Abstract
Den foreliggende oppfinnelsen vedrører lagringsstabile flytende formuleringer av palonosetron for å redusere kjemoterapi- og radioterapi- indusert emesis med palonosefron. Formuleringene er i særdeleshet nyttige ved fremstilling av intravenøs og orale flytende medikamenter.
Description
Den foreliggende oppfinnelsen vedrører lagringsstabile flytende palonosetronformuleringer som er spesielt nyttige ved fremstilling av injiserbare og orale medikamenter, på formen av farmasøytiske løsninger som omfatter
a) fra 0,03 mg/ml til 0,2 mg/ml palonosetronhydroklorid; og
b) en farmasøytisk akseptabel bærer med en pH på fra 4,0 til 6,0 som omfatter en
chelatdanner.
Emesis er en uheldig konsekvens av cytotoksisk terapi, radioterapi og postoperative miljøer som drastisk påvirker livskvaliteten til mennesker som gjennomgår slike behandlinger. I de senere årene har en klasse av legemidler som refereres til som 5-HT3 (5-hydroksytryptamin)-reseptorantagonister blitt utviklet, og denne legemiddelklassen behandler slike emesis ved å motvirke cerebrale funksjoner som er forbundet med 5-HT3-reseptoren. Legemidler innenfor denne klassen inkluderer ondansetron, granisetron, alosetron, tropisetron og dolasetron. Disse 5-HT3-antagonistene administreres ofte intravenøst kort tid før kjemoterapi og radioterapi påbegynnes, og kan administreres mer enn én gang under en kjemoterapi- eller radio-terapisyklus. I tillegg tilføres de ofte som tabletter eller orale eliksirer enten som et supplement til en intravenøs administrering, eller for å forenkle hjemmebruk av legemidlet dersom pasienten selv administrer det kjemoterapeutiske regimet.
På grunn av at noen kjemoterapeutiske midler kan indusere emesis for perioder på flere dager selv når de kun administreres én gang, ville det være ønskelig å administrere et emesishemmende legemiddel slik som en 5-HT3-antagonist hver dag inntil risikoen for emesis i det alt vesentlige har gitt seg. Denne klassen av 5-HT3-antagonister har ikke vist seg å være spesielt nyttig når det gjelder å møte dette behovet, fordi 5-HT3-reseptorantagonistene som for tiden finnes på markedet har vist seg å være mindre effektive til å kontrollere utsettelse av kvalme og oppkast enn hva de er ved kontrollering av akutt emesis. Nylig er det utført en klinisk undersøkelse vedrørende palonosetron, som er en ny 5-HT3-reseptorantagonist rapportert i US patent nr. 5 202 333. Disse undersøkelsene har vist at legemidlet er en størrelsesorden kraftigere enn de fleste eksisterende 5-HT3-reseptorantagonistene, og at det har en overraskende halveringstid på omtrent 40 timer, og at den er effektiv for å redusere forsinket start av kvalme indusert ved kjemoterapeutiske midler. Imidlertid har formulering av palonosetron som flytende formuleringer ikke vist seg å være noen lett sak, hvilket typisk skyldes lagringsstabilitetsspørsmål. US patent nr. 5 202 333 beskriver en intravenøs palonosetronformulering i eksempel 13 som inneholder de følgende ingrediensene:
Formuleringen har en pH på 3,7 og en lagringsstabilitet som er mindre enn den ett til to års perioden som er påkrevd av helsemyndighetene i forskjellige land.
Ondansetron, dets anvendelse og medikamenter fremstilt ved ondansetron er beskrevet i US patenter nr. 4 695 578, 4 753 789, 4 929 632, 5 240 954, 5 344 658, 5 578 628, 5 578 632, 5 922 749, 5 622 720, 5 955 488 og 6 063 802. Det distribueres kommersielt av GlaxoSmithKline som Zofran® og er angitt for forebyggelse av postoperativ kvalme og oppkast (PONV), kreftkjemoterapi-indusert kvalme og oppkast (CINV), og radioterapi-indusert kvalme og oppkast (RTNV) og det er tilgjengelig som en injeksjon,
tabletter og løsning, og som zofran ODT® (ondansetron) orale desintegrerende tabletter.
Granisetron, dets anvendelse og medikamenter framstilt med granisetron er beskrevet i US patenter nr. 4 886 808, 4 937 247, 5 034 398 og 6 294 548. Det distribueres kommersielt av Roche Laboratories Inc. som Kytril®, og er angitt for forebyggelse av kvalme og oppkast forbundet med kjemoterapi eller stråleterapi, og tilbys på tablett-form, oral løsning og som en injeksjon.
Alosetron, dets anvendelser og medikamenter fremstilt med alosetron er beskrevet i US patenter 5 360 800 og 6 284 770. Det distribueres kommersielt av GlaxoSmithKline som Lotronex®.
Tropisetron er kommersielt tilgjengelig som Navoban® (Novartis) CAS -89565-68-4 (tropisetron); CAS - 105826-92-4 (tropisetron hydroklorid) og det er angitt for behandling av PONV og CINV.
Dolasetron, dets anvendelser og medikamenter framstilt med ondansetron er beskrevet i US patenter nr 5 011 846 og 4 906 755. Det distribueres kommersielt av Aventis Pharmaceuticals Inc. Som Anzemet®, og er angitt for forebyggelse av både PONV og CINV, og det tilbys på formen av en tablett eller en intravenøs løsning.
Det finnes derfor et behov for en palonosetronformulering med økt stabilitet og derved økt lagringstid. Det er også et behov for et passende konsentrasjonsområde for både 5-HT3-reseptorantagonisten og dets farmasøytisk akseptable bærere som ville lette fram-stillingen av en formulering med den økte stabiliteten.
Det er et formål ved den foreliggende oppfinnelsen å tilveiebringe en palonosetron-hydrokloridformulering med økt farmasøytisk stabilitet for å forebygge og/eller redusere emesis.
Det er et annet formål med oppfinnelsen å tilveiebringe et akseptabelt konsentrasjonsområde som vil stabilisere en formulering som inneholder palonosetronhydroklorid.
Det er ytterligere et formål med den foreliggende oppfinnelsen og tilveiebringe en palonosetronformulering som vil tillate lenger lagring.
Det er også et formål med oppfinnelsen å tilveiebringe en palonosetronformulering som ville tillate terminal sterilisering.
Oppfinnerne har gjort en rekke funn som underbygger en overraskende effektiv og allsidig formulering og forebygging av emesis ved å anvende palonosetron. Disse formuleringene er stabile i perioder på mer enn 24 måneder ved romtemperatur, og kan således lagres uten kjøling, og fremstilles ved å benytte ikke-aseptiske, terminale steriliseringsprosesser.
Ved et aspekt har oppfinnerne funnet at formuleringene som inkluderer den aktive ingrediensen palonosetron, ved visse tilfeller kun krever en tiendedel av mengden av andre tidligere kjente forbindelser for å behandle emesis, hvilket overraskende mulig-gjør bruk av palonosetronkonsentrasjoner langt under de som vanligvis vil forventes. Således tilveiebringer oppfinnelsen en farmasøytisk stabil løsning for å forebygge eller redusere emesis som omfatter a) fra 0,03 mg/ml til 0,2 mg/ml palonosetronhydroklorid; og b) en farmasøytisk akseptabel bærer med en pH fra 4,0 til 6,0 som omfatter en chelatdanner.
Oppfinnerne har funnet at ved å regulere formuleringens pH og/eller eksipient-konsentrasjoner er det mulig å øke stabiliteten til palonosetronformuleringer. I en annen utførelsesform tilveiebringer oppfinnelsen en farmasøytisk stabil løsning for å forebygge eller redusere emesis som definert over som også omfatter fra 10 til 100 millimol sitratbuffer; og fra 0,005 til 1,0 mg/ml EDTA.
Oppfinnerne har videre funnet at tilsetningen av mannitol og en chelatdanner kan øke stabiliteten til palonosetronformuleringer. Ved en ytterligere utførelsesform tilveiebringer oppfinnelsen derfor en farmasøytisk stabil løsning for å forebygge eller redusere emesis som definert over, der den farmasøytisk akseptable bæreren omfatter mannitol.
Definisjoner
"Medisinglass" betyr en liten glassbeholder som er lukket med best egnet propp og forsegling, eller egnede primærbeholdere kan anvendes, for eksempel, men ikke begrenset til, forhåndsfylte sprøyter. Medisinglass betyr også en forseglet (lukket) medikamentbeholder som anvendes kun én gang, og inkluderer knuselig og uknuselige glassmedisinglass, knuselig plastmedisinglass, miniatyrglass med skrukork, og en hvilken som helst annen type beholder av en størrelse som er i stand til å inneholde kun én enhetsdose palonosetron (typisk omtrent 5 ml).

"Palonosetron" betyr (3aS)-2,3,3a,4,5,6-Heksahydro-2-[(S)-l-Azabisyklo[2.2.2]okt-3-yl]2,3,3a,4,5,6-heksahydro-l-okso-l//benz[de]isoquinohn, og foreligger fortrinnsvis som monohydroklorid. Palonosetronmonohydroklond kan representeres ved den

Konsentrasjoner - når konsentrasjoner av palonosetron her gis, er konsentrasjonen målt med hensyn til vekt av den frie basen. Konsentrasjoner av alle andre ingredienser gis basert på vekten av ingrediens tilsatt til løsningen.
"Farmasøytisk akseptabel" betyr hvilke som er brukbare ved fremstilling av en farma-søytisk sammensetning som generelt er trygg, ikke-toksisk og verken biologisk eller på annen måte uønsket og inkluderer alle de som er akseptable for veterinærbruk så vel som humanfarmasøytisk anvendelse.
"Farmasøytisk akseptable salter" betyr salter som er farmasøytisk akseptable, som definert over, og som innehar den ønskede farmakologiske aktiviteten. Slike salter inkluderer syreaddisjonssalter dannet med uorganiske syrer slik som hydrokloridsyre, hydrobromsyre, svovelsyre, salpetersyre, fosforsyre og lignende; eller med organiske syrer slik som eddiksyre, propionsyre, heksansyre, heptansyre, syklopetanpropionsyre, glykolsyre, pyrodrusyre, melkesyre, malonsyre, ravsyre, eplesyre, maleinsyre, fumar-syre, vinsyre, sitronsyre, benzosyre, o-(4-hydroksybezoyl)benzosyre, kanelsyre, mandelsyre, metansulfonsyre, etansulfonsyre, 1,2-etandisulfonsyre, 2-hydroksyetan-sulfonsyre, benzensulfonsyre, p-klorbenzensurlfonsyre, 2-naftalensulfonsyre, p-toluen-sulfonsyre, kamfersulfonsyre, 4-metylbisyklo[2.2.2]okt-2-en-l-karboksylsyre, gluko-hetonsyre, 4,4'-metylenbis(3-hydroksy-2-en-l-karboksylsyre), 3-fenylpropionsyre, trimetyleddiksyre, tertiær butyleddiksyre, laurylsvovelsyre, glukonsyre, glutaminsyre, hydroksynaftolsyre, sahcylsyre, stearinsyre, muconsyre og lignende.
I tillegg kan farmasøytisk akseptable salter dannes når et surt proton som er tilstede er i stand til å reagere med uorganiske eller organiske baser. Akseptable uorganiske baser inkluderer natriumhydroksid, natriumkarbonat, kaliumhydroksid, aluminiumhydroksid og kalsiumhydroksid. Akseptable organiske baser inkluderer etanolamin, dietanolamin, triethanolamin, trometamin, N-metylglukamin og lignende.
Det faktum at palonosetron i noen tilfeller kan formuleres i konsentrasjoner på kun omtrent en tiendedel av mengden av andre kjente forbindelser for behandling av emesis, muliggjør overraskende anvendelse av konsentrasjoner av palonosetron som er langt under de som vanligvis ville forventes. Således tilveiebringer oppfinnelsen en farmasøy-tisk stabil løsning for forebyggelse eller reduksjon av emesis som omfatter a) fra 0,03 mg/ml til 0,2 mg/ml palonosetronhydroklorid; og b) en farmasøytisk akseptabel bærer med en pH på fra 4,0 til 6,0 som omfatter en chelatdanner. En fremgangsmåte for å formulere en farmasøytisk stabil løsning av palonosetronhydroklorid omfatter å blande fra 0,03 mg/ml til 0,2 mg/ml palonosetronhydroklond med en farmasøytisk akseptabel bærer med en pH på fra 4,0 til 6,0 som omfatter en chelatdanner. I alternative utførelses-former inkluderer formuleringen palonosetronhydroklorid i en konsentrasjon på mest optimalt omtrent 0,05 mg/ml.
En særlig fordel forbundet med en lavere dosering av intravenøs palonosetron er evnen til å administrere legemidlet i en enkel intravenøs bolus i løpet av en kort, atskilt tids-periode. Denne tidsperioden strekker seg generelt fra omtrent 10 til omtrent 60 sekunder eller fra omtrent 10 til omtrent 40 sekunder, og mest foretrukket er fra omstrent 10 til 30 sekunder. Ved en særskilt utførelsesform leveres palonosetron i medisinglass som omfatter 5 ml løsning, hvilket er likt med omtrent 0,25 mg palonosetron med en konsentrasjon på omtrent 0,05 mg/ml.
Oppfinnerne har videre funnet at ved å regulere formuleringens pH og/eller eksipientens konsentrasjoner er det mulig å øke stabiliteten til palonosetronformuleringer. I alternative utførelsesformer er pH fra 4,5 til 5,5, og mest optimalt fra omtrent 5,0. Det finnes mange eksempler for en fagmann på området på egnede løsninger for å regulere pH i en formulering. To eksempler på slike løsninger er natriumhydroksid og saltsyreløsrung, hvorved begge disse kunne anvendes for å regulere pH i formuleringen.
I en annen utførelsesform tilveiebringer oppfinnelsen en farmasøytisk stabil løsing for å forebygge eller redusere emesis som omfatter fra 0,03 til 0,2 mg/ml palonosetronhydroklorid og (i) fra 10 til 100 millimol sitratbuffer, og/eller (ii) fra 0,005 til 1,0 mg/ml EDTA i en farmasøytisk akseptabel bærer med en pH på fra 4,0 til 6,0. En fremgangsmåte for å formulere en farmasøytisk stabil løsning av palonosetronhydroklorid omfatter å blande fra 0,03 til 0,2 mg/ml palonosetronhydroklorid og (i) fra 10 til 100 millimol sitratbuffer, og/eller (n) fra 0,005 til 1,0 mg/ml EDTA i en farmasøytisk akseptabel bærer med en pH på fra 4,0 til 6,0. Sitratbufferen kan være på formen av sitronsyre og/eller et salt av sitronsyre slik som trinatriumsitrat. Ved forskjellige utførelsesformer kan områdene for én eller flere av de forannevnte ingrediensene modifiseres som følger: • Formuleringen kan omfatte palonosetronhydroklorid i en konsentrasjon på fra 0,03 mg/ml til 0,2 mg/ml palonosetronhydroklond, og mest optimalt omtrent 0,05 mg/ml. • Formuleringen kan omfatte sitratbuffer i en konsentrasjon på fra 10 til 40 millimol, eller 15-30 millimol.
Formuleringen kan omfatte EDTA i en konsentrasjon på fra 0,005 mg/ml til 1,0
mg/ml eller 0,3 til 0,7 mg/ml, og mest optimalt omtrent 0,5 mg/ml.
Oppfinnerne har videre funnet at tilsetningen av mannitol og en chelatdanner kan øke stabiliteten til palonosetronformuleringer. I ytterligere en annen utførelsesform tilveiebringer oppfinnelsen derfor en farmasøytisk stabil løsning for å forebygge eller redusere emesis som omfatter a) palonosetronhydroklorid i en konsentrasjon fra 0,03 mg/ml til 0,2 mg/ml, og b) en farmasøytisk akseptabel bærer med en pH på fra 4,0 til 6,0, der den farmasøytisk akseptable bæreren omfatter en chelatdanner og mannitol. En fremgangsmåte for å formulere en farmasøytisk stabil løsning av palonosetronhydroklorid ved en konsentrasjon på 0,03 mg/ml til 0,2 mg/ml omfatter å blande a) palonosetronhydroklorid og b) en farmasøytisk akseptabel bærer med en pH fra 4,0 til 6,0, der den farma-søytisk akseptable bæreren omfatter en chelatdanner og mannitol. Chelatdanneren er fortrinnsvis EDTA, og ved forskjellige utførelsesformer er chelatdanneren til stede i en konsentrasjon på fra 0,005 til 1,0 mg/ml eller fra 0,05 mg/ml til 1,0 mg/ml eller fra 0,3 til 0,7 mg/ml eller mest optimalt omtrent 0,5 mg/ml. Ved forskjellige utførelsesformer er mannitol til stede i en konsentrasjon på fra 10,0 mg/ml til 80,0 mg/ml, fra 20,0 mg/ml til 60,0 mg/ml eller fra 40,0 til 45,0 mg/ml.
Injiserbare formuleringer formuleres typisk som vandige løsninger hvori vann er den primære eksipienten. Orale formuleringer vil generelt skille seg fra injiserbare formuleringer ved det ytterligere nærværet av smaksstoffer, fargestoffer eller viskositetsreguler-ende midler. Naturlige eller syntetiske søtningsstoffer inkluderer blant annet mannitol, sorbitol, sukrose, sakkarin, aspartam, acelsulfam K, eller syklamat. Disse midlene er generelt til stede i en konsentrasjon i et overskudd av 100 mg/ml eller 250 mg/ml når det benyttes som søtningsstoffer, i motsetning til konsentrasjonen av mannitol på 41,5 mg/ml som er beskrevet i noen av utførelsesformene ifølge oppfinnelsen, der mannitol kun tjener som et tonisifiserende middel.
Formuleringene ifølge den foreliggende oppfinnelsen er særlig egnede for anvendelse ved injiserbare og orale flytende formuleringer, men det vil forstås at løsningene kan ha alternative anvendelser. De kan for eksempel anvendes som mellomprodukter ved frem-stillingen av andre farmasøytiske doseringsformer. På lignende måte kan de ha andre administreirngsruter som inkluderer intranasal administrering eller inhalering. Injiserbare formuleringer kan benytte enhver rute inkludert intramuskulær, intravenøs eller subkutan.
Oppfinnelsen tilveiebringer anvendelse av
i) en pH av en løsning på fra 4,0 til 6,0,
ii) fra 10 til 100 mmol sitratbuffer;
iii) fra 0,005 til 1,0 mg/ml EDTA, og
iv) mannitol
for å tilveiebringe stabilitet til en injiserbar løsning av palonosetron eller et farmasøytisk akseptabelt salt derav, der palonosetronet eller et farmasøytisk akseptabelt salt derav er til stede i en konsentrasjon på fra 0,03 mg/ml til 0,2 mg/ml.
Ytterligere andre utførelsesformer vedrører forbedringer ved forenklingen av hvordan palonosetronformulenngen kan lagres eller fremstilles. I særdeleshet har oppfinnerne funnet at formuleringene ifølge den foreliggende oppfinnelsen tillater lagring av produktet i forlengede perioder ved romtemperatur. En fremgangsmåte for å lagre en eller flere beholdere hvori det forefinnes en løsning av palonosetronhydroklorid omfatter: a) å tilveiebringe et rom som omfatter den eller de nevnte beholderne; b) å regulere eller opprettholde temperaturen i rommet til høyere omtrent 10,15 eller 20°C; og c) å lagre de nevnte beholderne i det nevnte rommet i 1 måned, 3 måneder, 6 måneder, 1 år, 18 måneder, 24 måneder eller mer (fortrinnsvis ikke utover 36 måneder), hvori (i) palonosetronhydrokloridet foreligger i en konsentrasjon på fra 0,03 mg/ml til 0,2 mg/ml, (ii) pH i løsningen er 4,0 til 6,0, (ni) løsningen omfatter fra 10 til 100 millimol sitratbuffer og fra 0,005 til 1,0 mg/ml EDTA.
Stabiliteten til de forannevnte formuleringene egner seg selv også godt for terminale steriliseringsprosesser ved fremstillingsprosessen. En fremgangsmåte for å fylle en container hvori en løsning av palonosetronhydroklorid forefinnes som omfatter: a) å tilveiebringe én eller flere stabile åpne beholdere (fortrinnsvis 5 ml medisinglass); b) å fylle de nevnte beholderne med en løsning av palonosetron i et ikke-aseptisk miljø; c) å lukke de nevnte fylte beholderne; og d) å sterilisere de nevnte lukkede, fylte
beholderne, hvori (i) palonosetronhydrokloridet foreligger i en konsentrasjon på fra
0,03 mg/ml til 0,2 mg/ml, (ii) pH i løsningen er fra 4,0 til 6,0, (iii) løsningen omfatter fra 10 til 100 millimol sitratbuffer og fra 0,005 til 1,0 mg/ml EDTA.
Eksempler
Eksempel 1 • Stabilisering av pH
En studie ble utført for å bestemme effekten av pH på formuleringer som inneholder palonosetronhydroklorid, ved å måle stabiliteten ved 80°C ved pH 2,0, 5,0, 7,4 og 10,0. Resultatene indikerer at palonosetronhydroklorid er mer stabil ved pH 5,0.
Eksempel 2. Stabilisering av konsentrasjonsområder
En formuleringsoptimahsenngsstudie ble gjennomført ved å benytte en eksperimentell designprogramvare. Ved 24 timer ble mengder av legemiddelprodukt analysert for å undersøke det passende konsentrasjonsområdet for palonosetronhydroklorid (0,05 mg/ml til 5,0 mg/ml), sitratbuffer (0 til 80 mm) og EDTA (0 til 0,10%). Nivået av EDTA og sitratbuffer ble utvalgt basert på de optimale formuleringene, som ble vist å formuleres med EDTA 0,05% og 20 mm sitratbuffer ved pH 5,0. Resultatene fra denne studien indikerer at palonosetronkonsentrasjonen også er en kritisk faktor ved kjemisk stabilitet, der den høyeste stabiliteten er å se ved de laveste palonosetronkonsentra-sj onene.
Eksempel 3: Tonisifiseringsmiddél
Formuleringer av palonosetronhydroklorid i sitratbuffer ble fremstilt inkludert enten
a) natriumklorid eller b) mannitol. Palonosetronhydrokloridformuleringen inkludert mannitol viste fortreffelig stabilitet. Det optimale nivået av mannitol som er nødvendig
for en isotonisk løsning ble funnet å være 4,15%.
Eksempel 4: Formulering I
Det følgende er en representativ farmasøytisk formulering som inneholder palonosetron som er nyttig for intravenøse formuleringer, eller andre flytende formuleringer for legemidlet.
Eksempel 5 • Formulering II
Det følgende er en representativ farmasøytisk formulering som inneholder palonosetron som er nyttig for orale formuleringer eller andre flytende formuleringer av legemidlet.
Eksempel 6: Stabilitet av palonosetron uten deksametason
Den fysikalske og kjemiske stabiliteten av palonosetron HC1 ble studert ved konsentrasjoner på 5 ug/ml og 30 ug/ml i 5% dekstroseinjeksjon, 0,9% natriumkloirdinjeksjon, 5% dekstrose i 0,45% natriumkloridinjeksjon, og dekstrose 5% i melkesur Ringers injeksjon. Blandingene ble evaluert i løpet av 14 dager ved 4°C i mørket og i 48 timer ved 23 °C under fluoriserende lys.
Testprøvene av palonosetron HC1 ble fremstilt i polyvinylklorid (PVC)-poser av infusjonsløsninger i konsentrasjoner på 5 og 30 |ig/ml. Evalueringer av fysikalsk og kjemisk stabilitet ble utført på prøver tatt initielt og etter 1, 3, 5, 7 og 14 dager med lagring ved 4°C og etter 1, 4,24 og 48 timer ved 23°C. Fysikalsk stabilitet ble vurdert ved å benytte visuelle observasjoner ved normalt romlys og ved å benytte en høy-intensitetsmonoretningslysstråle. I tillegg ble turbiditet partikkelinnhold målt elektronisk. Den kjemiske stabiliteten av legemidlet ble evaluert ved å anvende en stabilitetsindikerende høyytelsesvæskekromatografisk (HPLC) analyseteknikk.
Alle prøver var fysikalsk stabile gjennom hele studien. Løsningen forble klar, og få eller ingen forandring i partikkelbyrde og disnivået ble funnet. I tillegg forekom lite eller intet tap av palonosetron HC1 i alle prøvene ved alle temperaturer under hele studieperioden.
Eksempel 7: Stabilitet av palonosetron med deksametason.
Den fysikalske og kjemiske stabiliteten av palonosetron HC1 0,25 mg blandet med deksametason (som natriumfosfat) 10 mg eller 20 mg i 5% dekstroseinjeksjon eller 0,9% natriumkloirdinjeksjon i polyvinylklorid (PVC)-miniposer, og også blandet med deksametason (som natriumfosfat) 3,3 mg i 5% dekstroseinjeksjon eller 0,9% natriumkloridinjeksjon i polypropylensprøyter ved 4°C i mørket i 14 dager og ved 23°C eksponert for normalt laboratoriefluoriserende lys i løpet av 48 timer ble studert.
Tekstprøvene av palonosetron HC1 5 ug/ml med deksametason (som natriumfosfat) 0,2 mg/ml og også 0,4 mg/ml ble fremstilt i polyvinylklorid (PVC)-miniposer av hver infusjonsløsning. I tillegg ble palonosetron HC125 ug/ml med deksametason (som natriumfosfat) 0,33 mg/ml i hver infusjonsløsning fremstilt som 10 ml testløsning i 20 ml polypropylensprøyter. Evalueringer av fysikalsk og kjemisk stabilitet ble utført og prøver tatt initielt og etter 1, 3, 7 og 14 dager med lagring ved 4°C og etter 1, 4, 24 og 48 timer ved 23°C. Fysikalsk stabilitet ble vurdert ved å benytte visuell observasjon i normalt romlys og ved å benytte en høyintensitetsmonoretningslysstråle. I tillegg ble turbiditet og partikkelinnhold målt elektronisk. Kjemisk stabilitet for legemidlet ble evaluert ved å anvende en stabilitetsindikerende høyytelsesvæskekromatografisk (HPLC) analyseteknikk.
Alle prøvene var fysikalsk kompatible gjennom hele studien. Løsningene forble klare, og liten eller ingen forandring i partikkelbyrden og disnivået ble funnet.
