Abstract
Description
細胞や組織などの培養の過程においては、複数の異なる培養環境や培養条件が必要になる場合がある。また、複数の異なる培養環境や培養条件で培養した細胞を、後の工程で混合する場合もある。これらの場合には、培養された細胞を複数の容器間で移し替る必要があった。
ここで、細胞の移し替えや培養液の追加の際におけるコンタミネーションを防止可能な技術として、複数の培養容器を接続することによって密閉系の細胞培養キットを構成し、これを用いて細胞培養を行うことが提案されている(特許文献1,2参照)。
すなわち、このような細胞培養キットを用いた場合、接続する培養容器の数が多くなると、チューブが絡まったり、あるいは培養容器が入り乱れてどれがどれかがわからなくなったりするなど、やはりその取り扱いが煩雑になるという問題があった。
また、最近では、自動細胞培養装置を用いた細胞培養方法も提案されているが、このような複数の容器からなる細胞培養キットの自動細胞培養装置への取付け作業は、非常に煩雑になってしまうという問題もあった。
現在は、技術的には、ガス透過化性を有するフィルム材からなる培養バッグの内面に抗CD3抗体を固相化して活性化用の培養バッグを作製し、これを増幅用の培養バッグに接続して細胞培養キットを構成し、自動細胞培養装置を用いてリンパ球の培養を行うことも可能になっている。
しかしながら、このような活性化用の培養バッグは、フラスコに比べてコストが相当大きくなるため、フラスコから活性化用の培養バッグへの置き換えは、簡単に行うことができなかった。
すなわち、本発明は、複数の異なる培養環境や培養条件で培養した細胞を、簡便な作業で移し変え可能な複室培養容器、及びこれを用いた細胞培養方法の提供を目的とする。
血液から分離した白血球及び培養液を前記接着系細胞培養室に注入して静置し、前記白血球における接着系細胞を前記接着系細胞培養室内の底面に接着させ、次いで、前記接着系細胞培養室から前記活性化培養室に、前記白血球におけるリンパ球と培養液を移送して、前記活性化培養室においてリンパ球を活性化させ、前記接着系細胞培養室に樹状細胞への誘導因子を含む培養液を注入して前記接着系細胞を培養し、前記接着系細胞を樹状細胞に誘導し、前記活性化培養室から前記増幅培養室に活性化されたリンパ球及び培養液を移送して、前記増幅培養室においてリンパ球を増幅させる方法としてある。
[第一実施形態]
まず、本発明の第一実施形態に係る複室培養容器及び細胞培養方法について、図1を参照して説明する。
本実施形態の複室培養容器は、ガス透過性を有する材料を加工してなる複室培養容器であって、細胞を培養するための培養室が二つ以上形成され、各培養室は、少なくとも一つの他の培養室と連通し、かつ複室培養容器の外部と連通するための外部接続部を少なくとも一つ備えたことを特徴とする。
培養室11には、複室培養容器1の外部と連通するための外部接続ポート111と、他の培養室と連通するための室間接続ポート112と、サンプリングを行うためのサンプリングポート113が備えられている。
外部接続ポート111は、図示しないが、チューブ2を介して培養液供給容器等に接続され、培養室11への培養液の供給のためなどに用いられる。
室間接続ポート112は、培養室12の室間接続ポート122にチューブ2を介して接続され、培養室12の内容液を培養室11に移送するために用いられる。
サンプリングポート113は、培養室11で培養された細胞のサンプリングを行うために用いられる。
外部接続ポート121は、図示しないが、チューブ2を介して細胞供給容器等に接続され、培養室12への細胞の注入のためなどに用いられる。
室間接続ポート122は、培養室11の室間接続ポート112にチューブ2を介して接続され、培養室12の内容液を培養室11に移送するために用いられる。
このようなガス透過性材料をヒートシール加工や圧空成形、真空成形などのシート成形することによって、本実施形態の複室培養容器1を製造することができる。
本実施形態の複室培養容器1による培養用途は特に限定されないが、例えば、リンパ球の培養、DC-LAK療法(樹状細胞、リンパ球の培養)、ES細胞の培養、iPS細胞の培養などに好適に用いることが可能である。
リンパ球の培養を行う場合、培養室11は、その内面に特別な処理を行うことなく、リンパ球を増幅させるための増幅培養室として用い、培養室12は、その内面に抗CD3抗体をコーティング(固相化)して、リンパ球を活性化させるための活性化培養室として用いることができる。
そして、図1に示すように、培養室11と培養室12を接続するチューブ2にクリップなどの流路開閉手段を取付けて、チューブ2における流路の開閉を制御する。なお、流路開閉手段としては、例えばピンチバルブや二方活栓などを用いることもできる。
その後、チューブ2における流路を開放して、培養室12から培養室11に活性化されたリンパ球及び培養液を移送し、培養室11においてリンパ球を増幅させる。
このとき、リンパ球の増幅に併せて、培養室11に培養液を追加し、約4日から10日間程度で活性化されたTリンパ球を大量に得ることができる。
なお、培養室12の内面にその他の生理活性物質をコーティングして、その他の細胞を活性化させた後、これを培養室11に移送して、増幅させることもできる。
ES細胞の培養を行う場合、培養室11は、その内面にマトリゲルコート処理を行って、ES細胞を増幅させるためのES細胞培養室として用い、培養室12は、マイトマイシンC処理を施したMEF(マウス胎児線維芽細胞)を充填し、MEFを培養するためのMEF培養室として用いることができる。
そして、予め準備したヒトES細胞を培養室11に播種して、MEF-conditioned培養液(bFGFを添加しておく)により37℃、2%二酸化炭素濃度の条件下で培養して、ヒトES細胞を増幅させることができる。
次に、本発明の第二実施形態に係る複室培養容器及び細胞培養方法について、図2を参照して説明する。
本実施形態の複室培養容器1aは、同図に示すように、培養室11と培養室12に加えて、さらに培養室13を備えている。
培養室13には、複室培養容器1aの外部と連通するための外部接続ポート131と、他の培養室と連通するための室間接続ポート132が備えられている。
外部接続ポート131は、図示しないが、チューブ2を介して細胞供給容器等に接続され、培養室13への細胞の注入のためなどに用いられる。室間接続ポート132は、培養室11の室間接続ポート112及び培養室12の室間接続ポート122にチューブ2を介して接続され、培養室13の内容液を培養室12等に移送するために用いることができる。培養室12における外部接続ポート121は、図示しないが、チューブ2を介して培養液供給容器等に接続され、培養室12に培養液を供給するためなどに用いることができる。
その他の点については、第一実施形態の複室培養容器1と同様の構成にすることができる。
培養室11は、その内面に特別な処理を行うことなく、リンパ球を増幅させるための増幅培養室として用い、培養室12は、その内面に抗CD3抗体をコーティングして、リンパ球を活性化させるための活性化培養室として用いる。また、培養室13は、その内面に接着系細胞を培養するための表面処理を行って、接着系細胞培養室として用いる。この接着系細胞培養室は、白血球をリンパ球と接着系細胞に分離して接着系細胞を培養するために用いられる。そして、図2に示すように、培養室11と培養室12を接続するチューブ2上と、培養室12と培養室13を接続するチューブ2上にクリップなどの流路開閉手段を取付けて、チューブ2における流路の開閉を制御する。
次に、培養室12と培養室13を接続するチューブ2における流路を開放して、培養室13から培養室12に、白血球における浮遊系細胞(リンパ球)と培養液を移送する。そして、培養室12において、リンパ球を活性化させる。
このようにして接着系細胞から誘導された未熟樹状細胞に、癌抗原を提示して約2日間培養し、成熟した樹状細胞を得ることができる。
その後、培養室11と培養室12を接続するチューブ2における流路を開放して、培養室12から培養室11に活性化されたリンパ球及び培養液を移送し、培養室11においてリンパ球を増幅させる。このとき、リンパ球の増幅に併せて、培養室11に培養液を追加し、約4日間程度で活性化されたTリンパ球(T細胞)を大量に得ることができる。
最終的には、これらの細胞を調製して、樹状細胞とリンパ球をそれぞれヒトに投与することができる。
次に、本発明の第三実施形態に係る複室培養容器及び細胞培養方法について、図3,及び図4を参照して説明する。
本実施形態の複室培養容器は、その一つの辺に、チューブを用いて接続された二つ以上の室間接続部が備えられ、各培養室における室間接続部が備えられた辺と、複室培養容器における当該一つの辺との距離が、各培養室の並び順に大きく又は小さくなるように、各培養室が形成されたことを特徴とする。その他の点については、第一実施形態の複室培養容器1と同様の構成にすることができる。
これによって、複室培養容器1bを傾けるだけで、培養室12から培養室11への内容物の移送を、好適に行うことが可能となる。
これに対して、本実施形態の複室培養容器1bによれば、培養室12における内容物の残存を大きく低減することが可能になっている。
次に、本発明の第四実施形態に係る複室培養容器及び細胞培養方法について、図5を参照して説明する。
本実施形態の複室培養容器は、各培養室を他の培養室と連通するための流路が、複室培養容器内において、フィルム材をヒートシール加工して形成されたことを特徴とする。
移送流路3は、物理的手段により開閉を制御できる。この物理的手段としては、例えば押圧部材などを用いることができ、その押込量を制御することで、移送流路3を開閉することができる。
次に、本発明の第五実施形態に係る複室培養容器及び細胞培養方法について、図6を参照して説明する。
本実施形態の複室培養容器1dは、図6に示すように、細胞を活性化させるための活性化培養室である培養室12と、細胞を増幅させるための増幅培養室である培養室11と、細胞をサンプリングするためのサンプリング室14と、細胞を回収するための回収室15とを備えている。
さらに、サンプリング室14と回収室15の周囲には、これらをそれぞれ容易に切り離し可能にするための、ミシン目、スリット、スコア、切欠等からなる易切断部4が設けられている。また、このような易切断部4を、各培養室を切り離すために設けても良い。
第一実施形態と同様に、培養室11を、その内面に特別な処理を行うことなく、リンパ球を増幅させるための増幅培養室として用い、培養室12を、その内面に抗CD3抗体を固相化して、リンパ球を活性化させるための活性化培養室として用いる。
また、サンプリング室14を、培養室11における培養細胞をサンプリングするために用い、回収室15を、培養室11における培養細胞の一定量を回収するために用いる。
リンパ球を活性化した後、培養室12と培養室11間の移送流路3を開放して、培養室12からリンパ球と培養液を培養室11に押し出して、活性化したリンパ球を培養室11に移送させ、培養室11においてリンパ球を増幅させる。このとき、リンパ球の増幅に併せて、外部接続ポート111を介して、培養液供給容器から培養室11に必要な培養液を追加することができる。この培養液の追加は、無菌接合装置などを用いることで無菌的に行うことも可能である。また、培養液の成分を改良し、培養室11に追加培養液を予め充填しておくようにすることもできる。さらに、本実施形態の複室培養容器1dに培養液供給室を形成して、培養液供給室から培養室11に培養液を追加可能にすることもできる。
また、本実施形態の複室培養容器1dは、流路が一体化された複室培養容器であるため、シールバーの設計を行うのみで、種々の室や流路を簡便に低コストで作製することが可能である。また、自動培養装置にセットする際にも、複雑なチューブの取り付けが不要になることから、作業効率が高まるだけでなく、取り付けミス等のリスクも低減することが可能になっている。
例えば、上記各実施形態を組み合わせたものにしたり、接着系細胞の移送のために、接着系細胞を培養室の内面から剥離するための酵素溶液を培養室に供給するための酵素溶液供給容器、及びその酵素の働きを阻害するための阻害剤溶液を培養室に供給するための阻害剤溶液供給容器を備えたものにしたりするなど、適宜変更することが可能である。

Claims (11)
- ガス透過性を有する材料を加工してなる複室培養容器であって、細胞を培養するための培養室が二つ以上形成され、各培養室は、少なくとも一つの他の培養室と連通し、かつ前記複室培養容器の外部と連通するための外部接続部を少なくとも一つ備えたことを特徴とする複室培養容器。
- 前記各培養室に、前記他の培養室と連通するための室間接続部がそれぞれ備えられ、前記室間接続部同士がチューブを用いて接続されたことを特徴とする請求項1記載の複室培養容器。
- 前記各培養室を前記他の培養室と連通するための流路が、前記複室培養容器内において、ガス透過性を有するフィルム材をヒートシール加工して形成されたことを特徴とする請求項1記載の複室培養容器。
- 前記培養室として、少なくとも、細胞を活性化させるための活性化培養室と、細胞を増幅させるための増幅培養室とを備えたことを特徴とする請求項1~3のいずれかに記載の複室培養容器。
- 前記培養室として、細胞を活性化させるための活性化培養室と、細胞を増幅させるための増幅培養室とを備えると共に、細胞をサンプリングするためのサンプリング室と、細胞を回収するための回収室とを備え、前記活性化培養室と前記増幅培養室を連通する前記流路が形成され、前記増幅培養室と前記サンプリング室間、及び前記増幅培養室と前記回収室間のそれぞれに、流路が、前記複室培養容器内において、ガス透過性を有するフィルム材をヒートシール加工して形成されたことを特徴とする請求項3記載の複室培養容器。
- 前記各培養室、前記サンプリング室、及び前記回収室からなる群から選択された少なくともいずれかの室を切り離すための易切断部を設けたことを特徴とする請求項5記載の複室培養容器。
- 全部又は一部の前記培養室に、それぞれ異なる培養基材を備えたことを特徴とする請求項1~6のいずれかに記載の複室培養容器。
- 前記複室培養容器が略長方形状であり、前記複室培養容器の一つの辺に、チューブを用いて接続された二つ以上の前記室間接続部が備えられ、前記各培養室における前記室間接続部が備えられた辺と、前記複室培養容器の一つの辺との距離が、前記各培養室の並び順に大きく又は小さくなるように、前記各培養室が形成されたことを特徴とする請求項1~7のいずれかに記載の複室培養容器。
- 請求項8に記載の複室培養容器を用いた細胞培養方法であって、
前記各培養室における前記室間接続部が備えられた辺と、前記複室培養容器の一つの辺との距離が大きい培養室ほど上方に位置させ、かつ前記室間接続部が備えられた前記複室培養容器における辺が斜め下向きになるようにしつつ、前記複室培養容器の一つの角が最下端に位置するように前記複室培養容器を傾けて、上方に位置する培養室から下方に位置する培養室に向けて、内容物の移送を行うことを特徴とする細胞培養方法。 - 請求項1~3のいずれかに記載の複室培養容器を用いた細胞培養方法であって、
前記複室培養容器における前記培養室として、リンパ球を活性化させるための抗CD3抗体が固相化された活性化培養室と、リンパ球を増幅させるための増幅培養室とが形成され、
リンパ球及び培養液を前記活性化培養室に注入して、前記活性化培養室においてリンパ球を活性化させ、
前記活性化培養室から前記増幅培養室に活性化されたリンパ球及び培養液を移送して、前記増幅培養室においてリンパ球を増幅させることを特徴とする細胞培養方法。 - 請求項1~3のいずれかに記載の複室培養容器を用いた細胞培養方法であって、
