本発明はトランスジェニックカイコ、および該カイコを用いた非天然アミノ酸含有タンパク質の製造方法に関する。
本願は、2017年4月12日に、日本に出願された特願2017-79294号に基づき優先権を主張し、その内容をここに援用する。

石油、金属、セラミックス等の天然資源は、将来の枯渇が懸念されている。枯渇のおそれの無い次世代素材としてシルクタンパク質が挙げられている。
シルクはそれ自身有用な特性を多く備えるが、その特性を改変することで、より有用な素材として産業上活用されると期待される。
例えば、既存のシルク素材を上回る強靭なシルク素材の開発や、デバイスのウェラブル化が進む中で、シルクタンパク質へのデバイス導入等が期待されている。また、生体親和性が高いシルクタンパク質の医療材料や化粧品材料等への応用が期待されている。
シルクはそれ自身有用な特性を多く備えるが、その特性を改変することで、より有用な素材として産業上活用されると期待される。
例えば、既存のシルク素材を上回る強靭なシルク素材の開発や、デバイスのウェラブル化が進む中で、シルクタンパク質へのデバイス導入等が期待されている。また、生体親和性が高いシルクタンパク質の医療材料や化粧品材料等への応用が期待されている。
シルクの特性を改変する方法として、カイコの遺伝子組換え技術を用いてシルクタンパク質の特性を改変させる方法が挙げられる。なかでも、タンパク質への非天然アミノ酸導入法は、非天然アミノ酸を構成要素として使用でき、天然にはない特性を有するタンパク質を創出できることから注目される。
タンパク質への非天然アミノ酸導入法には、二つの方法が知られている。一つは「部位特異的非天然アミノ酸導入法」である。この方法では、非天然アミノ酸の投与、及びターゲットタンパク質遺伝子配列の改変を必要とし、改変した配列のコドンに対応するアンチコドンを有するtRNA、及びそのtRNAを非天然アミノ酸でアミノアシル化するためのaaRS(アミノアシル-tRNA合成酵素)を用いる。しかしながら、この部位特異的非天然アミノ酸導入法では、ターゲットタンパク質遺伝子と目的のaaRS遺伝子の両方を改変させなければならず工程が煩雑である。また、ターゲットタンパク質遺伝子の配列が既知のものでなくてはならないという制限がある。
一方、二つ目の方法として「残基特異的非天然アミノ酸導入法」が知られている。この方法では、非天然アミノ酸の投与に加え、アミノ酸結合部位に変異を導入した変異型aaRSの強制発現によって非天然アミノ酸の取り込み効率を向上させる。この残基特異的非天然アミノ酸導入法は、ターゲットタンパク質遺伝子の配列を改変させなくとも実施可能なため、部位特異的非天然アミノ酸導入法に比べ利用が簡便であり、ターゲットタンパク質遺伝子の配列が未知であっても利用可能という利点等があるため、利用価値の高い技術である。
一方、二つ目の方法として「残基特異的非天然アミノ酸導入法」が知られている。この方法では、非天然アミノ酸の投与に加え、アミノ酸結合部位に変異を導入した変異型aaRSの強制発現によって非天然アミノ酸の取り込み効率を向上させる。この残基特異的非天然アミノ酸導入法は、ターゲットタンパク質遺伝子の配列を改変させなくとも実施可能なため、部位特異的非天然アミノ酸導入法に比べ利用が簡便であり、ターゲットタンパク質遺伝子の配列が未知であっても利用可能という利点等があるため、利用価値の高い技術である。
本発明者は、残基特異的非天然アミノ酸導入法を改良し、天然アミノ酸と大きく構造が異なる非天然アミノ酸等のある種の非天然アミノ酸(例えばp‐N3‐Phe)を含有するタンパク質をカイコにおいて製造することに成功した(非特許文献1参照)。

更に、少量の非天然アミノ酸で、高効率に非天然アミノ酸を含有するタンパク質の製造方法が求められている。
本発明は、上記事情に鑑みてなされたものであり、少量の非天然アミノ酸で、高効率に非天然アミノ酸含有タンパク質を製造できるトランスジェニックカイコ、及び該トランスジェニックカイコを用いた非天然アミノ酸含有タンパク質の製造方法を提供する。
本発明者は、上記課題を解決すべく鋭意研究した結果、tRNAに所望の非天然アミノ酸を結合させる活性を有するタンパク質をコードするDNAを絹糸腺特異的に発現するトランスジェニックカイコを見出し、本発明を完成させた。
すなわち、本発明は、下記の特徴を有するトランスジェニックカイコ、及び該トランスジェニックカイコを用いた非天然アミノ酸含有タンパク質の製造方法を提供するものである。
(1)以下の(a)~(c)のいずれか一つのアミノ酸配列を含む配列からなり、且つ、フェニルアラニン特異的tRNAに所望の非天然アミノ酸を結合させる活性を有するタンパク質をコードするDNA、及び、該DNAを絹糸腺特異的に発現させるプロモーターを有することを特徴とするトランスジェニックカイコ。
(a)配列番号1に示されるアミノ酸配列のアミノ酸番号432位に変異を有するアミノ酸配列、
(b)前記(a)のアミノ酸番号432位以外の部位において1乃至数個のアミノ酸が欠失、挿入、置換若しくは付加されたアミノ酸配列、

本発明によれば、少量の非天然アミノ酸で、高効率に非天然アミノ酸含有タンパク質を製造できるトランスジェニックカイコ、及び該トランスジェニックカイコを用いた非天然アミノ酸含有タンパク質の製造方法を提供できる。
《トランスジェニックカイコ》
本発明のトランスジェニックカイコは、以下の(a)~(c)のいずれか一つのアミノ酸配列を含む配列からなり、且つ、フェニルアラニン特異的tRNAに所望の非天然アミノ酸を結合させる活性を有するタンパク質をコードするDNA、及び、該DNAを絹糸腺特異的に発現させるプロモーターを有することを特徴とするトランスジェニックカイコである。
(a)配列番号1に示されるアミノ酸配列のアミノ酸番号432位に変異を有するアミノ酸配列、
(b)前記(a)のアミノ酸番号432位以外の部位において1乃至数個のアミノ酸が欠失、挿入、置換若しくは付加されたアミノ酸配列、

tRNAのアミノアシル化は、それぞれのアミノ酸に対応するアミノアシル-tRNA合成酵素(aaRS)によって触媒される。aaRSはタンパク質を構成する20種のアミノ酸間で厳密な基質特異性を有している。例えば、フェニルアラニンに対応するaaRSは、フェニルアラニル-tRNA合成酵素(PheRS)と呼ばれる。前記(a)~(c)のアミノ酸配列を有するタンパク質をコードするDNAは、カイコPheRSのαサブユニットに変異を有するDNAであって、フェニルアラニン特異的tRNAに所望の非天然アミノ酸を結合させる活性を有するタンパク質をコードするDNAである。
このフェニルアラニン特異的tRNAに所望の非天然アミノ酸を結合させる活性を有するタンパク質は、該活性を有していればよいのであって、前記(a)~(c)のいずれかの配列に加えて、タンパク質タグ等の任意の配列(例えば、ヒスチジンタグ、HAタグ、GFP等)をさらに有していてもよい。
前記(a)~(c)のいずれかの配列を有するタンパク質をコードするDNAを絹糸腺で発現させることで、絹糸腺で作られるタンパク質に対し本来のコドンとは通常は対応しないアミノ酸(非天然アミノ酸)を導入することが可能である。更に、絹糸腺で作られるタンパク質に対し、少量の該非天然アミノ酸で、高効率に該非天然アミノ酸を導入することができる。
このフェニルアラニン特異的tRNAに所望の非天然アミノ酸を結合させる活性を有するタンパク質は、該活性を有していればよいのであって、前記(a)~(c)のいずれかの配列に加えて、タンパク質タグ等の任意の配列(例えば、ヒスチジンタグ、HAタグ、GFP等)をさらに有していてもよい。
前記(a)~(c)のいずれかの配列を有するタンパク質をコードするDNAを絹糸腺で発現させることで、絹糸腺で作られるタンパク質に対し本来のコドンとは通常は対応しないアミノ酸(非天然アミノ酸)を導入することが可能である。更に、絹糸腺で作られるタンパク質に対し、少量の該非天然アミノ酸で、高効率に該非天然アミノ酸を導入することができる。
実施例において後述するように、本発明のトランスジェニックカイコは、前記(a)のアミノ酸番号432位が、アラニン、バリン、ロイシン、メチオニン、スレオニン、システイン、又はチロシンであることが好ましく、アラニン、バリン、メチオニンがより好ましい。
即ち、本発明のトランスジェニックカイコは、以下の(d)~(h)のいずれか一つのアミノ酸配列を含む配列からなり、且つ、フェニルアラニン特異的tRNAに所望の非天然アミノ酸を結合させる活性を有するタンパク質をコードするDNA、及び、該DNAを絹糸腺特異的に発現させるプロモーターを有することが好ましい。
(d)配列番号2(Phe432Val)に示されるアミノ酸配列、
(e)配列番号3(Phe432Ala)に示されるアミノ酸配列、
(f)配列番号4(Phe432Met)に示されるアミノ酸配列、

ここで、欠失、挿入、置換若しくは付加されたアミノ酸の数としては、1~50個が好ましく、1~20個がより好ましく、1~10個が更に好ましく、1~5個が特に好ましく、1~2個が最も好ましい。
同一性としては、85%以上が好ましく、90%以上がより好ましく、95%以上が更に好ましく、99%以上が特に好ましい。

更に、本発明のトランスジェニックカイコは、アミノ酸番号432位の変異に加えて、アミノ酸番号407位に変異を有していてもよい。アミノ酸番号407位における変異としては、アラニン又はグリシンが好ましい。
前記絹糸腺で作られる非天然アミノ酸含有タンパク質は特に制限されることはなく、カイコが絹糸腺において生産するシルクタンパク質が好ましい。また、絹糸腺において任意のタンパク質をコードする外来性のDNAを強制発現させ、任意のタンパク質を絹糸腺に生産させてもよい。
絹糸腺で作られるタンパク質は繭糸としてカイコ体外に分泌されるため、特別の回収方法を用いずとも回収が容易であり、また多量のタンパク質を容易に得ることができる。
また、前記DNAをトランスジェニックカイコ全体に発現させることによって生じる毒性を回避するという観点から、本発明においては、絹糸腺において前記DNAを発現させるプロモーターが用いられる。絹糸腺は、前部絹糸腺、中部絹糸腺及び後部絹糸腺に大別される。ここで前記DNAを絹糸腺特異的に発現させるプロモーターとしては、前記DNAを絹糸腺特異的に発現させることができるプロモーターであれば特に制限されず、上記三つの部位のいずれか一つ又は二つ以上の組み合わせからなる部位で発現させることができるプロモーターであってもよい。
中部絹糸腺特異的に発現させるプロモーターとして、例えば、セリシン1タンパク質またはセリシン2タンパク質をコードするDNAのプロモーターが挙げられる。中部絹糸腺特異的に発現させるプロモーターを用いることにより、中部絹糸腺で生産されるタンパク質に非天然アミノ酸を導入することができる。カイコの中部絹糸腺で生産されるタンパク質としてはセリシンが挙げられる。
また、後部絹糸腺特異的に発現させるプロモーターとしてはフィブロインL鎖(FibL)タンパク質をコードするDNAのプロモーターが挙げられる。後部絹糸腺特異的に発現させるプロモーターを用いることにより、後部絹糸腺で生産されるタンパク質に非天然アミノ酸を導入することができる。カイコの後部絹糸腺で生産されるタンパク質はフィブロインであり、フィブロインがカイコの繭糸タンパク質の大部分を占めているため、後部絹糸腺特異的に発現させるプロモーターを用いることで、シルクタンパク質の性質の制御がより容易となる。
中部絹糸腺特異的に発現させるプロモーターとして、例えば、セリシン1タンパク質またはセリシン2タンパク質をコードするDNAのプロモーターが挙げられる。中部絹糸腺特異的に発現させるプロモーターを用いることにより、中部絹糸腺で生産されるタンパク質に非天然アミノ酸を導入することができる。カイコの中部絹糸腺で生産されるタンパク質としてはセリシンが挙げられる。
また、後部絹糸腺特異的に発現させるプロモーターとしてはフィブロインL鎖(FibL)タンパク質をコードするDNAのプロモーターが挙げられる。後部絹糸腺特異的に発現させるプロモーターを用いることにより、後部絹糸腺で生産されるタンパク質に非天然アミノ酸を導入することができる。カイコの後部絹糸腺で生産されるタンパク質はフィブロインであり、フィブロインがカイコの繭糸タンパク質の大部分を占めているため、後部絹糸腺特異的に発現させるプロモーターを用いることで、シルクタンパク質の性質の制御がより容易となる。
前記プロモーターを用いる際には、同時に、前記DNAの発現、その後のタンパク質への翻訳等を制御するための他の配列を用いてもよく、そのような配列としては例えば3’UTR(非翻訳領域)、5’UTR配列等が挙げられる。
本発明のトランスジェニックカイコの作出に用いるホストのカイコとしては、後述する本発明の非天然アミノ酸含有タンパク質の製造方法において、非天然アミノ酸を含むタンパク質を製造できるカイコであれば特に制限されず、例えば、遺伝子組換え作成用として広く用いられるw1-pnd等の系統を用いても構わないが、飼料として投与された非天然アミノ酸のカイコ個体への取り込み量を増加させるという観点から、人工飼料摂食性に優れたカイコを用いることが好ましい。例えば、日01号、日509号、日510号、日603号、日604号、中604号、中605号等の品種が好ましく、非天然アミノ酸を含む人工飼料の1個体あたりの摂食量が多い白/CS(系統名:MCS601)がより好ましい。
また、本発明のトランスジェニックカイコを実用系統と交配したF1交雑種を作出してもよい。実用系統としては、日509号×日510号、日603号×日604号、中509号×中510号、中604号×中605号等が挙げられる。

また、本発明のトランスジェニックカイコの作出に用いるカイコとしては、前述のとおり特に制限されず、非休眠卵を産下する性質のカイコまたは休眠卵を産下する性質のカイコの、どちらも用いることが可能である。休眠卵を産下する性質のカイコを用いる場合には、「低温暗催青法」を用いて非休眠卵を産下させることができる。すなわち、胚子を胸肢発生期以後、特に剛毛発生期以後を15℃程度に保護する。この低温期間は、日数にして10日以上必要である。胚の反転から頭部着色の頃までを暗黒に保つ。その後孵化した幼虫を飼育して得たカイコ蛾は非休眠卵を産下する。(蚕種総論 著者:高見丈夫 発行:全国蚕種協会 (昭和44年5月31日))。
低温暗催青法以外の方法として、幼虫期の光条件をコントロールすることで非休眠卵を産下させる方法(特開2003-88274参照)を採択してもよい。
低温暗催青法等の方法を用いても非休眠卵が得られないカイコを使用する場合は、カイコ卵を浸酸処理することで、卵を休眠打破してもよい。前記浸酸処理の具体的な方法として、採卵から3~4時間後のカイコ卵を室温で30分間程度塩酸に浸漬させ、水洗により塩酸を洗い流した後、卵を風乾させることが挙げられる。前記塩酸は、例えば比重1.11のものを用いることができる(Ai-Chun Zhao et.al., Insect Science (2012) Vol.19, pp.172-182.参照)。
トランスジェニックカイコの作出方法としては、例えばカイコ卵へDNAを導入する方法が挙げられる。カイコ卵へのDNAの導入は、例えば、カイコの発生初期卵へ、トランスポゾンをベクターとして注射する方法(Tamura,T. ,et.al.,Nature Biotechnology,(2000)Vol.18, pp.81-84.参照)に従って行うことができる。
例えば、トランスポゾンの逆位末端反復配列(Handler,A.M.,et.al., Proc. Natl. Acad. Sci. U.S.A.,(1998)Vol.95, pp.7520-7525.参照。)の間に上記DNAを挿入したベクターとともに、トランスポゾン転移酵素をコードするDNAを有するベクター(ヘルパーベクター)をカイコ卵に導入する方法が挙げられる。ヘルパーベクターとしては、pHA3PIG (Tamura, T., et.al., Nature Biotechnology,(2000)Vol.18, pp.81-84.参照。)が挙げられるが、これに限定されるものではない。
本発明におけるトランスポゾンとしては、piggyBacが好ましいが、これに限定されるものではなく、マリナー(mariner)、ミノス(minos)等を用いることもできる(Shimizu,K.,Insect Mol.Biol.,(2000)Vol.9,pp.277-281.参照。)。
また、本発明のトランスジェニックカイコの作出にはバキュロウイルスベクターを使用することもできる(Yamao,M.,et.al., Genes Dev.(1999)Vol.13,pp.511-516.参照。)。
上述したトランスポゾンをベクターとして注射する方法、バキュロウイルスベクターを使用する方法、及びその他のカイコ卵へのDNAの導入方法は前記アミノ酸配列を有するタンパク質をコードするDNAに対して用いてもよく、また、本発明のトランスジェニックカイコを用いた非天然アミノ酸含有タンパク質の製造において、製造される任意のタンパク質をコードするDNAに対して用いてもよい。
カイコ卵へのDNAの導入は、当業者においては一般的な方法により実施することができる。例えば、以下のように、特開2012-187123号公報に記載の方法で実施することができる。
カイコ卵へDNA注入用の管を使用して直接卵内へDNAを導入することが可能であるが、好ましい態様としては、前もって物理的または化学的に卵殻に穴を空け、該穴からDNAを導入する。

本発明において、物理的に卵殻に穴を空ける方法としては、例えば針、微小レーザー等を用いて穴を空ける方法が挙げられる。中でも、針を用いた方法によって卵殻に穴を空ける方法が好ましい。用いられる針としては、カイコの卵殻に穴を空けることができるものであれば、その材質、強度等は、特に制限されない。一方、化学的に卵殻に穴を空ける方法としては、例えば薬品(次亜塩素酸等)等を用いて穴を空ける方法が挙げられる。
本発明において、DNA導入方法としては、上記のカイコ卵に物理的または化学的に穴を空け、DNA注入用の管を該穴から卵内に挿入し、DNAを注入する工程を、針とDNA注入用の管が一体型となったマニュピュレーターを使用して行うことが好ましい。マニュピュレーターを構成要素の1つとする装置を使用できる。
このような装置しては、解剖顕微鏡、照明装置、可動式のステージ、顕微鏡に金具で固定した粗動マニュピュレーター、このマニュピュレーターに付けたマイクロマニュピュレーター、DNAを注射するための空気圧を調整するインジェクターから構成されるものが挙げられる。なお、使用される装置としては、特許第1654050号公報に記載の装置または該装置を改良した装置が挙げられる。
カイコ卵にDNAが導入されたか否かは、例えば、注射したDNAを卵から再度抽出して測定する方法(Nagaraju,J., et.al., Appl.Entomol.Zool.,(1996)Vol.31,pp.589-598.参照。)や、注射した遺伝子としてのDNAの卵内での発現を見る方法(Tamura,T.,et.al.,(1990)Jpn. J. Genet.,Vol.65,pp.401-410.参照。)等によって確認することができる。カイコ卵に導入されたDNAが、カイコの染色体に組込まれ、遺伝子組換えカイコが得られたか否かは、たとえばDNAを注射した卵から育った成虫を交配し(相互に、あるいは非組換えカイコと)受精卵を得、その産卵後6~10日にマーカー遺伝子(たとえば、EGFPや、DsRed,CFP,YFPなど)の発現を蛍光顕微鏡で観察することや、体色マーカー遺伝子(たとえば、Bm-aaNATなど)の発現を目視で確認することによって可能である。
カイコ卵に導入したDNAがカイコにおいて発現しているかどうかは、インジェクションした卵(G0世代)から成長した個体が産んだ卵(G1世代)については、マーカー遺伝子の発現を蛍光顕微鏡で観察して確認する。なお、カイコからトータルRNAを抽出し、抽出したRNAをテンプレートに、RT-PCRにより導入遺伝子の確認をしてもよい。
また、カイコに導入したDNAがカイコ個体のどの部位で発現しているのかを確認する方法は、例えば、幼虫から組織を摘出し、各組織別に発現を確認する方法や、CFP、GFP、EGFP、YFP、DsRED等の既知のレポーター遺伝子等を用い、蛍光顕微鏡を用いてカイコ個体を観察する方法等によって確認することができる。
《非天然アミノ酸含有タンパク質の製造方法》
本発明の非天然アミノ酸含有タンパク質の製造方法は、本発明のトランスジェニックカイコに非天然アミノ酸を投与する工程を有する。ここで非天然アミノ酸とは、タンパク質を構成しないアミノ酸を意味する。タンパク質を構成するアミノ酸とは、20種の標準天然アミノ酸、セレノシステイン、及びピロリジンを意味する。すなわち、本発明において、非天然アミノ酸とは、タンパク質を構成するこれらアミノ酸の類縁体(修飾アミノ酸)、又はタンパク質を構成するこれらアミノ酸以外の任意のアミノ酸を意味する。
本発明の非天然アミノ酸含有タンパク質の製造方法は、本発明のトランスジェニックカイコに非天然アミノ酸を投与する工程を有する。ここで非天然アミノ酸とは、タンパク質を構成しないアミノ酸を意味する。タンパク質を構成するアミノ酸とは、20種の標準天然アミノ酸、セレノシステイン、及びピロリジンを意味する。すなわち、本発明において、非天然アミノ酸とは、タンパク質を構成するこれらアミノ酸の類縁体(修飾アミノ酸)、又はタンパク質を構成するこれらアミノ酸以外の任意のアミノ酸を意味する。
本発明のトランスジェニックカイコはフェニルアラニン特異的tRNAに所望の非天然アミノ酸を結合させる活性を有するタンパク質をコードするDNA配列を有する。そのため、用いられる非天然アミノ酸はフェニルアラニン類縁体であることが好ましい。フェニルアラニン類縁体としては、フェニルアラニンの側鎖の少なくとも一つ以上の水素原子が置換基によって置換されていることが好ましく、フェニルアラニン側鎖のベンゼン環の水素原子が置換されていることがより好ましく、前記ベンゼン環のパラ位が置換されていることが特に好ましい。前記置換基としては、例えば、フッ素、塩素、臭素、ヨウ素等のハロゲン原子;エチニル基;シアノ基;アジド基等の置換が挙げられる。
即ち、フェニルアラニン類縁体としては、下記一般式(1)で表されるアミノ酸が好ましい。

本発明のトランスジェニックカイコを用いた非天然アミノ酸含有タンパク質の製造にあたって、タンパク質に含有される非天然アミノ酸の種類及び含有量を確認する方法としては、種々の方法を選択できるが、質量分析法による分析を行うのが好ましく、MALDI-TOF-MS法により質量スペクトルを測定する方法が特に好ましい。
本発明によれば、製造される非天然アミノ酸含有タンパク質に、本来のタンパク質の性質や機能を向上又は改変させることができる。更には、本来のタンパク質にない新たな性質や機能を付与することができる。例えば、非天然アミノ酸の導入手法をシルクタンパク質に適用することで、シルクタンパク質に非天然アミノ酸を含有させ、本来のシルクにはない性質や機能を付与することができる。
本発明の非天然アミノ酸含有タンパク質の製造方法を用いることで、従来の改変手法における問題点を解決できる。例えば、化学反応にともなう毒性の懸念が小さい、「後加工」であるため機能性分子が失活しないなどの利点があり、特異的な化学反応が可能な官能基の導入により、従来の「化学的方法」によらない精密かつクリーンなタンパク質の化学改変を行うことが可能である。例えば、アジド基又はエチニル基を有する非天然アミノ酸をシルクタンパク質にとりこませることで、エチニル基又はアジド基との反応を介して、任意のタンパク質、ペプチド、薬剤、色素、糖鎖、架橋分子、キレート分子等の分子をシルクに結合させることが可能である。このような改変により、シルクの物性そのものを変化させることが可能である。従って、カイコが絹糸腺において生産するタンパク質の有用性を飛躍的に増大させる。また、シアノ基やフッ素など特徴的な赤外吸収や核磁気共鳴を示す原子(団)を残基特異的に導入することで、これまで不可能であった局所的なシルクの構造情報が得られる。
前記シルク材料の形態は特に制限されず、例えば、溶液状、パウダー状、繊維状、スポンジ状、フィルム状、ブロック状などの形態が挙げられる。
本発明で用いる非天然アミノ酸は、天然アミノ酸と同時に用いることができ、用いる非天然アミノ酸は1種類のみでもよく、2種類以上の非天然アミノ酸を同時に用いることもできる。
本発明におけるカイコの飼育方法は、当業者において一般的なカイコの飼育方法に従い、非天然アミノ酸を含有する飼料を与え、行うことができる。例えば、「文部省(1978) 蚕種製造.pp193、実教出版社、東京」に記載の方法に従って飼育を行うことができる。
また、光によって化学反応が引き起こされる官能基を有する非天然アミノ酸をカイコに給餌する場合、非天然アミノ酸に光が照射されない条件下で、カイコを飼育することが好ましい。好ましい飼育方法として、光を一切照射しない連続暗期環境下でカイコを飼育する方法が挙げられる。または、カイコには光が照射されていてもよいが、前記の光によって化学反応が引き起こされる官能基を有する非天然アミノ酸には光が照射されない条件下で、カイコを飼育する方法が挙げられ、例えば、給餌する餌の周囲の空間を物理的に光源から遮蔽し、該非天然アミノ酸をカイコに与え、さらに、カイコが絹糸腺から該非天然アミノ酸を含有するタンパク質を生産する際に、該タンパク質に光が照射されない環境でカイコを飼育する等の方法が挙げられる。
本発明のトランスジェニックカイコを用いたタンパク質製造方法は、残基特異的非天然アミノ酸導入法である。したがって、本発明によれば該カイコに投与する非天然アミノ酸の種類を変えるだけで、同一の系統のトランスジェニックカイコから任意の種類の非天然アミノ酸を含有するタンパク質を製造することができる。
次に実施例を示して本発明をさらに詳細に説明するが、本発明は以下の実施例に限定されるものではない。
[変異型カイコPheRSの作製]
p-N3-Pheの認識に優れる変異型カイコPheRSを作製するため、ヒトPheRSの立体構造解析(Structure of human cytosolic phenylalanyl-tRNA synthetase: evidence for kingdom-specific design of the active sites and tRNA binding patterns. Finarov I, Moor N, Kessler N, Klipcan L, Safro MG. Structure. 2010 Mar 10;18(3):343-53.)の情報から、Asn404→Ala、Pro405→Gly、Tyr406→Ala、Glu408→Ala、Phe432→Alaの5つの変異体を作製し、これらの変異体がPheRSとしての活性を有しているか調べたところ、全てPheRSとしての活性を有していることが認められた。
これらの変異型カイコPheRS、又はThr407Gly、若しくはThr407Alaの変異型カイコPheRSと、酵母tRNAPheと、を共発現する大腸菌を、p-N3-Pheを含む培地で培養した。大腸菌に増殖阻害が認められれば、p-N3-Pheがタンパク質合成に取り込まれたものと判断できる。作製した変異体の中で、Thr407Gly、Thr407Ala、Phe432Alaで顕著な増殖阻害が認められた。 更に、Phe432X(X=20種類のアミノ酸)、とThr407X’(X’=Thr,Gly,Ala)組み合わせた変異体を作製し、各変異体の増殖阻害を検討した。結果を図1に示す。図1中、色が濃いほど強い増殖阻害が認められたことを示す。
図1に示すように、Phe432Val, Phe432Ala, Thr407Ala/Phe432Val, Thr407Ala/Phe432Metの4つがp-N3-Pheの認識に優れていることが確認された。
[変異型カイコPheRSの作製]

[組み換えベクターの作製]
当業者の常識の範囲である既存の手法に準じ、配列番号1に示されるアミノ酸配列をコードするDNA(配列番号7)を用いて、配列番号2(Phe432Val)、配列番号3(Phe432Ala)、配列番号5(Thr407Ala/Phe432Val)、及び配列番号6(Thr407Ala/Phe432Met)に示されるアミノ酸配列をコードするDNA(BmFRS1*;(配列番号8:配列番号2(Phe432Val)に示されるアミノ酸配列をコードするDNA、配列番号9:配列番号3(Phe432Ala)に示されるアミノ酸配列をコードするDNA、配列番号10:配列番号5(Thr407Ala/Phe432Val)、配列番号11:配列番号6(Thr407Ala/Phe432Met)に示されるアミノ酸配列をコードするDNA))を有するpiggyBacベクター(5塩基の5’UTR配列を含む)(図2)をそれぞれ作製した。なお、上記ベクターにはフィブロインL鎖(FibL)由来のプロモーターおよび3’UTR配列をそれぞれ、上記BmFRS1*の5’および3’側に付加した。FibL由来のプロモーター配列を付加することにより、目的遺伝子を後部絹糸腺で特異的に発現させることが可能である。これにより、変異型遺伝子が幼虫の全身で発現することで生じると想定される幼虫の発育不全を回避することができた。
当業者の常識の範囲である既存の手法に準じ、配列番号1に示されるアミノ酸配列をコードするDNA(配列番号7)を用いて、配列番号2(Phe432Val)、配列番号3(Phe432Ala)、配列番号5(Thr407Ala/Phe432Val)、及び配列番号6(Thr407Ala/Phe432Met)に示されるアミノ酸配列をコードするDNA(BmFRS1*;(配列番号8:配列番号2(Phe432Val)に示されるアミノ酸配列をコードするDNA、配列番号9:配列番号3(Phe432Ala)に示されるアミノ酸配列をコードするDNA、配列番号10:配列番号5(Thr407Ala/Phe432Val)、配列番号11:配列番号6(Thr407Ala/Phe432Met)に示されるアミノ酸配列をコードするDNA))を有するpiggyBacベクター(5塩基の5’UTR配列を含む)(図2)をそれぞれ作製した。なお、上記ベクターにはフィブロインL鎖(FibL)由来のプロモーターおよび3’UTR配列をそれぞれ、上記BmFRS1*の5’および3’側に付加した。FibL由来のプロモーター配列を付加することにより、目的遺伝子を後部絹糸腺で特異的に発現させることが可能である。これにより、変異型遺伝子が幼虫の全身で発現することで生じると想定される幼虫の発育不全を回避することができた。
[トランスジェニックカイコの作出と組換え遺伝子の発現確認]
受精卵へのpiggyBacベクターのインジェクションおよびその後の組換え体のスクリーニング等は、当業者の常識の範囲である既存の方法に準じて行った。なお、白/CS(系統名:MCS601)は休眠卵を産下し、前述の低温暗催青法による非休眠化ができない系統であるため、前述の浸酸処理によって卵の休眠打破を行った。ゲノム中に挿入された変異型遺伝子のコピー数を、リアルタイムPCR法を用いて解析し、変異型遺伝子をホモで有するトランスジェニックカイコの系統を確立した。これらのトランスジェニックカイコの系統を、それぞれH06(Phe432Val)、H07(Phe432Ala)、H08(Thr407Ala/Phe432Val)、H09(Thr407Ala/Phe432Met)と名付けた。

[p-N3-Phe含有シルクタンパク質の製造]
(実験例1)
上記トランスジェニックカイコH06~H09を、5齢起蚕から2日目まで通常組成の人工飼料を給餌した後、3日目以降、0.1%(dry diet換算)のp-N3-Phe(一般式(1)で表されるアミノ酸において、R=N3)を含む人工飼料を給餌した。3日目以降のカイコの飼育は、p-N3-Pheの光反応を避けるため、遮光条件下にて行った。対照として、H03(Thr407Ala)を用いた。カイコの一頭当たりのフィブロイン生産量とp-N3-Pheの取り込み量(質量分析ピークの強度比)を図3~4に示す。
図3~4において、縦軸(左)は、棒グラフで表されたカイコの一頭当たりのフィブロイン生産量を示し、縦軸(右)は、丸プロットで表されたフィブロイン中のp-N3-Pheのピーク強度の割合を示す。
(実験例1)
上記トランスジェニックカイコH06~H09を、5齢起蚕から2日目まで通常組成の人工飼料を給餌した後、3日目以降、0.1%(dry diet換算)のp-N3-Phe(一般式(1)で表されるアミノ酸において、R=N3)を含む人工飼料を給餌した。3日目以降のカイコの飼育は、p-N3-Pheの光反応を避けるため、遮光条件下にて行った。対照として、H03(Thr407Ala)を用いた。カイコの一頭当たりのフィブロイン生産量とp-N3-Pheの取り込み量(質量分析ピークの強度比)を図3~4に示す。
図3~4において、縦軸(左)は、棒グラフで表されたカイコの一頭当たりのフィブロイン生産量を示し、縦軸(右)は、丸プロットで表されたフィブロイン中のp-N3-Pheのピーク強度の割合を示す。
フィブロイン中のp-N3-Pheのピーク強度は、以下の方法で算出した。
トランスジェニックカイコの繭層を約50 mg/mLの濃度になるように8M LiBr水溶液に35℃で40分間加熱して溶解させた。この溶解液を8M 尿素水溶液に約4.5 mg/mL
の濃度になるように希釈し、さらに還元剤を含むサンプルバッファーと混合して約3 mg/mLの濃度に調整した後、室温で30分以上静置して還元処理を行った。還元処理した試料液をSDS-PAGEで分離し、FibLのバンドを切り出して脱色・洗浄・乾燥処理した後、トリプシンにより37℃で一晩ゲル内消化した。生じたペプチド断片を抽出して乾燥固化した後、0.1%TFA水溶液とアセトニトリルの2:1混合液に溶解した。この溶解液をHCCAで飽和させた0.1%TFA水溶液とアセトニトリルの2:1混合液と混合し、MALDIターゲットプレート上に滴下して乾燥させ、測定データを取得した。
p-N3-Pheを投与したトランスジェニックカイコにおいて、Pheからp-NH2-Pheへの置換にともなう質量増加(理論値:14N1H1H-1H = +15.01 Da)が観察される。N3基はSDS-PAGEサンプル調製時の還元処理及び/あるいはMALDI-TOF-MS測定中にNH2基へ還元されたと考えられる。係る測定データからPhe含有ペプチド断片に対するp-N3-Phe含有ペプチド断片の相対的ピーク強度を求めた。

p-N3-Pheを投与したトランスジェニックカイコにおいて、Pheからp-NH2-Pheへの置換にともなう質量増加(理論値:14N1H1H-1H = +15.01 Da)が観察される。N3基はSDS-PAGEサンプル調製時の還元処理及び/あるいはMALDI-TOF-MS測定中にNH2基へ還元されたと考えられる。係る測定データからPhe含有ペプチド断片に対するp-N3-Phe含有ペプチド断片の相対的ピーク強度を求めた。
図4に示されるように、0.1%のp-N3-Pheを含む人工飼料を与えたH09におけるフィブロインの生産量は、H03におけるフィブロインの生産量と同等であった。 更に、図3~4に示されるように、カイコが生産したフィブロイン中のp-N3-Pheの取り込み量は、H06~H09がH03に対して優位に多かった。
(実験例2)
トランスジェニックカイコH06、H07において、給餌する人工飼料中のp-N3-Phの含有割合を変えたときのカイコの単位量当たりのフィブロイン生産量、及びp-N3-Pheの取り込み量を確認した。結果を図5~6に示す。図5~6において、横軸は、人工飼料中のp-N3-Pheの含有割合(dry diet換算)を示し、縦軸(左)は、棒グラフで表されたカイコの単位量当たりのフィブロイン生産量を示し、縦軸(右)は、丸プロットで表されたフィブロイン中のp-N3-Pheのピーク強度の割合を示す。
トランスジェニックカイコH06、H07において、給餌する人工飼料中のp-N3-Phの含有割合を変えたときのカイコの単位量当たりのフィブロイン生産量、及びp-N3-Pheの取り込み量を確認した。結果を図5~6に示す。図5~6において、横軸は、人工飼料中のp-N3-Pheの含有割合(dry diet換算)を示し、縦軸(左)は、棒グラフで表されたカイコの単位量当たりのフィブロイン生産量を示し、縦軸(右)は、丸プロットで表されたフィブロイン中のp-N3-Pheのピーク強度の割合を示す。
図5において、H06では、合成飼料中に0.05%のp-N3-Pheを含有することで、p-N3-Pheを導入したフィブロインを約100mg産生することができ、取り込まれたp-N3-Pheのピーク強度の割合が約0.15であった。非特許文献1におけるH03の結果と比較すると、H06は、必要なp-N3-Pheの含有割合が、H03の1/10で済み、かつ、p-N3-Pheの導入効率が、H03の3倍であった。
同様に、図6において、H07では、合成飼料中に0.05%のp-N3-Pheを含有することで、フィブロイン生産量は約80mgに減少したが、取り込まれたp-N3-Pheのシグナル強度の割合が約0.16であった。
(実験例3)
トランスジェニックカイコH06を一般品種日509×日510と交配し、その交雑種H06×(日509×日510)を得た。H06×(日509×日510)の雄幼虫および雌幼虫それぞれに対し、5齢起蚕から2日目まで通常組成の人工飼料を給餌した後、3日目以降、異なるp-N3-Phe含有割合の人工飼料を給餌した。それぞれの条件について、カイコの単位量当たりのフィブロイン生産量、及びp-N3-Pheの取り込み量を確認した。結果を図7に示す。図7において、横軸は、人工飼料中のp-N3-Pheの含有割合(dry diet換算)を示し、縦軸(左)は、棒グラフで表されたカイコの単位量当たりのフィブロイン生産量を示し、縦軸(右)は、丸プロットで表されたフィブロイン中のp-N3-Pheのピーク強度の割合を示す。
図7に示されるように、0.05%のp-N3-Pheを含む人工飼料を与えたH06×(日509×日510)におけるフィブロイン生産量は雄幼虫で約170mg、雌幼虫で約160mgと、H06におけるフィブロイン生産量の約1.6~1.7倍であった。一方で、フィブロイン中のp-N3-Pheの取り込み量は雄幼虫で約0.14、雌幼虫で約0.16と、H06と同等であり、交雑による取り込み量の減少は見られなかった。
上記の実験結果から、本発明に示すトランスジェニック系統を一般品種と交雑することによって、p-N3-Pheの取り込み量を減少させることなくフィブロイン生産量を向上させられることが示された。
トランスジェニックカイコH06を一般品種日509×日510と交配し、その交雑種H06×(日509×日510)を得た。H06×(日509×日510)の雄幼虫および雌幼虫それぞれに対し、5齢起蚕から2日目まで通常組成の人工飼料を給餌した後、3日目以降、異なるp-N3-Phe含有割合の人工飼料を給餌した。それぞれの条件について、カイコの単位量当たりのフィブロイン生産量、及びp-N3-Pheの取り込み量を確認した。結果を図7に示す。図7において、横軸は、人工飼料中のp-N3-Pheの含有割合(dry diet換算)を示し、縦軸(左)は、棒グラフで表されたカイコの単位量当たりのフィブロイン生産量を示し、縦軸(右)は、丸プロットで表されたフィブロイン中のp-N3-Pheのピーク強度の割合を示す。
図7に示されるように、0.05%のp-N3-Pheを含む人工飼料を与えたH06×(日509×日510)におけるフィブロイン生産量は雄幼虫で約170mg、雌幼虫で約160mgと、H06におけるフィブロイン生産量の約1.6~1.7倍であった。一方で、フィブロイン中のp-N3-Pheの取り込み量は雄幼虫で約0.14、雌幼虫で約0.16と、H06と同等であり、交雑による取り込み量の減少は見られなかった。
上記の実験結果から、本発明に示すトランスジェニック系統を一般品種と交雑することによって、p-N3-Pheの取り込み量を減少させることなくフィブロイン生産量を向上させられることが示された。
以上で説明した各実施形態における各構成及びそれらの組み合わせ等は一例であり、本発明の趣旨を逸脱しない範囲で、構成の付加、省略、置換、およびその他の変更が可能である。また、本発明は各実施形態によって限定されることはなく、請求項(クレーム)の範囲によってのみ限定される。
本発明のトランスジェニックカイコを用いて製造された非天然アミノ酸含有タンパク質、例えば非天然アミノ酸含有シルクタンパク質は、テキスタイル・医療分野・機能性材料等の分野に広く適用可能であり、それぞれの用途により適した改変シルクを提供できる可能性を有している。また、本発明によって新たなシルクの用途を開拓できる可能性がある。
Claims (12)
- 以下の(a)~(c)のいずれか一つのアミノ酸配列を含む配列からなり、且つ、フェニルアラニン特異的tRNAに所望の非天然アミノ酸を結合させる活性を有するタンパク質をコードするDNA、及び、該DNAを絹糸腺特異的に発現させるプロモーターを有することを特徴とするトランスジェニックカイコ。
(a)配列番号1に示されるアミノ酸配列のアミノ酸番号432位に変異を有するアミノ酸配列、
(b)前記(a)のアミノ酸番号432位以外の部位において1乃至数個のアミノ酸が欠失、挿入、置換若しくは付加されたアミノ酸配列、
(c)前記(a)のアミノ酸番号432位以外の部位において、80%以上の同一性を有するアミノ酸配列 - 前記(a)のアミノ酸番号432位が、アラニン、バリン、ロイシン、メチオニン、スレオニン、システイン、又はチロシンである請求項1に記載のトランスジェニックカイコ。
- 以下の(d)~(h)のいずれか一つのアミノ酸配列を含む配列からなり、且つ、フェニルアラニン特異的tRNAに所望の非天然アミノ酸を結合させる活性を有するタンパク質をコードするDNA、及び、該DNAを絹糸腺特異的に発現させるプロモーターを有する請求項1又は2に記載のトランスジェニックカイコ。
(d)配列番号2に示されるアミノ酸配列、
(e)配列番号3に示されるアミノ酸配列、
(f)配列番号4に示されるアミノ酸配列、及び、
(g)配列番号2~配列番号4のいずれかに示されるアミノ酸配列のアミノ酸番号432位以外の部位において1乃至数個のアミノ酸が欠失、挿入、置換若しくは付加されたアミノ酸配列、
(h)配列番号2~配列番号4のいずれかに示されるアミノ酸配列のアミノ酸番号432位以外の部位において、80%以上の同一性を有するアミノ酸配列 - さらに、前記(a)~(h)のアミノ酸番号407位に変異を有する請求項1~3のいずれか一項に記載のトランスジェニックカイコ。
- 前記アミノ酸番号407位の変異が、アラニン又はグリシンである請求項4に記載のトランスジェニックカイコ。
- 前記DNAを絹糸腺特異的に発現させるプロモーターは、前記DNAを後部絹糸腺特異的に発現させるプロモーターである請求項1~5のいずれか一項に記載のトランスジェニックカイコ。
- 前記DNAを絹糸腺特異的に発現させるプロモーターは、フィブロインL鎖由来のプロモーターである請求項1~6のいずれか一項に記載のトランスジェニックカイコ。
- 請求項1~7のいずれか一項に記載のトランスジェニックカイコと、実用系統とを交配してなることを特徴とするF1交雑種。
- 請求項1~7のいずれか一項に記載のトランスジェニックカイコ又は請求項8に記載のF1交雑種に非天然アミノ酸を投与する工程を有することを特徴とする非天然アミノ酸含有タンパク質の製造方法。
- 前記非天然アミノ酸は、フェニルアラニン類縁体である請求項9に記載の非天然アミノ酸含有タンパク質の製造方法。
- 前記Rはアジド基又はエチニル基である請求項11に記載の非天然アミノ酸含有タンパク質の製造方法。
