I fransk patentskrift nr. 2.437.213 og tilleggspatent nr. 2.466.252 er det beskrevet fremstilling av konjugerte anticancer-produkter oppnådd ved kobling ved kovalent binding av ricin's A-kjede til en proteinstruktur såsom et antistoff, et immunoglobulin eller et fragment av immunoglobulin med evne til selektivt å gjenkjenne et antigen på overflaten av bæreceller som man vil oppnå, f.eks. kreftceller. Hovedegenskapen ved disse konjugerte forbindelse er å være cytotoksiske midler som er spesifikke for celler som det tas sikte på.
Anvendelse av antistoffer rettet mot antigenene for differ-ensiering av kreftceller hadde allerede gjort det mulig å oppnå konjugerte forbindelser som oppvise en bemerkelsesverdig spesifisitet overfor aktuelle celler. Karakterisering av antigener som er assosiert med kreftcellene, og oppnåelse av monoklonale antistoffer rettet mot disse antigener gjør det mulig å frem-stille konjugerte forbindelser som oppviser øket spesifisitet overfor celler som er bærere av disse antigener.
Foreliggende oppfinnelse angår således en fremgangsmåte for fremstilling av produkter som er nyttige som medikamenter, produkter som er kalt konjugerte forbindelser og oppnådd ut fra ricin's A-kjede og et human-antimelanom-monoklonalt antistoff.
Med konjugerte forbindelser forstås blandede kunstige molekyler i hvilke ricin's A-kjede er assosiert ved en kovalent binding av disulfidtype til et human-antimelanom-antistoff med evne til selektivt å gjenkjenne et antigen som er assosiert med kreftcellene.
Oppnåelse av ren ricin's A-kjede er beskrevet i tidligere patenter. Fremstilling av monoklonale antistoffer rettet mot human-melanomer er omtalt i den vitenskapelige litteratur (det henvises spesielt til Proceedings of the National Academy of Sciences 77(4), 2183-7, 1980).
For å realisere de konjugerte forbindelser er det nødvendig at de proteiner som skal kobles, hver bærer minst ett svovelatom som er naturlig egnet eller kunstig gjort egnet til å skape den krevede disulfidbinding.
Ricin's A-kjede oppviser naturlig et eneste svovelatom som tillater den ønskede kobling. Det er det i tiolfunksjonen i cysteinresten som er inkludert i A-kjeden og som sikrer bindingen av denne A-kjede til B-kjeden i det komplette toksin.
Antimelanom-antistoffet har verken fri tiolfunksjon eller andre svovelatomer som kan benyttes for kobling. Det passer altså å innføre kunstig på molekylet til immunoglobulin et eller flere svovelatomer som til slutt vil kunne bli engasjert i disulfidbindingen som skal etableres med et eller flere molekyler i ricin's A-kjede.
Fremgangsmåten i henhold til oppfinnelsen er karakterisert ved at man omsetter ricin's A-kjede, idet kjeden er representert ved formelen RASH, med et derivat av antistoffet P97 av formel AC-S-S-X hvor X betyr et aktivator-radikal og AC betyr antistoffet P97.
Fremstillingen av den konjugerte forbindelse kan represen-teres ved følgende reaksjonsskjerna:

Antistoffet som er substituert med et aktivert svovelatom oppnås ved å gå ut fra selve antistoffet, ved substituering ved hjelp av et reagens som selv er bærer av et aktivert svovelatom, i henhold til følgende skjema:

R betegner en gruppe som samtidig kan bære substituentene Y og - S - S - X
X betegner aktivator-radikalet.
Den funksjonelle gruppe Y er en funksjon som har evne til å binde seg på kovalent måte til hvilken som helst av de funksjoner som bæres av de laterale kjeder i aminosyrene som det protein som skal substitueres består av. Blant disse er de endestående aminfunksjoner i lysyl-radikalene som inneholdes i proteinet, spesielt angitt. I dette tilfelle kan Y særlig representere: en karboksylgruppe som kan binde seg til aminfunksjoner i proteinet i nærvær av et koblingsmiddel som f.eks. et karbodiimid og særlig et vannløselig derivat som etyl-1-(dietylamino-3-propyl)-3-karbodiimid, et karboksylsyreklorid som kan reagere direkte med aminfunksjonene for å acylere dem, en ester betegnet "aktivert" som f.eks. en ester av orto-eller para-, nitro- eller dinitrofenyl eller dinitrofenyl eller en ester av N-hydroksyravsyreimid som reagerer direkte med aminfunksjonene for å acylere dem, et internt anhydrid av en dikarboksylsyre som f.eks. ravsyreanhydrid, som reagerer spontant med aminfunksjonene og skaper amidbindinger alkylgruppe som reagerer med amingruppene i proteinet i henhold til følgende reaksjon:
X betyr en funksjonell gruppe med evne til å reagere med et fritt tiolradikal. Spesielt kan X bety en 2-pyridylgruppe eller 4-pyridylgruppe som eventuelt er substituert med alkyl, halogen eller karboksyl. X kan også bety en fenylgruppe som fortrinnsvis er substituert med en eller flere nitro- eller karboksylgrupper. X kan videre representere en alkoksykarbonylgruppe, f.eks. metoksykarbonylgruppen.
Radikalet R betyr ethvert radikal med evne til samtidig å bære substituentene Y og S - S - X. Det må velges slik at det ikke omfatter funksjoner som under sluttreaksjonene er tilbøyelig til å interferere med de reagenser som benyttes og de produkter som syntetiseres. Spesielt kan gruppen R være en gruppe -(CH2)n hvor n ligger mellom 1 og 10, eller en gruppe:

betyr en rettkjedet eller forgrenet alkylgruppe med 1-5 karbonatomer, spesielt tert.-butylgruppen.
Omsetningen av forbindelsen Y-R-S-S-X med immunoglobulin utføres i homogen væskefase, oftest i vann eller en pufferløsning. Når løseligheten av reagensene krever det, er det mulig å tilsette i reaksjonsmiljøet opp til 20 volum*, av et organisk løsningsmiddel som er blandbart med vann, f.eks. en alkohol, spesielt tert.-butanol.
Reaksjonen utføres ved omgivelsestemperatur i et tidsrom som varierer mellom noen timer og 24 timer. Etter dette tillater en dialyse å fjerne produktene med lav molekylvekt og særlig overskuddet av reagenser. Denne fremgangsmåte gjør det mulig å innføre et antall substituentgrupper pr. mol protein mellom 1 og 5 hvis proteinet er et immunoglobulin av klasse G, mellom 1 og 15 hvis proteinet er et immunoglobulin av klasse M.
Ved anvendelse av slike forbindelser utføres koblingen med ricin's A-kjede i nærvær av vandig løsning av de to proteiner ved en temperatur som ikke overstiger 30°C i et tidsrom som varierer mellom noen timer og 1 dag. Den oppnådde løsning dialyseres for fjerning av produktene med lav molekylvekt, og så kan den konjugerte forbindelse renses ved forskjellige kjente metoder.
Det eksempel som følger gjør det mulig å forstå oppfinnelsen bedre.
Eksempel 1
En konjugert forbindelse oppnådd ved å gå ut fra human-antimelanom-antistoff (antistoff rettet mot antigen P97) substituert med en aktivert disulfidgruppe og ricin's A-kjede.
a) Human- antimelanom- antistoff ( anti P97)
Dette antistoff ble oppnådd ved den metode som er beskrevet
i Proceedings of National Academy of Science 1980, 22 (4), 2183-7 .
Det ble underkastet en siste rensing ved dialyse mot puffer PBS (10 mM fosfat, 140 mM natriumklorid, pH: 7,4).
b) Ricin' s A- kiede
Ricin's A-kjede ble fremstilt og renset slik som angitt i
franske patentskrifter nr. 78 27838 og tillegg nr. 79 24655.
c) Aktivert human- antimelanom- antistoff
Til 0,5 ml av en løsning av 20 mg/ml (pyridyl-2-disulfanyl)-3-propionsyre i tert.-butanol tilsettes 0,2 ml av en løsning av 60 mg/ml etyl-1-(dimetylamino-3-propyl)-3-karbodiimid, og dette får henstå i 3 minutter ved omgivelsestemperatur.
Man tilsetter 30 pl av den således oppnådde løsning til 1,66 ml av en løsning av antimelanom-antistoff, nemlig 2,4 mg/ml i puffer PBS. Man lar inkuberingen foregå i 20 timer ved 30°C.
Man dialyserer så løsningen kontinuerlig i 3 timer mot
21 liter puffer PBS ved 4°C. Man oppnår således 4 mg aktivert antistoff i en konsentrasjon av 2,3 mg/ml.
Ved spektrofotometrisk dosering ved 343 nm av 2-tion-pyridin frigjort ved utbytting med det reduserte glutation konstaterer man at man har oppnådd et antistoff som bærer 2,6 aktivator-grupper pr. mol antistoff.
d) Konjugert forbindelse
Til 1,3 ml av en løsning av aktivert antistoff i puffer PBS
(konsentrasjon 2,3 mg/ml, f.eks. 3 mg aktivert antistoff), tilsettes 0,52 ml av en løsning av ricin's A-kjede i den samme puffer (konsentrasjon 5,8 mg/ml), og man utfører inkubering ved 35°C i 20 timer.
Man kromatograferer reaksjonsblandingen på gelkolonne Sephadex G 100. I hver fraksjon bestemmer man konsentrasjonen av antistoff ved spektrofotometri ved 280 nm og konsentrasonen av A-kjeden ved dens inhiberende kraft på proteinsyntesen målt på et acellulært system. Man kombinerer de identiske fraksjoner som inneholder den konjugerte forbindelse og oppnår således ca. 1 mg konjugert forbindelse i en konsentrasjon av 0,2 mg/ml.
De analytiske bestemmelser som er utført gjør det mulig å vise at løsningen inneholder 50 pg/ml av biologisk aktiv A-kjede, f.eks. ca. 1,4 mol av A-kjede pr. mol antistoff.
Et studium som ble utført ved cytofluorometri har bl.a. gjort det mulig å vise at det anvendte antimelanom-antistoff, det tilsvarende aktiverte antistoff og den konjugerte forbindelse av dette antistoff sammen med ricin's A-kjede viser histogrammer av fluorescens som ligger svært nær, hvilket tillater fastslåelse av at antistoffet ikke har gjennomgått noen endring av betydning under aktiveringsreaksjonene og koblingen som den er utsatt for, og spesielt at det forblir i stand til, i den konjugerte forbindelse, å gjenkjenne antigenet P97 som det er rettet mot.
Den konjugerte forbindelse, oppnådd som beskrevet ovenfor i henhold til oppfinnelsen, er studert hva angår dens biologiske egenskaper og særlig dens anticancervirkning.
1) Inhibering av proteinsvntesen
Den fundamentale biologiske egenskap hos ricin's A-kjede er å inhibere den cellulære proteinsyntese ved endring av under-enheten ribosomal 60 S.
Man har her anvendt en cellemodell. Denne test måler effekten av de undersøkte substanser på inkorporering av 14
C-leucin i de kreftceller som dyrkes.
De anvendte celler tilhører stammen M 144 fra et human-melanom som bærer antigenet P97. Disse celler preinkuberes, etter trypsinerng, minst 24 timer i den hensikt å tillate reekspresjon av antigener fra den eventuelt endrede overflate. Etter tilsetning av den substans som skal undersøkes utfører man en ny inkubasjon. Ved slutten av inkubasjonen foretar man

målinger av inkorporeringsgraden av C-leucin ved de således behandlede celler.
Denne måling utføres i henhold til en teknikk som er tilpasset den teknikk som er beskrevet i Journal of Biological Chemistry 1974, 249 (11), 3557-62 ved anvendelse av traceren

C-leucin for bestemmelse av proteinsyntesegraden. Målingen av den inkorporerte radioaktivitet utføres her på hele cellene som er isolert ved filtrering.
Ut fra disse målinger kan man trekke opp kurver for effekter/doser hvor det på abscissen vises konsentrasjon for de undersøkte substanser og på ordinaten inkorporeringen av 1 C-leucin uttrykt i prosent inkorporering av kontrollceller i fravær av den substans som skal undersøkes.
Man kan således for hver substans som undersøkes, bestemme 14
den konsentrasjon som inhiberer 50% inkorporering av C-leucin eller "inhiberende konsentrasjon 50" (CI 50).
De resultater som er oppnådd med den konjugerte forbindelse som er fremstilt i ovenstående eksempel (ITHM) er vist på
figur 1. På denne figur er det likeledes vist kurver som er oppnådd henholdsvis med ricin (R), ricin's A-kjede (AR) og en konjugert forbindelse ricin's A-kjede - antistoff antiradikal dinitrofenyl (DNP) (DS 10), konjugert forbindelse som ikke viser noen affinitet for de testede celler.
Man kan på denne figur konstatere at den konjugerte forbindelse som er undersøkt (ITHM) viser en sterk cytotoksisk aktivitet (CI 50 = 5x10 M), ca. 400 ganger større enn for ricin's A-k]ede.
Forøvrig har den konjugerte forbindelse anti-DNP (DS 10)
ikke effekt på cellene M 1477 opp til den høyeste konsentrasjon som er undersøkt (10<-6>M). Derimot viser den samme konjugerte -9 forbindelse DS 10 seg cytotoksisk med en CI 50 nær 10 M hvis den bringes i nærvær av de samme celler M 1477 som på forhånd på kunstig måte er gjort bærer av radikalene DNP. Disse to sist-nevnte konklusjoner viser den spesifikke karakter ved den cytotoksiske aktivitet hos den konjugerte forbindelse ITHM.
2) Inhibering av kolonidannelse
Dyrkningscellene av melanom M 1477 fjernes fra sin bærer med en løsning av Versene (puffer PBS som inneholder etylendiamin-tetraeddiksyre) eller ved trypsinering. Disse celler insemineres med 2x10 4 celler pr. Petri-skål med diameter 5 cm som inneholdt følgende dyrkningsmedium: Substrat RPMI 1640 (Mérieux) tilsatt glutamin 2 mM, natrium-bikarbonat 2 g (1, 15% inaktivert kalvefetalserum (Seromed) og antibiotika (penicillin, streptomycin og amfotericin B).
Etter 24 timer behandles disse celler med varierende konsentrasjoner av den konjugerte forbindelse som skal under-søkes .
For sammenligning utføres den samme forsøksrekke med ricin, på den ene side og ricin's A-kjede på den annen side, og endelig med en konjugert forbindelse som ikke er spesifikk for denne celle cellestamme (konjugert forbindelse mellom ricin's A-kjede og et antistoff anti-DNP (DS 10)).
Etter nye 24 timer fjernes kulturmediet og erstattes med det samme friske miljø, fri for enhver cytotoksisk substans. 10 til 15 dager senere bestemmes antall kolonier som har utviklet seg, etter farvning med en løsning av krystallfiolett, ved hjelp av automatisk koloniteller (system Artek 880).
Denne metode gjør det mulig å bestemme en så liten mengde som 10 viable celler pr. skål samtidig som dette har kunnet bli verifisert ved benyttelse av kontrollkulturer. Det er likeledes vist at dannelsen av kolonier er strengt proporsjonal med startkonsentrasjonen av celler, i det minste innen grensene IO1

til 10 celler pr. ml.
De oppnådde resultater er vist på figur 2, hvor det på ordinaten logaritmisk er avsatt antall kolonier pr. skål, og på abscissen konsentrasjonen av produktet. Forsøkene er utført for ricin (R), ricin's A-kjede (AR) og det konjugerte produkt fremstilt i henhold til oppfinnelsen (ITHM).
Det konjugerte produkt ITHM viser høy aktivitet mens den
_9 siste melanom-celle er drept i en konsentrasjon av ca. 2 x 10 M av en konjugert forbindelse. Denne konsentrasjon er helt ut

sammenlignbar med verdien for ricin (1 x 10 M) mens det for ricin's A-kjede må være 10 ^ M for at den samme effekt skal oppnås.
Man kan likeledes se at den konjugerte forbindelse DS 10 ikke har noen aktivitet opp til 2 x 10 — 8 M som er den sterkeste konsentrasjon som den testes ved.
Konklusjonene fra dette forsøk bekrefter fullt ut erfaringen med inhibering av proteinsyntesen.
De konjugerte forbindelser som er fremstilt i henhold til oppfinnelsen viser alstå sterkt spesifikk virkning overfor cellestammer av human-melanom. De kan altså anvendes i human-terapi for behandling av melanomer og eventuelt andre sykelige tilstander, cancerøse eller ikke, som ville være ømfintlige overfor det anvendte antistoff.
Disse konjugerte forbindelser er tilpasset for injiserbar administrering. De kan anvendes alene eller i tilknytning til en annen behandling av kreftsykdommer og spesielt i tilknytning til andre immunodepressive medikamenter for å retardere og svekke den naturlige immunreaksjon hos pasienten overfor det fremmedprotein i hans organisme som den konjugerte forbindelse utgjør.
Med hensyn til å fjerne alle kreftceller, så må behandlingen utføres med tilstrekkelig dose av en konjugerte forbindelse, og behandlingens varighet må bestemmes i hvert tilfelle alt etter hvordan pasienten og hans sykdomstilstand er.
Claims (2)
1. Fremgangsmåte for fremstilling av en forbindelse som er et molekyl i hvilket ricin's A-kjede er knyttet, ved en kovalent binding av disulfidtype, til et human-antimelanom-antistoff P97, karakterisert ved at man omsetter ricin's A-kjede, idet kjeden er representert ved formelen RASH, med et derivat av antistoffet P97 av formel AC-S-S-X hvor X betyr et aktivator-radikal og AC betyr antistoffet P97.
2. Fremgangsmåte som angitt i krav 1, karakterisert ved at radikalet X betyr en gruppe som er egnet til å reagere med et fritt tiolradikal og særlig en 2- eller 4-pyridylgruppe som eventuelt er substituert, en fenylgruppe eller en alkoksykarbonylgruppe.
