R<9> er et hydrogenatom, azido, eller resten -OR13 eller -NR<14>R15 (med R<13> i betydning et hydrogenatom eller en eventuelt med en hydroksygruppe substituert C^- q-alkylgruppe og med R<14> og R<15> i betydningen hydrogenatom eller en C1_6-alkylgruppe), samt deres fysiologisk fordragelige salter.
Det er kjent fra (DE-OS 2 854 353, DE-OS 3 202 118) at stilbenderivater har farmakologiske virkninger ved topisk og systemisk terapi av neoplasier, akne, psoriasis og andre dermatologiske forstyrrelser. Deres virkning er imidlertid ikke alltid tilfredsstillende.
Det ble nå funnet at 1-substituerte tetraliner med formel
I samt deres fysiologisk fordragelige salter, har et bedre virkningsspektrum.
Forbindelsene kan ifølge oppfinnelsen fremstilles ved at man omsetter en karbonylforbindelse med formel II hvori R<1>, R<2> og R<3> har de ovenfor angitte betydninger, med en fosforforbindelse med formel III
hvor R<21> er hydrogenatom, en Ci_4-alkylgruppe, nitrilgruppen eller en gruppe -COOR<22>, og R<20> og R22 betyr en C1_3<->alkylgruppe ifølge Wittig-Horner, og om ønsket,
overføres erholdte syreazider med formel I i syreamider ved omsetning med tilsvarende aminer;
overføres erholdte nitriler med formel I i amider ved hydrolyse, eller i aldehyder ved DIBAH-reduksjon,
eller i aminer ved LiAlH4-reduksjon, som igjen kan overføres i karboksylsyreamider ved omsetning med tilsvarende karboksylsyre eller reaktivt derivat derav;
overføres erholdte karboksylsyrer med formel I i estere ved forestring med tilsvarende alkohol, eller i alkoholer ved reduksjon, som igjen kan overføres i etere derav ved foretring med den tilsvarende alkohol, eller karboksylestere ved omsetning med tilsvarende karboksylsyre eller reaktivt derivat derav, eller i syreazider ved omsetning med aktiveringsmiddel og deretter metallazid.
Hensiktsmessig arbeider man i et løsningsmiddel i nærvær av de vanlige basiske forbindelser for Wittig-Horner-reaksjoner.
Omsetningen ifølge Wittig-Horner går ved en temperatur på opptil 100°C, hensiktsmessig ved 20 til 50°C. Omsetningen kan utføres ved et atmosfæretrykk eller i en lukket beholder under høyere trykk, eventuelt under oppvarming til det angitte temperaturområdet.
Man kan utføre denne omsetningen i nærvær av et fortynnings-eller løsningsmiddel, f.eks. en lavere mettet dialkyleter, di-alkylglykoleter eller cyklisk eter, såsom dietyleter, etyl-tert.-butyleter, 1,2-dimetoksyetan, tetrahydrofuran eller dioksan,
et aromatisk hydrokarbon såsom benzen eller et alkylbenzen, såsom toluen eller xylen, eller et mettet alifatisk hydrokarbon, såsom heksan, heptan eller isooktan, et lavere alifatisk keton, såsom aceton, metyletylketon eller metylisobutylketon, en dialkylformamid såsom dimetyl- eller dietylformamid, eller i blandinger av de nevnte løsningsmidler. Fortrinnsvis anvender man cykliske etere som dioksan eller tetrahydrofuran samt spesielt dimetylformamid eller blandinger derav, hvorunder omsetningen i alminnelighet forløper ved en temperatur opptil 30°C.
Omsetningen foretas i nærvær av et deprotoneringsmiddel
for fosfatet (III). Alkalimetallhydrider og alkalimetallamider er egnet, spesielt av natrium og kalium, natrium- og kalium-saltene av dimetylsulfoksyd, alkyllitiumforbindelser såsom n-butyllitium eller alkalimetallalkoholater, fortrinnsvis natrium-metanolat og natriumetanolat.
Man kan få forbindelsen fremstilt også
ved Wittig-reaksjon av et fosfoniumsalt med formel IV

hvorunder R har den nevnte betydning.
Wittig- eller Wittig-Horner-reaksjonen gir normalt blandin-
ger av sterisk isomere (E/Z)-olefiner.
E/Z-Isomerblandingene med overveiende Z-andel omlagres under innvirkning av lys ved den olefiniske dobbeltbinding til blandinger med høyere andel av (E)-isomerer. Ut fra de erholdte (E/Z)-isomerblandinger med nå gunstigere (E)-innhold fremstilles med fordel rene (E)-forbindelser med formel (I), fortrinnsvis ved krystallisering eller en kromatografisk metode såsom søyle- eller preparativ HPLC-kromatografi.
Fotoisomeriseringen utføres fortrinnsvis i løsning. Egnede løsningsmidler er polare protiske og aprotiske løsningsmidler, f.eks. metanol, etanol, eddikester, tetrahydrofuran, aceton. Konsentrasjonen av bestrålt løsning ligger på 0,1 til 50, fortrinnsvis 1 til 15 vektprosent.
Man kan bestråle i nærvær av sensibilisatorer såsom f.eks. acetofenon, 4-metoksyacetofenon, propiofenon, benzen, aceton, benzofenon, benzil, Michlers keton. Særlig foretrukket er her aceton.
Som lyskilde for utførelse av den nevnte fotoreaksjon kan man med fordel anvende kunstige stråler hvis emisjon i det minste delvis ligger i området fra 200 til 600 nm, fortrinnsvis i området fra 300 til 400 nm. Med fordel anvendes kvikksølv-damp, fluor, xenon eller wolframlamper, lysstoffrør eller kull-buelamper.
Bestrålingstemperaturen er avhengig av typen av det anvendte løsningsmiddel. Særlig foretrukket er området fra +10 til +30°C. Strålingsvarmen kan ledes vekk ved lampekjøling og/eller kjøling av reaksjonsblanding, hvorunder det i lampekjølekretsen kan anvendes destillert vann eller på kjent måte filtrerende løs-ninger med tilsetninger.
De nødvendige ketoner og aldehyder med formel II for Wittig-Horner-reaksjonen lar seg f.eks. fremstille ved acylering av de tilsvarende tetrahydrotetrametylnaftaliner i nærvær av en Lewis-syre. Særlig egnet som acyleringsmiddel er acylhaloge-nidet, fortrinnsvis acylkloridene. Foretrukne Lewis-syrer er jern(III)-klorid, aluminium(III)klorid og titan(IV)klorid. Formyleringen kan med fordel utføres med heksametylentetramin/- trifluoreddiksyre. Tetrahydrotetrametylnaftalinene er beskrevet i US-pat- 3.44 2.640 og US-pat. 3 .499 . 751, eller kan fremstilles etter de deri beskrevne fremgangsmåter ut fra 2,5-diklor-2,5-dimetylheksan og et tilsvarende substituert benzen ved Friedel-Craft-alkylering.
Fosfoniumsaltene med formel IV kan fremstilles som følger: En karbonylforbindelse med formel II reduseres først ved hjelp av et kompleks metallhydrid såsom natriumborhydrid eller litiumaluminiumhydrid til den tilsvarende alkohol. Denne halogeneres med et fosforhalogenid, såsom fosfortribromid eller fosforoksy-klorid i nærvær av en base såsom pyridin. Etterfølgende omsetning med trifenylfosfin gir fosfoniumsalt med formel IV.
Benzosyreesteren med den generelle formel I hvori n = 0 og R betyr en karboalkoksygruppe overføres hvis dette er ønskelig
i de frie karboksylsyrer og deres fysiologisk fordragelige salter ved esterforsåpning. Omvendt kan man naturligvis forestre den frie syre på kjent måte.
Hensiktsmessig utføres forsåpningen/forestringen i nærvær av et fortynnings- eller løsningsmiddel, f.eks. en dialkyl-glykoleter eller cyklisk eter, såsom 1,2-dimetoksyetan, tetra-hydrof uran eller dioksan, et lavere alifatisk keton, såsom aceton, metyletylketon eller metylisobutylketon, eller i en lavere alifatisk alkohol, såsom metanol, etanol, propanol eller isopropanol, eventuelt i nærvær av vann, henholdsvis i blandinger av de nevnte løsningsmidler med vann.
Foretrukne løsningsmidler er vandige blandinger av etanol og metanol, hvorunder omsetningen utføres ved reaksjonsblandingens kokepunkt.
Forsåpningen finner fortrinnsvis sted i nærvær av alkali,såsom alkalimetallhydroksyder, alkalimetallkarbonater og -hydrogenkarbonater, spesielt av natrium og kalium, organiske tertiere baser, såsom pyridin eller lavere trialkylaminer, såsom tri-metyl- eller trietylamin i blandinger med vann. Derunder anvendes basen i forhold til esteren i støkiometrisk mengde eller i et lite overskudd. Fortrinnsvis anvendes natrium- eller kaliumhydroksyd.
Amidene fremstilt ifølge oppfinnelsen kan fremstilles på
i og for seg kjent måte idet man først overfører de tilsvarende benzosyrer i karbonylaktive derivater, f.eks. i syrehalogenidene,
-azidene, -imidazolidene, eller -anhydridene, O-acyl-N,N'-di-cykloheksylisoureaer eller p-nitrofenylestere, og behandler

disse med aminer HNR R . I tilfelle av spesielt reaktive
aminer, fremfor alt ammoniak, er en direkte amidolysé av estere (med resten -OR"'"<0>) foretrukket.
Et halogenid av en karboksylsyre, fortrinnsvis syrekloridet, kan ved reaksjon med 2-aminoetanol og etterfølgende cyklisering overføres i oksazolinderivat med formel I.
En karboksylsyre, en karboksylsyreester eller en karbonsyre-amid med formel I kan reduseres på i og for seg kjent måte til de tilsvarende alkoholer, henholdsvis aminer. Med fordel utfø-res reduksjonen ved hjelp av metallhydrid eller alkalimetall-hydrid i nærvær av et egnet løsningsmiddel. Som metallhydrider anvendes fortrinnsvis komplekse metallhydrider såsom litiumaluminiumhydrid eller diisobutylaluminiumhydrid. Som løsnings-middel anvendes ved arbeider med litiumaluminiumhydrid etere, såsom dietyleter, dioksan eller tetrahydrofuran. Utfører man reduksjonen med diisobutylaluminiumhydrid eller et alkoksy-natriumaluminiumhydrid, foretrekkes anvendelsen av hydrokarboner såsom heksan eller toluen.
Et amin eller en alkohol med formel I kan oppnås på i og for seg kjent måte med et alkanoylhalogenid eller -anhydrid,
et aralkylhalogenid eller et anhydrid eller et aroyl- eller heteroaroylhalogenid eller anhydrid, hensiktsmessig i et inert fortynnings- eller løsningsmiddel, f.eks. en lavere alifatisk keton såsom aceton, metyletylketon eller metylisobutylketon,
et dialkylformamid såsom dimetylformamid eller dietylformamid eller med overskudd av acyleringsmiddel som fortynnings- eller løsningsmiddel overføres i de tilsvarende amider og estere. Fortrinnsvis utføres omsetningen i nærvær av en base som syre-bindende middel i et temperaturområde som ligger mellom -20°C
og reaksjonsblandingens kokepunkt. Egnede baser er alkalimetallkarbonater, -hydrogenkarbonater, -hydroksyder eller alkoholater, spesielt av natrium og kalium, basiske oksyder såsom aluminiumoksyd eller kalsiumoksyd, organiske tertiere baser såsom pyridin, eller lavere trialkylaminer såsom trimetyl-eller trietylamin. Derunder kan basene anvendes i katalytisk mengde eller i støkiometrisk mengde, henholdsvis i lite overskudd i forhold til det anvendte alkyleringsmiddel.
En alkohol med formel I kan omsettes med alkylhalogenider 12 12 12
R -J, R Br eller R -Cl i nærvær av alkalimetallhydrider, fortrinnsvis natriumhydrid, eller i nærvær av alkyllitium-forbindelse, fortrinnsvis n-butyllitium i et organisk løsningsmiddel såsom tetrahydrofuran, dioksan, 1,2-dimetoksyetan, metyl-tert.-butyleter eller ved anvendelse av natriumhydrid også i dimetylformamid i et temperaturområde mellom -10°C og 40°C til en eter med formel I.
En alkohol med formel I kan med egnede oksydasjonsmidler, fortrinnsvis mangan(IV)-oksyd, eventuelt på et uorganisk bære-materiale såsom kieselgel eller aluminiumoksyd oksyderes til et aldehyd med formel I. Med fordel arbeider man i et inert organisk løsningsmiddel , f.eks. et hydrokarbon såsom heksan eller i en eter såsom f.eks. tetrahydrofuran eller i blandinger av de nevnte løsnings- og fortynningsmidler i temperaturområdet mellom -10°C og 30°C. Den nødvendige reaksjonstid er i det vesentlige avhengig av oksydasjonsaktiviteten til det anvendte mangan(IV)-oksyd.
Et aldehyd med formel I kan man også oppnå ved reduksjon av det tilsvarende nitril med formel I med diisobutylaluminiumhydrid i et løsningsmiddel, fortrinnsvis i toluen, heksan, tetrahydrofuran eller blandinger av disse løsningsmidler i et temperaturområde mellom -4 0°C og romtemperatur.
En karbonylforbindelse med formel I (i tilfelle n = 0) kan det omsettes med en fosforforbindelse med formel VI eller VII
hvori R 20 har den angitte betydning i en Wittig-Horner-reaksjon, og hensiktsmessig arbeider man i et løsningsmiddel, fortrinnsvis tetrahydrofuran, dimetylformamid eller dimetylsulfoksyd, og i nærvær av vanlige baser for slike defineringer som f.eks. natriumhydrid eller natriumetanolat. Omsetningen forløper ved en temperatur opptil 100°C, hensiktsmessig ved 20 til 50°C.
Nitril-henholdsvis estergruppen omvandles deretter om ønsket ifølge de foran- og etterfølgende beskrevne metoder i andre funksjonelle grupper.
Et nitril med formel I kan på i og for seg kjent måte under syre eller med fordel basekatalyse forsåpes til den tilsvarende karboksylsyre. Som baser anvendes fortrinnsvis alkalimetallhydroksyder, spesielt kaliumhydroksyd som anvendes i overskudd. Som løsningsmiddel anvendes som regel alkoholer som er
blandbare med vann som f.eks. metanol, etanol, isopropanol eller n-butanol. Omsetningen utføres normalt ved reaksjonsblandingens kokepunkt.
Ut fra nitrilene med formel I kan man ved addisjon av et azid, f.eks. et alkalimetallazid, fortrinnsvis natriumazid i nærvær av aluminiumklorid eller ammoniumklorid fremstille de tilsvarende tetrazoler med formel I. Som løsningsmiddel anvender man fortrinnsvis cykliske etere såsom dioksan eller tetrahydrofuran, samt spesielt dimetylformamid og blandinger derav, hvorunder omsetningen generelt forløper ved en.temperatur fra 60 - 100°C.
Noen av forbindelsene fremstilt ifølge oppfinnelsen har et surt hydrogenatom og kan derfor med baser overføres på vanlig måte i et fysiologisk fordragelig, godt vannløselig salt. Egnede salter er f.eks. ammonium-, alkalimetall-, spesielt av natrium, kalium og litium, jordalkalimetallsalter, spesielt av kalsium eller magnesium, samt salter med egnede organiske baser såsom med lavere alkylaminer, f.eks. metylamin, etylamin eller cykloheksylamin, eller med substituerte lavere alkylaminer, spesielt hydroksysubstituerte alkylaminer, såsom dietanolamin, trietanolamin eller tris(hydroksymetyl)-aminometan, samt med piperidin eller morfolin.
Eventuelt overføres de erholdte aminer med formel (I) ifølge kjente fremgangsmåter i syreaddisjonssaltet av en fysiologisk fordragelig syre. Som vanlig fysiologisk fordragelig organisk eller uorganisk syre kommer f.eks. saltsyre, hydrogen-bromid, fosforsyre eller svovelsyre i betraktning, og som organiske syrer f.eks. oksalsyre, maleinsyre, fumarsyre, melke-syre, vinsyre, eplesyre, sitronsyre, salicylsyre, adipinsyre eller benzosyre eller er å finne i Fortschritte der Arznei-mittelforschung Band 10, s. 224-225, Birkhauser Verlag,
Basel og Stuttgart, 1966.
Forbindelsene fremstilt ifølge oppfinnelsen og deres fysiologisk fordragelige salter kan på grunn av sine farmakologiske egenskaper finne anvendelse ved topisk og systemisk terapi og også profylakse av prekanzeroser og karzinomer i huden, slim-hinnen og de innvendige organer, samt ved topisk og systemisk terapi av akne, psoriasis og andre dermatologiske sykdommer som har forbindelse med patologisk forandret horndannelse samt ved behandling av revmatiske sykdommer, spesielt sådanne av betennel-ses- eller degenerativ karakter som angriper ledd, muskler, sener og andre deler av bevegelsesapparatet. Et foretrukket indikasjonsområde er ved siden av terapi av dermatologiske sykdommer den profylaktiske og terapeutiske behandling av prekanzeroser og tumorer.
De farmakologiske virkninger kan f.eks. demonstreres i de etterfølgende forsøksmodeller. Forbindelsene fremstilt ifølge oppfinnelsen opphever keratiniseringen som begynner etter vitamin-A-mangel in vitro på marsvintreakealvev. Keratiniseringen tilhører den tidlige fase av karcinogenese som i en lig-nende teknikk in vivo etter igangsettelse med kjemiske forbindelser inhiberes ved energirik stråling eller etter viral celle-transformasjon med forbindelsene med formel (I) fremstilt ifølge oppfinnelsen. Denne metodikk er beskrevet i Cancer Res. 36, 964-972 (1976) eller Nature 250, 64-66 (1974) og Nature 253, 47-50 (1975).
Dertil inhiberer forbindelsen fremstilt ifølge oppfinnelsen formeringshastigheten til bestemte maligne forandrede celler. Denne metodikk er beskrevet i J. Nati. Cancer Inst. 60, 1035-1041(1978), Experimental Cell Research 117, 15-22 (1978) og Proe. Natl.Acad. Sei. USA 77, 2937-2940 (1980).
En antiartritiske virkning av forbindelsen fremstilt ifølge oppfinnelsen kan på vanlig måte bestemmes i dyreeksperiment i Adjuvans-artritis-modellen. Den dermatologiske aktiviteten, f.eks. for behandlingen av akne, kan blant annet påvises ved den komedolytiske aktivitet og evnen til å redusere antall cyster i Rhino-mus modellen.
Denne metode er beskrevet av L.H. Kligman et al i The Journal of Investigative Dermatology 73, 354-358 (1978) og av J.A. Mezick et al. i "Models of Dermatology", Ed. Maibach, Lowe, Vol. 2, 5.59-63, Karger, Basel 1985.
Forsøkssubstansen i egnet vehikel ble brakt topisk på
hele ryggpartiet til Rhino-musen (100 yl) en gang daglig i fem på hverandre følgende dager pr. uke i to uker. Omtrent 72 timer
etter siste behandling ble rygghuden fjernet og lagt 18 timer ved 4-6°C i 0,5%-ig eddiksyre. Deretter ble en ca. 2 x 5 cm stor flate skåret ut, epidermet skrellet av, brakt på et objekt-glass (med den dermale side oppad) og spylt vannfri ved hjelp av alkohol/xylen inntil epidermet syntes gjennomsiktig. Prøven ble fiksert ved overtrekning med Permount og vurdert mikrosko-pisk. Diametere av 10 utriculi ble målt i 5 fritt valgte felt og derfra ble ved sammenligning med den ubehandlede kontroll-gruppe den gjennomsnittlige reduksjon av utriculustverrsnittet beregnet. Den følgende tabell viser de oppnådde resultater.
De følgende eksempler anskueliggjør oppfinnelsen:
A. Fremstilling av utgangsforbindelser
2-Formyl-5,6,7,8-tetrahydro-l-alkoksy-3,5,5,8,8-pentametyl-naftalin
340 ml Dimetylsulfoksyd og 32,2 g kaliumhydroksyd røres
5 minutter, deretter tilsetter man i rekkefølge 0,1 mol 2-formyl--5, 6,7,8-tetrahydro-3,5,8,8-pentametyl-l-naftol og 0,24 mol alkylhalogenid, hvorunder temperaturen stiger noe. Man lar dette stå natten over ved romtemperatur, rører og ekstraherer deretter med eter/vann. Eterfasen vaskes flere ganger med vann, tørkes (Na2SO^) og inndampes. Det således erholdte råprodukt renses om nødvendig ved destillasjon.
Med denne fremgangsmåten ble de følgende forbindelser fremstilt: 1- Etoksy-2-formyl-5,6,7,8-tetrahydro-3,5,5,8,8-pentametyl-naftalin, smp. 130-1323°C (0,2 mbar), utbytte 87%.
2- Formyl-5,6,7,8-tetrahydro-3,5,5,8,8-pentametyl-l-propyl-oksynaftalin, utbytte 100%(rått).
1-(Metyl)etoksy-2-formyl-5,6,7,8-tetrahydro-3,5,5,8,8--pentametylnaftalin, smp. 112-117°C (0,1 mbar), utbytte 70%. 1- Butyloksy-2-formyl-5,6,7,8-tetrahydro-3,5,5,8,8-penta-metylnaftalin, smp. 140°C (0,2 mbar), utbytte 82%.
2- Formyl-l-heksyloksy-5,6,7,8-tetrahydro-3,5,5,8,8-penta-metylnaftalin, utbytte 100% (rått).
B. Fremstilling av sluttprodukter
Eksempel 1
(E)-4-C2-(5,6,7,8-Tetrahydro-l-metoksy-3,5,5,8,8-pentametyl--2-naftyl)-1-etenylJ benzosyreetylester
Til en suspensjon av 25 ml absolutt dimetylsulfoksyd og 0,75 g (25 mmol) natriumhydrid (80%-ig, først befridd for 20%-ig paraffindel med petroleter), ble med begynnelse ved romtemperatur tildrvppet en løsning av 7,5 g (25 mmol) 4-karboksyetyl-benzylfosforsyredietylester i 12 ml dimetylsulfoksyd. Deretter rørte man enda 30 minutter ved 35 - 40°C. Så ble i løpet av 10 minutter 3 g (12,5 mmol) 2-formyl-5,6,7,8-tetrahydro-l--metoksy-3,5,5,8,8-pentametyl-naftalin oppløst i 12 ml dimetylsulfoksyd og 1 ml tetrahydrofuran dryppet til. Man lot røre 3 timer etterpå ved romtemperatur. Så ble satsen helt i 300 ml vann og surgjort med 2 N saltsyre. Den klebrige resten ble skilt fra, rørt med metanol og filtrert. Etter søylekromato-grafisk rensing av det faste stoff (kiselgel; toluen) fikk man 1,9 g av tittelforbindelsen, smp. 123,2°C.
HPLC-Analysen (Si 60, 5 ym, 150 bar, n-heptan/eddikester (97:3), t_ = 6,7 min) har en isomerrenhet større enn 98%.
Eksempel 2
(E)-4-f2-(5,6,7,8-Tetrahydro-l-metoksy-3,5,5,8,8-pentametyl--2-naftyl)-1-etenyl]benzonitril
Til en suspensjon av 3 g (0,1 mol) natriumhydrid (80%-ig, først befridd for 20%-ig paraffinandel med petroleter), i 100 ml absolutt dimetylsulfoksyd ble ved 2 5 til 40°C tildryppet en løs-ning av 25,2 g (0,1 mol) 4-cyanobenzylfosfonsyredietylester i 50 ml dimetylsulfoksyd. Man rørte enda 1 time og deretter ble en løsning av 12 g (50 mmol) 2-formyl-5,6,7,8-tetrahydro-l--metoksy-3,5,5,8,8-pentametyl-naftalin i 100 ml dimetylsulfoksyd dryppet til. Man lot røre natten over ved romtemperatur. Deretter ble reaksjonsblandingen helt i 1,2 1 vann og surgjort med 2 N saltsyre. Det dannede oljeaktige produkt skilt fra, rørt med litt metanol, og det dannede krystallisat ble filtrert fra. Det faste stoff ble ettervasket med metanol på filteret. Etter tørking ble 13,8 g av tittelforbindelsen tilbake, smp. 120°C.
Eksempel 3
(E)-4-f 2-(5,6,7,8-Tetrahydro-l-metoksy-3,5,5,8,8-penta-metyl-2-naf tyl) -1-etanyl] benzosyre
5,1 g (15 mmol) (E)-4-f2-(5,6,7,8-tetrahydro-l-metoksy--3,5,5,8,8-pentametyl-2-naftyl)-1-etenyl] benzonitril fra eksempel 2, 75 ml etanol og 75 ml 10 N natronlut ble oppvarmet under tilbakeløp (inntil slutten av omsetningen, ca. 3 timer). Reak-sjonsblandingen ble etter avkjøling helt i 750 ml is/vann og nøytralisert med kons. saltsyre. Det dannede faste stoff ble filtrert fra, vasket nøytralt med vann, ettervasket med metanol og tørket i nitrogenstrøm. Man fikk 3,5 g av tittelforbindelsen, smp. 222-223°C (etanol/H20).
Forbindelsene som er angitt i den etterfølgende tabell
ble enten fremstilt ved Wittig-Horner-reaksjon (analogt eksempel 2) eller ved forsåpning av de tilsvarende nitriler (analogt eksempel 3):

Eksempel 21
(E)-4-[2-(5,6,7,8-Tetrahydro-l,3-dimetoksy-5,5,8,8-tetra-metyl-2-naftyl)-1-etenyl]benzosyreazid
Til en løsning av 7,7 g (20 mmol) av karboksylsyren fra eksempel 14 i 35 ml aceton dryppet man ved 0°C først 3,4 ml trietylamin i 17 ml aceton, og deretter 2,6 ml (27 mmol) klormaursyre-etylester. Man lot røre 4 0 minutter videre ved 0°C og dryppet så en løsning av 2 g (31,4 mmol) natriumazid i 4,3 ml vann til dette, likeledes ved 0°C. Så rørte man 2 timer ved 0°C, og deretter filtrerte man krystallisatet fra og ettervasket med vann og etanol. Etter tørking ble 6,6 g av tittelforbindelsen tilbake, smp. 125-126°C (spaltning).
Eksempel 22
(E)-4-[2-(5,6,7,8-Tetrahydro-l,3-dimetoksy-5,5,8,8-tetra-metyl-2-naftyl)-1-etenyl]benz(2-hydroksyetyl)amid
Til en løsning av 3 g (8 mmol) av syreazidet som er beskrevet i det forutgående eksempel 21 i 200 ml abs. THF dryppet man 16 ml etanolamin. Etter 1,5 timers henstand inndampet man satsen delvis og helte i vann. Man surgjorde med 2 N HC1, filtrerte det utfelte faste stoff fra, vasket flere ganger med vann og litt metanol og tørket. Man fikk således 2,7 g av tittelforbindelsen, smp. 201-203°C.
Eksempel 2 3
(E)-4-[2-(5,6,7,8-Tetrahydro-l,3-dimetoksy-5,5,8,8-tetra-metyl-2-naftyl)-1-etenyl]benz(n-butyl)amid
Analogt eksempel 2 2 fikk man ut fra 3 g (7,7 mmol) av syreazidet som er beskrevet i eksempel 21 og 30 ml n-butylamin 2,3 g av tittelforbindelsen, smp. 137-140°C etter omkrystallisering fra metanol.
Eksempel 24
(E)-4- [2-(5,6,7,8-Tetrahydro-l,3-dimetoksy-5,5,8,8-tetra-metyl-2-naftyl)-1-etenyl]benzamid
En blanding av 2 g (5,3 mmol) av nitrilet fra eksempel 5,
5 g kaliumhydroksydpulver og 40 ml tert.-butanol ble oppvarmet i 20 minutter under tilbakeløp. Etter avkjøling helte man reaksjonsblandingen på mettet natriumkloridløsning og ekstraherte to ganger med eter. Resten man fikk etter tørking (Na2S04) og inndampning av eterfasene ga etter omkrystallisering fra metanol 0,9 g av tittelforbindelsen, smp. 190-194°C.
Eksempel 2 5
(E)-4-[2-(5,6,7,8-Tetrahydro-l,3-dimetoksy-5,5,8,8-tetra-me tyl- 2-na f tyl ) -1-etenyl]benzosyremetylester
Til en løsning av 11,8 g (30 mmol) av karboksylsyren fra eksempel 14 i 120 ml abs. tetrahydrofuran og 2,7 ml (56 mmol) pyridin dryppet man 7,2 ml (60 mmol) tionylklorid i 10 ml tetrahydrofuran. Etter en reaksjonstid på 3 timer filtrerte man pyridinhydrokloridet fra og dryppet filtratet til 10 ml abs. metanol. Man lot røre ved romtemperatur natten over, helte så
i vann og ekstraherte tre ganger med eter. Eterfasene ble vasket to ganger med vann, tørket (Na2S0^) og inndampet. Resten ga etter omkrystallisering fra metanol 5,5 g av tittelforbindelsen, smp. 140-142°C.
Eksempel 26
(E)-4-[2-(5,6,7,8-Tetrahydro-l,3-dimetoksy-5,5,8,8-tetra-metyl-2-naftyl)-1-etenyl]benzaldehyd
Til en løsning av 10,2 g (27 mmol) av nitrilet fra eksempel 5 i 100 ml abs. eter satte man ved romtemperatur 47,3 ml (57 mmol) DIBAH-løsning (20%-ig i heksan). Man lot rørei ytterlige-re 40 minutter og dryppet deretter 150 ml mettet vinsyreløsning til. Etter 1 time ekstraherte man tre ganger med eter, vasket eterfasene to ganger med vann, tørket (Na2SO^) og inndampet. Resten ble omkrystallisert to ganger fra isopropanol. Man fikk 4,5 g av tittelforbindelsen, smp. 108°C.
Eksempel 2 7
(E)-4-f2-(5,6,7,8-Tetrahydro-l,3-dimetoksy-5,5,8,8-tetra-metyl-2-naftyl)-1-etenyl]benzylalkohol
Til en suspensjon av 1,6 g (42 mmol) litiumaluminiumhydrid i 200 ml abs. eter dryppet man en suspensjon av 14 g (35,5 mmol) karboksylsyre fra eksempel 14 i 116 ml eter, hvorunder blandin-gen kokte svakt. Man rørte 3 timer under tilbakeløp. Etter av-kjøling satte man i rekkefølge 50 ml eddikester, 100 ml vann og 150 ml 2 N HC1 til. Fasene ble adskilt og den vandige fase ekstrahert to ganger med eter. De samlede eterfaser ble vasket nøytrale, tørket (Na2S0^) og inndampet. Råproduktet ble først omkrystallisert fra heptan, og deretter underkastet en lynkro-matografi (Si 60, heptan med stigende andeler eddikester). Man fikk således 6,5 g av tittelforbindelsen, smp. 102-104°C.
Eksempel 28
(E)-4 — f2—(5,6,7,8-Tetrahydro-l,3-dimetoksy-5,5,8,8-tetra-metyl-2-naftyl)-1-etenyl<]> benzylmetyleter
Til en suspensjon av 0,3 g (11 mmol) natriumhydrid i 15 ml tørt dimetylformamid dryppet man 3,8 g (10 mmol) av benzylalko-holderivatet fra eksempel 27 oppløst i 10 ml tørt dimetylformamid. Man lot røre inntil hydrogenutviklingen (ca. 1 time) var ferdig, og dryppet deretter til 1,56 g (11 mmol) jodmetan under iskjøling. Man oppvarmet deretter 5 timer ved 60°C. Neste dag dryppet man vann til og filtrerte det utfelte faste stoff fra. Dette ble løst i varm metanol. Ved avkjøling til romtemperatur dannet det seg to faser. Den øvre (metanol)-fase ble oppvarmet igjen og så stilt i kjøleskap. De utfelte krystaller ble filtrert fra og tørket. Man fikk således 2,1 g av tittelforbindelsen, smp. 66-6 8°C.
Eksempel 29
(E)-4- [2-(5,6,7,8-Tetrahydro-l,3-dimetoksy-5,5,8,8-tetra-metyl-2-naftyl)-1-etenyl] benzylacetat 1 g (2,7 mmol) Benzylalkoholderivat fra eksempel 27 ble oppløst i 5 ml pyridin og blandet med 1 ml eddiksyreanhydrid. Man lot røre natten over ved romtemperatur, blandet så med is/vann og filtrerte det resulterende faste stoff, hvilket ble vasket med vann, 0,5 N HC1 og igjen med vann. Etter tørking ble 1 g av tittelforbindelsen tilbake, smp. 73-74°C.
Eksempel 30
(E)-4-C 2-(5,6,7,8-Tetrahydro-l,3-dimetoksy-5,5,8,8-tetra-metyl-2-naftyl)-1-etenyl] benzylamin
Til en suspensjon av 3 g (75 mmol) litiumaluminiumhydrid
i 150 ml abs. eter dryppet man ved romtemperatur 10 g (27 mmol) av nitrilet fra eksempel 5, oppløst henholdsvis oppslemmet i 170 ml eter (ca. 20 minutter). Man oppvarmet 3,5 timer ved tilbakeløp, hydrolyserte neste dag den avkjølte reaksjonsblanding med vann og natriumsulfatløsning og ekstraherte den vandige fase igjen tre ganger med eter. De samlede eterfaser ble vasket
med vann, tørket (Na2S04) og inndampet. Det ble 9,8 g av den rene tittelforbindelse tilbake, smp. 59-60°C.
Eksempel 31
N-Acetyl-(E)-4-[2-(5,6,7,8-tetrahydro-l,3-dimetoksy-5,5,8,8-tetrametyl -2-naftyl)-1-etenyl] benzylamin
Til en løsning av 3 g (8 mmol) av benzylaminderivater fra eksempel 30, 1,6 g (16 mmol) trietylamin og 50 mg DMAP (4-n.N-dimetylaminopyridin) i 25 ml abs. tetrahydrofuran dryppet man 1 ml acetylklorid i 5 ml abs. tetrahydrofuran ved 20°C. Etter 1 time ved 0°C helte man dette i 100 ml vann og ekstraherte med metylenklorid. Den organiske fase ble tørket og inndampet. Den oljeaktige rest ble oppvarmet med heptan; etter avkjøling ble den ovenstående heptanfase helt av og prosessen gjentatt flere ganger. Til slutt krystalliserte 0,8 g av tittelforbindelsen ut fra heptanløsningen, og denne ble filtrert fra,
smp. 126-127°C.
Eksempel 32
(E,E)-4-[ 2-(5,6,7,8)-Tetrahydro-1,3-dimetoksy-5,5,8,8-tetrametyl-2-naftyl)-1-etenyl] kanelsyreetylester
Til en suspensjon av 0,5 g (16,6 mmol) natriumhydrid i 25 ml abs. tetrahydrofuran dryppet man en løsning av 3,3 g (14,8 mmol) dietylfosfonoeddiksyreetylester i 15 ml abs. tetrahydrofuran. Man rørte 1 time til og dryppet så en løsning av 2,8 g (7,4 mmol) av aldehydet som er beskrevet i eksempel 26 i 15 ml absolutt tetrahydrofuran til. Man lot røre enda i 16 timer, helte dette i vann og surgjorde. Den vandige fase ble skilt fra og ekstrahert to ganger med eter. De samlede eterekstrakter ble vasket en gang med vann, tørket (Na2SOij) og inndampet. Resten (4,8 g) ble omkrystallisert fra heptan, og man fikk 0,5 g av tittelforbindelsen, smp. 205-207°C.
Claims (5)
1. Analogifremgangsmåte ved fremstilling av terapeutisk aktive 1-substituerte tetraliner med formel I
hvori
n er tallet 0 eller 1,
R<1> betyr en hydroksy- eller C^g-alkoksygruppe,
R<2> og R<3> hydrogen- eller C1_4-alkyl eller metoksygrupper,
R<5> et hydrogenatom eller en C1_4-alkylgruppe,
R<6> et hydrogenatom, et nitril eller resten -CHR<7>R<8> eller -COR<9>, hvori R<7> betyr et hydrogenatom, R<8> et hydrogenatom eller resten -OR1<0> eller -NR1:LR1<2> (med R<10> i betydningen et hydrogenatom, en C1_4-alkyl eller en C1_4-alkanoylgruppe),
R<9> er et hydrogenatom, azido, eller resten -OR<13> eller -NR<14>R<15 >(med R<13> i betydning et hydrogenatom eller en eventuelt med en hydroksygruppe substituert C^.g-alkylgruppe og med R<14> og R<15> i betydningen hydrogenatom eller en C^.g-alkylgruppe), samt deres fysiologisk fordragelige salter,
karakterisert ved at man omsetter en karbonylforbindelse med formel II
hvor R<1>, R<2> og R<3> har de angitte betydninger, med en fosforforbindelse med formel III
hvori R<21> er hydrogenatom, en C1_4-alkylgruppe, nitrilgruppen eller en gruppe -COOR<22>, og R20 og R22 betyr en C^_3-alkylgruppe ifølge Wittig-Horner,og om ønsket,
overføres erholdte syreazider med formel I i syreamider ved omsetning med tilsvarende aminer;
overføres erholdte nitriler med formel I i amider ved hydrolyse, eller i aldehyder ved DIBAH-reduksjon,
eller i aminer ved LiAlH4-reduksjon, som igjen kan overføres i karboksylsyreamider ved omsetning med tilsvarende karboksylsyre eller reaktivt derivat derav;
overføres erholdte karboksylsyrer med formel I i estere ved forestring med tilsvarende alkohol, eller i alkoholer ved reduksjon, som igjen kan overføres i etere derav ved foretring med den tilsvarende alkohol, eller karboksylestere ved omsetning med tilsvarende karboksylsyre eller reaktivt derivat derav, eller i syreazider ved omsetning med aktiveringsmiddel og deretter metallazid.
2. Fremgangsmåte ifølge krav 1 ved fremstilling av (E)-4-[2- -(5,6,7,8-tetrahydro-3,5,5,8,8-pentametyl-l-propyloksy-2-naftyl)-1-etenyl]benzosyre, karakterisert ved at man anvender tilsvarende substituerte utgangsmaterialer.
3. Fremgangsmåte ifølge krav 1 ved fremstilling av (E)-4-[2- -l-butyloksy-5,6,7,8-tetrahydro-3,5,5,8,8-pentametyl-2-naftyl)- -1-etenyl]benzonsyre, karakterisert ved at man anvender tilsvarende substituerte utgangsmaterialer.
4. Fremgangsmåte ifølge krav 1 ved fremstilling av (E)-4-[2- -(l-etoksy-5,6,7,8-tetrahydro-3,5,5,8,8-pentametyl-2-naftyl)-1-etenyl]benzosyre, karakterisert ved at man anvender tilsvarende substituerte utgangsmaterialer.
5. Fremgangsmåte ifølge krav 1 ved fremstilling av (E)-4-[2- -(5,6,7,8-tetrahydro-l,3-dimetoksy-5,5,8,8-tetrametyl-2-naftyl)-1-etenyl]benzosyre, karakterisert ved at man anvender tilsvarende substituerte utgangsmaterialer.
