Description
Przedmiotem wynalazku jest bakteryjny srodek owadobójczy, który jako substancje czynna zawiera Bacillus thuringiensis patotyp C, zwlaszcza Bacillus thuringiensis var. tenebrionis, albo otrzymana z niego dzialajaca owadobójczo toksyne.2 145 191 Nowa odmiane Btt wyodrebniono z chorych stadiów rozwoju pospolitego maczniaka mly¬ narka (Tenebriomolitor L., Coleoptera: Tenebrionidae).Czysta kulture przechowuje sie w tempe¬ raturze 25-30°C na pozywce agarowej w skosnych rurkach. Suche zarodniki zamrazano w celu zakonserwowania typu. Oznaczanie prowadzono na podstawie danych morfologicznych i biochemicznych.Wyodrebniona bakteria produkuje na pozywce agarowej duze, nierówne, bialawe kolonie, których komórki zarodnikuja po 18-24 godzinach. Wegetatywna komórka jest zaopatrzona w rzeski, ma postac laseczki o wymiarach okolo 1,0 X 4-8/im. Reaguje ona gramo-dodatnio. Zarod¬ nikowa komórkamacierzysta obok przykrancowo ulozonego elipsoidalnego zarodnika (0,8 X 1,4- -1,6 /im) zawiera parasporalny krysztal o plaskiej postaci typu plytki i o konturze równolegloboku, rombu albo kwadratu (dlugosc krawedzi okolo 0,8-1,5 /im).W fazie wegetatywnej nastepuje wzrost Bacisllus fakultativ takze beztlenowo. Zarodnikowa¬ nie jest natomiast scisle tlenowe. Optymalna temperatura do wzrostu wegetatywnych komórek wynosi 25-30°C. Trzymanie w autoklawie przez 15 minut w temperaturze 121°C, co odpowiada nadcisnieniu 102kPa, dezaktywuje calkowicie zarodniki i równiez krystaliczna toksyne. Przez naswietlanie promieniami nadfioletowymi (UV) przy 254 na zarodniki w odpowiedniej dozie ulegaja dezaktywacji, natomiast krystaliczna toksyna nie ulega dezaktywacji.Biochemiczne sprawnosci wegetatywnych komórek sa nastepujace. Ze zdolnych do rozkladu weglowodorów, jak np. glikozy, tworzy sie kwas fermentacyjny i acetylometylokarbinol, a nie tworzy sie gaz. Przy zakwaszeniu, oprócz glikozy, rozklada sie mannoza i sacharoza, natomiast nie rozkladaja sie laktoza, celobioza, salicyna, arabinoza, ksyloza albo mannit. Eskulina ulega hydro¬ lizie, tak samo skrobia. Mozna wykryc nastepujace enzymy, takie jak katalaza, reduktaza azota¬ nowa dihydrolaza argininy i proteinaza, natomiast nie mozna bylo wykazac dekarboksylazy lizyny, dezaminazy fenyloalaniny, ureazy i lecytynazy.Do celów produkcyjnych na skale laboratoryjna stosowano wytrzasarke inkubacyjna. W kolbie Erlenmyer'a o pojemnosci 250 ml poddano fermentacji 50 ml srodowiska. Srodowiskomialo nastepujacy sklad: wyciag z drozdzy 0,5%, trypton 0,5%, glikoza 0,1%, KH2PO4 0,08% w wodo¬ ciagowej wodzie pH 7,0. Jako temperature inkubacji stosowano 25-30°C. Po zarodnikowaniu kultury, które mozna kontrolowacmikroskopowo, odwirowano biomase i przemyto ja woda albo roztworem buforowym.Dzialanie Btt na larwy Coleoptera nastepuje wedlug schematu, któryjest znany z patotypu A: w ciagu kilku godzin nastepuje zahamowanie wyzerki, a po kilku dniach, zaleznie od dawki, umieralnosci. Dzialanie toksyczne polega na tym, ze nablonek srodkowego jelita zostaje trwale uszkodzony przez rozpuszczona w soku jelitowym krystaliczna toksyne. Uszkodzone komórki jelita rozluzniaja sie wzajemnie oraz od blony podstawowej i kieruja sie do swiatla jelita. Na skutek zerwania sie bariery jelitowej komórki bakteryjne moga wnikac do hemocelu i tam dokonczyc proces dezyntegracji przez smiertelna posocznice.Nowa odmiana Btt, patotyp C, której prototyp mozna bylo wyodrebnic z martwej poczwarki maczniaka mlynarka, zostala zdeponowana w miemieckim zbiorze mikroorganizmów pod nume¬ rem DSM 2803. Wedlug badania serologicznego var. tenebrionis, szczep DSM 2803, nalezy do serotypu H 82, 8b.Nowa odmiana Btt, patotyp C, wykazuje selektywne dzialanie owadobójcze na Coleoptera, na przyklad na hurmaka olszowca albo stonke ziemniaczana. Dzialanie w przypadku larw jest znacznie silniejsze nizw w przypadku dojrzalych owadów.Nie mozna zaobserwowac zadnego dzialania przeciwko owadom luskoskrzydlym, na przy¬ klad przeciwko molikowi macznemu (Ephestia Kiihniella Zell.), molikowi kapusty (Plitella xylo- stella L.) i takze nie przeciwko owadom dwuskrzydlym, na przyklad komarowi przenoszacemu zólta febre (Aedes aegypti L.). W nizej podanej tablicy zebrane sa wyniki przeprowadzonych biotestów, a mianowicie toksyczne dzialanie Btt na rózne chrzaszcze, owady luskoskrzydle wzgled¬ nie owady dwuskrzydle.Wlasciwa podstawa dzialania owadobójczego w przypadku Bacillus thuringiensis jest toksyna krystaliczna.145 191 Tabela Badany zwiazek Hurmak olszowiec Stonka ziemniaczana Molik maczny Molik kapustny Komar przenoszacy zólta ferfoc Stosowana dawka (równowazniki zarodników) 106/cm2 Kf/cm2 2,5X10P/cm3 4X107/cm2 5X10e/cm3 Dzialanie toksyczne + + — — — W celu zastosowania jako srodka owadobójczego Bacillus thuringiensis var. tenebrionis albo otrzymywany z niego, dzialajacy owadobójczo preparat w znany sposób zadaje sie stosowanymi zwykle dodatkami, np. nosnikami, srodkami zwiekszajacymi przyczepnosc, srodkami zwilzaja¬ cymi itd. i przeprowadza w odpowiednia postac uzytkowa.Takspreparowany srodek owadobójczy mozna stosowac wpostaciproszku zawiesinowego, zawie¬ siny, jako granulat albo inne. Korzystnajest oczyszczona, zarodnikujacakultura w postaci zawie¬ siny z dodatkiem srodka zwilzajacego, na przyklad 0,1% CitowetR (firmy BASF). Na hektar przeznaczonego do traktowania pola rozpyla sie 1012 —1015, zwlaszcza 1013 —1014 zarownikówi równowazna ilosc toksycznych krysztalów. W ten sposób udaje sie ochrona, na przyklad pola ziemniaczanego 2-4 tygodni przed stonka ziemniaczana. W celu ochrony podczas calkowitej fazy wzrostu nalezy stosowac dalsze traktowania w wymienionym odstepie czasu. Nie zaobserwowano dzialania przeciwko niedocelowym organizmom,jak na przyklad przeciw organizmom owadozer- nym i pszczolom modnym.Zastrzezenia patentowe 1. Srodek owadobójczy, zawierajacy substancje czynna i znane dubstancje dodatkowe, zna¬ mienny tym, ze jako substancje czynna zawiera Bacillus thuringiensis, patotyp C albo otrzymana z niego toksyne. 2.
