発明のGTWY高含有素材は平均点が1.4となり、苦味が低減されていることが確認された。
1.試験方法
実施例5で調製したGTWY高含有素材を添加した牛乳の官能評価を行った。市販の牛乳にGTWY高含有素材2を0.67%添加したものを調製した(GTWY含量が2.3mg/100g)。比較対象として、実施例5の比較例で調製したGTWY素材を1.45%添加した牛乳サンプル(GTWY含量が1.1mg/100g)と、GTWY素材を加えない牛乳を用意した。10名のパネラーにサンプル3品を試飲してもらい、以下に示す4点法で苦味およびおいしさを評価した。

1点 ややおいしくない

10名の平均点を比較した結果、比較例で調製したGTWY素材を加えたサンプルでは苦味が感じられたほか、おいしさの評価が低下していた。一方、GTWY高含有素材2を加えたサンプルでは苦味がほとんど感じられず、GTWY素材を添加しないサンプルとのおいしさの評価の差は僅かであった。このことから、GTWY高含有素材を用いることで製品の風味を損なわずにGTWYを高濃度摂取することが可能であることが確認された。
1.試験方法
当該特許技術に使用可能な酵素を調べるため、実施例1で用いたプロテアーゼ P「アマノ」3SDとは異なるアスペルギルス・メレウス由来のプロテアーゼ(Protease from Aspergillus melleus (シグマ社))を用いて、酵素反応の初期pHがGTWYの生成に与える影響を調べた。
市販のWPCを13%(w/w)となるように50℃に加温したイオン交換水に溶解し、NaOHにてpHを6.5~9.0に調整した。Protease from Aspergillus melleus (シグマ社)を0.1%(w/v)となるように添加して反応を開始し、反応開始2時間後(反応後)にサンプルを採取して反応液のpHとGTWY含量を測定した。
初期pHと反応後のGTWY量とpHを表6に示す。プロテアーゼ P「アマノ」3SDを用いた場合と同様に、初期のpHがアルカリ側となるほどGTWYの生成量を高められることが確認された。したがって、本件発明において、アスペルギルス・メレウス由来のプロテアーゼは種類の制限なく広く利用可能であることが確認された。
実施例5で調製したGTWY高含有素材2を10g、ラクトース33.5g、可溶性でんぷん56.0g、ステアリン酸マグネシウム0.5gを混和後、常法により造粒し、カプセルに充填して、本発明のGTWY含有カプセル剤を製造した。
690gの脱イオン水に実施例5で調製したGTWY高含有素材2を10g溶解し、40℃まで加熱後、ウルトラディスパーサー(ULTRA-TURRAX T-25;IKAジャパン社製)にて9,500rpmで20分間撹拌混合した。マルチトール100g、酸味料2g、還元水飴20g、香料2g、脱イオン水176gを添加した後、100mlのガラス瓶に充填し、95℃、15秒間殺菌後、密栓し、本発明のGTWY含有飲料10本(100ml入り)を調製した。
実施例5で調製したGTWY高含有素材2を100g、牛乳9.9kgを混合し、40℃まで加熱後、TKホモミクサー(TK ROBO MICS;特殊機化工業社製)にて、6,000rpmで10分間撹拌混合した。130℃で2秒間、加熱殺菌した後、10℃以下まで冷却して本発明のGTWY含有乳飲料10kgを製造した。
実施例5で調製したGTWY高含有素材2を100g、脱脂粉乳1600g、グルコース300g、脱イオン水7700gを混合し、95℃で2時間保持することで加熱殺菌した。これを37℃まで冷却し、乳酸菌スターター(Lb.casei)を300g植菌し、攪拌混合後、37℃に保持したインキュベーター内でpH4.0まで発酵させた。pH4.0に到達後10℃以下まで冷却し、本発明のGTWY含有発酵乳10kgを製造した。
ゴーダチーズ9.5kg、チェダーチーズ9.5kg、実施例5で調製したGTWY高含有素材2を100g、クエン酸ナトリウム200g、脱イオン水700gを混合し、85℃で乳化した。乳化後にチーズをカルトンに充填して2昼夜、5℃で冷却して、本発明のGTWY含有チーズ20kgを製造した。
ナタネ硬化油4.5kg、レシチン40g、モノグリセリン脂肪酸エステル10g、ソルビタン脂肪酸エステル10gを混合し、油相を調製した。次に、実施例5で調製したGTWY高含有素材2を100g、脱脂粉乳308g、カゼインナトリウム10g、シュガーエステル20g、リン酸塩10g、キサンタンガム5g、脱イオン水4.987kgを混合して、水相を調製した。水相を65℃に加温し、70℃に加温した油相を少量ずつ攪拌しながら添加し、TKホモミクサー(TK ROBO MICS;特殊機化工業社製)にて、6,000rpmで10分間撹拌混合した。これを均質機で均質処理して本発明のGTWY含有クリーム10kgを製造した。
はちみつ2000g、実施例5で調製したGTWY高含有素材2を100g、脱脂粉乳770g、マスカルポーネ300g、液状水飴700g、グラニュー糖500g、生クリーム250g、バター200g、加糖卵黄400g、ゼラチン40g、寒天15g、ローカストビーンガム120g、脱イオン水4605gを混合して、プリンミックスとした。このプリンミックスをTKホモミクサー(TK ROBO MICS;特殊機化工業社製)にて、6,000rpmで10分間撹拌混合し、60℃に加熱して溶解した後、100gずつ容器へ充填して冷却することで、本発明のGTWY含有プリン100個を製造した。
実施例5で調製したGTWY高含有素材2を1kg、脱脂粉乳8.3kg、脱イオン水90kgを混合し、40℃まで加熱後、TKホモミクサー(TK ROBO MICS;特殊機化工業社製)にて、6,000rpmで10分間撹拌混合した。この溶液を噴霧乾燥して本発明のGTWY含有粉乳約10kgを製造した。
清涼飲料の製造工程中で酵素処理を行うことにより、GTWY含有清涼飲料を試作した。市販のWPC16gを蒸留水84gに溶解し、50℃に加温した後、NaOHにてpHを8.5に調整した。プロテアーゼ P「アマノ」3SDを0.1%(w/v)となるように添加し、50℃にて30分酵素反応を行った。別の容器で、脱脂粉乳10g、砂糖100g、クエン酸5g、クエン酸三ナトリウム2.5g、ペクチン5gを蒸留水778mlに溶解しておき、酵素処理したWPC溶液と混和した後、90℃で20分間加温殺菌して、100mlのプラスチックボトルに充填した。
また、本発明のGTWY含有組成物は、タンパク質の過剰な分解を抑制しつつGTWYを効率的に生成しているため、平均分子量が500以上であり、風味が良好であるという利点がある。このため、飲食品に添加しても飲食品本来の風味に悪影響を与えることなくペプチド含量を高めることが出来、利用価値が高い。
Claims (16)
- ホエイタンパク質含有素材にプロテアーゼを作用させてペプチド組成物を製造する方法であって、前記プロテアーゼを作用させる反応開始時の反応液中のpHが7.0より大きいことを特徴とする前記製造方法。
- 反応時間を5分以上5時間未満とする請求項1に記載の製造方法。
- ホエイタンパク質がβ-ラクトグロブリンである請求項1又は2に記載の製造方法。
- プロテアーゼがアスペルギルス・メレウス由来である請求項1~3のいずれかに記載の製造方法。
- ペプチド組成物がGTWY含有組成物である請求項1~4のいずれかに記載の製造方法。
- 前記ペプチド組成物がGTWY含有組成物であって、平均分子量が500以上であり、WYに対するGTWYのモル比率が2.0以上である請求項1~5のいずれかに記載の製造方法。
- ホエイタンパク質含有素材にプロテアーゼを作用させてペプチド組成物を製造する方法であって、プロテアーゼを作用させる反応液中のpHを6.6以上に保持することを特徴とする前記製造方法。
- 反応時間を5分以上5時間未満とする請求項7に記載の製造方法。
- ホエイタンパク質がβ-ラクトグロブリンである請求項7又は8に記載の製造方法。
- プロテアーゼがアスペルギルス・メレウス由来である請求項7~9のいずれかに記載の製造方法。
- ペプチド組成物がGTWY含有組成物である請求項7~10のいずれかに記載の製造方法。
- 前記ペプチド組成物がGTWY含有組成物であって、平均分子量が500以上であり、WYに対するGTWYのモル比率が2.0以上である請求項7~11のいずれかに記載の製造方法。
- ホエイタンパク質含有素材にアスペルギルス・メレウス由来のプロテアーゼを作用させてGTWY含有組成物を製造する方法におけるGTWYの分解抑制方法であって、プロテアーゼを作用させる反応開始時の反応液中のpHを7.0より大きくすることによりGTWYの分解を抑制することを特徴とする前記方法。
- ホエイタンパク質由来のペプチド組成物であって、平均分子量が500以上であるペプチド組成物。
- 前記ペプチド組成物がGTWY含有組成物であって、WYに対するGTWYのモル比率が2.0以上である請求項14に記載のペプチド組成物。
- 請求項15に記載のペプチド組成物を含む飲食品であって、飲食品中のWYに対するGTWYのモル比率が2.0以上である飲食品。
