本発明に係る細胞培養用足場材料は、ポリビニルアルコール誘導体部と、ペプチド部とを有するペプチド含有ポリビニルアルコール誘導体を含む。上記ペプチド含有ポリビニルアルコール誘導体は、1種のみが用いられてもよく、2種以上が併用されてもよい。
上記ポリビニルアルコール誘導体部は、上記ペプチド含有ポリビニルアルコール誘導体において、上記ポリビニルアルコール誘導体に由来する構造部分である。上記ポリビニルアルコール誘導体は、ポリビニルアルコールによって誘導される化合物である。上記ポリビニルアルコール誘導体は、ポリビニルアセタール樹脂であることが好ましく、上記ポリビニルアルコール誘導体部は、ポリビニルアセタール樹脂部であることが好ましい。すなわち、上記ペプチド含有ポリビニルアルコール誘導体は、ポリビニルアセタール樹脂部と、上記ペプチド部とを有するペプチド含有ポリビニルアセタール樹脂であることが好ましい。上記ポリビニルアルコール誘導体及び上記ポリビニルアセタール樹脂は、それぞれ1種のみが用いられてもよく、2種以上が併用されてもよい。
上記ペプチド部は、上記ペプチド含有ポリビニルアルコール誘導体において、上記ペプチドに由来する構造部分である。上記ペプチド部は、アミノ酸配列を有する。上記ペプチド部を構成するペプチドは、オリゴペプチドであってもよく、ポリペプチドであってもよい。上記ペプチドは、1種のみが用いられてもよく、2種以上が併用されてもよい。
上記リンカー部は、上記ペプチド含有ポリビニルアルコール誘導体において、上記リンカーに由来する構造部分である。上記リンカー部は、上記ポリビニルアルコール誘導体部と上記ペプチド部との間に位置する。上記ポリビニルアルコール誘導体部と上記ペプチド部とが、上記リンカー部を介して結合している。上記リンカー部は、リンカー(架橋剤)によって形成される。上記リンカーは、1種のみが用いられてもよく、2種以上が併用されてもよい。
構成(B):上記ペプチド含有ポリビニルアルコール誘導体の水酸基価が1100mgKOH/g以下である。
以下の器具及び試薬を用意する。

(1)300mLの三角フラスコに、試料5gと溶媒100mLとを加えて、試料を溶媒に溶解させる。溶解後、JIS K8001に従って調整したフェノールフタレイン溶液を数滴添加する。
(2)25mlビュレットに滴定液を充填して滴定を行い、フェノールフタレイン溶液のうすい紅色呈色が30秒間続いたときを終点とする。
(3)同様にして空試験も行う。
(4)下記式(X)により酸価を算出する。

(3)放冷後、エタノール5mLで壁面を洗い流して希釈する。希釈後、フェノールフタレイン溶液を数滴添加する。
(4)25mlビュレットに滴定液を充填して滴定を行い、フェノールフタレイン溶液のうすい紅色呈色が30秒間続いたときを終点とする。
(5)同様にして空試験も行う。
(6)下記式(Y)により水酸基価を算出する。

本発明に係る細胞培養用足場材料は、細胞を培養するために用いられる。本発明に係る細胞培養用足場材料は、細胞を培養する際の該細胞の足場として用いられる。
本発明に係る細胞培養用容器は、容器本体と、上述した細胞培養用足場材料とを備え、上記容器本体の表面上に、該細胞培養用足場材料が配置されている。上記細胞培養用容器は、細胞の培養領域の少なくとも一部に上記細胞培養用足場材料を備える。
ポリビニルアセタール樹脂(PVB1)の作製:
攪拌装置を備えた反応機に、イオン交換水2700mL、平均重合度1700、鹸化度99モル%のポリビニルアルコールを300重量部投入し、攪拌しながら加熱溶解し、溶液を得た。得られた溶液に、触媒として、塩酸濃度が0.2重量%となるように35重量%塩酸を添加した。次いで、温度を15℃に調整し、攪拌しながらn-ブチルアルデヒド22重量部を添加した。次いで、n-ブチルアルデヒド148重量部を添加し、白色粒子状のポリビニルアセタール樹脂(ポリビニルブチラール樹脂)を析出させた。析出してから15分後に、塩酸濃度が1.8重量%になるように35重量%塩酸を添加した後、50℃に加熱し、50℃で2時間保持した。次いで、溶液を冷却し、中和した後、ポリビニルブチラール樹脂を水洗し、乾燥させて、ポリビニルアセタール樹脂(ポリビニルブチラール樹脂(PVB1)、平均重合度1700、アセタール化度(ブチラール化度)70モル%、水酸基量27モル%、アセチル化度3モル%)を得た。
得られたポリビニルアセタール樹脂99重量部と、アクリル酸(リンカー)1重量部とをTHF300重量部に溶解し、光ラジカル重合開始剤の存在下で、紫外線照射下で20分間反応させ、ポリビニルアセタール樹脂とアクリル酸とをグラフト共重合させることにより、リンカーを導入した。リンカーを導入したポリビニルアセタール樹脂1重量部をブタノール19重量部に溶解させた。得られた溶液150μLを、エアダスターで除塵したφ22mmのカバーガラス(松浪社製「22丸No.1」)の表面上に吐出し、スピンコーターを用いて2000rpm、20秒回転させた後、60℃で60分間加熱して、表面が平滑な樹脂膜を得た。
Arg-Gly-Asp-Serのアミノ酸配列を有する直鎖状のペプチド(アミノ酸残基数4個、表ではRGDSと記載)を用意した。このペプチド1重量部と、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド塩酸塩(縮合剤)1重量部とを、カルシウム及びマグネシウムの双方を含まないリン酸緩衝生理食塩水に該ペプチドの終濃度が1mMとなるよう添加し、ペプチド含有液を作製した。このペプチド含有液1重量部を、スピンコートした樹脂膜(リンカーを導入したポリビニルアセタール樹脂)に添加し、反応させて、リンカーのカルボキシル基と、ペプチドのArgのアミノ基とを脱水縮合した。このようにして、ポリビニルアセタール樹脂部と、リンカー部と、ペプチド部とを有するペプチド含有ポリビニルアセタール樹脂を作製した。
得られたペプチド含有ポリビニルアセタール樹脂とカバーガラスとの積層体を、φ22mmのポリスチレンディッシュに配置することにより細胞培養用容器を得た。
実施例1と同様にして、樹脂膜(リンカーを導入したポリビニルアセタール樹脂)を得た。
Arg-Gly-Asp-Phe-Lysのアミノ酸配列を有する環状のペプチド(アミノ酸残基数5個、ArgとLysとが結合することにより環状ペプチド骨格を形成、PheはD体、表ではc-RGDfKと記載)を用意した。このペプチド1重量部と、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド塩酸塩(縮合剤)1重量部とを、カルシウム及びマグネシウムの双方を含まないリン酸緩衝生理食塩水に該ペプチドの終濃度が1mMとなるように添加し、ペプチド含有液を作製した。このペプチド含有液1重量部を、スピンコートした樹脂膜(リンカーを導入したポリビニルアセタール樹脂)に添加し、反応させて、リンカーのカルボキシル基と、ペプチドのLysのアミノ基とを脱水縮合した。このようにして、ポリビニルアセタール樹脂部と、リンカー部と、ペプチド部とを有するペプチド含有ポリビニルアセタール樹脂を作製した。
実施例1と同様にして、細胞培養用容器を得た。
実施例1と同様にして、樹脂膜(リンカーを導入したポリビニルアセタール樹脂)を得た。
Gly-Arg-Gly-Asp-Serのアミノ酸配列を有する直鎖状のペプチド(アミノ酸残基数5個、表ではGRGDSと記載)を用意した。ペプチドの添加量を10重量部にしたこと以外は、実施例1と同様にして、ペプチド含有ポリビニルアセタール樹脂を作製した。
実施例1と同様にして、細胞培養用容器を得た。
リンカーの導入において、70重量部のPVB1と30重量部のアクリル酸(リンカー)とを用いたこと、ペプチド部の形成において、ペプチドの添加量を30重量部に変更したこと以外は、実施例3と同様にして、ペプチド含有ポリビニルアセタール樹脂及び細胞培養用容器を作製した。
リンカーの導入において、50重量部のPVB1と50重量部のアクリル酸(リンカー)とを用いたこと、ペプチド部の形成において、ペプチドの添加量を40重量部に変更したこと以外は、実施例3と同様にして、ペプチド含有ポリビニルアセタール樹脂及び細胞培養用容器を作製した。
リンカーの導入において、30重量部のPVB1と70重量部のアクリル酸(リンカー)とを用いたこと、ペプチド部の形成において、ペプチドの添加量を70重量部に変更したこと以外は、実施例3と同様にして、ペプチド含有ポリビニルアセタール樹脂及び細胞培養用容器を作製した。
ポリビニルアセタール樹脂(PVB2)の作製:
攪拌装置を備えた反応機に、イオン交換水2700mL、平均重合度1700、鹸化度99モル%のポリビニルアルコールを300重量部投入し、攪拌しながら加熱溶解し、溶液を得た。得られた溶液に、触媒として、塩酸濃度が0.2重量%となるように35重量%塩酸を添加した。次いで、温度を15℃に調整し、攪拌しながらn-ブチルアルデヒド22重量部を添加した。次いで、n-ブチルアルデヒド143重量部を添加し、白色粒子状のポリビニルアセタール樹脂(ポリビニルブチラール樹脂)を析出させた。析出してから15分後に、塩酸濃度が1.8重量%になるように35重量%塩酸を添加した後、50℃に加熱し、50℃で2時間保持した。次いで、溶液を冷却し、中和した後、ポリビニルブチラール樹脂を水洗し、乾燥させて、ポリビニルアセタール樹脂(ポリビニルブチラール樹脂(PVB2)、平均重合度1700、アセタール化度(ブチラール化度)69モル%、水酸基量28モル%、アセチル化度3モル%)を得た。
得られたポリビニルアセタール樹脂70重量部と、アクリル酸(リンカー)30重量部とをTHF300重量部に溶解し、光ラジカル重合開始剤の存在下で、紫外線照射下で20分間反応させ、ポリビニルアセタール樹脂とアクリル酸とをグラフト共重合させることにより、リンカーを導入した。リンカーを導入したポリビニルアセタール樹脂1重量部をブタノール19重量部に溶解させた。得られた溶液150μLを、エアダスターで除塵したφ22mmのカバーガラス(松浪社製「22丸No.1」)の表面上に吐出し、スピンコーターを用いて2000rpm、20秒回転させた後、60℃で60分間加熱して、表面が平滑な樹脂膜を得た。
得られた樹脂膜(リンカーを導入したポリビニルアセタール樹脂)を用いたこと以外は、実施例4と同様にして、ペプチド含有ポリビニルアセタール樹脂を作製した。
実施例1と同様にして、細胞培養用容器を得た。
ポリビニルアセタール樹脂(PVB3)の作製:
攪拌装置を備えた反応機に、イオン交換水2700mL、平均重合度1700、鹸化度99モル%のポリビニルアルコールを300重量部投入し、攪拌しながら加熱溶解し、溶液を得た。得られた溶液に、触媒として、塩酸濃度が0.2重量%となるように35重量%塩酸を添加した。次いで、温度を15℃に調整し、攪拌しながらn-ブチルアルデヒド22重量部を添加した。次いで、n-ブチルアルデヒド138重量部を添加し、白色粒子状のポリビニルアセタール樹脂(ポリビニルブチラール樹脂)を析出させた。析出してから15分後に、塩酸濃度が1.8重量%になるように35重量%塩酸を添加した後、50℃に加熱し、50℃で2時間保持した。次いで、溶液を冷却し、中和した後、ポリビニルブチラール樹脂を水洗し、乾燥させて、ポリビニルアセタール樹脂(ポリビニルブチラール樹脂(PVB3)、平均重合度1700、アセタール化度(ブチラール化度)67モル%、水酸基量30モル%、アセチル化度3モル%)を得た。
得られたポリビニルアセタール樹脂を用いたこと以外は実施例7と同様にして、リンカーを導入した。
得られた樹脂膜(リンカーを導入したポリビニルアセタール樹脂)を用いたこと以外は、実施例4と同様にして、ペプチド含有ポリビニルアセタール樹脂を作製した。
実施例1と同様にして、細胞培養用容器を得た。
ポリビニルアセタール樹脂(PVB4)の作製:
攪拌装置を備えた反応機に、イオン交換水2700mL、平均重合度1700、鹸化度99モル%のポリビニルアルコールを300重量部投入し、攪拌しながら加熱溶解し、溶液を得た。得られた溶液に、触媒として、塩酸濃度が0.2重量%となるように35重量%塩酸を添加した。次いで、温度を15℃に調整し、攪拌しながらn-ブチルアルデヒド22重量部を添加した。次いで、n-ブチルアルデヒド100重量部を添加し、白色粒子状のポリビニルアセタール樹脂(ポリビニルブチラール樹脂)を析出させた。析出してから15分後に、塩酸濃度が1.8重量%になるように35重量%塩酸を添加した後、50℃に加熱し、50℃で2時間保持した。次いで、溶液を冷却し、中和した後、ポリビニルブチラール樹脂を水洗し、乾燥させて、ポリビニルアセタール樹脂(ポリビニルブチラール樹脂(PVB4)、平均重合度1700、アセタール化度(ブチラール化度)50モル%、水酸基量47モル%、アセチル化度3モル%)を得た。
得られたポリビニルアセタール樹脂を用いたこと以外は実施例7と同様にして、リンカーを導入した。
得られた樹脂膜(リンカーを導入したポリビニルアセタール樹脂)を用いたこと以外は、実施例4と同様にして、ペプチド含有ポリビニルアセタール樹脂を作製した。
実施例1と同様にして、細胞培養用容器を得た。
実施例4と同様にして、樹脂膜(リンカーを導入したポリビニルアセタール樹脂)を得た。
Gly-Arg-Gly-Asp-Ser-Proのアミノ酸配列を有する直鎖状のペプチド(アミノ酸残基数6個、表ではGRGDSPと記載)を用意した。このペプチドを用いたこと以外は実施例4と同様にして、ポリビニルアセタール樹脂部と、リンカー部と、ペプチド部とを有するペプチド含有ポリビニルアセタール樹脂を作製した。
実施例1と同様にして、細胞培養用容器を得た。
平均重合度1700、鹸化度90モル%のポリビニルアルコール99重量部と、アクリル酸(リンカー)1重量部とをエタノール300重量部に溶解し、光ラジカル重合開始剤の存在下で、紫外線照射下で20分間反応させ、ポリビニルアルコールとアクリル酸とをグラフト共重合させることにより、リンカーを導入した。Gly-Arg-Gly-Asp-Serのアミノ酸配列を有する直鎖状のペプチド(アミノ酸残基数5個、表ではGRGDSと記載)を用いたこと、リンカーのカルボキシル基と、ペプチドのGlyのアミノ基とを脱水縮合したこと以外は、実施例1と同様にして、ペプチド含有ポリビニルアルコール誘導体及び細胞培養用容器を作製した。
実施例1と同様にして、樹脂膜(リンカーを導入したポリビニルアセタール樹脂)を得た。比較例2では、ペプチド部を形成しなかった。また、実施例1と同様にして、細胞培養用容器を得た。
天然物由来の足場材の作製:
リン酸バッファー(PBS)中に5μg/mlに調整したVitronectin(コーニング社製)溶液をφ35mmディッシュに1ml添加した。そこにφ22mmのカバーガラス(松浪社製「22丸No.1」)を浸漬させ、37℃で1時間養生することでVitronectinが表面に平滑に吸着した天然物由来の足場材(表中ではVTNと記載)を得た。
実施例1と同様にして、細胞培養用容器を得た。なお、Vitronectinは乾燥すると変性し、接着性能が大きく低下してしまうため、細胞培養用容器の作製後すぐにPBS溶液で浸漬した。
(1)水酸基価及び酸価の測定
JIS K0070に記載の中和滴定法で、上述した方法により、細胞培養用足場材料の水酸基価及び酸価を測定した。
得られたペプチド含有ポリビニルアセタール樹脂の樹脂膜(実施例1~10及び比較例1)、及びリンカーを導入したポリビニルアセタール樹脂の樹脂膜(比較例2)をPBS溶液に30分間浸漬した。浸漬後の樹脂膜を原子間力顕微鏡(AFM、Bruker社製「Dimension XR」)により観察した。QNMモードでピークセットポイントを2nNに設定する測定条件とし、1μm×1μmの範囲を観察した。得られた高さマッピング画像と弾性率マッピング画像とを比較して海島構造の有無を判断した。なお、表では、海島構造が観察される場合に「A」と記載し、海島構造が観察されない場合に「B」と記載した。
(3-1)iPS細胞の播種及び培養
以下の液体培地及びROCK(Rho結合キナーゼ)特異的阻害剤を用意した。
ROCK-Inhibitor(Y27632)
以下の液体培地及び添加剤を用意した。液体培地であるMesenCult-ACF Plus Mediumに終濃度が1Xとなるように、MesenCult-ACF Plus 500X Supplement及びL-Glutamine 200mmol/L(100X)を加えた。
MesenCult-ACF Plus 500X Supplement(Stemcell technologies社製)
L-Glutamine 200mmol/L(100X)(富士フイルム和光純薬社製)
細胞播種から24時間経過後の細胞を位相差顕微鏡(オリンパス社製「IX73」、10×20倍)を用いて観察した。観察において、細胞培養用容器内の最も平均的な接着形態を示す視野の画像を取得した。得られた画像から、シェイプファクター(SF)を求めることにより、仮足の伸展性を評価した。なお、シェイプファクター(SF)とは、細胞を培養した後の細胞の平面視における領域の形状評価係数であり、下記式により求められる。
○:SFが0.1以上0.6以下
×:SFが0.6を超え1以下
細胞播種から5日経過後の細胞を位相差顕微鏡にて観察した。細胞塊が播種時よりも大きくなっているか否かを目視にて確認した。
○:播種時と比べて細胞塊が大きくなっており、細胞の増殖が確認できる
×:播種時と比べて細胞塊が変化しない、又は、細胞塊が剥離しており、細胞塊が存在しない
細胞播種から5日経過後の細胞数をセルカウンター(Chemometec社製「NucleoCounter Nc-3000」)を用いて求めた。次に、下記式を用いて参考例Aに対する細胞増殖率を求めた。
AA:参考例Aに対する細胞増殖率が100%以上
A:参考例Aに対する細胞増殖率が50%以上100%未満
B:参考例Aに対する細胞増殖率が10%以上50%未満
C:参考例Aに対する細胞増殖率が10%未満

Claims (9)
- ポリビニルアルコール誘導体部と、ペプチド部とを有するペプチド含有ポリビニルアルコール誘導体を含み、
水酸基価が、1100mgKOH/g以下である、細胞培養用足場材料。 - ポリビニルアルコール誘導体部と、ペプチド部とを有するペプチド含有ポリビニルアルコール誘導体を含み、
前記ペプチド含有ポリビニルアルコール誘導体の水酸基価が、1100mgKOH/g以下である、細胞培養用足場材料。 - 前記ペプチド部が、細胞接着性のアミノ酸配列を有する、請求項1又は2に記載の細胞培養用足場材料。
- 前記細胞接着性のアミノ酸配列が、RGD配列、YIGSR配列、又はPDSGR配列を少なくとも有する、請求項3に記載の細胞培養用足場材料。
- 前記細胞接着性のアミノ酸配列が、下記式(1)で表されるRGD配列を少なくとも有する、請求項3又は4に記載の細胞培養用足場材料。

前記式(1)中、Xは、Gly、Ala、Val、Ser、Thr、Phe、Met、Pro、又はAsnを表す。 - 前記ペプチド部が、3個以上10個以下のアミノ酸により構成される、請求項1~5のいずれか1項に記載の細胞培養用足場材料。
- 前記ポリビニルアルコール誘導体部と前記ペプチド部とが、リンカー部を介して結合している、請求項1~6のいずれか1項に記載の細胞培養用足場材料。
- 前記ペプチド含有ポリビニルアルコール誘導体が、ポリビニルアセタール樹脂部と、前記ペプチド部とを有するペプチド含有ポリビニルアセタール樹脂である、請求項1~7のいずれか1項に記載の細胞培養用足場材料。
- 容器本体と、
請求項1~8のいずれか1項に記載の細胞培養用足場材料とを備え、
前記容器本体の表面上に、前記細胞培養用足場材料が配置されている、細胞培養用容器。
