Claims (20)

1. Способ увеличения экспрессии рекомбинантного белка в культуре клеток, включающий культивирование клеток, которые экспрессируют рекомбинантный белок, в безбелковой среде, которая не содержит олигопептиды, причем среда содержит аминокислоты, витамины, органические и неорганические соли и источников углеводов и дополнена по меньшей мере 2 мг/л путресцина.2HCl.
2. Способ по п.1, где среда дополнена L-глутамином, аскорбиновой кислотой, этаноламином, селенитом натрия и неионным поверхностно-активным веществом.
3. Способ по п.1 или 2, где среда дополнена Fe(II) или Cu(II) в количестве, большем, чем количество в основной DMEM/HAM's F12 (1:1).
4. Способ по любому из предшествующих пунктов, где среда дополнена L-аспарагином, L-цистеином, L-цистином, L-пролином и L-триптофаном.
5. Способ по любому из предшествующих пунктов, где среда содержит путресцин в концентрации от примерно 1,0 мг/л до примерно 20 мг/л.
6. Способ по любому из предшествующих пунктов, где среда дополнительно содержит один или более из группы, состоящей из кадаверина, спермидина, спермина, агматина и орнитина.
7. Способ по любому из предшествующих пунктов, где среда дополнительно содержит орнитин, спермин или их комбинацию.
8. Способ по любому из предшествующих пунктов, где среда содержит буферное вещество.
9. Способ по п.8, где буферное вещество выбрано из одного или нескольких из бикарбонатов натрия, антиоксидантов, стабилизаторов или ингибиторов протеазы.
10. Способ по любому из предшествующих пунктов, где рекомбинантный белок выбран из группы, состоящей из фактора свертывания крови VII, фактора свертывания крови VIII, фактора свертывания крови IX, vWF, ADAMTS13 и фурина.
11. Способ по п.10, где рекомбинантный белок представляет собой фактор свертывания крови VIII.
12. Способ по п.10, где рекомбинантный белок представляет собой vWF.
13. Способ по любому из предшествующих пунктов, где клетки выбраны из группы, состоящей из клеток млекопитающих, клеток насекомых, клеток птиц, бактериальных клеток и дрожжевых клеток.
14. Способ по любому из предшествующих пунктов, где клетки представляют собой клетки СНО, клетки 293 или клетки BHK.
15. Способ по любому из предшествующих пунктов, где комбинация клетка/белок выбрана из группы, состоящей из клетки СНО/фактор свертывания крови VIII, клетки СНО/фактор свертывания крови VII, клетки CHO/ADAMTS13, клетки СНО/фурин и клетки 293/фактор свертывания крови IX.
16. Способ по п.15, где комбинация клетка/белок представляет собой клетки CHO/фактор свертывания крови VIII.
17. Способ по любому из предшествующих пунктов, где путресцин получен синтетическим путем.
18. Способ по любому из предшествующих пунктов, где путресцин происходит от источника, отличного от гидролизата белка.
19. Способ по любому из предшествующих пунктов, где среда химически определена.
20. Способ по любому из предшествующих пунктов, где клетки культивируют способом, выбранным из группы, состоящей из периодического культивирования, периодического культивирования с подпиткой, перфузионного культивирования и хемостатического культивирования.
