本発明の一実施形態は、式(1)で表される化合物又はその医薬上許容可能な塩を含む、ETA受容体拮抗剤に関する。エンドセリン受容体には、ETA受容体及びETB受容体が知られているが、ETB受容体を阻害すると血中エンドセリンの上昇、末梢性浮腫の発生等の問題が生じることがある。本実施形態のETA受容体拮抗剤は、好ましくは、エンドセリンB受容体を対照として、エンドセリンA受容体を選択的に阻害することができる。

血管攣縮としては、例えば、脳血管攣縮、及び血管攣縮性狭心症を挙げることができる。
6-ブロモ-5-メチルイソベンゾフラン-1(3H)-オン
3-ブロモ-4-メチル安息香酸(1.5g、7.01mmol)及び1,1-ジブロモメタン(20mL)の混合物にりん酸水素二カリウム(3.6g、21.0mmol)及び酢酸パラジウム(II)(157mg、0.70mmol)を室温で加え、反応混合物を140℃で48時間攪拌した。反応混合物を室温とし、セライトを用いて酢酸エチルで洗浄しながら濾過した。濾液を減圧下溶媒留去し、残渣をエタノールで粉砕洗浄し、標記化合物(1.3g)を得た。

6-ブロモ-1,1,5-トリメチル-1,3-ジヒドロイソベンゾフラン
6-ブロモ-5-メチルイソベンゾフラン-1(3H)-オン(1.00g、4.41mmol)及びテトラヒドロフラン(10mL)の混合物にメチルマグネシウムヨージド(3.0Mジエチルエーテル溶液、4.5mL、13.5mmol)を-15℃で加え、同温で2時間攪拌した。反応混合物に氷冷水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(615mg)を得た。

4,5-ジメチルイソオキサゾール-3-アミン(1.0g、8.92mmol)及びピリジン(20.0mL)の混合物に、4-ジメチルアミノピリジン(109mg、0.892mmol)及び2-クロロピリジン-3-スルホニルクロリド(2.84g、13.4mmol)を0℃でゆっくり加え、反応混合物を50℃で16時間攪拌した。反応混合物に氷冷水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させた。減圧下で溶媒を留去し、標記化合物(1.50g)を粗体として得た。これ以上の精製をすることなく、次の反応に用いた。

アルゴン雰囲気下、2-クロロ-N-(4,5-ジメチルイソオキサゾール-3-イル)-N-(メトキシメチル)ピリジン-3-スルホンアミド(400mg、1.21mmol)、トリエチルアミン(0.50mL、3.62mmol)、トリイソプロピルシリルアセチレン(440mg、2.41mmol)及びTHF(5.00mL)の混合物に、ビス(トリフェニルホスフィン)パラジウム(II)ジクロリド(84.6mg、0.121mmol)及びヨウ化銅(I)(11.5mg、0.0362mmol)を加え、反応混合物を55℃で6時間攪拌した。反応混合物に氷冷水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下で溶媒を留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(0.150g)を得た。

N-(4,5-ジメチルイソオキサゾール-3-イル)-N-(メトキシメチル)-2-((トリイソプロピルシリル)エチニル)ピリジン-3-スルホンアミド(150mg、4.3mmol)及びTHF(3.00mL)の混合物に、テトラブチルアンモニウムフルオリド(1mol/Lテトラヒドロフラン溶液、0.942mL、12.9mmol)を0℃でゆっくり加え、反応混合物を室温で2時間攪拌した。反応混合物に氷冷水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下で溶媒を留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(0.060g)を得た。

N-(4,5-ジメチルイソオキサゾール-3-イル)-N-(メトキシメチル)-2-((3,3,6-トリメチル-1,3-ジヒドロイソベンゾフラン-5-イル)エチニル)ピリジン-3-スルホンアミド(90mg、0.18mmol)及びメタノール(5mL)の混合物に6mol/L塩酸(1mL)を0℃で加え、反応混合物を50℃で4時間攪拌した。反応混合物を室温とし、氷冷水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣を高速液体クロマトグラフィー(Omega Ps C18(250x21.2mm)5μm、0.1%トリフルオロ酢酸水溶液/アセトニトリルーメタノール(1:1)混合液)により精製し、標記化合物(30mg)を得た。

2-ヒドロキシ-5-メチルベンズアルデヒド(10g、73.5mmol)及びエタノール(50mL)の混合物にアセトヒドラジド(5.44g、73.5mmol)を室温でゆっくり加え、反応混合物を90℃で16時間攪拌した。反応混合物を減圧下溶媒留去し、残渣をヘキサンで洗浄し、標記化合物(10g)を粗体として得た。これ以上の精製をすることなく、次の反応に用いた。

1-(2-(ヒドロキシメチル)-4-メチルフェニル)エタン-1-オール(1.6g、9.64mmol)及びテトラヒドロフラン(16mL)の混合物に、60%水素化ナトリウム(1.07g、11.2mmol)を0℃でゆっくり加え、同温で30分間攪拌した。反応混合物にりん酸トリメチル(3.5g、24.1mmol)を室温で加え、同温で16時間攪拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(1.0g)を得た。

1,5-ジメチル-1,3-ジヒドロイソベンゾフラン(700mg、4.73mmol)及びジクロロメタン(7mL)の混合物に、N-ブロモスクシンイミド(842mg、4.73mmol)及びトリフルオロメタンスルホン酸(700mg、4.73mmol)を0℃で加え、室温で2時間攪拌した。反応混合物に氷冷水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(0.7g)を得た。

アルゴン雰囲気下、N-(4,5-ジメチルイソオキサゾール-3-イル)-2-エチニル-N-(メトキシメチル)ピリジン-3-スルホンアミド(0.7g、2.18mmol)及びN,N-ジメチルホルムアミド(5mL)の混合物に6-ブロモ-1,5-ジメチル-1,3-ジヒドロイソベンゾフラン(0.54g、2.40mmol)、よう化銅(I)(83mg、0.436mmol)、トリエチルアミン(0.6mL、4.36mmol)及びテトラキス(トリフェニルホスフィン)パラジウム(0)(250mg、0.218mmol)を加え、反応混合物を80℃で4時間攪拌した。反応混合物を室温とし、氷冷水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣を高速液体クロマトグラフィー(Luna C8(250x50mm)10μm、0.1%ギ酸水溶液/アセトニトリル)により精製し、標記化合物(50mg)を得た。

5-メチル-1,3-ジヒドロイソベンゾフラン(1.00g、7.45mmol)、メタノール(5.00mL)及びエタノール(5.00mL)の混合物に硫酸銀(4.65g、14.9mmol)及びヨウ素(1.89g、14.9mmol)を0℃でゆっくり加え、同温で1時間攪拌した。反応混合物に氷冷水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下で溶媒を留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(0.60g)を得た。

アルゴン雰囲気下、N-(4,5-ジメチルイソオキサゾール-3-イル)-2-エチニル-N-(メトキシメチル)ピリジン-3-スルホンアミド(2.50g、7.78mmol)、5-ヨード-6-メチル-1,3-ジヒドロイソベンゾフラン(2.23g、8.56mmol)、トリエチルアミン(2.19mL、15.6mmol)及びトルエン(30.0mL)の混合物に、ビス(トリフェニルホスフィン)パラジウム(II)ジクロリド(899mg、0.778mmol)及びヨウ化銅(I)(148mg、0.778mmol)を加え、反応混合物を65℃で6時間攪拌した。反応混合物に氷冷水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下で溶媒を留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(1.60g)を得た。

5-クロロ-6-ヨードイソベンゾフラン-1(3H)-オン(200mg、0.680mmol)、ジクロロメタン(30mL)の混合物にジイソブチルアルミニウムヒドリド(20%トルエン溶液、1.7mL、2.0mmol)を-78℃で加え、同温で2時間攪拌した。反応混合物に飽和塩化アンモニウム水溶液を加え、セライトを用いて酢酸エチルで洗浄しながら濾過した。濾液を水、および飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(70mg)を得た。

1,1,5-トリメチル-1,3-ジヒドロイソベンゾフラン(0.60g、3.7mmol)とジクロロメタン(10mL)の混合物に、N-ヨードスクシンイミド(1.60g、7.40mmol)、トリフルオロメタンスルホン酸(1.10g、7.40mmol)を0℃でゆっくり加え、同温で2時間攪拌した。反応混合物に10%チオ硫酸ナトリウム水溶液を加え、ジクロロメタンで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(0.30g)を得た。

4-クロロ-5-メチルイソオキサゾール-3-アミン(2.98g、22.6mmol)とピリジン(50mL)との混合物に、4-ジメチルアミノピリジン(0.69g、5.66mmol)、2-クロロピリジン-3-スルホニル クロリド(4.0g、18.8mmol)を0℃でゆっくり加え、反応混合物を55℃で16時間攪拌した。反応混合物に氷冷水を加え、酢酸エチルで抽出した。有機層を2N塩酸、および飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させた。減圧下溶媒留去し、標記化合物(1.90g)を粗体として得た。これ以上の精製をすることなく、次の反応に用いた。

(E)-N’-(2-ヒドロキシ-4-メチルベンジリデン)アセトヒドラジド(4.6g、24mmol)、テトラヒドロフラン(50mL)の混合物に酢酸鉛(IV)(11.7g、26.3mmol)を0℃で加え、反応混合物を室温で16時間攪拌した。セライトを用いて酢酸エチルで洗浄しながら濾過し、濾液を飽和炭酸水素ナトリウム水溶液および飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(2.0g)を得た。

1,6-ジメチル-1,3-ジヒドロイソベンゾフラン
1-(2-(ヒドロキシメチル)-5-メチルフェニル)エタン-1-オール(0.90g、5.4mmol)とテトラヒドロフラン(10mL)の混合物に、60%水素化ナトリウム(0.52g、13.6mmol)を0℃でゆっくり加え、同温で30分間攪拌した。反応混合物にりん酸トリメチル(1.5mL、13.6mmol)を室温で加え、同温で16時間攪拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(0.6g)を得た。

5-ブロモ-1,6-ジメチル-1,3-ジヒドロイソベンゾフラン
1,6-ジメチル-1,3-ジヒドロイソベンゾフラン(600mg、4.05mmol)とジクロロメタン(10mL)の混合物に、N-ブロモスクシンイミド(721mg、4.05mmol)、トリフルオロメタンスルホン酸(1.8g、12.2mmol)を0℃で加え、室温で2時間攪拌した。反応混合物に氷冷水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)により精製し、標記化合物(0.3g)を得た。

2-((1,6-ジメチル-1,3-ジヒドロイソベンゾフラン-5-イル)エチニル)-N-(4,5-ジメチルイソオキサゾール-3-イル)-N-(メトキシメチル)ピリジン-3-スルホンアミド
アルゴン雰囲気下、N-(4,5-ジメチルイソオキサゾール-3-イル)-2-エチニル-N-(メトキシメチル)ピリジン-3-スルホンアミド(0.40g、1.24mmol)、N,N-ジメチルホルムアミド(5mL)の混合物に5-ブロモ-1,6-ジメチル-1,3-ジヒドロイソベンゾフラン(0.337g、1.49mmol)、よう化銅(I)(47mg、0.249mmol)、トリエチルアミン(0.36mL、2.49mmol)、テトラキス(トリフェニルホスフィン)パラジウム(0)(143mg、0.124mmol)を加え、反応混合物を80℃で4時間攪拌した。反応混合物を室温とし、水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣を高速液体クロマトグラフィー(Luna C18(250x50mm)10μm、0.1%ギ酸水溶液/アセトニトリル)により精製し、標記化合物(25mg)を得た。

2-((1,6-ジメチル-1,3-ジヒドロイソベンゾフラン-5-イル)エチニル)-N-(4,5-ジメチルイソオキサゾール-3-イル)ピリジン-3-スルホンアミド
2-((1,6-ジメチル-1,3-ジヒドロイソベンゾフラン-5-イル)エチニル)-N-(4,5-ジメチルイソオキサゾール-3-イル)-N-(メトキシメチル)ピリジン-3-スルホンアミド(30mg、0.064mmol)、メタノール(5mL)の混合物に6mol/L塩酸(0.5mL)を0℃で加え、反応混合物を50℃で4時間攪拌した。反応混合物を室温とし、氷冷水を加え、酢酸エチルで抽出した。有機層を飽和炭酸水素ナトリウム水溶液、および飽和食塩水で洗浄し、硫酸ナトリウムで乾燥させ、減圧下溶媒留去した。残渣を高速液体クロマトグラフィー(Luna C18(250x50mm)10μm、0.1%ギ酸水溶液/アセトニトリル)により精製し、標記化合物(5mg)を得た。

エンドセリンA受容体の阻害作用を以下の方法で検討した。
ヒトEndothelin A受容体(Accession Number NP_001948.1)を強制発現させたCHO-K1-mt aequorin細胞を抗生剤無添加の培地で18時間培養した後、PBS-EDTA(5mM EDTA)で処理し、遠心分離後(2分、405×g、室温)アッセイバッファー(DMEM/HAM‘s F12 with HEPES+0.1% BSA protease free)に懸濁させた。
エンドセリンB受容体の阻害作用を以下の方法で検討した。
ヒトEndothelin B受容体(Accession Number NP_000106.1)を強制発現させたCHO-K1-mt aequorin細胞を抗生剤無添加の培地で18時間培養した後、PBS-EDTA(5mM EDTA)で処理し、遠心分離後(2分、405×g、室温)アッセイバッファー(DMEM/HAM‘s F12 with HEPES+0.1% BSA protease free)に懸濁させた。
1.ETBのIC50値/ETAのIC50値が5000より大きい。
2.1.0nMで50%以上のETA阻害作用を示し、かつ5000nMで50%より小さいETB阻害作用を示す。
ETBのIC50値/ETAのIC50値が1000より大きく、5000以下の場合「+」とした。
結果を表1-3に示す。
CYP2C9に対する阻害作用を以下の方法により評価した。
(1)凍結しておいた肝ミクロソーム画分を氷上で溶解し、終濃度が0.5mg/mLになるようにPotassium phosphate buffer(PPB)で希釈した。
(2)評価用基質(Diclofenac:Sigma Aldrich社D6899)を終濃度2.5μMとなるように添加した。
(3)ゆっくりと混和し、37±1℃で5分間インキュベートし、反応液とした。
(4)反応液を801μLずつチューブに分取し、0.9μLのDMSOで段階希釈した評価化合物を終濃度9.1~20000nM(1/3希釈8濃度)になるように添加した。このとき、陰性コントロール群には0.9μLのDMSOを添加し、陽性コントロール群には終濃度1μMになるようにDMSOで調整したSulfaphenazole(Sigma Aldrich社S0758)を0.9μL添加した。
(5)混和後、270μLずつ別のチューブ2本に移し、37±1℃で5分間振盪水浴でインキュベートした。
(6)30μLの10mM NADPHを添加し、反応を開始し、37℃±1℃で10分間インキュベートした。
(7)振盪水浴からチューブを取り出し、300μLの反応停止液を加えて反応を停止し、混和後1021×gで20分間4℃で遠心分離した。
(8)評価用基質の代謝量をLC-MS/MSにて求めることで、評価化合物の各濃度の阻害率を算出した。
(9)IC50値をGraphPad Prism5を用いて算出した。IC50値が1μM以下であれば「++」、1μMより大きく、かつ10μM以下の範囲であれば「+」、10μMよりも大きければ「-」とした。結果を表2に示す。
ヒト肝ミクロソームにおける代謝安定性を以下の方法により評価した。
1.材料
(1)ヒト肝ミクロソーム:20mg/mL
(2)試験化合物:1.1mM DMSO溶液
(3)リン酸カリウム緩衝液:66.7mM(pH7.4)
(4)NADPH溶液:10mM in リン酸カリウム緩衝液
(5)クエンチング溶液:内部標準物質としてワルファリン入りの0.5%ギ酸-アセトニトリル溶液
プロピレンチューブ中に、971.5μLのリン酸カリウム緩衝液と27.5μLのヒト肝ミクロソームを入れ、懸濁させた。ここに、1μLの試験化合物を加え、この混合物のうち180μLを別のチューブに移した。混合物を37℃で5分間プレインキュベーションし、その後、20μLのNADPH溶液(インキュベーション時間30分の場合)又は20μLのリン酸カリウム緩衝液(インキュベーション時間0分の場合)を加えた。インキュベーション後に、200μLのクエンチング溶液を加え、反応を停止させた。続いて、3220×gで20分間遠心分離し、200μLの上清中の試験化合物の未変化体濃度をLC-MS/MSで測定した。得られた未変化体のピーク面積に基づき、インキュベーション時間0分を100%として未変化体の残存率(%)を算出した。
PAMPAを用いた膜透過性試験を検討した。具体的な方法は以下のとおりである。
(1)ドデカン液中にレシチンを2%(w/v)となるように加え、完全に溶解させた。(2)5μLのレシチン/ドデカン液をドナープレートに添加した。この時ピペットチップが膜に触れないようにした。
(3)すぐに150μLの評価化合物(10μM)を含むドナー溶液(5%DMSO-PBS)をドナープレートの各ウェルに添加し、300μLの水性バッファーをPTFEアクセプタープレートの各ウェルに添加した。
(4)評価化合物を含むドナー溶液で満たしたドナープレートをアクセプタープレートの上にゆっくりと設置し、膜の下面が全てのウェルでバッファーと接していることを確認した。
(5)蓋をして、装置を16時間室温でインキュベートした。
(6)インキュベート後、アクセプタープレートの膜の上面と下面から100μLずつ溶液を回収し、等量の内部標準を含むアセトニトリルを添加した。
(7)評価化合物の濃度を測定し、透過速度を算出した。
結果を表4に示す。膜透過速度の値が1x10-8cm/sec以下であれば「C」、1x10-8cm/secより大きく,かつ10x10-8cm/sec以下の範囲であれば「B」、10x10-8cm/secよりも大きければ「A」とした。
8~20週齢の雄性Wistarラットに、鎮痛剤処置とウレタン麻酔下にて、気管へ気道確保用カテーテル処置、頸動脈へ血圧測定用カテーテル処置、大腿静脈へBigET-1投与用カテーテル処置、十二指腸へ被験物質投与用カテーテル処置を行った。血圧および心拍数が安定したことを確認後、溶媒(0.5%メチルセルロース400溶液)または溶媒で調整した被験物質30mg/kg(5mL/kg)を十二指腸内に投与し、その1時間後にBigET-1(20μg/kg)を投与し、BigET-1投与60分後まで血圧をモニタリングした。各群n=6で実施し、解析はBigET-1投与5分前から60分後まで、5分毎に20秒間の収縮期血圧、拡張期血圧を求め、BigET-1投与5分前からの差分(デルタ値)を用いてBigET-1投与後60分間のAUC0-60min(mmHg*min)を算出した。溶媒投与群と被験物質投与群のAUCの比較にはStudentのt検定を用いた。
