Description
Foreliggende oppfinnelse angår fremstilling av a-sulfo-3-tienylmetylpenicillin med formelen:
samt farmasøytisk akseptable salter derav. Disse forbindelser har verdifulle antibakterielle egenskaper og kan anvendes som tera-peutiske midler for behandling av dyr og fjærkre, mer spesielt ved behandling av infeksjonssykdommer forårsaket av gram-
positive og gram-negative bakterier.
Mange penicilliner har vist seg meget effektive ved behandlinger av infeksjoner forårsaket av bakterier, men disse forbindelser har alle en eller flere av de følgende ulemper:
(1) de er ustabile i vandige syrer,
(2) de mangler evne til en effektiv bekjempelse av
Pseudomonas infeksjoner,
(3) de er alle ineffektive overfor såkalte penicillin G-resistente bakteriestammer (f.eks. mange stammer av Staphylo-
coccus aureus)
(4) de er ustabile overfor penicillinase fremstilt av forskjellige mikroorganismer.
Forbindelsen som fremstilles ifølge oppfinnelsen og dens
salter har i tillegg til sin meget sterke aktivitet overfor gram-positive bakterier, også meget høy resistens overfor de-komponering av syrer eller penicillinase, og spesielt høy
aktivitet overfor gram-negative mikroorganismer, f.eks. av slekten Pseudomonas.
Ifølge foreliggende oppfinnelse fremstilles forbindelsen med formel I og dens salter ved at 6-aminopenicillansyre eller et silylert derivat derav omsettes med et acyleringsmiddel avledet fra en karboksylsyre med formelen:
hvoretter, dersom nevnte silylerte derivat er benyttet, silylgruppen fjernes fra det acylerte produkt for tilveiebringelse av penicillinforbindelsen, og, om ønsket, omdannelse av et således erholdt produkt til et farmasøytisk akseptabelt salt derav.
Nevnte acyleringsmidler innbefatter syrehalogenider (f.eks. klorid, bromid, etc.) tilsvarende karboksylsyren med formel II, det tilsvarende karboksylsyreanhydrid, spesielt blandede anhydrider fremstilt fra karboksylsyren med formel II og en sterkere syre, f.eks. en lavere-alkyl- eller aralkyl-mono-ester av karbonsyre (f.eks. benzyloksykarbonsyre, etoksykarbon-syre, etc), en eddiksyre substituert med en eller flere elektron-tiltrekkende grupper (f.eks. dikloreddiksyre, trikloreddiksyre, etc), eller en alkan- eller aren-sulfonsyre (f.eks. toluen-sulfonsyre, metansulfonsyre, etc). I tillegg til dette kan nevnte acyleringsmidler også innbefatte det tilsvarende syreazid eller den tilsvarende aktive ester eller tioester, f.eks. en ester fremstilt fra karboksylsyren med en fenol eller en tiofenol (f.eks. p-nitrofenol, 2,4-dinitrofenol, pentaklorfenol, tiofenol). Videre kan acyleringsmidlet innbefatte karboksylsyren koblet med N,N-dimetylklorforminiumklorid, et N,N-karbonyldi-triazol, en karbodiimido-reagens (f.eks. N,N'-dicykloheksyl-karbodiimid, N,N'-diisopropylkarbodiimid eller N-cykloheksyl-N<1->
(2-morfolinoetyl)karbodiimid), en keteniminreagens eller en isoksa-zolium-saltreagens. Et annet acyleringsmiddel avledet fra karboksylsyren med formel II, er det tilsvarende azolid, dvs.
det tilsvarende syreamid hvor amidnitrogenet inngår i en aro-matisk femleddet ring inneholdende minst to nitrogenatomer (f.eks. imidazol, pyrazol, benzimidazol, etc). Blant disse acylerings-
midler er det spesielt foretrukket å anvende syrehalogenider,
og da spesielt syreklorider.
Acyleringsmidlet fremstilles ved kjente metoder. Det tilsvarende syrehalogenid kan lett fremstilles ved først å omsette tiofeneddiksyre med svoveltrioksyd eller dets dioksan-kompleks i dikloretan ved 10-60°C, og deretter behandle den fremstilte karboksylsyre II med et halogeneringsmiddel (f.eks. tionylklorid, fosforoksyklorid, fosforoksybromid, etc.) ved 0-100°C, mer foretrukket ved 10-40°C, i nærvær av en katalysator (f.eks. dimetylformamid, etc).
Det skal bemerkes at 6-aminopenicillansyren kan enten være 6-aminopenicillansyre selv eller et salt av denne. Vanligvis er det foretrukket at syren anvendes i form av et salt, f.eks. natriumsalt, kaliumsalt, trietylaminsalt, etc.
Som kjent omfatter silylerte derivater av 6-aminopenicillansyre mono-silylderivater og di-silylderivater, og begge to kan anvendes i foreliggende oppfinnelse, skjønt sistnevnte vanligvis vil gi bedre resultater enn førstnevnte forbindelse. Foretrukne eksempler på silylgruppen i de silylerte derivater
er følgende:
hvor R^, R_ og R, er en lavere-alkylgruppe med 1-5 karbonatomer, benzyl, fenetyl, cykloheksyl, fenyl eller tolyl.
Acyleringen i foreliggende fremgangsmåte utføres fortrinnsvis ved å omsette 6-aminopenicillansyre med acyleringsmidlet i et egnet oppløsningsmiddel i nærvær av en egnet base.
Acyleringen kan utføres ved romtemperatur, under romtemperatur eller endog over romtemperatur, men ettersom 6-aminopenicillansyre og dets acyleringsprodukter har en tendens til å bli dekomponert ved forhøyede temperaturer, er det imidlertid foretrukket at reaksjonen utføres ved temperaturer under 50°C, fortrinnsvis ved temperaturer fra -20 til +40°C så langt reaksjonsblandingen beholder flytende form.
Egnede baser er organiske baser (f.eks. trietylamin, tributylamin, pyridin, piperidin, morfolin, etc), samt uorganiske baser (f.eks. natriumbikarbonat, kaliumbikarbonat, natrium-hydroksyd, kaliumhydroksyd, natriumkarbonat, kaliumkarbonat, etc.). Mengden av basen er vanligvis 1-3 mol pr. mol av nevnte acyleringsmiddel. Ethvert oppløsningsmiddel som ikke forstyrrer reaksjonen kan anvendes for acyleringsreaksjonen, og som sådan kan man anvende vann, dioksan, aceton, dimetylformamid, tetrahydrofuran, kloroform, etylendiklorid, toluen, benzen eller blandinger av disse. Når man imidlertid anvender silylerte derivater, er det foretrukket å anvende et inert oppløsnings-middel, f.eks. eter, dioksan, benzen, tetrahydrofuran eller kloroform.
Det skal bemerkes at hydrolyse eller alkoholyse av et acyleringsprodukt fremstilt ved reaksjonen mellom et acyleringsmiddel og et silylert derivat av 6-aminopenicillansyre, lett kan utføres ved å behandle acyleringsproduktet med vann eller en lavere-alifatisk alkohol (f.eks. metanol, etanol, etc.) hvorved man oppnår forbindelsen med formel I.
Etter reaksjonen kan den ønskede forbindelse innvinnes fra reaksjonsblandingen ved hjelp av i og for seg kjente metoder. Hvis det er ønskelig, kan de fremstilte produkter renses på
kjent måte, f.eks. ved omkrystallisasjon eller kromatografi.
Det skal bemerkes at penicillinforbindelsen med formel I har to syrefunksjonelle grupper, dvs. karboksyl- og sulfogruppene. På grunn av forskjellen med hensyn til relativ surhetsgrad på disse syregrupper, er det mulig å fremstille enten et surt salt eller et normalt salt.
ct-karbonatomet i acylgruppen i forbindelsen med formel I vil utgjøre et asymmetrisk karbonatom, og det vil da eksistere to optiske isomere, og slike individuelle isomere former såvel som blandinger derav, omfattes av oppfinnelsen. Når acylerings-produktene oppnås som en blanding av isomere, så kan disse, hvis det er ønskelig, spaltes i de respektive isomere ved kromatograf-eringsteknikk eller ved omkrystallisasjon, hvor man anvender kjent teknikk. Alternativt kan optisk aktive penicilliner fremstilles ved å anvende et optisk aktivt acyleringsmiddel fremstilt fra en spaltet karboksylsyre med formel II. Den optiske spalting av karboksylsyren utføres ved at syren underkastes en spalting
på kjent måte, f.eks., ved å utkrystallisere et optisk, aktivt basesalt (f.eks. brucin, kinin) av karboksylsyren.
Den fremstilte forbindelse med formel I har sterkere inhiberende aktivitet mot Pseudomonas-mikroorganismer enn ct-sulfobenzylpenicillin. Forbindelsen med formel I har således uventede og utmerkede egenskaper overfor tidligere kjente beslektede forbindelser. For å illustrere dette har man foretatt forsøk hvor følgende data ble oppnådd:
Infektiøs test-mikroorganisme:
Pseudomonas aeruginosa IFO 3448.
Testede forbindelser:
Forbindelse A: a-sulfo-3-tienylmetylpenicillin-dinatriumsalt Forbindelse B: a-sulfobenzylpenicillin-dinatriumsalt
Hver forbindelse A og B forelå i sin DL-form.
Test metode: Agar-fortynningsmetoden
Resultater:
Fra de ovenfor angitte resultater vil det fremgå at forbindelse A som fremstilles ifølge foreliggende oppfinnelse har bedre antibakteriell aktivitet enn a-sulfobenzylpenicillin.
Forbindelsen med formel I har lav toksisitet overfor pattedyr og tolereres meget god, selv i store doser. De kan til-føres oralt i kapsler eller tabletter eller parenteralt i opp-løsninger eller suspensjoner. Ved behandling av Pseudomonas-infeksjoner i pattedyr, såsom mus, mennesker eller lignende,
kan penicillinene tilføres ved vanlige metoder i mengder varierende fra 5 til 50 mg/kg/døgn, mer vanlig fra 10 til 30 mg/kg/døgn, i oppdelte doser, f.eks. to-fire ganger pr. døgn.
Følgende eksempel illustrerer oppfinnelsen. Det in-
KBr — 1 frarøde absorbsjonsspektrum (IR) er uttrykt som v , cm ,
og det kjernemagnetiske resonansspektrum (NMR) er uttrykt som 6 (i D20, 60MC).
Eksempel
Fremstilling av dinatriumsaltet av a-sulfo-3-tienylmetyl-penicillin.
Til en suspensjon av 1,08 g 6-aminopenicillansyre i 8 ml vann tilsettes 1,43 g natriumbikarbonat. Etter at blandingen er oppløst, tilsettes gradvis en oppløsning av 1,30 g a-sulfo-3-tienylacetylklorid i 10 ml dietyleter. Blandingen omrøres ved en temperatur på omkring 0°C i 1 time. Det vandige lag vaskes to ganger med 10 ml porsjoner eter og innstilles til pH 1,2 med en kationutvekslerharpiks av polystyren-sulfonsyretypen under konstant avkjøling. Deretter vaskes oppløsningen to ganger med 15 ml porsjoner etylacetat, fulgt av ekstraksjon to ganger med 15 ml porsjoner n-butanol. Ekstraktene kombineres og vaskes to ganger med 15 ml porsjoner vann og deretter ekstraheres de med en vandig oppløsning av natriumbikarbonat. Ekstraktet innstilles til pH 6,7, vaskes med eter og lyofiliseres for dannelse av det ovenfor angitte sluttprodukt hvilket gir 1,0 g som et fargeløst pulver. Dette produkt viser følgende data etter om-krystallisering fra vann/aceton:

1045 (-S03Na)
NMR: 1,52, 1,57, 1,65 (6H, 2-(CHj)2), 4,27, 4,29 (1H, 3-H)
5,29 (1H, 10-H), 5,53-5,62 (2H, 5,6-H), 7,24-7,67

- Analogifremgangsmåte til fremstilling av terapeutisk aktivt a-sulfo-3-tienylmetylpenicillin med formelen:samt farmasøytisk akseptable salter derav, karakterisert ved at 6-aminopenicillansyre,et salt eller silylert derivat derav omsettes med et acyleringsmiddel avledet fra en karboksylsyre med formelen: hvoretter, dersom nevnte silylerte derivat er benyttet, silylgruppen fjernes fra det acylerte produkt for tilveiebringelse av penicillinforbindelsen, og, om ønsket, omdannelse av et således erholdt produkt til et farmasøytisk akseptabelt salt derav.
