Nærværende oppfinnelse vedrører fremstilling av et terapeutisk aktive fenoksypropanolaminderivater og deres fremstillings-fremgangsmåte.
Derivatene fremstilt ifølge oppfinnelsen har den generelle formel (I)
hvori R^ betyr en forgrenet eller uforgrenet C^-<C>^<g->alkylgruppe, en C^_ q cykloalkylgruppe, en forgrenet eller uforgrenet C^_^-alkylgruppe substituert med en fenylgruppe, med en fenylgruppe substituert med en C-^^-ålkoksygruppe; R, betyr en forgrenet eller uforgrenet C-^-C^g-alkylgruppe, en C^.g-cykloalkylgruppe, en forgrenet eller uforgrenet C-^-C^-alkylgruppe substituert med en fenylgruppe, med en fenoksygruppe, med en fenyl- eller fenoksygruppe substituert med en eller flere halogenatomer, såsom fluor, klor eller brom, med en eller flere forgrenede eller uforgrenede C^_3~alk<y>l eller C-^^-alkoksygrupper, med en metylendioksy-gruppe;

1-[4-(2-n-oktylaminoetyl)fenoksy]-3-propylamino-2-propanol 1-[4-(2-n-oktylaminoetyl)fenoksy]-3-n.oktylamino-2-propanol 1-[4-(2-n-oktylaminoetyl)fenoksy]-3-(4-fenylbutyl)amino-2-propanol
Når derivatene ifølge formel I foreligger som addisjonssalter med syrer, kan de omdannes ifølge vanlige fremgangsmåter til deres frie base eller i salter med andre syrer.
De mest vanlig anvendte salter er addisjonssalter med ikke-giftige, farmasøytisk anvendelige syrer, slik som egnete uor-ganiske syrer, f.eks. saltsyre, svovelsyre eller fosforsyre/ eller med egnete organiske syrer, slik som alifatiske, cyklo-alifatiske, aromatiske, aralifatiske eller heterocykliske, karb-oksyl- eller sulfonsyrer, f.eks. maursyre, eddiksyre, pronionsyre, ravsyre, glykolsyre, glukonsyre, melkesyre, maleinsyre, vinsyre, sitronsyre. askorbinsyre, glukoronsyre, malinsyre, fumarsyre, pyrodruesyre, aspartinsyre, glutaminsyre, benzosyre, anthranil-syre, hydroksybenzosyre, salicylsyre, fenyleddiksyre, mandel-syre, embonsyre, metansulfonsyre, etansulfonsyre, pantotensyre, toluensulfonsyre, sulfanilsyre, cykloheksylaminosulfonsyre, stearinsyre, alginsyre, p-hydroksypro<p>ionsyre. ft-hydroksysmør-syre, oksalsyre, malonsyre, galaktarsyre, galakturonsyre. Disse salter kan også stamme fra naturlige eller ikke naturliqe aminosyrer slik som lysin, glycin, arginin, ornitin, asparagin, glutamin, alanin, valin, treonin, serin, leucin, cystein og lignende.
Egnete estere av fenoksypropanalaminer ifølge oppfinnelsen er f.eks. estere avledet fra alifatiske karboksylsyrer med opp til 20 karbonatomer, slik som eddiksyre, palmitin-, stearin-
eller oleinsyre, og estere avledet fra aromatiske karboksylsyrer med opp til 10 karbonatomer, slik som benzosyre, såvel som deres addisjonssalter med syrer.
Forb]LtL&elsene .med formel I omfatter minst ett asymmetrisk kar-bonatom og eksisterer således som optiske isomerer eller race-mater.
De optiske isomerer kan erholdes ved gjenoppløsning av racema-ter ifølge tradisjonelle fremgangsmåter, f.eks. ved dannelse av diastereoisomere salter ved innvirkning av optisk aktive sy-
rer, slik som vinsyre, diacetylvinsyre, tartranilsyré, diben-zoylvinsyre, ditoluoylvinsyre og separering av den diastereoisomere blanding f.eks. ved krystallisasjon eller kromatogra-
fi, deretter frigivelse av de optisk aktive baser fra disse salter. De optisk aktive forbindelser med formel I kan også erholdes ved anvendelse av optisk aktive utgangsprodukter.
De foretrukne optiske isomerer av forbindelsene med formel I
er de med (S) konfigurasjonen ved karbonatomets nivå, hvilket bærer alkoholfunksjonen i oksypro<p>anolaminkjeden.
Generelt oppviser derivatene ifølge oppfinnelsen noen aktivitet på det kardiovaskulære system, f.eks. aktiviteter som hemmer blodplateagglutinering, antihypertensiv, ft-blokkerende, lokalt anestetisk, antispasmodisk, perifer, vasodilatorisk, hypolipidemisk antithrombosisk, antiarrhytmisk aktivitet og/ eller aktiviteter på sentralnervesystemet, f.eks. en beroli-gende aktivitet.
Disse egenskaper tillater også at man kan tenke på bruk av
de nye forbindelser i profylaksen og behandlingen av hypertensjon og kardiovaskulære virkninger, slik som athero-sclerose og angina pectoris, myokardial infarkt og rytme-forstyrrelser.
Mere spesielt er det blitt funnet at noen av de nye forbindelser var utstyrt med en meget spesiell aktivitet over-
for blodplateagglutinering. Faktisk hemmer noen produkter in vitro blodplateagglutineringsfenomenet foranlediget av kollagen og "Thrombofix". En viktig forskjell ble imidlertid observert vedrørende agglutineringshemningen, når ADP ble anvendt som agglutineringsmiddel. Mens produkter, slik som aspirin og suloktidil bare hemmer den andre fase av blodplateagglutine-ringsf enomenet (frigivelsesreaksjon) er noen produkter ifølge oppfinnelsen i stand til å inhibere begge faserav agglutinering. En lignende virkning iakttas bare med prostacyklinora-denosin.
Igjen, slik som prostacyklin er noen forbindelser med formel
I i stand til å desaggregere blodplatene som tidligere
var agglutinert med ADP, hvilket skiller dem fra andre anti-agglutineringsmidler, slik som aspirin, indomethacin, dipyri-damol og suloktidil, som har vist seg som fullstendig inaktive i så henseende.
Dessuten er det blitt funnet at noen forbindelser har en viktig virkning ved selektivt å blokke P^-adrenergiske reseptorer. Forbindelsene med denne selektive virkning har en høyere spesifisitet overfor kardiale fl-adrenergiske reseptorer (3-^) enn overfor (3-adrenergiske respetorer i perifere ' blodkar og bronkialmuskler (ft-?).
Denne kardioselektivitet tillater påtenkning av anvendelse av de nye forbindelser for pasienter som lider av astma og kroniske pulmonale forstyrrelser, såvel som de som lider av perifere arterie-insufficienser. Dessuten oppviser produktene ifølge oppfinnelsen hverken noen p-mimetisk aktivitet (delvise antagonister), eller membranvirkninger, slik som det kunne sluttes fra fraværet av modifikasjoner i hyppigheten av hjertekammerkontraksjoner i enhver retning.
Det er også blitt iakttatt at de nye forbindelser
samtidig var utstyrt med en viktig blokkerende aktivitet for 3^ adrenergiske reseptorer og med en viktig perifer vasodilatorisk aktivitet.
Det er for tiden kjent at perifere vasodilatoriske midler har en økende interesse ved behandlingen av hypertensjon. Det har imidlertid syntes som om nevnte midler har uønskede bivirk-ninger, slik som for eksempel tachycardi, som det er nødvendig å redusere ved samtidig administrasjon av et p-blokkerende middel. Et molekyl med samtidig 3-^ blokkerende egenskaper og perifere vasodilatoriske egenskaper er således av virkelig terapeutisk interesse.
Forbindelsene fremstilles ved at man omsetter en forbindelse med formelen

med en forbindelse R3Z', hvori R3 har den ovenfor gitte betydning og Z' har en av betydningene for Z, forutsatt at ved omsetningen, er Z eller Z' en NF^-gruppe, og når C = X<1 >betyr C = 0, er Z en NH2~gruppe, og det erholdte amid reduseres til det tilsvarende amin med den generelle formel I.
EKSEMPEL 1
1-[4-(2-n.oktylaminoetyl)fenoksy]-3-isopropylamino-2-propanol.
a) Til 18 g p-benzyloksyfenyletylamin, 15 ml trietylamin og 250 ml kloroform ble 15 g oktanoinsyreklorid langsomt tilsatt
under røring.
Etter at tilsetningen er fullstendig ble blandingen rørt ytter-ligere 3 til 4 timer, deretter ble oppløsningsmidlet inndampet i vakuum, resten ble fortynnet med vann og ekstrahert med kloroform. Den organiske fase ble tørket, filtrert og inndampet. Således ble 25 g N-oktanoyl-p-benzyloksyfenyletylamin erholdt. Utbytte: 90 % smp. (°C): 123-124.
b) 25 g av det foregående produkt, 500 ml etanol, 2,5 g 10% Pd/C og 2 ml konsentrert HC1 ble hydrogenert ved 2 5°C i 2
timer. Etter filtrering av katalysatoren ble oppløsningsmidlet inndampet i vakuum og resten størknet ved tilsetning av petro-leumeter. Således ble 17 g N-oktanoyl-p<->hydroksyfenyletylamin erholdt.
Utby-te: 90% smp. (°C): 60-63.
c) En oppløsning som inneholder 19 g av det foregående produkt, 95 ml etanol og 2,38 g NaOH ble langsomt tilsatt til en
oppløsning som inneholder 20 g epiklorhydrin, 80 ml dioksan og 20 ml vann og oppvarmet til 75°C.
Etter tilsetning (ca. 1 time) ble denne temperatur opprettholdt i 5 minutter, derpå ble oppløsningen kjølt, filtrert og inndampet til tørrhet. Resten ble ekstrahert med kloroform og den organiske fase ble vasket med 10%'ig NaOH og vann, og derpå inndampet. Den-faste rest .ble tørket og 20 g N-oktanoyl-4-(2,3-epoksypropoksy)fenyletylamin ble således oppnådd.
Utbytte: 85% sm<p>. (°C): 113-114.
d) 10 g av det foregående produkt, 50 ml etanol og 15 g isopropylamin ble tilbakeløpsbehandlet i 5 timer. Etter kjøling
ble oppløsningsmidlet og overskytende amin inndampet i vakuum,
og resten størknet ved tilsetning av pentan. Omkrystallisasjon ble utført fra en blanding av aceton og eter og således ble 9
g 1-[4-(2-N.oktanoylaminoetyl)fenoksy]-3-isopropylamino-2-propanol erholdt.'
Utbytte: 76% smp. (°C): 108-110.
e) 1,5 g av det foregående produkt oppløst i 10 ml THF ble langsomt tilsatt til en diboranoppløsning i THF, og blandingen
ble bragt til 40-50°C i 3 timer. Etter kjøling ble 10 ml vann langsomt tilsatt, og oppløsningen inndampes til tørrhet og i vakuum. Resten ble tatt opp med eter og en strøm av vannfritt HC1 ble ført gjennom oppløsningen. Bunnfallet ble filtrert og tørket og 1,1 g 1-[4-(2-n-oktylaminoe tyl)fenoksy]-3-isopropyl-amino-2-propanol(dihydroklorid) ble erholdt.
Utbytte: 60% smp. (°C): 236-240 (spaltning).
EKSEMPEL 2
1-[4-(2-n-oktylaminoetyl)fenoksy]-3-n.oktylamino-2-propanol
a) 20 g N-oktanoyl-4-(2,3-epoksypropoksy)fenyletylamin, 250 ml etanol og 20,4 g n-oktylamin ble tilbakeløpsbehandlet i 4
timer. Etter kjøling ble oppløsningsmiddel og overskytende amin inndampet i vakuum. Den eneste rest størknet ved tilsetning av n-pentan og omkrysta!lisert fra aceton. Således ble 16 r5 g 1-[4-(2-n.oktanoylaminoetyl)fenoksy]-3-n-oktylamino-2-propanol erholdt.
Utbytte: 58% smp. (°C): 111-112.
b) 10,4 g av det foregående produkt oppløst i 120 ml THF
ble langsomt tilsatt til en oppløsning av 200 ml diboran i
THF (IM). Temperaturen ble holdt ved 10-15°C under tilsetningen, og deretter hevet til 40-50°C i 4 timer. Etter kjøling ble 300 ml vann langsomt tilsatt, og oppløsningen ble konsentrert i vakuum. Resten ble fortynnet med kloroform, vasket med vann og tørket på MgSO^. Etter filtrering og inndampning av oppløsningsmidlet i vakuum ble resten fortynnet med en blanding av metanol og kloroform og surqjort med en oppløsning av eter mettet med HC1. Det slik erholdte bunnfall ble filtrert og omkrystallisert fra metanol. Således ble 8,3 g 1-[4-(2-n.ok-tylaminoetyl)fenoksy]-3-n-oktylamino-2-pronanol(dihydroklorid) erholdt.
Utbytte: 72% smp. (°C): 325 (spaltning)
EKSEMPEL 3 1-/4-[2-(2-fenyletyl)aminoetyl]fenoksY7-3-isopro<p>ylamino-2-propanol a) Til 13 5 g p-benzyloksyfenyletylamin, 11,5 ml trietylamin og 250 ml kloroform, ble 11,5 g fenylacetylklorid langsomt tilsatt under røring. Etter tilsetning ble blandingen også rørt i 4 timer, og deretter ble oppløsningsmidlet inndampet i vakuum og resten ble fortynnet med vann og ekstrahert med kloroform. Den organiske fase ble tørket, filtrert og inndampet. Således ble 17 g N-fenacetyl-p-benzyloksyfenyletylamin erholdt. Utbytte: 85% smp. (°C): 114-115. b) 19 g av det foregående produkt, 500 ml etanol 2 g 10%<1>ig Pd/C og 1 ml konsentrert HC1 ble hydrogenert (3,5 kg/cm 2) ved
250°C i 2 timer. Etter filtrering av katalysatoren ble oppløs-ningsmidlet inndampet i vakuum og resten ble gjort fast ved tilsetning av eter.
Således ble 12 g N-fenylacetyl-p-hydroksyfenyletylamin erholdt. Utbytte: 80 % smp (°C): 105-110.
c) En oppløsning som inneholder 12 g av det foregående produkt, 62 ml etanol og 1,88 g NaOH ble langsomt tilsatt til en
oppløsning som inneholder 13 g e<p>iklorhydrin, 53 ml dioksan og 13 ml vann og bragt til 75°C. Etter tilsetningen ble denne
temperatur opprettholdt i 10 minutter, derpå ble oppløsningen kjølt, filtrert og inndampet til tørrhet i vakuum. Resten ble ekstrahert med kloroform og den organiske fase ble vasket med 10% NaOH, deretter vann og inndampet. Således ble 10 g N-fen-acetyl-4-(2,3-epoksypropoksy)fenyletylamin erholdt, hvis homo-genitet ble sjekket med TLC og strukturen med NMR.
d) 10 g av det foregående produkt, 50 ml etanol og 75 g isopropylamin ble tilbakeløpsbehandlet i 5 timer. Etter kjøling
ble oppløsningsmiddel og overskytende amin inndampet i vakuum og resten ble gjort fast ved tilsetning av pentan. Det faste stoff ble vasket med eter, filtrert og tørket i vakuum. Således ble 12 g 1-/4-[2-(fenacetyl)-aminoetyl]fenoksyy-3-isopropylarai-no-2-propanol erholdt.
Utbytte: 90% smp. (°C) 100.
e) 8 g av det foregående produkt oppløst i 60 ml vannfritt THF ble langsomt tilsatt til en opnlø-sning av diboran i THF (IM). Etter tilsetning ble blandingen bragt til 40-50°C i 3 timer. Etter kjøling ble 200 m-l vann langsomt tilsatt, oppløs-ningen ble ekstrahert med" kloroform, tørkét på MgSO^, filtrert og inndampet. Resten ble tatt opp med. en blanding av metanol og eter gjennom hvilken en strøm av tørr gassholdig HC1 ble ført^ Det erholdte bunnfall ble filtrert og omkrystallisert fra en blanding av metanol og aceton. Således ble 6 g 1-/4"-[2 —(2-fenyletyl)aminoetyl]fenoksy7-3-isopropylamino-2-propanol (dihydroklorid) erholdt.
Utbytte: 60% smp. (°C) 225-227
EKSEMPEL 4 1- [4- (2-n-oktylaminoetyl)fenoksy]-3-(4-fenylbutylamino)-2-propanol a) 20 g N-oktanoyl-4-(2,3-epoksypropoksy)fenyletylamin, 250 ml etanol og 22,4 g 4-fenylbutylamin ble tilbakeløpsbehandlet i 4 timer. Etter kjøling ble oppløsningsmidlet og det overskytende amin inndampet i vakuum. Den oljeaktige rest erholdt på denne måte ble gjort fast ved tilsetning av n-pentan, og der<p>å ført gjennom en silikagelkolonne (eluering med en metanol-kloro-formblanding 1/5). Etter inndampning av elueringsoppløsnings-midlet, ble det faste stoff omkrystallisert fra aceton. Således ble 18,4 g 1-[4-(2-n.oktanoylaminoetyl)fenoksy]-4-(4-fenylbu-tylamino)-2-propanol erholdt.
Utbytte: 62% smp. (°C): 107-108.
b) 10 g av det foregående produkt oppløst i 100 ml vannfritt THF ble langsomt tilsatt til en oppløsning av 150 ml diboran i
THF (IM). Etter tilseting ble blandingen oppvarmet til 40-50°C
i 4 timer og behandlet ved å følge den foran beskrevne fremgangsmåte. Til slutt etter omkrystallisasjon fra metanol ble 5
g 1-[4-(2-n-oktylaminoetyl)fenoksy]-3-{4-fenylbutylamino)-2-propanol erholdt.
Utbytte: 47% smp. (°C): 308-310 (med spaltning)
EKSEMPEL 5 1-/4-[ 2-(4-fenylbutylamino)etyl]fenoksv7-3-n.butylamino-2-pfopanol a) 28,1 g N-(4-fenylbutyl)—4'-hydroksyfenylacetamid ble opp-løst i 200 ml etanol som inneholder 8 g natriumhydroksyd. Til-bakeløp finner sted i 1 time. b) 30 g epiklorhydrin, 100 ml dioksan og 25 ml vann ble blandet. Oppløsningen ble oppvarmet til 75°C, deretter ble den tidligere oppløsning tilsatt dertil. Oppløsningen ble tilbakeløps-behandlet i 1 time, deretter ble op<p>løsningen kjølt, filtrert
og inndampet til tørrhet i vakuum.
c) Den slik erholdte rest ble oppløst i 200 ml etanol, behandlet med 7,5 ml n-butylamin og tilbakeløpsbehandlet i 8 timer. Oppløsningsmidlet og overskytende amin ble inndampet og resten ble gjort fast ved tilsetning av pentan. Det faste stoff ble vasket med eter, filtrert og tørket i vakuum. Således ble N-(4-fenylbutyl)-4'-(3-n.butyl-amino-2-hydroksypro<p>oksy)-fenyl-acetamid erholdt.
d) 4 g av det foregående produkt oppløst i 50 ml vannfritt THF ble langsomt tilsatt til en oppløsning av diboran i THF (IM). Etter tilsetning ble blandingen, bragt til 40-50°C i 3 timer. Etter kjøling ble 200 ml vann langsomt tilsatt, oppløsningen ble ekstrahert med kloroform,, tørket på MgS04, filtrert og inndampet. Resten ble tatt opp i en blanding av metanol og eter hvorigjennom en strøm av tørr gassholdig HC1 ble ført. Det slik erholdte bunnfall ble filtrert og omkrystallisert fra en blanding av metanol og aceton. Således ble 1-/3-[2-(4-fenylbutyl-amino) etyl]fenoksy7 -3-n.butylamino-2-propanol erholdt. Utbytte: 62 %

produktene ifølge oppfinnelsen ble underkastet en rekke bio-kjemiske og farmakologiske prøver.
LD5Q ble bestemt ifølge Litchfield og Wilcoxon metoden (J. Pharmacol. Exp. Ther. 96, 99, 1949) og angitt i mg/kg for oral administrasjon til mus. Generelt viste produktene ifølge oppfinnelsen seg å være lite toksiske.
Virkningen på oppførslen ble studert under anvendelse av en metode avledet fra S. Irwin (Gordon Res. Conf. on Medicinal Chem. 133, 1959). Stoffene i suspensjon i en 1% tragantgummi ble administrert oralt ved hjelp av en intragastrisk sonde til grupper på 5 hannmus (Charles River DC1 stamme, fastet i 18 timer). Hvis den tilgjengelige mengde stoff tillater det er dosene 3000, lCTOO og 300 mg/kg. I det tilfelle at den sistnevnte dose er aktiv, undersøkes virkningen av drogen ved 100, 3&, 10 og mulgivis 3 mg/kg. Oppførslen studeres 2, 4, 6 og 24 timer etter behandling. Observasjonen forlenges når symptomene ved-rarer på dette tidspunkt. Dødelighetene opptegnes i 14 dager etter behandlingen. Ingen av de prøvete produkter har bevir-ket en unormal oppførsel hos mus ved doser lavere enn 300 mg/kg.
Den antihypertensive aktivitet ble undersøkt ved oral administrasjon til ikke-anestitiserte spontant hypertensive■rotter, hvis systoliske arterietrykk måles ved nivået for den midlere coccygeale arterie ifølge den pletysmografiske metode (J-. Robba, G. Lambelin, A.F..Schaepdryver, Arch, int. Pharmacodyn, 2Q0, 182, 1972). Arterietrykket måles hvert 30. minuttr fra 2 timer før til 3 timer etter oral administrasjon av produkter undersøkt ved 60 mg/kg eller av en placebo (1% tragantgummi). Bare rotter med et systolisk trykk på 180 til 220 mg Hg anvendes. To rotter anvendes pr. produkt. Behandlingene administreres uten at den person som foretar målin<g>ene kjenner til dette. De antihypertensive virkninger opptegnes ifølge en poengberegning som følger:
Under prøvebetingelsene har a-metyldopa en poengsum på +-V+ ved 100 mg/kg, reserpln en poengsum på +++ ved 3 mg/kg og gua-nidin en poengsum på +++ ved 60 mg/kg. Forbindelser 2, 3, 5,
8 og 10 ble undersøkt og viste seg å være aktiv.
Den antispasmodiske aktivitet for de undersøkte substanser ble undersøkt i sammenligning med marsvinileumkontraksjoner forårsaket av histamin og acetylcholin.
Disse erfaringer viser klart en muskulotrop aktivitet. Respon-sen på det kontraherende middel (sub-maksimal kontraksjon) erholdes hvert 5 minutt før og etter injeksjon av økende konsen--7 -4
trasjoner av prøvete produkter (10 M å 10 M). Inhiberings-prosentandelen under innflytelse av prøvete produkter beregnes og den teoretiske konsentrasjon som gir 50%'s inhibering bestemmes grafisk for hvert forsøk. -Disse verdier uttrykkes i
-log CI50 (M) .
Forbindelser 1, 2, 5, 6, 7, 8, 10, 11, 12, 14, 21, 22, 23, 26, 27, 33 og 34 ble undersøkt og viste seg å være aktive.
Den perifere vasodilatoriske aktivitet ble målt på anestetiser-te hunner ved nivået for femoral arteriesirkulasjon. For dette formål overhelles den femorale arterie, hvis kollateraler er blitt underbunnet, ved konstant hastighet med blod tatt fra aorta. Overhellingstrykket målt ved nivået for den femorale arterie varierer således som en funksjon av motstanden i det overhelte område. De prøvete produkt og de tilsvarende op<p>løs-ningsmidlér injiseres direkte ved en dose på 30 jag/ kg. Idet den kardiale ytelse holdes kontant måles således en vasodilasjon ved en reduksjon av øerfusjonstrykket. Sistnevnte poengbereg-nes i sammenligning med virkningen av papaverin, regnet som en referanse og injisert én gang pr. gruppe på 4 produkter. Når interessant prøves produktene med andre doser under de samme betingelser.
Den vasodilatoriske aktivitet angies som følger:
Forbindelse 1, 2, 3, 9, 11, 12, 14, 15, 17, 20, 21, 22, 23, 24, 25, 26, 28, 31, 33 og 34 ble undersøkt og viste seg å være aktiv.
Generelt har forbindelsene ifølge op<p>finnelsen en 3-lytisk aktivitet. Forbindelse 5 ble omhyggelig studert i. så måte.
Virkningen på (3-^ kardiale reseptorer måles ved inhibering av den kronotrope response overfor noradrenalin på marsvinhjer-teforkamre og av den inotrope response overfor iso<p>roterenol på marsvins venstre hjerteforkamre, som ble elektrisk stimu-lert. Kardioselektiviteten bestemmes ved å måle den antagoni-stiske virkning ved nivået for ø2 rese<p>torer på marsvinluft-rør. For dette formål kontraheres en luftrørskjede av en maksimal histaminkonsentrasjon og inhibering av avspenning ved hjelp av isoprotenol måles. Verdiene beregnet for kardioselektivitet erholdes ved å betrakte forskjellen mellom pA2_verdier for den kronotope respons og avspenningen på den ene side (1. kolonne) og for den inotrope response og avspenning på den andre side (2. kolonne). For disse forskjellige forsøk beregnes pA2 ifølge metoden til Van Rossum (Arch.int.Pharmacodyn., 143, 299-330, 1963). Resultatene er gitt i tabell II for forbindelse 5 i tabell I.
Forskjellen for pA2 observert på hjerteforkamre og luftrør indikerer en kardioselektivitet for forbindelsen ifølge oppfinnelsen som er lik den for referanseproduktene: metoprolol og atenolol.
Dessuten har produktene ifølge oppfinnelsen vist seg in vivo og ex vivo å være sterke inhibitorer av blodplateag lutinering,og konsentrasjonene som forårsaker 50%'s inhibiering (CI5o) er lavere enn eller sammenlignbare med de erholdt med aspirin og -indomethacin. Denne inhiberingsvirkning iakttas når agglutinering er forårsaket av en rekke midler, slik som "Thrombofax collagen", ADP og thrombin.
Blodplateagglutineringen måles ved å følge den turbidimetriske metode, slik som beskrevet av G.V.R. Born et M.J. Cross (J. Physiol., 168, 178-195, 1973). Det sitrerte plasma rik på men-neskeblodplater forinkuberes i 3 minutter før tilsetning av det induserende middel. Variasjonene i optisk transmisjon opptegnes i en periode på 10 minutter i et "Chronolog" agglutino-meter. Prøven foretas in duplo i nærvær av produktet og det tilsvarende oppløsningsmiddel. Resultatene noteres sem inhiberings-prosentandel av den maksimale agglutineringsamplitude i for-hold til kontroller. Produktkonsentrasjonen [gitt i mikromol (jiM eller 10 som forårsaker en inhibering av blodplateagglutinering på 50% under forsøksbetingelsene (CI^Q) beregnes fra resultater oppnådd med 6 forskjellige konsentrasjoner.
Tabell III angir resultatene erholdt for forskjellige produkter (konsentrasjon 5,10 ~*M og C^q) på. inhiberingen av blodplateagglutinering indusert av"Thrombofax". De fleste av de prøvete produkter (spesielt forbindelsene 1, 2, 4, 6 og 7) har en høyere inhiberingsaktivitet énn aspirin og indomethacin. Ett produkt, nemlig forbindelse 1, ble mere omhyggelig undersøkt. Tabell IV gir for dette produkt ci5q verdier for agglutinering indusert av ADP, kollagen og thrombin ved sammenligning med verdier erholdt for aspirin (ASA). I de følgende tabeller til-svarer tallene i den første kolonne tallene for forbindelsene i tabell I . Fra disse resultater kan det sluttes at forbindelse 1 inhiberer blodplateagglutinering på en viktig måte når "Thrombofax", collagen og thrombin anvendes som induserende midler. Disse iakttatte virkninger er større enn de erholdt med aspirin. Dessuten forårsaker spesielt forbindelse 1 og 7 ved 100 ^iM en inhibering av de to serier av agglutineringen forårsaket av ADP, idet dette fenomen inntil nå bare er blitt iakttatt for prostacyklin (PGI2) og adenosin (Moncada et al., J. Physiol., 2P, 273, 1977).Faktisk inhiberer de kjente agglutine-ringsmidler bare den andre serie av agglutineringen indusert av ADP (frigjøringsreaksjon). På den andre side viser forbindelse 1, 2 og 4 det ekseptionelle spesielle trekk å forårsake blokkering og reversering av agglutineringsfenomenet indusert av ADP. Således forårsaker ved en dose på 2,10 disse produkter en reversibilitet på 70%, 70% og 60% henholdsvis.
Denne virkning er blitt beskrevet i tidligere dokumenter bare for prostacyklin (PGI2) (Moncada et al., Prostaglandins, 12, 715, 1976) .
Aktiviteten for disse produkter blé vurdert ved produksjonen
av blodplatemalondialdehyd (MDA), som er en stabil metabolitt av arachidonsyre, for å påvise en mulig virkning på blodplate-cyklooksygenase. Metoden består i å inkubere i 3 minutter ved 37°C human<p>lasma rikt på blodplater med produktet som skal un-dersøkes. Deretter stimuleres produksjonen blodplater av MDA ved tilsetning av N-etylmaleimid ImM. MDA måles ved kolorimetri etter reaksjon med tiobarbitursyre ifølge den automatiserte metode beskrevet av Roncucci et al. (Thromb. Res., 1£, 3, 1979). Resultatene ved 5,10~<5> M er angitt i tabell V. Ved konsentrasjoner hvor blodplateagglutinering inhiberes fullstendig (tabell III), påvirkes produksjonen av blodplater MDA ikke vesentlig av de forskjellige prøvete <p>rodukter. Disse resultater indikerer at inhiberingen av blodplateagglutinering ikke er en følge av en Inhibering av cyklooksygenase. Dette faktum bekreftes ved nivået for vaskulær cyklooksygenase. Faktisk inhiberer hverken forbindelse 1 el. forbindelse 7 produksjonen av vaskulært prostacyklin i rotter. Forsøket er blitt utført ved å administrere produkter intravenøst med 5 og 10 mg/kg. Rotter ble avlivet 10 minutter etter intravenøs administrasjon, abdominal aorta og indre cava vena separeres og produksjon av prostacyklin vurderes ved anvendelse av blodplateagglutinering som bio-forsøk. (Bunting S. et al., Pro-
staglandins, 12 (6), 713, 1976).
Antiagglutineringsaktiviteten for disse produkter ble bekref-tet in vivo på aper etter oral administrasjon. De undersøkte produkter ble administrert med en gastrisk sonde som en 1%'ig tragantgummi. Blodet ble blandet med en 3,8 %'ig trinatrium-citratoppløsning før administrasjon og 2, 4, 6 og 24 timer etter administrasjon. Den enkelte orale anvendte dose var fra 1,25 til 5 mg/kg for forbindelse 1 og var 2 mg/kg for forbindelse 5 og 7. Forsøket ble utført på to rhesus-aper for hvert pro-dukt med to tilsvarende aper for placebo. Blodplateagglutinering ble indusert med collagen (individualiserte doser på 1 jiq til 4pg pr. ml) og kvantiteten bestemt ved måling av den maksimale amplitude når det gjelder verdiene før behandling. Tabell VI viser at forbindelse 1 allerede forårsaker, ved en meget lav dose, slik som 1,25 mg/kg, en merkbar inhibering 4 eller 6 timer etter administrering. Ved høyere doser (2, 2,5 og 5 mg/kg) vil virkningen stadig vedvare 24 timer etter behandling. Det er ingen virkning etter 4 8 timer. Forsøkene ut-ført med forbindelse 5 og 7 administrert med en enkelt oral dose på 2 mg/kg viser at det for begge nevnte produkter også eksisterer en inhibering av blodplateagglutinering målt 4 timer etter administrering.
Det skal bemerkes at aspirin ikke har noen virkning ved en dose lavere enn 5 mg/kg oralt.
For administrering av nye forbindelser ifølge oppfinnelsen vil den daglige dose være fra 10 mg til 2 g oralt og fra 0,1 mg til 100 mg parenteralt.
Produktene ifølge oppfinnelsen kan anvendes som forskjellige galeniske former. Følgende eksempler er ikke begrensende og vedrører galeniske formuleringer som inneholder et aktivt produkt.
