本発明においてCdc7阻害剤とは、Cdc7キナーゼ活性を阻害する薬剤をいい、低分子化合物、ポリペプチド、タンパク質、核酸(siRNA、miRNA、アプタマー等)、その他の高分子化合物等のCdc7キナーゼを阻害する薬剤を包含する。

本発明においてM期促進剤とは、細胞が細胞周期のG2/Mチェックポイントを通過して分裂期であるM期に移行するまでの過程を促進する機能を有する薬剤を言う。
G2/MチェックポイントはG2期からM期に移行する際の制御を行うチェックポイントであり、この制御がDNA損傷などで活性化されるとM期開始が阻害され、細胞はG2期に留まる。DNA損傷応答においては、リン酸化を受け活性化されたATR(ataxia-telangiectasia mutated related)がChk1をリン酸化して活性化し、次いで活性型Chk1がCdc25のリン酸化をダウンレギュレートし、リン酸化Cdc25によるCdk1の脱リン酸化を阻害するため、Cdk1は高リン酸化された不活性な状態に保たれ、細胞はM期に進行せずに細胞周期が停止する。

Cdc7阻害剤とM期促進剤の相乗的効果を調べるためWee1阻害剤であるMK-1775をCdc7阻害剤(化合物I-D又は化合物I-E)と共投与した。
COLO205(RCB2127、RIKEN BRC)は10cmディッシュで10%FCS(Equitech-bio)5%ペニシリンストレプトマイシン(GIBCO 15140)添加RPMI-1640メディウム(SIGMA R8758)を用いて培養した。良く増殖している状態(70-90%コンフルエント)の細胞からメディウムを除き、トリプシン(TrypL Express, GIBCO社製)2mLで処理して細胞をディッシュからはがし、同じメディウムで回収した。
回収した各細胞を6ウェルプレート(BD Falcon、カタログ番号 353046)に1ウェル 1mlあたり2.5x105細胞となるように蒔いた。5%CO2インキュベーター内で一晩培養した。翌日、メディウム500μlに添加したい化合物の1,000倍濃度のストック溶液を1.5μl添加したものを各ウェルに加え、最終濃度が1μMの条件下でさらに48~72時間培養を行った。
細胞を48~72時間化合物と反応後、細胞をトリプシン(TrypL Express、GIBCO社製)で処理しウェルからはがし、DPBSで洗った後、1mlの冷エタノールを添加、染色まで-30度で保存した。細胞は染色前に一度DPBSで洗い、ヨウ化プロピジウム(PI)溶液で遮光し室温で30分以上静置した。PI染色を行った細胞はFACSを用いて細胞周期及びSubG1で表される細胞死の割合の検討を行った。
まず、リン酸化されたヒストンH3のアップレギュレーションに対するMK-1775の濃度を調べ、次いでM期促進を引き起こす濃度範囲でCdc7阻害剤の添加に加えてMK-1775を大腸がん細胞であるColo205細胞の培養物に添加した。
BLISSスコアは、薬剤併用における相乗効果の評価によく用いられる基準の一つである(Borisyら Proc.Natl.Acad.Sci.USA.100(13):7977-7982(2003)、Buckら Mol.Cancer.Ther.5(11)(2006))。このBLISSスコアを用いて、Cdc7阻害剤とWee1阻害剤のがん細胞に対する増殖阻害に対する併用効果を解析した。
具体的には、Colo205細胞について、化合物(I-D)の0.1μMに対して、DMSO(対照)、Wee1阻害剤であるMK-1775の0.25μM、0.5μM、及び0.75μMをそれぞれ共投与して細胞死を起こした細胞数を調べた。化合物(I-E)の0.1μMに対しても、DMSO(対照)、MK-1775の各濃度をそれぞれ共投与して細胞死を起こした細胞数を調べた。結果を図1に示す。
化合物(I-D)又は化合物(I-E)とMK-1775(Wee1阻害剤)の相乗効果が正常な細胞でも観察されるかどうかを調べるために、正常ヒト皮膚線維芽細胞(NHDF)細胞を当該組み合わせ剤で処理して調べた。
具体的には、膵がん細胞株であるPanc-1細胞について、化合物(I-D)の0.1μMに対して、DMSO(対照)、MK-1775の0.5μM、及び0.75μMをそれぞれ共投与して細胞死を起こした細胞数を調べた。化合物(I-E)の0.1μMに対しても、DMSO(対照)、MK-1775の各濃度をそれぞれ共投与して細胞死を起こした細胞数を調べた。結果を図3に示す。Panc-1細胞(膵がん細胞株)についても、化合物(I-D)又は化合物(I-E)により誘導される細胞死は、Wee1阻害剤の共投与により相乗的に増加することがわかった。
化合物(I-E)とMK-1775の配合薬剤内の組成(μMでの比)を、0.01:1.0、0.03:0.1、及び0.3:0.1のように、1:100~3:1の範囲で変えて共投与し、Colo205細胞の細胞死を起こした細胞数を調べた。結果を図4に示す。化合物(I-E):MK-1775=1:100~3:1の範囲で、化合物(I-E)により誘導される細胞死は、Wee1阻害剤の共投与により十分相乗的に増加することがわかった。
Cdc7阻害剤として化合物(I-A)を用いた以外は、実施例1と同様に、MK-1775と共投与してColo205細胞について細胞死を起こした細胞数を調べた。結果を図5に示す。他のCdc7阻害剤として化合物(I-A)を用いても、誘導される細胞死は、Wee1阻害剤としてのMK-1775の共投与により相乗的に増加することがわかった。
Cdc7阻害剤として化合物(I-B)を用い、化合物(I-B)の濃度を0.3μMとした以外は、実施例1と同様に、MK-1775と共投与してColo205細胞について細胞死を起こした細胞数を調べた。結果を図6に示す。他のCdc7阻害剤として化合物(I-B)を用いても、誘導される細胞死は、Wee1阻害剤としてのMK-1775の共投与により相乗的に増加することがわかった。
Cdc7阻害剤として化合物(I-C)を用い、化合物(I-C)の濃度を0.1μM又は0.3μMとした以外は、実施例1と同様に、MK-1775と共投与してColo205細胞について細胞死を起こした細胞数を調べた。結果を図7に示す。化合物(I-C)を用いても、誘導される細胞死は、Wee1阻害剤としてのMK-1775の共投与により相乗的に増加することがわかった。
Cdc7阻害剤として化合物(I-F)又は化合物(I-G)を用い、その濃度を0.1μM又は0.3μMとし、MK-1775の濃度を0.5μMとした以外は、実施例1と同様に、MK-1775と共投与してColo205細胞について細胞死を起こした細胞数を調べた。結果を図8に示す。化合物(I-F)又は化合物(I-G)を用いても、誘導される細胞死は、Wee1阻害剤としてのMK-1775の共投与により相乗的に増加することがわかった。ここで、上述のように、化合物(I-G)は特表2009-531373、国際公開第2007/110344号明細書に記載の化合物F26であり、化合物(I-F)はメルク社、カタログ番号:217707-5MGCNのCdc7/Cdk9阻害剤である。
Cdc7阻害剤として化合物(I-D)又は化合物(I-E)を用い、その濃度を0.3μM又は0.1μMとし、M期促進剤としてWee1阻害剤であるMK-1775の代わりに、Chk1阻害剤であるSCH900776を用い、その濃度を3.3μMとした以外は、実施例1と同様に、両者を組み合わせて共投与して、Colo205細胞について細胞死を起こした細胞数を調べた。結果を図9に示す。他のM期促進剤としてSCH900776(SCH)を用いても、誘導される細胞死は、化合物(I-D)又は化合物(I-E)との共投与により相乗的に増加することがわかった。
Cdc7阻害剤として化合物(I-D)又は化合物(I-E)を用い、その濃度を0.1μM、0.03μM又は0.3μMとし、M期促進剤としてWee1阻害剤であるMK-1775の代わりに、Chk1阻害剤であるUCN-01を用い、その濃度を333nMとした以外は、実施例1と同様に、両者を組み合わせて共投与して、Colo205細胞について細胞死を起こした細胞数を調べた。結果を図10に示す。他のM期促進剤としてUCN-01を用いても、誘導される細胞死は、化合物(I-D)又は化合物(I-E)との共投与により相乗的に増加することがわかった。
Cdc7阻害剤として化合物(I-D)又は化合物(I-E)を用い、その濃度を0.1μM、0.03μM又は0.3μMとし、M期促進剤としてWee1阻害剤であるMK-1775の代わりに、Hsp90阻害剤であるゲルダナマイシンを用い、その濃度を10nM又は33nMとした以外は、実施例1と同様に、両者を組み合わせて共投与して、Colo205細胞について細胞死を起こした細胞数を調べた。結果を図11に示す。他のM期促進剤としてゲルダナマイシンを用いても、誘導される細胞死は、化合物(I-D)又は化合物(I-E)との共投与により相乗的に増加することがわかった。
BLISSスコアを用いて、Cdc7阻害剤とWee1阻害剤(MK-1775)のがん細胞に対する増殖阻害に対する併用効果を解析した。
ヒト乳腺癌MDA-MB-231細胞(ATCC社No.HTB-26)は、T75フラスコ中、10%ウシ胎仔血清(FBS)(Equitech-Bio社)および5%ペニシリンストレプトマイシン(ナカライ社)を添加したLeibovitz’s L-15培地(Life Technologies社、#11415-064)(以下、増殖培地)を用いて37℃インキュベーター内で培養した(CO2なし)。
MDA-MB-231細胞を細胞密度0.035×106cells/mLになるように、新鮮な増殖培地に再懸濁したのち、細胞懸濁液を96ウェル細胞培養プレート(Falcon、#353075)の各ウェルに100μLずつ添加し、37℃のインキュベーター内において1晩培養した(1ウェル当たり3,500個の細胞)。播種の約24時間後、培地を除去し、新しい増殖培地を80μL添加した。
第1試験化合物であるCdc7阻害剤(化合物(I-D)又は化合物(I-E)の10mM DMSOストック溶液を、10濃度(10mM、5mM、2.5mM、1.25mM、0.625mM、0.313mM、0.156mM、0.0781mM、0.0391mM、0.0195mM)にDMSOで希釈し、それぞれの溶液を増殖培地で更に100倍希釈して、上記プレートの各ウェルに10μL添加した(1000倍希釈となり、最終投与濃度の範囲は19.5~10000nM)。第2試験化合物のWee1阻害剤(MK-1775)の10mM DMSOストック溶液を、8濃度(1mM、0.5mM、0.25mM、0.125mM、0.0625mM、0.0313mM、0.0156mM、0.00781mM)にDMSOで希釈し、それぞれの溶液を更に増殖培地で100倍希釈して、上記プレートの各ウェルに10μL添加した(1000倍希釈となり、最終投与濃度の範囲は7.81~1000nM)。またDMSOを増殖培地で100倍希釈し、上記プレートに20μL添加したものをコントロールウェルとした。
化合物の添加後、72時間インキュベートしたのち、マイルドホルム(R)10N(和光純薬、#133-10311)を100μLずつ各ウェルに添加して室温で45分インキュベートし、細胞を固定した。各ウェルを100μLのリン酸緩衝生理食塩水(PBS)で3回洗浄し、2μMになるようにPBSで希釈したHoechst33342(Life Technologies社、#H3570)を100μLずつ各ウェルに添加して室温で30分インキュベートし、核を染色した。100μLのPBSで3回洗浄後、細胞イメージアナライザー(ArrayScan VTI HCS Reader、Thermo scientific社)を用いて核の数(細胞数)を測定した(Ex/Em=350/461nm)。
72時間後のコントロールウェルの平均値を非処理対照細胞数(Gunt72)とし、化合物処理したウェルの平均値を化合物処理細胞数(G72)とした。また、細胞播種の24時間後に培地交換しただけのウェルを初期値としてカウントし、初期細胞数(G0)とした。
細胞増殖率G=((G72-G0)/(Gunt72-G0)
また各ウェルの細胞増殖のパーセント阻害率は、以下の式で計算し、結果を図12および図13に示した。
細胞増殖阻害率GI(%)=100×(1-G)
各ウェルの理論的なBLISSindependence(BLISSin)は、第1試験化合物および第2試験化合物が各々単剤として作用したと考えたとき(つまり、どちらか一方の化合物濃度が0のときの細胞増殖率)、以下の式から算出できる。
BLISSin=G(第1試験化合物)×G(第2試験化合物)
BLISSスコア=100×(BLISSin-G)
本試験の結果を図14および15に示す。

- Cdc7阻害剤とM期促進剤とを含む薬剤組成物。
- 前記Cdc7阻害剤が、下式(I)で示されるフラノン誘導体又はその薬学的に許容される塩である、請求項1記載の薬剤組成物。
(式中、Aは、-COOR1又は水素原子を表し、R1は、水素原子、置換基を有してもよい炭化水素基、又は置換基を有してもよい複素環を表し、R2及びR3は同一又は異なってもよく、水素原子、置換基を有してもよい炭化水素基、置換基を有してもよいフェニル基、置換基を有してもよい複素環、置換基を有してもよい複素環式縮合環、又は置換基を有してもよいアミノ基を表す。あるいは、R2及びR3は、これらが結合している窒素原子と共に、置換基を有してもよい複素環又は置換基を有してもよい複素環式縮合環を形成してもよい。R4は、水素原子又はハロゲン原子を表す。ただし、Aが-COOR1を表す場合は、R2及びR3は同時に置換基を有してもよいアミノ基にならず、Aが水素原子を表す場合は、R3は水素原子を表す。) - 前記Aが-COOR1である、請求項2に記載の薬剤組成物。
