Description
SAMMENDRAG AV OPPFINNELSEN
Den foreliggende oppfinnelsen tilveiebringer nye forbindelser som er modulatorer av metobotropiske glutamatreseptorer - undertype 5 ("mGluR5") som kan brukes for behandling av lidelser i det sentrale nervesystemet så vel som andre lidelser som er modulert av mGluR5-reseptorer.
BAKGRUNN FOR OPPFINNELSEN
Glutamat, den viktigste aminosyretransmitteren i det sentrale nervesystemet (CNS) hos pattedyr, kontrollerer eller styrer eksitatorisk synaptisk nevrotransmisjon gjennom aktiveringen av ionotropiske glutamatreseptorer reseptorkanaler (iGluRer, det vil si NMDA, AMPA og kainat) og metabotropiske glutamatreseptorer (mGluRer). iGluRer er ansvarlige for rask eksitatorisk transmisjon (Nakanishi S. et al., (1998) Brain Res. Rev., 26: 230-235) mens mGluRer har en mer modulerende rolle som bidrar til finjusteringen av den synaptiske effekten. Glutamat er assosiert med mange fysiologiske funksjoner så som langtids forsterkning (LTP), en prosess man antar underbygger læring og hukommelse, men også kardiovaskulær regulering, sensorisk persepsjon og utvikling av synaptisk plastisitet. I tillegg spiller glutamat en viktig rolle i patofysiologien i forskjellige nevrologiske og psykiatriske sykdommer, spesielt når det foreligger en ubalanse i den glutamatergiske nevrotransmisj onen.
mGluRer er syv transmembran G-proteinkoblede reseptorer. De åtte reseptorene som hører til denne familien er klassifisert i tre grupper (gruppene I, II & III) etter sin sekvenshomologi og farmakologiske egenskaper (Schoepp D.D. et al. (1999) Neuropharmacology, 38: 1431-1476). Aktivering av mGluRer fører til en rekke forskjellige intracellulære responser og aktivering av forskjellige overføringskaskader. Blant mGluR-reseptorene er mGluR5-undertypen av spesiell interesse for en motbalanse av underskuddet eller overskuddet av nevrotransmisjon i nevropsykiatriske sykdommer. mGluR5 hører til gruppe I og dens aktivering starter cellulære responser gjennom G-proteinmedierte mekanismer. mGluR5 er koblet til fosfolipase C og stimulerer fosfoinositidhydrolyse og intracellulær kalsiummobilisering.
Det er blitt vist at mGluR-proteiner er lokalisert i postsynaptiske elementer som ligger inntil den postsynaptiske tetthetten (Lujan R. et al. (1996) Eur. J. Neurosci. 8: 1488-500; Lujan R. et al. (1997) J. Chem. Neuroanat, 13: 219-41) og er også påvist i presynaptiske elementer (Romano C. et al. (1995) J. Comp. Neurol. 355: 455-69). mGluR5-reseptorer kan derfor modifisere de postsynaptiske responser til en nevrotransmitter eller kan regulere nevrotransmitterfrigjøring.
I sentralnervesystemet foreligger det tallrike mGluR5-reseptorer, i alt vesentlig i hjernebarken, hippocampus, kaudat-putamen og kjerneakkumbens. Ettersom disse hj erneområdene har vist seg å være involvert i følelser, motiverende prosesser og i tallrike aspekter av den kognitive funksjonen, så er det predikert at mGluR5-modulatorer vil kunne være av terapeutisk interesse.
En rekke kliniske indikasjoner er blitt foreslått som mål for utviklingen av undertype selektive mGluR-modulatorer. Disse inkluderer epilepsi, nevropatisk og inflammatorisk smerte, tallrike psykiatriske lidelser (for eksempel angst og schizofreni), bevegelseslidelser (for eksempel Parkinsons sykdom) og nevrobeskyttel se (slag og hodeskade), migrene og misbruk/medikamentavhengighet (for oversikt, se Brauner-Osborne H. et al. (2000) J. Med. Chem. 43: 2609-45; Bordi F. og Ugolini A. (1999) Prog. Neurobiel. 59: 55-79; Spooren W. et al. (2003) Behay. Pharmacol. 14: 257-77).
mGluR5-reseptorer er ansett som et mulig medikamentmål for behandling av psykiatriske og nevrologiske lidelser så som angstlidelser, oppmerksomhetslidelser, spiseforstyrrelser, sinnsstemningslidelser, psykotiske lidelser, kognitive lidelser, personlighetsforstyrrelser og stoffmisbruksrelaterte lidelser.
Annen forskning indikerer at mGluR5-modulering spiller en rolle ved behandlingen av såkalt fragilt X-syndrom (Laura N. Antar et al. The Journal of Neuroscience, 17. mars 2004, 24-11, 2648-2655, Weiler I.J., Proe. Nati. Acad. Sei. USA, 1997, 94, 5395-5400), overvekt og gastro-spiserørsreflukssykdom (Blackshaw L.A. et al., presentasjon ved konferansen om nevrogastroenterologi & mortalitet, Madison, Wisconsin, 14. november 2001).
De internasjonale patentpublikasjonene WO03/104206, GB2124227, WO03/050087 og WO03/013247 beskriver 3-fenoksyprop-l-ynyl og 3-pyridinoksyprop-l-ynyl med herbicidale egenskaper. I U56166060 er 4-(5-fenylpent-l-ynyl)-lH-imidazol beskrevet som en lb-histaminantagonist.
De internasjonale patentpublikasj onene WO99/02497, WO01/16121 og WO02/46166 beskriver heteroaryletynylforbindelser og deres anvendelse som metabotropiske glutamatreseptorerantagonister. De internasjonale patentpublikasj onene WO2005/044265, WO2005/044266 og WO2005/044267 beskriver en klasse av 3-(pyridin-2-yl)prop-2-ynylderivativer som kan brukes ved GERD-indikasj on.
Forbindelser med den generelle formel I har sterk aktivitet og selektivitet med hensyn til mGluR5-reseptoren. Forbindelsene ifølge den foreliggende oppfinnelsen kan oppvise fordelaktige egenskaper fremfor tidligere kjente forbindelser. Forbedringer er blitt observert med hensyn til en eller flere av de følgende egenskaper ved oppfinnelsens forbindelser: Styrken på målet, selektiviteten for målet, løseligheten, biotilgjengeligheten, hjerneinntrengningen og aktiviteten i atferdsmodeller på psykiatriske og nevrologiske lidelser.
DETALJERT BESKRIVELSE AV OPPFINNELSEN
Forbindelser og sammensetninger i henhold til oppfinnelsen, samt deres anvendelse i fremstilling av et medikament for behandling eller forebygging av medisinske tilstander er definert i kravene.
Den foreliggende oppfinnelsen inkluderer begge de mulige stereoisomerene og inkluderer ikke bare rasemiske forbindelser men også de individuelle enantiomerene.
Definisjoner av begreper
Det er i det etterfølgende gitt definisjoner på de forskjellige begreper som er brukt i beskrivelsen og kravene for å beskrive den foreliggende oppfinnelsen.
For å unngå tvil, er det underforstått at i den foreliggende beskrivelsen betyr "Ci-C6<M>en karbongruppe med 1, 2, 3, 4, 5 eller 6 karbonatomer. "Co-C6<M>betyr en karbongruppe med 0, 1, 2, 3, 4, 5 eller 6 karbonatomer.

Slike kondenserte ringsystemer kan inkludere en ring som helt eller delvis er umettet så som en benzenring hvorved det dannes kondenserte ringsystemer så som benzokondenserte karbosykler. Sykloalkyl inkluderer slike kondenserte ringsystemer som for eksempel spirokondenserte ringsystemer. Eksempler på sykloalkyl inkluderer syklopropyl, syklobutyl, syklopentyl, sykloheksyl, dekahydronaftalen, adamantan, indanyl, fluorenyl, 1,2,3,4-tetrahydronaftalen og lignende. Begrepet "(C3-C7)sykloalkyl" kan for eksempel være syklopropyl, syklobutyl, syklopentyl, sykloheksyl, sykloheptyl og lignende.
"Solvat" refererer seg til et kompleks med variabel støkiometri dannet av et oppløst stoff (for eksempel en forbindelse med formel I) og et løsemiddel. Det er foretrukket at løsemiddelet er et farmasøytisk akseptabelt løsemiddel så som vann og at løsemiddelet ikke påvirker den biologiske aktiviteten til det oppløste stoffet.
Foretrukne forbindelser ifølge den foreliggende oppfinnelsen er: 2-(4-(pyridin-2-yl)but-3-ynyl)-imidazo[l,2-a]pyridin

Den foreliggende oppfinnelsen angår de farmasøytisk akseptable syreaddisjonssaltene av forbindelsene med formel (I) eller farmasøytisk akseptable bærere eller eksipienter.
Den foreliggende oppfinnelsen angår en fremgangsmåte som kan brukes for å behandle eller forebygge lidelser i det perifere eller sentrale nervesystemet som definert i kravene.
Den foreliggende oppfinnelsen angår farmasøytiske sammensetninger som kan
tilveiebringe fra omtrent 0,01 til 1000 mg av den aktive bestanddelen pr enhetsdose. Sammensetningene kan administreres på enhver egnet måte. For eksempel kan dette skje oralt i form av kapsler osv., parenteralt i form av løsninger for injeksjon, topisk
i form av salver eller hudvann, okulært i form av øyedråper, rektalt i form av stikkpiller, intranasalt eller transkutant i form av leveringssystemer så som plastere.
De farmasøytiske formuleringene ifølge oppfinnelsen kan fremstilles ved hjelp av kjente fremgangsmåter og typen av den farmasøytiske sammensetningen som anvendes vil være avhengig av den forønskede administrasjonsveien. Den totale daglige dosen vil vanligvis variere fra omtrent 0,05 til 2000 mg.
SYNTESEMETODER
Forbindelser med den generelle formel I kan fremstilles ved fremgangsmåter som er velkjente innenfor organisk synteseteknikk og som delvis er angitt i de følgende synteseskjemaer. I alle de skjemaer som er beskrevet i det etterfølgende så er det underforstått at det kan brukes beskyttende grupper for følsomme eller reaktive grupper når dette er nødvendig, noe som skjer i overensstemmelse med generelle prinsipper innen den organiske kjemien. Beskyttende grupper kan manipuleres eller endres ifølge standardmetoder innenfor den organiske synteseteknikk (Green T.W. og Wuts P.G.M. (1991) Protecting Groups in Organic Synthesis, John Wiley et Sons). Disse gruppene kan fjernes på ethvert hensiktsmessig trinn av syntesen ved å bruke fremgangsmåter som tør være kjent for personer med faglig innsikt innenfor organisk kjemi. mGluR5- bindingsprøve
Aktiviteten til forbindelser ifølge den foreliggende oppfinnelsen ble undersøkt ved å bruke en radioligandbindende teknikk ved å bruke hele rottehjerner og tri ti ert 2-metyl-6-(fenyletynyl)pyridin ([<3>H]-MPEP) som en ligand ved å bruke lignende fremgangsmåter til det som er beskrevet i F. Gasparini et al. Bioorg. Med. Chem. Lett. 2002, 12, 407-409 og i J. F. Anderson et al. J. Pharmacol. Exp. Ther. 2002, 303, 3, 1044-1051.
• Membranfremstilling:
Hjernebarken ble dissekert ut fra hjernene til 200-300 g Sprague-Dawley-rotter (Charles River Laboratories, L'Arbresle, Frankrike). Vevet ble homogenisert i 10 volumer (vol/vekt) av iskald 50 mM Hepes-NaOH (pH 7,4) ved å bruke et Polytron homogeniseringsapparat (Kinematica AG, Luzern, Sveits) og så sentrifugert i 30 minutter ved 40000 g (4°C). Supernatanten ble kastet og pelleten ble vasket to ganger med fornyet suspensjon i 10 volumer 50 mM HEPES-NaOH. Membranene ble så oppsamlet ved sentrifugering og vasket før en avsluttende suspensjon på nytt i 10 volumer av 20 mM HEPES-NaOH, pH 7,4. Proteinkonsentrasjonen ble bestemt ved hjelp av Bradfordmetoden (Bio-Rad proteinprøve, Reinach, Sveits) med storfeserumalbumin som standard.
[<3>H]-MPEP-bindingseksperimenter:
Membranene ble opptint og på nytt suspendert i en bindende buffer som inneholdt 20 mM HEPES-NaOH, 3 mM MgCh, 3 mM CaCh, 100 mM NaCl, pH 7,4. Konkurranseundersøkelser ble utført ved innkubering i 1 time ved 4°C: 3 nM [<3>H]-MPEP (39 Ci/mmol, Tocris, Cookson Ltd, Bristol, UK), 50 ug membran og et konsentrasjonsområde fra 0,003 nM - 30 uM av forbindelsene med et totalt reaksjonsvolum på 300 ul. Den ikke-spesifikke bindingen ble definert ved å bruke 30 uM MPEP. Reaksjonen ble avsluttet ved rask filtrering gjennom glassfiberfilterplater (Unifilter 96-brønners GF/B filterplater, Perkin-Elmer, Schwerzenbach, Sveits) ved å bruke 4 x 400 ul iskald buffer i et celleinnhøstingsapparat (Filtermate, Perkin-Elmer, Downers Grove, USA). Radioaktiviteten ble bestemt ved væskescintillasjonsspektrometri ved å bruke en 96 brønners plateleser (TopCount, Perkin-Elmer, Downers Grove, USA).
• Dataanalyse:
Inhiberingskurvene ble utviklet ved å bruke Prism GraphPad-programmet (Graph Pad Software Inc, San Diego, USA). ICso-bestemmelsene ble utført fra de oppnådde data ved en 8-punkts konsentrasjonsresponskurve ved å bruke en ikke-lineær regresj onsanaly se.
Den etterfølgende tabellen viser de midlere ICso-verdiene slik disse ble oppnådd ved minst tre uavhengige eksperimenter med utvalgte molekyler, hvor det hver gang ble utført to parallelle forsøk.
Forbindelsene ifølge den foreliggende oppfinnelsen viser høy affinitet for mGluR5-reseptoren. Som allosteriske modulatorer kan de brukes for produksjon av medikamenter, da spesielt for å forebygge eller behandle lidelser i
sentralnervesystemet så vel som andre lidelser som er modulert av denne reseptoren.
Forbindelsene ifølge oppfinnelsen kan administreres enten alene eller i kombinasjon med andre farmasøytiske midler som er effektive ved behandlingen av de tilstander som er nevnt ovenfor.
De følgende ikke-begrensende eksemplene er tenkt å illustrere oppfinnelsen. De fysiske data som er gitt for de eksemplifiserte forbindelsene er i overensstemmelse med den angitte strukturen på disse forbindelsene.
Klinkekulenedgravningsmodell på angst
Angstmodeller i gnagere er brukt som standardtester for å demonstrere anksiolytisk lignende egenskaper til nye forbindelser. Mus har en tendens til å grave ned uskadelige nye gjenstander når de møter slike i et prøvebur.
Klinkekulenedgravingsatferd i mus blir redusert ved hjelp av forbindelser som er effektive angstdempende midler hos mennesker. Klinkekulenedgraving hos mus er således blitt brukt som en modell for prediksjonen av angsdempendelignende effekter av forbindelser (Millan, M.J. et al., Neuropharmacology, 42: 677-684
(2002)).
Selektive negative allosteriske modulatorer (allosteriske antagonister) for den metabotropiske glutamatreseptorundertype 5 (mGluR5), har tidligere vist seg å redusere klinkekulenedgraving hos mus (Spooren, W.P. et al., Journal of Pharmacology and Experimental Therapeutics, 295: 1267-1275 (2000)). Disse resultatene viser at klinkekulenedgravingstesten er en brukbar modell for å påvise det angstdempende potensialet for forbindelser som er antagonister for mGluR5. Slike forbindelser kan brukes ved behandling av angst og beslektede lidelser.
Forsøksdyr: De foreliggende undersøkelser ble utført i overensstemmelse med de dyrebehandlings- og bruksbestemmelser som er nedfelt hos Addex Pharmaceuticals og EEC-direktivene for beskyttelse av dyr som brukes eksperimentelt og for andre vitenskapelige formål (86/609/EEC og etterfølgende revisjoner). C57BL6/j hannmus (20-30 g) med en alder på 7 uker da de ble levert ble plassert i grupper under kontrollert temperatur og fuktighet i en 12 timers lys/mørkesyklus i minst 5 dager før bruk. Musene hadde adgang til mat og vann ad libitum, bortsett fra under klinkekulenedgravingseksperimentene.
Bedømmelse av klinkekulenedgraving: Effekten av forbindelsene på klinkekulenedgraving hos mus ble testet. Den dagen testen ble gjennomført ble dyrene merket på halen og veid i et separat rom 1 time før medikamentadministrering. Testforbindelsen eller bærervæsken ble administrert po 60 minutter før selve testen. Klinkekulenedgravingen ble testet i et separat eksperimentrom. For denne testen ble musene plassert individuelt i klare plastbur (16 x 22 x 14 cm) med 5 cm sagmugg og 10 klinkekuler som var jevnt fordelt langs veggene i buret. Musene ble så overlatt til seg selv uforstyrret i burene i 30 minutter. Etter fjerning av musene fra prøveburene, ble antall nedgravde klinkekuler talt. En klinkekule ble ansett å være nedgravd hvis 2/3 eller mer var dekke.
Administrering av forbindelsene: Testforbindelsene ble løst i en løsning av 80% 0,1 N saltsyre og 20% Tween 80 og justert til pH 6 med 1 M NaHCOs. Testforbindelsene ble administrert ved en oral mageslange (po) i et volum på 10 ml/kg. Forbindelse-bærervæskebehandlede mus mottok det ekvivalente volumet av bærervæskeløsningen po i fravær av tilsatt forbindelse.
Statistiske analyser: Statistiske analyser ble utført ved å bruke GraphPad PRISM versjon 4.01 statistisk programvare (GraphPad, San Diego, CA, USA). Dataene ble analysert ved å bruke en enveis variasjonsanalyse (ANOVA) fulgt av Bonferroni-korrigerte multiple sammenligninger eller t-tester hvis bare 2 grupper var tilstede. Signifikansnivået ble satt på p < 0,05.
Effekt av forbindelsene på klinkekulenedgraving i mus
Resultatene viser at forbindelsen i henhold til oppfinnelsen svekker klinkekulenedgraving i mus og antyder at forbindelsene kan brukes ved behandling av angst.
Vogel-konfliktdrikkemodellen på angst i rotter
Angstmodeller hos gnagere brukes som standardtester for å demonstrere anksiolytisk lignende egenskaper for nye forbindelser. Vogelkonfliktdrikkemodellen involverer konflikten mellom tørst og mottak av milde sjokk ved drikking av vann (straffedrikking). Rotter som var holdt vekk fra vann ble plassert i et kammer og ble periodevis utsatt for sjokk når de drakk vann. Sjokkene undertrykker drikking mens angstdempende midler reverserer denne sjokkinduserte undertrykkingen av drikking. Vogelkonfliktdrikkemodellen ble først foreslått som en screeningmodell for angstdempende midler (Vogel, J.R. et al., Psychopharmacologia (Berl.), 21: 1-7 (1971)) og allment akseptert som en robust modell for testing av forbindelsers anksiolytisklignende egenskaper (Millan, M.J. og Brocco M., European Journal of Pharmacology, 463: 67-96 (2003)).

Sprague-Dawley hannrotter (350 g) 7-9 uker gamle på testtidspunktet ble plassert i grupper i en temperatur- og fuktighetskontrollert forsøksanordning i en 12 timers lys/mørkesyklus i minst 5 dager før bruk. Rottene hadde adgang til for ad libitum, bortsett fra under Vogelkonfliktdrikkemodelleksperimentene. Rottene hadde adgang til vann ad libitum inntil 48 timer før selve testingen.
Bedømmelse av Vogelkonfliktdrikking: Effekten av forbindelsene på drikking i Vogelkonflikdrikkemodellen hos rotter ble testet. Testkamrene var plassert i lydsvekkende bokser og hvert kammer inneholdt en drikketube av rustfritt stål og et gulv av et stålnett (MedAssociates, Georgia, Vermont, USA). 48 timer før testingen ble rottene plassert i prøvekamrene i 10 minutter. Vann ble fjernet fra rottene umiddelbart etter tilvenningssesjonen. 24 timer før testingen ble rottene igjen plassert i testkamrene og fikk lov til å drikke i 4 minutter. Rottene fikk så fri adgang til vann i 1 time, hvoretter vannet ble fjernet. På testdagen ble rottene brakt til testrommet minst 30 minutter før selve testingen. Rottene ble plassert individuelt i testkamrene i en periode på 5 minutter. Rottene fikk et sjokk for hver 20. slikking på drikketuten. Antall straffede drikkinger ble telt automatisk ved hjelp av datamaskingrensesnittet. Antallet straffede drikkinger ble sammenlignet mellom behandlingsgruppene. Et økende antall straffede drikkinger i rotter behandlet med en forbindelse ble tolket som en anksiolytisklignende effekt.
Administrering av forbindelsene: Testforbindelsene ble enten løst i en løsning av 80% 0,1 N saltsyre, 20% Tween 80 og justert til pH 6 med 1 M NaHCOs (eksempel 256) eller en 7,5% arabikumgummi/EhO-løsning (eksempel 130). Prøveforbindelsene ble administrert ved hjelp av en oral mageslange (po) i et volum på 3 ml/kg. Forbindelse-bærervæskebehandlede rotter mottok et tilsvarende volum av bærervæskeløsningen po i fravær av tilsatt forbindelse.
Statistiske analyser: Det ble utført statistiske analyser ved å bruke GraphPad PRISM versjon 4.01 statistikkdataprogrammet (GraphPad, San Diego, CA, USA). Dataene indikerer at forbindelsene i henhold til oppfinnelsen er angstdempende i Vogelkonfliktdrikketesten.
Sammendrag av atferdsresultater
Resultatene viser at forbindelser i henhold til oppfinnelsen er effektive i spesifikke modeller på angst hos gnagere. Disse resultatene antyder at forbindelsene ifølge den foreliggende oppfinnelsen kan brukes ved behandling av angstlidelser og beslektede lidelser og sykdommer i sentralnervesystemet.
EKSEMPLER
Hvis intet annet er angitt, ble alle utgangsforbindelser oppnådd fra kommersielle firmaer og brukt uten ytterligere rensing.
Spesifikt kan de følgende forkortelser være anvendt i eksemplene og i hele den etterfølgende beskrivelsen.
Alle referanser til saltløsning refererer seg til en mettet vandig løsning av NaCl. Hvis intet annet er angitt, så er alle temperaturer angitt i °C (grader Celsius). Alle reaksjoner ble utført i en inert atmosfære ved romtemperatur hvis intet annet er angitt.
Den mikrobølgeovn som er anvendt, er et apparat fra Biotage (Optimizer™) utstyrt med en intern kontrollenhet som styrer reaksjonstemperatur og trykk og opprettholder den forønskede temperatur ved hjelp av datastyrt kontroll.
<*>H NMR-spektra ble målt ved hjelp av et Brucker 500 MHz apparat. Kjemiske skift er uttrykt i deler pr million (ppm, 5 enheter). Koblingskonstantene er i hertzenheter (Hz). Alle massespektra ble målt ved hjelp av elektrosprayioniserings (ESI) -metoder.
De fleste reaksjonene ble kontrollert ved hjelp av tynnsjiktkromatografi på 0,25 mm Macherey-Nagel silikagelplater (60F-2254) og visualisert med UV-lys. Flashkolonnekromatografi ble utført på silikagel (220-440 mesh, Fluka).
Smeltepunktsbestemmelsene ble utført i et Buchi B-540-apparat.

33(A) Etyl imidazof 1. 2- alpyridin- 2- karboksylat
En løsning av 2-aminopyridin (2 g, 21 mmol) og etylbrompyruvat (4,14 g, 21 mmol) i 31 ml etanol ble kokt under tilbakeløp i 24 timer. Løsemiddelet ble fordampet og resten ble løst i et minimalt volum av vann. Løsningen ble nøytralisert (pH = 8) med mettet NaHCCtø. Det vandige laget ble ekstrahert med AcOEt, hvoretter det organiske laget ble vasket med mettet NaCl. Løsemiddelet ble fjernet under redusert trykk, og dette ga 2,51 g etyl imidazo[l,2-a]pyridin-2-karboksylat (utbytte: 62%) som en oransje olje som kan brukes uten ytterligere rensing.
LCMS (RT): 0,72-1,39 min; MS (ES+) ga m/z: 191,1.

En tørr rundkolbe ble tilsatt LiAlFf.4 (650 mg, 17 mmol) i 52 ml vannfri THF under røring. Løsningen ble så avkjølt til 0°C og så dråpevis tilsatt en løsning av etyl imidazo[l,2-a]pyridin-2-karboksylat (2,5 g, 13 mmol) i 3 ml tørr THF. Løsningen ble grønn og reaksjonsblåndingen ble så rørt ved 0°C i 30 minutter og deretter i 2 timer ved romtemperatur. Reaksjonen ble stoppet ved suksessivt å tilsette 0,52 ml vann, 0,52 ml NaOH 1 M og 3 x 0,52 ml vann. Løsningen ble så filtrert gjennom celite. Det organiske laget ble fordampet, og dette ga 2,25 g av en oransje væske. Resten ble renset ved flashkromatografi på silikagel (forpakket 70 g silikagelkolonne DCM/MeOH: 95/5 som elueringsmiddel), og dette ga 1 g 2-hydroksymetylimidazo[l,2-a]pyridin (utbytte: 51%) som en brun olje.

En rundkolbe som inneholdt 2-hydroksymetylimidazo[l,2-a]pyridin (800 mg, 5,4 mmol) i 8 ml DCM ble ved romtemperatur tilsatt tionylklorid (1,96 ml, 27 mmol). Løsningen ble klar og 10 minutter senere hadde det dannet seg et bunnfall. Reaksjonsblandingen ble rørt ved romtemperatur i 2 timer, hvoretter løsemiddelet ble fjernet under redusert trykk, og dette ga 1,10 g av et brunaktig fast stoff som klorhydratformen av 2-(klormetyl)imidazo[l,2-a]pyridin (utbytte: 100%).
40 ml mettet NaHC03ble tilsatt klorhydratformen av 2-(klormetyl)imidazo[l,2-ajpyridin og det vandige laget ble ekstrahert med etylacetat. De organiske lagene ble slått sammen og vasket med saltløsning, tørket over MgS04, hvoretter løsemiddelet ble fjernet under redusert trykk, og dette ga 877 mg 2- (klormetyl)imidazo[l,2-a]pyridin (utbytte: 80%) som et brunlig fast stoff (smeltepunkt 84-85°C).

En suspensjon av etyl 5-bromimidazo[l,2-a]pyridin-2-karboksylat (1,50 g, 5,6 mmol), Pd(PPh3)4(322 mg, 0,279 mmol) i en blanding av 17 ml toluen, 6 ml NaHC03og 4 ml MeOH tilsatt fenylborsyre ved romtemperatur. Den resulterende reaksjonsblandingen ble holdt på 80°C i 12 timer, avkjølt og fortynnet med 50 ml vann. Uløselige faste stoffer ble frafiltrert og fasene ble skilt. Den vandige fasen ble ekstrahert med etylacetat, hvoretter de samlede organiske lag ble vasket med vann, hvoretter løsemiddelet ble fjernet under redusert trykk. Ifølge den generelle protokollen som er beskrevet i J. Heterocyclic. Chem., 1988, 25, 129-137, så ble en løsning av 5-fluorpyridin-2-amin (1,80 g, 16 mmol) i 24 ml EtOH tilsatt l,3-diklorpropan-2-on (2,03 g, 16 mmol) og den resulterende blandingen ble rørt over natten ved 80°C. Løsemiddelet ble fordampet og resten ble løst i en minimal mengde med vann. Løsningen ble så nøytralisert (pH = 8) med mettet NaHC03. Det vandige laget ble ekstrahert med AcOEt, hvoretter det organiske laget ble vasket med mettet NaCl. 75 (D) 2-( 4-( 5- fluorDvridin- 2- vnbut- 3- vnvnimidazori. 2- alDvridin Et tørt reaksjonsrør som inneholdt en suspensjon av kobberjodid (6,5 mg, 0,03 mmol) og 2,3 ml trietylamin ble tilsatt 2-brom-5-fluorpyridin (120 mg, 0,68 mmol) og Pd(PPh3)2Ch (39 mg, 0,03 mmol) under N2. Det ble oppnådd en gul suspensjon etter 5 minutters røring ved romtemperatur. En løsning av 2-(but-3-ynyl)imidazo[l,2-a]pyridin (120 mg, 0,68 mmol) i 0,5 ml trietylamin ble så tilsatt under N2. F ORMULERINGSEK SEMPLER
Typiske eksempler på oppskrifter for formuleringen ifølge den foreliggende oppfinnelsen er som følger:
1) Tabletter
I dette eksempelet kan forbindelsen i henhold til oppfinnelsen erstattes med den samme mengden av ethvert av de beskrevne eksemplene..
2) Suspensjon
Det ble fremstilt en vandig suspensjon for oral administrering slik at hver milliliter inneholdende fra 1 til 5 mg av ett av de ovenfor beskrevne eksempler, 50 mg natriumkarboksymetylcellulose, 1 mg natriumbenzoat, 500 mg sorbitol og vann til 1 ml.
3) Injiserbar sammensetning
Det ble fremstilt en parenteral sammensetning ved å røre sammen 1,5 vekt% av den aktive bestanddelen ifølge den foreliggende oppfinnelsen i 10 volum% propylenglykol og vann.
4) Salve

karakterisert vedat kan eksistere som optiske isomerer og hvor nevnte forbindelse er enten den rasemiske blandingen eller en eller begge de individuelle optiske isomerene.
3. Forbindelsen ifølge ethvert av kravene 1 eller 2,
karakterisert vedat nevnte forbindelse er valgt fra: 2-(4-(pyridin-2-yl)but-3-ynyl)-imidazo[l,2-a]pyridin

6- fluor-2-(4-(pyridin-2-yl)but-3-ynyl)-imidazo[l,2-a]pyridine 2-(4-(5-fluorpyridin-2-yl)but-3-ynyl)-imidazo[l,2-a]pyridine og farmasøytisk akseptable salter av disse.
