Description
Foreliggende oppfinnelse angår fremstilling av nye, terapeutisk aktive fenylpiperazinderivater med formelen:
hR v2or er R f1 eneyr l besnom zyelv, ekntlouerblet nezyr ls, ubCs,t_i,t-uaelkryt l meed lle1r -2 båsdube sR ti1toug-enter som består av halogen, trifluormetyl, C^_^-alkyl, Cj _^-alkoksy, hydroksy, nitro, amino, acetamido eller metansulfonamido; R^ er hydrogen eller ^-alkyl; Q er furan, tiofen, oksazol eller tiazol; m er 1-3 og n er 0 eller 1, forutsatt at når m er 2 og n er 0, kan R 1 ikke være metyl når Q er en tiazol-5-yl-gruppe; eller et farmasøytisk akseptabelt salt av et slikt derivat.
Foretrukne grupper av forbindelser med formel (I), er de med en eller flere av de følgende kjennetegn:
(a) m er 1 og n er 0,
(b) Q er tiazol,
(c) Q er oksazol,
(d) Q er tiofen,
(e) Q er furan,
(f) R 1 er Cj^-alkyl såsom metyl eller i-propyl,
(g) R^ er fenyl,

(h) R er benzyl,
(i) R 2er fenyl substituert med en eller to metylradikaler.
Forbindelsene med formel (I) fremstilles ifølge foreliggende oppfinnelse ved at man
(a) kondenserer en forbindelse med formelen:
hvor R 1 , R 3, Q, m og n har de ovenfor angitte betydninger, og hvor L representerer en avspaltende gruppe, fortrinnsvis halogen, med en forbindelse med formelen: hvor R 2 har den ovenfor angitte betydning, eller (b) reduserer en forbindelse med formelen: hvor R 1 , R 2, Q og m har de ovenfor angitte betydninger, hvorved man får dannet en forbindelse hvor n er 0; eller (c) ringslutter en forbindelse med formelen: 1 2 3 hvor R , R , R , m og n har de ovenfor angitte betydninger, slik at man får dannet en forbindelse med formel (I) hvor Q er oksazol, eller (d) når R 1 eller R 2 er fenyl substituert med amino, reduserer den tilsvarende nitro til amino som kan acetyleres eller metansulfoneres når fenylgruppen er substituert med acetamido eller metansulfonamido, eller (e) når R 1 eller R 2 er fenyl som er substituert med hydroksy, behandler den tilsvarende alkoksysubstituerte fenyl med reduserende midler,
og, om ønsket, omdanner en således erholdt forbindelse til et farmasøytisk akseptabelt salt derav.
Den ovennevnte kondensasjon (a) utføres fortrinnsvis i nærvær av en protonakseptor så som en base, f.eks. natriumkarbonat. Man kan bruke polare oppløsningsmidler så som alka-noler, f.eks. etanol, som er egnet oppløsningsmiddel for reaksjonen som fortrinnsvis utføres ved oppvarming under tilbakeløp. Den avspaltende gruppe L kan være et hvert radi-kal som er kjent for å være effektivt i slike kondensasjons-reaksjoner, og man kan nevne at halogenatomer, f.eks. klor, eller metylsulfonyl eller substituert benzensulfonylgrupper gir gode resultater i denne reaksjon. Reaksjonen utføres fortrinnsvis i temperaturområdet fra 20 til 150°C. Forbindelser med formel (VI) er enten kjente (se J.A.C.S. 56 470-1
(1934) og 53 1470-3(1931)) eller kan fremstilles ved kjent
teknikk.
Reduksjonen (b) av amidet med formel (VIII) kan ut-føres ved å bruke kjemiske reduksjonsmidler så som I^Hg eller litiumaluminiumhydrid. Eteroppløsningsmidler så som tetrahydrofuran kan brukes i reaksjonen som fortrinnsvis utføres ved temperaturer fra 0 til 80°C. Amidene med formel (VIII) kan fremstilles ved en reaksjon mellom et syreklorid (fremstilt f.eks. fra den tilsvarende syre ved en reaksjon med tionylklorid) med følgende formel: og et fenylpiperazin med formel:
Forbindelser med formel (X) vil normalt bli fremstilt ved å omsette den tilsvarende karbonylforbindelse med formel: med en passende fenylforbindelse med formel
i nærvær av en protondonor, f.eks. eddiksyre.
Den ovennevnte ringslutningsreak-sjon (c) og de reaksjonsbetingelser som er nødvendig for å utføre den ønskede ringslutning, vil være velkjent. Det vises
i denne forbindelse til Chemical Reviews, 75, 389-437 (1975) eller Advances in Heterocyclic Chemistry 17 (1974).
Forbindelser med formel (X)
kan således ringsluttes til forbindelser med formel:
ved å bruke dehydratiseringsmidler så som polyfosforsyre, fosforoksyklorid eller konsentrert svovelsyre. Reaksjonen utføres fortrinnsvis i temperaturområdet fra 80 til 180°C.
Forbindelser med formel (X) kan fremstilles ved å omsette det tilsvarende bromderivat med formel: og den tilsvarende base med formel
i nærvær av en syreakseptor så som natriumkarbonat.
Forbindelser med formel (XI) kan i seg selv fremstilles ved å omsette et amin med formel: med det tilsvarende syreklorid med formel
ved å bruke dimetylformamid som oppløsningsmiddel.
Forbindelser med formel (I) fremstilt ved de forannevnte fremgangsmåter kan isoleres som sådan eller i form av syreaddisjonssalter.
Syreaddisjonssaltene er fortrinnsvis farmasøytisk akseptable, ikke-toksiske addisjonssalter med egnede syrer, så som uorganiske syrer som saltsyre, hydrobromsyre, sal-petersyre, svovelsyre eller fosforsyre, eller organiske syrer så som organiske karboksylsyrer, eksempelvis kan nevnes glykolsyre, maleinsyre, hydroksymaleinsyre, eplesyre, vin-syre, sitronsyre, salicylsyre, o-acetoksybenzosyre, nikotin-syre eller isonikotinsyre, eller organiske sulfonsyrer så
som metansulfon-, etansulfon-, 2-hydroksyetansulfon-, toluen-p-sulfon- eller naftalen-2-sulfonsyre. Bortsett fra nevnte farmasøytisk ekseptable syreaddisjonssalter inngår også andre salter i forannevnte definisjon av syreaddisjonssalter, f. eks. de som kan dannes med pikrin eller oksalsyre. Videre kan syreaddisjonssaltene brukes for rensing av forbindelsene eller for fremstilling av andre, f.eks. farmasøytisk akseptable syreaddisjonssalter av andre syrer eller kan brukes for identifikasjon, karakterisering eller rensing av basene.
Et syreaddisjonssalt kan omdannes til en fri forbindelse på kjent måte, f.eks. ved behandling med en base så som et metallhydroksyd eller alkoksyd, f.eks. et alkalimetall-eller jordalkalimetallhydroksyd, eksempelvis kan nevnes litiumhydroksyd, natriumhydroksyd, kaliumhydroksyd eller kalsiumhydroksyd, med et metallkarbonat så som et alkali-metall- eller jordalkalimetallkarbonat eller -hydrogenkarbo-nat, med ammoniakk, eller med en hydroksylionebytningsharpiks eller ved hjelp av en annen egnet reagens.
Et syreaddisjonssalt kan omdannes til andre syre-addis jonssalter på kjent måte, f.eks. så kan et salt med en uorganisk syre behandles med et metallsalt, f.eks. et natrium-, barium- eller sølvsalt, og en syre i et egnet oppløsnings-middel hvori det resulterende uorganiske salt er oppløselig, og kan således fjernes fra reaksjonsmediet. Et syreaddisjonssalt kan også omdannes til andre syreaddisjonssalter ved behandling med anioniske utbytningsharpikser.
Forbindelser med formel (I) og deres farmasøytisk akseptable salter har vist seg å ha verdi ved profylaktisk og terapeutisk behandling av forskjellige lidelser som innbe-fatter hypersensitivitet, så som astma. Forbindelsene har lav toksitet.
som kan brukes i farmasøytiske preparater ifølge foreliggende oppfinnelse, er laktose, dekstrose, sukrose, sorbitol, manni-tol, propylenglycol, flytende parafin, myk hvit parafin, kaolin, smeltet silisiumdioksyd, mikrokrystallinsk cellulose, kalsium-silikat, silisiumdioksyd, polyvinylpyrrolidin, ceto<g>tearylalko-hol, stivelse, modifiserte stivelser, gummi acacia, kalsiumfos-fat, kakaosmør, etoksylerte estere, olje av teobroma, arachis-olje, alginater, tragakant, gelatin, sirupper, metylcellulose, polyoksyetylensorbitanmonolaurat, etyllaktat, metyl og propyl hydroksybenzoat, sorbitantrioleat, sorbitansesquioleat og oleylalkohol samt drivmidler så som triklormonofluormetan, di-klordifluormetan og diklortetrafluoretan. I forbindelse med
tabletter kan man tilsette et smøremiddel for å gi bedre passa-sje og binde sammen de pulveriserte ingrediensene, og denne blanding kan puttes i en form og stanses ut i form av tabletter. For slike formål kan man bruke aluminium, magnesium eller kal-siumstearater, talkum eller mineralolje.
De følgende eksempler illustrerer oppfinnelsen.
Eksempel 1
(a) 2- acetoksytioacetamid.
En strøm av tørr H^S-gass ble ført raskt inn i en oppløsning av 150 g (l,52 mol) 2-acetoksyacetonitril og 20 ml trietanolamin i 500 ml tørr etanol under kraftig røring. Den indre temperatur steg til ca. 55°C. Gass-strømmen ble opprett-holdt i 3 timer hvoretter en tynnsjiktkromatografiprøve indi-kerte at reaksjonen var fullstendig. Overskudd av H^S-gass ble blåst av med en strøm av nitrogen, og etylalkoholen ble fordampet og man fikk et residuum. Dette noe klebrige krystallinske residuum ble ekstrahert med kokende eter, og en liten uoppløse-lig rest ble kastet. De samlede ekstrakter ble behandlet med avfargende trekull, filtrert og fordampet til ca. k00 ml. Ved av-kjøling til 0°C fikk man ca. 170 g av et noe klebrig 2-acetoksytioacetamid. Dette ble brukt i neste trinn uten ytterligere karakterisering og rensning. (b) 2- hydroksvmetyl- 4- fenyltiazol.
39,9 g (0,2 mol) 2-bromacetofenon og 30 g (0,225 mol) 2-acetoksytioacetamid ble oppløst i 150 ml dioksan. Blandingen
ble rørt og oppvarmet på et dampbad i 15 minutter og man fikk en krystallinsk masse. kO ml vandig 5N saltsyre ble tilsatt, og opp-varmingen fortsatte i ytterligere 30 minutter. Dioksanen ble fordampet under vakuum, og residuumet ble behandlet med en vandig Na2C0^ oppløsning. Det faste stoff ble oppsamlet, vasket med vann og tørket. Etter omkrystallisering fra benzen veiet produktet 33,5 g (87$) og hadde et sm.p. på 88-89°C.
(c) 2- klormetyl- 4- fenyltiazol.
6,25 g (0,052 mol) tionylklorid ble dråpevis tilsatt en rørt oppløsning av 9,55 g (0,05 mol) 2-hydroksymetyl-4-fenyltiazol og 4,0 g (o,051 mol) pyridin i 60 ml tørr benzen ved romtemperatur. Blandingen ble rørt og så oppvarmet til koking i løpet av 1 time. Den avkjølte reaksjonsblanding ble så rystet med 2 x 70 ml HgO og tørket over vannfritt magnesiumsulfat. Oppløsningsmidlet ble fjernet under vakuum og den gjenværende røde olje ble ekstrahert med 40~60°C petroleter (lOO ml) idet man kastet et lite tjæreaktig residuum. Ekstraktet ble behandlet med avfargende trekull, filtrert og fordampet, og man fikk en gul-rød olje som utkrystalliserte ved avkjøling.
Utbytte 9,9 g 94,5$.
Det ble ikke utført ytterligere rensning, og materi-alet ble brukt direkte i neste trinn.
(d) 1-( 4- fenyl- 2- tiazolylmetyl)- 4- fenylpiperazin.
En blanding av urent (4,19 g» 0,02 mol) 2-klormetyl-4-fenyltiazol og 3,24 g (0,02 mol) N-fenylpiperazin og finpulverisert vannfritt natriumkarbonat i 60 ml absolutt etanol ble rørt og kokt under tilbakeløp i 8 timer. Oppløsningsmidlet ble fordampet til tørrhet, og det faste residuum suspendert i 100 ml vann og ekstrahert med 2 x 100 ml CH^Cl^. Ekstraktene ble vasket med vann og tørket over vannfritt magnesiumsulfat. Ved fordampning av oppløsningsmidlet og omkrystallisering av residuumet fra benzen/60-80°'s petroleterblanding (med trekullbehandling) fikk man det forønskede produkt som veide 5|03 g (75$ utbytte), og det haddet et sm.p. på 142-143°C.

Eksempel 20
(a) N- fenacylkloracetamid.
25 g (0,226 mol) kloracetylklorid ble langsomt tilsatt en rørt oppløsning av 25,9 S (0,151 mol) fenacylaminhydroklorid i 80 ml vannfri dimetylformamid (DMF) ved romtemperatur. Den indre temperatur steg til ca. 40°C. Blandingen ble rørt i 4 timer, hvoretter hovedmengden av DMF ble fordampet under vakuum. Residuumet ble behandlet med 200 ml vann, og det krystallinske bunnfall oppsamlet, vasket med vann og tørket. Utbytte 24,75 g (77$), sm.p. 118-119°C.
(b) 2- klormetyl- 5- fenyloksazol.
22,6 g N-fenacylkloracetamid (0,107 mol) ble blandet med polyfosforsyre (200 g), og massen ble oppvarmet under røring i 30 minutter ved 150°C. Den varme væsken ble så under kraftig
røring helt over i 800 ml vann. Den dannede blanding ble ekstrahert med 3 x 100 ml kloroform, ekstraktene ble vasket ved rysting med vann og så tørket over magnesiumsulfat. Kloroformen ble fordampet, og den gjenværende olje ekstrahert med 400 ml kokende
60-80°C petroleter. Den resulterende oppløsning ble behandlet med avfargende trekull for å fjerne tjæreaktige stoffer og så fordampet, hvorved man fikk en blekt gul olje som utkrystalliserte raskt. Utbytte 18,8g (91$), sm.p. 70-71°C.
(c) 1-( 5- fenyl- oksazol- 2- ylmetyl)~ 4- fenylpiperazin.
En oppløsning av 3,87g (0,02 mol) 2-klormety1-5-fenyloksazol og 3,25 g (0,02 mol) N-fenylpiperazin i 60 ml absolutt etanol ble kokt og rørt med finpulverisert vannfritt natriumkarbonat (5 g) i 6 timer.
Oppløsningen ble fordampet til tørrhet og behandlet med 60 ml vann. Den oppnådde suspensjon ble ekstrahert med 2 x 70 ml diklormetan. Ekstraktene ble tørket over magnesiumsulfat og fordampet til tørrhet. Det faste residuum ble utkrystallisert fra 80-100°'s petroleter ved hjelp av avfargende trekull.
Utbytte 4,67 g, sm.p. 96°C.

Eksempel 36
1-( 5- fenyloksazol- 2- ylmetyl)- 4-( 4- aminofenyl)- piperazin.
En oppløsning av 7,28 g (0,02 mol) 1-(5-fenyloksazol-2-ylmetyl)-4-(4-nitrofenyl)-piperazin i 50 ml iseddik ble hydrogenert over 5$ palladium-på-trekull-katalysator (lOO mg) ved atmosfærisk trykk og romtemperatur inntil opptak av hydrogen var fullstendig. Oppløsningsmidlet ble avdampet under vakuum, og residuumet behandlet med vandig natriumbikarbonatoppløsning og dette ga et hvitt krystallinsk faststoff. Vekt etter tør-king var 6,54 g ( 98%), sm.p. 130 C.
Eksempel 37
1-( 5- fenyloksazol- 2- ylmetyl)- 4-( 4- acetamidofenyl)- piperazin.
4-aminofenylforbindelsen fra eksempel 36 i en mengde på 3,0 g (0,009 mol) ble oppvarmet på et dampbad med 5 ml iseddik og 10 ml eddiksyreanhydrid i 1 time. Blandingen ble helt over i 200 ml vann, kraftig rørt og det oppsamlede bunnfall ble vasket med vann og tørket. Etter omkrystallisering fra metanol veide produktet 2,45 g og smeltet ved 196-197°C.
Eksempel 38
1-( 5- fenyloksazol- 2- ylmetyl)- 4-( 4- hydroksyfenyl)- piperazin.
5,02 g (0,066 mol) propan-1-tiol ble langsomt tilsatt en rørt suspensjon av 50$ natriumhydrid/mineraloljebland-ing (3,17 g, 0,066 mol) i 140 ml vannfri dimetylformamid.. Opp-løsningen av natriumpropan-1-tiolat som derved ble dannet, ble tilsatt 7,7g (0,022 mol) 1-(5-fenyloksazol-2-ylmetyl)-4-(4-metoksyfenyl)-piperazin. Oppløsningen ble oppvarmet under til-bakeløp på et oljebad i 10 timer under en nitrogenatmosfære. Blandingen ble helt over i 600 ml vann og ekstrahert med 3 x 200 ml etylacetat. Ekstraktene ble rystet flere ganger med vann, tørket og fordampet til et hvitt faststoff. Etter omkrystallisering fra dioksan/60-80°C petroleter veide produktet 5,5g (74,5$), og hadde et sm.p. på 189-190°C.
Eksempel 39
1-^2-(5-fenyloksazol-2-yl)-etyl7-4-fenylpiperazin.
(a) N- fenacyl- 3- brompropanamid.
25 g (0,146 mol) 3-brompropionylklorid ble langsomt tilsatt en rørt suspensjon av fenacylaminhydroklorid (17,15 g, 0,1 mol) i 50 ml tørr dimetylformamid ved romtemperatur. Den indre temperatur steg til 40°C. Blandingen ble rørt i 2 timer og nevnte DMF ble fjernet under vakuum. Residuet ble fortynnet med 200 ml vann og det krystallinske faste stoff oppsamlet, vasket med vann og tørket. Produktet veide 9,49 g (35$), og
smeltet ved llk°C.
(b) N-fenacyl- 3-( 4- fenylpiperazin- l- yl)- propanamid.
En oppløsning av 10,2 g (0,038 mol) N-fenacyl-3-brompropanamid og 12,0 g (0,074 mol) N-fenylpiperazin i 100 ml tørr DMF ble rørt sammen med 15,0 g vannfritt finpulverisert natriumkarbonat i 18 timer ved romtemperatur. Tempteraturen ble så hevet til 80°C og holdt der i 12 timer for å få en fullstendig reaksjon. Oppløsningsmidlet ble fordampet under vakuum, og residuumet fortynnet med 100 ml vann. Blandingen ble ekstrahert med 3 x 100 ml kloroform. Ekstraktene ble rystet med 2 x 100 ml 2N saltsyre og det organiske lag kastet. Det vandige lag ble gjort basisk med en NaOH-oppløsning til pH 9 og ekstrahert med 3 x 75 ml CHCl^. Ekstraktene ble vasket med vann og
tørket. Etter fjerning av oppløsningsmidlet ble den gjenværende viskøse gummi behandlet med eter og man fikk et oki-farvet fast stoff. Etter omkrystallisering fra isopropanol veide produktet 6,Og og smeltet ved 126°C.
(c ) 1-/1?- (5-f enyloksazol-2-yl)-etyl7-4-fenylpiperazin.
4,03 g (0,011 mol) N-fenacyl-3-(4-fenylpiperazin-l-yl ) -pr opanamid ble blandet med 36 g polyfosforsyre og rørt og oppvarmet ved l40°C i 30 minutter. Blandingen ble tilsatt 100 ml vann, avkjølt og ekstrahert med 2 x 30 ml kloroform. Ekstraktene ble kastet, og den vandige fase gjort basisk til pH 9 med 50$ vandig NaOH-oppløsning. Den dannede emulsjon ble ekstrahert med 3 x 100 ml kloroform, ekstraktene ble tørket og fordampet. Det krystallinske residuum ble omkrystallisert fra 60-80°C petroleter. Utbytte 2,6 g, sm.p. 84-85°C.

1-/5-(3-trifluormetylfenyl)-furan-2-ylmetyl7-4-fenylpiperazin. (a) 2- hydroksymetyl- 5-( 3- trifluormetylfenyl)- furan.
2-(3-trifluormetylfenyl)-furan (j. Chem. Soc. (c) I968, 2737 og Acta Chem Scand. 24, 2379 (1970)) (21,2 g, 0,1 mol) ble rørt i 100 ml tetrahydrofuran, avkjølt til -40°C hvoretter 0,1 mol i en oppløsning i heksan av n-butyllitium ble dråpevis tilsatt. Etter røring i 1 time ved -40°C ble 3,3 g (0,11 mol)
para-formaldehyd langsomt tilsatt. Blandingen ble så rørt ved
- kO°C i 30 minutter, hvoretter temperaturen langsomt steg opp-over. Ved 10°C startet en eksotermisk reaksjon og temperaturen steg gradvis til 40°C over 10 minutter og falt så tilbake til 20 oC. Etter røring i ytterligere 1 time ble den klare brune oppløsningen helt over en blanding av is og vann, som så ble justert til pH 4 med fortynnet saltsyre og ekstrahert via etylacetat. Det tørrede acetatekstrakt ble fordampet, og man fikk en brun gul viskøs olje som ble destillert i vakuum, og man fikk 2-hydroksymetyl-5-(3-trifluormetylfenyl)-furan som en farveløs væske med k.p. på 125°C/0,5 mm Hg (l6,2 g) som ved henstand ga krystaller med et sm.p. på 45°C.t
(b) 2- brommetyl- 5-( 3- trifluormetylfenyl)- furan.
4,84 g (0,02 mol) av hydroksymetylforbindelsen fremstilt som beskrevet ovenfor, ble oppløst i 30 ml dimetylformamid, avkjølt til 0-5°C, hvoretter tionylbromid i en mengde på 1,7 ml (0,02 mol) ble dråpevis tilsatt under røring. Etter 2 timer ble den grønlige til gule oppløsningen helt over på is og vann og ekstrahert via dietyleter. Eterekstraktet ble vasket med NaCl (mettet oppløsning) tørket over en molekylær sil 3A og fordampet og man fikk en lys brun olje som langsomt utkrystalliserte
(^,6 g).
(c) 1-/3-(3-trifluormetylfenyl)-furan-2-ylmetyl7-4-fenylpiperazin.
0,015 mol av brommetylproduktet fremstilt som beskrevet ovenfor ble rørt i 50 ml dioksan sammen med 2,75 g (0,017 mol) N-fenylpiperazin i nærvær av 1,5 g vannfritt natriumkarbonat,
og blandingen ble kokt under tilbakeløp i 7 timer, helt over i isvann og ekstrahert med kloroform. Kloroformekstraktet ble vasket med vann deretter med en mettet saltoppløsning og tilslutt fordampet, dette ga en olje som ved behandling med eterholdig saltsyre ga et lyst gult krystallinsk faststoff 2,1 g. Dette hydroklorid ble oppløst i 15 ml kloroform, rørt sammen med 15 ml vann hvoretter man tilsatte en mettet oppløsning av natriumkarbonat inntil man fikk alkalisk reaksjon. Fasene ble skilt, og kloroformlaget ble fordampet og man fikk en halmgul olje (l,7g)
som langsomt utkrystalliserte seg, og som så ble omkrystallisert fra petroleter 60/80°C, og man fikk den ovennevnte forbin-delse: 1-/3-(3-trifluormetylfenyl)-furan-2-ylmetyl7-4-fenylpiperazin, 1,2 g, sm.p. 94°C.
På lignende måte som beskrevet tidligere, ble føl-gende forbindelser fremstilt bortsett fra at de angitte hydroksy-metylmellomprodukter ble omdannet til klormetylforbindelser via tionylklorid.

of Chemistry, 26, 1147 (1973) (6,67 g, 0,03 mol) ble kokt under tilbakeløp i 100 ml benzen sammen med 4,5 ml tionylklorid i 1,5 time. Etter fjerning av tionylkloridoverskuddet, ble det gjenværende syreklorid i benzen dråpevis tilsatt en blanding av 7,23 g (0,03 mol) N-(3-bromfenyl)-piperazin og 4,5 ml (ca. 0,03 mol) trietylamin i 100 ml benzen. Etter røring i 1 time ved romtemperatur ble blandingen rystet med 100 ml vann. Den utskilte ben-zenfasen ble ytterligere vasket med en mettet saltoppløsning og så fordampet, dette ga en olje som lett lot seg utkrystallisere. ■ Produktet ble omkrystallisert fra etylacetat / 60-80°C petroleter (1/3 <V>/v), hvorved man fikk 1-/3-(4-klorfenyl)-2-furoyl7-
4-(3-bromfenyl)-piperazin, 12,3 g , sm.p, 130°C.
Det ovennevnte tertiære amid med en vekt på 4,45 g (0,01 mol) ble oppløst i 30 ml tørr tetrahydrofuran og dråpevis lander røring og avkjøling til 0°C tilsatt en oppløsning av di-boran i tetrahydrofuran (20 ml, 1 M oppløsning) (N.B. reaksjonen ble utført under nitrogen). Temperaturen steg til ca. 8°C, og den resulterende uklare oppløsning ble kokt under tilbakeløp i 1 time. Man fikk så en klar blekt gul oppløsning som ble oppvarmet på et dampbad i 15 minutter med 5N HC1 (20 ml). Etter ny avkjøling ble oppløsningen gjort alkalisk med 4N NaOH og ekstrahert med dietyleter. Eterekstraktet ble etter tørking over magnesiumsulfat fordampet, og man fikk en olje som ble omdannet til sitt monohydroklorid, hvorved man fikk den forønskede forbindelse i en vekt på 2,8 g med et sm.p. 217°C.

6,0 g (0,015 mol) 1-(5-fenyl-2-furoyl)-4-(3-trifluor-metylf enyl )-piperazin og 1,28 g (0,034 mol) natriumborhydrid ble rørt i 25 ml diglym under nitrogen hvoretter man dråpevis under kontinuerlig røring tilsatte nylig destillert bortrifluorideterat (6 ml, 0,045 mol) i 20 ml diglym. Den resulterende klare opp-løsning ble rørt ved romtemperatur i 3 timer og så fordampet for fjerning av nevnte diglym. Residuumet ble forsiktig fortynnet med 100 ml vann fulgt av 20 ml 5N saltsyre, og blandingen ble oppvarmet på et dampbad i 15 minutter. Blandingen ble avkjølt og gjort alkalisk med 4N natriumhydroksyd og ekstrahert med dietyleter. Det tørrede eterekstrakt ble fordampet og man fikk en olje som ikke lot seg utkrystallisere. Produktet ble omdannet til hydrokloridet så igjen omdannet til den frie base som i eksempel 40. Basen ble oppnådd som en lettflytelig olje som med sterk avkjøling utkrystalliserte og ga et sm.p. på 56°C.

De fraksjoner som inneholdt den forønskede forbindelse ble slått sammen, fordampet, og det krys-; tallinske faste stoff ble omkrystallisert fra 6o/80°C petroleter og man fikk den forønskede forbindelse med et sm.p. på 110°C.

(0,03 mol) beskrevet som angitt ovenfor, ble omsatt som beskrevet i eksempel 40 (b), men man brukte tionylklorid istedenfor tionylbromid. Kloretylforbindelsen ble oppnådd som en halmgul olje med vekt på 6,2 g og utkrystallisert ved henstand og hadde da et sm.p. på kO°C. Produktet ble utrørt i 5° ml dioksan sammen med 4,8 g ('0,03 mol) N-f enylpiperazin i nærvær av 3»° S vannfritt natriumkarbonat, og blandingen ble kokt under tilbakeløp i 24 timer. Blandingen ble frafiltrert natriumkarbonat, og filtratet fordampet noe som ga en blekt gul olje som ble oppløst i varm 40/60°C petroleter og kjørt gjennom en kolonne av sili-siumdioksydgel idet man brukte benzen/etylacetat 4:1 <V>/v som euleringsmiddel. Det første rasktbevegende bånd inneholdt uom-satt kloretylforbindelse (3»5 g)« De langsomt bevegelige bånd inneholdt den forønskede forbindelse som etter omkrystallisering fra etylacetat hadde et sm.p. på 124°C.

(b) 1-/5-(5-fenylfuran-2-yl)prop-l-yl7-4-fenylpiperazin-monohydroklorid
8,0 g (0,04 mol) av hydroksypropylforbindelsen ble oksydert til det tilsvarende keton, ved hjelp av den fremgangsmåte som er beskrevet av S.L. Huang et al J.O.C. kl, 3329 (1976). Ketonet ble oppnådd som en brun olje og veide k, 0 g, og dette
ble omsatt med N-fenylpiperazin og natriumcyanoborhydrid som angitt i eksempel 70, men i 3 døgn. Etter rensing ved hjelp av kolonnekromatografi ble den forønskede forbindelse oppnådd som en halmgul olje som ikke lot seg utkrystallisere, og som derfor ble omdannet til sitt monohydroklorid (etylacetat-hydro-genklorid) (l,3 g)> sm.p. 206°C.

En blanding av 7,4 g (0,05 mol) 4-klormetyl-2-metyltiazol, 12,05 g (0,05 mol) 1-(3-bromfenyl)piperazin og 10 g vannfritt natriumkarbonat ble suspendert i 150 ml absolutt etanol. Blandingen ble rørt og kokt under tilbakeløp i 8 timer. Oppløsningsmidlet ble deretter fordampet, og residuumet ble tilsatt 100 ml vann. Den dannede emulsjon ble ekstrahert méd diklormetan, og de organiske ekstrakter ble tørket over magnesiumsulfat og fordampet til en olje. Denne ble så oppløst i varm bensen, behandlet med avfargende trekull og filtrert. Filtratet ble fordampet slik at man fikk den forønskede forbindelse som en olje som fullstendig utkrystalliserte seg ved henstand, sm.p. 82°C (omkrystallisert fra eter).

(a) 2- n- propyl- 4- klormetyltiazol
En oppløsning av 30,96 g (0,3 mol) tiabutanamid i absolutt etanol (200 ml) ble langsomt tilsatt en oppløsning av 38,1 g (0,3 mol) av 1,3-dikloraceton i 100 ml etanol. Blandingen ble kokt under tilbakeløp i 1 time hvoretter oppløsnings-midlet ble fordampet. Residuumet ble behandlet med et overskudd av mettet natriurabikarbonatoppløsning og så ekstrahert med eter. Ekstraktene ble tørket og fordampet hvoretter man fikk en brun olje som i vakuum ble destillert til en blekt gul olje med. et k.p. pa 50 C/0,06 mm Hg. Denne olje ble omdannet til den forønskede forbindelse i form av et hydrokloridsalt ved å bruke hydrogenkloridholdig eter, sm.p. 60°C med dekomponering.
På lignende måte fremstilte man følgende forbindelser:

k.p. ll6-H8°C/0,09 mm Hg.
(c) 2- isopropyl- 4- klormetyltiazol- hydroklorid
sm.p. 58°C (dekomp.)

1-( 2- n- propyltiazol- 4- ylmetyl)- 4- fenylpiperazin
3,18 g (0,015 mol) av 2-n-propyl-4-klormetyltiazol-hydroklorid, 2,43 S (0,015 mol) 1-fenylpiperazin og 10 g vannfritt natriumkarbonat ble utrørt i 60 ml absolutt etanol, og blandingen ble kokt under tilbakeløp i 6 timer. Suspensjonen ble filtrert, og filtratet fordampet til en gul olje. Denne ble oppløst i kokende benzen og behandlet med avfargende trekull. Etter filtrering ble benzenen fjernet, og man fikk en gul olje som utkrystalliserte i en mengde på 4,27 g. Dette stoff ble omkrystallisert fra petroleter (k.p. 60-80°C) ved 0°C, og man fikk et hvitt krystallinsk fast stoff som smeltet ved 38-39°C.

Claims (1)
- Analogifremgangsmåte for fremstilling av terapeutisk aktive fenylpiperazinderivater med formelen:hR v2oer r R f1 eneyr l besnom zyelv, ekntluoerblt enezyr ls, ubCs1t_i,t-uaelkryt l meed lle1r -2 båsdube stR i1tuogen-ter som består av halogen, trifluormetyl, C1_4~alkyl, C. ,-alkoksy, hydroksy, nitro, amino, acetamido eller metansulfonamido; R 3 er hydrogen eller C^_^-alkyl; Q er furan, tiofen, oksazol eller tiazol; m er 1-3 og n er 0 eller 1, forutsatt at når m er 2 og n er 0, kan R ikke være metyl når Q er en tiazol-5-yl-gruppe; eller et farmasøytisk akseptabelt salt av et slikt derivat; karakterisert ved at man: (a) kondenserer en forbindelse med formelen:hvor R 1 , R 3, Q, m og n har de ovenfor angitte betydninger,og hvor L representerer en avspaltende gruppe fortrinnsvis halogen, med en forbindelse med formelen:hvor R 2 har den ovenfor angitte betydning, eller (b) reduserer en forbindelse med formelen:hvor R 1 , R 2, Q og m har de ovenfor angitte betydninger, hvorved man får dannet en forbindelse hvor n er 0; eller (c) ringslutter en forbindelse med formelen: 12 3hvor R , R , R , m og n har de ovenfor angitte betydninger, slik at man får dannet en forbindelse med formel (I) hvor Q er oksazol, eller 1 2 (d) når R eller R er fenyl substituert med amino, reduserer den tilsvarende nitro til amino som kan acetyleres eller metansulfoneres når fenylgruppen er substituert med acetamido eller metansulfonamido, eller (e) når R 1 eller R 2 er fenyl som er substituert med hydroksy, behandler den tilsvarende alkoksysubstituerte fenyl med reduserende midler,og, om ønsket, omdanner en således erholdt forbindelse til et farmasøytisk akseptabelt salt derav.
