pH調整剤の例としては、アンモニア、ジエタノールアミン、ピリジン、N-メチル-D-グルカミン、トリス(ヒドロキシメチル)アミノメタン等の有機アミン;水酸化カリウム、水酸化ナトリウム等のアルカリ金属水酸化物;塩化カリウム、塩化ナトリウム等のアルカリ金属ハロゲン化物;硫酸、リン酸、塩酸、炭酸等の無機酸又はそのアルカリ金属塩;コリン等の4級アンモニウムカチオン、あるいはこれらの混合物(例えば、リン酸緩衝生理食塩水等の緩衝液)を挙げることができる。これらの中でも、アンモニア、ジエタノールアミン、水酸化ナトリウム、コリン、N-メチル-D-グルカミン、トリス(ヒドロキシメチル)アミノメタンが好ましく、特にアンモニア、ジエタノールアミン、水酸化ナトリウム及びコリンが好ましい。
好ましくは、本発明のゾルはさらに溶媒、pH調整剤を含む。溶媒、pH調整剤の具体例は上記のとおりである。より好ましくは、本発明のゾルは、コーティング膜形成用ゾルであって、コーティング膜形成用組成物の一態様である。
本発明のコーティング膜を形成するための基体としては、ガラス、金属含有化合物若しくは半金属含有化合物、活性炭又は樹脂を挙げることができる。金属含有化合物若しくは半金属含有化合物は、例えば基本成分が金属酸化物で、高温での熱処理によって焼き固めた焼結体であるセラミックス、シリコンのような半導体、金属酸化物若しくは半金属酸化物(シリコン酸化物、アルミナ等)、金属炭化物若しくは半金属炭化物、金属窒化物若しくは半金属窒化物(シリコン窒化物等)、金属ホウ化物若しくは半金属ホウ化物などの無機化合物の成形体など無機固体材料、アルミニウム、ニッケルチタン、ステンレス(SUS304、SUS316、SUS316L等)が挙げられる。
例えば人工透析器のフィルター用コーティング膜とする場合は、上記素材で作成された例えば直径0.1乃至500μmの中空糸形状をしたフィルターの内側に本発明に係るコーティング膜形成用組成物を通液し、その後乾燥工程、洗浄工程(熱水(例えば40℃乃至95℃)洗浄等)を経て作製することができる。
必要に応じて、滅菌のためにγ線、エチレンオキサイド、オートクレーブ等の処理がされる場合もある。
また、本発明のコーティング膜は、フラスコ、ディッシュ、プレート等の細胞培養容器や、蛋白質の付着を抑えた各種研究用器具のコーティング膜として有用である。
また、本発明のコーティング膜は、化粧品用材料、コンタクトレンズケア用品用材料、スキンケア用繊維加工剤、生化学研究用診断薬用材料、臨床診断法で広く用いられている酵素免疫測定(ELISA)法やラテックス凝集法における非特異的吸着を抑制するためのブロッキング剤、酵素や抗体などの蛋白質を安定化するための安定化剤としても有用である。
さらに本発明のコーティング膜は、トイレタリー、パーソナルケア用品、洗剤、医薬品、医薬部外品、繊維、防汚材向けのコーティング膜としても有用である。

式(B)で表される化合物の、上記共重合体を形成するモノマー全体に対する割合は、3モル%乃至80モル%であり、好ましくは5モル%乃至70モル%であり、さらに好ましくは8モル%乃至65モル%である。また、式(B)で表される化合物は、2種以上であってもよい。
式(C)で表される化合物の、上記共重合体を形成するモノマー全体に対する割合は、上記式(A)及び(B)の割合を差し引いた残部全てでも良いし、上記式(A)及び(B)と上記第4成分との合計割合を差し引いた残部であってもよいが、例えば1モル%乃至90モル%であり、好ましくは3モル%乃至88モル%であり、さらに好ましくは5モル%乃至87モル%であり、最も好ましくは50モル%乃至86モル%である。また、式(C)で表される化合物は、2種以上であってもよい。

本発明に係るコーティング膜形成用組成物は、所望の共重合体を、所望の溶媒にて所定の濃度に希釈することにより調製してもよい。
さらに本発明に係るコーティング膜形成用組成物は、本発明の共重合体含有ワニスから調製してもよい。一の実施態様では、本発明の共重合体含有ワニスは、上記式(A)及び(B)で表される化合物を、溶媒中で、両化合物の合計濃度0.01質量%乃至20質量%にて反応(重合)させる工程を含む製造方法により調製することができる。
水への溶解性、イオンバランス及びモノマーとの相互作用を考慮した場合、2,2’-アゾ(2-メチル-N-(2-ヒドロキシエチル)プロピオンアミド、2,2’-アゾビス(N-(2-カルボキシエチル)-2-メチルプロピオンアミジン)n-水和物、4,4’-アゾビス(4-シアノペンタノイックアシッド)、2,2’-アゾビス[2-(2-イミダゾリン-2-イル)プロパン]ジヒドロクロリド、2,2’-アゾビス[2-(2-イミダゾリン-2-イル)プロパン]ジスルファートジヒドレート、2,2’-アゾビス[2-(2-イミダゾリン-2-イル)プロパン]、2,2’-アゾビス(2-アミジノプロパン)ジヒドロクロリド及びペルオキソ二硫酸から選ばれることが好ましい。

アシッドホスホオキシエチルメタアクリレート(製品名;ホスマーM、ユニケミカル(株)製、乾固法100℃・1時間における不揮発分:91.8%、アシッドホスホオキシエチルメタクリレート(44.2質量%)、リン酸ビス[2-(メタクリロイルオキシ)エチル](28.6質量%)、その他の物質(27.2質量%)の混合物)1.00gにエタノール39.76g、メタクリル酸2-(ジメチルアミノ)エチル(東京化成工業(株)製)0.76g、メタクリル酸ブチル(東京化成工業(株)社製)3.22g、2,2’-アゾビス(2,4-ジメチルバレロニトリル)(製品名;V-65、和光純薬工業(株)社製)0.25gを、20℃以下に保ちながら順に加えた。十分に攪拌して均一となった上記全てのものが入った混合液を滴下ロートに導入した。一方で、別途純水29.82g、エタノール29.82gを冷却管付きの3つ口フラスコに入れ、これを窒素フローし、撹拌しながらリフラックス温度まで昇温した。この状態を維持しつつ、上記混合液を導入した滴下ロートを3つ口フラスコにセットし、0.5時間かけて混合液を純水とエタノールの沸騰液内に滴下した。滴下後、24時間上記環境を維持した。24時間後に冷却することで固形分約4.50質量%の共重合体含有ワニス105.00gを得た。得られた透明液体のGPCにおける重量平均分子量は約9,200であった。
アシッドホスホオキシエチルメタアクリレート(製品名;ホスマーM、ユニケミカル(株)社製、乾固法100℃・1時間における不揮発分:91.8%、アシッドホスホオキシエチルメタクリレート(44.2質量%)、リン酸ビス[2-(メタクリロイルオキシ)エチル](28.6質量%)、その他の物質(27.2質量%)の混合物)1.00gを20℃以下に冷却しながらコリン(48-50%水溶液:東京化成工業(株)社製)1.18gと、純水8.49gを加えて均一になるまで撹拌した。この混合溶液にメタクロイルコリンクロリド80%水溶液(東京化成工業(株)社製)1.26g、メタクリル酸ブチル(東京化成工業(株)社製)3.22g、2,2’-アゾビス(2,4-ジメチルバレロニトリル)(製品名;V-65、和光純薬工業(株)社製)0.03g、エタノール37.33gを20℃以下に保ちながら順に加えた。十分に攪拌して均一となった上記全てのものが入った混合液を、滴下ロートに導入した。一方で、別途純水14.00gとエタノール55.99gを冷却管付きの3つ口フラスコに加えて窒素フローし、撹拌しながらリフラックス温度まで昇温した。この状態を維持しつつ、上記混合液を導入した滴下ロートを3つ口フラスコにセットし、0.5時間かけて混合液を純水とエタノールの沸騰液内に滴下した。滴下後、24時間上記環境を維持した状態で加熱撹拌する。24時間後に冷却することで固形分約5.00質量%の共重合体含有ワニス124.00gを得た。得られたコロイド状液体のGFCにおける重量平均分子量は約42,000であった。

アシッドホスホオキシエチルメタアクリレート(製品名;ホスマーM、ユニケミカル(株)社製、乾固法100℃・1時間における不揮発分:91.8%、アシッドホスホオキシエチルメタクリレート(44.2質量%)、リン酸ビス[2-(メタクリロイルオキシ)エチル](28.6質量%)、その他の物質(27.2質量%)の混合物)4.75gを35℃以下に冷却しながらコリン(48-50%水溶液:東京化成工業(株)社製)5.69gを加えて均一になるまで撹拌した。この混合液にメタクロイルコリンクロリド80%水溶液(東京化成工業(株)社製)5.97g、メタクリル酸ブチル(東京化成工業(株)社製)6.54g、2,2’-アゾビス(2,4-ジメチルバレロニトリル)(製品名;V-65、和光純薬工業(株)社製)0.08g、エタノール139.95gを35℃以下に保ちながら順に加えた。十分に攪拌して均一となった上記全てのものが入った混合液を、滴下ロートに導入した。一方で、別途純水103.94gとエタノール112.10gを冷却管付きの3つ口フラスコに加えて窒素フローし、撹拌しながらリフラックス温度まで昇温した。この状態を維持しつつ、上記混合液を導入した滴下ロートを3つ口フラスコにセットし、1時間かけて混合液を純水とエタノールの沸騰液内に滴下した。滴下後、24時間上記環境を維持した状態で加熱撹拌する。24時間後に冷却することで固形分約4.16質量%の共重合体含有ワニス379.03gを得た。得られたコロイド状液体のGFCにおける重量平均分子量は約8,600であった。
アシッドホスホオキシエチルメタアクリレート(製品名;ホスマーM、ユニケミカル(株)社製、乾固法100℃・1時間における不揮発分:91.8%、アシッドホスホオキシエチルメタクリレート(44.2質量%)、リン酸ビス[2-(メタクリロイルオキシ)エチル](28.6質量%)、その他の物質(27.2質量%)の混合物)25.00gを35℃以下に冷却しながらコリン(48-50%水溶液:東京化成工業(株)社製)29.95gを加えて均一になるまで撹拌した。この混合液にメタクロイルコリンクロリド80%水溶液(東京化成工業(株)社製)20.95g、メタクリル酸ブチル(東京化成工業(株)社製)28.67g、2,2’-アゾビス(2,4-ジメチルバレロニトリル)(製品名;V-65、和光純薬工業(株)社製)0.70g、エタノール110.84gを35℃以下に保ちながら順に加えた。さらに、2,2’-アゾビス(N-(2-カルボキシエチル)-2-メチルプロピオンアミジン)n-水和物(和光純薬社製品名;VA-057、和光純薬工業(株)社製)0.70gを純粋27.71gに溶解させた水溶液を35℃以下に保ちながら上記の溶液に加え、十分に攪拌して均一となった上記全てのものが入った混合液を、滴下ロートに導入した。一方で、別途純水56.81gとエタノール131.62gを冷却管付きの3つ口フラスコに加えて窒素フローし、撹拌しながらリフラックス温度まで昇温した。この状態を維持しつつ、上記混合液を導入した滴下ロートを3つ口フラスコにセットし、1時間かけて混合液を純水とエタノールの沸騰液内に滴下した。滴下後、24時間上記環境を維持した状態で加熱撹拌する。24時間後に冷却することで固形分約19.86質量%の共重合体含有ワニス432.97gを得た。得られたコロイド状液体のGFCにおける重量平均分子量は約8,500であった。

アシッドホスホオキシエチルメタアクリレート(製品名;ホスマーM、ユニケミカル(株)製、乾固法100℃・1時間における不揮発分:91.8%、アシッドホスホオキシエチルメタクリレート(44.2質量%)、リン酸ビス[2-(メタクリロイルオキシ)エチル](28.6質量%)、その他の物質(27.2質量%)の混合物)10.00gに純水68.88gを加え十分に溶解した。次いで、エタノール29.52g、メタクリル酸2-(ジメチルアミノ)エチル7.63g(東京化成工業(株)製)、2,2’-アゾビス(N-(2-カルボキシエチル)-2-メチルプロピオンアミジン)n-水和物(和光純薬社製品名;VA-057、和光純薬工業(株)製)0.09gを、20℃以下に保ちながら、ホスマーMの水溶液に順に加えた。十分に攪拌して均一となった上記全てのものが入った混合液を、滴下ロートに導入した。一方で、別途純水373.89g、エタノール29.52gを冷却管付きの3つ口フラスコに入れ、これを窒素フローし、撹拌しながらリフラックス温度まで昇温した。この状態を維持しつつ、上記混合液を導入した滴下ロートを3つ口フラスコにセットし、0.5時間かけて混合液を純水とエタノールの沸騰液内に滴下した。滴下後、24時間上記環境を維持した状態で加熱撹拌することで固形分約3.5質量%の共重合体含有ワニス509.60gを得た。得られた透明液体のGFCにおける重量平均分子量は約280,000であった。
半導体評価用の市販のシリコンウエハをそのまま用いた。
バーコート法により作成された、市販のポリエーテルスルホン(PES)のフィルム(約0.1mm)を約1cm角にカットしたものをPESフィルムとした。
Au蒸着された水晶振動子(Q-Sense,QSX304)を、UV/オゾン洗浄装置(UV253E、フィルジェン株式会社製)を用いて10分間洗浄し、直後に1-デカンチオンール(東京化成工業(株)製)0.1012gをエタノール100mlに溶解した溶液中に24時間浸漬した。エタノールでセンサー表面を洗浄後自然乾燥し、ポリ(オキシ-1,4-フェニレンスルホニル-1,4-フェニレン)(Aldrich社製)1.00gを1,1,2,2-テトラクロロエタン99.00gに溶解したワニスをスピンコーターにて3500rpm/30secで膜センサー側にスピンコートし、205℃/1min乾燥することでQCMセンサー(PES)とした。
Au蒸着された水晶振動子(Q-Sense,QSX304)を、UV/オゾン洗浄装置(UV253E、フィルジェン株式会社製)を用いて10分間洗浄し、直後に2-アミノエタンチオール(東京化成工業(株)製)0.0772gをエタノール1000mlに溶解した溶液中に24時間浸漬した。エタノールでセンサー表面を洗浄後自然乾燥し、ポリスチレン(Aldrich社製)1.00gをトルエン99.00gに溶解したワニスをスピンコーターにて3500rpm/30secで膜センサー側にスピンコートし、120℃/1min乾燥することでQCMセンサー(PS)とした。
上記合成実施例1で得られた共重合体含有ワニス1.00gに、純水10.78g、エタノール4.89gを加えて十分に攪拌し、コーティング膜形成組成物を調製した。pHは5.2であった。得られたコーティング膜形成組成物中に、上記シリコンウェハ又はPESフィルムをディップし、オーブンにて45℃、12時間乾燥させた。その後、コーティング膜上に付着している未硬化の膜形成用組成物をPBSと純水で十分に洗浄を行って、コーティング膜が形成されたシリコンウェハ、又はPESフィルムを得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ30Åであった。
また、上記コーティング膜形成用組成物を3500rpm/30secでQCMセンサー(PES)にスピンコートし、乾燥工程として45℃のオーブンで12時間ベークした。その後、洗浄工程として過剰についた未硬化のコーティング膜形成用組成物をPBSと超純水にて各2回ずつ洗浄し、表面処理済QCMセンサー(PES)を得た。
上記合成実施例2で得られた共重合体含有ワニス1.00gに、純水10.78g、エタノール4.89gを加えて十分に攪拌し、コーティング膜形成組成物を調製した。pHは5.3であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ、PESフィルム又は表面処理済QCMセンサー(PES)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ81Åであった。
上記合成実施例3で得られた共重合体含有ワニス6.50gに、エタノール203.88g、アンモニア水(28%水溶液、関東化学(株)製)0.17gを加えて十分に攪拌し、コーティング膜形成組成物を調製した。pHは10.3であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ、PESフィルム又は表面処理済QCMセンサー(PES)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ120Åであった。
上記合成実施例4で得られた共重合体含有ワニス6.50gに、エタノール192.83g、アンモニア水(28%水溶液、関東化学(株)製)0.22gを加えて十分に攪拌し、コーティング膜形成組成物を調製した。pHは11.2であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ、PESフィルム又は表面処理済QCMセンサー(PES)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ82Åであった。
上記合成実施例5で得られた共重合体含有ワニス6.50gに、エタノール203.02、アンモニア水(28%水溶液、関東化学(株)製)0.27gを加えて十分に攪拌し、コーティング膜形成組成物を調製した。pHは11.2であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ、PESフィルム又は表面処理済QCMセンサー(PES)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ119Åであった。
上記合成実施例6で得られた共重合体含有ワニス9.00gに、コリン(48-50%水溶液:東京化成工業(株)社製)0.10gとエタノール141.00gを加えて十分に攪拌し、コーティング膜形成組成物を調製した。pHは12.8であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PES、PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ109Åであった。
上記合成実施例7で得られた共重合体含有ワニス9.00gに、コリン(48-50%水溶液:東京化成工業(株)社製)0.13gとエタノール141.00gを加えて十分に撹拌し、コーティング膜形成組成物を調製した。pHは13.2であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PES、PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ57Åであった。
上記合成実施例8で得られた共重合体含有ワニス9.00gに、コリン(48-50%水溶液:東京化成工業(株)社製)0.18gとエタノール141.00gを加えて十分に撹拌し、コーティング膜形成組成物を調製した。pHは12.6であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PES、PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ44Åであった。
上記合成実施例9で得られた共重合体含有ワニス18.00gに、1mol/L塩酸(1N)(関東化学(株)社製)0.57gと純水17.06g、エタノール39.82gを加えて十分に撹拌し、コーティング膜形成組成物を調製した。pHは3.6であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ51Åであった。
上記合成実施例10で得られた共重合体含有ワニス8.00gに、1mol/L塩酸(1N)(関東化学(株)社製)1.03gと純水45.26g、エタノール105.62gを加えて十分に撹拌し、コーティング膜形成組成物を調製した。pHは3.5であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ54Åであった。
上記合成実施例10で得られた共重合体含有ワニス10.00gに、1mol/L塩酸(1N)(関東化学(株)社製)1.19gと純水26.78g、エタノール62.54gを加えて十分に撹拌し、コーティング膜形成組成物を調製した。pHは3.6であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ112Åであった。
上記合成実施例10で得られた共重合体含有ワニス50.00gに、1mol/L塩酸(1N)(関東化学(株)社製)1.74gと純水45.00g、エタノール105.00gを加えて十分に撹拌し、コーティング膜形成組成物を調製した。pHは3.7であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ165Åであった。
上記合成実施例10で得られた共重合体含有ワニス5.00gに、クエン酸(関東化学(株)社製)1.29gと純水28.53g、エタノール66.55gを加えて十分に撹拌し、コーティング膜形成組成物を調製した。pHは3.5であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ359Åであった。
上記合成実施例10で得られた共重合体含有ワニス5.00gに、酢酸(関東化学(株)社製)1.85gと純水28.53g、エタノール66.55gを加えて十分に撹拌し、コーティング膜形成組成物を調製した。pHは3.5であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ291Åであった。
上記合成実施例10で得られた共重合体含有ワニス5.00gに、リンゴ酸(関東化学(株)社製)1.55gと純水28.53g、エタノール66.55gを加えて十分に撹拌し、コーティング膜形成組成物を調製した。pHは3.5であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ342Åであった。
上記合成実施例10で得られた共重合体含有ワニス4.00gに、1mol/L塩酸(1N)(関東化学(株)社製)0.57gとクエン酸(関東化学(株)社製)0.40gと純水22.47g、エタノール53.74gを加えて十分に撹拌し、コーティング膜形成組成物を調製した。pHは3.5であった。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ又は表面処理済QCMセンサー(PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ300Åであった。
上記PESフィルムをそのまま用いた。
上記QCMセンサー(PES)をそのまま用いた。
上記比較合成例1で得られた共重合体含有ワニス1.00gに、純水7.27g、エタノール3.39gを加えて十分に攪拌し、コーティング膜形成用組成物を調製した。実施例1と同様の方法にて、コーティング膜が形成されたシリコンウェハ、PESフィルム又は表面処理済QCMセンサー(PES、PS)を得た。上記シリコンウェハを用いて光学式干渉膜厚計でコーティング膜の膜厚を確認したところ44Åであった。
上記QCMセンサー(PS)をそのまま用いた。
(血小板溶液の調製)
3.8質量%クエン酸ナトリウム溶液0.5mLに対して、健康なボランティアより採血した血液4.5mLを混和した後、遠心分離にて[冷却遠心機5900((株)久保田製作所製)、1000rpm/10分、室温]上層の多血小板血漿(PRP)を回収した。引き続き、下層について遠心分離を行い(上記遠心機、3500rpm/10分、室温)、上層の乏血小板血漿(PPP)を回収した。多項目自動赤血球分析装置(XT-2000i、シスメックス(株)製)にてPRPの血小板数を計測後、PPPを用いてPRPの血小板濃度が30×104cells/μLになるように調製した。
各実施例、比較例のPESフィルムを24穴平底マイクロプレート(コーニング社製)に配置した。これらの基板を配置したプレートのウェル内に、上記血小板濃度に調製したPRP溶液300μLを添加した。5%二酸化炭素濃度を保った状態で、37℃で24時間、CO2インキュベーター内にて静置した。所定の静置時間が経過した後、プレート内のPRPを除き、PBS3mLにて5回洗浄した。その後、2.5体積%グルタルアルデヒドのPBS溶液2mLを添加し、4℃で一昼夜静置後、グルタルアルデヒドのPBS溶液を除き、超純水(Milli-Q水)3mLで5回洗浄した。さらに、70%エタノール水(v/v)1mLで3回洗浄し、風乾した。
上記血小板付着実験を行った各実施例、比較例のPESフィルムに、イオンスパッター(E-1030、(株)日立ハイテクノロジーズ製)にてPt-Pdを1分間蒸着した。その後、電子顕微鏡(S-4800、(株)日立ハイテクノロジーズ製)にて血小板の付着を1,000倍で観察した。電子顕微鏡にてPESフィルムの中心部から半径2mm以内5箇所(1箇所あたりの面積:縦95μm×横126.5μm=11,385[μm2])の血小板付着数を計測した。各箇所の計測値を平均することで血小板付着数とした。その結果を下記表1に示す。
各実施例及び比較例によって得た表面処理済QCMセンサー(PES、PS)を散逸型水晶振動子マイクロバランスQCM-D(E4、Q-Sense社製)に取り付け、周波数の変化が1時間で1Hz以下となる安定したベースラインを確立するまでPBSを流した。次に、安定したベースラインの周波数を0Hzとして約10分間PBSを流した。引き続き、フィブリノゲンをPBSで100μg/mlに希釈した溶液、又は41010 - Basal Medium Eagle (BME), no Glutamine(サーモフィッシャーサイエンティフィック株式会社製)に15wt%のウシ血清(FBS)、L-Glutamine、抗生物質としてペニシリンとストレプトマイシンを添加した溶液を約30分流し、その後再びPBSを約20分流した後の11次オーバートーンの吸着誘起周波数のシフト(Δf)を読み取った。分析のためにQ-Tools(Q-Sense社製)を使用して、吸着誘起周波数のシフト(Δf)を、Sauerbrey式で説明される吸着誘起周波数のシフト(Δf)を単位面積当たりの質量(ng/cm2)と換算したものを生体物質の付着量として表2に示す。比較例と比較し、実施例は1桁又は2桁低い各種タンパク質吸着量を示した。なお、フィブリノゲンはPESセンサー、FBS由来の生体物質はPSセンサーをそれぞれ被吸着物質とした。
実施例又は比較例にて調製したコーティング膜形成用組成物を用い、下記のコーティング法により平底96穴細胞培養プレート(BDバイオサイエンス社製、#351172)のウェルにコーティング膜を形成した。
コーティング方法は、上記コーティング膜形成組成物を1ウェルあたり200μL添加し、60分間放置後に過剰の液を除去して50℃で一晩乾燥させた。その後、滅菌水を1ウェルあたり200μL添加後、除去して洗浄を行った。同様に、さらに2回洗浄を行った。
陽性対照のサンプルとしては、市販の細胞低接着プレート(コーニング社製、#3474)を用いた。
細胞は、マウス胚線維芽細胞C3H10T1/2(DSファーマバイオメディカル社製)を用いた。細胞の培養に用いた培地は、10%FBS(HyClone社製)とL-グルタミン-ペニシリン-ストレプトマイシン安定化溶液(SIGMA-ALDRICH社製)を含むBME培地(サーモフィッシャーサイエンティフィック社製)を用いた。細胞は、37℃/CO2インキュベーター内にて5%二酸化炭素濃度を保った状態で、直径10cmのシャーレ(培地10mL)を用いて2日間以上静置培養した。引き続き、本細胞をPBS5mlで洗浄した後、トリプシン-EDTA溶液(インビトロジェン社製)1mLを添加して細胞を剥がし、上記の培地10mLにてそれぞれ懸濁した。本懸濁液を遠心分離(株式会社トミー精工製、型番LC-200、1000rpm/3分、室温)後、上清を除き、上記の培地を添加して細胞懸濁液を調製した。
上記にて調製したプレートに対して、それぞれの細胞懸濁液を2×104cells/wellとなるように各100μL加えた。その後、5%二酸化炭素濃度を保った状態で、37℃で4日間CO2インキュベーター内にて静置した。
培養4日間後、コーティングした平底96穴細胞培養プレートに対する細胞の付着を倒立型顕微鏡(オリンパス社製、CKX31)による観察に基づき比較した。また、Cell Counting Kit-8溶液(同仁化学研究所製)を1ウェル当たり10μL添加し、37℃で2時間CO2インキュベーター内にて静置した。その後、吸光度計(MolecularDevices社製、SpectraMax)で450nmの吸光度を測定した。その結果を下記表3に示す。
各実施例の各コーティング膜形成用組成物中のゾル粒子径の測定は、動的光散乱光度計(DLS、大塚電子社製、製品名:DLS-8000DLTKY)を用いて測定した。結果を表4に示す。
Claims (23)
- 下記式(A)、(B)及び(C):

Ta、Tb、Tc、Ua1、Ua2、Ub1、Ub2及びUb3は、それぞれ独立して、水素原子又は炭素原子数1乃至5の直鎖若しくは分岐アルキル基を表し;
Qa及びQbは、それぞれ独立して、単結合、エステル結合又はアミド結合を表し、Qcは、単結合、エーテル結合又はエステル結合を表し;
Ra及びRbは、それぞれ独立して、ハロゲン原子で置換されていてもよい炭素原子数1乃至10の直鎖若しくは分岐アルキレン基を表し、Rcは、炭素原子数1乃至18の直鎖若しくは分岐アルキル基、炭素原子数3乃至10の環式炭化水素基、炭素原子数6乃至10のアリール基、炭素原子数7乃至14のアラルキル基又は炭素原子数7乃至14のアリールオキシアルキル基(ここで、前記アリール部分は、ハロゲン原子で置換されていてもよい炭素原子数1乃至5の直鎖若しくは分岐アルキル基で置換されていてもよい)を表し;
An-は、ハロゲン化物イオン、無機酸イオン、水酸化物イオン及びイソチオシアネートイオンからなる群から選ばれる陰イオンを表し;

- 乾燥工程後に得られた膜を、さらに水及び電解質を含む水溶液からなる群より選ばれる少なくとも1種の溶媒で洗浄する工程を含む、請求項5乃至7の何れか1項に記載のコーティング膜。
- 生体物質の付着抑制能を有する、請求項5乃至8の何れか1項に記載のコーティング膜。
- 下記式(a)で表される有機基を含む繰り返し単位と、下記式(b)で表される有機基を含む繰り返し単位と、下記式(c)で表される有機基を含む繰り返し単位とを含む共重合体:
