Description
本願は、2020年2月5日に、日本に出願された特願2020-018312号に基づき優先権を主張し、その内容をここに援用する。
(1) (A)ステリル基を導入したヒアルロン酸誘導体と、
(B)ヒドロキシ基、カルボキシ基、アミノ基、アミド基、カルバメート基、ウレア基およびチオール基からなる群から選ばれる少なくとも一種類の官能基を有する極性基含有化合物と、
を含むヒアルロン酸誘導体組成物であって、
(A)ヒアルロン酸誘導体に対する前記ステリル基の導入率が0.1%以上35%未満である、ヒアルロン酸誘導体組成物。
(2) 前記(B)極性基含有化合物が、ヒドロキシ基を少なくとも一つ有する極性基含有化合物である、前記(1)に記載のヒアルロン酸誘導体組成物。
(3) 前記(B)極性基含有化合物の含有量が、前記(A)ヒアルロン酸誘導体の質量に対して0.001質量ppm以上1000質量ppm未満である、前記(1)又は(2)に記載のヒアルロン酸誘導体組成物。
(4) 前記(B)極性基含有化合物が複数の極性基を有している化合物である、前記(1)~(3)いずれかに記載のヒアルロン酸誘導体組成物。
(5)前記(B)極性基含有化合物が、アルコールである前記(1)~(4)いずれかに記載のヒアルロン酸誘導体組成物。
(6) 前記アルコールが、エタノール、イソプロパノール、及び、多価アルコールからなる群より選択される、前記(5)に記載のヒアルロン酸誘導体組成物。

本実施形態のヒアルロン酸誘導体組成物は、(A)ステリル基を導入したヒアルロン酸誘導体(以下、「(A)ヒアルロン酸誘導体」と称する場合がある)と、(B)(B)ヒドロキシ基、カルボキシ基、アミノ基、アミド基、カルバメート基、ウレア基およびチオール基からなる群から選ばれる少なくとも一種類の官能基を有する極性基含有化合物と、を含む。(A)ヒアルロン酸誘導体に対するステリル基の導入率が0.1%以上35%未満である。
(A)ヒアルロン酸誘導体において、ステリル基は、ヒアルロン酸に対して直接的に結合していてもよく、リンカーを解して結合されていてもよい。
ここでいう「リンカー」とは、遺伝子工学により導入し得る任意のペプチドリンカー、又は合成化合物リンカーを用いることができるが、本実施形態の(A)ヒアルロン酸誘導体においては、ペプチドリンカーが好ましい。ペプチドリンカーの長さは特に限定されず、目的に応じて当業者が適宜選択することが可能であるが、好ましい長さは2アミノ酸以上(上限は特に限定されないが、通常、30アミノ酸以下、好ましくは20アミノ酸以下)であり、特に好ましくは15アミノ酸である。(A)ヒアルロン酸誘導体に含まれるペプチドリンカーは、全て同じ長さのペプチドリンカーを用いてもよく、異なる長さのペプチドリンカーを用いてもよい。
本明細書において使用される「ステリル基」という用語は、ステロイド骨格を有する基であれば特に制限されない。ここでステロイドとしては、具体的には、コレステロール、コレスタノール、カンペスタノール、エルゴスタノール、スチグマスタノール、コプロスタノール、スチグマステロール、シトステロール、ラノステロール、エルゴステロール、シミアレノール、胆汁酸、テストステロン、エストラジオール、プロゲストロン、コルチゾール、コルチゾン、アルドステロン、コルチコステロン、デオキシコルチステロン等が挙げられる。ステリル基としては、コレステリル基、スチグマステリル基、ラノステリル基、エルゴステリル基等が挙げられ、中でも、コレステリル基(特に、コレスタ-5-エン-3β-イル基)が好ましい。
(A)ヒアルロン酸誘導体に対するステリル基の導入率(以下、単に「ステリル基導入率」と称する場合がある)は0.1%以上35%未満であり、5%以上33%以下が好ましく、6%以上22%以下がより好ましく、6%以上20%以下がさらに好ましい。
ステリル基導入率が上記範囲内であることで、ヒアルロン酸誘導体が純水中又は低塩濃度下では良好に溶解しつつ、生理食塩濃度下で凝集して沈殿を形成するという性質を有するようになる。ステリル基導入率が上記範囲内であることで、ヒアルロン酸誘導体組成物中の(A)ヒアルロン酸誘導体を薬物と複合化させた、ヒアルロン酸誘導体-薬物結合体組成物は、体内に投与(例えば、皮下投与)する際に、投与後に凝集する特長を生かした沈殿型の徐放製剤となり得る。

(A)ヒアルロン酸誘導体の分子量は特に限定はされないが、局所投与における拡散遅延由来の徐放機能を期待する観点や前述した生理食塩濃度下における沈殿形成能という観点からは、分子量の比較的大きい(A)ヒアルロン酸誘導体が好ましく、最終剤形が溶液製剤の場合はシリンジアビリティの観点から分子量の比較的小さい(A)ヒアルロン酸誘導体が好ましい。このような(A)ヒアルロン酸誘導体の分子量としては、1,000(1kDa)以上1,000,000(1,000kDa)未満が好ましく、3kDa以上500kDa以下がより好ましく、5kDa以上300kDa以下がより好ましく、5kDa以上120kDa以下がさらに好ましく、10kDa以上100kDa以下が特に好ましい。(A)ヒアルロン酸誘導体の分子量は、一般的には、対応する分子量を有する原料を使用することにより調節することができる。

さらに、別の態様において、Zは、-NH-[CH(-Za)-CONH]n-1-CH(-Za)-CO-で表されるペプチドリンカーであってもよく、ここで、nは2以上30以下の整数であり、Zaは、それぞれ独立に、H2N-CH(-Za)-COOHとして表されるα-アミノ酸中の置換基を表す。当該ペプチドリンカーは、N末端にてグルクロン酸部分のカルボキシ基に結合し、C末端にて基-N(-Ra)-Y-X1に結合する。当該ペプチドリンカーのアミノ酸残基として利用できるアミノ酸の例としてはα-アミノ酸、例えばアラニン、アルギニン、アスパラギン(Asn)、アスパラギン酸、システイン、グルタミン、グルタミン酸、グリシン(Gly)、ヒスチジン、イソロイシン、ロイシン(Leu)、リジン、メチオニン、フェニルアラニン(Phe)、プロリン、セリン、スレオニン、トリプトファン、チロシン、バリンといった天然型(L型)のアミノ酸、それらのD体等が挙げられ、合成されたアミノ酸を含む全てのα-アミノ酸を用いることができる。すなわち、Zaとしては、例えば、-CH3、H2NC(NH)NH(CH2)3-、H2NCOCH2-等が挙げられる。また、n個のZは、同一でも異なっていてもよい。nは、2以上30以下の整数であるが、2以上10以下が好ましく、2以上4以下がより好ましい。ペプチドリンカーの好ましい例としては、例えば、-Gly-Phe-Leu-Gly-、-Asn-Phe-Phe-、-Phe-Phe-、Phe-Gly-等が挙げられる。
一般式(I)において、Yは-(CH2)n1-及び-(CH2CH2O)m1-CH2CH2-(ここで、n1は、2以上20以下の整数であり、2以上15以下の整数が好ましく、2以上12以下の整数がより好ましく、2以上6以下の整数がさらに好ましい。m1は、1以上4以下の整数である)からなる群より選択される基が好ましい。具体的には、-(CH2)2-、-(CH2)6-、-(CH2)8-、-(CH2)12-、又は、-(CH2CH2O)2-CH2CH2-が好ましい。また、純水中乃至低塩濃度下では高い溶解性を実現させつつ、生理食塩濃度下では高い沈殿形成能を示させるという観点からは、Yは-(CH2)2-、-(CH2)6-、-(CH2)8-及び-(CH2)12-からなる群より選択される基が好ましく、-(CH2)6-がより好ましい。
Yaとしては、-CH2-又は-CH2-CH2-が好ましい。
Ybとしては、-CH2-CH2-、-CH(CH3)CH2-、2-ブテン-1,4-ジイル、ヘプタ-2,4-ジエン-1,6-ジイル又はオクタ-2,4,6-トリエン-1,8-ジイルが好ましく、-CH2-CH2-又は-CH(CH3)CH2-がより好ましい。
Rとしては、コレステリル基が好ましい。

別の態様において、(A)ヒアルロン酸誘導体は、繰り返し単位(I)、繰り返し単位(Ia)及び繰り返し単位(II)から実質的になるヒアルロン酸誘導体であってもよい。
一般式(II)において、Q+はカルボキシ基と水中で塩を形成するカウンターカチオンであれば特に限定されず、2価以上の場合は価数に応じて複数のカルボキシ基と塩を形成する。カウンターカチオンの例としては、リチウムイオン、ナトリウムイオン、ルビジウムイオン、セシウムイオン、マグネシウムイオン、カルシウムイオン等の金属イオン;式:N+RjRkRlRm(式中、Rj、Rk、Rl及びRmは、それぞれ独立に、水素原子及びC1-6アルキルからなる群より選択される)で表されるアンモニウムイオン等が挙げられる。中でも、Q+は、ナトリウムイオン、カリウムイオン、又はテトラアルキルアンモニウムイオン(例えば、テトラn-ブチルアンモニウムイオン等)が好ましい。Rj、Rk、Rl及びRmは、C1-6アルキルからなる群より選択される同一の基であることが好ましく、n-ブチル基が好ましい。
(B)極性基含有化合物は、ヒドロキシ基、カルボキシ基、アミノ基、アミド基、カルバメート基、ウレア基およびチオール基からなる群から選ばれる少なくとも一種類の官能基を有する。
(B)極性基含有化合物の含有量が上記下限値以上であることで、ヒアルロン酸誘導体組成物の生理食塩濃度下での沈殿形成能をより高くすることができ、一方、上記上限値以下であることで、純水中乃至低塩濃度下における(A)ヒアルロン酸誘導体の凝集をより効果的に抑制することができ、ヒアルロン酸誘導体組成物水溶液の0.22μmフィルターを用いた滅菌ろ過のろ過性をさらに向上させることができる。
(B)極性基含有化合物の含有量は、ガスクロマトグラフィー-質量分析(GC-MS)法により測定することができ、具体的には後述する実施例に記載された方法に従って測定することができる。
中でも、アルコールとして、エタノール、イソプロパノール、及び、多価アルコールからなる群より選択される1種以上のアルコールが好ましく、多価アルコールがより好ましく、エチレングリコールがさらに好ましい。

本実施形態の医薬組成物は、薬物と担体を含み、上記ヒアルロン酸誘導体組成物を担体として含む。本実施形態の医薬組成物において、担体と薬物は、水素結合、イオン結合、ファンデルワース力等の非共有結合等により直接的又は間接的に結合して、複合体を形成しており、互いに遊離していない状態であることが望ましい。当該医薬組成物を生体内に投与した際には、担体から薬物が徐々に遊離し、良好な徐放性を期待できる。
本実施形態の医薬組成物に含まれる薬物としては、特に限定されず、例えば、タンパク質、ペプチド、多糖類、核酸、低分子化合物等が挙げられる。本実施形態の医薬組成物は、薬理活性を有するタンパク質、ペプチド、核酸等のバイオ医薬品又は低分子化合物が、ヒアルロン酸誘導体組成物中の(A)ヒアルロン酸誘導体と、複合体を形成して含まれることが好ましい。
低分子化合物としては、例えば、制癌剤(例えば、アルキル化剤、代謝拮抗剤、アルカロイド等)、免疫抑制剤、抗炎症剤(ステロイド剤、非ステロイド剤系抗炎症剤等)、抗リウマチ剤、抗菌剤(β-ラクタム系抗生物質、アミノグリコシド系抗生物質、マクロライド系抗生物質、テトラサイクリン系抗生物質、新キノロン系抗生物質、サルファ剤等)等が挙げられる。

核酸としては、例えば、DNA、RNA、アンチセンス核酸、デコイ核酸、リボザイム、低分子干渉RNA、核酸アプタマー等が挙げられる。
多糖類としては、例えば、レンチナン、シゾフィラン、パヒマラン、プスチュラン、イーストグルカン、イーストマンナン、マリナクタン、カードラン、デキストラン硫酸、ヘパリン、カラゲナン、イヌリン、コンドロイチン、コンドロイチン硫酸等が挙げられる。
本実施形態の医薬組成物は、分散性微粒子溶液であってもよく、沈殿性懸濁液であってもよく、凍結乾燥体であってもよい。分散性微粒子溶液である場合は、体内に投与する前は溶液状態でありながら体内に投与(例えば、皮下投与)することにより投与後に体内のその場で凝集する特長を有する沈殿型の徐放製剤となりうる。沈殿性懸濁液である場合は、活性成分がバーストリリースされにくいことを特長とする沈殿型の徐放製剤となりうる。この場合、シリンジアビリティも付与することが可能であることから、沈殿物の大きさは200μm以下が好ましく、100μm以下がより好ましく、20μm以下がさらに好ましい。また、凍結乾燥体の場合は、医師が投与前に生理食塩水等の等張液を加えて投与液を用事調製するタイプの沈殿型の徐放製剤となりうる。この場合、溶液状態では不安定な活性成分を含む医薬組成物に適していると考えられる。
本実施形態のヒアルロン酸誘導体-薬物結合体組成物は、上記ヒアルロン酸誘導体-薬物結合体組成物と1以上の薬物を含み、上記ヒアルロン酸誘導体組成物に含まれる(A)ヒアルロン酸誘導体に、上記1以上の薬物が結合している。
(ヒアルロン酸誘導体の分子量)
ヒアルロン酸誘導体の分子量は、サイズ排除クロマトグラフィー多角度光散乱検出器(SEC-MALS)により決定された重量平均分子量である。ヒアルロン酸誘導体組成物(20mg)を超純水(10mL)に溶解して室温で12時間以上撹拌し、ヒアルロン酸誘導体組成物水溶液(2mg/mL)を得た。このヒアルロン酸誘導体組成物水溶液(750μL)に対して300mM ヒドロキシプロピル-β-シクロデキストリン(HP-β-CD)水溶液(750μL)加えて振とう機を用いて10秒間混合し、37℃にて1時間インキュベートした。そして、得られた試料をSEC-MALS測定に供して重量平均分子量を決定した。SEC-MALS測定の条件を以下に示す。

(ステリル基導入率)
ヒアルロン酸誘導体のステリル基導入率は、1H-NMR測定により決定した。まず、ジメチルスルホキシド-d6(99.9v/v%、0.05v/v%のトリメチルシリル(TMS)含有、富士フイルム和光製)と20%重塩酸(99.5v/v%、富士フイルム和光製)とを質量比99:1にて混合し、測定溶媒を調製した。続いて、この測定溶媒(0.6mL)にヒアルロン酸誘導体組成物(2mg)を添加し、超音波バスにて30分間処理して完全に溶解させて1H-NMR測定に供した。1H-NMR測定は、フーリエ変換核磁気共鳴装置(FT-NMR装置)(ECS400、日本電子製)を用いてサンプル温度85℃にて実施した。ステリル基導入率は、N-アセチル-D-グルコサミンのアセチル基由来のピーク(COCH3、1.6ppm以上2.0ppm以下、3H)の積分値と、コレステリル基中のメチル基由来のピーク(CH3、0.7ppm、3H)の積分値より、以下に示す式を用いてヒアルロン酸ユニットに対するコレステリル基の導入率を算出した。なお、N-アセチル-D-グルコサミンのアセチル基由来のピークが含まれる1.6ppm以上2.0ppm以下付近のピークにはコレステリル基由来のピーク(5H)が重なっているため、1.6ppm以上2.0ppm以下付近のピークの積分値からコレステリル基メチル由来のピーク(0.7ppm)の積分値を5/3倍したものを差し引いて算出した値(即ち、積分値(1.6ppm以上2.0ppm以下)-積分値(0.7ppm)×5/3)をN-アセチル-D-グルコサミンのアセチル基由来のピークの積分値として、ステリル基導入率の計算に使用した。

ヒアルロン酸誘導体組成物中の(B)極性基含有化合物の含有量は、ガスクロマトグラフィー-質量分析(GC-MS)法により測定した。
まず、ヒアルロン酸誘導体組成物(16.0mg)を超純水(2.0mL)に添加し、12時間以上撹拌して溶解させて8.0mg/mLのヒアルロン酸誘導体組成物水溶液を得た。その後、得られた水溶液をGC-MS測定に供し、外部標準法によって測定試料中の(B)極性基含有化合物の含有量を求めた。そののち、添加したヒアルロン酸誘導体の量に対する測定試料中の(B)極性基含有化合物の質量比を、ヒアルロン酸誘導体に対する(B)極性基含有化合物の含有量として算出した。定量分析に必要な検量線は、市販標品を用いて作成した。GC-MS測定の条件を以下に示す。

ヒアルロン酸誘導体を0.1gをガラスチューブに入れ、200℃で10分間加熱脱着を行い、揮発成分を-130℃にてトラップした。その後、冷却されていたトラップを250℃で急速加熱し、GC-MS測定を行った。
濃度既知のエチレングリコール標準品を同様の条件で測定し、検出ピーク面積値から検量線を作成した。検量線からサンプル中のエチレングリコールの質量を算出した。その後、使用した(A)ヒアルロン酸誘導体の質量に対するサンプル中のエチレングリコールの質量比を、(A)ヒアルロン酸誘導体に対する(B)極性官能基含有化合物の含有量として算出した。

イソプロパノールおよびエタノールの場合は、温度条件を50℃(5分)→10℃/分→240℃(6分保持)とした以外は、エチレングリコールと同様に測定した。
(沈殿率)
ヒアルロン酸誘導体組成物の沈殿率は、以下に示す手順により決定した。まず、600mM塩化ナトリウム入りの40mMリン酸緩衝液(pH 7.4)を調製した(濃縮緩衝液と呼ぶ)。次に、ヒアルロン酸誘導体組成物に超純水を加え、12時間以上撹拌して溶解させてヒアルロン酸誘導体組成物水溶液を得た。この水溶液を、ろ過チューブ(ポアサイズ:5.0μm;UFC40SV25;メルク社製)に入れて遠心分離機にかけ、ろ過処理した。続いて、ろ過済みヒアルロン酸誘導体水溶液(600μL)と濃縮緩衝液(200μL)を混合した沈殿形成サンプル(N数:3)、及び、ろ過済みヒアルロン酸誘導体水溶液(600μL)と超純水(200μL)を混合したリファレンスサンプル(N数:2)を調製した。その後、沈殿形成サンプル及びリファレンスサンプルを37℃にて20分間インキュベートし、そののちに遠心分離機(2,000×g、10分間)にかけた。遠心分離後のサンプルそれぞれの上澄みを200μL採取し、いずれも300mMヒドロキシプロピル-β-シクロデキストリン(HP-β-CD)水溶液(200μL)を加えて2倍希釈したうえで、37℃にて2時間インキュベートした。さらに、全てのサンプルに対して10mM HP-β-CD水溶液(1000μL)を加えて希釈し、シリンジフィルター(ポアサイズ:0.45μm)により処理した。そして、得られた沈殿形成サンプル及びリファレンスサンプルをサイズ排除クロマトグラフィー(SEC)測定に供した。ヒアルロン酸誘導体組成物の沈殿率は、沈殿形成サンプル及びリファレンスサンプルのSECクロマトグラムのヒアルロン酸誘導体由来のピーク(7分付近)の面積値を用いて、以下に示す式に基づいて算出した。

(滅菌ろ過性)
滅菌ろ過性は、ヒアルロン酸誘導体組成物水溶液を0.22μmフィルターで処理した際のろ過最大量により評価した。すなわち、ヒアルロン酸誘導体組成物を超純水に溶解して0.9mg/mLとし、得られたヒアルロン酸誘導体組成物水溶液を0.22μmフィルター(ステリカップ-GP S2GPU05RE、有効ろ過面積40cm2、メルク社製)により膜が目詰まりして水溶液が透過しなくなるまでろ過しつづけた。そして、ろ過済みのヒアルロン酸誘導体組成物水溶液の体積を測定し、有効ろ過面積あたりのろ過済みのヒアルロン酸誘導体組成物水溶液の体積を計算してろ過最大量とした。
[実施例1]
(ヒアルロン酸誘導体組成物HA-a1の製造)
ヒアルロン酸誘導体組成物を次の工程1~工程3に従って調製した。
(コレステリル 6-アミノヘキシルカーバメート塩酸塩の合成)
コレステリル 6-アミノヘキシルカーバメート塩酸塩(Chol塩酸塩)を次に示す工程1-1、続いて工程1-2に従って合成した。
コレステリルクロロホルメート(3.37g、7.5mmol)の無水ジクロロメタン(20mL)の溶液に、アルゴン雰囲気下、トリエチルアミン(TEA、1.05mL)を加えて撹拌した。氷冷下で、6-(t-ブトキシカルボニル)アミノ-1-アミノヘキサン(1.12mL、5mmol)を滴下して加え、そのまま氷冷下で30分間攪拌後、室温まで昇温し、当該混合物を一晩撹拌した。反応混合物を、超純水及び飽和食塩水で洗浄し、無水硫酸マグネシウムで乾燥後、減圧下で溶媒を留去した。得られた残渣をシリカゲルカラムクロマトグラフィー(溶離液:酢酸エチル:n-ヘキサン=1:4)で精製し、目的物のフラクションを合わせて溶媒を減圧下留去した。
得られた残渣を酢酸エチル(40mL)に溶解し、4N塩酸/酢酸エチル溶液(40mL)を加えて室温で一晩撹拌した。生じた沈殿物を遠心分離により回収した。得られた固体を酢酸エチルにて4回洗浄後、減圧下で乾燥し、コレステリル 6-アミノヘキシルカーバメート塩酸塩(Chol塩酸塩)1.2gを得た。生成物の1H-NMRスペクトル(CS400 日本電子製、EtOH-d6)を図1に示す。
(ヒアルロン酸のテトラブチルアンモニウム(TBA)塩の調製)
ヒアルロン酸のTBA塩(HA-TBA)を次に示す工程2-1、続いて工程2-2に従って調製した。
DOWEX(登録商標)50WX-8-400(アルドリッチ社製)を超純水に懸濁させ、デカンテーションにより樹脂を超純水で3回程度洗浄した。40質量%テトラブチルアンモニウムヒドロキシド水溶液(TBA-OH)(アルドリッチ社製)を樹脂のカチオン交換能に対し約1.5倍モル等量加え、30分間撹拌した。余剰のTBA-OH溶液をデカンテーションにより除去した後、さらに過剰の超純水で洗浄することで、TBA塩化したカチオン交換樹脂を得た。
分子量50,000(50kDa)の原料ヒアルロン酸ナトリウム塩(HA-Na)を15mg/mLの濃度で超純水に溶解した。「(1)工程2-1」でTBA塩化したカチオン交換樹脂の懸濁液を、HAユニット(ユニット分子量401.3)のモル数に対し樹脂のイオン交換能換算で5倍モル等量添加した。15分間撹拌した後、0.45μmのフィルターを用いて濾過を行い、濾液を凍結乾燥し、ヒアルロン酸のTBA塩(HA-TBA)を白色固体として得た。生成物の1H-NMRスペクトル(ECS400 日本電子製、EtOH-d6)を図2に示す。
「2.(2)工程2-2」で調製したHA-TBAの無水DMSO溶液(10mg/mL)を調製した。その後、HA-TBA中に存在する二糖繰り返し単位(HAユニット)に対する、「1.工程1」で合成したChol塩酸塩の添加量がモル比(Chol塩酸塩/HAユニット)で15/100となるように添加した。次に、HAユニットに対する4-(4,6-ジメトキシ-1,3,5-トリアジン-2-イル)-4-メチルモルホリニウムクロリド(DMT-MM)の添加量がモル比(DMT-MM/HAユニット)で21.6/100となるように加え、室温で一晩撹拌した。反応溶液は、0.3M 酢酸アンモニア/DMSO溶液、0.15M NaCl水溶液、超純水の順で透析(スペクトラポア4(スペクトラム社製)、分画分子量(MWCO):12,000以上14,000以下)した。得られた透析液に対し、エチレングリコール(EG)を、組成物中の含有量が0.0062質量ppmとなるように加え、その後凍結乾燥して目的物(HA-C6-Chol)を白色固体として得た。生成物の1H-NMRスペクトルを図3に示す。N-アセチル-D-グルコサミンのアセチル基由来のピーク(COCH3、1.6ppm以上2.0ppm以下、3H)、コレステリル基中のメチル基由来のピーク(CH3、0.7ppm、3H)が確認された。
(ヒアルロン酸誘導体組成物HA-a2~HA-a25及びHA-b1~HA-b2の製造)
「3.工程3」において、HAユニットに対するChol塩酸塩の添加量、及び、HAユニットに対するDMT-MMの添加量がモル比で下記表に示す比率となるようにし、アルコールの種類及び(A)ヒアルロン酸誘導体に対する(B)極性基含有化合物の含有量が下記表に示すようにした以外は、実施例1と同様の方法を用いて、各ヒアルロン酸誘導体組成物を得た。得られたヒアルロン酸誘導体組成物の1H-NMR測定を行い、各ヒアルロン酸誘導体組成物に含まれるヒアルロン酸誘導体においていずれも、N-アセチル-D-グルコサミンのアセチル基由来のピーク(COCH3、1.6ppm以上2.0ppm以下、3H)、コレステリル基中のメチル基由来のピーク(CH3、0.7ppm、3H)が確認された。
(アルコールの種類及び略称)
イソプロパノール:IPA
エタノール:EtOH
また、ステリル基導入率が異なるヒアルロン酸誘導体組成物HA-a1、HA-a17及びHA-a18(実施例1、17及び18)において、ステリル基導入率が低くなるほど、6mg/mLのNaCl含有濃縮緩衝液での沈殿率がより良好になる傾向がみられた。
また、アルコールの含有量が異なるヒアルロン酸誘導体組成物HA-a19及びHA-a23(実施例19及び23)において、アルコールの含有量が少なくなるほど、ろ過最大量が増加する傾向がみられた。
また、アルコールの種類が異なるヒアルロン酸誘導体組成物HA-a23~HA-a25(実施例23~25)において、エチレングリコールを使用した場合に、1.2mg/mLのNaCl含有濃縮緩衝液での沈殿率が良好であった。
Claims (15)
- (A)ステリル基を導入したヒアルロン酸誘導体と、
(B)ヒドロキシ基、カルボキシ基、アミノ基、アミド基、カルバメート基、ウレア基およびチオール基からなる群から選ばれる少なくとも一種類の官能基を有する極性基含有化合物と、
を含むヒアルロン酸誘導体組成物であって、
(A)ヒアルロン酸誘導体に対する前記ステリル基の導入率が0.1%以上35%未満である、ヒアルロン酸誘導体組成物。 - 前記(B)極性基含有化合物が、ヒドロキシ基を少なくとも一つ有する極性基含有化合物である、請求項1に記載のヒアルロン酸誘導体組成物。
- 前記(B)極性基含有化合物の含有量が、前記(A)ヒアルロン酸誘導体の質量に対して0.001質量ppm以上1000質量ppm未満である、請求項1又は2に記載のヒアルロン酸誘導体組成物。
- 前記(B)極性基含有化合物が複数の極性基を有している化合物である、請求項1~3いずれか一項に記載のヒアルロン酸誘導体組成物。
- 前記(B)極性基含有化合物が、アルコールである請求項1~4いずれか一項に記載のヒアルロン酸誘導体組成物。
- 前記アルコールが、エタノール、イソプロパノール、及び、多価アルコールからなる群より選択される、請求項5に記載のヒアルロン酸誘導体組成物。
- 前記アルコールが、多価アルコールである、請求項6に記載のヒアルロン酸誘導体組成物。
- 前記アルコールが、エチレングリコールである、請求項7に記載のヒアルロン酸誘導体組成物。
- 前記(A)ヒアルロン酸誘導体が、下記一般式(I)で表される繰り返し単位を有する、請求項1~8のいずれか一項に記載のヒアルロン酸誘導体組成物。
Zは、直接結合、又は2個以上30個以下のアミノ酸残基からなるペプチドリンカーを表し;
X1は、以下の式:
-NRb-R、
-NRb-COO-R、
-NRb-CO-R、
-NRb-CO-NRc-R、
-COO-R、
-O-COO-R、
-S-R、
-CO-Ya-S-R、
-O-CO-Yb-S-R、
-NRb-CO-Yb-S-R、及び
-S-S-R、
で表される基からなる群より選択される基であり;
Ra、Rb及びRcは、それぞれ独立に、水素原子、C1-20アルキル、アミノC2-20アルキル及びヒドロキシC2-20アルキルからなる群より選択され、ここで当該基のアルキル部分は、-O-及び-NRf-からなる群より選択される基が挿入されていてもよく;
Rfは、水素原子、C1-12アルキル、アミノC2-12アルキル及びヒドロキシC2-12アルキルからなる群より選択され、当該基のアルキル部分は-O-及び-NH-からなる群より選択される基が挿入されていてもよく;
Rは、ステリル基であり;
Yは、C2-30アルキレン、又は-(CH2CH2O)m-CH2CH2-であり、ここで、当該アルキレンは、-O-、-NRg-及び-S-S-からなる群より選択される基が挿入されていてもよく;
Rgは、水素原子、C1-20アルキル、アミノC2-20アルキル及びヒドロキシC2-20アルキルからなる群より選択され、当該基のアルキル部分は-O-及び-NH-からなる群より選択される基が挿入されていてもよく;
Yaは、C1-5アルキレンであり;
Ybは、C2-8アルキレン又はC2-8アルケニレンであり;
mは、1以上100以下の整数である。 - 前記Rがコレステリル基である、請求項9に記載のヒアルロン酸誘導体組成物。
- 前記(A)ヒアルロン酸誘導体の分子量が1,000以上1,000,000未満である、請求項1~10のいずれか一項に記載のヒアルロン酸誘導体組成物。
- 薬物と、担体を含み、前記担体が、請求項1~11のいずれか一項に記載のヒアルロン酸誘導体組成物である、医薬組成物。
- 前記薬物が(A)ヒアルロン酸誘導体と複合体を形成する、請求項12に記載の医薬組成物。
- 前記薬物が、薬理活性を有するタンパク質、ペプチド又は核酸である、請求項12又は13に記載の医薬組成物。
- 請求項1~11のいずれか一項に記載のヒアルロン酸誘導体組成物および薬物を含み、前記ヒアルロン酸誘導体組成物に含まれる(A)ヒアルロン酸誘導体に、前記薬物が結合した、ヒアルロン酸誘導体-薬物結合体組成物。
