Description
また本発明は、乳化剤とフィチン酸とを含み、かつpH3.5以下である、対象への抗菌性付与用液状組成物を提供する。
該液状組成物が、乳化剤とフィチン酸とを含み、かつpH3.5以下である、
殺菌又は静菌方法を提供する。
さらに本発明は、液状組成物を対象に噴霧もしくは塗布するか、又は該液状組成物に対象を浸漬させることを含み、
該液状組成物が、乳化剤とフィチン酸とを含み、かつpH3.5以下である、
対象への抗菌性付与方法を提供する。
さらに本発明は、乳化剤とフィチン酸とを含み、かつpH3.5以下である液状組成物の、対象への抗菌性付与のための使用を提供する。
さらに本発明は、乳化剤とフィチン酸とを含み、かつpH3.5以下である液状組成物の、対象への抗菌性付与用液状組成物の製造のための使用を提供する。
〔液状組成物の調製〕
室温下で、乳化剤(カプリル酸モノグリセリド又はカプリン酸モノグリセリド)、エタノール及び滅菌済み精製水を1:2:2の割合で混合して溶液を調製し、これに各種有機酸、及び残余のエタノールと滅菌済み精製水を添加して、表1記載の液状組成物(溶液)を調製した。得られた液状組成物のpHを測定した。また、下記の手順で各液状組成物の殺菌又は静菌特性を評価した。
(1)サンプル菌液の調製
対象菌には、4種の乳酸菌Weissella viridescens、Leuconostoc mesenteroides、Leuconostoc citreum、及びLeuconostoc pseudomesenteroidesを用いた。対象菌のシングルコロニーを、白金耳を用いて数コロニー釣菌し、TSB培地に植菌して30℃で20時間種培養したものを乳酸菌液(107cfu/mL)とした。得られた乳酸菌液を、滅菌済み精製水10mLに0.1mL混合し、さらにこれを10倍希釈した。4種の乳酸菌の10倍希釈液を等量混合してサンプル菌液を調製した。
塗布直後及び18時間後の液状組成物による静菌特性を調べた。
滅菌プラスチックシャーレを二群に分けた。一方の群では、各液状組成物を1.0mLずつ空のシャーレに入れ、コンラージ棒で均一に塗布し、次いでこれにサンプル菌液を1.0mL接種し、20秒間コンラージ棒で混合しながら塗り広げた(0h群)。もう一方の群では、各液状組成物を1.0mLずつ空のシャーレに入れ、コンラージ棒で均一に塗布した後、安全キャビネット内で18時間風乾し、水分とエタノールを完全に蒸発させた。これにサンプル菌液を1.0mL接種し、20秒間コンラージ棒で混合しながら塗り広げた(18h群)。初発菌数は105~106cfu/mLに調整した。
サンプル菌液を塗布したシャーレを、5分間静置後、シャーレ内の液体を採取し、MRS寒天培地を用いて菌数を測定した。
(2)で測定した菌数をもとに、下記評価基準にて各組成物(液体群及び乾燥群)の殺菌又は静菌特性を評価した。
A:初発の1/1000以下に菌数が減少
B:初発の1/100以下~1/1000超に菌数が減少
C:初発の1/10以下~1/100超に菌数が減少
D:菌数が初発の1/10よりも多い
試験例1と同様の手順で、但しフィチン酸の濃度を変えて、表2~3の液状組成物(溶液)を調製し、殺菌又は静菌特性を評価した。
試験例1と同様の手順で、但し乳化剤の濃度を変えて、表4~5の液状組成物(溶液)を調製し、殺菌又は静菌特性を評価した。
様々な素材上における液状組成物の殺菌又は静菌作用を調べた。
試験例1と同様の手順で、液状組成物(カプリル酸モノグリセリド 0.15%、フィチン酸 0.40%、エタノール 40.00%、水 残余;単位は質量%;pH2.00)、及びサンプル菌液を調製した。
対象素材として、金属(ステンレス板)、ゴム(ゴム手袋)、プラスチック(番重ふた)を準備した。ステンレス板及び番重ふたは、10cm四方に切断して使用した。ゴム手袋は、10cm四方のプラスチック板に皺のないように伸ばして両面テープで貼り、余った部分はカットして使用した。
サンプル菌液を塗布した対象素材を5分間静置後、無菌パウチに入れ、上から滅菌水10mLを加え、熱シールして上下に5回程度軽く振った。パウチを開封して中の液体を採取し、MRS寒天培地で菌数を測定し、試験例1と同様の基準で殺菌又は静菌特性を評価した。
Claims (14)
- 乳化剤とフィチン酸とを含み、かつpH3.5以下であり、該乳化剤がカプリル酸モノグリセリド及びカプリン酸モノグリセリドから選択される少なくとも1種である、
対象に付着させ乾燥させることで該対象に抗菌性を付与するための液状組成物。 - 前記乳化剤の濃度が0.005質量%以上である、請求項1記載の組成物。
- 前記フィチン酸の濃度が0.005質量%以上である、請求項1又は2記載の組成物。
- 前記乳化剤と前記フィチン酸との質量比が1:0.005~1:200である、請求項1~3のいずれか1項記載の組成物。
- さらにエタノールを含有する、請求項1~4のいずれか1項記載の組成物。
- さらに水性溶媒を含有する、請求項1~5のいずれか1項記載の組成物。
- 対象の面積100cm2あたり0.1~5.0mLの用量で使用される、請求項1~6のいずれか1項記載の組成物。
- 液状組成物を対象に噴霧もしくは塗布するか、又は該液状組成物に対象を浸漬させることにより、該液状組成物を該対象に付着させること、及び
該液状組成物が付着した対象を乾燥させること、
を含み、
該液状組成物が、乳化剤とフィチン酸とを含み、かつpH3.5以下であり、該乳化剤がカプリル酸モノグリセリド及びカプリン酸モノグリセリドから選択される少なくとも1種である、
対象への抗菌性付与方法。 - 前記液状組成物における前記乳化剤の濃度が0.005質量%以上である、請求項8記載の方法。
- 前記液状組成物における前記フィチン酸の濃度が0.005質量%以上である、請求項8又は9記載の方法。
- 前記液状組成物における前記乳化剤と前記フィチン酸との質量比が1:0.005~1:200である、請求項8~10のいずれか1項記載の方法。
- 前記液状組成物がさらにエタノールを含有する、請求項8~11のいずれか1項記載の方法。
- 前記液状組成物がさらに水性溶媒を含有する、請求項8~12のいずれか1項記載の方法。
- 前記液状組成物が対象の面積100cm2あたり0.1~5.0mLの用量で使用される、請求項8~13のいずれか1項記載の方法。
