Description
本發明係有關為了從含有不同聚合度之寡聚原花青素(oligomeric proanthocyanidine,亦有稱為寡聚原花色素之情況)之原料獲得美白效果等高之寡聚原花青素之精製方法、寡聚原花青素聚合度之調整方法及以寡聚原花青素作為有效成分之透明質酸酶抑制劑及膠原酶抑制劑等。
以獲得美白、改善皮膚粗糙等美容效果為目的,廣泛使用皮膚外用劑、化粧品。已知具有美白作用等之種種物質,惟,合成品長期用於人類之皮膚時,有很多安全性之問題,因此,尋求源自天然之成分且效果高者。
寡聚原花青素(以下,亦稱為「原花青素」或「OPC」)為在葡萄、蘋果、可可等植物中含量多之多酚之一種,亦稱為「縮合型丹寧」或「黃烷-3-醇聚合物」之化合物。OPC之構造通常為以黃烷-3-醇作為構成單位,而為黃烷-3-醇在4-6位或4-8位縮合或聚合之結合樣式。OPC為根據上述結合樣式進行縮合或聚合之
二聚物以上之聚合體之總稱。
有OPC具有抗氧化作用、美白作用等種種生理活性之報告。例如於專利文獻1記載有從在康科特紅葡萄(Concord red grape)之果實粉碎物中加入精製水,於50至60℃萃取24小時之萃取物獲得之葡萄萃取物具有酪胺酸酶活性抑制作用及活性氧消除作用。於專利文獻2記載有聚合度為12之縮合型丹寧顯示美白作用及酪胺酸酶活性抑制效果。於專利文獻3記載有酒糟乙醇萃取物顯示梅納褐變反應(Maillard reactions)抑制活性,在該萃取物中含有兒茶酸、前花青素B及前花青素C。於專利文獻4揭示含有原花青素之梅納褐變反應抑制劑。於專利文獻5及6記載有將從製造酒之殘留物及其萃取物用於抗炎症劑、抗氧化劑、肌膚年輕化劑等化粧品配合物及/或醫藥品配合物製造用之活性物質組成物。
關於該等生理活性與OPC聚合度活性之相關性大都不是很瞭解,惟,對於例如美白作用,有聚合度為約12之OPC之效果高之報告(專利文獻2)。因此,尋求可以高純度調製具有所期待生理活性之OPC之技術。
從植物原料將OPC分離精製通常經由以熱水或有機溶劑萃取葡萄等植物原料而進行。惟,於萃取中,由於與聚合度無關的萃取出原花青素混合物,要從獲得之萃取液將作為目的之具有生理活性之OPC以高純度分離及精製有困難。因此,尋求從原料以更簡單、效率佳且以高純度調製具有所期待生理活性之OPC之方法。
又,預防或改善隨著年齡增加發生之皺紋及鬆弛
等,在美容上為重要之課題。為皮膚老化徵兆代表之皺紋、鬆弛之原因雖然尚未完全明瞭,其原因之一可列舉隨著年齡增加,組織中透明質酸之量減少,導至皮膚失去水嫩感。因此,抑制將透明質酸分解之透明質酸酶之活性,對於維持皮膚真皮中透明質酸之量,保持水嫩且具張力的肌膚有效。又,認為皮膚中之膠原減少為引起皮膚皺紋或鬆弛之主要原因。膠原酶為參予膠原分解之酵素,與皮膚彈性降低或產生皺紋等有關。
因此,抑制透明質酸酶及膠原酶之活性,給予皮膚彈力或張力,在防止皮膚老化上很重要。例如於專利文獻7揭示含有將原花青素之每個構成單體之没食子酸付加率為10%(莫耳比)以下之原花青素作為有效成分之膠原酶抑制劑或肌質改善劑。又,於專利文獻8揭示含有將葡萄種子及/或葡萄渣滓之萃取物作為有效成分之透明質酸酶抑制劑。惟,仍尋求對於透明質酸酶及膠原酶具有更優越抑制效果,可更有效地改善皺紋或鬆弛等肌膚老化,安全性高之化粧品等。

本發明之目的係有鑑於上述課題,提供可將具有美白作用等種種生理活性之寡聚原花青素(OPC)以簡便之操作、即可高純度地從原料分離(萃取)及精製之精製方法。本發明又以提供在從原料萃取OPC時,調整萃取液中OPC聚合度之方法為目的。本發明更以提供將經由上述精製方法獲得之OPC作為有效成分之安全性高且發揮優越效果之黑色素生成抑制劑、透明質酸酶抑制劑及發揮優越效果之膠原酶抑制劑等為目的。
本發明人等有鑑於上述課題,進行深入研究之結果發現從含有複數種OPC之原料萃取OPC時,根據萃取溶劑中醇之濃度可萃取出不同聚合度之OPC。例如,只要萃取溶劑中醇之濃度為約30至70v/v%,即可萃取出聚合度高之OPC。另一方面,萃取溶劑中醇之濃度若未達30v/v%,例如在約20v/v%以下,則萃取出聚合度低之OPC。更具體而言,若使用約30至70v/v%之醇水溶液進行萃取,則萃取出平均聚合度為8以上之OPC。又,若使用約20v/v%以下之醇水溶液進行萃取,則萃取出平均聚合度未達8之OPC。上述根據萃取溶劑中醇之濃度,萃取出之OPC不同聚合度之為令人驚奇之知識。根據該等方法,可簡單地調製收率佳且高純度之具有各種生理活性之OPC。又,經由調整萃取中使用之溶劑之醇濃度,可調整所獲得萃取液中OPC之聚合度。
本發明人等另發現經由上述精製方法,從源自葡萄之原料獲得之OPC具有黑色素生成抑制作用、透明質酸酶抑制作用及膠原酶抑制作用,另,OPC之聚合度越高,該等作用越強。例如使用約30至60v/v%,尤其是約30至50v/v%之醇水溶液萃取出之OPC純度特別高,而發揮甚高之黑色素生成抑制效果、透明質酸酶抑制效果及膠原酶抑制效果。例如透明質酸酶及膠原酶參予皺紋、鬆弛及皮膚之老化,對於該等酵素具有強烈抑制活性之OPC作為防止皺紋、鬆弛效果及防止老化效果高之化粧品等甚有用。
本發明人等以上述見解為基礎進行深入研究,因而完成本發明。
亦即,本發明係有關以下之(1)至(25)。
(1)一種寡聚原花青素之精製方法,其特徵為包含將含有不同聚合度之複數種寡聚原花青素之原料,以水或醇水溶液作為萃取溶劑進行萃取之萃取步驟,於該萃取步驟中,依據目的寡聚原花青素之聚合度設定該萃取溶劑之醇濃度,以萃取具有目的聚合度之寡聚原花青素。
(2)如上述(1)所述之精製方法,其中,該萃取溫度為70至未達100℃。
(3)如上述(1)或(2)所述之精製方法,其中,該醇為乙醇。
(4)如上述(1)至(3)中任一項所述之精製方法,其中,該精製方法為(A)平均聚合度為5至未達8之寡聚原花青素之精製方法或為(B)平均聚合度為8以上之寡聚原花青素之精製方法。
(5)如上述(1)至(4)中任一項所述之精製方法,其中,該(A)精製方法為平均聚合度為5至未達8之寡聚原花青素之精製方法,係將萃取溶劑之醇濃度作成0至20v/v%或(B)精製方法為平均聚合度為8以上之寡聚原花青素之精製方法,係將萃取溶劑之醇濃度作成30至60v/v%。
(6)如上述(1)至(5)中任一項所述之精製方法,其中,該精製方法為平均聚合度為8至10之寡聚原花青素之精製方法,萃取溶劑為40至50v/v%之乙醇水溶液。
(7)如上述(1)至(6)中任一項所述之精製方法,其中,該含有不同聚合度之複數種寡聚原花青素之原料為源自植物者。
(8)如上述(7)所述之精製方法,其中,該源自植物之原料為至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料。
(9)一種具有黑色素生成抑制作用之寡聚原花青素之精製方法,包含將至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料,於70至未達100℃,在30至60v/v%之醇水溶液中進行萃取之萃取步驟。
(10)一種透明質酸酶抑制劑,係將寡聚原花青素作為有效成分,該寡聚原花青素係將至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料,在30至60v/v%之醇水溶液中進行萃取而得之萃取液或萃取物中所含有者。
(11)如上述(10)所述之透明質酸酶抑制劑,其中,該萃取溫度為70至未達100℃。
(12)一種膠原酶抑制劑,係將寡聚原花青素作為有效
成分,該寡聚原花青素係將至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料,在30至60v/v%之醇水溶液中進行萃取而得之萃取液或萃取物中所含有者。
(13)如上述(12)所述之膠原酶抑制劑,其中,該萃取溫度為70至未達100℃。
(14)如上述(10)至(13)中任一項所述之抑制劑,其中,該醇為乙醇。
(15)如上述(10)至(14)中任一項所述之抑制劑,其中,該醇水溶液為40至50v/v%之乙醇水溶液。
(16)如上述(10)至(15)中任一項所述之抑制劑,其中,該寡聚原花青素之平均聚合度為8至10。
(17)一種透明質酸酶抑制劑,係將平均聚合度為8至10之寡聚原花青素作為有效成分。
(18)一種膠原酶抑制劑,係將平均聚合度為8至10之寡聚原花青素作為有效成分。
(19)如上述(17)或(18)所述之抑制劑,其中,該寡聚原花青素係至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料中所含有者。
(20)如上述(17)至(19)中任一項所述之抑制劑,其中,該寡聚原花青素之平均聚合度為8.5至9.5。
(21)如上述(17)至(20)中任一項所述之抑制劑,其中,該寡聚原花青素係將至少1種自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料,於70至未達100℃,在30至60v/v%之醇水溶液中進行萃取而得之萃取液或萃取物中所含
有者。
(22)如上述(19)至(21)中任一項所述之抑制劑,其中,該葡萄為赤霞珠(Cabernet Sauvignon)。
(23)如上述(10)至(22)中任一項所述之抑制劑,其中,該抑制劑為皮膚外用劑或化粧品。
(24)一種寡聚原花青素聚合度之調整方法,其特徵為包含將含有不同聚合度之複數種寡聚原花青素之原料,以水或醇水溶液作為萃取溶劑進行萃取之萃取步驟,於該萃取步驟中,依據目的寡聚原花青素之聚合度設定該萃取溶劑之醇濃度,以萃取具有目的聚合度之寡聚原花青素。
(25)如上述(24)所述之精製方法,其中,該方法為(A)經由將萃取溶劑之醇濃度作成0至20v/v%,將萃取液中寡聚原花青素之平均聚合度調整為5至未滿8或(B)經由將萃取溶劑之醇濃度作成30至60v/v%,將萃取液中寡聚原花青素之平均聚合度調整為8以上者。
根據本發明之精製方法,可將顯示優越美白作用等生理活性之OPC從原料以簡單且以高收率、高純度進行分離及精製。經由該等方法精製之OPC由於為高純度,若作為醫藥品、化粧品、飲食品、醫藥部外品等之材料使用,在提昇該醫藥品、化粧品、飲食品、醫藥部外品等之機能性有用。例如,於本發明之精製方法中,由於從源自葡萄之原料,使用約30至60v/v%之醇水溶液為萃取溶劑,獲得之OPC具有優越之透明質酸酶抑制作用、膠原酶抑制作用、黑色素生成抑制作用等,因而發揮優越之
防止皺紋、鬆弛之效果、防止老化之效果、美白效果等,適合用於皮膚外用劑、化粧品、醫藥部外品等。
又,根據本發明,可提供安全性高且發揮優越之黑色素生成抑制作用之黑色素生成抑制劑、發揮透明質酸酶抑制效果之透明質酸酶抑制劑及發揮優越膠原酶抑制效果之膠原酶抑制劑。
以下,對本發明作詳細之說明。
本發明寡聚原花青素之精製方法包含將含有不同聚合度之複數種寡聚原花青素之原料以水或醇水溶液作為萃取溶劑,進行萃取之萃取步驟。
本發明之方法只要可達成本發明之效果,亦可包含萃取步驟以外之步驟。
作為含有不同聚合度之複數種寡聚原花青素之原料較好為源自植物之原料,可列舉例如葡萄、可可、蘋果、松、野櫻莓、荔枝等植物之果實、果皮、種子、殻、葉、樹皮及其處理物等。該等可單獨使用,亦可將2種以上組合使用。本發明中使用之原料亦可為該等植物之處理物。處理物可列舉例如粉碎物、破碎物、磨碎物、乾燥物、凍結乾燥物、壓搾物、其濃縮物或壓搾後之固體成分等。
本發明中之原料較好為源自葡萄之原料,更好為例
如至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料等。葡萄較好為例如為葡萄酒原料之品種,可列舉例如為夏多內(Chardonnay)、甲州等白葡萄酒原料之葡萄;為赤霞珠、康克德(Concord)、梅洛(Merlot)、貝利-A(Muscat Bailey A)等紅葡萄酒原料之葡萄等。其中,較好為紅葡萄酒原料之葡萄,更好為赤霞珠、梅洛等,最好為赤霞珠。本發明中之原料亦適合使用例如在製造紅葡萄酒(red wine,亦即紅酒)或白葡萄酒(white wine,亦即白酒)中產生之葡萄果實之壓搾物(渣滓)等。於製造紅葡萄酒中產生之葡萄果實之壓搾物通常為於紅葡萄酒製造步驟中將葡萄果實破碎,將含有果肉、果皮及種子之果汁(葡萄破碎物)發酵後獲得之發酵液壓搾後之渣滓(果皮及種子)等。於製造白葡萄酒中生成之壓搾物通常為於白葡萄酒製造步驟中從葡萄果實壓搾果汁後之渣滓(葡萄之果皮及種子)等。其中,本發明之原料適合使用於製造紅葡萄酒中產生之葡萄果實之壓搾物(渣滓)。更好為赤霞珠、梅洛等葡萄果實之壓搾物,最好為赤霞珠之葡萄果實之壓搾物。若將於製造葡萄酒中產生之葡萄壓搾物作為原料,從可將原本作為廢棄物處理之葡萄壓搾物有效利用之點而言,較佳。
本發明中之萃取溶劑為水或醇水溶液。醇通常為碳原子數約1至4之低級醇,可列舉例如甲醇、乙醇、正丙醇、異丙醇、正丁醇、2-丁醇、丁二醇等。該等係可使用1種或2種以上。其中,較好為乙醇。
於萃取步驟之萃取,通常在原料中添加作為萃取溶劑之水或規定濃度之醇水溶液進行。於萃取步驟中,較好將原料浸漬於萃取溶劑中,亦可進行適當攪拌、掁動等操作。萃取通常
以分批式或連續式進行,較好為分批式。
萃取步驟中原料與萃取溶劑之比例並無特別限制,例如,以重量比計,對於原料1,萃取溶劑較好為約5至30,更好為約5至15,最好為約5至10。
於本發明之精製方法,萃取步驟中之萃取溫度較好為約30至未達100℃(約30℃以上約未達100℃),更好為約70至未達100℃(約70℃以上約未達100℃),最好為約70至90℃,又以約70至未達90℃最好。若為該等溫度,則寡聚原花青素以外之物質之溶出少,可將寡聚原花青素安定萃取,較佳。萃取時間較好為約0.5至4小時,更好為約1至2小時。萃取可在常壓或加壓下進行,較好在常壓下進行。
於上述萃取步驟,依據目的寡聚原花青素之聚合度設定該萃取溶劑之醇濃度,以萃取具有目的聚合度之寡聚原花青素。對應萃取步驟中使用之萃取溶劑中之醇濃度,從原料中萃取不同聚合度之寡聚原花青素。因此,經由適當設定作為萃取溶劑使用之水或醇水溶液之醇濃度,可效率佳的獲得含有作為目的之特定聚合度之寡聚原花青素之萃取液。
萃取溶劑之醇濃度只要依據作為目的之寡聚原花青素,亦即從原料中萃取出之寡聚原花青素之聚合度等加以設計即可,例如較好設定在約0至70v/v%之範圍。將萃取溶劑之醇濃度作成0%為使用水作為萃取溶劑之意思。亦即,於本發明較好在萃取溶劑中使用水或醇濃度為約70v/v%以下之醇水溶液。萃取溶劑之醇濃度只要在該濃度範圍,經由調整萃取中使用之萃取溶劑之醇濃度,可萃取出具有目的聚合度之寡聚原花青素。例如,為
了獲得高聚合度(通常,平均聚合度在約8以上,較好平均聚合度為約8至10)之寡聚原花青素,萃取溶劑之醇濃度較好在約30至70v/v%之範圍,更好為約30至60v/v%,最好為約30至50v/v%,又以在約40至50v/v%之範圍更佳。只要在該等濃度範圍,由於可獲得高純度之寡聚原花青素而較佳。萃取溶劑中使用2種以上之醇時,醇之總濃度較好在上述之範圍。
例如,欲獲得平均聚合度在約5至未達8(約5以上約未達8)之寡聚原花青素時,於萃取步驟中較好將萃取溶劑之醇濃度作成約0至20v/v%,更好作成約0至10v/v%。亦即,作為萃取溶劑,較好使用水或醇濃度在約20v/v%以下之醇水溶液,更好為水或醇濃度在約10v/v%以下之醇水溶液。萃取溶劑之醇濃度若在上述範圍內,則通常從原料係萃取出平均聚合度為約5至未達8之寡聚原花青素。本發明之精製方法適合作為該等平均聚合度為約5至未達8之寡聚原花青素之精製方法。
又,欲獲得例如平均聚合度為超過約6,未達約8之寡聚原花青素時,於萃取步驟中較好將萃取溶劑之醇濃度作成約1至20v/v%,更好作成約10至20v/v%。亦即,作為萃取溶劑較好使用上述濃度之醇水溶液。萃取溶劑之醇濃度若在上述範圍,則通常從原料萃取出平均聚合度為超過約6、未達約8之寡聚原花青素。
在經由上述萃取步驟獲得之萃取液中,通常含有不同聚合度之各種聚合度之寡聚原花青素,惟,經由使用上述規定濃度之醇水溶液作為萃取溶劑,可萃取出平均聚合度在上述範圍之聚合度之寡聚原花青素。例如,如上所述,若將萃取溶劑之醇
濃度作成約30至70v/v%,則通常可獲得含有平均聚合度在約8以上之高聚合度之寡聚原花青素之萃取液。
寡聚原花青素之平均聚合度通常係將寡聚原花青素之數平均分子量除以兒茶酸之分子量(Mw290)而求得。寡聚原花青素之平均聚合度可經由公知之方法求得。例如可經由實施例進行之寡聚原花青素聚合度之測定方法等,從凝膠過濾層析等尺寸大小排除層析之測定結果,使用GPC軟體(島津製作所(股)公司製造)等,算出數平均分子量,將獲得之數平均分子量除以兒茶酸之分子量(Mw290)進行計算。
本發明之精製方法亦適用於作為平均聚合度在約8以上之寡聚原花青素之精製方法。為了獲得平均聚合度在8以上之寡聚原花青素,於萃取步驟中使用之萃取溶劑之醇濃度較好作成約30至70v/v%。亦即,作為萃取溶劑較好使用醇濃度為約30至70v/v%之醇水溶液。又,若使用上述濃度之醇水溶液作為萃取溶劑,通常經由萃取步驟萃取出之寡聚原花青素之平均聚合度之上限通常為約10。
萃取平均聚合度在約8以上之寡聚原花青素時,於萃取步驟中更好將作為萃取溶劑之醇水溶液之醇濃度作成約30至60v/v%。另,若考慮到萃取出之寡聚原花青素之純度,則作為萃取溶劑之醇水溶液之醇濃度較好為約30至50v/v%,更好為約40至50v/v%。只要在該濃度範圍,即可獲得更高純度之寡聚原花青素。
萃取溶劑之醇濃度可根據原料等適當設定,例如於使用源自葡萄之原料時,萃取溶劑之醇濃度若為約30至70v/v%,
則通常萃取出平均聚合度為約8至10之寡聚原花青素。又,於使用源自葡萄之原料時,若使用醇濃度為約30至60v/v%之醇水溶液作為萃取溶劑,則通常可有效率的萃取出平均聚合度為約8.5至10,更好為約8.5至9.5,最好為約8.7至9.5之寡聚原花青素而較佳。
以白葡萄酒用之葡萄作為原料時,若萃取溶劑之醇濃度為約40至60v/v%,較好為約50v/v%,則通常可從原料萃取出平均聚合度在約9以上之寡聚原花青素。以紅葡萄酒用之葡萄作為原料時,若萃取溶劑之醇濃度為約40至60v/v%,則通常可從原料萃取出平均聚合度在9以上之寡聚原花青素。
本發明之另一較佳形式為較好將萃取溶劑之醇濃度作成約40至60v/v。萃取溶劑之醇濃度若為約40至60v/v,則通常可從原料萃取出平均聚合度為約9至10之寡聚原花青素。又,若考慮萃取出之寡聚原花青素之純度,萃取溶劑之醇濃度較好為約40至50v/v%。
於本發明精製方法之較佳形式,係於萃取步驟中較好使用約30至70v/v%,更好約30至60v/v%,最好約30至50v/v%,特別好為約40至50v/v%之乙醇水溶液作為萃取溶劑進行萃取。若使用該等濃度之乙醇水溶液,則通常可從萃取液中萃取出平均聚合度為約8以上,較好平均聚合度為約8.5以上,更好為約8.7以上之高聚合度且高純度之寡聚原花青素。該等精製方法特別適合作為以高純度精製平均聚合度在8以上之寡聚原花青素之方法。
本發明精製方法之最佳形式為於萃取步驟中使用約
40至50v/v%之乙醇水溶液作為萃取溶劑進行萃取。以該等精製方法作為平均聚合度為約8至10之寡聚原花青素之精製方法特別佳。
於本發明之精製方法,依據萃取步驟中使用之萃取溶劑之醇濃度,可從原料萃取出不同聚合度之寡聚原花青素。因此,本發明之精製方法亦可用於作為從原料以高純度萃取目的聚合度之寡聚原花青素之方法。
於上述萃取步驟,經由適當設定萃取中使用之萃取溶劑之醇濃度,可調整獲得之萃取液中寡聚原花青素之聚合度。因此,本發明之精製方法適用於調整萃取液中寡聚原花青素聚合度之方法。
包含將含有不同聚合度之複數種寡聚原花青素之原料,將水或醇水溶液作為萃取溶劑進行萃取之萃取步驟,於該萃取步驟,依據目的寡聚原花青素之聚合度設定該萃取溶劑之醇濃度,以萃取具有目的聚合度之寡聚原花青素之寡聚原花青素聚合度之調整方法亦包含於本發明中。
於上述調整方法中之萃取步驟及其較佳之形式與上述之精製方法中之萃取步驟相同。
例如,於萃取步驟中若將萃取溶劑之醇濃度作成約0至20v/v%,則從原料萃取出平均聚合度為約5至未達8之寡聚原花青素,經由此,可將萃取液中寡聚原花青素之平均聚合度作成約5至未達8。又,若將萃取溶劑之醇濃度通常作成約30至70v/v%,較好約30至60v/v%,更好約30至50v/v%,則從原料萃取出平均聚合度為約8以上之寡聚原花青素。經由此,可將萃取液
中寡聚原花青素之平均聚合度作成約8以上。因此,依據萃取溶劑之醇濃度,可將萃取液中寡聚原花青素之聚合度調整在所期待之範圍。經由(A)將萃取溶劑之醇濃度作成約0至20v/v%,將萃取液中寡聚原花青素之平均聚合度調整為約5至未達8或(B)將萃取溶劑之醇濃度作成約30至60v/v%(較好約30至50v/v%),將萃取液中寡聚原花青素之平均聚合度調整為約8以上之調整方法為本發明較佳實施形式之一。萃取出之寡聚原花青素之平均聚合度上限通常為約10。
本發明之精製方法及調整方法亦可適當包含萃取步驟以外之步驟。例如,於萃取步驟之後可經由過濾、離心分離等適當之方法將萃取液回收。回收之萃取液可以原狀作為醫藥、化粧品、飲食品、醫藥部外品等之材料使用,必要時可進行除去溶劑、OPC成分之濃縮、乾燥、粉末化等之處理。另,在經濃縮之萃取液之乾燥、粉末化時,可適當使用於醫藥品或化粧品領域等使用之賦形劑等。
又,經由上述萃取步驟獲得之萃取液或萃取液之濃縮物等經由超濾等膜處理或吸附層析等各種層析處理,獲得含有更高純度之寡聚原花青素之畫分(fraction)。吸附劑可使用離子交換樹脂、苯乙烯/二乙烯基苯系、甲基丙烯酸系等之吸附劑、親水性乙烯聚合物、修飾葡聚糖凝膠、聚丙烯醯胺凝膠、反相系矽膠等。吸附畫分以醇水溶液、醇等之溶離液溶出,回收。溶離液較好為甲醇、乙醇、丙醇、丙酮等。獲得之畫分之主成分係經由萃取步驟從原料萃取出之寡聚原花青素。對於獲得之含有寡聚原花青素之畫分,可另進行濃縮、乾燥、粉末化等處理或可作成1,3-
丁二醇溶液等。
含有經由上述精製方法獲得之寡聚原花青素之畫分(以下,亦稱為OPC畫分)為以高純度含有特定聚合度之OPC,具有各種生理活性,適用於醫藥品、化粧品、醫藥部外品、飲食品等。
例如,於上述精製方法若原料使用源自葡萄之原料等,於萃取步驟使用約30至60v/v%之醇水溶液作為萃取溶劑,則可獲得含有具有優越黑色素生成抑制作用、透明質酸酶抑制作用及膠原酶抑制作用之高純度OPC之萃取液或OPC畫分。該等萃取液及OPC畫分對具有美白作用、透明質酸酶抑制作用或膠原酶抑制作用之醫藥品、化粧品、飲食品、醫藥部外品等係有用者。本發明之精製方法亦適用於該等具有優越美白作用、透明質酸酶抑制作用及膠原酶抑制作用之寡聚原花青素或以高純度含有該寡聚原花青素之畫分之精製方法、調製方法或製造方法。
於包含將至少一種選自由葡萄果實、葡萄種皮及葡萄之種子所成群組之源自葡萄之原料以約70至未達100℃,於約30至60v/v%之醇水溶液中進行萃取之萃取步驟之具有黑色素生成抑制作用之寡聚原花青素之精製方法亦包含於本發明中。本發明之精製方法及其較佳之形式等與上述之平均聚合度為約8以上(較好約8至10)之寡聚原花青素之精製方法相同。獲得之寡聚原花青素之黑色素生成抑制作用可以公知之方法,例如實施例中記載之方法等進行評估。
經由本發明精製方法獲得之具有黑色素生成抑制作用之寡聚原花青素及以高純度含有該寡聚原花青素之畫分等,係
對人體為安全,且發揮優越之黑色素生成抑制效果者。經由上述精製方法獲得之寡聚原花青素或含有該寡聚原花青素之萃取液、其處理物(萃取物)、畫分等作為黑色素生成抑制劑或美白劑,適用於美白用皮膚外用劑、化粧品、飲食品、醫藥部外品等,發揮優越之預防或抑制成為雀斑或雀卵斑發生原因之黑色素之生成等之效果。
以經由上述精製方法獲得之寡聚原花青素作為有效成分之黑色素生成抑制劑或美白劑亦包含於本發明中。含有經由上述精製方法獲得之寡聚原花青素之萃取液、該萃取液之處理物(萃取物)、含有寡聚原花青素之畫分等可以原狀作為黑色素生成抑制劑或美白劑使用,亦可另添加其他之成分使用。本說明書中、黑色素生成與黑色素產生為相同意義。
以寡聚原花青素作為有效成分,該寡聚原花青素係將至少一種選自由葡萄果實、葡萄種皮及葡萄之種子所成群組之源自葡萄之原料在約30至60v/v%之醇水溶液中進行萃取而獲得之萃取液或萃取物中所含有者之透明質酸酶抑制劑亦為本發明之一。
以寡聚原花青素作為有效成分,該寡聚原花青素係將至少一種選自由葡萄果實、葡萄種皮及葡萄之種子所成群組之源自葡萄之原料在約30至60v/v%之醇水溶液中進行萃取而獲得之萃取液,或萃取物中所含有者之膠原酶抑制劑亦為本發明之一。
透明質酸酶抑制作用及膠原酶抑制作用可以公知之方法進行評估。例如透明質酸酶抑制作用可經由以實施例中記載之方法測定透明質酸酶之抑制率等進行評估。膠原酶抑制作用可
以例如實施例中記載之方法或前田有美惠等(食品衛生學雜誌31(3),233-237(1990))中記載之方法等進行評估。
於上述透明質酸酶抑制劑及膠原酶抑制劑中,寡聚原花青素之平均聚合度較好在約8以上,更好為約8至10,最好為約8.5至10,又以約9至10最佳。又,於另外之較佳形式,寡聚原花青素之平均聚合度為約8.5至9.5,更好為約8.7至9.5。只要含有平均聚合度在該等範圍之聚合物,則即使寡聚原花青素中有單體、二聚物、其他片段存在,亦可獲得優越之透明質酸酶抑制活性、優越之膠原酶抑制活性,因而較佳。
為本發明透明質酸酶抑制劑及膠原酶抑制劑之有效成分之寡聚原花青素係於上述精製方法中使用至少一種選自由葡萄果實、葡萄種皮及葡萄之種子所成群組之源自葡萄之原料作為原料,於該萃取步驟中將作為萃取溶劑之醇水溶液之醇濃度作成約30至70v/v%獲得之萃取液或萃取物中含有者。較好為將醇水溶液之醇濃度作成約30至60v/v%,更好為約30至50v/v%,最好為約40至50v/v%獲得之萃取液或萃取物中含有之寡聚原花青素。萃取中較佳之條件等係與上述之平均聚合度在約8以上(較好約8至10)之寡聚原花青素之精製方法中之條件等相同。例如,萃取溫度較好為約30至未達100℃,較好為約70至未達100℃,更好為約70至90℃,最好為約70至未達90℃。醇較好為乙醇。源自葡萄之原料及其較佳形式亦與作為上述精製方法之原料使用之源自葡萄之原料及其較佳形式相同。
本發明亦包含將平均聚合度為約8至10之寡聚原花青素作為有效成分之透明質酸酶抑制劑。
本發明亦包含將平均聚合度為約8至10之寡聚原花青素作為有效成分之膠原酶抑制劑。
於本發明,只要含有平均聚合度為約8至10之聚合物,即使寡聚原花青素中有單體、二聚物、其他片段存在,亦獲得優越之透明質酸酶抑制活性、優越之膠原酶抑制活性。平均聚合度為約8至10之寡聚原花青素較好為含於至少一種選自由葡萄果實、葡萄種皮及葡萄之種子所成群組之源自葡萄之原料中者。該等源自葡萄之原料中含有之寡聚原花青素可經由例如上述之精製方法從原料獲得。寡聚原花青素之平均聚合度之下限較好為約8.5。
於上述透明質酸酶抑制劑及膠原酶抑制劑中,寡聚原花青素較好為平均聚合度為約8.5至9.5,更好平均聚合度為約8.7至9.5之寡聚原花青素。又,該等寡聚原花青素較好為將至少一種選自由葡萄果實、葡萄種皮及葡萄之種子所成群組之源自葡萄之原料於約70至未達100℃,於約30至60v/v%之醇水溶液中進行萃取獲得之萃取液或萃取物中所含有者。萃取之較佳形式等係與上述精製方法中之較佳形式等相同。
源自葡萄之原料及其較佳形式係與上述之精製方法中者相同。葡萄較好為赤霞珠。
上述透明質酸酶抑制劑及膠原酶抑制劑除了必須成分之寡聚原花青素之外,亦可含有該等以外之成分。
本發明抑制劑之製造可使用含有以上述萃取步驟獲得之寡聚原花青素之萃取液或萃取物。上述萃取物較好可使用例如從回收之萃取液除去溶劑,進行OPC分之濃縮、乾燥、粉末化
等處理而獲得之萃取液之濃縮物、乾燥物、粉末等處理物等。又,將經由上述萃取步驟獲得之萃取液或其濃縮物等進行上述之超濾等膜處理或吸附層析等各種層析處理,獲得之含有更高純度OPC之畫分亦可作為本發明之萃取物使用。
含有經由上述精製方法獲得之寡聚原花青素之萃取液或萃取物可以原狀作為透明質酸酶抑制劑或膠原酶抑制劑使用,亦可添加其他之成分使用。
上述透明質酸酶抑制劑及膠原酶抑制劑、上述之黑色素生成抑制劑及美白劑係適合作為例如皮膚外用劑等之醫藥品、醫藥部外品、化粧品等使用。
例如將透明質酸酶抑制劑、膠原酶抑制劑、黑色素生成抑制劑及美白劑作為皮膚外用劑等時,劑型等並無特別限制,可作成例如軟膏、乳霜、乳液、化粧水、美容液、敷膜、浴用劑、洗臉料等。將透明質酸酶抑制劑、膠原酶抑制劑、黑色素生成抑制劑及美白劑作成化粧品時,其形態並無特別限制,可作成例如化粧水、乳液、美容液、乳霜、敷膜等之皮膚化粧品;化粧底粧液、化粧底霜、粉狀、液狀或乳霜狀之粉底霜等化粧化粧品;護手霜、腳霜、身體乳液等之身體用化粧品;洗臉料、身體皂等。
上述透明質酸酶抑制劑及膠原酶抑制劑、上述之黑色素生成抑制劑及美白劑例如必要時亦可為含有通常皮膚外用劑或化粧品中配合之成分及/或添加劑之組成物。
配合成分可列舉美白劑、保濕劑、抗氧化劑、油性成分、紫外線吸收劑、界面活性劑、增黏劑、醇類、粉末成分、色劑、水、各種皮膚營養劑、防腐/殺菌劑、收斂劑、香料等之各
種成分、添加劑等,必要時可將該等之1種或2種以上適當配合。可於上述本發明中之有效成分,例如可於上述寡聚原花青素或含有以上述精製方法獲得之寡聚原花青素之萃取液或萃取物中適當配合該等通常使用之成分、添加劑等,根據常法,可獲得皮膚外用劑、化粧品、醫藥部外品等。
以下,記載上述配合成分之一例,惟,本發明之配合成分不只限於以下者,即使為下述以外者,亦可使用醫藥品、化粧品、醫藥部外品等中使用之成分及添加劑。
美白劑可列舉例如熊果苷、麴酸及其衍生物、氫醌衍生物、鞣花酸及其衍生物、傳明酸(Tranexamic acid)及其衍生物、間苯二酚衍生物、半胱胺酸及其衍生物、松樹皮萃取物等、2,5-二羥基苯甲酸衍生物、煙鹼衍生物、α-羥酸等。

於皮膚外用劑及化粧品之配合量,寡聚原花青素較好為約0.000006至1質量%,更好為約0.0000125至0.5質量%,最好為約0.0001至0.01%,又以約0.0005至0.0025質量%最佳。
本發明之透明質酸酶抑制劑及膠原酶抑制劑、上述之黑色素生成抑制劑及美白劑之使用方法係根據使用對象之皮膚狀況等而異,只要為例如以下之方法即可。
亦即,可1日數次(例如約1至5次,較好約1至3次)、適量(例如約0.05至2g)塗抹於皮膚。又,較好將組成物以使寡聚原花青素(較好為平均聚合度為約8至10之寡聚原花青素)之1日使用量為約0.00025至0.25mg,更好為約0.005至0.2mg之方式塗抹。塗抹期限並無特別限定。
本發明之透明質酸酶抑制劑及膠原酶抑制劑係對人體為安全且發揮優越之透明質酸酶抑制或膠原酶抑制效果者。因此,本發明之抑制劑適合用於預防或抑制皺紋、鬆弛用之皮膚外用劑或化粧品、預防或防止老化用之皮膚外用劑或化粧品等,發揮優越之預防或抑制皺紋、鬆弛、預防或防止皮膚老化等之效果。
以下,列舉實施例對本發明作更詳細之說明,惟,本發明不只限於該等例。
實施例中,醇濃度(%)若無特別說明,為體積%(v/v%)。
以下之調製例中,白葡萄酒用葡萄之壓搾物係使用於製造白葡萄酒步驟中獲得之葡萄壓搾物,亦即,從葡萄果實壓搾果汁後之渣滓(葡萄之果皮及種子)。紅葡萄酒用葡萄之壓搾物係使用於製造紅葡萄酒步驟中獲得之葡萄壓搾物,亦即,將葡萄果實破碎後之破碎液(含有果肉、果皮及種子之果汁)發酵後,將該發酵液壓搾獲得之渣滓(果皮及種子)。
在製造白葡萄酒步驟中獲得之夏多內(葡萄品種)之壓搾物(夏多內之果皮及種子250g)中加入10倍量之水作為萃取溶劑,於80℃加熱萃取1小時。將獲得之熱水萃取液經由尼龍篩孔(日本理化學器械(股)公司製造,商品名:NRS-200)粗過濾,獲得萃取液1。又,對於過濾後之殘渣再次以相同條件進行萃取、過濾,將獲得之萃取液2與先前之萃取液1合併後減壓濃縮至400mL。將獲得之濃縮液進行矽藻土過濾,濾液通過填充有丙烯酸系樹脂 安貝利(Amberlite)XAD(商標登記)-7HP(歐加隆(股)公司製造,100mL)之管柱後將水(200mL)通液,除去餘留之糖類及有機酸(將糖類及有機酸溶出)。之後使用70%乙醇水溶液(400mL)作為洗提液,將多酚從管柱中溶出。獲得之溶出液減壓濃縮後供給凍
結乾燥,獲得1.62g之源自白葡萄酒用葡萄之原花青素畫分1。
於上述(1)方法之加熱萃取中除了各自使用10%、20%、30%、50%及70%乙醇水溶液替代水作為萃取溶劑之外,進行與上述(1)相同之操作,將源自白葡葡萄酒用葡萄之原花青素加以萃取及精製。經由此,獲得源自白葡萄酒用葡萄之原花青素畫分2至6,各自分別為2.03g(萃取溶劑10%乙醇水溶液)、2.06g(萃取溶劑20%乙醇水溶液)、2.57g(萃取溶劑30%乙醇水溶液)、2.80g(萃取溶劑50%乙醇水溶液)及2.32g(萃取溶劑70%乙醇水溶液)。
在製造紅葡萄酒步驟中獲得之赤霞珠(葡萄品種)之壓搾物(赤霞珠之果皮及種子30g)中加入10倍量之水作為萃取溶劑,於80℃加熱萃取1小時。將獲得之熱水萃取液經由尼龍篩孔(日本理化學器械(股)公司製造,商品名:NRS-200)粗過濾,獲得萃取液。將獲得之萃取液減壓濃縮至10mL後,以水稀釋至45mL,於冷藏庫靜置3天。接著,將該萃取液供離心分離(3000rpm,5分鐘),將上澄液通過填充有丙烯酸系樹脂 安貝利XAD(商標登記)-7HP(歐加隆(股)公司製造,20mL)之管柱後將水(40mL)通液,除去餘留之糖類及有機酸(將糖類及有機酸溶出)。之後使用70%乙醇水溶液(80mL)作為洗提液,將多酚從管柱中溶出。獲得之溶出液減壓濃縮後凍結乾燥,獲得46mg之源自紅葡萄酒用葡萄之原花青素畫分1。
於上述(1)方法之加熱萃取中,除了各自使用10%、20%、30%、50%、60%及70%乙醇水溶液替代水作為萃取溶劑之外,進行與上述(1)相同之操作,將源自紅葡萄酒用葡萄之原花青素加以萃取及精製。經由此,獲得源自白葡萄酒用葡萄之原花青素畫分2至8,各自分別為58mg(萃取溶劑10%乙醇水溶液)、78mg(萃取溶劑20%乙醇水溶液)、89mg(萃取溶劑30%乙醇水溶液)、97mg(萃取溶劑40%乙醇水溶液)、104mg(萃取溶劑50%乙醇水溶液)、99mg(萃取溶劑60%乙醇水溶液)及76mg(萃取溶劑70%乙醇水溶液)。
為了評估因品種導至之源自葡萄之原花青素畫分之不同,使用以下品種之葡萄壓搾物,將源自葡萄之原花青素加以萃取及精製。
.夏多內之壓搾物(調製例1使用之白葡萄酒用葡萄之壓搾物)
.甲州之壓搾物(甲州之果皮及種子)
.赤霞珠之壓搾物(調製例2使用之紅葡萄酒用葡萄之壓搾物)
.康科特葡萄之壓搾物(康科特葡萄之果皮及種子)
.梅洛之壓搾物(梅洛之果皮及種子)
.貝利A葡萄之壓搾物(貝利A葡萄之果皮及種子)
在葡萄酒製造步驟獲得之葡萄壓搾物夏多內、赤霞珠、康科特葡萄、梅洛、貝利A葡萄及甲州各個之果皮及種子中加入15倍量之水作為萃取溶劑,於80℃加熱萃取1小時,將該熱水萃取液經由尼龍篩孔(日本理化學器械(股)公司製造,商品名:NRS-200)粗過濾,獲得萃取液1。對於過濾後之殘渣再次以相同條件進行萃取、過濾,將獲得之萃取液2與先前之萃取液1合併後進行矽藻土過濾,濾液通過填充有丙烯酸系樹脂 安貝利XAD(商標登記)-7HP(歐加隆(股)公司製造,100mL)之管柱後將水(管柱容積(CV)之2倍量)通液,除去餘留之糖類及有機酸(將糖類及有機酸溶出)。之後使用70%乙醇水溶液(CV之3倍量)作為洗提液,將多酚從管柱中溶出。獲得之溶出液減壓濃縮後供給凍結乾燥,源自葡萄酒用葡萄之原花青素畫分收率各自分別為0.832%(夏多內)、0.833%(赤霞珠)、0.762%(康科特葡萄)、1.075%(梅洛)、0.617%(貝利A葡萄)及1.094%(甲州)。
將調製例3獲得之源自夏多內之原花青素畫分及源自赤霞珠之原花青素畫分各自使用Sephadex LH-20(商品名,Pharmacia fine chemicals公司製造)進行精製及分畫,分成單純多酚畫分、兒茶酸/表兒茶酸畫分、低分子寡聚原花青素畫分及高分子寡聚原花青素畫分4種。移動相依序使用水(CV之3倍量)、20%乙醇水溶液(CV之3倍量)、35%乙醇水溶液(CV之3倍量)、55%乙醇水溶液(CV之3倍量)、75%乙醇水溶液(CV之3倍量)及70%丙酮水溶液(CV之3倍量)。
分別獲得經由20%乙醇水溶液溶出之畫分為單純多
酚畫分,經由35%乙醇水溶液溶出之畫分為兒茶酸/表兒茶酸畫分,經由55%乙醇水溶液溶出之畫分為低分子寡聚原花青素畫分,經由75%乙醇水溶液溶出之畫分作為高分子寡聚原花青素畫分。
評估於上述調製之源自葡萄酒用葡萄之壓搾物之原花青素畫分之方法表示於下。
源自葡萄酒用葡萄之壓搾物之原花青素畫分之寡聚原花青素純度以日本專利第4659407號表示之方法為基準之方法求得。具體而言,以下述之方法求得純度。
首先,在上述調製例調製之源自葡萄酒用葡萄之壓搾物之原花青素畫分各1.0mg中加入0.6N鹽酸/丁醇1.0mL,於90℃進行反應2小時,將寡聚原花青素類分解為花青素。對於獲得之反應溶液以後述之高速液體層析儀之分析條件進行分析,將反應溶液中含有之花青素量定量後經由下述表示之計算式,算出寡聚原花青素純度。又,標準物質使用原花青素B-1(PB-1、船越(股)公司製造:NIU-N210)。

寡聚原花青素之平均聚合度係從上述HPLC之測定
結果使用GPC軟體(島津製作所(股)公司製造),算出數平均分子量,將獲得之數平均分子量除以兒茶酸之分子量(Mw 290)而計算。
含有10%胎牛血清之易格(eagle)最小必須培養基(以下,簡稱為培養基)。
製作試驗濃度200倍之DMSO溶液,試驗時以培養基稀釋200倍,用於試驗。對照組係製作DMSO溶液,陽性對照組係製作10mM熊果苷(Arbutin)之DMSO溶液,任何一種在試驗時均以培養基稀釋200倍,用於試驗。
第1天:將1.5×106個之B16黑色素瘤細胞與5mL之培養基一同接種於60mm×15mm培養皿,於二氧化碳恆溫箱(37℃、CO2分壓5%)培養。又,試驗以n=2進行。
第2天:交換含有試驗試料之培養基,於二氧化碳恆溫箱培養3天。
第5天:與第2天相同,交換含有試驗試料之培養基,於二氧化碳恆溫箱再培養2天。
第7天:將培養之細胞破碎,測定黑色素量及蛋白量。
除去培養基,將細胞用5mL之PBS(-)洗浄。除去洗浄液後加入0.02%EDTA/PBS(-)1mL,於37℃保溫5分鐘後進行吸
量,將細胞懸濁。將細胞懸濁液0.9mL放入2.0mL容量之埃彭道夫管(eppendorf tube)中,離心(5000rpm,1分鐘)後除去上清液。添加PBS(-)0.9mL,以試管震盪器(vortex mixer)將細胞分散,離心(5000rpm,1分鐘)後除去上清液。添加50mM磷酸緩衝液(pH6.8)0.45mL,以試管震盪器將細胞分散後用超音波破碎裝置(Cosmobio公司製造,型號UCD-250HSA),在冰冷條件下將細胞破碎。
破碎條件:輸出功率250W 30秒開/30秒關/共5分鐘
將細胞破碎液400μL放入2.0mL容量之埃彭道夫管中,添加等量之4N氫氧化鈉,於60℃加熱2小時,進行黑色素之溶出處理。將處理液在96洞微盤3個洞中每個添加200μL,測定492nm之吸光度,尋求平均值。各試料均以n=2進行試驗,另尋求該等之平均值,從預先使用黑色素(Nacalai科技公司製造)作成之黑色素校正曲線算出黑色素量。
將細胞破碎液35μL以等量之50mM磷酸緩衝液(pH6.8)稀釋,在96洞微盤3個洞中每個添加5μL,使用DC蛋白質化驗盒II(Bio/Rad公司製造)進行反應,測定690nm之吸光度,尋求平均值。各試料均以n=2進行試驗,另尋求該等之平均值,從預先使用試驗盒之牛血清白蛋白作成之蛋白校正曲線算出蛋白量。
將對照組(DMSO)之黑色素量、蛋白量作為100,算
出空白、熊果苷(陽性對照)及各試料之黑色素量及蛋白量之比。另,以對照組(DMSO)之(黑色素量/蛋白量)比作為100,算出熊果苷(陽性對照)及試料之(黑色素量/蛋白量)比,從100扣除該值,尋求熊果苷(陽性對照)及試料之黑色素生成抑制率。又,於試驗例1、3及4中,將試料濃度以公比2倍稀釋,尋求抑制率,從各試驗濃度之黑色素生成抑制率經由內插法尋求抑制50%之試料濃度「IC50(μg/mL)」。「IC50(μg/mL)」表示抑制50%黑色素生成必需之濃度,呈現更低值之化合物即作為抑制劑之活性更高。
源自葡萄酒用葡萄之寡聚原花青素之透明質酸酶抑制試驗為改變文獻(前田有美惠等,食品衛生學雜誌31(3),233-237(1990))之方法,以下述之方法實施。透明質酸經由透明質酸酶分解為N-乙醯基己糖胺。藉由對-二甲胺基苯甲醛(和光純藥工業(股)公司製造,以下,簡稱為p-DAB)標識之發色,將還原末端之N-乙醯基葡糖胺以吸光度定量,測定透明質酸酶抑制活性。
在溶解於10%DMSO之試料溶液40μL中將溶解於0.1M乙酸緩衝液(pH4.0)之1000U/mL透明質酸酶(Sigma公司製造)20μL混合,於37℃預備加溫20分鐘。添加溶解於相同緩衝液之0.5mg/mL化合物48/80(Sigma公司製造)40μL,於37℃靜置20分鐘,將透明質酸酶活化。於該溶液中添加0.8mg/mL透明質酸鉀溶液100μL使最終濃度為0.4mg/mL,於37℃進行反應40分鐘後添加0.4N氫氧化鈉溶液40μL,冰冷使反應停止。在反應液中加入以6N氫氧化鈉將pH值調整為9.1之0.8M硼酸溶液40μL,於100℃煮沸3分鐘。在冰中冷卻至室溫,於此,添加遮光之10mg/mL
對-二甲胺基苯甲醛(p-DAB)溶液1.2mL,於37℃進行反應20分鐘後測定585nm之吸光度(A585)。試料之透明質酸酶抑制活性以經由下述式求得之抑制率表示。

於比較對照,使用400μg/mL(莫耳濃度0.873mmM)之(-)-表兒茶酸-3-O-沒食子酸酯(和光純藥(股)公司製造,以下,簡稱為EGCG)40μL替代試料溶液。
源自葡萄酒用葡萄之寡聚原花青素之膠原酶抑制試驗係改變文獻(Wunsch人等,Hoppe Seylers Z Physiol Chem.,333,149-51(1963))之方法,以下述之方法實施。
酵素溶液為將膠原酶TypeIV(Sigma公司製造)10mg溶解於蒸餾水1mL使成為100μg/mL(55.1unit/mL)之濃度,於使用時稀釋50倍使用。基質溶液係溶解於使PZ-肽(4-苯基偶氮基-苯甲氧基羰基-Pro-Leu-Gly-Pro-D-Arg-OH)(Pz-Pro-Leu-Gly-Pro-D-Arg-OH,Sigma公司製造)之濃度成為0.50μg/Ml之含有20nmol/L氯化鈣之Tris鹽酸緩衝液(pH7.1)使用。將酵素溶液20μL及基質溶液160μL在溶解於10%DMSO之試料溶液20μL混合,於37℃進行反應30分鐘。接著,加入25mM檸檬酸溶液400μL,將反應停止,以乙酸乙酯2mL萃取反應液中之Pz-Pro-Leu。將獲得之
乙酸乙酯層波長320nm之吸光度對照乙酸乙酯進行測定。又,各試料之抑制活性以經由下述式求得之抑制率算出。
膠原酶抑制率(%)={1-(a-b)/(c-d)}×100
a:添加試料時,反應30分鐘後之吸光度
b:添加試料時,反應0分鐘後之吸光度
c:未添加試料,反應30分鐘後之吸光度

於上述式中,膠原酶之活性完全被抑制時,膠原酶抑制率(%)為100%。亦即呈現高「抑制率(%)」之化合物作為抑制劑之活性更高。
於膠原酶抑制試驗,陽性對照使用添加800μg/mL(莫耳濃度0.176mM)之異戊基膦醯基-甘胺醯-L-脯胺醯-L-丙胺酸二鉀鹽(Isoamylphosphonyl-Glycyl-L-Proly-L-Alaine,dipotassium salt;C15H26K2N3O6P(Elastin Products Company,Inc.製造,以下,簡稱為IP304)20μL(反應時之濃度為80μg/mL)替代試料溶液。
對調製例1調製之源自白葡萄酒用葡萄之原花青素畫分1至6,根據上述評估方法評估效能等時獲得表1表示之結果。
可知以30%至50%含水乙醇水溶液萃取出之源自白葡萄酒用葡萄之原花青素畫分具有甚高之黑色素生成抑制作用。其活性認為與所含寡聚原花青素之純度及其聚合度有關。又可知若以30%至50%含水乙醇水溶液進行萃取,則原料中所含之高聚合度寡聚原花青素可以高收率且高純度萃取。
對調製例2調製之源自紅葡萄酒用葡萄之原花青素畫分1至8,根據上述評估方法評估效能等,獲得表2表示之結果。
可知以30%至60%含水乙醇水溶液萃取出之源自紅葡萄酒用葡萄之原花青素畫分具有甚高之黑色素生成抑制效果及透明質酸酶抑制作用。又可知若以30%至60%含水乙醇水溶液進行萃取,則原料中所含之高聚合度寡聚原花青素可以高收率且高純度萃取。
對調製例3調製之源自葡萄酒用葡萄之原花青素畫分,根據上述評估方法評估效能等,獲得表3表示之結果。
此次進行研究之源自各種葡萄之原花青素畫分之黑色素生成抑制及透明質酸酶抑制活性與寡聚原花青素之純度或聚合度成比例。其中,已知從夏多內、赤霞珠及甲州獲得之源自葡萄之原花青素畫分可獲得甚高的美容效果。
對調製例4調製之源自葡萄酒用葡萄之原花青素畫分,根據上述評估方法評估效能等,獲得表4及表5表示之結果。源自夏多內之原花青素畫分之結果表示於表4,源自赤霞珠之原
花青素畫分之結果表示於表5。表4及表5之移行率為以調製例4調製之源自葡萄酒用葡萄之原花青素畫分中所含之單純苯酚畫分量、兒茶酸/表兒茶酸畫分量、低分子OPC畫分量及高分子OPC畫分量之合計作為100時,從單純苯酚畫分量、兒茶酸/表兒茶酸畫分量、低分子OPC畫分量及高分子OPC畫分量計算之各個畫分之比率。例如單純苯酚畫分之移行率係經由以下之公式計算。下述式中,「兒茶酸/表兒茶酸畫分」簡稱為「兒茶酸畫分」。
從試驗例1至4,對於源自葡萄之原花青素畫分之分畫物進行黑色素生成抑制效果及透明質酸酶抑制之評估時,於任一種高分子寡聚原花青素畫分均確認效果甚高。可知要獲得高美容效能,即使於寡聚原花青素中,具有高聚合度之萃取物亦為必要。根據上述之研究,明瞭以30v/v%至60v/v%,較好30v/v%至50v/v%之含水乙醇水溶液萃取出之源自葡萄之原花青素畫分含有豐富之具有高聚合度之寡聚原花青素,可知要獲得高純度之具有高美容效果之寡聚原花青素,萃取之溶劑是重要的。又,尤其是若使用30至60%之乙醇水溶液,可效率佳的萃取寡聚原花青素且獲得含有許多高聚合度之寡聚原花青素之萃取物。
將調製例2調製之源自紅葡萄酒用葡萄之原花青素畫分1至8,以800μg/mL之10%DMSO溶液調整,經由上述(5)膠原酶抑制試驗之評估方法評估各畫分之膠原酶抑制活性,獲得表6表示之結果。表6表示之膠原酶抑制率為從上述膠原酶抑制率之計算式求得之值。陽性對照(IP304)之膠原酶抑制率(從上述膠原酶抑制率之計算式求得之值)為71.1%。
將調製例3調製之熱水萃取之源自葡萄酒用葡萄之原花青素畫分,以800μg/mL之10%DMSO溶液調整,經由上述(5)膠原酶抑制試驗之評估方法評估各畫分之膠原酶抑制活性,獲得表7表示之結果。表7表示之膠原酶抑制率為從上述膠原酶抑制率之計算式求得之值。陽性對照(IP304)之膠原酶抑制率(從上述膠原酶抑制率之計算式求得之值)為84.0%。
將調製例4調製之源自赤霞珠之原花青素畫分,以800μg/mL之10%DMSO溶液調整,經由上述(5)膠原酶抑制試驗之評估方法評估各畫分之膠原酶抑制活性,獲得表8表示之結果。表8表示之膠原酶抑制率為從上述膠原酶抑制率之計算式求得之值。陽性對照(IP304)之膠原酶抑制率(從上述膠原酶抑制率之計算式求得之值)為84.6%。表8之移行率與表5中之移行率相同。
從試驗例5至7對於源自葡萄之原花青素畫分之分畫物進行膠原酶抑制之評估時,任何一種在高分子寡聚原花青素畫分確認高效果。因此明瞭寡聚原花青素中具有高聚合度之萃取物具有高美容效果。例如以30v/v%至60v/v%,較好30v/v%至50v/v%之含水乙醇水溶液萃取出之源自葡萄之原花青素畫分,含有豐富之具有高聚合度之寡聚原花青素,顯示甚高之膠原酶抑制活性。
根據本發明可以高純度精製在飲食品、化粧品或醫藥品等領域有用之寡聚原花青素。例如,經由本發明精製方法獲得之寡聚原花青素具有優越之美白作用、透明質酸酶抑制作用及膠原酶抑制作用,在化粧品、醫藥品等領域有用。
Claims (16)
- 一種寡聚原花青素之精製方法,其特徵為:包含將含有不同聚合度之複數種寡聚原花青素之原料,以醇水溶液作為萃取溶劑進行萃取之萃取步驟;其中,該原料為至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料,於該萃取步驟中,設定萃取溫度為70至未達100℃,並設定該萃取溶劑之醇濃度為30至60v/v%,以萃取平均聚合度為8以上之寡聚原花青素。
- 如申請專利範圍第1項所述之精製方法,其中,該醇為乙醇者。
- 如申請專利範圍第1項或第2項所述之精製方法,其中,該精製方法為平均聚合度為8至10之寡聚原花青素之精製方法,而萃取溶劑為40至50v/v%之乙醇水溶液者。
- 一種具有黑色素生成抑制作用之寡聚原花青素之精製方法,其特徵為包含將至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料,於70至未達100℃,在30至60v/v%之醇水溶液中,萃取平均聚合度為8以上之寡聚原花青素之萃取步驟。
- 一種透明質酸酶抑制劑,係將平均聚合度為8以上之寡聚原花青素作為有效成分,該寡聚原花青素係將至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料,於70至未達100℃,在30至60v/v%之醇水溶液中進行萃取而得之萃取液或萃取物中所含有者。

- 如申請專利範圍第5項或第6項所述之抑制劑,其中,該醇為乙醇者。
- 如申請專利範圍第5項或第6項所述之抑制劑,其中,該醇水溶液為40至50v/v%之乙醇水溶液者。
- 如申請專利範圍第5項或第6項所述之抑制劑,其中,該寡聚原花青素之平均聚合度為8至10者。
- 一種透明質酸酶抑制劑,係將至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料中所含有的平均聚合度為8至10之寡聚原花青素作為有效成分。
- 一種膠原酶抑制劑,係將至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料中所含有的平均聚合度為8至10之寡聚原花青素作為有效成分。
- 如申請專利範圍第10項或第11項所述之抑制劑,其中,該寡聚原花青素之平均聚合度為8.5至9.5者。
- 如申請專利範圍第10項或第11項所述之抑制劑,其中,該寡聚原花青素係將至少1種自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料,於70至未達100℃,在30至60v/v%之醇水溶液中進行萃取而得之萃取液或萃取物中所含有者。
- 如申請專利範圍第10項或第11項所述之抑制劑,其中,該葡萄為赤霞珠(Cabernet Sauvignon)者。
- 如申請專利範圍第10項或第11項所述之抑制劑,其中,該抑制劑為皮膚外用劑或化粧品者。
- 一種寡聚原花青素聚合度之調整方法,其特徵為包含將含有不同聚合度之複數種寡聚原花青素之原料,以醇水溶液作為萃取溶劑進行萃取之萃取步驟;其中,該原料為至少1種選自由葡萄果實、葡萄種皮及葡萄種子所成群組之源自葡萄之原料,於該萃取步驟中,設定萃取溫度為70至未達100℃,並設定該萃取溶劑之醇濃度為30至60v/v%,以萃取平均聚合度為8以上之寡聚原花青素。
