本発明の第1実施例であるイオントラップ質量分析装置と該装置により実施される特徴的な質量分析方法について、添付図面を参照して説明する。
続いて、スペクトル比較処理部53は、上記のようなニュートラルロスの解析により抽出されたピーク群に含まれる複数のピークの強度をそれぞれ求める。また、もう一方のマススペクトル上で、上記ピーク群に含まれる各ピークと同じ質量電荷比に現れるピークを抽出して、そのピークの強度もそれぞれ求める。
図8(a)中のピーク(1)は、目的化合物がイオン化して得られるプロトン付加イオンピーク[M+H]+である。ヘリウムによる弱い脱離促進作用によって、一部の糖は分析中に脱離する。図8(a)中の主要イオンピーク群(2)~(5)は、目的化合物からの糖の脱離により生じたものであり、それぞれ [M+H]+-Sia、[M+H]+-2Sia、[M+H]+-3Sia、及びN-結合型糖ペプチドに特徴的な、糖が全て脱離したペプチドイオン、HexNAc環開裂付加ペプチドイオン、及びHexNAc付加ペプチドイオンで構成されるトリプレットピークである。当然、目的化合物が不明である状況では、これらのピークの帰属は不明である。
次に、本発明の第2実施例であるイオントラップ質量分析装置と該装置により実施される特徴的な質量分析方法について、添付図面を参照して説明する。この第2実施例のイオントラップ質量分析装置は、特に、遊離性の高い糖が付加した糖ペプチド(具体的にはN-結合型糖ペプチド)の構造解析を目的としたものであり、第1実施例とは本発明におけるイオン非選択フラグメンテーション促進手段に相当する手段が相違する。また、この第2実施例のイオントラップ質量分析装置では、単にマススペクトル上のピークの帰属を決定するのみならず、そのピーク帰属結果を利用してプリカーサイオンを決定し、さらにそのプリカーサイオンについてのMS2分析(擬似MS3分析)を実行して得られたマススペクトルを用いて糖ペプチドを構成するペプチドの同定及び糖鎖構造解析も行っている。

- 分析対象である化合物由来のイオンを質量分析する質量分析器と、該質量分析器により質量電荷比に応じて分離されたイオンを検出する検出器と、を具備する質量分析装置において、
a)目的化合物由来のイオンに対し、イオン選択操作を伴わずにフラグメンテーションを促進するイオン非選択フラグメンテーション促進手段と、
b)前記目的化合物由来のイオンについて前記イオン非選択フラグメンテーション促進手段によるフラグメンテーション促進操作を行わずに前記質量分析器により質量分析を実行して第1のマススペクトルを取得する第1分析実行手段と、
c)前記目的化合物由来のイオンについて前記イオン非選択フラグメンテーション促進手段によるフラグメンテーション促進操作を行った後に前記質量分析器により質量分析を実行して第2のマススペクトルを取得する第2分析実行手段と、
d)前記第1のマススペクトルと前記第2のマススペクトルとを比較することにより、前記目的化合物由来のピークの帰属を判断するデータ処理手段と、

a)前記目的化合物由来のイオンに対し前記質量分析器により質量分析を実行して第1のマススペクトルを取得する第1分析実行ステップと、
b)前記目的化合物由来のイオンに対し、イオン選択操作を伴わずにフラグメンテーションを促進するイオン非選択フラグメンテーション促進操作を行った後に前記質量分析器により質量分析を実行して第2のマススペクトルを取得する第2分析実行ステップと、
c)前記第1のマススペクトルと前記第2のマススペクトルとを比較することにより、前記目的化合物由来のピークの帰属を判断するデータ処理ステップと、
を有することを特徴とする質量分析方法。
