Description
また、このような処理でも除去の困難なアレルゲン性蛋白を除去する方法として、天然ゴムラテックスにアルカリプロテアーゼを添加して蛋白分解処理を施した後に、エキソペプチダーゼ活性を有するプロテアーゼを添加して蛋白分解処理を施し、蛋白質及びその分解物を除去処理する方法が提案されている(特許文献2参照)。
特許文献4に記載の方法によれば、ゴムの樹から採取後7日以内の新鮮な天然ゴムラテックス(Fresh NRラテックス)に尿素系化合物からなる蛋白質変成剤を添加して蛋白質除去処理を行うことにより、その窒素含有率を0.004%とすることができる。
したがって、天然ゴム中に含まれるアレルゲン性蛋白質をさらに除去した天然ゴム及びそのラテックス、並びにそれらの効率的な製造方法が望まれていた。
天然ゴムラテックスの試料中に存在する全ての蛋白質およびその分解物(以下、「全蛋白質」という。)中には、ヒト血清中に抗体を産生し得る「抗原蛋白質」の群が含まれる。また、ヒト血清中に産生する抗体は、アレルギー反応を誘発し得るIgEクラスの抗体と、アレルギー反応を誘発しないIgEクラスの抗体とに分類される。ここで、「抗原蛋白質」の中で、アレルギー反応の原因となり得るIgEクラスの抗体を産生する抗原蛋白質を、他の抗原蛋白質と区別するために「アレルゲン性蛋白質」と定義する。
1.天然ゴムラテックスに尿素系化合物、界面活性剤および極性有機溶媒を添加し、当該ラテックス中の蛋白質を変性処理した後に除去することを特徴とする、天然ゴムラテックスを乾燥して得られる固形ゴム中の蛋白質の含有量が、RRIM試験法により測定した窒素含有率において0.001%以下のレベルである蛋白質フリー天然ゴムラテックスの製造方法。
2.前記極性有機溶媒として、炭素数1~5の低級脂肪族アルコール、炭素数3~4のケトン、炭素数1~5のカルボン酸、炭素数1~5のカルボン酸のエステルから選択された1種又は2種以上の混合物を、天然ゴムラテックスのゴム分に対して0.001~30重量%用いることを特徴とする、1.に記載の蛋白質フリー天然ゴムラテックスの製造方法。
3.前記変性処理した蛋白質の除去を遠心分離処理により行うことを特徴とする、1.又は2.に記載の蛋白質フリー天然ゴムラテックスの製造方法。
4.1.~3.のいずれかに記載された製造方法で得られた蛋白質フリー天然ゴムラテックスに酸を添加して凝固することを特徴とする、蛋白質フリー天然ゴムの製造方法。
5.1.~3.のいずれかに記載された製造方法で得られた蛋白質フリー天然ゴムラテックスを乾燥することを特徴とする、蛋白質フリー天然ゴムの製造方法。
6.天然ゴムラテックスを乾燥して得られる固形ゴム中の蛋白質の含有量が、RRIM試験法により測定した窒素含有率において0.001%以下のレベルであり、改良ローリー法により測定した天然ゴムラテックスを乾燥して得られる固形ゴム中の蛋白質量が、0.5μg/g以下のレベルであることを特徴とする蛋白質フリー天然ゴムラテックス。
7.天然ゴム中の蛋白質の含有量が、RRIM試験法により測定した窒素含有率において0.001%以下のレベルであり、改良ローリー法により測定した天然ゴムラテックスを乾燥して得られる固形ゴム中の蛋白質量が、0.5μg/g以下のレベルであることを特徴とする蛋白質フリー天然ゴム。
(1)アレルギーを誘発するおそれがきわめて低い蛋白質フリー天然ゴムラテックスを短時間で効率良く得ることができる。
(2)蛋白質の除去に不向きな高アンモニア天然ゴムラテックスを原料として使用した場合でも、蛋白質フリー天然ゴムラテックスを効率良く製造することができる。
(3)本発明により得られる蛋白質フリー天然ゴムラテックスは、手袋等の家庭用品、手術用手袋、カテーテル等の医療用具、授乳用具、避妊具等に幅広く利用できるものである。したがって、安全性の高いこれらの製品の製造原料としてきわめて有用である。
(原料天然ゴムラテックス)
原料天然ゴムラテックスとしては、ゴムの樹から採取された後、濃縮処理が施されていない天然ゴムラテックス(フィールドラテックス)、ゴムの樹から採取後14日以内の新鮮な天然ゴムラテックス、市販の高アンモニア天然ゴムラテックスのいずれを使用してもよい。ここで、新鮮な天然ゴムラテックスとは防食処理されていないラテックスを意味する。このような天然ゴムラテックスとしては、好ましくはゴムの樹から採取後3箇月以内、特に好ましくは採取後7日以内、最も好ましくは採取後3日以内のラテックスを使用する。また、ラテックス中のゲル含有量が好ましくは40%以下、特に好ましくは10%以下のラテックスを使用する。
極性有機溶媒としては、水に混和するものが好ましい。好ましい極性有機溶媒としては、例えば、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、tert-ブタノール、1-ペンタノール、2-ペンタノール、3-ペンタノール等の炭素数が1~5の低級脂肪族アルコール;アセトン、メチルエチルケトン等の炭素数3~4のケトン;酢酸、プロピオン酸等の炭素数1~5のカルボン酸;酢酸エチル等の前記炭素数1~5のカルボン酸のエステル(炭素数1~5の低級アルキルエステルが好ましい)が挙げられる。これらの極性有機溶媒は単独で、又は2種以上を混合して使用することができる。
天然ゴムラテックスに対する極性有機溶媒の配合割合は、天然ゴムラテックスのゴム分に対して0.001~30重量%、特に0.01~10重量%、更に0.05~1重量%とすることが好ましい。極性有機溶媒の配合割合がゴム分に対して0.001重量%未満では、アレルゲン性蛋白質の変成処理効率を十分に改善することができない。一方、極性有機溶媒の配合割合がゴム分に対して30重量%を超えると、蛋白質変性処理工程でゴム分が凝固するといった問題が生じる。
蛋白質変性剤としては、尿素系化合物或いは尿素複塩を使用する。好ましい尿素系化合物としては、次の一般式(1)で表される尿素誘導体及び尿素複塩が用いられる。

天然ゴムラテックス中に含まれるアレルゲン性蛋白質の除去前処理は、前述の極性有機溶媒と変性剤及び界面活性剤を原料天然ゴムラテックスに添加し、約1分~5時間、好ましくは約1分~2時間、更に好ましくは約1分~1時間処理することによって行われる。蛋白質変性剤の添加量は、使用する変性剤の性質に応じて適宜選択することができる。通常、原料天然ゴムラテックスのゴム分に対して約0.001~10重量%の蛋白質変性剤を添加する。
極性有機溶媒および蛋白質変性剤で処理された天然ゴムラテックスは、さらに遠心分離等の手段により、ゴム分とアレルゲン性蛋白質を分離し、除去することにより精製され、工業用原料として使用可能な、アレルギーを誘発するおそれのない蛋白質フリー天然ゴムラテックスが得られる。
アレルゲン性蛋白質の除去処理を遠心分離により行う場合には、遠心分離処理の回数は1回以上行う。通常は、ゴム分の損失および歩留まりの低下に伴う不利益を被ることのない範囲で、遠心分離処理を2回以上行うことが好ましい。
蛋白質フリー天然ゴムラテックスを製造する際には、蛋白質の除去前処理を施す前にまたは前処理中に、安定化剤として界面活性剤をラテックス中に添加することが好ましい。とりわけ、原料となる高アンモニア天然ゴムラテックスのpHを中性領域に調整して蛋白質の除去処理を行う際には、ゴム分の凝固を防止するために界面活性剤の添加が望まれる。
(アニオン界面活性剤)
アニオン界面活性剤には、例えばカルボン酸系、スルホン酸系、硫酸エステル系、リン酸エステル系等が挙げられる。カルボン酸系のアニオン界面活性剤としては、例えば炭素数が6~30の脂肪酸塩、多価カルボン酸塩、ロジン酸塩、ダイマー酸塩、ポリマー酸塩、トール油脂肪酸塩などが挙げられる。中でも、炭素数10~20のカルボン酸塩が好適である。カルボン酸系のアニオン界面活性剤の炭素数が6を下回ると蛋白質および不純物の分散・乳化作用が不十分となるおそれがあり、炭素数が30を超えると水に分散させにくくなるおそれがある。
硫酸エステル系界面活性剤としては、例えばアルキル硫酸エステル塩、ポリオキシアルキレンアルキル硫酸エステル塩、ポリオキシアルキレンアルキルフェニルエーテル硫酸塩、トリスチレン化フェノール硫酸エステル塩、ポリオキシアルキレンジスチレン化フェノール硫酸エステル塩等が挙げられる。
ノニオン界面活性剤には、例えばポリオキシアルキレンエーテル系、ポリオキシアルキレンエステル系、多価アルコール脂肪酸エステル系、糖脂肪酸エステル系、アルキルポリグリコシド系等が挙げられる。
ポリオキシアルキレンエーテル系のノニオン界面活性剤としては、例えばポリオキシアルキレンアルキルエーテル、ポリオキシアルキレンアルキルフェニルエーテル、ポリオキシアルキレンポリオールアルキルエーテル、ポリオキシアルキレンスチレン化フェノールエーテル、ポリオキシアルキレンジスチレン化フェノールエーテル、ポリオキシアルキレントリスチレン化フェノールエーテル等が挙げられる。前記ポリオールとしては炭素数2~12の多価アルコールが挙げられ、例えばプロピレングリコール、グリセリン、ソルビトール、シュクロース、ペンタエリトリトール、ソルビタン等が挙げられる。
糖脂肪酸エステル系のノニオン界面活性剤としては、例えばショ糖、グルコース、マルトース、フラクトース、多糖類の脂肪酸エステル等が挙げられ、これらのポリアルキレンオキサイド付加物も使用可能である。
カチオン界面活性剤には、例えばアルキルアミン塩型、アルキルアミン誘導体型およびそれらの第4級化物、ならびにイミダゾリニウム塩型等が挙げられる。アルキルアミン塩型のカチオン界面活性剤としては、第1級アミン、第2級アミンおよび第3級アミンの塩が挙げられる。アルキルアミン誘導体型のカチオン界面活性剤は、エステル基、エーテル基、アミド基のうちの少なくとも1つを分子内に有するものであって、例えばポリオキシアルキレン(AO)アルキルアミンおよびその塩、アルキルエステルアミン(AO付加物を含む)およびその塩、アルキルエーテルアミン(AO付加物を含む)およびその塩、アルキルアミドアミン(AO付加物を含む)およびその塩、アルキルエステルアミドアミン(AO付加物を含む)およびその塩、アルキルエーテルアミドアミン(AO付加物を含む)およびその塩等が挙げられる。
アルキルアミン誘導体型カチオン界面活性剤のうち、酢酸塩の具体例としては、例えばココナットアミンアセテート、ステアリルアミンアセテート等が挙げられる。上記アルキルアミン塩型およびアルキルアミン誘導体型カチオン界面活性剤におけるアルキル基は特に限定されるものではないが、通常炭素数8~22の、直鎖状、分岐鎖状またはゲルベ状のものが挙げられる。
具体的には、ラウリルトリメチルアンモニウムハライド、セチルトリメチルアンモニウムハライド、ステアリルトリメチルアンモニウムハライド等のアルキルトリメチルアンモニウムハライド;ジステアリルジメチルアンモニウムハライド等のジアルキルジメチルアンモニウムハライド;トリアルキルメチルアンモニウムハライド;ジアルキルベンジルメチルアンモニウムハライド;アルキルベンジルジメチルアンモニウムハライド等が挙げられる。
本発明の脱アレルゲン化天然ゴムラテックスの製造方法においては、上記例示の各成分のほかにも、必要に応じて他の添加剤を配合することができる。かかる他の添加剤としては、例えばpH調整剤としての、リン酸第一カリウム、リン酸第二カリウム、リン酸ナトリウム等のリン酸塩;酢酸カリウム、酢酸ナトリウム等の酢酸塩;硫酸、酢酸、塩酸、硝酸、クエン酸、コハク酸等の酸類またはその塩;アンモニア、水酸化ナトリウム、水酸化カリウム、炭酸ナトリウム、炭酸水素ナトリウム等が挙げられる。
また、酵素としての、リパーゼ、エステラーゼ、アミラーゼ、ラッカーゼ、セルラーゼ等が挙げられる。さらに、分散剤としての、スチレンスルホン酸共重合物、ナフタレンスルホン酸ホルマリン縮合物、リグニンスルホン酸、多環式芳香族スルホン酸共重合物、アクリル酸および無水マレイン酸のホモポリマー/共重合物、イソブチレン-アクリル酸、イソブチレン-無水マレイン酸共重合物等が挙げられる。
本発明によれば、天然ゴムラテックスを乾燥して得られる固形ゴム中の蛋白質を、RRIM試験法により測定した窒素含有率として0.001%以下とし、改良ローリー法により測定した天然ゴムラテックスを乾燥して得られる固形ゴム中の蛋白質濃度として、0.5μg/g以下のレベルにすることができる。
(改良ローリー法)
改良ローリー法(ASTM D5712-99)は、蛋白質溶液をアルカリ性条件下でCu2+と反応させ、その反応物をFolin試薬で還元する2つのステップからなり、アレルゲン性蛋白質を含む全蛋白質の定量分析法としてよく用いられる方法である。
本発明では、バイオ・ラッド社の蛋白質定量キットを用いて、蛋白質の量を改良ローリー法により測定した。具体的には、試料に含まれる蛋白質をリン酸緩衝溶液を用いて抽出した。抽出した蛋白質を、Cu2+と反応させ、反応物をFolin試薬で還元し、750nmの吸光度を測定した。測定された吸光度から、ウシγグロブリンを標準物質として作製した検量線を用いて、ウシγグロブリン換算量として求めた。
また、本発明では、蛋白質フリー天然ゴムラテックスをキャスティングなどの方法によって乾燥させることにより、蛋白質フリー天然ゴムからなるフイルムや各種の成型品を製造することができる。
以下の例では、界面活性剤として、アニオン界面活性剤ラウリル硫酸ナトリウム(SLS:キシダ化学工業製)を使用した。
原料ラテックスとして、ゴールデンホープ社(マレーシア)製のゴム分濃度60.2重量%、アンモニア分0.7重量%の高アンモニア天然ゴムラテックスを使用し、これをゴム分の濃度が30重量%となるように水で希釈した。このラテックスのゴム分100重量部に対して、アニオン界面活性剤SLS3.3重量部およびエタノール0.083重量部を添加した。次いで、このラテックスのゴム分100重量部に対して変性剤として尿素0.3重量部を添加し、25℃で60分間攪拌することによって変性処理を行った。
変性処理を完了したラテックスについて10000rpmで30分間遠心分離処理を施した。こうして分離した上層のクリーム分を、1%SLS-0.025%エタノール水溶液にゴム分濃度が30%になるよう分散し、2回目の遠心分離処理を上記と同様にして行った。さらに、得られたクリーム分を1%SLS-0.025%エタノール水溶液に再分散し、3回目の遠心分離処理を上記と同様にして行った。得られたクリーム分をゴム分の濃度が30重量%となるように1%界面活性剤水溶液に再分散させることによって、蛋白質フリー天然ゴムラテックスを得た。
実施例1において、極性有機溶媒としてエタノールに代えて同量の2-プロパノールを使用したほかは、実施例1と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例1において、極性有機溶媒としてエタノールに代えて同量のアセトンを使用したほかは、実施例1と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例1において、界面活性剤としてSLSに代え同量のノニオン界面活性剤「レオドールTW-O120V」(花王社製:ポリオキシエチレンソルビタンモノオレート)を使用したほかは、実施例1と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例1において、極性有機溶媒としてエタノールに代えてアセトン8.3重量部を使用したほかは、実施例1と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例5において、原料ラテックスとしてゴムの樹から採取後1日の新鮮天然ゴムラテックスを使用したほかは、実施例5と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例1において、極性有機溶媒としてエタノールに代えて同量の酢酸エチルを使用したほかは、実施例1と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例5において、界面活性剤としてSLSに代えて同量のノニオン界面活性剤「マイドール10」(花王社製:アルキルグルコシド)を使用したほかは、実施例5と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例1において、極性有機溶媒としてエタノールに代えて0.83重量部の酢酸を使用したほかは、実施例1と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例1において、極性有機溶媒として0.083重量部のエタノールと0.083重量部の酢酸を使用したほかは、実施例1と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例1において、極性有機溶媒を使用しなかったほかは、実施例1と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例4において、極性有機溶媒を使用しなかったほかは、実施例4と同様にして蛋白質フリー天然ゴムラテックスを得た。
実施例6において、極性有機溶媒を使用しなかったほかは、実施例6と同様にして蛋白質フリー天然ゴムラテックスを得た。
天然ゴム中に含まれる蛋白質の量を表す指標として、上記実施例及び比較例において得られた蛋白質フリー天然ゴムラテックスをシャーレ上にキャストし、乾燥することにより固形ゴムを作製し、窒素含有率測定用のサンプルとした。
また、対照サンプルとして、実施例1で原料として使用した高アンモニア天然ゴムラテックス(対照例1)から、同様に直接キャストフイルムを作製した。実施例、比較例および対照例の各サンプルについて、その窒素含有率(N%)をRRIM試験法(Rubber
Reseach Institute of Malaysia(1973),’SMR Bulletin No.7’)によって、次の手順により測定した結果を表1に示す。RRIM試験法は、Kjeldahl法とも呼ばれている試験法で、蛋白質やアミノ酸等の窒素を定量する標準的な方法である。
固形状天然ゴムに、硫酸銅、硫酸カリウムおよびセレンからなる触媒と硫酸を添加し、1時間程度加熱した後、水酸化ナトリウム水溶液を加えて水蒸気蒸留を行った。留出した窒素分をホウ酸アンモニウムとして捕捉し、希硫酸で滴定することで窒素含有率を求めた。
また、実施例4においても、極性有機溶媒を使用しない比較例2に比べて天然ゴムラテックス中の窒素含有量は大幅に減少しており、極性有機溶媒の効果が明白である。
そして、アレルゲン蛋白質の量も検出限界以下に減少している。(実施例5,6及び比較例1,3参照)
実施例1~3、比較例1ならびに対照例1のゴムラテックスから生ゴムフィルムを作成し、赤外吸収スペクトルを測定した結果を図1に示す。
図1において、横軸は波数(cm-1)、縦軸は吸収強度を表す。また、Aは実施例1、Bは実施例2、Cは実施例3、Dは比較例1、そしてEは対照例1から得られたフィルムのスペクトルを表す。
また、短鎖のペプチド結合に由来する3320cm-1のピークも確認されず、赤外線吸収スペクトルで検出可能な蛋白質は、実質的に全て除去されていることが判明した。
Claims (7)
- 天然ゴムラテックスに尿素系化合物、界面活性剤および極性有機溶媒を添加し、当該ラテックス中の蛋白質を変性処理した後に除去することを特徴とする、天然ゴムラテックスを乾燥して得られる固形ゴム中の蛋白質の含有量が、RRIM試験法により測定した窒素含有率において0.001%以下のレベルである蛋白質フリー天然ゴムラテックスの製造方法。
- 前記極性有機溶媒として、炭素数1~5の低級脂肪族アルコール、炭素数3~4のケトン、炭素数1~5のカルボン酸、炭素数1~5のカルボン酸のエステルから選択された1種又は2種以上の混合物を、天然ゴムラテックスのゴム分に対して0.001~30重量%用いることを特徴とする、請求項1に記載の蛋白質フリー天然ゴムラテックスの製造方法。
- 前記変性処理した蛋白質の除去を遠心分離処理により行うことを特徴とする、請求項1又は2に記載の蛋白質フリー天然ゴムラテックスの製造方法。
- 請求項1~3のいずれかに記載された製造方法で得られた蛋白質フリー天然ゴムラテックスに酸を添加して凝固することを特徴とする、蛋白質フリー天然ゴムの製造方法。
- 請求項1~3のいずれかに記載された製造方法で得られた蛋白質フリー天然ゴムラテックスを乾燥することを特徴とする、蛋白質フリー天然ゴムの製造方法。
- 天然ゴムラテックスを乾燥して得られる固形ゴム中の蛋白質の含有量が、RRIM試験法により測定した窒素含有率において0.001%以下のレベルであり、改良ローリー法により測定した天然ゴムラテックスを乾燥して得られる固形ゴム中の蛋白質量が、0.5μg/g以下のレベルであることを特徴とする蛋白質フリー天然ゴムラテックス。
- 天然ゴム中の蛋白質の含有量が、RRIM試験法により測定した窒素含有率において0.001%以下のレベルであり、改良ローリー法により測定した天然ゴムラテックスを乾燥して得られる固形ゴム中の蛋白質量が、0.5μg/g以下のレベルであることを特徴とする蛋白質フリー天然ゴム。
