Abstract
ヒトを含めた哺乳類動物などを対象に、例えば無機物質などの経口摂取可能なマテリアルで屁及び/又は大便の独特な悪臭の効果的な低減を実現することを目的に、ナノサイズ化されたコロイド状の白金微粒子を有効成分として含む、経口摂取用の屁及び/又は大便の悪臭低減剤を提供する。
Description
本発明は、屁及び/又は大便の独特な悪臭を効果的に低減する悪臭低減剤に関する。
ヒトを含めた哺乳類動物などの屁と大便にはヒトに強い不快感を与える独特な悪臭があり、美容的な観点や介護の容易化などの観点若しくはペットの衛生的飼育のし易さの観点から悪臭を軽減する方法が望まれている。この悪臭は硫化物(硫化水素、二酸化硫黄、硫化炭化水素)、インドール、スカトール、亜鉛、アンモニアなどにより生じるものと想像されており、例えば、ビフィズス菌によりアンモニアやインドールなどを低減し大便の悪臭を軽減する方法は知られている(非特許文献1)。しかしながら、ビフィズス菌などを除くと、経口摂取可能な例えば無機物などのマテリアルで屁と大便の独特な悪臭を効果的に低減するものは従来無かった。
外気にフレグランスを噴霧して便臭などを打ち消すものはあったが(特許文献1)、マッシュルームから抽出される機能性食品素材とされるシャンピニオンエキスは口臭、体臭、便臭の消去に効果的とされたがその根拠が認められず、平成21年に公正取引委員会によって排除命令が提出された。
このような事情から、安全に経口投与可能で、効果的にヒトを含めた哺乳類動物などの屁と大便の悪臭を低減する手段の提供が望まれていた。
一方、ナノサイズの白金微粒子を含むスーパーオキサイドアニオン分解剤が知られており(特許文献2)、このような白金微粒子を含む口腔内洗浄材なども知られているが(特許文献3)、白金微粒子が屁及び/又は大便の独特な悪臭を効果的に低減することは全く知られていなかった。

本発明の課題は、ヒトを含めた哺乳類動物などを対象に、例えば無機物質などの経口摂取可能なマテリアルで屁及び/又は大便の独特な悪臭の効果的な低減を実現することにある。
本発明者は、白金微粒子を経口摂取させると、驚くべきことに屁と大便の独特の悪臭を効果的に低減できることを見出した。
本発明により、白金微粒子を有効成分として含む、経口摂取用の屁及び/又は大便の悪臭低減剤が提供される。
本発明の悪臭低減剤は、屁及び/又は大便中の硫化水素の低減用の悪臭低減剤である場合がある。
また、本発明の悪臭低減剤は、ナノサイズ化されたコロイド状の白金微粒子を有効成分として含む悪臭低減剤である場合がある。
また、本発明の悪臭低減剤は、前記白金微粒子の平均粒径が10nm以下である悪臭低減剤である場合がある。
さらに、本発明の悪臭低減剤は、水1000ml当たり1nmole~100mmoleの白金微粒子を含む悪臭低減剤である場合がある。
また、本発明は、白金微粒子を有効成分として経口摂取する、屁及び/又は大便の悪臭を低減する方法を提供する。
さらに、本発明は、屁及び/又は大便の悪臭を低減する方法に使用するための白金微粒子を提供する。
本発明の白金微粒子は、屁及び/又は大便中の硫化水素の低減用の悪臭低減剤に使用するための白金微粒子である場合がある。
また、本発明の白金微粒子は、ナノサイズ化されたコロイド状の白金微粒子を有効成分として含む悪臭低減剤として使用するための白金微粒子である場合がある。
また、本発明の白金微粒子は、前記白金微粒子の平均粒径が10nm以下である悪臭低減剤として使用するための白金微粒子である場合がある。
さらに、本発明の白金微粒子は、水1000ml当たり1nmole~100mmoleの白金微粒子を含む悪臭低減剤として使用するための白金微粒子である場合がある。
また、本発明は、悪臭低減剤の製造における白金微粒子の使用を提供する。
本発明の悪臭低減剤は、ヒトを含む哺乳類動物などに対して経口摂取させることができ、体内に吸収されることがなく、ほぼ全量がそのまま排泄されるので安全である。例えば、クエン酸で還元されクエン酸をそのまま保護材とするナノサイズ化された白金微粒子を含む懸濁液として服用させることができ、屁及び/又は大便の独特な悪臭を効果的に消臭することができる。
本発明は、白金微粒子を有効成分として含む、経口摂取用の屁及び/又は大便の悪臭低減剤である。
本明細書において、「白金微粒子」は、白金の微粒子を言い、白金黒のような粉状の白金微粒子、保護剤を使用した白金微粒子、保護材を使用せずに界面活性剤を使用した白金微粒子、保護剤や界面活性剤も何も使用しない白金微粒子、及び、一原子粒子が含まれる。
白金微粒子の製造方法は種々知られており(例えば、特公昭57-43125号公報、特公昭59-120249号公報、及び特開平9-225317号公報、特開平10-176207号公報、特開2001-79382号公報、特開2001-122723号公報など)、当業者はこれらの方法を参照することによって微粒子を容易に調製することができる。例えば、貴金属微粒子の製造方法として、沈殿法又は金属塩還元反応法と呼ばれる化学的方法、あるいは燃焼法と呼ばれる物理的方法などを利用できる。液相レイザーアブレーション法や保護材を使用せずに界面活性剤で懸濁する方法を採用することもできる。本発明の悪臭低減剤としては、いずれの方法で調製された微粒子を用いてもよい。微粒子化の方法は、例えば特許第4058072号の明細書に説明されているので、この公報の開示を参照として本明細書の開示に含める。
また、例えば、保護剤を使用した白金微粒子の場合、微細金属粒子同士が凝集しないように保護する物質として金属保護剤を使用して、白金ナノ粒子が水中で均一に分散するようにすることができる。微細金属粒子の保護剤の例としては、ポリビニルピロリドン、ポリアクリル酸、ペクチン、クエン酸などが挙げられ、ペクチンとクエン酸は食品中の成分であることから、ペクチンやクエン酸が好ましい。
また、本発明の悪臭低減剤における白金は白金を含む合金であってもよいが、好ましいのは白金自体である。合金を形成する金属としては、例えば、金、ルテニウム、ロジウム、パラジウム、オスミウム、イリジウムなどが挙げられる。白金微粒子とともに、これらの他の金属の1種又は2種以上の微粒子を含む複数種金属のコロイド状態の微粒子懸濁物を使用してもよい。
白金微粒子としては、比表面積が大きく、表面反応性に優れたコロイド状態を形成可能な微粒子が好ましい。例えばマイクロメーターレベルの粒径の微粒子やナノレベルの粒径の微粒子とすることができる。微粒子の粒径は特に限定されないが、50nm以下の平均粒径を有する微粒子を用いることができ、好ましくは平均粒径が20nm以下、さらに好ましくは平均10nm以下、特に好ましくは平均粒径が1~6nm程度の微粒子を用いることができる。これらの微粒子を水性媒体中に安定な懸濁状態で含む分散液も好ましい。水性媒体としては、水のほか、生体に対する毒性が低く水と任意の割合で混じり合う有機溶媒、例えば、エタノール、エチレングリコールなどを用いることができる。好ましくは水性媒体として水を用いることができる。
本発明の悪臭低減剤は、例えば、水1000ml当たり1nmole~100mmole、好ましくは水1000ml当たり10nmole~50mmole程度の白金微粒子をコロイド状態で含むように調製されることが好ましい。白金濃度が100ppm~300ppm程度とすることが好ましく、200ppm程度とすることが特に好ましい。
上記の濃度でコロイド状の白金微粒子を含む悪臭低減剤をヒトを含む哺乳類動物に経口投与することができる。例えば、200ppmのコロイド状の白金微粒子を含む悪臭低減剤を一日当たり50~5000μl程度、好ましくは一日当たり200μl程度経口的に摂取することにより、腸管内において所望の作用が発現された結果、排泄される屁及び/又は大便の悪臭が顕著に低減する。すなわち、本発明の白金微粒子の経口摂取量は、1日当たり最大で1x10-3gに相当する。
白金微粒子の安全性については、静脈内投与での肝障害に関する報告がある(Y.Yagmagishi, A. et al.,Pharmazie 68:178-182, 2013)。具体的には、マウスに、粒径1nm以下と15nmの白金ナノ粒子(sub-nanosized platinum particles; 以下、各々snPt、nPtと記載)を静脈内投与して24時間後に、肝障害の有無を血中内に漏出したアラニンアミノ基転移酵素(alanine transaminase; ALT)とアスパラギン酸アミノ基転移酵素(aspartate transaminase)で測定した結果、投与量が15~20mg/kgでsnPtのみが肝障害を示した。一方、粒径15nmのnPtでは、同じ投与量でも肝障害は見られなかったとしている。この15~20mg/kgの静脈内投与は、体重60kgのヒトでは約1gとなる。経口摂取からこれ程の白金微粒子の量が体内に入るには、小腸からの吸収率が最大で0.1%(下記実施例に本発明の白金微粒子をヒトが経口摂取した場合、99.96%が糞中に回収されたことが示されている)と仮定すると、1,000gを経口摂取することになる。したがって、本発明の悪臭低減剤の経口摂取量は安全性が高いものと理解される。
本発明の悪臭低減剤は、健康食品として飲用することができ、それ自体を医薬としても使用できる。
健康食品としては、例えば、上記の200ppmのコロイド状の白金微粒子を含む悪臭低減剤を、茶、ジュース、清涼飲料水、ドリンクに添加して、これら形態で飲用できる。
本発明の悪臭低減剤を医薬として使用する場合には、上記の200ppmのコロイド状の白金微粒子を含む経口用液剤としてのみならず、薬学的に許容可能な医薬品添加剤を添加し、錠剤、カプセル剤、顆粒剤、散剤等の固形製剤として医薬製剤を製造し、患者に投与することができる。
前記医薬製剤の調製に使用される薬学的に許容可能な医薬品添加剤は、例えば、当業者に公知の、安定化剤、抗酸化剤、pH調整剤、緩衝剤、懸濁剤、乳化剤、界面活性剤等の添加物を添加して、調製することができる。これらの医薬品添加剤の種類及びその用法・用量は、医薬品添加物事典2016( 日本医薬品添加剤協会編、薬事日報社、2016年2月) などに記載され、これらの記載に従って医薬製剤を調製し、使用することができる。
1.白金ナノ粒子の製造
白金ナノ粒子は、クエン酸還元法で作製した。43.8 mlの水に、16.6mM 塩化白金酸(H2PtCl6)を4ml添加し、油浴(オイルバス)で加熱した。温度が100°Cに達した時点で、77.21mMのクエン酸三ナトリウム(trisodium Citrate; C6H5Na3O7)を8.6ml添加し、そのまま100°Cで反応液が黒色に変化するまで概ね1時間30分程加熱を継続した。反応液が黒色に変化したら加熱を停止し、反応液の温度が室温にまで下げ、最後に水10mlを加え、白金濃度1mMの白金ナノコロイド液66.4mlを得た。白金微粒子の粒径は、4.7±1.5nmであった。

2.白金ナノ粒子の屁臭と大便臭に対する低減効果の評価
ナノサイズ化された白金微粒子を作製し、経口摂取して糞中から白金金属を回収するとともに、屁と大便独特な悪臭の減少を定量化した。
白金ナノコロイドとしては、1mlあたり平均粒径6nm以下の白金微粒子を白金として180μgを含み、凝集した白金粒子を実質的に含まない白金ナノコロイド水を経口摂取用液として用いた。白金ナノコロイド0.9560%(白金;0.00019%、クエン酸三ナトリウム0.0028%)、クエン酸0.0045%、及び水99.0395%を含み、白金含有量は172±1.5ppm、pHは3.71であった。
ナノサイズ化された白金微粒子を作製し、経口摂取して糞中から白金金属を回収するとともに、屁と大便独特な悪臭の減少を定量化した。

クエン酸で還元されクエン酸をそのまま保護材とするナノサイズ化された白金微粒子(4.7±1.5nm)は、経口摂取で消化管から殆ど吸収されずに大腸にまで到達し糞中に99.96%は排泄されるという安全性が確保され、揮発性有機化合物(Volatile Organic Compounds;VOC)、煙、におい等、特にアンモニア、トルエン、硫化水素を鋭敏に検知し、新鮮な空気を1として、空気中のにおいの変化を総合的に判断するものとしてCIAQ(Composite Index of Air Quality)で数値化し表示するにおいモニターGT300-VOC(販売元:株式会社エムケー・サイエンティフィック)を用いて、ナノサイズ化された白金金属粒子の経口摂取前と経口摂取3週間後で屁と大便の悪臭を数値化し比較した。ヒト体重60kg当たり40μg/日の割合でナノサイズ化された白金微粒子を3週間経口摂取したところ、においモニターで計測された数値がナノサイズ化された白金金属粒子を3週間経口摂取後に危険率(p)0.01%未満で有意に減少し、屁と大便の悪臭がナノサイズ化された白金微粒子の経口摂取で統計学的有意に減少することが示された(表1)。なお、統計学的解析は、Studentのt-test検定法を使用した。
3.大便中の硫化水素低減効果の評価
(株)ガステック社製気体採取器セット(型式GV100S)を用いて、ナノサイズ化された白金微粒子の経口摂取前と後で大便から飛散するアンモニアと硫化水素を測定し、数値化して比較した。

具体的には、ヒトの大便、100~200gを琺瑯製おまるに分取し、秤量後素早くプレスオンシール(GLAD社製)で覆い気密性を維持しながら、概ね3分間後にそれぞれの検知管を用いておまる上空に保持された気体の濃度(ppm)を定量した。
