Analogifremgangsmåte for fremstilling av antivirale amin-derivater.
Description
Virusinfeksjoner som angriper pattedyr, inkludert
mennesker, er normalt smittsomme sykdommer som kan forårsake store menneskelige lidelser og økonomiske tap. Uheldigvis er opp-
dagelsen av antivirale forbindelser langt mer komplisert og vanskelig enn oppdagelsen av antibakterielle og antifungale midler. Dette er delvis forårsaket av den nære strukturelle likhet mellom vira og strukturen til visse essensielle cellulære bestanddeler som f.eks. ribonuklein- og deoksyribonuklein-syrene.
Ikke desto mindre er det i litteraturen beskrevet mange ikke-virale "antivirale midler", dvs. substanser "som kan frembringe enten en beskyttende eller terapeutisk effekt til en klar påvisbar fordel for den virusinfiserte vert, eller ethvert materiale som signifikant kan øke antistoff-dannelse, forbedre ikke-spesifikk motstand, fremskynde rekonvalesensen eller undertrykke symptomene"
(Herrman et al., Proe. Soc. Exptl. Biol. Med., 103, 625 (1960)). Listen over rapporterte antivirale midler omfatter for å nevne
noen få, interferon og syntetiske materialer som f.eks. amantadin-hydroklorid, pyrimidiner, biguanider, guanidin, pteridin og metisazon. På grunn av det temmelig smale område av virusinfeksjoner som kan behandles med hver av de for tiden kommersielt tilgjengelige antivirale midler, er nye, syntetiske antivirale midler alltid velkomne som potensielt verdifulle tillegg til utrustningen i medisinsk teknologi.
Pattedyrceller produserer en substans som svar på virus-infeksjonen, som gjør cellene i stand til å motstå formeringen av forskjellige vira. De virus-motvirkende eller virus-inngripende substansene refereres til som "interferoner". Interferonene er qlykoproteiner som kan være forskjellige når det gjelder fysikalsk-kjemiske egenskaper, men som alle oppviser samme biologiske egenskaper, nemlig at de inhiberer et bredt område av ikke-beslektede vira, har ingen toksisk eller annen skadelig virkning på cellene og er art-spesifikke (Lockart, Frontiers of Biology, vol. 2, "Interferons", utgitt av av Finter, W.B. Saunders Co. , Philadelphia, 1966, sidene 19-20).
Ingen gjennomførbar, økonomisk metode er ennå utviklet for fremstilling av eksogent interferon for rutinemessig klinisk bruk mot virusinfeksjoner. En alternativ måte å produsere interferon på er derfor forsøkt, som omfatter administrering av en ikke-viral substans til det dyr som skal beskyttes eller behandles, hvilken stimulerer eller induserer produksjon av interferon i cellene.
Det interferon som fremstilles på denne måte refereres til som "endogent" interferon.
US-patent 2.738.351 viser at forbindelser med den generelle formel:
hvor R, og R ? hver kan være alkyl, aralkyl, aryl, cykloalkyl, nitrosubstituert aryl, halogen-substituert aryl, alkyl-substituert aryl eller alkoksy-substituert aryl. X, Y og Z kan hver være oksygen, svovel eller sulfonyl, ALK er lineær eller forgrenet alkylen med fra 1 til 6 karbonatomer, og B kan være di(lavere)-alkylamino, piperidino, morfolino, pyrrolidino, (lavere-alkyl)-pyrrolidino, ISJ'-alky 1-piperazino eller pipekolino, er lokal-anestetiske midler. I tillegg beskriver diskusjonen av alternative syntetiske veier (se kol. 1, 11. 57-70 i nevnte patent) mellom-produkter med ovenstående formel hvor B er amino og (lavere-alkyl)-amino. Ingen av forbindelsene som er spesielt oppregnet i nevnte patent inneholder imidlertid alkyl-R^ eller -P<2 større enn n-pentyl. Ikke i noen av disse forbindelser er ennvidere både og R ? alkyl, og uåde X og Y oksygen.

Den oppfinnelse som beskrives her, omfatter fremstilling av de nye antivirale forbindelsene med formlene I og II. Farmasøytiske preparater kan inneholde en antiviral effektiv mengde av en forbindelse med formlene I og II som essensiell aktiv ingrediens i en farmasøytisk godtagbar bærer og anvendes for profylaktisk (forebyggende) bekjempelse av en virusinfeksjon hos et pattedyr ved administrering av en mengde av en forbindelse med formel I eller II som effektivt profylaktisk be-kjemper nevnte virusinfeksjon, og kan anvendes for indusering av produksjon av interferon i et pattedyr ved å administrere.,
en effektiv mengde av en forbindelse med formel I og II for
å indusere produksjon av interferon.
Forbindelsene fremstilt ifølge oppfinnelsen oppviser antiviral aktivitet mot en lang rekke vira in vivo hos pattedyr og in vitro i vevkulturer fra pattedyr. Minst en vesentlig del av denne aktivitet kommer av disse forbindelsers evne til å indusere produksjonen av interferon i cellene, dvs. endogent interferon.
Med "farmasøytisk godtagbare" syreaddisjonssalter menes de salter som er ikke-toksiske i de administrerte doser. Farmasøytisk godtagbare syreaddisjonssalter som kan anvendes, omfatter slike vann-løselige og vann-uløselige salter som f.eks. hydrokloridet, hyd-robromidet, fosfatet, nitratet, sulfatet, acetatet, heksafluorfosfatet, citratet, glukonatet, benzoatet,
propionatet, butyratet, sulfosalicylatet, maleatet, lauratet, malatet, fumaratet, succinatet, oksalatet, tartratet, amsonatet (4,4<1>-diamino-stilben-2,2'-disulfonatet), pamoatet (1,1<1->metylen-bis-2-hydroksy-3-naftoatet), stearatet, 3-hydroksy-2-naftoatet, p-toluensulfonatet, metansulfonatet, laktatet og suraminsaltene.
Spesielt verdifulle er følgende forbindelser og deres farmasøytisk godtagbare syreaddisjonssalter: 1,3-di-0- (n-heksadecyl)-2-0-(3-aminopropyl)-glycerol, 1,3-di-0-(n-heksadecyl)-2-0-(meta-aminometylbenzyl)-glycerol, 1,3-di-0-(n-heksadecyl)-2-0-(meta-aminometylfenyl)-glycerol.
Forbindelsene med formlene I og II ovenfor fremstilles ifølge oppfinnelsen ved at
(a) en forbindelse med formelen
reduseres, eller en forbindelse med formelen reduseres, hvor Y' er valgt fra alkylen med 1 til 3 karbonatomer; hvor den venstre binding er forbundet med 0; og
hvor q er som ovenfor angitt og hvor venstre
binding er forbundet med 0, for fremstilling av en forbindelse med formel I eller II hvor er hydrogen; eller
(b) en forbindelse med formelen

Fagmannen vil forstå at flere forbindelser med formlene I og II kan fremstilles ved å bruke selvfølgelige variasjoner av de ovenfor beskrevne syntesemetoder.
Den antivirale aktiviteten til forbindelsene fremstilt ifølge oppfinnelsen ble bestemt ved hjelp av to uavhengige metoder.
I den første administreres testforbindelsen til mus intraperitonealt 18 til 24 timer før de tilføres en dødelig dose av encefalomyokarditt (EMC) virus. Overlevelsesdata noteres i 10 dager efter tilførselen og sammenlignes med data for ubeskyttede dyr. Fremgangsmåten ifølge hvilken medisinen gis 18 til 24 timer tidligere, og på et sted som er tydelig forskjellig fra virus-infeksjonsstedet, skal eliminere lokaleffekter mellom medisin og virus og identifisere bare de forbindelser som gir en systemisk antiviral respons.
I den andre fremgangsmåte behandles enkeltskikt fra nese-polyppceller hos mennesker som er dyrket på mikrotiter-plater, med testforbindelsen omtrent 18 timer før behandling med en dødelig dose av vesikuløs stomatittvirus (VSV). Testforbindelsen vaskes vekk fra enkeltskiktene før virusbehandlingen. Kulturvæske som ekstraheres fra platene efter en inkubasjonstid efter virusbehandlingen titreres på mengden infektuøst virus som foreligger i mikrotiter-plater av L-929 musefibroblast. Sammenligning gjøres med virusutbyttedata for kulturvæske som ekstraheres fra ubeskyttede polyppceller.
Videre ble mange av forbindelsene fremstilt ifølge oppfinnelsen testet på sin evne til å øke den kjente antivirale aktivitet for polyinosin:polycytidy1-syre. Endelig ble visse av forbindelsene testet på sin evne til å indusere sirkulerende interferon hos mus efter parenteral administrering, ved å bruke den metode som er beskrevet av Hoffman, W.W. et al., Antimicrobial Agents and Chemotherapy, 3, 498-501 (1973).
Parenteral, lokal eller intranasal administrering av de ovenfor beskrevne aminer til et pattedyr før det ut-
settes for et infektiøst virus gir rask motstand mot viruset. Administreringen bør fortrinnsvis finne sted fra ca. 2 dager til
ca. 1 dag før eksponering for viruset, selv om dette vil variere noe med den spesielle dyreart og det spesielle infektiøse virus.
Når materialene fremstilt ifølge oppfinnelsen administreres, brukes de lettest og mest økonomisk i en dispergert form i en godtagbar bærer. Når det sies at dette materiale er dispergert, betyr det at partiklene kan være av molekylær størrelse og holdt i virkelig løsning i et egnet løsningsmiddel, eller at partiklene kan være av kolloidal størrelse og dispergert i en væskefase i form av en suspensjon eller emulsjon. Uttrykket "dispergert" betyr også at partiklene kan være blandet med og spredt i en fast bærer,
slik at blandingen foreligger i form av et pulver eller et støv. Dette uttrykk skal også omfatte blandinger som er egnet for bruk som spray, omfattende løsninger, suspensjoner eller emulsjoner av midlene fremstilt ifølge oppfinnelsen.
Når materialene fremstilt ifølge oppfinnelsen administreres parenteralt (subkutant, intramuskulært, intraperitonealt), brukes de i en mengde av fra ca. 1 mg/kg legemsvekt til ca. 250 mg/kg legemsvekt. Det favoriserte område er fra ca. 5 til ca. 100 mg/kg legemsvekt og det foretrukne område fra ca. 5 til ca. 50 mg/kg legemsvekt. Doseringen er naturligvis avhengig av hvilket pattedyr som behandles og hvilken spesiell amin-forbindelse som brukes, og må bestemmes av den person som er ansvarlig for administreringen av forbindelsen. Vanligvis vil små doser bli administrert til å begynne med, og
med gradvis økning av doseringen inntil den optimale mengde er bestemt for den spesielle pasient som behandles.
Bærere som er egnet for parenteral injeksjon kan enten være vandige som f.eks. vann, isotonisk saltløsning, isotonisk dekstrose, Ringer's løsning, eller ikke-vandige som f.eks. fete oljer av vegetabilsk opprinnelse (bomullsfrø, peanøtt-olje,
mais, sesam) og andre ikke-vandige bærere som ikke vil innvirke på effektiviteten til preparatet og er ikke-toksiske i det volum eller den mengde de brukes (glycerol, etanol, propylenglykol, sorbitol). Videre kan det fremstilles blandinger som er egnet for improvisert fremstilling av løsninger før administrering.
Slike blandinger kan omfatte flytende fortynningsmidler som f.eks. propylenglykol, dietylkarbonat, glycerol, sorbitol.
Ved praktisering av den nasale administreringsmåten av forbindelsene fremstilt ifølge oppfinnelsen kan enhver praktisk metode brukes for å bringe det antivirale midlet i kontakt med pattedyrets luftveier. Effektive metoder omfatter administrering av midlet ved hjelp av intranasale eller nese-svelg-dråper og ved inhalering slik det avleveres fra en forstøvningsinnretning eller en aerosol. Slike metoder for administrerinq er av praktisk betydning fordi de utgjør en enkel, sikker og effektiv metode for utførelse av oppfinnelsen. For intranasal administrering av midlet, vanligvis i en godtagbar bærer, er en konsentrasjon av midlet mellom 1,0 og 100 mg/ml tilfredsstillende. Konsentrasjoner i området av ca. 30 til 50 mg/ml tillater administrering av et passende volum av materialet.
For lokal påføring brukes de antivirale midlene mest hensiktsmessig i en godtagbar bærer for å tillate lett og reguler-bar applikasjon og bedre absorpsjon. Også her er konsentrasjoner i området fra ca. 1,0 til ca. 250 mg/ml tilfredsstillende. Vanligvis vil det i de to ovenstående metoder for administrering bli administrert en dose i området av ca. 1,0 til ca. 250 mg/kg legemsvekt, og fortrinnsvis fra ca. 5,0 til ca. 50 mg/kg legemsvekt.
Forbindelsene som anvendes i foreliggende oppfinnelse,
kan anvendes alene, dvs. uten andre medisiner, som blandinger av flere enn en av de her beskrevne forbindelser, eller i kombinasjon med andre medisinske midler som f.eks. analgetika, anestetika, antiseptika, dekongestanter, antibiotika, vaksiner, buffermidler og uorganiske salter, for å skaffe ønskelige farmakologiske egenskaper. Videre kan de administreres i kombinasjon med hyaluronidase for å unngå eller i det minste minske lokal irritasjon og øke absorpsjonshastigheten for forbindelsen. Hyaluronidase-mengder på minst ca. 150 (U.S.P:)-enheter er effektive i dette henseende, selv om større eller mindre mengder naturligvis kan brukes.
De materialer fremstilt ifølge oppfinnelsen som er vann-uløselige, omfattende de som er lite og/eller vanskelig løselige i vann, administreres for å oppnå optimale resultater,
som preparater, f.eks. suspensjoner, emulsjoner, som tillater dannelse av partikkelstørrelser på mindre enn ca. 20y. Partikkel-størrelsene i blandingene påvirker tilsynelatende deres biologiske aktivitet ved bedre absorpsjon av det aktive materiale. Ved sammenblanding av disse matex-ialer brukes forskjellige overflateaktive midler og beskyttelseskolloider. Egnede overflateaktive midler er partialestrene av vanlige fettsyrer, som f.eks. laurin-, olje- og stearinsyre, med heksitolanhydrider oppnådd fra sorbitol, og polyoksyetylenderivatene av slike esterprodukter. Slike produkter selges under varemerkene "Span" og "Tween", respektive, og er tilgjengelige fra ICI United States Inc., Wilmington, Del. Celluloseetere, spesielt cellulosemetyleter ("Methocel", til-gjengelig fra Dow Chemical Co., Midland, Mich) er meget effektive som beskyttelseskolloider for anvendelse i emulsjoner inneholdende materialene ifølge oppfinnelsen.
For å oppnå optimale resultater administreres de vann-løselige materialer som er beskrevet her, i vandig løsning.
Typisk administreres materialene i fosfatbufret saltløsning. De vann-uløselige forbindelsene administreres i blandinger av den type som er beskrevet ovenfor eller i forskjellige andre blandinger som anført tidligere. Dimetylsulfoksyd tjener som en egnet bærer for vann-uløselige forbindelser. En representativ blanding av slike forbindelser omfatter sammenblanding av 25 til 100 mg av den valgte medisin som emulsjon ved å smelte og blande den med like deler av polysorbat 80 og glycerol til hvilken varmt (80°C)
vann tilsettes under kraftig omrøring. Natriumklorid tilsettes i en konsentrert løsning til en sluttkonsentrasjon på 0,14M og natriumfosfat med pH 7 tilsettes til en sluttkonsentrasjon på 0,01M for eksempelvis å gi følgende representative blanding:
I visse tilfeller, som når det opptrer klumper av medisinpartiklene, brukes ultralydbehandling for å skaffe en homogen blanding.
Følgende eksempler illustrerer oppfinnelsen.
Eksempel 1
1, 3- di- 0-( n- heksadecyl)- 2- 0- ( 3- aminopropy1)- glycerol- hydroklorid A. 1, 3- di- 0-( n- heksadecy l)- 2-0-( 2- cyanoetyl)-glycerol
En blanding av 1,3-di-0-(n-heksadecyl)-glycerol (80 g, 148 mmol), akrylonitril (1,49 kq, 28,1 mol) og vandig 2N natriumhydroksyd (1,2 1) ble oppvarmet til 50°C. Tetrabutylammoniumhydroksyd (19,2 g 40 vekt%ig vandiq løsning, 29,15 mmol) ble langsomt tilsatt, hvilket fikk temperaturen i den eksoterme reaksjonsblandingen til å stige til ca. 80 til 90°C. Reaksjonsblandingen ble så omrørt i 20 minutter uten ytre oppvarmning, fulgt av avkjøling til 20°C og tilsetning av vann (1,0 1). Et fast materiale, en blanding av uomsatt og cyanoetylert 1,3-di-0-(n-heksadecyl)-glycerol ble isolert og igjen behandlet med ny akrylonitril (1,49 kg, 28,1 mol), vandig 2N natriumhydroksyd (1,2 1) og tetrabutylammoniumhydroksyd (19,2 g, 40%ig vandig løsning, 29,15 mmol) i 20 minutter med omrøring ved 50°C, fulgt av avkjøling og tilsetning av vann (1,0 1). Det resulterende 1,3-di-0-(n-heksadecyl)-2-0-(2-cyanoetyl)-glycerol-faststoff ble frafiltrert, vasket i rekkefølge med vann, metanol og acetonitril og tørret [82 g, 93% utbytte, sm.p. 45-46°C,

B. Tittelforbindelsen
En blanding av 1,3-di-0-(n-heksadecyl)-2-0-(2-cyanoetyl)-glycerol (20,5 g, 34,5 mmol), tetrahydrofuran (200 ml), etanol (10 ml) og Raney-nikkel-katalysator (3 g) ble mettet med ammoniakkgass ved 0 til 5°C og så hydrogenert (3,5 kg/cm ) i en Paar hydrogenator i 3 timer ved romtemperatur. Blandingen ble så filtrert, katalysatoren vasket med tetrahydrofuran (50 ml) og totalfiltratet ble inndampet i vakuum til en olje. Denne fremgangsmåte ble gjentatt ytterligere tre ganger med ferske reaktanter og katalysator og det ble oppnådd ialt 77 g olje. Oljen ble oppløst i eter (500 ml)
og løsningen vasket med 2 vekt%ig, vandig ammoniumhydroksyd-
løsning (500 ml), tørret (MgSO^), filtrert og inndampet i vakuum for å gi et fast stoff. Faststoffet ble oppløst i metanol (300 ml) og løsningen mettet med hydrogenkloridgass og så inndampet i vakuum til et fast stoff. Dette faststoff ble krystallisert fra etylacetat for å gi det navngitte produkt med svak forurensning (63 g, 72% utbytte, sm.p. 69-70 C) og så omkrystallisert to ganger fra isopropanol:acetonitri1 (1:1, 800 ml),

Den kombinerte eterekstrakt ble vasket med vann (200 ml), tørret (MgSO^), filtrert og inndampet i vakuum for å gi 1,3-di-O-(n-heksadecyl)-2-0-(2-karboksyetyl)-glycerol-faststoff (4,5 g), som ble renset ved silikagelkromatografi (eluering med toluen:etanol)

(3,5 g, 5,7 mmol) ble oppløst i en blanding av benzen (55 ml) og konsentrert svovelsyre (5,89 g) . Hydrazoinsyre (6,34 ml av en 4,65 vekt%ig benzenløsning, 6,0 mmol) ble så tilsatt dråpevis og den resulterende blanding ble omrørt i 2 timer ved romtemperatur. Tynnskiktkromatografi (TLC) analyse viste omtrent 50% reaksjon av 2-karboksyetyl-forbindelsen. Ytterligere hydrazoinsyre (6,34 ml 4,65%ig benzenløsning, 6,0 mmol) ble tilsatt dråpevis og reaksjons-løsningen omrørt i ytterligere 16 timer ved 40°C. TLC-analyse viste nå at reaksjonen i hovedsak var fullstendig. Vann (50 ml)
og vandig 2N natriumhydroksyd ble så tilsatt og den resulterende blanding ekstrahert med eter (3 x 200 ml). Den kombinerte eterekstrakt ble tørret (Na-^SO^), filtrert, mettet med hydrogenkloridgass og inndampet i vakuum for å gi et fast stoff. Faststoffet ble renset ved silikagelkromatografi (eluering med kloroform:metanol) og omkrystallisert fra varm etylacetat

1,3-di-0-(n-heksadecyl)-2-0-(3-aminopropyl)-glycerol-/ hydroklorid (1,0 g, 1,6 mmol) ble tilsatt til en blanding av kaliumkarbonat (830 mg, 6,0 mmol) og benzen (75 ml). Acetylklorid (150 mg, 1,9 mmol) ble så tilsatt og den resulterende blanding omrørt i 1 time under tilbakeløp. Ytterligere acetylklorid (150 mg, 1,9 mmol) ble så tilsatt og reaksjonsblandingen omrørt i 1 time til under tilbakeløp. TLC-analyse viste at reaksjonen var i det vesentlige fullstendig. Reaksjonsblandingen ble avkjølt, vann (75 ml) tilsatt og den resulterende blanding ekstrahert med eter (3 x 100 ml). Den kombinerte eterekstrakt ble tørret (MgSO^), filtrert og inndampet i vakuum for å gi den navngitte forbindelsen [800 mg, 79% utbytte, sm.p. 53-54°C,

1,3-di-0-(n-heksadecyl)-2-0-(3-acetamidopropyl)-glycerol (700 mg, 1,1 mmol) ble oppløst i eter (100 ml) og behandlet med litiumaluminiumhydrid (500 mg, 13 mmol). Vann (100 ml) ble så tilsatt og blandingen ekstrahert med eter (2 x 100 ml). Den kombinerte eterekstrakt ble tørret (MgSO^), filtrert, behandlet med hydrogenkloridgass og inndampet i vakuum til et fast stoff som ble omkrystallisert fra varm etylacetat,

1, 3-di-0- (n-heksadecyl)-2-0-(3-aminopropyl)-glycerol-hydroklorid (700 mg, 1,1 mmol) ble oppløst i en løsning av eddiksyre (1,05 ml), natriumacetat (350 mg, 4,3 mmol) og aceton (1,3 ml). Natriumborhydrid (1,25 g, 33 mmol) ble tilsatt i små porsjoner
inntil TLC-analyse viste at all 3-amino-propyl-forbindelse var forbrukt. Reaksjonsblandingen ble så behandlet med vandig 2N natriumhydroksyd (20 ml) og vann (20 ml) og ekstrahert med eter
(3 x 40 ml). Den kombinerte eterekstrakt ble tørret (MgS04), filtrert, behandlet med hydrogenkloridgass og så inndampet i vakuum til et fast stoff som ble omkrystallisert fra varm etylacetat (210 mg, faststoffet inneholdt ca. 1/2 mol H^ O pr. mol av nevnte produkt, 28% utbytte, sm.p. 72-73°C,

Natriumhydrid (1,78 g av en 50 vekt%ig dispersjon i mineralolje, 37 mmol) ble tilsatt ved 60°C til en løsning av 1,2-di-0-(n-heksadecyl)-glycerol (10 g, 18,5 mmol) i N,N-dimetyl-formamid (100 ml), og den resulterende løsning omrørt i 20 minutter ved 60°C. Allylbromid (4,47 g, 37 mmol) ble så tilsatt dråpevis,
og den resulterende blanding omrørt i 3 timer ved 90°C, avkjølt, forsiktig fortynnet med vann (200 ml) for å stanse reaksjonen, og ekstrahert med eter (3 x 150 ml). Osmiumtetroksyd (90 mg, 0,354 mmol) ble tilsatt til en løsning av 1,2-di-0-(n-heksadecyl)-3-0-allyl-glycerol (4,5 g,
7,75 mmol) i tetrahydrofuran:vann (3:1, 120 ml), og den resulterende oppløsning omrørt i 5 minutter ved romtemperatur. Natriumperjodat
(9 g, 42 mmol) ole så tilsatt og reaksjonsløsningen omrørt i
16 timer ved romtemperatur under nitrogen. Reaksjons løsningen ble så fortynnet med vann (150 ml) og ekstrahert med eter (2 x 150 ml). Den kombinerte eterekstrakt ble vasket med vann (150 ml), tørret
(MgSO^) og inndampet i vakuum til en olje, som ble renset ved silikagelkromatografi (eluering med benzen:etylacetat,

C. Tittelforbindelse
Natriumcyanoborhydrid (0,1 g, 1,6 mmol) ble tilsatt til en løsning av 1,2-di-0-(n-heksadecyl)-3-0-formylmetyl-glycerol (1,5 g, 2,6 mmol) og isopropylamin (0,89 g, 15 mmol) i metanoi: tetrahydrofuran (1:1, 50 ml), og blandingen ble omrørt i 2 timer ved romtemperatur. Så ble pH justert til 6 med 5N metanolisk saltsyre, ytterligere natriumcyanoborhydrid (0,1 g, 1,6 mmol) tilsatt, og reaksjonsblandingen så omrørt i 60 timer ved romtemperatur, filtrert, behandlet med vandig, 3N natriumhydroksyd

Boranmetylsulfid (BMS)-kompleks (6,5 ml, 68,5 mmol) ble
tilsatt ved 0 til 5°C til en løsning av 1,3-di-O-(n-heksadecyl)-2-0-ally1-glycerol (10,82 g, 18,6 mmol) fremstilt som i eksempel 11A) i heksan (190 ml), og den resulterende løsning omrørt i 3 timer ved romtemperatur. Reaksjonsløsningen ble så avkjølt igjen til 6 til 5°c og etanol (17,3 ml) tilsatt dråpevis for å spalte gjenværende BMS. Reaksjonsløsningen ble så behandlet med vandig, 3N natrium-

(eluering med benzen:etanol) [5 g, 45% utbytte, sm.p. 29°C,

B. 1, 3- di- O-( n- heksadecyl)- 2- 0-[ 3-( p- tosyloksy) propyl]-glycerol
1,3-di-O-(n-heksadecyl)-2-0-(3-hydroksypropyl)-glycerol (8,0 g, 13,4 mmol) ble tilsatt ved 10°C til en løsning av p-toluensulfonylklorid (5,25 g, 27,5 mmol) og pyridin (10 ml) i metylenklorid (200 ml), og blandingen omrørt i 60 timer ved romtemperatur. Vann (200 ml) ble så tilsatt, metylenkloridet og den vandige fase separert og den sistnevnte ekstrahert med metylenklorid (2 x 150 ml). De tre metylenkloridskiktene ble kombinert, vasket med vann
(2 x 150 ml), tørret (MgSO^), filtrert og inndampet i vakuum. Det resulterende tosylat ble renset ved silikagelkromatografi (eluering med benzen) [3,0 g, 30% utbytte, olje,

På samme måte som beskrevet i eksempel 12B ble den ovennevnte forbindelse fremstilt ved å omsette 1,2-di-O-(n-heksadecyl )-glycerol med p-toluensulfonylklorid. Rensing ble gjennom-ført ved omkrystallisering fra etylacetat,

En blanding av 1,2-di-O-(n-heksadecyl)-3-0-(p-tosyl)-glycerol (1,4 g, 2,0 mmol), natrium-4-cyanofenolat (0,5 g, 3,5 mmol) og xylen (100 ml) ble omrørt i 16 timer med tilbakeløp. Siden reaksjonen fortsatt ikke var fullstendig, ble xylenet fjernet ved destillasjon og erstattet med N,N-dimety1formamid (100 ml), og den resulterende løsning omrørt i ytterligere 16 timer ved 150°C. (0,6 g, 0,93 mmol) ble tilsatt til en suspensjon av litiumaluminiumhydrid (0,3 g, 7,9 mmol) i eter (25 ml), og den resulterende blanding omrørt i 30 minutter ved romtemperatur. Vann (25 ml) ble så tilsatt forsiktig, eter- og vannfasen skilt, og den sistnevnte ekstrahert med eter (3 x 25 ml) og etylacetat (25 ml). De fem organiske ekstraktene ble kombinert, tørret (MgSO^), filtrert og inndampet i vakuum til en olje som ble oppløst i eter. Løsningen ble behandlet med hydrogenkloridgass, noe som forårsaket utfelning av et fast stoff

På samme måte som beskrevet i eksempel 12A og B ble 1,2-di-0-(n-heksadecyl)-3-0-allyl-glycerol omsatt med BMS, og de resulterende 2-hydroksypropyl- og 3-nydroksypropyl-forbindelser omdannet til de tilsvarende tosylåtene. B. 1,2-di-O-(n-h eksadecyl)- 3- 0-( 2- azidopropyl)- glycerol
Den resulterende blanding av tosylatene (3,0 g, 4,0 mmol)
ble oppløst i N,N-dimetylacetamid (50 ml) og behandlet med en løsning av natriumazid (0,326 g, 5,0 mmol) i vann (5 ml) i 16
timer ved 90°C. Reaksjonsløsningen ble så avkjølt, fortynnet med vann (200 ml) og ekstrahert med eter (2 x 150 ml). Den kombinerte eterekstrakt ble vasket med vann, tørret (MgSO.), filtrert og inn--1 dampet i vakuum til en olje [2 g, 81% utbytte, IR (CHC13) 2100 cm ,], en blanding av 2-azidopropyl- og 3-azidopropyl-forbindelsene, som ble brukt uten videre rensning i neste trinn.
C. Tittel forbindelse
Den resulterende blanding av azider (2 g, 3,2 mmol) ble oppløst i eter (100 ml), behandlet med litiumaluminiumhydrid (0,4 g, 10,5 mmol) og omrørt i 2 timer ved romtemperatur. Over-skudd hydrid ble ødelagt ved forsiktig tilsetning av etanol -(10 ml) og vann (150 ml), og blandingen så ekstrahert med eter (2 x 100 ml). Den kombinerte eterekstrakt ble tørret (MgSO^), filtrert, og konsentrert i vakuum til en olje (1,8 g), som ble renset ved silikagelkromatograf i (eluering med ben zen : etanol) og., så. omdannet til hydrokloridsaltet ved oppløsning og behandling med hydrogenkloridgass. Saltet ble omkrystallisert fra etylacetat (0,21 g, faststoff inneholdende ca. 1/2 mol ti^ O pr. mol av ovennevnte produkt, 10% utbytte, sm.p. 56-68°C,

Sammenblanding til en emulsjon ble utført ved å smelte
og oiande like deler av nevnte forbindelse, polysorbat 80 og glycerol, og så dispergere blandingen i varmt vann under kraftig omrøring. Blandingen ole så justert til sluttkonsentrasjoner på 0,14M natriumklorid og 0,01M natriumfos fat, pH 7. Videre fortynninger ole utført med 0,14M natriumklorid - 0,01M natriumfos fat,
pH 7 bufferløsning.
Tre grupper på 10 albino hunnmus (20-25 g legemsvekt)
ble gitt 0,5 ml intraperitoneale injeksjoner inneholdende doserings-
nivåer på 1,5, 5 og 15 mg av nevnte forbindelse/kg legemsvekt, resp. En fjerde kontrollgruppe på 10 mus ble ikke gitt slik injeksjon. 18 til 24 timer senere ble alle fire grupper tilført 0,2 ml ved subkutan injeksjon inneholdende 20 ganger LD^^, den doseringsmengde som forårsaker 50% døde hos ubeskyttede mus efter 10 dager, av encefalomyokarditt (EMC)-virus. Overlevelsesdata ble nedtegnet i de neste 10 dager og relativ overlevelse (S ) beregnet:
Antiviral aktivitet uttrykkes som relativ overlevelse (S^) i forsøksgruppene sammenlignet med kontrollen på den 10. dag efter tilførselen av virus. S rdefineres ved formelen:

Reduksjon av virus-ut bytte på menn eskelige polyppceller i n vitro med 1, 3-di- O-( n- he ksadecyl)-2-0-( 3- aminopropy1)glyce rol-hydr oklorid
Vekstmedium ble fremstilt ved å tilsette til Eagle's medium med minimum essensielle stoffer (100 ml), 100X konsentrert antibiotisk antimykotisk løsning (2 ml), 200 mM glutaminløsning (1 ml), 100X konsentrert løsning av ikke-essensielle aminosyrer (1 ml), 100 mM natriumpyruvatløsning (1 ml) og varme-inaktivert kalvefosterserum (10%). Hver brønn i 96-brønns mikrotiterplater ble tilsatt ca. 50000 menneskelige nesepolyppce1ler suspendert i 0,2 ml vekstmedium. Platene ble så inkubert i 8-10 dager ved 37°C i en 5%ig C02_atmosfære for å etablere enkeltskikt av celler.
Ved slutten av den 8-10 dagers cellevekstperiode ble sammenhengende monoskikt på platene vasket fire ganger med fosfatbufret saltløsning og straks efter behandlet med 0,2 ml pr. brønn av et vedlikeholdsmedium inneholdende 10, 5,0, 1,0,
0,5, 0,1 og 0 ug/ml av nevnte forbindelse., respektive. Vedlike-holdsmediet var identisk med vekstmediet beskrevet ovenfor,
bortsett fra at mengden av nevnte kalvefosterserum var 2%. Platene ble inkuoert i ytterligere 18 timer ved 37°C, og monoskiktene så vasket fire ganger med fosfatbufret saltløsning for å fjerne nevnte forbindelse, tilført et preparat inneholdende ca. 1000 ganger TCID^q, dvs. det doseringsnivå som forårsaker en 50%ig infeksjonsgrad i ubeskyttede kulturer, av vesikuløs stomatittvirus (VSV) i en 2 timers adsorpsjonsperiode (37°C), vasket fire ganger med fosfatbufret saltløsning for å fjerne uabsorberte viruspartikler,
og gjenmatet med 0,2 ml vedlikeholdsmedium pr. brønn. Platene ble så inkubert i 7 timer ved 37°C, og kulturvæsken fra 5-8 likedanne celler høstet fra hver plate, lagret frosne i reagensglass, og så titrert for bestemmelse av mengden infektuøst virus som var til stede i mikrotiterplatene av L-929 musefibroblast. L-929 muse-kulturer ble undersøkt mikroskopisk og analysert ca. 3-4 dager senere med følgende prosentuelle minskning i virusutbytte
(i forhold til kontrollen) bestemt for de fem testede konsentrasjoner av den nevnte forbindelse:

Eksempel 68
Evne for 1, 3- di- O-( n-he ksadecyl)-2- 0-( 3- aminopropyl)- qlycerol-hydroklorid til å indusere s irku lerende interferon
En blanding av like vektmengder av nevnte forbindelse, polysorbat 80 og glycerol ble smeltet og så homogenisert i varm 0,14M natriumklorid inneholdende 0,01M natriumfos fat, pH 7 (PBS) . Den resulterende olje-i-vann emulsjon kunne lett fortynnes med
PBS for administrering.
Sveitsiske hunnmus (20-25 g kroppsvekt) ble injisert (0,5 ml, intraperitonealt) en mengde av den ovenstående, fortynnede emulsjon inneholdende 25 mg av nevnte forbindelse pr. kg kroppsvekt. 8, 12, 16 og 20 timer efter injeksjonen ble det uttatt prøver av plasma fra fire mus som ble kombinert. Serie-fortynninger i L-15 (Leibovitz) medium inneholdende 5% kalvefoster-medium ole inkubert i mikrotiterplater over natten ved 37°C på sammenhengende monoskikt av L-929 musefibroblast. Monoskiktene ble så vasket med proteinfritt medium, tilført 10 ganger TCID_0# dvs. det doseringsnivå som forårsaker en 50%ig infeksjonsgrad hos ubeskyttede kulturer av vesikuløs stomatittvirus (VSV) i en times (37°C) adsorpsjonsperiode, vasket, behandlet på nytt med L-15-medium inneholdende 5% kalvefosterserum og så igjen inkubert
i 48 timer ved 37°C. L-929-kulturene ble så undersøkt mikroskopisk på viral cytopatologi og analysert for bestemmelse av plasma-interferon-nivået, det resiproke av plasmafortynningen, som gir 50% beskyttelse for L-929 monoskiktene.
Et andre forsøk fulgte ovenstående fremgangsmåte, bortsett fra at det ble injisert 10 mg av nevnte forbindelse pr. kg kroppsvekt i musene og at prøver av peritonealvask ble uttatt fra fire mus og samlet 6, 9, 12, 15 og 18 timer efter injeksjonen. Prøvene ble gjort ved å eksponere peritonealmembranet, injisere 1 ml Hank's balanserte saltløsning inneholdende 100 penicillin-enheter/ml og 100 g strptomycin/ml i peritoneal-hulen, kort massere buken og så aspirere peritonealvasken.

Økning av polyinosin- polycytidy1- syr e ( poly ( I:C))- indusert cellemotstand mot virusi nfeksjon ved hjelp av 1, 3- di-Q-( n- heksadecy1)- 2- 0-( 3- aminoprop yl)-gly cerol-hydro klorid
Vekstmedium ble fremstilt fra Eagle's medium med minimum av essensielle stoffer ved tilsetning av 100X konsentrert antibiotisk antimykotisk løsning (2 ml), 200 mM glutamin-løsning (1 ml) og varme-inaktivert kalvefosterserum (5%). L-929 musefibroblaster ble suspendert i vekstmediet og hver brønn i en 9 6-brønns mikro-titerplate ble tilsatt 0,2 ml av nevnte suspensjon inneholdende 20.000 til 30.000 celler. Platene ble inkubert i 2 til 4 dager ved 37°C i en 5%ig C02-atmosfære for å etablere monoskikt av celler. Platene ble vasket fire ganger med fosfatbufret saltløsning like før behandling.
Poly(I:C) ble fremstilt i konsentrasjoner på 5,0, 1,0,
0,2 og 0,04 u<g>/ml i det ovenfor beskrevne medium minus kalveserumet. 0,1 ml av hver fortynning ble kombinert i et rutearrangement på L-929-celle-monoskiktene med 0,1 ml fortynninger inneholdende
20,0, 4,0, 0,8, 0,16 og 0,032 ug av nevnte forbindelse pr. ml av nevnte serumfrie medium. Kontrollbrønner ble eksponert for enten poly(I:C) eller den ovennevnte forbindelse alene. Platene ble inkubert i 6 timer ved 37°C i en 5%ig C02-atmosfære, vasket fire ganger med fosfatbufret saltløsning, og matet på nytt med 0,1 ml pr. brønn av vekstmedium inneholdende 2% kalvefosterserum. Efter 18 timers videre inkubering ble platene undersøkt på toksisitet og så tilført 0,1 ml pr. brønn av en vesikuløs stomatittvirus (VSV)-suspensjon inneholdende 10 til 30 ganger TCIDd-O^ (vevkultur-infiserende dose som forårsaker 50% infeksjonsgrad). Platene ble inkubert i ytterligere 3 til 4 dager og så undersøkt mikroskopisk på cytopatogenisk effekt (CPE). Celler som var beskyttet mot virus, var fri for CPE. Den minimale beskyttende dose (MPD) for poly(I:C) ble notert og øket antiviral aktivitet forårsaket av kombinasjonen med den ovennevnte forbindelse oppført for hvert fortynningsnivå av nevnte forbindelse.
Merknad: Kombinasjon av poly(I:C) med nevnte forbindelse gir samme antivirale effekt som en økning av poly(I:C)-konsentrasjonen det angitte antall ganger ville gi.

På samme måte som beskrevet i eksempel 73 ble økning av poly(I:C)-indusert cellemotstand mot virusinfeksjoner bestemt for de nedenfor oppførte forbindelser.
Claims (4)
1. Analogifremgangsmåte for fremstilling av en anti-viral forbindelse med formelen
og de farmasøytisk godtagbare syreaddisjonssalter derav,
hvor
og R2 hver er valgt fra normal alkyl med fra 12 til 20 karbon
atomer og normal alkenyl med 12 til 20 karbonatomer som ikke har en dobbeltbinding i 1-stilling,
Y er valgt fra alkylen med 2 til 4 karbonatomer, hvor de to
valenser er på forskjellige karbonatomer; orto-, meta- og para-fenylendimetylen; og
hvor q er et helt tall fra 1 til 3, og den venstre binding er forbundet med 0, og
R3 er valgt fra hydrogen og alkyl med 2 til 4 karbonatomer, karakterisert ved at (a) en forbindelse med formelen
reduseres, eller en forbindelse med formelen
reduseres, hvor Y' er valgt fra alkylen med 1 til 3 karbonatomer;
, hvor den venstre binding er forbundet med 0; og
hvor q er som ovenfor angitt og hvor venstre
binding er forbundet med 0, for fremstilling av en forbindelse med formel I eller II hvor R3 er hydrogen; eller (b) en forbindelse med formelen
omsettes med hydrazo-syre for å danne en forbindelse med formel I eller II hvor R~ er hydrogen; eller (c) en forbindelse med formelen:
"CH2-0_ ,
hvor den venstre binding er forbundet med 0; og
hvor q er som ovenfor angitt og hvor venstre
binding er forbundet med 0, omsettes med en forbindelse med formelen NH^R-^ under reduserende betingelser; og eventuelt alkyleres en fremstilt forbindelse med formel I eller II hvor R^ er hydrogen; og
eventuelt omdannes en fremstilt forbindelse med formel I eller II til et farmasøytisk godtagbart syreaddisjonssalt derav.
2. Fremgangsmåte som angitt i krav 1, karakterisert ved at det,fremstilles en forbindelse med formel I hvor R^ og R^ hver er n-heksadecyl,
