本発明のフェニルイミダゾール化合物は、種々の方法により製造される。その好ましい一例を示せば次の通りである。

上記公知化合物(6)を、ハロゲン化物(7)と反応させることにより、化合物(4a)に変換できる。反応は、N,N-ジメチルホルムアミド(DMF)、N,N-ジメチルアセトアミド(DMA)、ジメチルスルホキシド(DMSO)等の不活性溶媒中、炭酸カリウム、炭酸ナトリウム等のアルカリ存在下、室温~100℃にて5~30時間程度で完了する。ここで、ハロゲン化物(7)は、化合物(6)1モルに対して1~2モル使用され、アルカリは化合物(6)1モルに対して1~3モル使用される。

2-ブロモ-1-[4-(4-ブロモ-2-フルオロベンジルオキシ)-3-メトキシフェニル]エタノンの製造
25.0 g (0.15 mol)の1-(4-ヒドロキシ-3-メトキシフェニル)エタノンと 15.4g(0.11 mol)の炭酸カリウムを80 mLのDMFに加えた液に、50.2 g (0.19 mol)の4-ブロモ-1-(ブロモメチル)-2-フルオロベンゼンを加え、室温下2時間、次いで60 ℃で16時間撹拌した。反応溶液に40 mLのメタノールを加え、60 ℃で1時間撹拌した後、200 mLの水を加え、析出した結晶を濾過し、水およびヘキサンで洗浄した。60 ℃で3時間真空乾燥し、52.7gの1-[4-(4-ブロモ-2-フルオロベンジルオキシ)-3-メトキシフェニル]エタノンを得た。
参考例1と同様にして、表1に記載の各化合物を製造した。
2-ブロモ-1-[4-(4-ブロモ-2-フルオロベンジルオキシ)-2-メトキシフェニル]エタノンの製造
25.3 g (0.17 mol)の1-(2,4-ジヒドロキシフェニル)エタノンと、44.6 g (0.17 mol)の4-ブロモ-1-ブロモメチル-2-フルオロベンゼンを、150 mLのDMFに溶かした溶液に、氷冷下11.8g(0.09 mol)の炭酸カリウムを加え、30分間、さらに室温で12時間撹拌した。再度、氷冷下、11.8g(0.09 mol)の炭酸カリウムを加え、30分間、さらに室温で12時間撹拌した。この反応溶液に100 mLのメタノールを加え、80 ℃で1時間撹拌下後、150 mLの水を加え、析出した結晶を濾過しメタノールで洗浄した。60 ℃で2時間真空乾燥し、43.5 gの1-[4-(4-ブロモ-2-フルオロベンジルオキシ)-2-ヒドロキシフェニル]エタノンを得た。(収率75%)
参考例8と同様にして、表2に記載の各化合物を製造した。
適当な出発原料を用い、参考例1または8と同様にして、表3に記載の各化合物を製造する。得られる化合物の構造を、表3に記載する。
4-[4-(4-ブロモ-2-フルオロベンジルオキシ)-3-メトキシフェニル]-2-[4-(トリフルオロメチル)フェニル]-1H-イミダゾール塩酸塩の製造
2.8 g (28.0 mmol)の炭酸水素カリウム、3.0 g (6.9 mmol)の参考例1の化合物及び1.6g (6.9 mmol)のp-トリフルオロメチルベンズアミジン塩酸塩二水和物を、4 mlの水と12 mlのTHFの混合液に加え、60 ℃で16時間撹拌した。室温まで温度を下げた反応溶液に70 mlの酢酸エチルを加え、30分間撹拌した。この溶液を、水、飽和食塩水で順次洗浄し、無水硫酸マグネシウムで乾燥した。溶媒を減圧留去後、ジエチルエーテルを加え析出した結晶をろ過し、70 ℃で1時間、真空乾燥し、3.0 gの目的化合物を得た。得られた化合物の構造と物性を、表4に記載する。
参考例1~12の化合物あるいは公知の2-ブロモ-1-(4-ヒドロキシ-3-メトキシフェニル)エタノン等を原料に用い、実施例1と同様にして、本発明化合物を製造した。得られた化合物の構造と物性を、表4に記載する。
参考例の化合物あるいは適当な出発原料を用い、実施例1と同様にして、本発明化合物を製造した。得られた化合物の構造と物性を、表5に記載する。
適当な出発原料を用い、実施例1と同様にして、本発明化合物を製造する。得られる化合物の構造を、表6に記載する。
ヒト骨格筋細胞及びマウス骨格筋細胞におけるLPL活性化効果の確認試験
プレートに播種後、培養、分化させたヒト骨格筋細胞(human skeletal muscle myoblasts細胞)又はマウス骨格筋細胞(C2C12細胞)を、本発明化合物を溶解した0.3% dimethyl sulfoxide (DMSO)含有培養液(濃度:10μMまたは30μM)に曝露した。曝露から8時間後に培地を除去し、10 mM Tris-HCl buffer (150mM NaCl,10μ/mL; ヘパリンナトリウム含有)を細胞に添加し、37℃、5%CO2存在下で10分間インキュベートした。LPL活性測定キット(Roar Biomedical Inc.製)を用いて、上清中のLPL活性を測定した。LPL活性は、上清とLPL基質を混合してから10分後~70分後における蛍光強度増加量で評価した。0.3%DMSO含有培養液に曝露した細胞の蛍光強度増加量の平均値を対照群として用いた。なお、実施例18、19、32、45および57の化合物についてはヒト骨格筋細胞を用い、その他の化合物についてはマウス骨格筋細胞を用いた。
LPL活性上昇率(%)=[(実験群平均値)-(対照群平均値)]/(対照群平均値)×100
結果を表7に示す。
正常ラットにおける脂質改善作用及び体重増加抑制効果の確認試験
8週齢SDラット(日本チャールス・リバー社:平均体重約350g)を用い、実験群には本発明化合物の投与量を100mg/kg体重に調製した5%アラビアゴム懸濁液を、5ml/kg体重の用量で経口投与し、対照群には5%アラビアゴム懸濁液(本発明化合物を含まない)を、5ml/kg体重の用量で経口投与した。群分けは、一群あたり5匹とし、8週齢で体重を指標に行った。被検化合物は、9週齢から毎日定時に5日あるいは2週間経口投与した。経口投与期間中に定期的に体重を測定し、体重増加抑制効果を観察した。被検化合物最終投与4時間後に静脈より血液を採取し、採取した血液から血漿を分離し、血液生化学的検査(トリグリセライド、HDL-コレステロール)を行った。
体重増加抑制率(%)=[(対照群平均値)-(実験群平均値)]/(対照群平均値)× 100
トリグリセライド低下率(%)=[(対照群平均値)-(実験群平均値)]/(対照群平均値)× 100
HDL-コレステロール上昇率(%)=[(実験群平均値)-(対照群平均値)]/(対照群平均値)× 100
食餌性肥満マウスにおける抗肥満作用の確認試験
AKR/Jマウスを用い、60%カロリー脂肪含有高脂肪食を摂取させ、食餌性の肥満モデルを作製した肥満マウスの実験群には本発明化合物の投与量が20mg/kg体重になるように調整した5%アラビアゴム懸濁液を、5ml/kg体重の割合で経口投与した。対照群には、本発明化合物を含まない5%アラビアゴム懸濁液を、5ml/kg体重の割合で経口投与した。群分けは、体重を指標に行った。被験液は、毎日定時に4週間投与した。経口投与期間中に体重を測定し、体重増加抑制効果を確認した。対照群および実験群の体重の測定値より、体重低下率(%)を次式に従って算出した。結果を表10に示す。

錠剤の調製
有効成分として実施例18で得た化合物を用いて、1錠当たりその300mgを含有する錠剤(10000錠)を、次の処方により調製した。

カプセル剤の調製
有効成分として実施例57で得た化合物を用いて、1カプセル当たりその200mgを含有する硬質ゼラチンカプセル剤(10000カプセル)を、次の処方により調製した。

Claims (24)
- 下記一般式(1)
であらわされるフェニルイミダゾール化合物。 - 一般式(1)においてR7およびR8が水素原子を示す請求項1に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR4がチエニル基を示す請求項1または2に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR4がフリル基を示す請求項1または2に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR4が、置換基として低級アルキル基、低級アルコキシ基、ハロゲン原子、カルボキシ基、低級アルコキシカルボニル基およびハロゲン置換低級アルキル基から選ばれる基を1または2個有することのあるフェニル基を示す請求項1または2に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR1が未置換のフェニル低級アルキル基または置換基としてハロゲン原子、シアノ基およびハロゲン置換低級アルキル基から選ばれる基を1または2個有するフェニル低級アルキル基である請求項1または2に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR1がベンジル基、4-シアノベンジル基、3-シアノベンジル基、2-シアノベンジル基、4-クロロベンジル基、4-トリフルオロメチルベンジル基、4-クロロ-2-フルオロベンジル基および4-ブロモ-2-フルオロベンジル基から選ばれる基である請求項6に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR1がベンジル基、4-シアノベンジル基、3-シアノベンジル基、2-シアノベンジル基、4-クロロベンジル基および4-ブロモ-2-フルオロベンジル基から選ばれる基である請求項6に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR4が、チエニル基、フリル基または置換基として低級アルキル基、低級アルコキシ基、ハロゲン原子、低級アルコキシカルボニル基およびハロゲン置換低級アルキル基から選ばれる基を1または2個有することのあるフェニル基である請求項1、2または6に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR4が、2-チエニル基、3-チエニル基、3-フリル基、フェニル基、4-フルオロフェニル基、3-フルオロフェニル基、3-フルオロ-4-メチルフェニル基、3,4-ジフルオロフェニル基、4-トリフルオロメチルフェニル基、3-トリフルオロメチルフェニル基、2-トリフルオロメチルフェニル基、4-クロロフェニル基、3-クロロフェニル基、3,4-ジクロロフェニル基、3,5-ジクロロフェニル基、3-クロロ-4-フルオロフェニル基、3-メトキシフェニル基、4-メトキシフェニル基、3,4-ジメトキシフェニル基、4-メトキシカルボニルフェニル基、4-カルボキシフェニル基、4-(1,1-ジメチルエチル)フェニル基、1-メチルエチル基および4-メチルフェニル基から選ばれる基である請求項1、2または6に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR4が、2-チエニル基、3-チエニル基、3-フリル基、フェニル基、4-フルオロフェニル基、3-フルオロフェニル基、3,4-ジフルオロフェニル基、4-トリフルオロメチルフェニル基、3-トリフルオロメチルフェニル基、2-トリフルオロメチルフェニル基、4-クロロフェニル基、3-クロロフェニル基、3,4-ジクロロフェニル基、3,5-ジクロロフェニル基、3-クロロ-4-フルオロフェニル基、3-メトキシフェニル基、4-メトキシカルボニルフェニル基、4-(1,1-ジメチルエチル)フェニル基、1-メチルエチル基および4-メチルフェニル基から選ばれる基である請求項10に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR1が、置換基としてハロゲン原子、シアノ基およびハロゲン置換低級アルキル基から選ばれる基を1または2個有することのあるピリジル低級アルキル基である請求項1または2に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR1が、5-トリフルオロメチル-2-ピリジルメチル基、2-ピリジルメチル基、3-ピリジルメチル基、5-クロロ-2-ピリジルメチル基および5-シアノ-2-ピリジルメチル基から選ばれる基である請求項12に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR1が、置換基としてハロゲン置換低級アルキル基を有することのあるピリジル低級アルキル基である請求項1または2に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR1が、2-ピリジルメチル基および6-トリフルオロメチル-3-ピリジルメチル基から選ばれる基である請求項14に記載のフェニルイミダゾール化合物。
- 一般式(1)においてR1がベンジル基、4-クロロベンジル基および4-ブロモ-2-フルオロベンジル基から選ばれる基であり、R4が、4-トリフルオロメチルフェニル基、4-フルオロフェニル基、3,4-ジフルオロフェニル基、4-クロロフェニル基、3,4-ジクロロフェニル基および4-メチルフェニル基から選ばれる基である請求項1または2に記載のフェニルイミダゾール化合物。
- 以下の化合物から選ばれる請求項1に記載のフェニルイミダゾール化合物:
