Abstract
Description
従来技術1)ラセミN-(ベンジルオキシカルボニル)-2-メチルプロリンとキニンから塩を形成させてアセトン中から晶析することにより、N-ベンジルオキシカルボニル-2-メチル-L-プロリンのキニン塩を取得する。続いて、アセトン/ジエチルエーテル混合溶媒から再結晶を6回繰り返し、得られるキニン塩をアンモニア水で解塩することにより、N-(ベンジルオキシカルボニル)-2-メチル-L-プロリンを取得する方法(非特許文献1)。
従来技術2)L-アラニンベンジルエステルと1-ブロモ-3-クロロプロパンを反応させて、更にボック化することにより、N-(3-クロロプロピル)-N-(tert-ブトキシカルボニル)-L-アラニンベンジルエステルを取得する。続いて、これを、ジメチルホルムアミド中、リチウムヘキサメチルジシラザンで処理して環化反応を行い、パラジウム触媒存在下に加水素分解することにより、N-(tert-ブトキシカルボニル)-2-メチル-D-プロリンを製造する方法(特許文献1)。

(c)化合物(1)と前記化合物(2)を前記溶媒中で混合後、濃縮する事により固体を析出させる方法。
(d)前記化合物(1)と前記化合物(2)を前記溶媒中で混合後、更に貧溶媒を添加する事により固体を析出させる方法。
(e)前記化合物(1)と前記化合物(2)を前記溶媒中で混合後、更に貧溶媒に濃縮置換する事により固体を析出させる方法。
なお前記解塩は、塩基性水溶液と化合物(3)とを接触させる事でも可能である。
カラム:ダイセル化学社製CHIRALCEL OJ-RH 150×4.6mm、移動相:アセトニトリル/リン酸水溶液(pH2.5)=2/8(容量比)、流速:0.7ml/min、検出器:UV210nm、カラム温度:30℃
N-(tert-ブトキシカルボニル)-2-メチル-D-プロリンの保持時間:7.1分
N-(tert-ブトキシカルボニル)-2-メチル-L-プロリンの保持時間:9.8分
なお実施例で得られる化合物(3)(塩)を上記分析法で分析すると、分析中に解塩し、N-(tert-ブトキシカルボニル)-2-メチル-プロリンとしての光学純度を知ることができる。

Claims (7)
- R1がtert-ブトキシカルボニル基である、請求項1に記載の製造法。
- R2がメチル基、又はエチル基であり、Arがフェニル基、又はp-クロロフェニル基である、請求項1又は2に記載の製造法。
- 請求項1~3のいずれかに記載の方法により製造した前記化合物(3)を解塩することを特徴とする、光学純度が向上した前記化合物(1)の製造法。
- R1がtert-ブトキシカルボニル基であり、R2がメチル基、又はエチル基であり、Arがフェニル基、又はp-クロロフェニル基である、請求項5に記載の光学活性2-メチルプロリン誘導体。
- 前記化合物(3)がN-(tert-ブトキシカルボニル)-2-メチル-L-プロリン/(S)-1-フェネチルアミン塩、N-(tert-ブトキシカルボニル)-2-メチル-D-プロリン/(R)-1-フェネチルアミン塩、N-(tert-ブトキシカルボニル)-2-メチル-L-プロリン/(S)-1-(p-クロロフェニル)エチルアミン塩、N-(tert-ブトキシカルボニル)-2-メチル-D-プロリン/(R)-1-(p-クロロフェニル)エチルアミン塩、N-(tert-ブトキシカルボニル)-2-メチル-L-プロリン/(S)-1-フェニルプロピルアミン塩、又はN-(tert-ブトキシカルボニル)-2-メチル-D-プロリン/(R)-1-フェニルプロピルアミン塩である、請求項5に記載の光学活性2-メチルプロリン誘導体。
