Description
Oppfinnelsen vedrører en forbindelse med formelen
hvori R3 er cyklopropyl, 4-fluorfenyl eller 2,4-difluorfenyl, og X er fluor eller klor, som anvendes til fremstilling av 6-f luor-7-halogen-kinolonkarboksylsyremellomprodukter for * fremstilling av 6-fluor-7-substituerte-kinolonkarboksylsyrer som har antibakteriell aktivitet, og fremgangsmåter for fremstilling av slike mellomprodukter.
Vanligvis fremstilles de antibakterielle 6-fluor-7-substituerte-kinolonkarboksylsyrene ved substitusjon av de tilsvarende 7-halogen-6-fluor-kinolonkarboksylsyremellom-.produktene, hvori halogenatomet fortrinnsvis er fluor eller klor. Fremgangsmåter i tidligere teknologi for fremstiling av slike 7-halogenmellomprodukter er beskrevet i U.S. patentene 4.563.459 og 4.623.650.
Den foreliggende oppfinnelse finner anvendelse i en ny fremgangsmåte for fremstilling av 7-halogen-kinolonkarboksyl-syremellomprodukter med formelen
hvori R3 og X er som definert i formel II, R4 er hydroksy, C^ C, alkoksy, amino eller C^ C^ alkylamino, og X er fluor eller klor.
En klasse mellomprodukter er nye forbindelser med
formelen
hvori Rx er hydrogen eller C1-C4 alkyl, R2 er jod, t-butylamino, cyklopropylamino, fenylamino, 4-fluorfenylamino eller 2,4-difluorfenylamino, og X er fluor eller klor under den forut-setningen at når R2 er jod, er Rx hydrogen. Foretrukne mellomprodukter i denne klassen er forbindelser med formelen V, hvori Rx er hydrogen, X er fluor, og R2 er jod, cyklopropylamin eller 2,4-difluorfenyl. Forbindelsene med formelen II fremstilles ifølge oppfinnelsen ved omsetning av en forbindelse med formelen

R5R6C = 0
hvori R5 og R6 begge er klor eller triklormetyloksy, eller R5 er klor og R6 er Ci-Cg alkoksy, triklormetyloksy, fenoksy eller fenoksy substituert med en substituent som er inert under reaksjonsbetingelsene såsom en, to eller tre av halogen, nitro, C1-C6 alkyl eller trifluormetyl. Fortrinnsvis er R5 og R6 begge triklormetoksy eller begge klor, eller R5 er klor og R6 er triklormetyloksy.
Forbindelsene med formel III fremstilles ved å omsette en forbindelse med formelen hvori X er fluor eller klor, og Hal er jod, brom eller klor, med en forindelse med formelen R3NH2, hvor R3 er etyl, t-butyl, cyklopropyl, fenyl, 4-f luorf enyl eller 2 ,4-dif luorf enyl, i nærvær av kobber eller en kobberforbindelse.
Forbindelsene med formelen IV hvori Hal er jod, er nye forbindelser. De kan fremstilles fra den kjente forbindelsen 2-amino-4,5-difluorbenzosyre ved omsetning med natriumnitritt i en løsning av fortynnet svovelsyre ved en temperatur på omlag -10 til 0°C og omgivende trykk. Den dannede diazonium-forbindelsen behandles deretter med en løsning av kaliumjodid i fortynnet svovelsyre ved en temperatur mellom omlag -10 og 0°C, og den resulterende mørke vellingen omrøres i omlag 12 til 24 timer ved langsom oppvarming til omgivende temperatur.
Fremstillingen av antranilsyreforbindelser med formelen III fra forbindelser med formelen IV foregår i nærvær av katalytiske mengder av kobber (0) eller en kobberforbindelse såsom kobber(II)oksyd eller kobber(I)oksyd, eller et kobber-salt såsom kobber(II)acetat, kobber(II)sulfat, kobber(II)-klorid, kobber (II) bromid, kobber (II) trif lat, kobber (I) klorid, kobber (I) bromid og kobber (I) trif lat. Kobberkatalysatoren er vanligvis tilstede i mengder på minst omlag 5 mol-%, og vanligvis omlag 10 til 20 mol-%. Reaksjonen er i nærvær av et inert, dipolart, aprotisk løsningsmiddel, såsom dimetylformamid, tetrahydrofuran, dimetoksyetan, N-metyl-pyrro-lidinon, dimetylacetamid eller dimetylsulfoksyd, og i nærvær av en organisk base såsom pyridin eller dimetylaminopyridin, eventuelt i nærvær av en tertiær aminbase såsom trietylamin eller diisopropyletylamin. Den organiske basen er vanligvis tilstede i mengder på 1 til 2 mol ekivalenter, vanligvis 1,5 mol ekvivalenter.
Reaksjonstemperaturen avhenger av hvorvidt Hal i formelen IV er jod, brom eller klor. Når Hal er jod, kan reaksjonen foregå ved omlag 10 til 40°C, og fordelaktig ved omgivende temperatur såsom omlag 2 0 til 25°C. Når Hal er brom, er reaksjonstemperaturen fra omlag 20 til 50°C. Når Hal er klor, er reaksjonstemperaturen omlag 50 til omlag 100°C, vanligvis omlag 70°C, og reaksjonen utføres i en lukket beholder som forårsaker et reaksjonstrykk på mellom en atmosfære og omlag to atmosfærer.
Når Hal i formelen IV er jod eller klor, foregår reaksjonen i det minste til å begynne med i fravær av luft, f.eks. ved innføring av en inert gass såsom nitrogen til reaksjonsbe-holderen, eller ved å utføre reaksjonen i en lukket beholder.
Det ble funnet at høye utbytter oppnås ved å anvende omlag 2 mol ekvivaltenter av reagenset med formelen R3NH2, omlag 1,5 mol ekvivalenter av den organiske basen pyridin i dimetylformamid, og omlag 0,2 mol ekvivalenter av kobberkatalysatoren.
Reaksjonen kan også utføres med en ekvivalent av R3NH2, en ekvivalent kobber eller dets salter og 1,5 ekvivalent pyridin i dimetylformamid.
Isatonsyreanhydridene med formelen II fremstilles fra forbindelser med formelen III ved omsetning med et reagens med formelen R5R6C=0, hvor R5 og R6 er som definert ovenfor. F.eks. er reagenset fosgen eller fortrinnsvis bis-(triklormetyl)-karbonat (trifosgen) som er kommersielt tilgjengelig og, som et fast stoff, lett å håndtere. Når reagenset ovenfor er et fast stoff, utføres reaksjonen i et inert løsningsmiddel såsom klorert alkan, f.eks. metylenklorid, kloroform, karbontetra-klorid eller dikloretan, eller et aromatisk løsningsmiddel såsom toluen, benzen eller xylen. Reaksjonen utføres ved omlag -10 til 15°C i omlag 15 minutter til 1,5 timer, vanligvis i en halv time. Når reagenset er fosgen, kan løsnings-middlet også være en vandig syre såsom saltsyre. Når reagenset er en væske såsom metylklorformat elle etylklorformat, kan løsningsmiddelet utelates, og et overskudd av reagenset kan anvendes isteden. Reaksjonsblandingen oppvarmes deretter til mellom omlag 150 og 200°C i omlag 18 til 24 timer.
Omsetningen for å danne isatonsyreanhydridene med formelen II foregår i nærvær av en organisk base såsom pyridin eller dimetylaminopyridin, eventuelt i nærvær av et tertiært amin såsom trietylamin eller diisopropyletylamin.
Kinolonene med formelen I fremstilles fra isatonsyreanhydridene II ved omsetning med minst omlag en ekvivalent av alkalimetallsaltet av C^ Cj alkyl 3-hydroksyakrylat. Alkali-metallet er natrium, litium eller kalium. Reaksjonen utføres i et dipolart aprotisk løsningsmiddel såsom dimetylformamid, tetrahydrofuran, dimetoksyetan, N-metylpyrrolidinon eller dimetylacetamid. Reaksjonstemperatuen ligger i området fra omlag 20 til 100°C, vanligvis omlag 50°C, og reaksjonstiden er omlag 1 til 24 timer, vanligvis omlag 1 time. Reaksjonen utføres fordelaktig i nærvær av et chelateringsmiddel for alkalimetallioner. Eksempler på egnede chelateringsmidler er N, N' -dimetylimidazolidinon, heksametylf osf orsyretriamid, N,N' -dimetylpropylenurea og tris [2-(metoksyetoksy) etyl] amin.
Kinolonene med formelen I hvori R4 er hydroksy, kan fremstilles fra de tilsvarende esterne med formelen I hvori R4 er C^-C^ alkoksy ved konvensjonell hydrolyse, f.eks. ved oppvarming med en syre såsom saltsyre.
De følgende eksemplene illustrerer oppfinnelsen.
Eksempel I
Til en 1 liters rundbunnet, firehals-kolbe utstyrt med mekanisk rører, to dråpetrakter og et termometer ble innført 20 g (86,71 mmol) av 2-amino-4,5-dif luorbenzosyre og en løsning av 12,3 ml konsentrert svovelsyre i 90 ml vann. Vellingen ble avkjølt til mellom 0 og -5°C i et is-acetonbad. En av dråpetraktene ble fylt med en løsning av 6,57 g (95,22 mmol) natriumnitritt i 30 ml vann og langsom tilsetning av. løsningen ble begynt. Den indre reaksjonstemperaturen steg aldri over 0°C, og hele løsningen ble innført i løpet av 5 minutter. Den andre dråpetrakten ble fylt med en løsning av 21,6 g (128,31 mmol) kaliumjodid i 4 5 ml av IN svovelsyre. Denne løsningen ble deretter tilsatt dråpevis over en periode på 10 minutter med den indre temperaturen ved eller under 0°C. Under tilsetningen frigjør reaksjonsblandingen nitrogengass som forårsaker noe skumming. Etter at tilsetningen var fullstendig, ble den mørke blandingen omrørt over natten under langsom oppvarming til romtemperatur. Reaksjonsblandingen ble behandlet med en løsning av 30 g natriumbisulfitt i 165 ml vann, og suspensjonen ble justert til pH 2,5 med 5 ml av 6N saltsyre. Det resulterende slam ble omrørt ved 0°C i 3 0 minutter og deretter filtrert. Rensing av det mørke materi-alet ble utført ved å løse det meste av det faste stoffet i etylacetat, etterfulgt av klaring og behandling med aktivert trekull. Etter filtrering gjennom filteraid (celitt) og inndamping, ble det oppnådd 20 g (82%) av 2-jod-4,5-difluor-benzosyre; sp: 126-127°C.
Eksempel II
Til en 35 ml rundbunnet, enhals-kolbe utstyrt med magnetisk rørestav og nitrogeninnløp ble fylt 45 mg (0,704 mmol) kobberbronze, 5 ml vannfri dimetylf ormamid (DMF) , 430 /il (5,28 mmol) pyridin og 537 /j1 (7,75 mmol) cyklopropylamin. Den resulterende oppslemmingen ble derette behandlet med en løsning av 1 g (3,52 mmol) 2-jod-4,5-difluorbenzosyre i 5 ml DMF, og blandingen ble omrørt over natten ved romtemperatur. Reaksjonsblandingen, nå nesten en løsning, ble klaret, og deretter tilsatt til vann (100 ml) ved pH 4,5. En velling dannes øyeblikkelig, men før filtrering ble blandingen en gang til justert til pH 4,5 med 6N saltsyre og avkjølt til 0°C. Filtrering av det hvite faste stoffet ga 0,720 g (95%) av 2-N-cyklopropylamino-4 , 5-dif luorbenzosyre; sp: 174-176 °C.
Eksempel III
I en 10 ml forseglbar trykkreaksjonskolbe utstyrt med magnetisk rører og teflonseptumlokk ble fylt en løsning av 1,0 g (5,19 mmol) 2-klor-4 , 5-dif luorbenzosyre, 792 jul , (11,43 mmol) cyklopropylamin, 800 mg (4,15 mmol) kobber (I) jodid og 630 /il (7,79 mmol) pyridin i 8,0 ml av N,N-dimetylacetamid. Flasken ble forseglet og ble oppvarmet til 70"C under omrøring i en periode på 16 timer. Reaksjonsblandingen fikk avkjøle seg til romtemperaur, og ble deretter tilsatt til 100 ml vann. Oppslemmingen ble justert til pH 13 med natriumhydroksyd-løsning, og ble omrørt i 15 minutter ved romtemperatur. Oppslemmmingen ble filtrert, og filtratet ble justert til pH 4,5 med konsentrert vandig HCl. Filtrering av den resulterende vellingen ga 451 mg (41%) av 2-cyklopropylamino-4,5-difluorbenzosyre; sp: 175-176°C.
Eksempel IV
I en 10 ml rundbunnet, enhals-kolbe utstyrt med en septum og magnetisk rørestav ble fylt en løsning av 100 mg (0,46 mmol) av 2-N-cykloproplamino-4, 5-dif luorbenzosyre og 62 jizl (0,44 mmol) av trietylamin i 2 ml metylenklorid. Løsningen ble avkjølt til 0°C og ble behandlet med en løsning av 4 5 mg (0,147 mmol) bis-(triklormetyl)-karbonat i 0,5 ml metylenklorid. Til slutt ble innført en katalytisk mengde av dimetylaminopyridin (10 mg) som en løsning i metylenklorid (0,5 ml). Etter omrøring ved 0°C i 1,5 timer, ble reaksjonsblandingen quenchet ved tilsetning av en liten mengde IN saltsyre. Den organiske fasen ble tørket over natriumsulfat, og dereter konsentrert til en gul olje for å gi 114 mg av N-cyklopropyl-6,7-difluor-2H-3 , l-benzoksazin-2,4(1H)dion (100%). Produktet ble krystallisert fra varm etanol; sp: 138-139°C.
Eksempel V
I en 15 ml rundbunnet, enhals-kolbe utstyrt med magnetisk rører og under nitrogenatmosfære ble tilsatt 60 mg (0,484 mmol) av natriumsaltet av metyl 3-hydroksyakrylat i 1,5 ml DMF. Den resulterende løsningen ble omrørt i nærvær av 4Å molekylsikter over natten, og deretter filtrert i et annet reaksjonskar utstyrt med kondensator, nitrogenledning og magnetrører. I blandingen ble fylt 52 /ul (0,467 mmol) av N,N'-dimetylimidazolidinon, og løsningen ble oppvarmet til 55°C. Til denne reaktoren ble tilsatt en løsning av 93 mg (0,388 mmol) av N-cyklopropyl-6, 7-difluor-2H-3,1-benzoksazin-2,4(1H)dion i 1,5 ml DMF. Reaksjonsblandingen ble omrørt ved 55°C i en time. Systemet fikk avkjøle seg til romtemperatur, og ble deretter tilsatt til 3 0 ml vann ved pH 4,0. pH i den resulterende oppslemmingen ble justert til 5,5, og blandingen ble avkjølt til 0°C og filtrert. Etter tørking ble det oppnådd 50 mg (46%) av metyl l-cyklopropyl-6,7-difluor-1,4-dihydro-4-okso-3-kinolinkarboksylat, sp: 223-224°C. Filtratet ble ekstrahert med metylenklorid og etter tørking og inndamping ble oppnådd ytterligere 47 mg (43,5%, totalt utbytte: 89,5%) av det ønskede produktet.
Eksempel VI
En oppslemming av trietylamin (15 ml, 0,11 mol), 2,4-difluoranilin (25 ml, 0,24 mol), kobberbronze (2,7 g, 0,04 mol) , i varm DMF (25 ml) ble behandlet med en løsning av 2-klor-4,5-difluorbenzosyre (23 g, 0,12 mol) i 25 ml DMF, og tempraturen ble holdt ved 85°C i 8 timer. Reaksjonsblandingen fikk avkjøle seg til romtemperatur, og ble deretter omrørt over natten. Reaksjonsblandingen ble inndampet i vakuum, og residuet ble fordelt mellom eter og vandig ammoniumklorid. Den organiske fasen ble vasket med 2N HC1 og mettet vandig litiumkloridløsning. Eteren ble tørket over natriumsulfat, behandlet med darko og deretter filtrert og inndampet. Residuet ble krystallisert fra heksaneter for å gi 21,6 g (62%) av 2-(2 , 4-dif luorf enylamino) -4 , 5-dif luorbenzosyre; sp: 215-216°C.

1. Forbindelse med formelen
karakterisert ved at R3 er cyklopropyl, 4-fluorfenyl eller 2,4-difluorfenyl, og X er fluor eller klor.
2. Fremgangsmåte for fremstilling av forbindelser med formel II ifølge krav 1,karakterisert ved omsetning av en forbindelse med formelen hvori X og R3 er som definert i krav 1 eller 2 med en forbindelse med formelen hvori R5 og R6 begge er klor eller triklormetyloksy, eller R5 er klor og R6 er C±- C6 alkoksy, triklormetyloksy, fenoksy eller fenoksy substituert med en substituent som er inert under reaksj onsbetingelsene.
3. Fremgangsmåte ifølge krav 2,karakterisert ved at R5 og R6 begge er triklormetyloksy eller klor, eller R5 er klor og R6 er triklormetyloksy.
