Pteridineverbindingen, werkwijze ter bereiding daarvan alsmede farmaceutische preparaten, die deze verbindingen bevatten.
Gebied van de uitvinding
De uitvinding heeft betrekking op pteridineverbindingen, die van de basisstructuur van het triamtereen (2,4,7-triamino-6-fenylpterldine) zijn afgeleid en die een diuretische, antikaliuretische, calcium-anta-5 gonistische en cardioprotectieve werking bezitten.
Stand der techniek
Het Britse octrooischrift 1.597.881 heeft betrekking op derivaten van het triamtereen, waarbij de para-plaats van 6-fenylrlng met een hydrofiele groep veretherd is. Het daarbij geopenbaarde concept staat in 10 tegenstelling tot het werk van Weinstock et al. (J.Med.Chem. 11, 573 (1968). Volgens dezen wordt de diuretische werkzaamheid op zijn hoogst door lipofiele substitutie bij triamtereenderivaten bevorderd, bij hydrofiel gesubstitueerde derivaten zou geen diuretische activiteit te verwachten zijn. In het Britse octrooischrift is een reeks verbindingen 15 aangegeven, die het geopenbaarde concept bewijzen. In het profiel van een werkzame stof, zoals deze aan het Britse octrooischrift ontleend kan worden, wordt intussen op talrijke parameters ingegaan, zoals bijvoorbeeld werkzaamheid, acute en chronische toxiciteit, verenigbaarheid met andere werkzame stoffen, farmacokinetiek en -dynamiek, metabolisme, 20 fysische en chemische parameters zoals stabiliteit, oplosbaarheid, toegankelijkheid e.a.
Onder de algemene formule van het Britse octrooischrift 1.597.881 vallen ook derivaten met basisch gesubstitueerde groepen en dergelijke verbindingen zijn ook beschreven. Bij de latere wetenschappelijke ver-25 dere bewerking van deze verbindingsklasse werden echter de hydroxy-triamtereenethers met alcoholische of zure groepen als bijzonder veelbelovend beschouwd. (Vergelijk H. Knauf et al., Therapiewoche 30, 6831-6847 (1980). Daar wordt ook o.a. vastgesteld: "Uit de genoemde resultaten kon de slotconclusie getrokken worden, 30 dat het beoogde doel, een werkzaam, in water oplosbaar triamtereenderi-vaat met gunstige farmacokinetische eigenschappen te verkrijgen, bereikbaar is, doordat men onder het aanhouden van het basisskelet van het triamtereen een geschikte zijketen met een carboxylgroep invoert, die aan het molecuul amfifiele eigenschappen verleent” en Ner kan der-35 halve vastgesteld worden, dat het gelukt is, tot de gewenste, snel werkzame, in water oplosbare triamtereenderivaten te geraken".

De hydroxytriamtereenethers met zure groepen, in het bijzonder het 2,4,7-triamino-6-[4-(4-carboxybutoxy)fenyl]-pteridine (in het volgende met Val-O-TA afgekort) laten bij intraveneuse toedoening water-diureti-5 sche, natriuretische en antikaliuretische werking zien. De door furose-mide bewerkstelligde kaliurese kan bijvoorbeeld met Val-O-TA nagenoeg volledig gecompenseerd worden.
De zuurderivaten laten echter bij perorale toediening bij ratten in hoeveelheden van ongeveer 50 mg/kg lichaamsgewicht geen hoopvolle 10 diuretische werking zien. De oplosbaarheden van de zuurderivaten met rechte keten liggen in het pH traject van 11-12 bij ongeveer 0,1%.
Er bestond derhalve de opgave een werkzame stof te vinden, die vanwege zijn eigenschappen zo veel mogelijk geen werking van de toepassingsmogelijkheden met zich mee mag brengen en waarvan de farmacologi-15 sche eigenschappen ten minste zo goed dienen te zijn als die van het triamtereen. Het zou ook te wensen zijn, dat de metabolisering geen nieuwe problemen zou opwerpen. In het bijzonder zou een verhoudingsgewijze goede in water oplosbare werkzame stof te verlangen zijn, die zonder problemen voor intraveneuse toediening geschikt is. Een derge-20 lijke werkzame stof zou zich als een partner voor combinatietherapie met het snel werkzame diureticum furosemide aanbieden, voorzover de farmacokinetische en farmacodynamische parameters daarvoor spreken. Oplossing
Gevonden werd, dat in de zin van de gestelde opgave pfteridinever-25 bindingen met de algemene formule 1, waarin R]_ en 1¾ onafhankelijk van elkaar waterstof of een eventueel vertakte alkylgroep met 1 tot 6 koolstofatomen of een benzylrest voorstellen, of waarin R]_ en R2 tezamen met het stikstofatoom en eventueel het erbij betrekken van een of twee andere heteroatomen een heterocyclisch 30 vijf- of zes-ringsysteem vormen, of waarin R^ en R2 tezamen met het stikstofatoom een rest

Onder heteroatomen in 40 de zin van de onderhavige definitie voor -NRiR2 als heterocyclisch 8500564 « * 3 ringsysteem kunnen stikstof en zuurstof en In de tweede pleats ook zwavel genoemd worden.
De verbindingen met de formule 1 bezitten een chlraal centrum. De onderhavige conclusies omvatten alle met betrekking tot de chiraliteit 5 onderscheidbare vormen.
Als fysiologisch aanvaardbare zuur-additiezouten in de zin van de onderhavige uitvinding komen die met anorganische zuurresten, bijvoorbeeld de chloriden, bromiden, sulfaten, carbonaten, fosfaten, of met organische carbon- resp. sulfonzuurresten, bijvoorbeeld zouten van 10 azijnzuur, propionzuur, oxaalzuur, malonzuur, barnsteenzuur, fumaar-zuur, maleïnezuur, melkzuur,· wijnsteenzuur, appelzuur, citroenzuur, benzo'ézuur, methaan- of ethaansulfonzuur, ethaandisulfonzuur in aanmerking; onder de zouten verdienen de tartraten, in het bijzonder de waters toftartraten, de voorkeur.
15 De bereiding van de verbinding met de formule 1 en de daarvan af geleide zuur-additiezouten kan op op zichzelf bekende wijze plaats hebben, bijvoorbeeld steunend op de werkwijze van R.6.W. Spickett en G.H.
Timmis (J.Chem.Soc. 2887-95 (1954) resp. volgens het Britse octrooi-schrift 1.397.881.
20 Daarbij wordt voordelig het gesubstitueerde benzylcyanide met de algemene formule 2, waarin en R2 dezelfde betekenis hebben als bij de algemene formule 1 onder katalyse met basen met 2,4,6-triamino- 5-nitrosopyrimidine tot de verbinding met de formule 1 omgezet. Uitvoering van de reactie kan in een geschikt reservoir, dat van koeler en * 25 roerder is voorzien, met voorkeur met een alkalimetaalalkanolaat als base en de overeenkomstige alcohol als oplosmiddel onder uitsluiting van vocht plaats hebben. Als alcohol is bijvoorbeeld een alkoxyalkanol zoals methoxyethanol geschikt gebleken. Doelmatig wordt in de apparatuur onder een droog beschermend gas uit alcohol en alkalimetaal het 30 alkoxide bereid. Men past het 2,4,6-triamino-5-nitrosopyrimidine en de verbinding met de formule 2, bij voorkeur in geringe overmaat toe en spoelt eventueel met de alcohol na. Men laat onder roeren en bij voorkeur bij verhoogde temperatuur, bijvoorbeeld onder terugvloeikoeling gedurende een bepaalde tijdsperiode, bijvoorbeeld 2 uren reageren en 35 laat vervolgens afkoelen, bijvoorbeeld gedurende een nacht. Het gevormde neerslag wordt afgefiltreerd, met een geschikt oplosmiddel zoals bijvoorbeeld aceton gewassen en doelmatig enige tijd in een droogkast bij voorkeur onder verhitten en onder verminderde druk gedroogd. De verbindingen kunnen eventueel verder gezuiverd, bijvoorbeeld herkris-40 talliseerd worden, bijvoorbeeld uit IN zoutzuur, waarbij dihydrochlori- 8500564

4 den verkregen worden.
De omzetting van de verbindingen met de formule 1 in de overeenkomstige zuur-additiezouten kan in het algemeen op op zichzelf bekende wijze door omzetting met het geschikte zuur uitgevoerd worden.
5 De bereiding van de gesubstitueerde benzylcyaniden met de algemene formule 2 heeft met voordeel door additie van aminen met de algemene formule 3, waarin R^ en R£ dezelfde betekenissen bezitten als bij formule 1» aan de epoxygroep van het 3-(4-cyaanmethylfenoxy)propyleen-oxide plaats.
10 Voordelig voegt men de aminen met de algemene formule 3 in over maat (richtwaarde ongeveer vijfvoudig voor secundaire, tienvoudig voor primaire aminen) toe aan een oplossing van de epoxyverbinding in een alcohol. De aminen kunnen ten dele ook in de vorm van een waterbevat-tende, bijvoorbeeld 40% fs oplossing toegepast worden. Bij secundaire 15 aminen met de formule 3 leidt eerder de toepassing in watervrij milieu, bijvoorbeeld in een alcohol, zoals bijvoorbeeld methanol, tot het doel. Bij aminen met bepaalde sterische hindering, zoals bijvoorbeeld het di-isopropylamine, verdient verwarming, bijvoorbeeld koken onder terug-vloeikoeling, aanbeveling.
20 De opbrengsten liggen in het algemeen bij ten minste 50 en ten hoogste 95%.
Het 3“(4“cyaanmethylfenoxy)propyleenoxide kan bijvoorbeeld volgens een op zichzelf bekende reactie door omzetting van 4-hydroxybenzylcya-nide met eplbroom- of epichloorhydrine bereid worden.

Voordelige werkingen van de uitvinding
De verbindingen met de formule 1 en de daarvan afgeleide zuur-25 additiezouten zijn geelachtige, kristallijne verbindingen, die in het UV-licht de van de pteridineverbindingen bekende sterke fluorescentie laten zien.
Zij zijn in water verrassend goed oplosbaar, in het bijzonder bij pH-waarden < 7. Goed oplosbaar zijn ook de zuur-addltiezouten van 30 de verbindingen met de formule 1.
De verbindingen met de formule 1 en de daarvan afgeleide zuur-additiezouten met fysiologisch aanvaardbare zuren zijn voor perorale, alsmede voor intraveneuse toediening zeer goed geschikt. De diurese-proeven werden volgens de volgende methodiek uitgevoerd: 35 Diureseproeven/methodiek:
Voor de proeven werden mannelijke Wistar-ratten met een lichaamsgewicht van ongeveer 130 g toegepast, waaraan 18 uren het voer was onthouden. Zij ontvingen onmiddellijk voor de intraveneuse toediening van de proefstof oraal een hoeveelheid van 20 ml/kg van een 0,9Z's 40 NaCl-oplossing. De intraveneuse toepassing bedroeg 0,001 tot 300 ymol 8500564 » * 6 werkzame stof in 4 ml/kg 0,9%’s NaCl-oplossing (pH 3). Toegepast werd onder lichte narcose met ether in de staartader. In de regel werd per proef zes proefdieren gebruikt. De perorale toediening heeft plaats door middel van een keelgatsonde in het gastro-intestinale gebied. De 5 dieren werden afzonderlijk in diuresekooien gebracht en de urine werd na 2,5 h opgevangen.
De bepaling van de elektrolyten (Na+/K*, Mg+^) had vlam-fotometrisch en door atoomabsorptiemeting met de elektrolyt-autornaat FL6 van de fa. Zeiss/Oberkochen plaats.
10 Dosis-reactiekrommen werden met behulp van de niet lineaire regressie-analyse door middel van het NONLIN-computerprogramma volgens C. Daniel en F.S* Wood in "Fitting Equations tot Data", J. Wiley &
Sons, New York, 1980, vastgesteld.
De bij de bepaling van de diuretische werkzaamheid toegepaste 15 grootheid ED50 (werkingssterkte = potentie) wordt gedefinieerd als de hoeveelheid werkzame stof per kg LG (LG * lichaamsgewicht) die voor de halfmaximale werking vereist is.
De volgende tabel licht de betere werkzaamheid toe van de verbindingen met de formule 1 (=2,4,7-triamino-6-[4-(2-hydroxy-3-dimethyl-20 aminopropoxy)-fenyl]pteridine; in het volgende als verbinding met de formule IA aangeduid, in vergelijking met triamtereen en het 2,4,7-tri-amino-6-[4-(3-dimethylaminopropoxy)-fenyl] pteridine [-=DES-0H] ED50 [pmol/kg lichaamsgewicht] 25 werkzame stof ^ Mg® triamtereen 4,40 1,87 - DES-OH 5,53 0,54 30 verbinding met de formule IA 9,36 0,24 0,40
Resultaten:
De verbinding met de formule 1 laat een uitstekende diuretische 35 werkzaamheid zien, zowel met betrekking tot de natriuretische alsook kalium- en magnesium-sparende eigenschappen.
Zij is daarin bijvoorbeeld duidelijk beter dan het triamtereen en de desoxyverbinding. Het kalium-sparende effect begint bij een tiende van de dosering, waarbij de werkzaamheid van triamtereen begint.

De nadruk verdienen de gunstige toxiciteitswaarden bijvoorbeeld bij intraveneuse toediening. Voorts is de goede eliminering bij intra-veneuse toediening van praktische betekenis. Bij de volgende toepassing van het begrip "werkzame stof met de formule 1 resp. IA enz.” dienen de 20 verbindingen met de algemene formule 1 en de daarvan afgeleide zuur-additiezouten verstaan te worden.
Volgens tot dusverre aanwezige onderzoekingen treedt geen metabo-lisering van de werkzame stoffen met de formule 1 op.
De hoge werkingssterkte veroorlooft een relatief geringe dosering 25 van een werkzame stof volgens de uitvinding met de formule 1, waarbij vanzelfsprekend het gewicht, de leeftijd, de constitutie en de algemene gezondheidstoestand van de patiënt in aanmerking genomen moeten worden.
In het algemeen is een dagelijkse dosis van 0,2 tot 1000 mg, bij voorkeur 1,5 tot 20 mg geschikt. De toediening kan in verschillende toedie-30 ningseenheden plaats hebben, bijvoorbeeld voor elke 2 mg vier maal daags. De toediening kan oraal of parenteraal plaats hebben.
De werkzame stoffen met de formule 1 zijn uitstekend geschikt als farmaceutica met diuretische, antikaliuretische, antimagnesiuretische, antihypertensieve en cardioprotectieve werking.
35 Zij zijn ook bijzonder voor combinatie met andere werkzame stoffen met vergelijkbare indicatie geschikt, voorzover zij daarmee verenigbaar zijn. Van aanzienlijke betekenis is de combinatie met snelwerkende diuretics zoals het furosemide (4-chloor-N-furfuryl-5-sulfamoylantranil-zuur volgens het Amerikaanse octrooischrift 3.058.882).
40 De nadruk verdient, dat de kaliuretische werking van furosemide 8500564 » 8 met ongeveer een tiende van de dosis van een werkzame stof met de formule 1 gecompenseerd kan worden. In het algemeen staan de toe te dienen hoeveelheden in de verhouding van 0,25 tot 100 gew.delen furosemide tot 1 gew.deel werkzame stof met de formule 1. Bij een aanbevolen dagelijk-5 se dosis van 2 x 40 mg peroraal furosemide verdient de toediening van 8 mg werkzame stof met de formule 1 aanbeveling. Een intraveneuse toediening van 20 mg furosemide komt overeen met de intraveneuse toediening van 2 mg van de werkzame stof met de formule 1.
Van bijzonder belang is ook de combinatie met calcium-antagonis-10 tisch werkzame stoffen, zoals zij ook in het Duitse Offenlegungsschrift 2.658.500 toegepast worden, in het bijzonder het Verapamil, Gallopamil, Nifedipin, Diltiazem. Daarbij staan in de combinatiepreparaten de cal-ciumantagonisten tot de werkzame stof met de formule 1 in een gew.verhouding 100:1 tot 0,1:1.
15 Van speciaal belang is ook de combinatie van de kalium-sparende werkzame stoffen met de formule 1 met andere diuretica. (Vergelijk Ullmanns Encyklopëdie der technischen Chemie, 4de druk, deel 10, blz. 181-186, Verlag Chemie, 1975).
Genoemd kunnen worden saluretica, in het bijzonder de benzothia-20 diazinederivaten, zoals het Chlorothiazid, het Hydrochlorothiazid en de Hydrochlorothiazid-analogen, in het bijzonder het Hydroflumethiazid, Thiabutazid, Bendroflumethiazid, Trichloromethiazid, Methylcyclothia-zid, Polythiazid, Cyclothiazid, Cyclopenthiazid, Ethiazid, Benzthiazid, Methylbenzylhydrochlorothiazid, voorts de sulfamoylsaluretica zoals het 25 Chlorthalidon, Mefrusid, Clopamid, Quinethazon, Chlorexolon.
Daarbij staan de werkzame stoffen met de formule 1, bijvoorbeeld de werkzame stof met de formule IA, tot de voor het afzonderlijke diure-ticum aanbevolen dosering in de gew.verhouding 2:1 tot 0,01:1.
In het bijzonder wordt naar de combinatie Hydrochlorothiazid in de 30 gew.verhouding werkzame stof met de formule IA tot het salureticum van 1:1 tot 0,05:1 verwezen.
Voorts is de werkzame stof met de formule 1 geschikt voor combinatie met 0-blokkers analoog de in het Britse octrooischrift 1.584.089 geleerde combinatiepreparaten met triamtereen, in het bijzonder voor 35 combinatie met Propranolol resp. de zuur-additiezouten daarvan, waarbij nog saluretica in de combinatie aanwezig kunnen zijn.
Daarbij staan de werkzame stoffen met de formule tot de p-blokkers in de gew.verhouding 2:D tot 10:D, waarbij D de aanbevolen dagelijkse dosis tot minus 50% van de aanbevolen dagelijkse dosis voorstelt.
40 De farmaceutische preparaten, die de nieuwe werkzame stof met de 8500564 9 formule 1 bevatten, kunnen op de gebruikelijke wijze bereid worden en zij kunnen de gebruikelijke dragers en hulpstoffen bevatten. Een uitvoeringsvorm van de uitvinding zijn vaste, voor orale toediening geschikte preparaten, zoals bijvoorbeeld tabletten, capsules, dragee's 5 enz. Voor de orale toepassing kunnen als dragers farmaceutisch indifferentie vaste stoffen, zoals bijvoorbeeld mannitol, melksuiker, organische en anorganische calciumzouten enz. toegepast worden. Als bindmiddel zijn o.a. polyvinylpyrrolidon, gelatine of cellulosederivaten geschikt. Als andere toevoegsels kunnen middelen voor het doen uiteen-10 vallen van tabletten, zoals bijvoorbeeld zetmeel of alginezuur, glijmiddelen, zoals bijvoorbeeld stearinezuur of zouten daarvan en anorganische stromingsmiddelen, zoals bijvoorbeeld talk of colloidaal kiezel-zuur, alsmede smaakcorrigentia enz. toegepast worden.
De werkzame stoffen kunnen met de hulpstoffen op gebruikelijke 15 wijze gemengd en nat of droog gegranuleerd worden. Al naar gelang de aard van de toegepaste toevoegsels kan eventueel ook door eenvoudig mengen een directe tabletteerbaarpoeder verkregen worden. Het granulaat of poeder kan direct in capsules worden afgevuld of op gebruikelijke wijze tot tabletkernen worden geperst.
20 Bij parenterale toediening kunnen de therapeutische middelen even tueel op de gebruikelijke wijze bereid en toegediend worden.
De volgende voorbeelden dienen ter toelichting van de bereiding van de verbindingen met de algemene formule 1 en de bereiding van de farmaceutische preparaten.
25 Bereiding van de verbindingen met de algemene formule 1
Als analytische methode wordt o.a. de dunnelaag-chromatografie op gerede DC-platen ("Merck", Kieselgel 60, F 254, artikel nr 5715) in verschillende elueermiddelen toegepast.

Voorbeeld I
Bereiding van 2,4,7-triamlno-6-[4-(3-dimethylamlno-2-hydroxypropoxy)-40 fenylj-pteridine («verbinding met de formule IA) 8500564 10 uitgangsmengsel: 45,0 g (192 mmol) 1-(4-cyaanmethylfenoxy)-3- dimethylaminopropanol-2 27,1 g (176 mmol) 2,4,6-triamine-5-nitrosopyrimidine 5 (TNP) 4,1 g (176 mmol) natrium 1125 ml 2-ethoxyethanol.
In een driehalsrondbodemkolf van 2 1 met magneetroerder, koeler, 10 droogbuis (met CaCl2~vulling) en thermometer werd uitgegaan van 925 ml 2-ethoxyethanol en de apparatuur werd 10 min. met argon gespoeld.
4,1 g Natrium werden aan de ethoxyethanol toegevoegd. Nadat alle natrium daarin was opgelost werden 27,1 g 2,4,6-triamino-5-nitrisopyrimi-dine en 45,0 g l-(4-cyaanmethylfenoxy)-3-dimethylaminopropanol-2 in de 15 reactiekolf gebracht en stofresten werden met 200 mi 2-ethoxyethanol weggespoeld. Het reaetiemengsel werd 2 uren onder terugvloeikoeling gekookt en een nacht op kamertemperatuur afgekoeld. Het gevormde neerslag werd over een D-4-frit afgezogen, met in totaal 1 1 aceton gewassen en 14 uren bij 60°C in een vacuumdroogkast (verminderde druk opgewekt met 20 een oliepomp) gedroogd.
Opbrengst ; 34,5 g (53,0% van de theorie).
Rf-waarde (elueermiddel 1) * 0,40.
De bereiding van de andere verbindingen met de algemene formule 1 kan analoog voorbeeld I uitgevoerd worden. Zij worden in tabel A opge-25 somd.
Voorbeeld II
Zuivering van het 2,4,7-triamino-6-[4-(3-dimethylamino-2-hydroxyw propoxy)fenyl]-pteridine («verbinding met de formule IA) en bereiding van het dihydrochloride-monohydraat 30 In een rondbodemkolf van 1 1 met koeler, schermverhittingskap en magneetroerder werden 15,0 g 2,4,7-triamino-6-[4-(3-dimethylamino-2-hy-droxypropoxy)fenyl]-pteridine met 675 ml IN zoutzuur tot koken verhit. Aan de bruihachtige, doorzichtige oplossing werden 5,0 g A-kool (actieve kool) toegevoegd en het mengsel werd 15 min. in kookhitte geroerd.
35 De actieve kool werd afgefiltreerd (vouwfilter), de oplossing werd opnieuw tot koken verhit en over een membraan gefiltreerd (0,2 pm, voorfilter van glasvezel). De oplossing werd een nacht onder uitsluiting van lucht tot kamertemperatuur afgekoeld. Daarbij werd een lichtgeel neerslag verkregen. Het neerslag werd afgezogen, tweemaal met elk 20 ml 40 water, driemaal met elk 50 ml aceton en tweemaal met elk 50 ml diethyl- 8500564 11 ether gewassen, 10 uren bij 60°G en 5 uren bij 105°C in een vacuum-droogkast (verminderde druk opgewekt met de oliepomp) gedroogd. Opbrengst: 11,95 g (64,0% van de theorie, berekend als dihydrochloride-monohydraat).

Bereiding van l-(4~cyaanmethylfenoxy)-3-dlmethylamlnopropaaol~2 uitgangsmengsel: 10,0 g (52,9 mmol) 3-(4-cyaanmethylfenoxy) 5 propyleenoxide (CPO) 29,8 g (264,5 mmol) 40%’s oplossing van dimethylamine in water 100 ml methanol.
10 10,0 g 3-*(4-cyaanmethylfenoxy)propyleenoxide werden in een drie- halsrondbodemkolf van 250 ml met druppeltrechter, thermometer en mag-neetroerder in 100 ml methanol opgelost. 29,8 g van een 40%'s oplossing van dimethylamine werden aan de oplossing langzaam (in verloop van 20 min.) druppelsgewijze toegevoegd. Het mengsel werd een nacht bij kamer-15 temperatuur geroerd. De methanol en de overmaat dimethylamine werden op een roterende verdamper verwijderd en de achtergebleven olie werd in 75 ml IN zoutzuur opgenomen. De oplossing werd driemaal met elk 50 ml di-chloormethaan geëxtraheerd. De doorzichtige, waterbevattende fase werd afgescheiden en 20 ml 4N natriumhydroxide werden toegevoegd. Daarbij 20 werd de waterbevattende fase troebel en er werd een olie afgescheiden. Ter betere afscheiding van de olie werden 30 g keukenzout toegevoegd. Het alkalische mengsel werd eenmaal met 100 ml en tweemaal met 50 ml dichloormethaan uitgeschud. De organische fasen werden verenigd en boven natriumsulfaat gedroogd. Nadat het natriumsulfaat was afgefil-25 treerd, werd het dichloormethaan op een roterende verdamper verwijderd. Opbrengst: 3,0 g (70,5Z van de theorie).
Andere gesubstitueerde benzylcyaniden met de formule 2 kunnen analoog bereid worden (zie tabel B).
Voorbeeld XVII
30 Bereiding van l-(4-cyaanmethylfenoxy)-3-diisopropylaminopropanol-2 uitgangsmengsel: 30,0 g (158,6 mmol) 3-(4-cyaanmethylfenoxy)- propyleenoxide 210,6 ml (1,58 mmol) diisopropylamine.

In een driehalsrondbodemkolf van 500 ml met koeler, droogbuis (CaCl2"vulling) en thermometer werden 30,0 g 3-(4-cyaanmethylfenoxy)-propyleenoxiden met 210,6 ml diisopropylamine 24 uren onder terugvloei-koeling gekookt. Het bij de reactie gevormde, witte neerslag werd, na-40 dat het reactiemengsel tot kamertemperatuur was afgekoeld, afgezogen en 8500564 16 verwijderd. Het doorzichtige filtraat werd op een roterende verdamper geconcentreerd. Het residu werd in 90 ml zoutzuur (32%'s) opgelost. De oplossing werd vijfmaal met elk 100 ml dichloormethaan geëxtraheerd. De organische fase werd verwijderd. Aan de zoutzure, water bevattende fase 5 werden langzaam ongeveer 80 ml 40%’s natriumhydroxide toegevoegd en er werd driemaal met elk 100 ml dichloormethaan uitgeschud. De verenigde organische fasen werden boven natriumsulfaat gedroogd. Het droogmiddel werd afgefiltreerd en het dichloormethaan werd op een roterende verdamper verwijderd.

Voorbeeld XVIII
Bereiding van l-(4-cyaanmethylfenoxy)-3-dibutylaminopropanol-2 uitgangsmengsel: 30,0 g (158,6 mmol) 3-(4-cyaanmethylfenoxy)- 15 propyleenoxide 133,5 ml ( 793 mmol) dibutylamine 300 ml methanol
Aan 30 g 3-(4-cyaanmethylfenoxy)propyleenoxide werden 133,5 ml bu-20 tylamine en 300 ml methanol toegevoegd en er werd 20 uren bij kamertemperatuur geroerd. Het reactiemengsel werd op een roterende verdamper geconcentreerd en het residu werd in 225 ml IN zoutzuur opgenomen. De zure oplossing werd in totaal met 300 ml dichloormethaan geëxtraheerd. De dichloormethaanfase werd verwijderd. Aan de water bevattende fase 25 werden ongeveer 70 ml 4N natriumhydroxide toegevoegd en er werd met 200 ml dichloormethaan uitgeschud. De dichloormethaanfase werd boven natriumsulfaat gedroogd. Het droogmiddel werd afgefiltreerd en het dichloormethaan werd op een roterende verdamper verwijderd.
Opbrengst : 16,9 g (35,2% van de theorie).

Voorbeeld XXVII
Bereiding van 3-(4-cyaanmethylfenoxy)propyleenoxlde uitgangsmengsel: 16,2 g ( 705 mmol) natrium 5 93,9 g ( 705 mmol) 4-hydroxybenzylcyanide 250 ml (3188 mmol) epichloorhydrine (295 g) 1 1 dimethylformamide (DMF) 2 1 abs* ethanol 10 16,2 g Natrium werden in een rondbodemkolf van 2 1 in 1 1 abs.etha nol opgelost. Vervolgens werd een oplossing van 93,9 g 4-hydroxybenzylcyanide in 1 1 abs.ethanol toegevoegd en werd goed gemengd. De doorzichtige oplossing werd op een roterende verdamper tot droog ingedampt (badtemperatuur 40eC, aanvankelijk verminderde druk opgewekt met water-15 straalpomp en later met de oliepomp). Het roodbruine residu werd onder een verminderde druk opgewekt met een oliepomp boven P^O^q gedroogd. Het gedroogde fenolaat werd in 1 1 DMF opgelost. Nadat weer ongeveer 100 ml DMF op de roterende verdamper (badtemperatuur 50eC, oliepomp) waren afgedestilleerd, werden 250 ml epichloorhydrine toegevoegd* 20 Het aanvankelijke doorzichtige reactiemengsel werd 3 uren in een olie-bad tot 100°C verhit. Het reactieverloop werd zo vervolgd (elueermid-del: ether:methanol 95:5). Het neergeslagen zout werd over een D4-frit afgescheiden en met ongeveer 200 ml DMF gewassen. De verenigde filtra-ten werden onder een verminderde druk (badtemperatuur 70°C; oliepomp) 25 ingedampt. De achterblijvende bruine olie werd zonder zuivering verder omgezet.
Opbrengst: 134 g (100% van de theorie).
Bereiding van orale toedieningsvormen van de werkzame stof met de formule 1 30 a) Bereiding van een granulaat met de verbinding met de fojramle 1_ als we£kzame_stof_____________ 46,3 gew.Z werkzame stof met de formule 1 78,28 gew.Z melksuiker-granulaat 35 6,72 gew.Z maïszetmeel 5.00 gew.Z talk 5.00 gew.Z calciumcarboxymethylcellulose 0,50 gew.Z magnesiumstearaat en 0,45 gew.Z aerosol 40 8500564 20 worden gemengd en tezamen gegranuleerd.

Uit 98 kg melksuiker (DIN 20) wordt met 14 kg isopropanol en 16 kg copolymeer van dimethylaminoethylmethacrylaat-methacrylzuurbutylester-5 methacrylzuurmethylester (50:30:20) in een menger met een gefluïdiseer-de laag een granulaat bereid, dat uit 98 gew.% melksuiker en 20 gew.% lak van het bovengenoemde copolymeer bestaat, c) BerejLding_ van ^e^t^b^et^tenmas^sa^
Het granulaat, dat de werkzame stof met de formule 1 bevat, wordt 10 met het granulaat volgens b) dat het melksuiker bevat, in een tuimel-menger tezamen met 5 gew.% talk, 5 gew.% calciumcarboxymethylcellulose, 6,72 gew.% maïszetmeel, 0,5 gew.% magnesiumstearaat en 0,02 gew.% aerosol gemengd. Vervolgens wordt de massa tot tabletten geperst. Het gerede tablet bevat ongeveer 2 gew.% werkzame stof met de formule 1.
15 Bereiding van een vulmassa voor capsules 4,45 gew.% van de werkzame stof met de formule 1 werden met 51,05 gew.% lactose, 42,5 gew.% maïszetmeel en 2 gew.% talk in een tuimelmen-ger gemengd. Vervolgens wordt de massa in harde gelatinecapsules afgevuld.
20 Bereiding van een orale toedieningsvorm met Verapamil
Granulaat van Verapamil en werkzame stof met de formule 1 a) j^e^eWing van helt ^er_apami^-^raimla.at
Verapamil wordt tezmen met polyvinylpyrrolidon op gebruikelijke wijze vochtig gegranuleerd. De samenstelling van het granulaat be-25 draagt: 96,5 gew.% Verapamil, 3,5 gew.% polyvinylpyrrolidon.
b) 69,45 gew.% werkzame stof met de formule 1, 22,74 gew.% melksuiker, 5,8 gew.% maïszetmeel, 0,25 gew.% aerosil en 1,76 gew.% polyvinylpyrrolidon als oplossing in een mengsel van isopropanol en gedestilleerd water in de verhouding 1:1 worden gemengd en tezamen gegranu- 30 leerd.
Bereiding van tabletmassa
Het Verapamil bevattende granulaat volgens a) en het werkzame stof bevattende granulaat volgens b) werden tezamen met 17,3 gew.% lactose, 7,9 gew.% maïszetmeel, 5 gew.% calciumcarboxymethylcellulose, 3,2 gew.% 35 talk, 0,6 gew.% magnesiumstearaat en 0,1 gew.% aerosil gemengd. Vervolgens wordt de massa tot tabletten geperst.
De gerede tabletten bevatten de werkzame stof met de formule 1 en Verapamil in de gew.verhouding 0,01 tot 10:1.
Bereiding van een injectie-oplossing van de werkzame stof 40 met de formule 1 8500564 21
a) Uit de werkzame £tof_met__de formule JL
0,5 gew./vol.Z werkzame stof met de formule 1 wordt tezamen met een voor het isotoon maken voldoende hoeveelheid natriumchloride in water "voor injectiedoeleinden” (Europ. Arzneibuch, deel 2) opgelost, de 5 oplossing wordt vrij van zwevende stoffen gefiltreerd, in ampullen gevuld en gesloten. De gesloten ampullen worden gesteriliseerd.
b) Met fu£os_emide 0,5 gew./vol.Z werkzame stof met de formule 1 en 2 gew./vol.Z fu-rosemide worden in 20 gew./vol.Z propaandiol-1,2 opgelost en met water 10 "voor injectiedoeleinden" tot het eindvolume aangevuld. De oplossing wordt vervolgens zoals in voorbeeld a) verder behandeld.

Werkwijze ter bereiding van de pteridineverbindingen met de 30 algemene formule 1 volgens conclusie 1, met het kenmerk, dat men 2,4,6-triamino-5-nitrosopyrimidine onder katalyse met basen met het gesubstitueerde benzylcyanide met de algemene formule 2, waarin en R2 dezelfde betekenissen bezitten zoals bij formule 1, omzet en eventueel in de zuur-additiezouten omzet.

Farmaceutische, in het bijzonder diuretisch, antlkaliuretisch, antihypertensief en cardioprotectief werkzame preparaten, met het kenmerk, dat zij pteridineverbindingen met de algemene formule 1, waarin en R2 onafhankelijk van elkaar waterstof of een eventueel vertakte alkylgroep met 1 tot 6 koolstofatomen of een benzylrest voor- 40 stellen, 8500564 ttlik......... of waarin en R2 tezamen met het stikstofatoom en eventueel onder het erbij betrekken van een of twee andere heteroatomen een heterocyclisch vijf- of zes-ringsysteem vormen, of waarin R^ en R2 tezamen met het stikstofatoom een rest 5 -N-C NH II H NH2 10 vormen, en de daarvan afgeleide, fysiologisch verdraagbare zuur-additiezouten als werkzame stof bevatten.
21. Farmaceutische prepraten volgens conclusie 20, met het kenmerk, dat zij de dagdosis van 0,2 tot 1000 mg werkzame stof bevatten.
22. Farmaceutische, in het bijzonder diuretisch, antikaliuretisch, antihypertensief en cardioprotectief werkzame preparaten volgens conclusie 20, met het kenmerk, dat zij de werkzame stof tezamen met furo-semide in de gew.verhouding 1 tot 0,25:100 bevatten.
23. Farmaceutische, in het bijzonder diuretisch, antikaliuretisch, 20 antihypertensief en cardioprotectief werkzame preparaten volgens conclusie 20, met het kenmerk, dat zij de werkzame stof tezamen met een calciumantagonist bevatten.
24. Farmaceutische preparaten volgens conclusie 23, met het kenmerk, dat zij de werkzame stof tezamen met Verapamil in de gew.verhou- 25 ding 0,01 tot 10:1 bevatten.
25. Farmaceutische, in het bijzonder diuretisch, antikaliuretisch, antihypertensief en cardioprotectief werkzame preparaten volgens conclusie 20, met het kenmerk, dat zij de werkzame stof tezamen met een salureticum bevatten.
26. Farmaceutische, in het bijzonder diuretisch, antikaliuretisch, antihypertensief en cardioprotectief werkzame preparaten volgens conclusie 25, met het kenmerk, dat zij de werkzame stof tezamen met Hydro-chlorothiazid in de gew.verhouding 1:1 tot 0,05:1 bevatten.
27. Farmaceutische, in het bijzonder diuretisch, antikaliuretisch, 35 antihypertensief en cardioprotectief werkzame preparaten volgens conclusie 20, met het kenmerk, dat zij de werkzame stof tezamen met een β-blokker, eventueel in combinatie met een salureticum bevatten. 28. l-(4-Cyaanmethylfenoxy)-3-aminopropanolen-2 met de algemene formule 2, 40 waarin R^ en R2 onafhankelijk van elkaar waterstof of een eventueel 8500564 vertakte alkylgroep met 1 tot 6 koolstofatomen of een benzylrest voorstellen, of waarin Rj_ en R£ tezamen met het stikstofatoom en eventueel onder het erbij betrekken van een of twee andere heteroatomen een heterocy-5 clische vijf- of zes-ringsysteem vormen.
29. Werkwijze ter bereiding van'de 1-(4-cyaanmethylfenoxy)-3-ami-nopropanolen met de algemene formule 2 volgens conclusie 28, met het kenmerk, dat men aan 3-(4-cyaanmethylfenoxy)-propyleenoxide het amine met de algemene formule 3, waarin R^ en R2 dezelfde betekenissen 10 bezitten als bij de algemene formule 2, addeert. 8500564 k __
