Definitions
- the present invention relates to antibodies to gremlin-l, and more particularly, to independence on bone morphogenic protein (BMP) or vascular endothelial growth factor receptor-2 (VEGFR2).
- BMP bone morphogenic protein
- VEGFR2 vascular endothelial growth factor receptor-2
- An antibody against gremlin-1 which has a therapeutic effect against cancer by inhibiting gremlin-1 in a conventional manner.
- Gremlin-l (20.7 kDa) is an antagonist of bone morphogenetic protein (BMP) and has a structure common to members of the cysteine rich region cysteine knot motif, and the TGF- ⁇ superfamily. It is a protein of 184 amino acids.
- the protein is evolutionarily conserved and the human gremlin gene is located on chromosome 15ql3-ql5 (Topol LZ et al., (1997) Mol. Cell Biol., 17: 4801-4810; Topol LZ et al., Cytogenet Cell Genet., 89: 79-84).
- Gremlin-1 has been reported with three isoforms as secretory proteins (Topol LZ et al., J. Biol. Chem., 275: 8785-8793).
- Subtype 1 is the most common subtype, and subtypes 2 and 3 delete amino acids 39-79 and 10-79, respectively.
- Gremlin-1 inhibits BMP from binding to receptors on the cell surface by forming heterodimers with BMP-2, BMP-4 and BMP-7 (Stanley E et al., (1998) Mech. Dev , 77: 173-184; Merino R et al., (1999) Development, 126: 5515-5522; Lap in DW et al., (2000) Curr. Opin. Nephrol. Hyper tens., 9: 469-472 ).
- gremlin-1 plays an important role in regulating BMP not only in the development of lung, limbs, and kidneys, but also in neural crest cell differentiation. (Lu 'et al., (2001) Dev.
- Gtemlin-1 not only antagonizes soluble ligands, but also interacts with BMP-4 precursor proteins in cells to downregulate BMP-4 mediated signaling activity in embryo lungs (Sun J et al., ( 2006) J. Biol. Chew., 281: 29349-29356.
- gremlin-1 interacts with the slit protein, one of the secreted axonal guidance proteins, and acts as an inhibitor of chemotaxis (monocytochemistry) (Chen B et al., ( 2004) J. Immunol., 173: 5914-5917). It has recently been reported that gremlin-1 binds to vascular endothelial growth factor receptor-2 (VEGFR2) and regulates angiogenesis in a BMP-independent manner (Mitola S et al., (2010) Blood, 116: 3677-3680. . .
- Gremlin-1 is overexpressed in various human tumors, including cervical, endometrial, lung, ovary, kidney, breast, colon, and pancreas cancer, but are (Namkoong H et al, (2006 ) BMC Cancer, ⁇ '74; Sha G et al., (2009) Fertil Steril., 91: 350-358), its role in tumorigenesis has not been studied in detail. While studying the tumor-specific properties of gremlin ⁇ 1, the inventors found that gremlin-1 interacts directly with cancer cells irrespective of BMP or VEGFR2, and based on this, an antibody capable of inhibiting gremlin-1 was identified.
