Abstract
Description
(a)N末端シグナルペプチド有りもしくは無しの配列番号:1のアミノ酸配列を含むかもしくはそれからなる(すなわち配列番号:21において示されるような成熟ペプチド配列を含み得るか又はそれからなり得る)、又は
(b)N末端シグナルペプチド有り又は無しの配列番号:1のアミノ酸配列(配列番号:21において示されるような)と比べて、1つ又は複数のアミノ酸の置換、修飾、欠失又は挿入を有する誘導体であり、それはグレムリン−1の活性を保持する(配列番号:20のアミノ酸配列)、
(c)そのバリアントであり、かかるバリアントが、典型的には配列番号:1(又は配列番号:20もしくは21)に対して、少なくとも約60%、70%、80%、90%、91%、92%、93%、94%又は95%の同一性(又は場合によっては約96%、97%、98%又は99%の同一性)を保持する、
グレムリン−1ポリペプチドも指し得る。換言すれば、かかるバリアントは、配列番号:1に対して約60%〜約99%の同一性、好適には配列番号:1に対して80%〜約99%の同一性、より好適には配列番号:1に対して90%〜約99%の同一性、及び最も好適には配列番号:1に対して約95%〜約99%の同一性を保持し得る。バリアントはさらに以下で記載される。

本発明の抗体、又はスクリーニング方法によって同定されるグレムリン−1を調節する薬剤は、医薬組成物において提供され得る。医薬組成物は通常は滅菌され、典型的には薬学的に許容される担体及び/又はアジュバントを含むだろう。本発明の医薬組成物は、薬学的に許容されるアジュバント及び/又は担体を追加で含み得る。
本発明の抗体、又は本発明のスクリーニング方法によって同定される調節薬剤は、グレムリン−1活性と関連する任意の病態の治療、予防又は寛解において使用され得る。例えば、グレムリン−1を介するシグナリングから全体又は部分的にもたらされる任意の病態である。換言すれば、本発明は、グレムリンによって媒介されるか又は影響を受ける病態の治療、予防又は寛解に関する。かかる病態としては、腎線維症(例えば糖尿病性腎症及び慢性移植腎症)及び特発性肺線維症を包含する線維症、肺動脈性肺高血圧症、血管新生並びにがん(例えば結腸直腸がん)が挙げられる。
タンパク質発現及び封入体調製
E.coli中での発現について至適化された短縮ヒトグレムリン−1コーディング配列(配列番号:20)を、BamHI/XhoIを使用して修飾pET32aベクター(Merck Millipore)の中へクローン化し、N末端6His−TEVタグ(pET−hグレムリン1)を備えたグレムリン配列をコードするベクターを生成した。
配列番号:2(イタリック体で示された6His−TEVタグの非グレムリン残基が存在する)。配列ナンバリングはUniProt O60565及び配列番号:1に基づいていた。
封入体を、変性バッファー(8Mの尿素、100mMのトリス、1mMのEDTA、10mMのNa2S4O6及び100mMのNa2SO3(pH8.5))中で再懸濁し、室温で16時間撹拌し、15分間21,000gの遠心分離によって清澄化した。
可溶化した封入体を、8Mの尿素、50mMのMES、200mMのNaCl、1mMのEDTA(pH6.0)中で平衡化したセファクリルS−200 26/60カラム(120mL)の上へロードした。グレムリン−1タンパク質を含有する画分を6Mの尿素、20mMのMES(pH6.0)により希釈し、HiTrap SP HP陽イオン交換カラムの上へロードし、1MのNaCl勾配により10カラム体積(10CV)にわたって溶出させた。精製し変性させたhグレムリン−1タンパク質を含有する画分をプールした。
変性した精製グレムリン−1タンパク質を、再折りたたみバッファー(50mMのトリス(pH8.5)、150mMのNaCl、5mMのGSH及び5mMのGSSG、0.5mMのシステイン、5mMのEDTA、0.5Mのアルギニン)へ、0.1mg/mlの最終濃度まで滴加し、定常的な撹拌により4℃で5日間インキュベーションした。5日後に、グレムリン−1タンパク質を20mMのHEPES、100mMのNaCl(pH7.5)に対して透析した。
グレムリン1タンパク質結晶を、6.6mg/mlのグレムリン1の溶液、及びpH4の0.1Mのクエン酸、1Mの塩化リチウム及び27%のポリエチレングリコール(PEG)6000を、1:1比で混合することによって、ハンギングドロップ法を使用して成長させた。データ収集の前に、結晶を、結晶化バッファーへの20%のグリセロールの添加によって凍結保護した。回析データをDiamond Light Sourceで収集し、XDSを使用してプロセシングした(Kabsch,Wolfgang(2010)Acta Crystallographica Section D 66,125−132)。回析データ統計を以下の表中で要約する。

背景
Hek Id1レポーター遺伝子アッセイは、クローン12 Hek293−Id1レポーター細胞を使用する。この細胞株を、Id1転写因子により安定的にトランスフェクションした。Id1はBMPシグナリング経路における転写因子である。グレムリンはBMPを結合し、BMPの受容体への結合を阻止し、レポーター遺伝子からのルシフェラーゼシグナルを低減することが公知である。したがって、このレポーターアッセイを使用して、抗グレムリン抗体をスクリーニングし、BMPとグレムリンの相互作用をブロックする任意のものがあるかどうかを調べることは可能である。この相互作用のブロッキングがあるならば、ルシフェラーゼシグナルの回復はこれらの細胞において観察される。
クローン12細胞を、10%のFCS、1×L−グルタミン及び1×NEAAを含有するDMEM中で培養した。細胞をハイグロマイシンB(200μg/ml)の存在下においても増殖させて、細胞がId1遺伝子発現を損失していないことを保証する。細胞を、0.5%のFCS、1×L−グルタミン及び1×NEAAを含有するDMEM中でアッセイした。ハイグロマイシンBは、細胞がアッセイ中である短い時間については必要とされない。
グレムリン−1の全長形態及び短縮形態をHek−Id1レポーター遺伝子アッセイにおいて試験して、BMP4/7に対するブロッキング活性を確認する。全長タンパク質についての結果を図6A中で示し、短縮タンパク質についての結果を図6B中で示す。

グレムリン1は、Hek−Id1レポーター遺伝子アッセイにおいてBMP 4/7シグナリングを阻害することができた。
抗グレムリン−1抗体は、免疫及びライブラリーパニングに由来した。ライブラリーを、献血から増幅されたV領域によるナイーブヒトライブラリーとしてインハウスで生成した。
抗体を、例3において記載されるHek−Id1レポーター遺伝子アッセイを使用して、及びSMADリン酸化の測定によって、スクリーニングした。SMAD1、5及び8は、BMPシグナリングに際してリン酸化される。したがって、グレムリン−1の阻害物質はSMADリン酸化を増加させる。
Hek−Id1レポーター遺伝子アッセイにおいて、免疫由来抗体との明らかなヒットはなかった(10倍過剰量の抗体をBMP4/7ヘテロダイマーに対して試験した)。結果を図7中で示す。

Ab7326 Fabによる複合体におけるヒトグレムリン−1の結晶構造を、2.1Åの分解能で解像した。Fab配列を以下に示す。
方法
ヒトグレムリン1についての抗グレムリンmIgGの親和性を、Biacore T200 system(GE Healthcare Bio−Sciences AB)上で表面プラズモン共鳴(SPR)技術を使用して、bia分子(biamolecular)相互作用分析によって決定した。抗グレムリンmIgGを固定化抗マウスFc表面によって捕捉し、グレムリン1を捕捉mIgGに対してタイトレーションした。捕捉リガンド(ヤギ抗マウスIgGのaffinipure F(ab’)2断片、Fc断片特異的、115−006−071、Jackson ImmunoResearch Inc.)を、約1600反応単位(RU)のレベルへ、600秒の活性化注入及び非活性化注入を使用して、アミンカップリング化学経由でCM4 Sensor Chipのフローセル2上に10mMのNaAc(pH5.0)中で50μg/mlで固定化した。HBS−EP+バッファー(0.01MのHEPES(pH7.4)、0.15MのNaCl、3mMのEDTA、0.05%のサーファクタントP20)を、10μL/分の最小流速で、ランニングバッファーとして使用した。参照表面を、捕捉リガンドを省いて、フローセル2のためのような表面の活性化及び非活性化によって、フローセル1上で調製した。
結合親和性(5つの決定についての平均KD値として取得した)は、100pM未満であることが見出された。
要約
TGFβスーパーファミリーにおける不均衡は、肺動脈性肺高血圧症(PAH)を包含する多くの肺病理学に強く関与している(Budd & Holmes Pharm Ther 2012)。グレムリン−1はPAHの発症及び進行に関与している(Thomas et al AJP 2009;Ciuclan et al AJP 2013)。最近の研究により、抗グレムリン−1抗体はPAHの前臨床低酸素状態/SU5416モデルにおける肺動脈性肺高血圧症の発症を阻害できることが実証された(Ciuclan et al AJP 2013)。本明細書において、PAHの前臨床的な低酸素状態/SU5416マウスモデルにおける血行動態及び血管リモデリングに対する抗グレムリン1抗体の効果を査定した。
肺高血圧症(PH)は、肺動脈において測定される圧が上昇する血行動態的状態である。臨床的には、これは、>25mmHgの平均安静時肺動脈圧(mPAP)、≧3のWood単位の肺血管抵抗(PVR)、及び≦15mmHgの肺動脈楔入圧によって定義される(Badesch et al 2009)。PHの病理学的特徴は多面であり、肺動脈圧、小動脈から中血管への血管リモデリング、右心室肥大、及び最終的な右心不全が挙げられる(Faber & Loscalzo 2004)。PHは、5つの主要なカテゴリー(第I群を包含する)へとWHOによって分類される。第I群は、特発性PAH及び遺伝性PAHに加えて、他の合併症(全身性硬化症等)と同時に生じる関連PAHを包含する、肺動脈性肺高血圧症を提示する(Simonneau et al 2009)。多くの素因となる突然変異が遺伝性PAH患者及び特発性PAH患者におけるPAHの発症へリンクしており、最も主なものは骨形成タンパク質受容体(BMPR2)への突然変異である(Budd & Holmes Pharm Ther 2012)。加えて、BMPにより活性化される下流のシグナル伝達構成要素のSmadsにおける突然変異もPAHを発症する患者において報告されている(Budd & Holmes Pharm Ther 2012)。より最近では、PAHのある患者において、BMP阻害物質(グレムリン(グレムリン−1及び2))のレベルが促進されることが、研究により同定された(Cahill et al Circ 2012)。PAHの発症におけるグレムリン−1についての機能的役割と合致して、グレムリン−1のハプロ欠損は、低酸素のマウス肺におけるBMPシグナリングを増大させ、血管リモデリングの減弱によって肺血管抵抗の低減を導く(Cahill et al Circ 2012)。これらの観察は、抗グレムリン−1抗体がマウスにおける慢性低酸素状態/SU5416誘導性の肺動脈性肺高血圧症を寛解することを実証したCiuclan及び共同研究者(AJP 2013)によって、さらに支持された。最近の研究は、非遺伝性の機構が全身性硬化症患者におけるBMPR2発現の低減に寄与し、PAHの発症に寄与し得ることを示唆する(Gilbane et al AJRCCM)。まとめると、BMPスーパーファミリー軸における不均衡は肺病理学(PAH)の発症を導き得る。

動物
C57Bl/6マウスを特定病原体除去施設中で収容し、食物及び水へ自由裁量でアクセスさせ、12時間の明/暗サイクルへ曝露した。本動物研究は、UK Home Office Animals(Scientific Procedures)Act 1986下で許可された。
8〜10週齢のC57Bl/6メスマウスを群へ割り付けた(表6)。すべての群を計量し、Ciuclan et al AJRCCM 2013中で記載されるように、100μlのベヒクル(脱イオン水中の0.5%のカルボキシメチルセルロース(CMC);0.9%の塩化ナトリウム;0.4%のポリソルベート80;0.9%のベンジルアルコール(NaCl))中の20mg/kgのSU5416を、皮下(s.c.)投与した。必要に応じて、表6中で略述されるように、PBS、30mg/kgのIgG1、又は30mg/kgの抗グレムリン−1のいずれかを含有する第2の皮下注入を投与した。一方で、低酸素状態+イマチニブ群におけるマウスにイマチニブを注入した餌を与えて、100mg/kg/日を送達した。次いで低酸素状態マウス群を常圧低酸素状態(10%のO2)チャンバーの中へ置く一方で、酸素正常状態群におけるマウスをチャンバーと同じ部屋中の酸素正常状態(21%のO2)条件で収容した。

RVSP及びMABPの血行動態測定値を、3週間の低酸素状態曝露及び表6中で略述されるような関連する薬物処理後に動物から得た。動物を1.5%のイソフルオランにより麻酔し、サーモスタットで37℃に制御した加熱用毛布の上へ仰臥位で置いた。第一に、右頸動脈を単離し、プレッシャーカテーテル(1.4Fの直径を備えたMillar mouse SPR−671NRプレッシャーカテーテル(Millar Instruments、英国))を右心室の中へ導入及び前進させて、RVSPを決定した。第二に、MABPを左総頚動脈の単離によって測定し、プレッシャーカテーテルを導入した。RVSP及びMABPの両方を、前較正したPowerLab system(ADInstruments(オーストラリア))で記録した。
スライドを脱ワックスし、キシレン及びエタノールの濃度勾配を使用して再水和した。スライドをメタノール中の0.3%のH2O2中に30分間浸して抗原を取り戻し、PBS中で3回洗浄し、1:30のPBS中の正常なウマ血清中で1時間ブロッキングした。1:100の濃度で抗αSMA一次抗体を各々のスライドへ添加し、4℃で一晩インキュベーションし、次いで5分間PBS中で3回リンスした。ビオチン化ウマ抗マウスIgG抗体(ラット吸着二次抗体)を1:200で希釈し、各々のスライドの上へピペットで移し、45分間インキュベーションし、次いで5分間PBS中で3回洗浄した。製造者の指示に従って、アビジンビオチン複合体アルカリフォスフォターゼ(phosphotase)(ABC−AP)を30分前に調製し、各々のスライド上に30分間置き、次いで5分間3回洗浄した。AP基質をキットの指示に従って調製し、各々のスライドの上へピペットで移し、放置して現像させ、次いで5分間3回洗浄した。1:100の濃度で抗vWF一次抗体を各々のスライドへ添加し、4℃で一晩インキュベーションし、次いで5分間PBS中で3回リンスした。ビオチン化ヤギ抗ウサギIgG二次抗体を1:200で希釈し、各々のスライド上に45分間放置し、次いで5分間PBS中で3回洗浄した。キットの指示に従って、ABCを30分前に調製し、各々のスライド上に30分間置き、次いで5分間3回洗浄した。DAB基質をキットの指示に従って調製し、各々のスライドの上へピペットで移し、放置して約5〜10分現像させ、次いで5分間3回洗浄した。スライドをヘマトキシリンにより約40秒対比染色し、脱水し、pertexを使用してカバーガラスによりマウントした。すべてのスライドを、Hamamatsu NanoZoomer 2.0−HT Slide Scanner(Hamamatsu、Welwyn Garden City、英国)によりデジタルスキャンした。
各々の低酸素状態群からランダムで取得した40を超える血管を、筋性化の程度について査定した。少なくとも4人の独立した観察者によって、血管をスコアリングした。0=非筋性化;1=部分筋性化;2=完全筋性化とし;各々の血管についての様式の値を決定した。完全筋性化、部分筋性化、又は非筋性化の血管のパーセンテージを決定し、平均±SEMをプロットした。一元配置ANOVAを遂行して、有意性*P<0.05;**P<0.01;***P<0.005;****P<0.001を決定する。
SU5416(20mg/kg)の投与、続いて慢性的な常圧低酸素状態(10%のO2)への曝露は、PBS単独群又はIgG1対照群の両方において、21日間の酸素正常状態/SU5416単独に比較して、RVSPの有意な(P<0.01)増加を導いた(図12)。有意差は、IgG1処理群とPBS処理群との間に観察されなかった。低酸素状態で維持されたSU5416を投与したマウスにおけるRVSPの上昇に対する薬物治療(イマチニブ)の効果は、対照群に比較して、RVSPの有意な(P<0.001)低減を導いた。抗グレムリン1は、IgG1対照群及びPBS対照群に比較して、RVSPの有意な(P<0.005)低減を導いた。抗グレムリン1の有意な効果は、酸素正常状態で維持された動物におけるRVSPに対して観察されなかった(図12)。
本明細書において、慢性低酸素状態/SU5416モデルにおけるPAHの発症に対する抗グレムリン−1抗体の効果を査定した。抗グレムリン−1抗体はRVSP(図12)及び血管リモデリング(図15)を有意に阻害したが、PAHの低酸素状態/SU5416のマウスモデルにおける体循環(MABP)圧(図13)に対する効果を示さなかった。酸素正常状態/SU5416処理マウスにおけるRVSPに対する、抗グレムリン−1抗体による有意な効果は、観察されなかった。抗グレムリン1の抗体の効果はCiuclan及び共同研究者(AJP 2013)によって行われた観察と合致し、それにおいて、抗グレムリン−1抗体によるアンタゴニズムは、PAHの慢性低酸素状態/SU5416マウスモデルにおいて、RVSPの上昇及び血管リモデリングを寛解することが実証された(図12及び15)。Ciuclan及び共同研究者とは対照的に、本発明者は、本研究において抗グレムリン−1抗体による右心肥大に対する有意な効果を認めなかった(図14)。しかしながら本研究において、本発明者は、低酸素状態/SU5416マウスにおける右心肥大の発症に対して、対照薬物イマチニブによる有意な効果を認めなかった。まとめると、本研究は、PAHの慢性低酸素状態/SU5416モデルにおける肺血管リモデリングの発症及びRVSPの上昇におけるグレムリン−1の役割並びにその治療における抗グレムリン−1抗体の使用を支持する。

- 抗体との複合体における、ヒトグレムリン−1の結晶。
- 前記抗体が、配列番号:18の重鎖及び配列番号:19の軽鎖を含む、請求項32に記載の結晶。
- 表1中の座標によって定義された、ヒトグレムリン−1の構造。
- 表1中のグレムリン−1の構造座標又は構造のホモログを定義する座標によって定義された、機械読み取り可能データによりコードされたデータストレージ材料を含む、機械読み取り可能データストレージ媒体。

- 請求項38、39又は40のいずれか一項に記載の使用によって同定される、薬剤。
- (a)表1中の構造座標からリガンド結合部位を同定するステップと;
(b)前記リガンド結合部位の少なくとも一部と相互作用する候補薬剤を同定するステップと;
(c)前記薬剤を得るか又は合成するステップと、
を含む、グレムリン−1活性の調節薬剤についてのスクリーニング方法。 - ステップ(a)及び(b)がインシリコで遂行される、請求項42に記載の方法。
- 前記薬剤がグレムリン−1活性の阻害物質である、請求項42又は43に記載の方法。
- 前記方法が、グレムリン−1の以下の残基:
Trp93;
Phe117;
Tyr119;
Phe125;
Tyr126、及び/又は
Phe138、
(ここで、残基ナンバリングは配列番号:1に基づく)

請求項42、43又は44のいずれか一項に記載の方法。 - 前記薬剤が、グレムリン−1のすべての以下の残基:Trp93、Phe117、Tyr119、Phe125、Tyr126及びPhe138と相互作用する、請求項45に記載の方法。
- 候補薬剤がX線結晶学によって決定される残基の6Å内にあるならば、前記薬剤がグレムリン−1と相互作用する、請求項45又は46に記載の方法。
- 前記リガンド結合部位がアロステリック部位である、請求項42、43又は44のいずれか一項に記載の方法。
- 前記方法が、グレムリン−1の以下の残基:Ile131、Lys147、Lys148、Phe149、Thr150、Thr151、Arg169、Lys174及びGln175(ここで、残基ナンバリングは配列番号:1に基づく)のうちの少なくとも1つと相互作用する候補薬剤を同定することを含む、請求項48に記載の方法。
- 前記薬剤が、グレムリン−1のすべての以下の残基:Ile131、Lys147、Lys148、Phe149、Thr150、Thr151、Arg169、Lys174及びGln175と相互作用する、請求項49に記載の方法。
- Lys147、Lys148、Phe149、Thr150、Thr151、Arg169、Lys174及びGln175は同じグレムリン−1モノマー上に所在し、Ile131は第2のグレムリン−1モノマー上に所在する、請求項49又は50に記載の方法。
- 候補薬剤がX線結晶学によって決定される残基の4Å内にあるならば、前記候補薬剤がグレムリン−1と相互作用する、請求項48〜51のいずれか一項に記載の方法。
- 前記薬剤が小分子又は抗体である、請求項42〜52のいずれか一項に記載の方法。
- 請求項42〜53のいずれか一項に記載の方法によって同定される、グレムリン−1調節薬剤。
