Abstract
Description
本発明は、骨形成タンパク質(BMP)または血管内皮成長因子受容体−2(VEGFR2)に非依存的な方式によりグレムリン−1を抑制することにより、癌または免疫疾患に対して治療効果を有するグレムリン−1に対する抗体を提供する。
また、本発明は、前記抗体及び薬学的に許容可能な添加剤を含む癌または免疫疾患の予防または治療用薬学的組成物を提供する。
さらに、本発明は、前記抗体またはこれを含む薬学的組成物をこれを必要とする対象に投与することを含む癌または免疫疾患の治療方法を提供する。
さらにまた、本発明は、前記抗体を含む癌または免疫疾患の診断用キットを提供する。
以下、本発明について詳細に説明する。
本発明は、グレムリン−1に対する抗体を提供する。前記抗体は、骨形成タンパク質(BMP)または血管内皮成長因子受容体−2(VEGFR2)に非依存的な方式によりグレムリン−1を抑制することにより、癌に対して治療効果を有することを特徴とする。前記抗体は、免疫抗体、キメラ抗体、ヒト抗体またはヒト化抗体であってもよいが、これらに制限されない。
(1)配列番号1のアミノ酸配列を有する軽鎖可変領域及び配列番号2のアミノ酸配列を有する重鎖可変領域を含む抗体;
(2)配列番号3のアミノ酸配列を有する軽鎖可変領域及び配列番号4のアミノ酸配列を有する重鎖可変領域を含む抗体;
(3)配列番号5のアミノ酸配列を有する軽鎖可変領域及び配列番号6のアミノ酸配列を有する重鎖可変領域を含む抗体;
(4)配列番号7のアミノ酸配列を有する軽鎖可変領域及び配列番号8のアミノ酸配列を有する重鎖可変領域を含む抗体;
(5)配列番号9のアミノ酸配列を有する軽鎖可変領域及び配列番号10のアミノ酸配列を有する重鎖可変領域を含む抗体;
(6)配列番号11のアミノ酸配列を有する軽鎖可変領域及び配列番号12のアミノ酸配列を有する重鎖可変領域を含む抗体;及び
(7)配列番号13のアミノ酸配列を有する軽鎖可変領域及び配列番号14のアミノ酸配列を有する重鎖可変領域を含む抗体。
前記抗体の軽鎖可変領域及び重鎖可変領域は、ヒトまたはウサギのものであってもよい。前記抗体の軽鎖可変領域及び重鎖可変領域はそれぞれ単独で用いられてもよく、両方が組み合わせられて用いられてもよく、この技術分野における公知の軽鎖不変領域及び重鎖不変領域とそれぞれ組み合わせられて用いられてもよく、これらの断片が用いられてもよい。なお、前記軽鎖可変領域及び重鎖可変領域は公知の方法により scFv(single−chain variable fragment)の形に製造されてもよい。
但し、下記の実施例は本発明を例示するためのものに過ぎないもので、本発明の範囲はこれらの実施例に限定されるものではない。
<1−1> 細胞培養
A549、HeLa、A172及びA431細胞は韓国細胞株銀行(ソウル、韓国)から入手し、ヒト臍帯静脈内皮細胞(HUVEC)はインビトロジェン社(カールスバッド、カリフォルニア州)から入手した。一方、A549、A172及びA431細胞は10%ウシ胎児血清(FBS)(GIBCO社製、グランドアイランド、ニューヨーク州)入りRPMI−1640培地(ウェルジーン社製、韓国)で、HeLa細胞は10%FBS入りMinimum Essential Medium(MEM)培地(ウェルジーン社製)で、そしてHUVECは内皮細胞成長培地−2(Endothelial Growth Medium−2;EGM−2)(ロンザ社製、ウォーカーズヴィル、メリーランド州)で培養した。
文献[Namkoong H et al., (2006) BMC Cancer 6: 74]の方法を参照して、ヒト子宮組織cDNAライブラリーからグレムリンcDNAを増幅させて得た。次いで、下記のPCRプライマーを用いたポリメラーゼ連鎖反応(PCR)法により前記グレムリンcDNAの5’及び3’末端にHindIII及びXhoI制限酵素部位を導入した。

グレムリン−1及びヒトIgG−Fc融合タンパク質をコードする遺伝子を文献[Park S et al., (2010) Clin Chim Acta 411: 1238−1242]に記載の方式に従い、オーバーラッピングPCR(overlapping PCR)法を用いて製造した。前記ポリメラーゼ連鎖反応(PCR)に用いられたグレムリン−1及びIgG−Fcに対するリンカープライマー配列は、下記の通りである:

<2−1> 免疫化
5μgのグレムリン−1−Fcを2mLのリン酸緩衝食塩水(PBS)と混合し、37℃で30分間培養して、これを2%スクワレン中に毒素が除去された内毒素であるMPL(monophosphorylate lipid A species)及びマイコバクテリアの細胞壁成分であるTDW及びCWSを含有する乳中水乳化液である補助剤(シグマ社製、セントルイス、ミズーリ州)に乳化させた後、ニュージーランド産の白いウサギに注入した。免疫化を3週おきに3回行った。免疫化されたウサギの抗体力価(titer)を二次抗体として西洋ワサビペルオキシダーゼ(HRP)共役のマウス抗ウサギIgG多クローン抗体(ピアス・ケミカル社製、ロックフォード、イリノイ州)を用いて酵素結合免疫吸着分析(ELISA)によって決定した。
オリゴ(dT)プライミング(priming)を有する第一鎖cDNA合成キットに対するスーパースクリプト(SuperscriptTM)IIIシステム(インビトロジェン社製)を用いて第一鎖cDNAを合成した。前記分離された5μgの総RNAを1μLの0.5μg/μLオリゴ(dT)、1μLの10mM dNTP及びジエチルピロカーボネート(DEPC)で処理された水と混合して最終的な体積を10μLに調整した。前記混合物を65℃で5分間培養し、氷中で冷却した。前記RNA試料に2μLの10倍濃度反応緩衝液、4μLの25mM MgCl2、2μLの100mMジチオトレイトール(DTT)、1μLのRNase OUT(リボヌクレアーゼ阻害剤)及び1μLのSuperscriptIII逆転写酵素を添加し、50℃で50分間培養した。85℃で5分間培養した後、氷中で冷却して反応を終了した。次いで、1μLのRNase Hを添加し、37℃で20分間培養した。前記第一鎖cDNAを−20℃で保管した。
ウサギの脾臟及び骨髄から得た第一鎖cDNAに対してExpand High Fidelity PCRシステム(ロシュ・モレキュラーシステムズ社製、インディアナ州、米国)を用いた30サイクルのPCRを行った。ウサギVL(9×Vk及び1×Vλ)コード配列の増幅のための10個のプライマー組み合わせ及びウサギVHコード配列(表1)の増幅のための4つの組み合わせを併用した。各反応において、1μLのcDNAを60pmolの各プライマー、10μLの10倍濃度反応緩衝液、8μLの2.5mm dNTP(プロメガ社製、マディソン、ウィスコンシン州)、0.5μLのTaq DNA重合酵素及び水と混合して最終的な体積を100μLに調整した。下記の条件下でPCRを行った:94℃で15秒、56℃で30秒及び72℃で90秒の30サイクル後に、72℃で10分間の最終延長。約350bpの長さを有する断片を1.5%アガロースゲルに搬入して電気泳動した後、QIAEXIIゲル抽出キット(キアゲン社製、バレンシア、カリフォルニア州)を用いて精製した。精製されたポリメラーゼ連鎖反応(PCR)産物をOD 260nmにおいて読み取って定量した(1OD単位=50μg/mL)。
2次PCRにおいて、1次VL産物をオーバーラップ延長PCRによって1次VH産物とランダムに連結した(表2)。長いリンカーの単鎖断片に対して少なくとも10回の反応を行った。各反応において、100ngの精製された軽鎖産物及び重鎖産物を60pmolの各プライマー、10μLの10倍濃度反応緩衝液、8μLの2.5mM dNTP(プロメガ社製)、0.5μLのTaq DNA重合酵素及び水と混合して最終的な体積を100μLに調整した。下記の条件下でPCRを行った:94℃で15秒、56℃で30秒及び72℃で2分の20サイクル後に、72℃で10分間の最終延長。約700bpの大きさの断片を1.5%アガロースゲルに搬入して電気泳動した後、QIAEXIIゲル抽出キット(キアゲン社製、バレンシア、カリフォルニア州)を用いて精製した。精製されたPCR産物をOD 260nmにおいて読み取って定量した(1OD単位=50μg/mL)。
PCR産物及びpComb3Xベクター(スクリプス研究所製)をクローニングのためにSfiI制限酵素で処理した。10μgの精製されたオーバーラップPCR産物を360ユニットのSfiI(1μgのDNA当たりに16ユニット、ロシュ・モレキュラーシステムズ社製)、20μLの10倍濃度反応緩衝液M及び水と混合して最終的な体積を200μLに調整した。20μgのpComb3Xベクターを120ユニットのSfiI(1μgのDNA当たりに6ユニット、ロシュ・モレキュラーシステムズ社製)、20μLの10倍濃度反応緩衝液M及び水と混合して最終的な体積を200μLに調整した。前記混合物を50℃で5時間培養した。約700bpの長さを有する切断された挿入物を1%アガロースゲルで精製し、約3,400bpの長さを有するベクターDNA及びpComb3Xベクター内に含有されている約1,600bpの長さを有する断片を0.6%アガロースゲルで精製した。
高効率のライゲーション及び形質転換のためのベクター並びに挿入物の適合性を評価するために、小規模のライゲーションを行った。ベクターDNAのSfiI処理物のみを含有するライゲーション反応を5%未満のライブラリーサイズであるバックグラウンドライゲーションに対して評価した。ライゲーションのために、140ngのSfiIで処理されて精製されたベクターDNA及び70ngのSfiIで処理されて精製されたPCR産物またはスタッファーDNA、4μLの5倍濃度リガーゼ緩衝液、1μLのT4リガーゼ(インビトロジェン社製)及び水を混合して総体積を20μLに調整した。前記ライゲーション混合物を室温下で4時間培養した。前記ライゲーションされた産物1μLを0.2cmキュベット及び遺伝子パルサー (Genepulser;バイオラッドラボラトリーズ製、ハーキュリーズ、カリフォルニア州)を用いて2.5kV、25μF及び200Ωで電気穿孔法によって50μLのER2738エレクトロコンピテント細胞(NEB)内に形質転換させた。前記細胞を3mLのSB培地に再懸濁させ、225〜250rpmで攪拌しながら37℃で1時間培養した。形質転換体の総数をLB+カルベニシリンプレートの上に1μL、10μL及び100μLを塗抹して決定した。理想的には、最終的なライブラリーの大きさはベクターDNAμgに対して少なくとも108集落形成単位(cfu)にならなければならず、5%バックグラウンドライゲーション未満でなければならない。
大腸菌ER2738菌株を15mLのSB入り50mLのポリプロピレンチューブで培養し、250rpmにて攪拌しながら37℃で一晩中成長させた。翌日に、2.5mLの培養物を500mLのSB、10mLの20%(w/v)グルコース及び5mLの1M MgCl2入り2Lフラスコ内に希釈させ、600nmにおけるODが0.8〜0.9に達するまで37℃で250rpmで攪拌した。次いで、培養物を予め冷却した500mLの遠心分離瓶に入れ、4℃で20分間3,000gにおいて遠心分離した。上澄み液を除去し、ペレットを予め冷却した30mLの10%(v/v)グリセロールに再懸濁させた。再懸濁されたペレットを遠心分離し、グリセロールで3回洗浄した後、5mLの10%グリセロールに再懸濁して−80℃で保管した。
<2−7>において製造された10μLのER2738懸濁液を10mLのSB培地に接種し、37℃で1時間250rpmにて攪拌した。新たに製造されたプレートから得た単一ヘルパーファージプラークをピペットチップを用いて培養物内に移した。前記感染された10mLの培養物をカナマイシン入りの予め加熱した(37℃)SB培地500mL入りの2Lの三角フラスコに移して最終的な濃度を70μg/mLに調整した後、37℃で一晩中250rpmにて攪拌した。翌日に、培養物を2,500gにおいて15分間遠心分離し、上澄み液を水槽中において70℃で20分間培養した。2,500gにおいて15分間遠心分離した後、上澄み液を新たな50mLのポリプロピレンチューブに移して4℃で保管した。
1.4μgのSfiIで切断したpComb3X、700ngの<2−5>において得た、SfiIで切断されたPCR産物、40μLの5倍濃度リガーゼ緩衝液、10μLのT4 DNAリガーゼ及び水(最終的な体積を200μLに調整する)を用いてライブラリーライゲーションを行った。前記ライゲーション混合物を室温下で一晩中培養した後、−80℃で一晩中エタノールで沈殿させた。マイクロ遠心分離器を用いて最高の速度にて4℃で15分間遠心分離した後、上澄み液を除去し、ペレットを1mLの70%(v/v)エタノールで洗浄して乾燥させた。ペレットを15μLの水に溶解させた。前記結さつされたライブラリーサンプルを300μLのエレクトロコンピテント大腸菌内に形質転換させ、5mLのSB培地において37℃で1時間培養した。前記培養物に100mLの予め加熱したSB培地及び3μLの100mg/mLカルベニシリンを添加した。50μg/mLのカルベニシリンを含有するLB培地に前記培養物0.1μL、1μL及び10μLを塗抹してライブラリーの大きさを決定した。培養物を250rpmにて1時間攪拌した。前記培養物に4.5μLの100mg/mLカルベニシリンを添加して1時間さらに攪拌した。前記培養物に2mLのVCSM13ヘルパーファージ、183mLの予め加熱したSB及び92.5μLの100mg/mLのカルベニシリンを添加して300rpm及び37℃で2時間攪拌した。前記培養物に280μLの50mg/mLのカナマイシンを添加し、前記培養物を300rpm及び37℃で一晩中攪拌した。翌日に、培養物を3,000gにおいて4℃で15分間遠心分離した。上澄み液を500mLの遠心分離瓶に移し、次いで、8gのポリエチレングリコール(PEG)−8000及び6gのNaClを添加した。氷中で30分間保管した後、上澄み液を15,000gにおいて4℃で15分間遠心分離した。上澄み液を廃棄し、ファージペレットを1%ウシ血清アルブミン(BSA)を含有するトリス緩衝食塩水(TBS)に再懸濁させた。
常磁性ビーズ(ダイナル・バイオテク社製、レイクサクセス、ニューヨーク州)を用いてバイオパニングを行った。3μgのグレムリン−1を室温下で20時間1×107ビーズと結合させた。前記ビーズをリン酸緩衝生理食塩水(Phosphate buffered saline;PBS)で4回洗浄し、ブロッキング緩衝液とともに室温下で1時間培養した。グレムリン−1と結合したビーズを<2−9>において得たscFvをディスプレイするファージとともに室温下で2時間培養した。前記洗浄段階を1次における3回から後続する4次においては10回に増やした。結合されたファージを50μLの0.1Mグリシン/HCl(pH2.2)を用いて溶出させ、3μLの2M Tris−Cl(pH9.1)によって中和させた。このようなファージ入り上澄み液を用いてER2738を感染させ、ファージミドを一晩中増幅するためにヘルパーファージVCSM13を用いてレスキュー(rescue)した。翌日に、<2−9>において上述したようにPEG−8000及びNaClを添加してファージを製造した。なお、前記ファージにより感染された培養物を50μg/mLのカルベニシリンを含有するLBプレートの上に塗抹して投入(input)及び生産(output)ファージ力価を決定した。
追加の分析のために選ばれた個別クローンからscFv結合を確認するために、精製された組換えグレムリン−1に対してscFvをディスプレイするファージを用いてELISAを行った。グレムリン−1によりコーティングされたマイクロタイタープレートをリン酸緩衝生理食塩水(PBS)中の3%のウシ血清アルブミン(BSA)を用いて37℃で1時間ブロッキングした。次いで、ファージ上澄み液をPBS中の6%BSAと同様に混合し、37℃で1時間培養した。0.05%のツイーン添加リン酸緩衝生理食塩水(Tween added Phosphate Buffered Saline;PBST)で洗浄した後、プレートをHRPの結合された抗M13抗体(1:5000希釈、ピアス・ケミカル社製)とともに培養した。色相反応のために、2,2’−アジノ−ビス(3−エチルベンゾチアゾリン−6−スルホン酸;ABTS)基質溶液(アムレスコ社製、ソロン、オハイオ州)を使用した。
scFv断片を全長IgGに転換させて過発現させた。本発明による抗体(O−13)の特異性をウェスタンブロット分析法を用いて決定した。本発明による抗体(O−13)の特異性を決定するために、pcDNA3.1/myc−His−グレムリン−1で形質感染されたHEK293F細胞の培養上澄み液をドデシル硫酸ナトリウム・ポリアクリルアミドゲル電気泳動(Sodium Dodecyl Sulfate−Polyacrylamide Gel Electrophoresis;SDS−PAGE)で分離した。ブロットを100ng/mLの本発明による抗体(O−13)と4℃で一晩中培養した後、HRPの結合された抗ヒトIgG(Fc特異的、1:1000希釈;ピアス・ケミカル社製)とともに室温下で1時間培養した。ブロットをメーカーのガイドラインに従い、向上された化学発光システム(ピアス社製)を用いて視覚化させた。ゲルをメーカーのガイドラインに従い、クマシーブリリアントブルーG−250(シグマ社製)を用いて視覚化させた。
前記過程により得た7種類の抗体をそれぞれR−80、R−88、O−1、O−13、O−26、J−1及びO−13(ヒト化)抗体と命名し、アミノ酸及び塩基配列を分析した。
配列分析の結果、前記R−80抗体は、配列番号1のアミノ酸配列を有する軽鎖可変領域及び配列番号2のアミノ酸配列を有する重鎖可変領域を含み、R−88抗体は、配列番号3のアミノ酸配列を有する軽鎖可変領域及び配列番号4のアミノ酸配列を有する重鎖可変領域を含み、O−1抗体は、配列番号5のアミノ酸配列を有する軽鎖可変領域及び配列番号6のアミノ酸配列を有する重鎖可変領域を含み、O−13抗体は、配列番号7のアミノ酸配列を有する軽鎖可変領域及び配列番号8のアミノ酸配列を有する重鎖可変領域を含み、O−26抗体は、配列番号9のアミノ酸配列を有する軽鎖可変領域及び配列番号10のアミノ酸配列を有する重鎖可変領域を含み、J−1抗体は、配列番号11のアミノ酸配列を有する軽鎖可変領域及び配列番号12のアミノ酸配列を有する重鎖可変領域を含み、O−13(ヒト化)抗体は、配列番号13のアミノ酸配列を有する軽鎖可変領域及び配列番号14のアミノ酸配列を有する重鎖可変領域を含む。
予備実験により、前記抗体のうち最も優れた効果を有するO−13抗体を今後の実験に供試した。
グレムリン−1が癌細胞と直接的に相互作用するかどうかを調べるために、グレムリン−1を4種類の癌細胞(A549、HeLa、A172及びA431)と共に培養した後、フローサイトメトリーにより分析した。
具体的に、付着細胞をトリプシンで処理し、リン酸緩衝食塩水(PBS)中の1%(w/v)ウシ血清アルブミン(BSA)で洗浄した。懸濁細胞を500×gにおいて2分間遠心分離して回収し、PBS中の1%(w/v)BSAで洗浄した。全ての細胞をPBS中の1%(w/v)BSAで100nMの最終濃度のヒスチジンヘキサマータグ(His−tag)を有するグレムリン−1(R&Dシステムズ社製、ミネアポリス、ミネソタ州)と共に37℃で1時間培養した。次いで、細胞をPBS中の1%(w/v)BSAで2回洗浄し、5μg/mlの最終濃度のFITC-結合されたHis抗体(アブカム社製、ケンブリッジ、イギリス)と共に暗所で37℃で30分間培養した。次いで、細胞をPBS中の1%(w/v)BSAで2回洗浄し、FACSCantoIIフローサイトメーター(BDバイオサイエンス社製、サンノゼ、カリフォルニア州)上において分析する前に500μLのPBSに再懸濁させた。
一方、グレムリン−1抗体(O−13)の中和効能を分析するために、細胞をヒスチジンヘキサマータグ(His−tag)を有するグレムリン−1 100nM及びPBS中の1%(w/v)BSA中の10μMの本発明による抗体(O−13)と共に37℃で1時間培養した後、FITCの結合されたHis抗体(アブカム社製)で探針させた。
前記実験結果を図1に示す。同図に示すように、グレムリン−1は癌細胞株と直接的に相互作用し、これはグレムリン−1抗体によって抑制された。
グレムリン−1と細胞株との相互作用がグレムリン−1の唯一に知られている細胞表面受容体であるVEGFR2によって媒介されるか否かを評価した。
グレムリン−1がヒト臍帯静脈内皮細胞(HUVEC)と結合するか否かを実施例3と同じ過程に従い、フローサイトメトリーによって分析した。前記結果を図2Aに示す。同図に示すように、グレムリン−1はHUVECと結合しなかった。HUVECがVEGFR2を発現するため、グレムリン−1がHUVECと結合しないということは、グレムリン−1がVEGFR2とは無関係に癌細胞に結合するということを示唆する。
A549細胞、HeLa細胞及びHUVEC細胞 における VEGFR2mRNA及びタンパク質の発現をRT−PCR及び免疫ブロット分析法を用いて確認した。
具体的に、RT−PCRのために、トリゾール試薬(インビトロジェン社製)を用いてメーカーのガイドラインに従い、A549細胞、HeLa細胞及びHUVECから総RNAを分離した。次いで、前記RNAからスーパースクリプト(SuperscriptR)III第一鎖(First−Strand)合成システム(インビトロジェン社製)を用いてcDNAを合成した。下記のプライマーを用いて2720サーマルサイクラー(アプライドバイオシステムズ社製、フォスターシティ、カリフォルニア州)においてRT−PCRを行った。RT−PCRは94℃で30秒、56℃で30秒及び72℃で1分間35サイクルで行った。

HUVEC、A549細胞及びHeLa細胞をプロテアーゼ抑制剤カクテル(シグマアルドリッチ社製、セントルイス、ミズーリ州)を含有する冷たい溶解緩衝液[50mM Tris−HCl(pH7.4)、150mM NaCl、2mM EDTA、1%Triton−X100、0.1%SDS、1mM PMSF]に溶解させた後、文献[Lee MS et al., (2008) Hybridoma (Larchmt) 27: 18−24]に記載の過程に従いウェスタンブロットを行った。このとき、一次抗体としてVEGFR−2(1:1,000希釈;セル・シグナリング・テクノロジー社製、ダンバース、マサチューセッツ州)及びβ−アクチン(1:10,000希釈;アプライドバイオロジカルマテリアルズ社製、リッチモンド、ブリティッシュコロンビア州)抗体を使用し、二次抗体として西洋ワサビペルオキシダーゼ(HRP)の結合された抗マウスIgG(1:1,000希釈;ピアス・ケミカル社製、ロックフォード、イリノイ州)またはHRPの結合された抗ウサギIgG(1:1,000希釈;ピアス・ケミカル社製)を使用した。ブロットを増強された化学発光システム(ピアス社製)を用いてメーカーのガイドラインに従い視覚化させた。
前記免疫ブロット分析実験結果を図2Cに示す。図2Cに示すように、HUVECとは異なり、A549及びHeLa細胞はVEGFR2を発現しなかった。前記結果もまた、グレムリン−1と癌細胞との相互作用がVEGFR2によって媒介されないということを示す。
グレムリン−1は、BMP−2、BMP−4及びBMP−7に特異的に結合してこれらの活性を抑制するBMP拮抗剤である。BMPは多くの組織の形態の形成及び恒常性において重要な役割を果たすと知られている多機能性成長因子である。また、BMP−2、BMP−4及びBMP−7は乳房癌及び前立腺癌をはじめとする様々な癌において頻繁に過発現される(Singh A et al., (2010) Cytokine Growth Factor Rev 21: 299−313; Chen D et al., (2004) Growth Factors 22: 233−241; Bobinac D et al., (2005) Croat Med J 46: 389−396)。BMP−4はBCC細胞の増殖を減少させ、グレムリン−1の添加はBMP−4の抗増殖効果を間接的に減少させることが報告されている(Sneddon JB et al., (2006) Proc Natl Acad Sci U S A 103: 14842−14847)。このため、BMPがグレムリン−1と癌細胞との相互作用に影響を及ぼすか否かを調べてみた。
まず、グレムリン−1がBMPと結合するか否かを調べるために、下記のようにして酵素免疫分析を行った。具体的に、マイクロタイタープレート(コーニング・コスター社製、ケンブリッジ、マサチューセッツ州)を100nMのBMP−2、BMP−4またはBMP−7(R&Dシステム社製)でコーティングし、PBS中の1%(w/v)脱脂油でブロッキングした。次いで、各ウェルにグレムリン−1−Fc(10nM)単独及びグレムリン−1−Fc(10nM)と500nMの本発明による抗体(O−13)との組み合わせを添加した。洗浄後に、プレートをHRPの結合されたIgG−Fcに特異的な抗体(1:5,000希釈;ピアス・ケミカル社製)とともに培養した。次いで、文献[Chung J et al., (2004) FASEB J 18: 361−363]に従い、2,2’−アジノ−ビス(3−エチルベンゾチアゾリン−6−スルホン酸;ABTS)基質溶液(アムレスコ社製、ソロン、オハイオ州)を用いて発色反応(coloring reaction)させ、405nmにおける吸光度を測定した。前記結果を図3Aに示す。図3Aに示すように、グレムリン−1はBMP−7を除くBMP−2及びBMP−4と相互作用することが分かり、本発明による抗体(O−13)はグレムリン−1とBMP−2またはBMP−4との相互作用にいかなる影響も及ぼさなかった。
A549細胞とグレムリン−1を反応させ、ここに10倍モル過量のBMP−2、BMP−4及びBMP−7を加えた後、実施例3の方法と同様にしてフローサイトメトリーを行った。前記結果を図3Bに示す。図3Bに示すように、BMPの存在はグレムリン−1のA549細胞との相互作用に影響を及ぼさなかった。これらの結果は、グレムリン−1にA549細胞とBMPとの相互作用を媒介する2つの別途のモチーフが存在することを示唆する。
<6−1> 細胞分散の分析
グレムリン−1処理時における癌細胞の形状を調べるために、A549細胞をクリスタルバイオレット染色法により分析した。具体的に、A549細胞を24ウェルプレート(1.0×104細胞/ウェル)に塗抹し、100nMのヒスチジンヘキサマータグ(His−tag)を有するグレムリン−1で3日間処理した。培地を除去し、細胞をリン酸緩衝生理食塩水(PBS)で洗浄した後、PBS中の4%パラホルムアルデヒドで10分間固定させた。細胞を蒸留水中の0.05%クリスタルバイオレットで30分間染色した。文献[Li XL et al., (2009) Cancer Chemother Pharmacol 64: 1097−1104]に開示された方法に従い染色溶液を除去し、細胞をPBSで3回洗浄した。ライカDFL290カメラ(ライカマイクロシステムズ社製、ヴェッツラー、ドイツ)を用いてイメージを得、これをライカ・アプリケーション・スイート・ソフトウェア(ライカマイクロシステムズ社製)を用いて分析した。
前記撮影されたイメージを図4Aに示す。図4Aに示すように、グレムリン−1で処理されたA549細胞はその形状が繊維芽細胞とほとんど同様であり、細胞が分散されたことが分かる。
E−カドヘリン(E-cadherin)の下向き調節は上皮間葉移行(epithelial−mesenchymal transition;EMT)と関連があり、癌が進行したときにE−カドヘリン及び上皮間葉移行(EMT)の抑制が共通して観察される(Perl AK et al., (1998) Nature 392: 190−193)。このため、免疫ブロット分析法及び免疫蛍光染色によりグレムリン−1処理によるE−カドヘリンの発現の変化を調べてみた。
免疫ブロット分析の場合、A549細胞(1.0×105細胞/ウェル)を60mmのディッシュの上に塗抹し、50%コンフルエンシーまで成長させた。前記細胞を100nMのヒスチジンヘキサマータグ(His−tag)を有するグレムリン−1で3日間処理した。前記細胞をプロテアーゼ抑制剤カクテル(シグマアルドリッチ社製、セントルイス、ミズーリ州)を含有する冷たい溶解緩衝液[50mM Tris−HCl(pH7.4)、150mM NaCl、2mM EDTA、1%Triton−X100、0.1%SDS、1mM PMSF]に溶解させた後、文献[Lee MS et al., (2008) Hybridoma (Larchmt) 27: 18−24]に記載の過程に従いウェスタンブロットを行った。このとき、一次抗体としてE−カドヘリン(1:1,000希釈;アブカム社製)及びβ−アクチン(1:10,000希釈;アプライドバイオロジカルマテリアルズ社製、リッチモンド、ブリティッシュコロンビア州)抗体を使用し、二次抗体として西洋ワサビペルオキシダーゼ(Horseradish peroxidase、HRP)の結合された抗マウスIgG(1:1,000希釈;ピアス・ケミカル社製、ロックフォード、イリノイ州)またはHRPの結合された抗ウサギIgG(1:1,000希釈;ピアス・ケミカル社製)を使用した。前記ブロットを増強された化学発光システム(ピアス社製)を用いてメーカーのガイドラインに従い視覚化させた。
また、免疫蛍光染色の場合、A549細胞(1.0×105細胞/ウェル)をポリL−リシン(100μg/ml、シグマ社製)によりコーティングされたガラスカバースリットの上に塗抹し、50%コンフルエンシーまで成長させた。細胞を100nMのヒスチジンヘキサマータグ(His−tag)を有するグレムリン−1で3日間処理し、PBSで洗浄した後、PBS中の4%パラホルムアルデヒドで常温下で30分間固定させた。固定された細胞をPBS中の0.2%トリトンX−100(ツイーン添加リン酸緩衝生理食塩水(PBST))で常温下で10分間透過させた後、PBST中の1%ゼラチンで常温下でブロッキングした。E−カドヘリン抗体(アブカム社製)後にアレクサ(Alexa)488の結合された二次抗体(インビトロジェン社製)を用いて免疫蛍光染色を行った。核は4’,6−ジアミジノ−2−フェニルインドール( DAPI)(rhodamine-phalloidin 1:1,000希釈;インビトロジェン社製)で染色し、アクチンフィラメントはローダミン・ファロイジン(1:1,000希釈;インビトロジェン社製)を用いて染色した。カバースリットを退色防止剤(anti-fading agent)を有する水溶性マウンティング培地(aqueous moutingmedium)を用いてガラススリットの上に載せた。次いで、LSM 5 PASCALレーザースキャニング顕微鏡(カール・ツァイス社製、ドイツ)を用いてイメージを得、これをLSM 5 PASCALソフトウェアを用いて分析した。
前記結果を図4Cに示す。図4Cに示すように、グレムリン−1で処理されたA549細胞においてE−カドヘリン(E-cadherin)の発現が大幅に減少された。
細胞の移動有無を確認するために、下記のようにして細胞移動分析を行った。具体的に、細胞を1ウェル当たりに1.0×105細胞の密度で24ウェルプレートに塗抹した。ピペットチップでスクラッチして傷を付けた。脱着された細胞を除去するために培地で洗浄した後、前記培養物に100nMのヒスチジンヘキサマータグ(His−tag)を有するグレムリン−1を24時間添加した。直後に、且つ、24時間後にライカDFL290カメラ(ライカマイクロシステムズ社製)を用いてイメージを分析した。スクラッチにより形成された面積を通じて細胞が移動した距離を24時間目に傷付幅を測定し、開始時点の傷付幅からこれを差し引くことにより決定した。次いで、前記得られた値を従来の技術に従い未処理の細胞の移動距離を100%に設定して%移動で表した。
本発明による抗体(O−13)の中和効能を決定するために、傷付いた細胞をヒスチジンヘキサマータグ(His−tag)を有するグレムリン−1単独または10μMの本発明による抗体(O−13)(または、10μMの対照群抗体)とともに24時間培養した。上述したようにして距離を決定した。
同じプロトコールを用いて、Mockの形質感染されたA549細胞及びグレムリン−1で形質感染されたA549細胞を塗抹し、傷を付けた。 Mock-A549細胞はいかなる処理なしに培養したのに対し、グレムリン−1−A549細胞は10μMの本発明による抗体(O−13)または対照群抗体の存在下で24時間培養した。上述したようにして距離を決定した。結果は、3回の独立した実験の代表値である。
前記測定結果を図4Dに示す。図4Dに示すように、グレムリン−1で処理した場合、A549細胞の移動が大幅に増大され、これは、中和抗体を添加したときに完全に消失した。
これらの結果は、グレムリン−1が癌細胞の分散及び移動を誘導し、これをグレムリン−1抗体を用いて抑制することができるということを示す。
<7−1> グレムリン−1mRNA及びタンパク質発現量の測定
実施例<1−2>において製造された「グレムリン−1−A549」細胞株及び「 mock-A549」細胞株に対して、グレムリン−1転写体及びタンパク質の数値変化を上述した逆転写酵素-ポリメラーゼ連鎖反応(RT−PCR)及びウェスタンブロットによって確認した。
前記実験結果を図5Aに示す。図5Aに示すように、 mock-A549細胞株においてはグレムリン−1mRNA及びタンパク質が発現されなかったのに対し、グレムリン−1−A549細胞株においてはその数値が急増した。
また、下記のように免疫ブロット分析方法に従い、E−カドヘリンの発現量を測定した。具体的に、グレムリン−1−A549細胞及びMock-A549細胞(1.0×105細胞/ウェル)を60mmディッシュの上に塗抹して50%コンフルエンシーまで成長させた。Mock-A549細胞を処理なしに培養し、グレムリン−1−A549細胞を10μMの本発明による抗体(O−13)または対照群抗体(パリビズマブ(Palivizumab)、シナジス、アボット・ラボラトリーズ製、アボットパーク、イリノイ州)の存在下で24時間培養した。細胞を溶解させ、実施例6に記載の免疫ブロット分析方法に従い分析した。
前記結果を図5Bに示す。図5Bに示すように、E−カドヘリン発現はMock-A549細胞と比較してグレムリン−1−A549細胞において減少し、中和抗体を添加した場合に僅かに増加した。
細胞外マトリックス(extra−cellular matrix:ECM)によりコーティングされた内側チャンバー(ケミコン社製、テメキュラ、カリフォルニア州)を用いてメーカーのガイドラインに従い細胞浸潤分析を行った。Mock-A549細胞及びグレムリン−1−A549細胞(3.0×105細胞/ウェル)を300μLの無血清培地に懸濁させた。プレートの底面ウェルに10%ウシ胎児血清(FBS)を含有する完全培地(500μL)を添加した。細胞を48時間培養した。非移動細胞を除去し、リン酸緩衝生理食塩水(PBS)で洗浄した。膜をPBS中の4%パラホルムアルデヒドで固定させ、クリスタルバイオレット染色溶液(ケミコン社製)で染色した。ライカDFL290カメラ(ライカマイクロシステムズ社製)を用いてイメージを得、これをライカ・アプリケーション・スイート・ソフトウェアを用いて分析した。移動された細胞を1ウェル当たりに4個の別途のフィールドにおいて計数した。次いで、得られた値をMock-A549細胞の細胞数を100%に設定して%浸潤で表した。
前記結果を図5Cに示す。図5Cに示すように、Mock-A549細胞と比較してグレムリン−1−A549が遥かに多量移動した。
実施例6に記載の方法に従い細胞移動分析を行った。Mock-A549細胞及びグレムリン−1−A549細胞を塗抹し、傷を付けた。Mock-A549細胞は何の処理なしに培養し、グレムリン−1−A549細胞は10μMの本発明による抗体(O−13)または対照群抗体の存在下で24時間培養した。次いで、上述したようにして距離を決定した。
前記測定結果を図5Dに示す。前記図5Dに示すように、グレムリン−1−A549細胞はMock-A549細胞と比較して増加された移動を示し、このような増加された移動は本発明による抗体(O−13)を添加したときに大幅に抑制された。
グレムリン−1が細胞成長に影響を及ぼすか否かを確認するために、細胞増殖の分析を行った。前記分析は、セルタイター96アクオス・ワン・ソリューション・セル・プロリフェレーション・アッセイ(CellTiter 96 Aqueous One Solution Cell Proliferation Assay)(プロメガ社製、マディソン、ウィスコンシン州)を用いて10%FBS入りRPMI−1640培地において96ウェルプレートで行った。具体的に、Mock-A549細胞及びグレムリン−1−A549細胞を1ウェル当たりに1,000の細胞密度で塗抹した。24時間後に細胞を無血清培地で2回洗浄し、3μMの本発明による抗体(O−13)が含有するかあるいは含有していない完全培地100μLで培養した。細胞増殖を492nmのフィルターを有する96ウェルプレートに対してラブシステムズ・マルチスカン・アセント・フォトメトリック・プレート・リーダー(Labsystems Multiskan Ascent Photometric plate reader)(サーモ・ラブシステムズ社製、フランクリン、マサチューセッツ州)を用いて決定した。実験は3回繰り返し行った。
前記測定結果を図5Eに示す。前記図5Eに示すように、グレムリン−1−A549細胞はMock-A549細胞と比較してさらに高い成長速度を示し、前記増大された成長速度は本発明による抗体(O−13)の添加によって抑制された。
腫瘍形成に対するグレムリン−1の効果を評価するために、下記のようにしてグレムリン−1−A549細胞またはMock-A549細胞をヌードマウス内に皮下注射した後、腫瘍体積を測定した。
全ての動物実験は実験動物資源ソウル大学協会及び使用委員会(Permit number:SNU−11−0207)の承認を受けた。グレムリン−1−A549細胞及びMock-A549細胞(1.0×106細胞/マウス)を4〜6週齢の雌無胸腺ヌードマウス(各処理群当たりに7匹)の右側わき腹に皮下注射した。腫瘍の形成及び大きさを腫瘍の長さ及び幅をノギスで測定して毎週評価した後、下記式を用いて腫瘍の体積を計算した(Tomayko MM et al., (1989) Cancer Chemother Pharmacol 24: 148−154):
腫瘍の体積=(長さ×幅×高さ)/2
前記測定結果を図5Fに示す。グレムリン−1−A549細胞を注射したマウスにおける腫瘍の体積は、 Mock-A549細胞を注射したマウスよりも急増し、注射してから14週目に略500mm3の腫瘍サイズ差を示す。この結果は、グレムリン−1の発現増加が腫瘍の形成に重要な役割を果たすということを示す。
免疫組織化学染色法を用いて正常組織及び癌組織(皮膚、乳房、リンパ節、肺、肝、食道、胃、結腸、直腸、腎臓、膀胱、前立腺、睾丸、子宮頸、子宮内膜及び甲状腺癌組織)を本発明による抗体(O−13)と反応させてグレムリン−1の発現量を比較した。
具体的に、本発明による抗体(O−13)(2mg/ml)0.5mlをFITC試薬と混合してメーカーのガイドラインに従い抗体をFITCでラベリングした(Pierce FITC antibody labeling kit, 53027)。正常組織及び癌組織を固定させた組織配列スライド(Tissue array slide;スーパーバイオチップ社製、BC8)に対してメーカーのガイドラインに従いパラフィン除去作業を行った。前記スライドを抗原賦活液(antigen retrieval solition; TEpH9.0)入りガラスジャー(glass jar)に入れた後、プレッシャークッカー(pressure cooker)で30分間加熱した。スライドを取り出して60〜65℃に冷却させ、蒸留水で洗浄し、冷たい95%エタノールに10分間浸漬した後、流水で10分間洗浄した。次いで、メーカーのガイドラインに従い、サーモ・ウルトラビ 液ジョン・LP・デテクション・システム(Thermo Ultra−vision LP detection system(サーモ社製)を用いて組織スライドに前記FITCによりラベリングされた抗体を反応させた。前記抗体は1:75で使用し、抗FITCマウスIgG(ライフスパン(LS)・バイオサイエンス社製)は1:200で使用した。次いで、HRPの結合された重合体(ラブビジョン社製)を15分間処理した後、DAB溶液を1分間処理した。マイヤ−ヘマトキシリン液(Mayer’s hematoxylin solution)で反対染色した後、顕微鏡で4倍率、200倍率及び400倍率の順に観察した。前記実験を行うに当たって、IgGkappa抗体を対照群として使用した。
Claims (16)
- グレムリン−1に対する抗体。
- 骨形成タンパク質(BMP)または血管内皮成長因子受容体−2(VEGFR2)に非依存的な方式によりグレムリン−1を抑制することにより、癌に対して治療効果を有することを特徴とする請求項1に記載のグレムリン−1に対する抗体。
- 前記抗体が、免疫抗体、キメラ抗体、ヒト抗体及びヒト化抗体よりなる群から選ばれるものであることを特徴とする請求項1に記載のグレムリン−1に対する抗体。
- 前記抗体が、軽鎖不変領域、重鎖不変領域、軽鎖可変領域及び重鎖可変領域を含み、前記軽鎖可変領域及び重鎖可変領域がそれぞれ下記のアミノ酸配列を有する群から選ばれることを特徴とする請求項1に記載のグレムリン−1に対する抗体:
(1)配列番号1のアミノ酸配列を有する軽鎖可変領域及び配列番号2のアミノ酸配列を有する重鎖可変領域;
(2)配列番号3のアミノ酸配列を有する軽鎖可変領域及び配列番号4のアミノ酸配列を有する重鎖可変領域;
(3)配列番号5のアミノ酸配列を有する軽鎖可変領域及び配列番号6のアミノ酸配列を有する重鎖可変領域;
(4)配列番号7のアミノ酸配列を有する軽鎖可変領域及び配列番号8のアミノ酸配列を有する重鎖可変領域;
(5)配列番号9のアミノ酸配列を有する軽鎖可変領域及び配列番号10のアミノ酸配列を有する重鎖可変領域;
(6)配列番号11のアミノ酸配列を有する軽鎖可変領域及び配列番号12のアミノ酸配列を有する重鎖可変領域;及び
(7)配列番号13のアミノ酸配列を有する軽鎖可変領域及び配列番号14のアミノ酸配列を有する重鎖可変領域。 - 前記抗体の軽鎖不変領域及び重鎖不変領域が、ヒトまたはウサギのものであることを特徴とする請求項4に記載のグレムリン−1に対する抗体。
- 前記抗体が、グレムリン−1が癌細胞に直接的に結合することを抑制することを特徴とする請求項1に記載のグレムリン−1に対する抗体。
- 前記抗体が、グレムリン−1に依存する細胞移動性(cell migration)、細胞浸潤性(cell invasion)及び細胞増殖(cell proliferation)を抑制することを特徴とする請求項1に記載の抗体。
- 前記抗体が、グレムリン−1に依存するE−カドヘリン(E-cadherin)の発現を阻害することを特徴とする請求項1に記載の抗体。
- 請求項1に記載の抗体及び薬学的に許容可能な担体を含む癌または免疫疾患の予防または治療用薬学的組成物。
- 前記癌が、前立腺癌、膵臓癌、肺癌、非小細胞肺癌、胃癌、肝癌、腎臓癌、卵巣癌、大腸癌、直腸癌、乳房癌、甲状腺癌、皮膚癌、骨癌、基底細胞癌、扁平細胞癌、鼻咽頭癌、膀胱癌、子宮癌、皮膚癌、食道癌及び頭頸部癌よりなる群から選ばれることを特徴とする請求項9に記載の薬学的組成物。
- 前記免疫疾患が、関節リウマチ(rheumatoid arthritis)、全身性進行性硬化症(Progressive systemic sclerosis、Scleroderma)、全身性紅斑性狼瘡(lupus)、アトピー皮膚炎、円形脱毛症(alopecia areata)、乾癬、天疱瘡、喘息、アフタ性潰瘍、慢性甲状腺炎、後天性再生不良貧血、原発性胆汁性肝硬変、潰瘍性大腸炎、ベーチェット病(Behcet’s disease)、クローン病、硅素肺症、石綿肺症、IgA腎臓疾患、連鎖球菌感染後糸球体腎炎(PSGN)、シェーグレン症候群(Sjogren Syndrome)、ギラン・バレー症候群(Guilian−Barre syndrome)、皮膚筋炎(dermatomyositis)、多発性筋炎(polymyositis)、多発性硬化症(multiple sclerosis)、自己免疫性溶血性貧血 (Autoimmune hemolytic anemia)、自己免疫性脳脊髄炎、重症筋無力症(Myasthenia gravis)、グレーブス病(Grave’s disease)、結節性多発動脈炎(Polyarteritis nodosa)、強直性脊椎炎(Ankylosing spondylitis)、線維筋痛症(Fibromyalgia syndrome)及び側頭動脈炎(Temporal arteritis)よりなる群から選ばれることを特徴とする請求項9に記載の薬学的組成物。
- 請求項1の抗体または請求項9の薬学的組成物をこれを必要とする対象に投与することを含むことを特徴とする癌または免疫疾患の治療方法。
- 前記抗体が、骨形成タンパク質(BMP)または血管内皮成長因子受容体−2(VEGFR2)に非依存的な方式によりグレムリン−1を抑制することにより、癌を治療することを特徴とする請求項12に記載の癌または免疫疾患の治療方法。
- 前記癌が、前立腺癌、膵臓癌、肺癌、非小細胞肺癌、胃癌、肝癌、腎臓癌、卵巣癌、大腸癌、直腸癌、乳房癌、甲状腺癌、皮膚癌、骨癌、基底細胞癌、扁平細胞癌、鼻咽頭癌、膀胱癌、子宮癌、皮膚癌、食道癌及び頭頸部癌よりなる群から選ばれることを特徴とする請求項12に記載の癌または免疫疾患の治療方法。
