Abstract
La presente invención se refiere a un procedimiento para la producción de butadieno, caracterizado porque comprende: (a) cultivar por fermentación en una cantidad suficiente de nutrientes y medio un organismo cicrobiano que no está presente en forma natural que produce alcohol crotílico produciendo así de manera biosintética alcohol crotílico, en donde dicho organismo microbiano comprende una ruta de alcohol crotílico que comprende al menos dos ácidos nucleicos exógenos que codifican para enzimas de la ruta alcohol crotílico expresadas en una cantidad suficiente para producir alcohol crotílico, en donde dicha ruta de alcohol crotílico comprende convertir crotonil-CoA a alcohol crotílico utilizando al menos un ácido nucleico exógeno de dichos dos ácidos nucleicos exógenos, en donde dichas enzimas de la ruta de alcohol crotílico se seleccionan del grupo que consisten en; (i) una crotonil-CoA reductasa (formada de aldehído), y una crotonaldehído reductasa (formadora de alcohol); (ii) na crotonil-CoA reductasa (formadora de alcohol); y (iii) una crotonil-CoA hidrolasa, sintetasa o transferasa, una crotonato reductasa, y una crotonaldehído reductasa (formadora de alcohol), y en donde dicha ruta de alcohol crotílico comprende adicionalmente al menos un ácido nucleico exógeno de dichos dos ácidos nucleicos exógenos que codifica para una enzima de la ruta de alcohol crotílico para convertir : (i) acetil-CoA a crotonilo-CoA utilizando una aceitil-CoA: acetil-CoA aciltransferasa, una acetoacetil-CoA reductasa y una 3-hidroxibutiril-CoA deshidratasa; (ii) gulutaconil-CoA a crotonil-CoA utilizando una glutaconil-CoA descarboxilasa; (iii) glutaril-CoA o crotonil-CoA utilizando una glutaril-CoA deshidrogenasa; (iv) 3-acminobutiril-CoA a crotoni-lCoA utilizando una 3- aminobutiril-CoA desaminasa; o (v) 4-hidroxibutiril-CoA a crotonil-CoA utilizando una 4-hidroxibutiril-CoA deshidratasa; y (b) convertir el alcohol crotílico producido al cultivar dicho microorganismo microbiano que no está presente en forma natural en el pasa (a) a butadieno utilizando un producto químico.
