Abstract
En la presente se proporcionan construcciones polipeptídicas de unión al antígeno multiespecíficas que comprenden al menos dos heterodímeros distintos, cada uno de los cuales comprende una cadena pesada y una cadena ligera. Al menos un heterodímero comprende una región Fab que comprende una cadena ligera lambda y al menos un heterodímero comprende una región Fab que comprende una cadena ligera kappa. Una o más de las cadenas pesadas y ligeras de inmunoglobulina que forman la construcción polipeptídica de unión al antígeno comprenden modificaciones de aminoácidos que fomentan el correcto emparejamiento entre las cadenas pesadas y ligeras para formar la construcción polipeptídica de unión al antígeno multiespecífica deseada. Las modificaciones de aminoácidos pueden estar en los dominios CH1 y/o CL, en los dominios VH y/o VL, o una combinación de estos.
