Abstract
Description
厚生労働省によると、冬の急性胃腸炎患者の10分の1はノロウイルス感染が原因している。ノロウイルスの感染経路はほとんどが経口感染であり、食品取扱者や調理器具からの2次汚染を防止するという観点から、ノロウイルスの失活化または不活化方法が重要である。
(a)エタノール、イソプロピルアルコール、又はこれらの混合物を、消毒剤全体に対して40~90%(w/w)
(b)乳酸を、消毒剤全体に対して0.1~2%(w/w)
(c)クエン酸を、消毒剤全体に対して0.01~2%(w/w)
(d)溶液中で亜鉛イオンを遊離する亜鉛含有化合物を、消毒剤全体に対して0.001~0.1%(w/w)

[発明が解決しようとする課題]
上記のように、厚生労働省によって、ノロウイルス失活化に効果があるとされている次亜塩素酸ナトリウム水溶液には、いくつかの問題があり、また、エタノールは、単独ではノロウイルス失活化に効果がなく、特殊な天然抽出物(グレープフルーツ種子抽出液)とフィチン酸の併用により、カリシウイルス用除菌液として知られているにすぎない。
上記課題を解決する本発明は以下のとおりである。

カリシウイルスの存在が疑われ、かつタンパク質を含有する物に、有効成分としてエタノールと酸を含み、pHが2.5~5.0の範囲にある水溶液からなる組成物を接触させることを含む、タンパク質共存下のカリシウイルスを不活化する方法。

前記カリシウイルスの存在が疑われ、かつタンパク質を含有する物が、施設または物品表面に存在し、
前記施設または物品表面の少なくとも一部または全部に水透過性及び/又は保持性のシートを被覆する工程、

本発明によれば、エタノール製剤(組成物)を用いて、タンパク質と共存するカリシウイルスを不活化できる方法を提供でき、特にノロウイルスの蔓延抑制に有効である。さらに、本発明によれば、医療や介護の現場において必要とされている、ノロウイルスなどのノンエンベロープウイルスを含む吐瀉物や糞尿等に対するウイルスの有効かつ安全な不活化方法を提供することができる。
本発明は、カリシウイルスの存在が疑われ、かつタンパク質を含有する物に、有効成分としてエタノールと酸を含み、pHが2.5~5.0の範囲にある水溶液からなる組成物を接触させることを含む、タンパク質共存下のカリシウイルスを不活化する方法に関する。
床等に飛び散った患者の吐瀉物や糞便を処理するときには、使い捨てのガウン(エプロン)、マスクと手袋を着用し汚物中のウイルスが飛び散らないように、糞便、吐しゃ物をペーパータオル等で静かに拭き取ることが適当である。拭き取った後は、床をシートで被覆し、被覆したシートに前記組成物を供給し、シート下の床を消毒する。さらに、シートで被覆されていない床も念のため前記組成物を噴霧する等して、飛び散った可能性がある飛沫も処理することが好ましい。あるいは、拭き取った後に、シートで被覆することなく、前記組成物で浸すように床をさらに拭き取ることもできる。シートを用いない方法は、飛び散った吐瀉物等が少量の場合には適当である場合がある。これらの処理後、必要により、水拭きをすることもできる。
以下、本発明を実施例に基づいてさらに詳細に説明する。
エタノール試薬特級99.5%をW/W%で80%、70%、60%、50%、40%、30%、20%となるようにM/100リン酸緩衝液pH7.2(以下、PBSと略記する)で希釈しサンプルを調製し、以下に示す試験方法にてサンプルの消毒効果(ウイルス不活化効果)を測定した。結果を表1に示す。
1)サンプルの消毒効果(ウイルス不活化効果)を測定するための使用ウイルスは、ネコカリシウイルスF9株を用いた。
2)サンプル1およびサンプル2の原液および2倍希釈液(M/100 リン酸緩衝食塩液pH7.2:PBSにて希釈)を用いた。消毒効果のエンドポイントが得られなかった場合は、さらにサンプルを希釈した。
3)それぞれのサンプル液900μlに、ウイルス液を100μl加え、室温にて1分間と30分間作用させた。
反応後100μl採取し、多量のタンパク質を含む日水製薬社製ブイヨン液(肉エキス5mg、ペプトン15mg/ml)900μlに加えた。日水製薬社製ブイヨンを用いたのは、作用後の試験サンプルの消毒効果の進行を阻止するためである。
4)用いたウイルスのウイルスコントロール群は、消毒薬サンプルの代わりにブイヨン液で希釈し感染価を測定した。
5)ブイヨン液で10倍に希釈したウイルス液は、ブイヨン液にてさらに10-1~10-4に希釈した(階段希釈液)。
6)6穴プレート上に4日間培養させたCrFK細胞の細胞増殖用培地(ウシ胎児血清10%含むMEM培地)をアスピレーターにて取り除き、各階段希釈したウイルス液について3穴の細胞を使用し、それぞれに100μl接種した。
7)インキュベータ内で1時間ウイルス吸着を行った後、ウシ胎児血清を含まない組織培養液(MEM培地;MINIMUM ESSENTIAL MEDIUM EAGLE (MODIFIED) WITH EARLE`S SALTS(MIP Biomedicals,Inc)を重炭酸ナトリウムにてpH7.1に調整)を1穴あたり3ml加え、そして取り除くことにより細胞の洗浄を行った。(検査サンプルの細胞への毒性を除くため)
8)0.7%寒天を含むMEM寒天培地(0.03%L-グルタミン、0.22%重炭酸ナトリウム含有)をプレート1穴あたり3ml加え(40℃)、寒天培地が固形化したら6穴プレートを反転し、5%CO2インキュベータにて72時間培養した。
9)培養終了後の6穴プレートに1穴あたり1.5mlの10%ホルマリンPBS液を加え1時間室温で放置し細胞固定後、流水で寒天培地を取り除き、メチレンブルー染色を行った。この方法により、ウイルス数をプラークとして数えることができるようになった。
10)消毒薬サンプルを加えないウイルスの感染価に対し、消毒薬を加えたウイルスの感染価を比較することで消毒効果を評価した。
表2に示す組成のサンプルを調製し、例1に記載した試験方法にてサンプルの消毒効果(ウイルス不活化効果)を測定した。結果を表3に示す。
表4に記載のサンプルにネコカリシウイルス原液を等量加え、室温1分間反応後、5倍濃度のブイヨン液(タンパク質量80mg/ml)で10倍に希釈し細胞毒性を消失させた。更に10倍段階希釈で10-6までサンプルの希釈列には希釈液にMEMを用いた。各希釈液をコンフルエントになった3日間培養ネコ肝臓細胞(CrFK)6穴プレートの3穴に各100μl接種し、ウイルス吸着90分後に、0.7%寒天を含むMEM培地を重層した。34℃、5%CO2培養インキュベータにて50時間培養した。10%ホルマリンを用いた数時間の固定後、寒天培地を除去し、メチレンブルー染色液を加え、染色後のプラーク数をカウントした。結果を表5に示す。
※次亜塩素酸ナトリウムは、純正化学(株)製であり、有効塩素濃度の測定値は5.6%である。
※ネコカリシウイルス:F9株
※サンプル1~4のエタノール濃度、サンプル1~8のクエン酸濃度およびサンプル9~13の次亜塩素酸ナトリウム濃度は、ウイルスと等量混合後2倍に希釈されるのでいずれも濃度は2分の1になる。
1.クエン酸とエタノールの混合製剤は、ネコカリシウイルスに対してクエン酸5%から1.25%まで優れたウイルス不活化効果を認めたが、0.625%では効果が減少した。
2.一方、エタノールを含まないクエン酸のみのグループでは、5%に於いても効果がなかった。
3.例1の結果を併せるとそれぞれ単体では効果のないものが、混合することにより効果を発揮したことが明らかになった。
4.次亜塩素酸ナトリウムのウイルス完全不活化には、最終濃度200ppmが必要であった。
表6に示すサンプルを調製し、例3に記載した方法に従って消毒効果(ウイルス不活化効果)を測定した。但し、ネコカリシウイルス液はタンパク質量の異なる普通ブイヨンを含むもの、および普通ブイヨンを含まないMEM培地のみ含むものを用いた。比較として次亜塩素酸ナトリウムについても同様の試験を行った。
1.ネコカリシウイルスにブイヨンタンパク質を加えることにより、消毒効果の減少が認められた。
2.クエン酸5%+エタノールではタンパク質共存下でも消毒効果は減少しなかった。
3.乳酸5%単独では、ウイルス不活化効果を認めなかったが、エタノールを加えることにより効果を認めた。
4.次亜塩素酸ナトリウムは、高濃度(最終濃度800ppm)であるにもかかわらず、タンパク質共存下では消毒効果が消失した。
5.自然界のウイルスは何らかの形でタンパク質と混在しているため、クエン酸5%+エタノールのような、タンパク質混在下のウイルスを不活化する効果は、非常に有用である。
ネコカリシウイルスに混在するタンパク質量と、不活化効果の相関性を調べた。表7に示すサンプルを調製し、例3に記載した方法に従って消毒効果(ウイルス不活化効果)を測定した。但し、ネコカリシウイルス液はタンパク質量の異なる普通ブイヨンを含むもの、および普通ブイヨンを含まないMEM培地のみ含むものを用いた。比較として次亜塩素酸ナトリウムについても同様の試験を行った。
1.クエン酸5%(2.5%)+エタノールは、20mg/mlのタンパク質を含むウイルスを完全に不活化した。
2.クエン酸2.5%(1.25%)+エタノールは、10mg/mlのタンパク質を含むウイルスを完全に不活化した。
3.クエン酸1.25%(0.625%)+エタノールは、4mg/mlのタンパク質を含むウイルスを103まで不活化したが完全に不活化していない。
4.次亜塩素酸ナトリウム1600ppm(800ppm)は、4mg/mlのタンパク質を含むウイルスを完全に不活化したが10mg/ml、20mg/mlのタンパク質を含むウイルスについては不活化できなかった。
5.次亜塩素酸ナトリウム800ppm(400ppm)、次亜塩素酸ナトリウム400ppm(200ppm)は、4mg/mlのタンパク質を含むウイルスに対しても不活化できなかった。
表8に示すサンプルを調製し、例3に記載した方法に従って消毒効果(ウイルス不活化効果)を測定した。但し、ネコカリシウイルス液はタンパク質量の異なる普通ブイヨンを含むもの、および普通ブイヨンを含まないMEM培地のみ含むものを用いた。比較として次亜塩素酸ナトリウムについても同様の試験を行った。但し、ネコカリシウイルスに10倍濃度の普通ブイヨン(170mg/ml)を加え、タンパク質量80mg/ml、40mg/ml、または20mg/mlを含むウイルス液を作製し、これを用いた。タンパク質量0mg/mlについてはMEM(タンパク質を含まない組織培養液)で代用した。濃度の異なるクエン酸、エタノール、次亜塩素酸ナトリウム溶液に、タンパク質量の異なるネコカリシウイルス液を等量加え、室温、60秒間反応させた。
1.クエン酸5%(2.5%)+エタノールは、最終濃度40mg/mlのタンパク質共存下でウイルスを完全に不活化した。
2.クエン酸2.5%(1.25%)+エタノールは、最終濃度10mg/mlのタンパク質共存下で約106の感染性の減少を示した。
3.次亜塩素酸ナトリウム3200ppm(1600ppm)は、10mg/mlのタンパク質共存下ではウイルスを完全に不活化したが、タンパク質濃度が高くなるにつれウイルスの不活化効果が急激に減少した。
4.次亜塩素酸ナトリウムは、1600ppm(800ppm)以下の濃度では、タンパク質共存下でウイルス不活化作用が消失することが判った。
RT-PCR法により抗ノロウイルスへの不活化効果の評価を行った。試験サンプルの組成は、エタノール68%、乳酸10%、水22%であり、試験材料としては糞便由来のノロウイルス(NV遺伝子2群に属する)を用いた。
材料

1.表9に示すサンプルとタンパク質量 80mg/ml,20mg/ml,10mg/ml,0mg/mlを含むネコカリシウイルス原液を100μlずつ等量混合し、室温で60秒間反応させた。
2.反応後直ちにサンプル・ウイルス混合液100μlを80mg/ml濃度のブイヨン液900μlに加え反応を止めた。これをMEM培地で10-2から10-5まで10倍階段希釈を行った。
3.培養3日目の6穴プレートに10倍階段希釈を行ったウイルス・サンプル液を100μl接種し、90分のウイルス吸着操作を行った後、0.7%寒天培地で重層し5%CO2インキュベーター内で34℃60時間培養した。
4.培養終了後10%ホルマリン固定、メチレンブルー染色により形成されたプラーク数を数え、感染価を判定した。
5.ウイルスコントロールは、サンプルの代わりにMEMを用い、方法はサンプルに準じた。
結果を表10に示す。
1.サンプル1についてはウイルス不活化効果を認め、タンパク質量10mg/mlの共存下に於いても、ウイルス感染性を完全に不活化した。
2.サンプル2についてはウイルス不活化を認めなかった。但し、後述の例11に示すように、サンプル2の組成でもウイルス液量に対する使用量を増大させるとウイルス不活化を認められるようになる。
3.サンプル3及び4についてはウイルス不活化効果を認めたが、サンプル1には及ばなかった。

1.サンプル1~6についてはウイルス不活化効果を認め、タンパク質量20mg/mlの共存下に於いても、ウイルス感染性を完全に不活化した。
2.サンプル7については、タンパク質量20mg/mlを含むウイルス液中のウイルスに対しては不活化効果が低い成績であったが10mg/mlより低濃度のタンパク質を含むウイルス液中のウイルスに対しては不活化効果を認めた。
材料
ウイルス:ネコカリシウイルス F9株
普通ブイヨン10倍濃度:160mg/ml
次亜塩素酸ナトリウム:有効塩素濃度 4%
1.表13に示すサンプルに加えるネコカリシウイルスに普通ブイヨンを加えることにより最終タンパク質濃度80mg/ml,40mg/ml,20mg/ml,0mg/mlとなるように調整した4種類のタンパク質濃度の異なるウイルス液を用意した。
2.サンプルごとに使用するウイルスは、タンパク質濃度の異なるウイルスでウイルス不活化活性を測定した。
3.サンプルを100μlとウイルス100μl等量混合し、ボルテックスにより室温25℃、60秒間攪拌作用させた後、100μlを採取し、900μlの80mg/mlタンパク質濃度のブイヨン液に加え10倍に希釈した。このような高濃度のブイヨン液を使用したのは、サンプルの作用停止とプラーク形成に用いるCrFK細胞に対する消毒薬の細胞毒性を無毒化させるためである。
4.10-2から10-5までの希釈液にはMEMを用い、6穴プレートに各希釈液を1希釈に付き2穴ずつ100μl接種した。
5.コントロールウイルス群は各濃度のタンパク質を含むウイルスにMEMを等量に加えた。
6.90分間の細胞へのウイルス吸着、寒天培地の重層、34℃で60時間培養した。
7.培養後のプレートに形成されたプラークは、ホルマリン固定の後、メチレンブルー染色しプラーク数により感染価を計算した。
タンパク質共存下でウイルスを不活化させる作用は、サンプル3が優れていると思われた。

1.ウイルス:薬剤=1:1の系において、サンプルB,F,Gについては、最終タンパク質10mg/ml存在下においてもウイルス不活化効果を認めた。
2.サンプルA,Cについてもウイルス不活化効果を認めたが、B,F,Gには及ばなかった。
3.ウイルス:薬剤=1:9の系において、サンプルA~Dについては、最終タンパク質濃度16mg/ml存在下においてもウイルス不活化効果を認めた。
4.サンプルEについてもウイルス不活化効果を認めたが、A~Dには及ばなかった。

- カリシウイルスの存在が疑われ、かつタンパク質を含有する物に、有効成分としてエタノールと酸を含み、pHが2.5~5.0の範囲にある水溶液からなる組成物を接触させることを含む、タンパク質共存下のカリシウイルスを不活化する方法。
- 前記カリシウイルスの存在が疑われ、かつタンパク質を含有する物が、施設または物品表面に存在し、
前記施設または物品表面の少なくとも一部または全部に水透過性及び/又は保持性のシートを被覆する工程、
前記シート上に前記組成物を供給して、シート下の前記表面に前記組成物を接触させる工程、及び
接触後、所定時間放置する工程

- 前記カリシウイルスの存在が疑われ、かつタンパク質を含有する物が、カリシウイルス感染者の吐瀉物または糞尿である請求項1~4のいずれかに記載の方法。
- 前記組成物におけるエタノールの濃度が、40~85%(w/w)の範囲である請求項1~6のいずれかに記載の方法。
- 前記酸が有機酸または無機酸である請求項1~7のいずれかに記載の方法。
- 前記酸の濃度が1~10%の範囲である請求項1~8のいずれかに記載の方法。
- 前記酸は、1~10%の濃度において、前記水溶液のpHが2.5~5.0の範囲を示す酸である請求項1~9のいずれかに記載の方法。
- 前記酸は、クエン酸、乳酸またはリン酸である請求項8に記載の方法。
- 前記組成物はポリグリセリン脂肪酸エステルをさらに含有する請求項1~11のいずれかに記載の方法。
- 前記カリシウイルスが、ノロウイルスである請求項1~12のいずれかに記載の方法。
- 前記カリシウイルスの存在が疑われ、かつタンパク質を含有する物の容量1に対して前記組成物の供給量(容量比)が、0.5~100の範囲である請求項1~13のいずれかに記載の方法。
